Hypomagnesaemic tetany was induced in non-lactating and lactating ewes by feeding them semi-synthetic low magnesium diets containing additional potassium chloride and citric acid. Aqueous and vitreous humour were sampled from one eye at the time of death (fresh) and from the second eye after the head had been stored at ambient temperature for 24 hours (24-hour). There were significant relationships between the concentrations of magnesium in cerebrospinal fluid and plasma and its concentrations in fresh aqueous humour and fresh vitreous humour. Magnesium concentrations of < 0.33 mmol/litre in fresh aqueous humour and < 0.50 mmol/litre in 24-hour aqueous humour were associated with severe hypomagnesaemia and tetany. However, the concentration of magnesium in aqueous humour is relatively unstable and, unless the time of death was known accurately, its interpretation would be difficult. Magnesium concentrations of < 0.60 mmol/litre in fresh vitreous humour and < 0.65 mmol/litre in 24-hour vitreous humour were associated with severe hypomagnesaemia and tetany in adult sheep. The concentration of magnesium in vitreous humour was relatively stable for up to 48 hours postmortem.  相似文献

Monoamines are important brain neurotransmitters. An investigation was carried out to determine if hypomagnesaemic tetany was associated with alterations in regional brain monoamine concentrations in bovines. The results, established in cows with normal magnesium status, demonstrated that regional differences existed in the distribution and concentration of brain monoamines in the adult bovine, which were similar to those in other species. In magnesium-deficient cows, severe hypomagnesaemia and lowered cerebrospinal fluid (CSF) magnesium concentrations were associated with significant alterations in monoamine concentrations in some brain regions. Alterations in 3,4-dihydroxyphenylalanine (DOPA) and dihydroxyphenylacetic acid (DOPAC) concentrations in the corpus striatum, and dopamine (DA) in the cerebral cortex and cerebellum were recorded. These regions play an important role in both voluntary and involuntary motor function, and therefore these alterations may play a role in the aetiology of hypomagnesaemic tetany. However, there was no significant change in DA concentrations in the corpus striatum (the main dopaminergic region in the brain) associated with hypomagnesaemia. In addition, a significantly lower norepinephrine (NE) concentration in the corpus striatum of hypomagnesaemic animals was also recorded. Norephinephrine is generally excitatory and therefore lowered NE concentrations would be expected to result in depression rather than stimulation of motor function.  相似文献

Administration of 1,25 dihydroxyvitamin D3 (1,25(OH)2D3) to lactating dairy cows resulted in increased dietary calcium absorption and elevated concentrations of plasma calcium. Dietary magnesium absorption was unaffected by 1,25(OH)2D3 however, plasma magnesium concentration was depressed. Injections of 1,25(OH)2D3 were effective in elevating plasma calcium concentrations in both normal and hypomagnesaemic cows. This indicates a potential use for 1,25(OH)2D3 to prevent and treat hypocalcaemic cows with or without concurrent hypomagnesaemia.  相似文献

Samples of ventricular and lumbar cerebrospinal fluid (CSF) were taken at intervals from six lactating dairy cows, made hypomagnesaemic by feeding a magnesium-deficient diet. Initially the ventricular CSF had a higher magnesium concentration than lumbar CSF but as the cows became hypomagnesaemic the magnesium concentration in ventricular CSF decreased more rapidly than that in lumbar CSF. Tetany occurred when the concentration of magnesium in ventricular CSF fell below about 0.5 mmol litre-1. Intravenous infusion of magnesium into two hypomagnesaemic cows increased the concentration of magnesium in ventricular CSF before there was any change in the lumbar CSF. Newly formed CSF from the ventricles appeared to be more sensitive to changes in the concentration of magnesium in plasma than lumbar CSF which equilibrates with the diffusible magnesium of the central nervous system.  相似文献

Magnesium, calcium, phosphate, total protein and free fatty acid concentrations, and a lipase activity using p-nitrophenylpalmitate NP-C16) as a substrate, were determined in the serum of 15 pregnant Aberdeen Angus cows grazing a Phalaris tuberosa pasture with a tendency to induce tetany. A gradual decrease in magnesium concentrations, but no definite change in the other parameters, was observed when the results were plotted directly against the time of the experiment. When the results were plotted as a function of the stage of pregnancy or lactation of the animals, magnesium concentrations showed a more steady decrease which was associated with increases in calcium concentrations and, at the lowest magnesium concentrations, with increases in phosphate concentrations. Total proteins and free fatty acids remained fairly constant and the highest lipase activity was associated with parturition. The lowest magnesium concentrations in serum did not correspond with high levels of either lipase or free fatty acids. The hypomagnesaemia observed in this experiment was attributed to the high potassium:calcium and magnesium ratio in the pasture and to the physiological condition (pregnant and lactating) of the animals. None of the parameters evaluated in this paper would be more sensitive than serum magnesium levels for the early detection of the conditions that predispose grazing cattle to grass tetany.  相似文献

The relationship between intensity of production and the occurrence of hypomagnesaemia and grass tetany in dairy cattle was investigated from 1966-1970 at the Ellinbank Dairy Research Station, Warragul, Victoria. Hypomagnesaemia and/or grass tetany occurred in cows grazing at both high (3.2 cows/hectare) and low (2.2 cows/hectare) stocking rates over the four-year period. Pasture Mg concentrations remained relatively high (0.2% dry matter) throughout this period, except in the low stocking rate pastures in one year, 1967. It is concluded that hypomagnesaemia and grass tetany was precipitated by a combination of low dry matter intake in winter and the effects of a gradual increase of K concentration in autumn and winter pastures, following the heavy application of K fertilizer. Under these conditions, MgO supplementation was inadequate to prevent grass tetany occurring in some animals.  相似文献

Hypomagnesaemic tetany in cows develops occasionally after an acute increase in K intake such as can occur when cows are transferred to spring grass. There is evidence that under these conditions plasma Mg concentrations are only transiently decreased. In this study the questions addressed were whether the plasma Mg concentration, indeed adapts to a high K intake as only dietary variable, and whether urinary Mg excretion is associated with this adaptation. Dry cows were fed rations containing either 26 or 50 g K/kg dm, the extra K being in the form of KHCO3. When the cows were acutely transferred from the low to the high K ration, plasma Mg concentrations fell slightly, but significantly from 0.86 to 0.76 mmol/l within five days, but rose again to 0.80 mmol/l after another 23 days, this rise being also statistically significant. None of the animals developed tetany. The decrease in plasma Mg concentration in individual animals after five days on the high-K ration ranged from 6 to 21%. The time course of urinary Mg excretion resembled that of plasma Mg concentration; minimum Mg excretion was seen after four to six days on the high-K ration with a subsequent increase thereafter. To explain the transient lowering of plasma Mg concentration, it is suggested that the K-induced decrease in Mg status caused a delayed increase in the carrier-mediated component of Mg absorption, which in turn caused an increase in urinary Mg excretion. When the cows were acutely switched from the high to the low K ration, plasma Mg concentration and urinary Mg excretion rose, but no transient changes were seen.  相似文献

Samples of blood and milk were taken from clinically ketotic cows at 12-hour intervals before and after treatment The concentration of beta-hydroxybutyrate was determined in plasma and milk, and the concentration of acetone was determined in milk. Measurement of milk acetone had the optimal combination of sensitivity, specificity and positive predictive value to identify ketotic cows and was subsequently used in a comparison of the fertility of ketotic and normal cows. Two samples of milk were taken approximately two weeks apart, between 12 and 60 days after calving, from 410 cows on three farms and the concentration of acetone in the milk was measured. Thirty cows with milk acetone concentrations of at least 0.4 mmol/litre were compared with paired control cows with milk acetone concentrations less than 0.3 mmol/litre. The ketotic cows had a significantly longer calving-to-conception interval and a significantly higher culling rate due to a failure to conceive (P<0.05).  相似文献

Sheep given a liquid diet low in calcium and magnesium by infusion directly into the abomasum developed concurrent hypocalcaemia and hypomagnesaemia, with plasma concentrations of calcium and magnesium decreasing to 2.0 and 0.4 mmol/l respectively. Treatment of these hypomagnesaemic sheep with 1,25 dihydroxyvitamin D3 (1,25(OH)2 D3) increased the plasma calcium, magnesium and phosphorus concentrations with plasma calcium increasing to 2.5 mmol/l and plasma magnesium to 0.6 mmol/l. Plasma magnesium increased despite a small but significant increase in the daily excretion of magnesium in the urine, and the amount of magnesium derived from either bone and/or intestine must have been greater than the amount lost in the urine. Since in other experiments we have demonstrated that plasma calcium remains within the normal range when a liquid diet adequate in magnesium but low in calcium is infused, these results imply that either synthesis of and/or end organ response to 1,25(OH2) D3 is impaired in magnesium deficient sheep.  相似文献

The hypothesis that the toxic effects of imidocarb mediated by reduced cholinesterase activity might be intensified by hypomagnesaemia was tested in calves. Hypomagnesaemia was induced in 12 males (50 kg) using an artificial milk based on a commercial nondairy coffee creamer. Although plasma magnesium levels reached 0.33 mmol litre-1 in two weeks no clinical signs were detected. In 12 control calves a daily magnesium supplement of 0.6 g was inadequate although the published requirement is 0.45 g; it was raised to 1.2 g to keep plasma magnesium normal. Lighter calves developed hypomagnesaemia more readily and fast-growing calves had lower plasma urea concentrations. Plasma calcium, but not plasma magnesium, showed significant positive correlation with plasma albumin. The only statistically significant effects of hypomagnesaemia were slight elevations of white cell count and plasma sodium. The hypomagnesaemic and normomagnesaemic calves were divided into two equal groups and treated with 3.3 mg kg-1 of imidocarb dipropionate or a placebo. The drug produced the expected clinical signs of mild toxicity and depression of cholinesterase but no other adverse effects. Transient slight depressions of plasma calcium and potassium concentration, a transient rise of plasma sodium and elevation of creatine kinase occurred. None of the effects of imidocarb treatment was intensified by hypomagnesaemia except, perhaps, constriction of the pupils; generally, hypomagnesaemic animals were affected less.  相似文献

The bone activity and bone mineral content in rib bones resected from 33 dairy cows between 3 and 8 h after parturition were measured, and the effects upon them of a deficient supply of dietary magnesium (Mg) during the last seven weeks of pregnancy were studied. The cows were fed a diet containing either 0.22% magnesium (low Mg) or 0.82% magnesium (high Mg) in the dry matter (DM), and the potassium content of both rations was increased to approximately 4.1% in the DM to reduce the absorption of magnesium. In the cows fed the low-Mg diet a fall in plasma Mg concentration was observed. In the low-Mg, low-parity cows the plasma Mg concentrations at parturition were higher than in the low-Mg, high-parity cows, i.e. 0.83 mmol/l and 0.54 mmol/l, respectively. After parturition four cows in the low-Mg, high-parity group showed clinical signs of hypocalcaemia but none of the other groups did so. The bone formation in low-parity cows was significantly (P less than 0.05) affected by Mg supply, with higher percentages of both trabecular surface covered by osteoid and osteoid volume in the low-Mg group. In the high-parity cows no significant differences in bone formation were found between the low- and high-Mg groups. An inadequate Mg supply resulted in a significantly (P less than 0.05) higher Ca content in the bone ash of low-parity cows and a significantly (P less than 0.05) higher bone ash percentage in the bone of high-parity cows.  相似文献

The association between the concentrations of various blood constituents and the responses of cows with milk fever following calcium borogluconate therapy was determined using analysis of variance. In cows which had not received herdsman treatment prior to sampling (n = 63), plasma potassium was the only constituent with a significant effect (p less than 0.01), the mean concentrations for cows classified as unassisted, assisted, or delayed, recoveries, and deaths ranging down from 4.3 to 2.8 mmol/l, respectively. In cows which had received treatment prior to sampling (n = 55) those making unassisted recoveries had significantly lower mean concentrations of serum total calcium than those making assisted recoveries (1.3 v 1.9 mmol/l, respectively, p less than 0.05), and cows making delayed recoveries had significantly lower mean plasma inorganic phosphate concentrations (0.3 mmol/l) than those making unassisted and assisted recoveries (both 0.7 mmol/l, p less than 0.05) and those subsequently dying (1.2 mmol/l, p less than 0.01). Response was not dependent on plasma potassium concentrations in the treated group of cows, and plasma sodium, serum magnesium, and erythrocyte sodium concentrations were not significant in either group of cows.  相似文献

Propionate was recently shown to increase leptin synthesis in rodents. To determine if a similar effect occurs in ruminants, propionate was administered to lactating dairy cows. In experiment 1, 31 cows were given an intrajugular Na propionate bolus (1,040 micromol/kg body weight), increasing plasma propionate from 160 to 5,680 microM and plasma insulin from 6.8 to 77.8 microIU/mL. Plasma leptin concentration decreased from 2.11 ng/mL before bolus to 1.99 ng/mL after dosing (P<0.05) with no differences in leptin concentrations at 20, 50, and 100 min post-bolus (P>0.10). In experiment 2, 12 cows were used in a duplicated 6 x 6 Latin square experiment to assess the dose-response effect of ruminal propionate infusion on plasma leptin concentration. Sodium propionate was infused at rates of 0, 260, 520, 780, 1040, or 1,300 mmol/h, while total short-chain fatty acid infusion rate was held constant at 1,300 mmol/h by addition of Na acetate to the infusate. Coccygeal blood was sampled following 18 h of infusion. Increasing the rate of propionate infusion linearly increased plasma propionate concentration from 180 to 330 microM (P<0.001) and plasma insulin concentration from 6.7 to 9.1 microIU/mL (P<0.05). There was a quadratic response in plasma leptin concentration (P=0.04) with a maximum at 780 mmol/h propionate, but leptin concentrations increased by no more than 8% relative to the 0 mmol/h propionate infusion. Leptin concentrations were correlated with insulin concentrations but not with propionate concentrations in plasma. Propionate is not a physiological regulator of leptin secretion in lactating dairy cows.  相似文献

Acid-base balance and electrolyte concentrations, including ionised calcium, were monitored during intravenous fluid therapy of 11 collapsed diarrhoeic suckler calves aged five to 10 days. Six healthy calves of similar age and type were used to provide control data. All the diarrhoeic calves were severely acidotic (TCO2<12 mmol/litre). Isotonic sodium bicarbonate (1-3 per cent) was administered until the metabolic acidosis was half corrected, as indicated by the TCO2 increasing to 17 to 24 mmol/litre when the infusion was changed to an extracellular volume replacement fluid containing 144 mmol/litre Na+, 35 mmol/litre HCO3-, 4 mmol/litre K+ and 113 mmol/litre Cl- which was administered until the calf was discharged. Milk feeding was started as soon as the calf had a suck reflex. The treatment was successful in 10 calves. At admission the diarrhoeic calves were hypocalcaemic compared with the control calves, but their ionised calcium was significantly higher, with significantly less calcium being protein bound. Treatment with isotonic sodium bicarbonate resulted in a significant improvement in acid-base balance, but both total and ionised calcium decreased significantly, the decrease in ionised calcium being proportionately greater owing partly to a significant increase in the protein binding of calcium. The mean total, bound and ionised calcium concentrations were all significantly lower in the treated calves after they had received isotonic sodium bicarbonate than in the control calves. Further treatment with replacement fluid had no significant effect on any of the parameters apart from pCO2 which increased significantly. Milk feeding had no significant effect on plasma calcium concentrations. The calves' mean ionised calcium concentration was significantly lower at the end of the treatment than before it, but there was no difference in the mean total and bound calcium concentrations. The calves' mean plasma potassium and magnesium concentrations decreased significantly during the course of the treatment.  相似文献

Electrolytes, metabolites, cortisol and reproductive hormones were measured in maternal plasma taken at least twice daily from three cases of bovine hydrops before, during and after parturition induced by dexamethasone or prostaglandin. Caesarean operations were required for two of the cases. Maternal plasma electrolytes remained within the normal range, but average potassium and creatinine concentrations were higher (9.2 and 0.68 mmol/litre, respectively) than normal (4.7 and 0.42 mmol/litre) in samples of amniotic fluid obtained at calving. Sodium (100 mmol/litre) and chloride (67 mmol/litre) in allantoic fluid were also higher than normal (53 and 20 mmol/litre, respectively). Conversely, creatinine concentrations were lower than normal in allantoic fluid (2.2 vs 13.8 mmol/litre). Oestradiol concentrations were lower than normal in maternal plasma (ranges: less than 20 to 140 pg/ml vs 30 to 440 pg/ml); maximum prostaglandin F metabolite (PGFM) concentrations were slightly elevated (ranges 1.1 to 2.0 ng/ml vs 0.4 to 0.9 ng/ml). Progesterone and cortisol concentrations remained within the normal range; the latter hormone increased markedly in parallel with raised PGFM concentrations. In two cases, the concentrations of reproductive hormones tended to be lower in the amniotic fluid than in the allantoic fluid. For example, progesterone concentrations were 42.8 and 14.9 ng/ml in the amniotic fluids vs 64.2 and 29.8 ng/ml in the allantoic fluids of the two cows; PGFM concentrations were 27.7 and 4.3 ng/ml vs 34.6 and 5.0 ng/ml, and oestradiol concentrations were 1.5 and 3.5 ng/ml vs 1.1 and 6.4 ng/ml in the two fluids, respectively.  相似文献

The influence of a deficient magnesium supply during the dry period on the ability of dairy cows to mobilise calcium at parturition was investigated. The daily rations pre-partum consisted of 6 kg grass hay, and 4 kg concentrates containing either 1.58 per cent magnesium (HMg) or 0.22 per cent magnesium (LMg). Nine cows of parity one to six received the LMg diet and 10 cows of parity one to seven the HMg diet. Both rations provided 1.40 times maintenance on an energy basis and contained 4 per cent potassium in the dry matter. Throughout the dry period the mean concentration of magnesium in the plasma of the LMg group was significantly lower than in the HMg group and at parturition the mean plasma concentrations were 0.65 mmol litre-1 and 1.16 mmol litre-1, respectively. After parturition two cows in the LMg group showed clinical signs of hypocalcaemia but none of the HMg group did so. Cows of the LMg group had a lower mean rate of calcium mobilisation (0.27 mmol min-1) than those of the HMg group (0.34 mmol min-1) but the difference could not be explained by differences in bone turnover as reflected by urinary hydroxyproline excretion. Such a reduced rate of calcium mobilisation may be a contributory factor in the high incidence of milk fever in some herds fed inadequate magnesium in the dry period.  相似文献

Severe hypomagnesaemia was induced in lactating cows and lactating sheep by feeding them magnesium-deficient diets for 17 and 14 days, respectively. Hypomagnesaemia in cows was associated with abnormally high rates of change in the numbers of leucocytes, neutrophils, monocytes and platelets. There were increases in the concentration of iron in the liver of the hypomagnesaemic ewes and in the heart of the hypomagnesaemic cows, which were not associated with a haemolytic process. The percentage of some of the peroxidisable fatty acids was lower in the heart tissue of hypomagnesaemic cows, but the reduction was not associated with significant lipid peroxidation.  相似文献

Thirty-seven of 53 diarrhoeic calves hospitalised for intravenous fluid therapy were classified as very severely acidotic (total carbon dioxide less than 8 mmol/litre) by using a Harleco apparatus. All the calves were given intravenously 10 to 20 litres of electrolyte solution which contained 144 mmol/litre sodium, 4 mmol/litre potassium, 113 mmol/litre chloride and 35 mmol/litre bicarbonate, and in addition the 37 very severely acidotic calves received 400 ml of 1M sodium bicarbonate in the first 5 litres of fluid administered. Sixteen of the 37 very severely acidotic calves had a distended right flank, suggesting the presence of a dilated fluid-filled viscus. Neither their history nor other clinical signs were useful predictors of the distension. The distended calves had significantly higher plasma concentrations of sodium and chloride, and significantly lower plasma creatinine concentrations than the calves which were not distended. Treatment was successful in all the 21 non-distended calves but four of the distended calves died despite treatment. The resolution of the distension in the successfully treated calves, coincided with a significant increase in plasma bicarbonate concentration and the passage of large amounts of malodorous mucoid faeces.  相似文献

Haematological and blood biochemical changes were studied in nine camels after maximal exercise over 4 or 5 km. There was a lack of splenic reserve for red blood cells, indicated by a minimal increase in haemoglobin concentration and haematocrit. There were marked increases in plasma lactate (to over 20 mmol/litre), plasma ammonia and plasma glucose and a pronounced decrease in circulating free fatty acids. There were small but significant increases in plasma calcium, magnesium, sodium, potassium, chloride and phosphate concentrations.  相似文献

Five cows in a herd of 15 cattle that had just been turned out onto lush pasture after having over-wintered on poor quality hay died suddenly. Biochemical profiles collected from the cadavers revealed reduced serum levels of magnesium, urea, and beta-hydroxybutycate. Classical grass tetany (hypomagnesemia) was diagnosed on postmortem examination.  相似文献
