Conservation and improvement strategies in farm animals should be based on a combination of genetic and phenotypic characteristics. Genotype data from 30 microsatellites were used to assess the genetic diversity and relationships among five Cuban cattle breeds (Siboney de Cuba, Criollo Cubano, Cebú Cubano, Mambí de Cuba and Taíno de Cuba). All microsatellite markers were highly polymorphic in all the breeds. The expected heterozygosity ranged from 0.67 ± 0.02 in the Taíno de Cuba breed to 0.75 ± 0.02 in the Mambí de Cuba breed, and the observed heterozygosity ranged from 0.66 ± 0.03 in the Cebú Cubano breed to 0.73 ± 0.02 in the Siboney de Cuba breed. The genetic differentiation between the breeds was significant (p < 0.01) based on the infinitesimal model (FST). The exact test for Hardy‐Weinberg equilibrium within breeds showed a significant deviation in each breed (p < 0.0003) for one or more loci. The genetic distance and structure analysis showed that a significant amount of genetic variation is maintained in the local cattle population and that all breeds studied could be considered genetically distinct. The Siboney de Cuba and Mambí de Cuba breeds seem to be the most genetically related among the studied five breeds.  相似文献

Conservation decisions based on neutral genetic diversity have been observed to promote retention of useful quantitative variation in biological populations. An experiment was undertaken to determine the association between microsatellite marker polymorphisms and phenotypic variation in semen production and cryosurvival traits in bulls. Thirty-five ejaculates were collected from ten bulls of two breeds and evaluated before and after cryopreservation for several semen traits. The bulls were also genotyped using a set of sixteen bovine-specific microsatellite marker loci. Fixation indices (FST), heterozygosity and Nei's genetic distance measures were computed from allele frequency data for each of the bulls. Molecular and phenotypic data were used to compute tri-distance matrices for the ten bulls and correlated using Mantel's test in GenAIEx 6.5. The study revealed extensive heterogeneity in semen traits, heterozygosity and FST values among the bulls. Large pairwise phenotypic and genetic distances were also observed. Correlation between pairwise genetic distances and phenotypic distances was significant and highly positive for sperm viability (r = .61, p < .001) and moderately positive for sperm motility (r = .40–42, p < .05) variables. For sperm morphology, ejaculate volume and sperm concentration, correlation with genetic distances was positive, low and not significantly different from zero (p > .05). A tendency for a triangular-shaped relationship between genetic and phenotypic distances for post-thaw motility and viability traits was also observed. Accordingly, association with neutral genetic diversity was absent for semen production traits and moderate to highly positive for sperm cryosurvival traits. Given these findings, conservation decisions based on neutral genetic diversity may capture variation in some adaptive traits, but not others.  相似文献

Four-week-old pigs of the Chester White, Duroc, Hampshire, Landrace, and Yorkshire breeds (n = 518) were vaccinated with a pseudorabies modified live-virus vaccine to determine whether genetic differences existed for immune response after vaccination. All pigs and their dams (sows) were tested before vaccination to determine preimmunization antibody titers, using a microtitration serum-neutralization (SN) test. The SN test results of sows were negative, as were preimmunization tests of the pigs. At 4 weeks after pigs were vaccinated, additional blood samples were collected from the pigs, and end-point SN titers were determined, using a 2-fold dilution scheme. Small, but statistically significant, breed differences existed for antibody response, with Yorkshire and Chester White pigs having the highest response, and Duroc and Landrace pigs, the lowest. Differences among sire progeny groups were small, but there were significant differences among dams. Genetic differences as seen by differences among breeds indicates that the efficacy of vaccines may vary from breed to breed and that vaccine trials should not neglect this potential source of variation.  相似文献

The genetic variability within the Uruguayan Creole horse and its relationship to a group of geographically or historically related breeds (Spanish Pure-bred, Barb, Quarter horse, Paso Fino, Peruvian Paso, Arabian and Thoroughbred horse), was evaluated using 25 loci (seven of blood groups, nine of protein polymorphisms and nine microsatellites) analyzed on a total of 145 Uruguayan Creole horses. In this study, blood group and protein polymorphism variants that are considered to be breed markers of Spanish Pure-bred and Barb horses were detected in the Creole breed. Conversely, some microsatellites and protein polymorphisms alleles were found uniquely in the Creole horse. American horse breeds together with Barb and Arabian horses clearly formed a separate cluster from the Spanish pure-bred and Thoroughbred breeds, as shown by an UPGMA dendrogram based on Nei's standard genetic distance. Data in this study provided evidence for considerable genetic variation within Uruguayan Creole horses and of a distinctive breed profile. Both traits were most likely inherited from the XVIth century Spanish horses, more closely related to Barb than to Spanish Pure-bred.  相似文献

Genetic evaluations for a multiple-breed population of beef cattle were used to estimate genetic trends for five breeds, and genetic differences and overlap among 14 breeds. Genetic evaluations studied were for direct contributions to birth weight, gain from birth to 200 and 365 d, and maternal contribution to gain from birth to 200 d. Almost all genetic trends were positive, but the magnitude of the trends varied among breeds. Trends were nonlinear between 1985 and 1995 for most breed and trait combinations. The rates of increase in genetic trends were generally higher for the lighter weight breeds, and lighter weight breeds had faster growth rate genetic trends at 1995 than the heavier breeds. Genetic trend estimates for yearling gain at 1995 were 2.46, 2.23, 1.73, 1.70, and 1.46 kg/yr for Angus, Hereford, Limousin, Charolais, and Simmental, respectively. Corresponding birth weight genetic trends were .130, .226, .049, .130, and .048 kg/yr. Mean genetic differences between breeds have been decreasing in magnitude due to these differences in genetic trends between heavier and lighter breeds. Genetic variation for the traits studied seemed to be greater within than between breeds for calves born and cows calving between 1993 and 1995. Genetic trends at 1995 suggest that ratios of within:between breed variation will increase and that across-breed genetic improvement initiatives for growth traits will become more important in the future.  相似文献

为研究中国地方绵羊品种之间的遗传多样性,利用6个微卫星位点,对中国6个绵羊品种群体进行遗传检测,计算了各品种群体的平均杂合度(He)和平均多态信息含量(PIC),同时分析了各品种群体内的遗传变异,进行了UPGMA聚类。结果表明,6个绵羊品种Ho为0.5446~0.7140,PIC为0.5211~0.7732;6个绵羊品种都具有较丰富的遗传多样性,其中岷县黑裘皮羊的遗传变异最高,小尾寒羊的遗传变异最低;小尾寒羊的近交系数最高,揭示该群体内近交程度可能较高。利用UPGMA方法构建了系统发生树,兰州大尾寒羊与岷县黑裘皮羊为一类,哈萨克羊与贵德黑裘皮羊为一类,泗水绵羊与小尾寒羊为一类,表明群体遗传结构与地理分布及其驯化历史相关。  相似文献

This article described the relationship among six Chinese indigenous sheep breeds.There were six microsatellite loci which used to analyze the genetic diversity.Based on the allele frequency of six sheep variations, we calculated the mean heterozygosity (Ho), which were from 0.5446 to 0.7140;The polymorphism information content (PIC) in six sheep breeds were from 0.5211 to 0.7732.Genetic variations within breed were analyzed in the study, and we clustered the UPGMA tree according to the DA genetic distance.The results showed that six breeds had relatively abundant genetic diversity, and MXB had the highest genetic variation, while SW had the lowest genetic variation.The inbreeding coefficient of SW was higher than other breeds, which revealed a higher inbreeding degree in this breed.Phylogenetic tree was constructed using UPGMA, which showed that LZD and MXB were classified as a class, HZK and GF were classified as a class, SIH and SW were classified as a class.The results revealed the genetic structure of breeds correlated with geographical distribution and history of domestication.  相似文献

Genetic variation at intron 1 of the interferon gamma gene has recently been associated with variation in faecal egg count in Romney, Merino and Soay breeds of sheep. The Texel breed is more resistant to gastrointestinal nematode infection than the Suffolk breed, based on faecal egg count. Hence, the objective of this experiment was to characterise the polymorphisms of intron 1 of the interferon gammagene in Suffolk and Texel sheep, and to determine if the characterised haplotype variants were associated with faecal egg count variation in these breeds. Intron 1 haplotypes and faecal egg measurements were determined for Suffolks (n=113) and Texels (n=135). Four haplotypes were identified in Suffolks (A, B, C and D), two haplotypes as previously described, and two previously unknown haplotypes. However, there was no association between interferon gamma intron 1 haplotype and faecal egg count in Suffolks. In contrast, two previously identified haplotypes (A and B) were present in the Texel breed, and the B haplotype was associated with resistance to nematode infection (P=0.02). These results suggest that intron 1 of the interferon gamma gene has an important role in resistance to nematode infection in the Texel breed.  相似文献

Understanding existing levels of genetic diversity of sheep breeds facilitates in situ and ex situ conservation activities. A comprehensive evaluation of US sheep breeds has not been previously performed; therefore, we evaluated the genetic diversity among and within 28 US sheep breeds. Both major and minor breeds were included in the analysis and consisted of 666 animals from 222 producers located in 38 states. The level of within-breed genetic diversity was variable and not dependent upon status of a breed as a major or minor breed. Bayesian cluster analysis indicated the breeds were grouped more by physiological differences (meat vs. wool production) rather than geographic origin. Results suggest several actionable items to improve in situ and ex situ conservation. The results clearly identify breeds in need of increased in situ and ex situ management (e.g., Hog Island and Karakul) and allow several suggestions for in situ management of flocks. Conversely, several of the breeds appear genetically similar and therefore require less emphasis on collecting germplasm samples for the gene bank. Commercially important breeds (e.g., Rambouillet and Suffolk) were found to have substantial variation, which should enable breeders to proceed, unencumbered by genetic diversity concerns, with selection strategies that maximize profit.  相似文献

The present study aimed at analyzing the genetic variability of indigenous goat breeds (Capra hircus) using the MHC-associated microsatellite markers BF1, BM1818, BM1258, DYMS1, and SMHCC1. The following breeds were included: Chinese Xuhuai, Indian Changthangi and Pashmina, Kenyan Small East African (SEA) and Galla, and Albanian Vendi. To examine genetic variability, the levels of heterozigosity, degrees of inbreeding, and genetic differences among the breeds were analyzed. The mean number of alleles ranged from nine in the Galla to 14.5 in the Vendi breed. The mean observed heterozygosity and mean expected heterozygosity varied from 0.483 in the Vendi to 0.577 in the Galla breed, and from 0.767 in the SEA to 0.879 in the Vendi breed, respectively. Significant loss of heterozygosity (p < 0.01) indicated that these loci were not in Hardy-Weinberg equilibrium. The mean FIS values ranged from 0.3299 in the SEA to 0.4605 in the Vendi breed with a mean value of 0.3623 in all breeds (p < 0.001). Analysis of molecular variance indicated that 7.14% and 4.74% genetic variation existed among the different breeds and geographic groups, whereas 92.86% and 95.26% existed in the breeds and the geographic groups, respectively (p < 0.001). The microsatellite marker analysis disclosed a high degree of genetic polymorphism. Loss of heterozygosity could be due to genetic drift and endogamy. The genetic variation among populations and geographic groups does not indicate a correlation of genetic differences with geographic distance.  相似文献

This study was conducted to evaluate the genetic diversity of Blonde d'Aquitaine, a well-muscled native French beef breed, and to understand the relationships between Blonde d'Aquitaine, Limousin and Salers. We also compared these three beef breeds to the Holstein dairy breed. For this purpose, a set of 16 microsatellite markers were investigated. The obtained results show that Blonde d'Aquitaine has a high level of genetic diversity. Our study shows also that the French beef breeds have genetic differentiation among them, with approximately 9% of the total variation owing to breed differences. Our results show also that Blonde d'Aquitaine and Salers populations are genetically more similar to each other than to the Limousin.  相似文献

Genetic polymorphisms in genes related to neurotransmitters or hormones affect personality or behavioral traits in many animal species including humans. In domestic animals, the allele frequency of such genes has been reported to be different among breeds and it may account for breed differences in behavior. In this study, we investigated breed differences in horses in the dopamine receptor D4 gene (DRD4), which has been reported to affect horse personality. We collected samples from seven horse breeds including those native to Japan and Korea, and compared the sequence of the DRD4 exon3 region among these breeds. We found that there were two types of polymorphisms (VNTR and SNPs) in the exon3 region, and some of them seemed to be breed-specific. In addition, we found that the allele frequency of G292A, reported to be associated with horse personality, differed greatly between native Japanese horses and Thoroughbred horses. The frequency of the A allele which is associated with low curiosity and high vigilance, was much lower in native Japanese horses (Hokkaido, 0.03; Taishu, 0.08) than in Thoroughbreds (0.62). This difference may account for breed differences in personality or behavioral traits. Further studies of the function of these polymorphisms and their effect on behavior are indicated.  相似文献

利用RFLP法检测了MyoD基因在10个中外猪种及部分杂交群体中的分布情况,分析了各群体内MyoD基因的遗传分布、遗传变异、群体杂合性等群体遗传信息,并进一步以各群体基因频率为基础,计算出群体间遗传距离和进化距离,根据进化距离对群体进行聚类,重建了系统发生树。结果表明:MyoD基因内含子1的DdeI酶切位点上不同基因型的分布在多数群体中都服从Hardy-Weinberg平衡,但在杜洛克和DLY群体中发生偏离。总体上讲,各试验群体的遗传多样性较丰富,群体遗传变异性较高,进化过程中受到自然选择压的作用,选择潜力较大。在系统发生树上,10个群体被分为4个分枝。分别是长白猪血缘、原始地方品种、杜洛克血缘和高原藏猪分枝。这一结果与各猪种的育种过程有较高的吻合性。说明部分功能基因的RFLP数据可用于近缘物种间的遗传分化研究。  相似文献

Italian Maremmana, Turkish Grey and Hungarian Grey breeds belong to the same Podolic group of cattle, have a similar conformation and recently experienced a similar demographic reduction. The aim of this study was to assess the relationship among the analysed Podolic breeds and to verify whether their genetic state reflects their history. To do so, approximately 100 single nucleotide polymorphisms (SNPs) were genotyped on individuals belonging to these breeds and compared to genotypes of individuals of two Italian beef breeds, Marchigiana and Piemontese, which underwent different selection and migration histories. Population genetic parameters such as allelic frequencies and heterozygosity values were assessed, genetic distances calculated and assignment test performed to evaluate the possibility of recent admixture between the populations. The data show that the physical similarity among the Podolic breeds examined, and particularly between Hungarian Grey and Maremmana cattle that experienced admixture in the recent past, is mainly morphological. The assignment of individuals from genotype data was achieved using Bayesian inference, confirming that the set of chosen SNPs is able to distinguish among the breeds and that the breeds are genetically distinct. Individuals of Turkish Grey breed were clearly assigned to their breed of origin for all clustering alternatives, showing that this breed can be differentiated from the others on the basis of the allelic frequencies. Remarkably, in the Turkish Grey there were differences observed between the population of Enez district, where in situ conservation studies are practised, and that of Bandirma district of Balikesir, where ex situ conservation studies are practised out of the original raising area. In conclusion, this study demonstrates that molecular data could be used to reveal an unbiased view of past events and provide the basis for a rational exploitation of livestock, suggesting appropriate cross-breeding plans based on genetic distance or breeding strategies that include the population structure.  相似文献

The objective of this study was to estimate the effects of breed, sex, and halothane genotype on fatty acid composition and several fatty acid indices of lipid extracted from porcine LM. Purebred Yorkshire (n = 436), Duroc (n = 353), Hampshire (n = 218), Spotted (n = 187), Chester White (n = 173), Poland China (n = 124), Berkshire (n = 256), and Landrace (n = 187) pigs (n = 1,934; 1,128 barrows and 806 gilts) from 1991, 1992, 1994, and 2001 National Barrow Show Sire Progeny Tests were used. Pigs were classified as the HAL-1843 normal (NN) genotype (n = 1,718) or the HAL-1843 carrier (Nn) genotype (n = 216). For statistical analysis, a mixed model was used that included fixed effects of breed, sex, halothane genotype, test, slaughter date, interaction of breed x sex, and random effects of sire and dam within breed. Breed significantly affected the concentration of individual fatty acids, total lipid content, and the values of several fatty acid indices of LM. Duroc pigs had the greatest (P < 0.01) content of total SFA. Total MUFA concentration in Poland China pigs was greater (P < 0.05) than in all other breeds except the Spotted (P > 0.05). The concentrations of total PUFA were greater (P < 0.01) in Hampshire, Landrace, and Yorkshire pigs compared with those of other breeds. Significant sex differences for individual fatty acids were detected. Compared with gilts, barrows had greater (P < 0.01) concentrations of SFA and MUFA but lower (P < 0.01) total PUFA. Halothane genotype was a significant source of variation for the percentages of some fatty acids. Pigs with the carrier (Nn) genotype had lower concentrations of SFA (P < 0.05) and MUFA (P < 0.01) but a greater concentration of PUFA (P < 0.01) compared with NN pigs. There were significant negative correlations between total lipid content and individual PUFA and significant positive correlations between lipid concentration and most individual SFA and MUFA. In conclusion, the results suggest that breed and sex are important sources of variation for fatty acid composition of LM.  相似文献

The distribution of genetic diversity and the genetic relationships among western Mediterranean horse breeds were investigated using microsatellite markers. The examined sample included seven Spanish and three Italian local horse breeds and populations, plus a Spanish Thoroughbred outgroup. The total number of animals examined was 682 (on average 62 animals per breed; range 20–122). The microsatellite marker set analysed provided 128 alleles (10.7 alleles per locus). Within‐breed genetic diversity was always high (>0.70), with breeds contributing about 8% of the total genetic variability. The mean molecular coancestry of the entire population examined was 0.205, Losino being the breed that contributed most. In addition to Nei's standard and Reynolds’ genetic distances, pair‐wise kinship distance and molecular coancestry were estimated. Remarkably similar breed rankings were obtained with all methods. Clustering analysis provided an accurate representation of the current genetic relationships among the breeds. Determining coancestry is useful for analysing genetic diversity distribution between and within breeds and provides a good framework for jointly analysing molecular markers and pedigree information. An integrated analysis was undertaken to obtain information on the population dynamics in western Mediterranean native horse populations, and to better determine conservation priorities.  相似文献

It was hypothesized in these studies that differences would exist in baseline immune and performance measures among different breeds of pigs, and that these differences would be affected by age of the pig. Baseline immune, plasma cortisol (CORT) concentrations, and performance measures were determined among Berkshire (n = 36), Duroc (n = 18), Meishan (n = 54), Landrace x Yorkshire (White X; n = 36), and Yorkshire (n = 36) pigs at 4, 8, and 12 wk of age. All piglets were weaned at 17 to 21 d of age and moved to a common nursery environment. Total white blood cell (WBC), leukocyte differential, plasma CORT, immunoglobulin G (IgG) concentrations, natural killer cytotoxicity, neutrophil phagocytosis (PHAGO), and chemotaxis (CHTX) were evaluated. At all ages, plasma CORT was greatest in Meishan pigs, and least in Yorkshires (P < 0.05). Plasma IgG increased with age for all breeds (age: P < 0.01; breed x age: P < 0.005), except that in Meishans, IgG decreased. Natural killer cytotoxicity was greatest (P < 0.05) among Meishan pigs. There were breed x age interactions for neutrophil PHAGO (P < 0.001) and CHTX (P < 0.001). Overall, Yorkshire pigs showed the greatest (P < 0.05) percentage of PHAGO but the least (P < 0.05) CHTX. White X pigs had the greatest (P < 0.05) CHTX response. Berkshire pigs had the greatest (P < 0.001) numbers of neutrophils. At 12 wk of age, Meishan pigs had the least BW gain (P < 0.001), and Durocs had the greatest G:F (P < 0.001). There were no significant sex differences for immune (P > or = 0.15), performance (P > or = 0.20), or CORT (P = 0.70) measures. Pig breed and age influenced both baseline immune measures and plasma CORT in growing pigs, suggesting that pig breed and age are important factors influencing the response to various stressors or infectious challenges.  相似文献

Sexual performance has been subjectively measured with a libido test during screening of rams before public sale and breeding at the U.S. Sheep Experiment Station from 1990 to 2000. The objective of this study was to determine whether sexual performance was genetically influenced. Sexual performance scores ranged from 1 to 6 with scores increasing from sexually inactive to highly sexually active in the presence of estrous ewes. The overall average score was 3.5+/-0.02. Records from four breeds (Columbia, n = 807; Polypay, n = 1,668; Rambouillet, n = 1,208; and Targhee, n = 1,002) were combined into one analysis because breeds had similar phenotypic variances. Total number of records was 4,685, which included a second sexual performance test on 1,212 rams in the following year. Variance components were estimated using a REML algorithm. Fixed effects were breed of ram, selection line within breed, and year by breed. A permanent environmental effect for ram was included to account for repeated observations on individual animals. Age and weight of the rams at time of the libido test were linear covariates and were breed-specific. Adjusted means for sexual performance scores did not differ among breeds (P > 0.05). Age was a significant effect (P < 0.01), with sexual performance score increasing 0.05 units for each month of age. The additive genetic variance was estimated as 0.54. The estimate of variance due to ram permanent environmental effects was 1.19. The residual variance was estimated to be 0.67. The heritability estimate was moderate (0.22+/-0.04) and repeatability was high (0.72). These results imply that one screening for sexual performance provides a reliable measure of sexual performance and that favorable response to selection for ram serving capacity may be expected.  相似文献

Genetic diversity was compared among eight dog breeds selected primarily for conformation (Standard Poodle, Italian Greyhound and show English Setter), conformation and performance (Brittany), predominantly performance (German Shorthaired and Wirehaired Pointers) or solely performance (field English Setter and Red Setter). Modern village dogs, which better reflect ancestral genetic diversity, were used as the standard. Four to seven maternal and one to two Y haplotypes were found per breed, with one usually dominant. Diversity of maternal haplotypes was greatest in village dogs, intermediate in performance breeds and lowest in conformation breeds. Maternal haplotype sharing occurred across all breeds, while Y haplotypes were more breed specific. Almost all paternal haplotypes were identified among village dogs, with the exception of the dominant Y haplotype in Brittanys, which has not been identified heretofore. The highest heterozygosity based on 24 autosomal microsatellites was found in village dogs and the lowest in conformation (show) breeds. Principal coordinate analysis indicated that conformation‐type breeds were distinct from breeds heavily used for performance, the latter clustering more closely with village dogs. The Brittany, a well‐established dual show and field breed, was also genetically intermediate between the conformation and performance breeds. The number of DLA‐DRB1 alleles varied from 3 to 10 per breed with extensive sharing. SNPs across the wider DLA region were more frequently homozygous in all pure breeds than in village dogs. Compared with their village dog relatives, all modern breed dogs exhibit reduced genetic diversity. Genetic diversity was even more reduced among breeds under selection for show/conformation.  相似文献
