1. We examined the effect of lipopolysaccharide (LPS), a component of Gram-negative bacteria, on food passage in the digestive tract of chickens (Gallus gallus) in order to clarify whether bacterial infection affects food passage in birds.

2. Food passage in the crop was significantly reduced by intraperitoneal (IP) injection of LPS while it did not affect the number of defecations, suggesting that LPS may affect food passage only in the upper digestive tract.

3. Similar to LPS, prostaglandin E2 (PGE2), one of the mediators of LPS, also reduced crop-emptying rate in chickens while it had no effect on the number of defecations.

4. Pretreatment with indomethacin, which is an inhibitor of cyclooxygenase (COX), a prostaglandin synthase, had no effect on LPS-induced inhibition of crop emptying.

5. IP injection of LPS did not affect the mRNA expression of COX2 in the upper digestive tract of chickens.

6. It is therefore likely that LPS and PGE2 reduced food passage rate in the crop by a prostaglandin-independent pathway in chickens.  相似文献

1. A study was conducted to estimate the prevalence and quantification by species of Campylobacter infection in broiler flocks at the end of the rearing period and to identify associated risk factors.

2. A questionnaire about the rearing period was completed and caecal samples were collected from 121 broiler flocks in Brittany, France, during 2008.

3. Campylobacter was isolated in 87 out of 121 flocks – a prevalence of 71.9% (95% CI, 63.7–80.1%), including 40.5% of Campylobacter jejuni and 29.8% of Campylobacter coli.

4. The average concentration, in positive flocks, was 7.96 log10 cfu/g and ranged from 3.15 to 10.32 log10 cfu/g.

5. The average concentration by species was: 7.57 log10 cfu/g for C. jejuni and 8.44 log10 cfu/g for C. coli.

6. There was a seasonal effect, with increased risk of Campylobacter colonisation in June, July and August (odds ratio (OR) = 9.59, 95% CI 1.15–79.75).

7. The other factors, associated with lower risk of Campylobacter colonisation, were the acidification of drinking water (OR = 0.33, 95% CI 0.13–0.86), antibiotic treatment at the beginning of the rearing period (OR = 0.20, 95% CI 0.07–0.55) and rodent control around the house (OR = 0.18, 95% CI 0.03–0.95).

8. The results show that hygiene practices and biosecurity measures could lead to a reduction in Campylobacter colonisation.  相似文献

1. Machine vision and artificial neural network (ANN) procedures were used to estimate live body weight of broiler chickens in 30 1-d-old broiler chickens reared for 42 d.

2. Imaging was performed two times daily. To localise chickens within the pen, an ellipse fitting algorithm was used and the chickens’ head and tail removed using the Chan–Vese method.

3. The correlations between the body weight and 6 physical extracted features indicated that there were strong correlations between body weight and the 5 features including area, perimeter, convex area, major and minor axis length.

5. According to statistical analysis there was no significant difference between morning and afternoon data over 42 d.

6. In an attempt to improve the accuracy of live weight approximation different ANN techniques, including Bayesian regulation, Levenberg–Marquardt, Scaled conjugate gradient and gradient descent were used. Bayesian regulation with R2 value of 0.98 was the best network for prediction of broiler weight.

2. Numbers of selected groups of faecal bacteria were not reduced by erythrosine.

1. The aim of this study was to determine the most likely time interval after infection with influenza virus H9N2 for co-infection with Escherichia coli to cause colibacillosis, the importance of lung load of E. coli and the involvement of respiratory phagocytes.

2. Specific pathogen free chickens were inoculated intranasally with 106EID50 of influenza virus or uninfected. After specified time intervals, 107 CFU E. coli or phosphate-buffered saline was inoculated. The presence of lesions, the number of respiratory phagocytes in the respiratory lavage fluid and the E. coli load in the lung were determined after different time intervals.

3. Compared with the number of lesions in chickens receiving only E. coli inoculation, the number lesions in co-infected chickens were increased at 0- and 3-d time intervals, but reduced in the groups at 6- and 9-d intervals between co-infection.

4. At 1–3 d after E. coli inoculation, the number of lesions chickens was correlated with the number of respiratory phagocytes harvested and related to the E. coli load in the lungs at 5 d.

5. These results suggest that the lesions caused by E. coli in chickens were increased within a 0–3 d interval following H9N2 virus inoculation and that this effect is related to the number of respiratory phagocytes.  相似文献

1. An experiment was conducted to compare the natural resistance of an indigenous breed of local village chickens to Salmonella gallinarum with two commercial breeds: ISA Brown and ISA White layers under experimental conditions.

2. A total of 72 chickens from each of these breeds were randomly distributed to 4 pens to provide equal numbers of two replicate pens maintained as infected and control (uninfected). All chickens in infected groups were inoculated orally with 1 × 108 CFU (1 ml dose) of a field isolate of S. gallinarum, at the age of 8 and 16 weeks given over 5 consecutive days. Growth performance, clinical signs, gross pathological lesions and antibody responses were measured.

3. A significantly higher mortality was observed in the brown layers compared with the white layers, and clinical signs and mortality were absent in village chickens. However, a large number of birds with gross lesions and high antibody titres were detected in village chickens, indicating that birds had the disease subclinically. Commercial breeds had a significantly higher body weight, feed intake and feed conversion efficiency.

4. There was a significantly lower proportion of positive reactors in village chickens in the whole-blood agglutination test (35%) compared to brown (100%) and white (90%) layers even after the second inoculation. Uninfected birds were negative in all groups. The indirect enzyme-linked immunosorbent assay confirmed these observations.

5. These results suggest that the indigenous breed had superior natural resistance to S. gallinarum than the commercial breeds.  相似文献

1. This experiment was conducted to evaluate the effects of replacing dietary cottonseed meal (CSM) or fermented cottonseed meal (FCSM) for soya bean meal (SBM) on growth performance, carcass characteristics, gastrointestinal microbial populations, and intestinal morphology of broiler chickens.

2. CSM was fermented with Bacillus subtilis, Aspergillus niger and A. oryzae for 7 d. A total of 300 one-d-old male Ross 308 broiler chickens were used in a 42-d experiment in which the birds were randomly allotted to one of 5 dietary treatments (containing 0%, 10% and 20% CSM or FCSM) in a completely randomised design. Birds were reared on litter floor and had free access to feed and water during the experiment.

3. Results indicated that the fermentation process significantly reduced crude fibre and free gossypol, while it increased crude protein content and lactic acid bacteria (LAB) count in CSM.

4. The use of FCSM instead of CSM significantly improved growth performance of broilers. The abdominal fat yield in treatments containing FCSM was significantly lower than in the other treatments. The increase in the population of LAB in the crop and decrease in the population of coliforms in the ileum of birds fed on diets containing FCSM were more significant than in other birds. Villi in the duodenum and jejunum of the birds fed on diets containing FCSM were significantly higher than for the other experimental groups.

5. The positive effects of diets containing FCSM on growth performance and intestinal health of broiler chickens showed that this processed source of protein can serve as an appropriate alternative for SBM in diets for broiler chickens.  相似文献

1. The effect of reducing food intake to 75% of the ad libitum intake was determined from hatching to 8 weeks in young Light Sussex chickens.

2. Restricted birds were lighter throughout the experiment.

3. Relative adrenal weight tended to be greater in restricted birds but the difference decreased with time.

4. There was no depletion of adrenal cholesterol: from week 5 there was a significantly greater amount in the adrenals of restricted birds.

5. After 1 week of restriction plasma corticosterone concentration was 73% greater than in controls. It decreased progressively, falling within the normal range at 5 weeks.

6. Restricted birds were hypoglycaemic from weeks 2 to 7 and hyper‐lipacidaemic throughout. A negative correlation between plasma glucose and free fatty acids was found.  相似文献

1. The protective effect of a natural Brazilian calcium montmorillonite (CaMont) against aflatoxins was studied in broiler chickens.

2. A total of 1056-d-old Cobb male broilers were housed in experimental pens (22 chickens per pen) for 42 d. Three levels of CaMont (0, 2.5 and 5 g/kg) and two levels of aflatoxins (0 and 3 mg/kg) were assayed. Each treatment had 8 replicate pens of 22 broiler chickens each.

3. Of all the chickens tested in the experiment, the ones treated with aflatoxins were the most adversely affected. CaMont treatment at concentrations of 2.5 and 5 g/kg improved body weight of chickens at 42 d of age by 13.3% and 22.7%, increased daily feed intake by 9.7% and 24.7%, and improved the productive efficiency index of chickens by 53% and 66.5%, respectively.

4. Dietary CaMont positively affected parameters such as weight of liver, heart and gizzard; however, serum potassium concentration decreased by 15.3% compared with that of chickens given only the aflatoxin-contaminated diet.

5. CaMont did not cause adverse effects in chickens that did not receive aflatoxins.

6. CaMont at pH 8.5 partially reduced the toxic effects of aflatoxins in broilers when included at levels of 2.5 and 5 g/kg in the diet.  相似文献

1. Campylobacter jejuni is the most common bacterial cause of human food-borne gastroenteritis in the world. A major source of human infection is the consumption of contaminated meat, particularly poultry. New control measures to reduce or eliminate this pathogen from the animal gastrointestinal tract are urgently required, and the use of probiotics as competitive exclusion agents is a promising biocontrol measure to reduce C. jejuni in the food chain.

2. In this study, we assessed the potential of Lactobacillus johnsonii FI9785, which has shown efficacy against Clostridium perfringens, to combat C. jejuni. The effect of prophylactic administration of L. johnsonii on the ability of C. jejuni to colonise chickens was determined.

3. Two doses of L. johnsonii given a week apart led to a reduction in C. jejuni colonisation in the caecal contents, but this biocontrol seemed reliant upon a high level of initial colonisation by the probiotic.

4. The microbial composition in the chicken gut was significantly altered by the probiotic treatment, as shown by denaturing gradient gel electrophoresis of 16S rRNA gene amplicons.

5. Together these results demonstrate the potential of this probiotic strain to be tested further as a competitive exclusion agent in poultry against C. jejuni.  相似文献

1. The effects of the dietary inclusion of a Bacillus-based direct-fed microbial (DFM) candidate on digesta viscosity, bacterial translocation, microbiota composition and bone mineralisation were evaluated in broilers consuming rye-based diets.

2. In the present study, control mash rye-based diets (CON) or Bacillus-DFM supplemented diets (TRT) were administered ad libitum to male broilers in three independent experiments.

3. In Experiments 1 and 2 (= 25/group), liver samples were taken to evaluate bacterial translocation, digesta samples were used for viscosity measurements and the intestinal microbial flora was evaluated from different intestinal sections to enumerate total recovered gram-negative bacteria (TGB), lactic acid bacteria (LAB) and anaerobic bacteria (TAB). Additionally, both tibias were removed for assessment of bone quality.

4. In Experiment 3, each experimental group had 8 replicates of 20 chickens (n = 160/group). Weekly, body weight (BW), feed intake (FI) and feed conversion ratio (FCR) were evaluated. At d 28-of-age, samples were taken to determine bacterial translocation, digesta viscosity and bone quality characteristics.

5. In all experiments, consumption of Bacillus-DFM reduced bacterial translocation to the liver and digesta viscosity. Additionally, DFM supplementation improved BW, bone quality measurements and FCR. Moreover, chickens fed on the Bacillus-DFM diet in Experiments 1 and 2 showed a significant reduction in the number of gram-negative and anaerobic bacteria in the duodenal content compared to control.

6. In summary, chickens fed on a rye-based diet without DFM inclusion showed an increase in bacterial translocation and digesta viscosity, accompanied by reduced performance and bone quality variables relative to the Bacillus-DFM candidate group. Hence, incorporation into the feed of a selected DFM ameliorated the adverse anti-nutritional effects related to utilisation of rye-based diets in broilers chickens.  相似文献

Background: Salmonella Typhimurium, a non-host-adapted Gram-negative intracellular pathogen, is capable of infecting a variety of animal hosts and humans.

Objective: This study utilized the prime-booster immunization strategy using Salmonella Typhimurium–LTB (S. Typhimurium–LTB) ghost with the aim of inducing a robust immune response for the prevention of avian salmonellosis. In addition, the effect of MontanideTM ISA 70VG adjuvant on S. Typhimurium–LTB ghost vaccination was investigated.

Animals and methods: A total of 75 chickens were divided into three groups (n=25) for intramuscular immunization: group A (non-immunized control injected with sterile PBS), group B (immunized with S. Typhimurium–LTB ghost), and group C (immunized with S. Typhimurium–LTB ghost plus MontanideTM ISA70VG adjuvant).

Results: Compared with group A, the immunized chickens (groups B and C) exhibited increased titers of antigen specific plasma IgG and intestinal secretory IgA antibodies. In addition, group C showed enhanced induction of the humoral immune response compared to group B. The populations of splenic CD3+CD4+ and CD3+CD8+ T-cells increased significantly in both immunized groups. In addition, increased mRNA expression of the Th1 cytokines, IFN-γ, and IL-2 were observed in S. Typhimurium antigen-stimulated peripheral blood mononuclear cells from groups B and C chickens. Chickens from both vaccinated groups showed significant protection against virulent S. Typhimurium oral challenge compared to non-vaccinated chickens and a lower challenge strain count was recovered from the internal organs of group C.

Conclusions: Injection of S. Typhimurium-LTB ghost with or without MontanideTM ISA70VG adjuvant is capable of inducing protective immunity against the virulent S. Typhimurium infection in chickens.  相似文献

1. An experiment was conducted to study the effect of two different diets on zootechnical performance and the major bacterial groups in association with the host mucosa and dispersed in the lumen contents of the small intestine of broiler chickens.

2. The two experimental diets were maize or sorghum-based. In addition to the total bacteria, bacterial groups belonging to the Enterobacteriaceae (Enterococcus and Lactobacillus) were quantified by real-time PCR.

3. There were no differences in body weight gain and feed intake, but feed conversion ratio increased for sorghum-fed broilers at 21 and 42 d of age.

4. The Enterococcus group decreased in all gut segments from 7 to 42 d, while the Lactobacillus group increased in both ecosystems. In the ileal mucosa, the enterobacterial counts decreased from 7 to 42 d in the maize-based diet, but remained stable in the sorghum-based diet.

5. The results shed light on the spatial and temporal distribution of bacterial groups that play important physiological roles in the small intestine of chickens. Specifically, the increased Enterobacteria population in the ileum is consistent with the relatively poor feed conversion in sorghum-fed broilers.  相似文献

1. An experiment was conducted to study the effect of yeast (Saccharomyces cerevisiae) cell wall (YCW) supplemented in diets of broiler chickens challenged with Escherichia coli lipopolysaccharide (LPS).

2. One-day-old broiler chicks were randomly distributed into 24 cages (6 replicate cages; 8 birds/cage) and were inoculated with 0 or 1 mg/kg body weight E. coli-LPS (d 4 and 9) and 0 or 500 mg YCW/kg feed, resulting in a 2 × 2 factorial arrangement of treatments. Experimental diets did not include coccidiostats, in-feed antibiotics or enzymes.

3. On d 21, the inoculation of E. coli-LPS reduced weight gain and feed intake and increased feed conversion ratio (FCR) of birds, an effect maintained until 28 d. In contrast, chickens given diets with YCW improved the FCR at both 21 and 28 d of age.

4. E. coli-LPS challenge reduced the relative weight of bursa of Fabricius, except when chickens were given YCW, which resulted in an interaction. Supplementation of broiler diets with YCW exacerbated the cellular immune response as measured by the delayed cutaneous hypersensitivity response test.

5. The results of this study suggested a benefit on feed efficiency when YCW was added to diets fed to broiler chickens challenged with E. coli-LPS. Part of the mode of action of YCW might be related to better maintenance of immune status in response to microbial challenge.  相似文献

1. The clinical severity, histological changes, indicators of gut leakiness and inflammatory cytokine profiles were studied in chickens with dextran sulphate sodium (DSS)-induced intestinal inflammation.

2. The experimental groups (1.25%, 1.5% and 2.5% DSS) showed clinical signs, such as loose stools and weight loss, which increased with additional treatment days and, as expected, the effects of DSS-induced intestinal inflammation were time and dose-dependent.

3. After 10 d, histological manifestations were evident, including goblet cell depletion, mucus layer loss, significantly shorter villi and a thinner total ileal mucosa.

4. The d(-)-lactate value, which was used as a gut leakiness indicator, was significantly increased in the 2.5% DSS group.

5. Expression of the inflammatory cytokines interleukin-1Beta, tumour necrosis factor alpha and interleukin-10 in the serum significantly increased with DSS treatment.

6. This study indicates that the experimental intestinal inflammation induced by DSS is an ideal model to study the pathogenic mechanisms of intestinal inflammation in chickens and to test the efficacy of therapies.  相似文献

1. Contaminated chicken meat remains an internationally important vehicle for human infection with Salmonella and Campylobacter spp. In addition, the last 20 years has seen an international pandemic of human salmonellosis caused by the contamination of eggs with Salmonella Enteritidis.

2. It has been a long held scientific view that Campylobacter spp. and most, if not all of the common zoonotic salmonella, are essentially commensal in chickens. They usually form part of the gut flora and contaminate chicken carcases, for example, by faecal spillage at slaughter. Even when certain salmonella serovars like S. Enteritidis are invasive in laying hens overt evidence of clinical disease is rare and the birds appear to behave normally.

3. Are these bacteria just ‘passing through’ the avian host and only transient members of the bacterial flora or is there a more dynamic perspective to this infection/colonisation process? Chickens mount antibody responses to both pathogens, which indicate something other than commensalism. Such immune responses, however, do not always result in the clearance of the pathogen.

4. Not all animals in a group will carry salmonella or campylobacter, even under experimental conditions, and will vary, especially those that are outbred, in their responses to pathogen challenge. Identifying the reasons behind this could have important implications for disease control.

5. Both salmonella and campylobacter are more likely to be found in animals, which are compromised and this may explain at least part of the variations seen. Animals are more susceptible to infection when they are in a poor environment, fed a poor diet and/or under physical or psychological stress.

6. Work in this area has naturally focused on pathogens of medical significance and has shown that neurotransmitters such as noradrenaline can markedly alter pathogen behaviour. Other host responses like Interferon γ can also affect host tissues in a way, which facilitates invasion by pathogens and may also interact directly with certain bacteria.

7. From a food safety perspective, there is evidence that egg contents contamination in ovo may be linked to transient stress in the hen. Current work at the University of Bristol on the epidemiology of campylobacter in broiler production is also showing a potential link between gut health and campylobacter colonisation and challenging the concept that these bacteria are common commensals.

8. The poor economic returns received by the egg and poultry industries mean that intensive production methods are common. Is it possible to rear chickens under these conditions in such a way as to exclude zoonotic pathogens like salmonella and campylobacter? Data from the UK strongly suggest that this is possible with the former pathogen. Can similar advances be achieved with campylobacter?  相似文献

1. A realised heritability of 0.23 was obtained in an Australorp flock (S) selected for five generations for high egg specific gravity.

2. A comparison with an unselected control flock (C) over 50 weeks of lay in the final generation indicated a number of statistically significant correlated responses in commercially important traits in the S line in addition to the direct response of + 0.004 in specific gravity.

3. With an increase in specific gravity, there was a decrease of 3.4 in the percentage of soft‐shelled eggs laid.

4. The weight and albumen height of eggs measured within 1 hour of lay declined by 1.8 g and 2.1 Haugh units respectively.

5. There were reductions in the weight and albumen height losses of eggs stored over a 10‐d period (C 0.74, S 0.62 g and C 17.9, S 15.0 Haugh units respectively), so that at the end of this period the albumen heights in both lines were the same.

6. Average body weight and daily food intake were less by 0.28 kg and 7.5 g respectively.

1. Proteolytic and amylase activities were assayed in the gut contents of germ‐free and conventional chickens aged 3 and 14 d.

2. The tendency was for the proteolytic activity to be increased and the amylase activity to be decreased in conventional chicks. These differences were statistically significant in the caecum.

3. All of the additional proteolytic activity detectable in the caecum of conventional chicks was inhibited by soyabean trypsin inhibitor. However, the increased activity was not detectable in the small intestine and was, therefore, not due to pancreatic trypsin.

1. The E3 ubiquitin protein ligase 1 (WWP1) gene, the mutation of which causes muscular dystrophy in chickens, is expressed not only in the pectoral muscle, but also in a number of tissues such as the kidney. Therefore, this study examined some parameters related to kidney function in muscular dystrophic (MD) chickens.

2. Plasma osmolality, Na+ and K+ concentrations, aldosterone levels, and the expression of aquaporin (AQP) 2, AQP3, and α subunits of the amiloride-sensitive epithelial sodium channel (αENaC) were analysed in the kidneys of 5-week-old MD chickens and White Leghorn (WL) chickens under physiological conditions or after one day of water deprivation.

3. Plasma osmolality, Na+ concentrations, and plasma aldosterone levels were significantly higher in MD chickens than in WL chickens. αENaC mRNA expression levels were lower in MD chickens than in WL chickens. AQP2 and AQP3 mRNA expression levels were similar in the two strains of chickens.

4. Plasma osmolality correlated with aldosterone levels and AQP2 and αENaC mRNA levels in WL chickens. In MD chickens, plasma osmolality correlated with AQP2 mRNA levels, but not with plasma aldosterone or αENaC mRNA levels.

5. These results suggest that neither water reabsorption nor the expression of AQP2 and AQP3 is impaired in MD chickens and that a WWP1 gene mutation may or may not directly induce an abnormality in Na+-reabsorption in the kidneys of MD chickens, potentially through αENaC.  相似文献
