The effects of different pretreatments on phytate and mineral contents were investigated in whole grain barley and oat tempe fermented with Rhizopus oligosporus. Different varieties of barley and oats were exposed to pretreatments such as pearling, rolling, moistening, autoclaving and soaking before fermentation. Pearling was the most effective pretreatment for reduction of phytate content for both oats and barley. Nevertheless, mineral contents were reduced, and most likely cell wall rich fractions were also reduced by this process. In the first experiments the phytate content reduction in the oats and barley samples were reduced by 74% (3.3 μmol/g, d.m.) and 89% (1.4 μmol/g, d.m.), respectively. However, to improve iron absorption the phytate levels should not exceed 0.5 μmol/g, and further phytate degradation was necessary. Therefore, in the final experiments barley samples were exposed to an optimised process with prolonged soaking at a higher temperature and the pearling residues were returned before fermentation. When the outer layers of the barley kernels were returned before fermentation the phytate content was successfully reduced by 97% to 0.4 μmol/g (d.m.) and Fe and Zn levels were well preserved.  相似文献

No difference in wheat phytase activity was observed when different types of acid were used to adjust the pH of wholemeal wheat flour suspensions to pH 5·0, the optimum for wheat phytase. When whole wheat bread was made without additives or after adjustment of the dough pH with acetic acid or lingonberry (traditional ingredients in bread making in Sweden), 64%, 96% and 83%, respectively, of the initial phytate was hydrolysed. A small but significant difference between breads with and without yeast or with deactivated yeast was found, indicating that yeast contributed some phytase activity under the conditions of bread making (pH 5·3–5·8 and 30–37°C). The optimum pH of yeast phytase was found to be 3·5. The isomers of IP5formed with purified wheat phytase or yeast phytase were studied using sodium phytate as substrate. Wheat phytase formed 1,2,3,4,5-IP5whereas yeast phytase formed 1,2,4,5,6-IP5. Determination of the isomers of inositol pentaphosphate demonstrated that the reduction in phytate levels in bread compared with wholemeal flour resulted from both wheat and yeast phytase activities.  相似文献

White and brown ragi (Eleusine coracana) varieties were analysed for tannin, phytate phosphorus, total phosphorus, iron, ionisable iron, zinc and soluble zinc content. White ragi had no detectable tannin while in brown varieties it ranged from 351 to 2392 mg per 100 g. Germination brought about a progressive decrease in tannin and phytate phosphorus and an increase in ionisable ion and soluble zinc content of grain ragi. Both in raw and germinated grain, ionizable iron was significantly higher in white than in brown varieties. While ionisable iron was inversely correlated with the level of tannin and phytate phosphorus, soluble zinc was negatively correlated with phytate phosphorus. After extraction of tannin, ionisable iron of brown ragi rose by 85%. On the other hand, in white varieties, addition of tannin extracted from brown ragi, resulted in a 52–65% decrease in ionisable iron content. These studies indicated that poor iron availability in ragi as judged by its low ionisable iron content was due to the presence of tannin in the grain.  相似文献

The present paper summarizes the current state of knowledge on molecular genetic approaches to increasing iron and zinc availability and vitamin content in cereals. We have also attempted to integrate the scientific issues into the wider context of human nutrition. In the cereal grain, iron and zinc are preferentially stored together with phytate in membrane-enclosed globoids in the protein storage vacuole (PSV) found in the aleurone and the embryo scutellum. The PSV is accordingly central for understanding mineral deposition during grain filling and mobilization of minerals during germination. Recent studies in Arabidopsis have led to the first identification of iron and zinc transporters of the PSV and further illustrate some of the dynamics associated with mineral and phytate transport and deposition into the vacuole. This provides new opportunities for modulating iron and zinc deposition in the cereal grain. Current strategies towards increasing the iron content of the endosperm are largely based on the expression of legume ferritin genes in an endosperm-specific manner. However, it is apparent that this approach, at least in rice, only allows a two- to three-fold increase in the iron content of the grain due to exhaustion of the iron stores in leaves. Further increases thus have to rely on additional uptake and transport of iron from the root. Phytate is generally considered to be the single most important anti-nutritional factor for iron and zinc availability. In the current paper we summarize attempts to increase phytase activity in the grain by transformation and evaluate the potential of this approach as well as the reduction of phytate biosynthesis for improving the bioavailability of iron and zinc. Vitamins constitute the second important group of micronutrients in grain and we discuss current efforts to increase the amounts of provitamin A, vitamin C and vitamin E.  相似文献

Pulque is made by fermenting the agave sap or aguamiel of Agave atrovirens with a whole array of microorganisms present in the environment including several lactic acid bacteria and yeasts such as Saccharomyces cerevisiae. Ascorbic acid was determined in pulque and aguamiel, respectively. Phytase activity in lees, liquid and freeze-dried pulque was assayed by measuring the appearance of phosphate from phytate by a colorimetric method likewise phosphate from phytate present in fresh corn tortilla was measured after in vitro incubation with pulque. Iron, zinc, calcium, magnesium and selenium contents were measured in pulque and corn tortilla as well as in nixtamalized corn flour (NCF), the latter is used to make instant tortilla, since corn provides most of the energy as well as most of the phytate in the Mexican rural diet. Pulque showed phytase activity but much less ascorbic acid and iron than previously reported; additionally, phytase in pulque hydrolyzed most of phytate’s corn tortilla. Lees, which is mostly made of pulque’s microbiota, significantly accumulated iron and zinc but no selenium. NCF was fortified with iron by the manufacturers but poorly blended. There were significant differences on selenium content between tortillas samples, apparently some soils in central Mexico are selenium deficient. Moderate pulque intake appears to increase the bioavailability of iron and zinc bound by phytate in corn.  相似文献

Mineral deficiencies are prevalent in human populations and the improvement of the mineral content in cereal products represents a possible strategy to increase the human mineral intake. Nevertheless, most of the inorganic phosphorus (Pi) present in mature cereal seeds (40–80%) is stored as phytate, an anti-nutritional factor that forms complexes with minerals such as Ca, Mg, Zn and Fe reducing their bioavailability. The present study was undertaken: (i) to determine the variation in phytate and mineral concentrations in the whole grains of 84 Italian durum wheat (Triticum durum Desf.) cultivars representative of old and modern germplasm; (ii) to estimate the magnitude of genotype × environment interaction effects; and (iii) to examine the interrelationships among mineral concentrations in durum wheat with the final aim to identify superior durum wheat cultivars that possess low phytate content and high concentration of mineral elements in their whole-wheat flour. The cultivars were grown in field trials during 2004–2005 at Foggia, Italy and during 2005–2006 at Foggia and Fiorenzuola d’Arda—Southern and Northern Italy. The phytate content was estimated indirectly by using a microtitre plate assay evaluating the Pi absorbance at 820 nm, while the Cu, Fe, Mn, Ca, K, Mg, Na and Zn mineral contents were determined by ICP/OES. The contents of Zn and Fe across years and locations ranged from 28.5 to 46.3 mg/kg for Zn with an average of 37.4 mg/kg and from 33.6 to 65.6 mg/kg for Fe with an average of 49.6 mg/kg. Pi grain content was between 0.46 and 0.76 mg/g showing a positive correlation with all minerals except Cu and Zn. Although breeding activity for Fe and Zn would be difficult because G × E interaction is prevalent, multi-location evaluation of germplasm collection help to identify superior genotypes to achieve this objective. The results here reported open the possibility of designing a specific breeding program for improving the nutritional value of durum wheat through the identification of parental lines with low-Pi and high minerals concentration in whole grains.  相似文献

Effects of bran concentration, bran particle size distribution, and enzyme addition – fungal phytase, fungal alpha-amylase – on the mixing and fermentative behaviour of wheat dough and on the amount of phytic acid remaining in bread have been investigated using a factorial design of samples 24. Bran concentration and bran particle size significantly affected all Farinograph parameters, whereas enzyme effects were particularly observed on both the water absorption of the flour and the parameters characterizing the overmixing. Water absorption was maximized in doughs with higher fine bran addition and/or in doughs with no enzymes, and was minimized in blends containing coarse added bran and alpha-amylase and/or alpha-amylase and phytase. alpha-Amylase addition had a significant positive effect on dough development and gassing power parameters during proofing. At low bran addition, phytate hydrolysis takes place to a greater extent than at high bran addition levels. Combination of bran with amylolytic and phytate-degrading enzymes could be advisable for overcoming the detrimental effect of bran on the mineral availability (phytase) or on the technological performance of doughs (alpha-amylase).  相似文献

Zinc and iron are important micronutrients for human health for which widespread deficiency occurs in many regions of the world including South Asia. Breeding efforts for enriching wheat grains with more zinc and iron are in progress in India, Pakistan and CIMMYT (International Maize and Wheat Improvement Centre). Further knowledge on genotype × environment interaction of these nutrients in the grain is expected to contribute to better understand the magnitude of this interaction and the potential identification of more stable genotypes for this trait. Elite lines from CIMMYT were evaluated in a multilocation trial in the eastern Gangetic plains (EGP) of India to determine genotype × environment (GE) interactions for agronomic and nutrient traits. Agronomic (yield and days to heading) data were available for 14 environments, while zinc and iron concentration of grains for 10 environments. Soil and meteorological data of each of the locations were also used. GE was significant for all the four traits. Locations showed contrasting response to grain iron and zinc. Compared to iron, zinc showed greater variation across locations. Maximum temperature was the major determinant for the four traits. Zinc content in 30–60 cm soil depth was also a significant determinant for grain zinc as well as iron concentration. The results suggest that the GE was substantial for grain iron and zinc and established varieties of eastern Gangetic plains India are not inferior to the CIMMYT germplasm tested. Hence, greater efforts taking care of GE interactions are needed to breed iron and zinc rich wheat lines.  相似文献

A pot-culture experiment was conducted to assess the bioavailability of iron from spinach cultivated in soil fortified with graded levels of iron and zinc (FeSO4 . 7H20 and ZnSO4 . 7H20, respectively). Applications of varying levels of iron to soil increased the total iron and phosphorus contents and decreased the zinc content (P<0.05). The effect of applying varying levels of zinc was the opposite of on the minerals in spinach. The ascorbic acid content was remarkably reduced with varying levels of iron and zinc. Higher levels of zinc and lower levels of iron in the soil increased the bioavailability of iron from spinach (P<0.05). In conclusion, the interactions of 15 ppm zinc with 30 ppm iron significantly enhanced the bioavailability of iron, total iron and zinc contents.  相似文献

Influence of irradiation (0.05–0.20 kGy) and germination (24–120 hours) in distilled and tap water on phytate, protein and amino acids of soybean, was studied. Phytate values significantly decreased with increasing germination period and irradiation dose (P<0.01). Irradiation independently decreased the original phytate (212.0 mg/100 g) to a range value of 205.0–190.0 mg/100 g depending upon dose level. Germination of unirradiated seeds for 120 hours in distilled and tap water lowered the phytate to 55.0 and 94.9 mg/100 g (74.1 and 55.2% reduction) respectively. Maximum destruction of phytate to levels of 20.5 and 50.9 mg/100 g (90.3 and 76.0% reduction) occurred during germination of 0.20 kGy samples for 120 hours in distilled and tap water respectively. Total protein content significantly increased during germination (P<0.05) and the increase was more in tap than distilled water. Germination for 120 hours of untreated seeds in tap water increased the essential and decreased non-essential amino acids while in the 0.10 kGy sample, increases in both cases were observed.  相似文献

Pearl millet is the most important staple food crop for millions of people across the world. Micronutrient malnutrition is the major problem for people living in the semi-arid regions of Africa. Identification of gene effects controlling the inheritance of grain Fe and Zn will be helpful in formulating suitable breeding strategies for biofortified pearl millet development. Hence, generation mean analysis was used to study epistasis and estimate gene effects for grain iron and zinc contents along with the agronomic and morphological traits. Six generations P1, P2, F1, F2, BC1P1 and BC1P2 were generated and were evaluated during the 2018–19 off season. Analysis of variance showed significant variability for all the traits in both generations. Six parameter model revealed predominance of additive gene effects for inheritance of grain iron concentration, and additive × additive type of non-allelic interactions. For grain zinc concentration additive gene effects were preponderant compared to non-additive gene effects, and only additive × dominance gene effects were significant among the three types of epistasis. Grain weight per plant was predominantly under non-additive gene effects and additive × additive and additive × dominance gene effects type of epistasis was detected in each cross. Likewise, for flowering non-additive gene effects were most important with the presence of dominance × dominance type of epistasis. For plant height, panicle circumference and length, additive × additive genes effects were the most important among the three type of non-allelic gene action.These findings can be helpful in enehancing the pearl millet breeding programs in Africa.  相似文献

The dehydrodiferulic acid content of different common and durum wheat grains and milling fractions was determined by an HPLC procedure. The 8-O-4′, 5–8′ benzofuran, 5–8′ and 5-5′ dehydrodimers were identified in all samples studied and occurred in decreasing relative amounts, respectively. Durum wheats were twice as concentrated in dimers as common wheats. An important genetic variation for dehydrodiferulic acid content was shown within durum wheat grains, whereas the agronomic conditions had no effect. There was 10 to 20 times more dehydrodiferulic acids in external layers (aleurone, bran) than in the starchy endosperm of durum wheat grains. The content and composition in dimers of the inner endosperm did not vary according to genotypes and cultivation conditions. The ratio of dehydrodimers to monomers of ferulic acid which represented an index of dimerisation, was 1·6 times higher in the external layers of the grain than in the endosperm. No relation was found, however, between the degree of ferulic acid dimerisation and the milling behaviour of durum wheat grains.  相似文献

Malnutrition related to micronutrient deficiency can create immense economic and societal problems. The objective of this study was to quantify the mineral element concentration distribution in milled fractions, using 43 common wheat (Triticum aestivum L.) cultivars sown in Jinan, China during the 2005–2006 crop season. All 43 cultivars had low Fe (average 28.2 mg Kg−1) and Zn (28.6 mg Kg−1) concentrations, and wide ranges of variation for mineral element concentrations. Highly significant effects among milling fractions and cultivars on all traits were observed, with fraction effect being the larger. There was an uneven distribution of mineral element concentrations in wheat grain. Shorts and bran fractions had the highest mineral element concentrations, whereas flours from break and reduction had low concentrations. Compared with those in the central endosperm, the concentration of inorganic phosphorus (Pi) decreased the most with decreasing flour yield, whereas the concentration of phytic acid P (PAP), phytase activity, and Ca decreased the least. Pi was the most concentrated element in the aleurone, whereas PAP, phytase activity, and Ca were the least, compared to those in the central endosperm. Milling technique through adjusting flour yield can be used to improve the element composition of flour.  相似文献

Decreased iron and zinc bioaccessibility, caused by the anti-nutrient phytic acid, is one of the leading reasons for micronutrient deficiency-related disorders. Biofortification of wheat with phytase gene to enhance iron and zinc bioaccessibility appears to be a fitting solution for this problem, especially in developing countries where most of the population belongs to the lower economic sector. However, societal views on crops, particularly crops that are genetically modified (GM) to express a new trait, needs to be changed. Risk assessment of GM crops can play a crucial role in fostering positive public perception, since it is imperative to ensure safety before allowing human consumption. The present study performed compositional and morphological risk assessment of T3 and T4 generations of phytase transgenic wheat by comparing their biochemical and morphological traits. Transgenic plants were analysed for their carbohydrate, protein, starch and phytic acid content along with iron bioaccessibility, zinc bioaccessibility and phytase enzyme activity. Morphological traits studied included plant height, seed number, seed weight and spike number. No significant differences were observed for carbohydrate, protein, starch content and for morphological traits; however, a significant increase was observed in phytase activity as well as iron and zinc bioaccessibility, which correlated with a significant decrease in phytic acid. These results demonstrate that phytase transgenic wheat is as native as local wheat varieties and can potentially increase iron and zinc bioaccessibility.  相似文献

Einkorn (Triticum monococcum L., subsp. monococcum), emmer (Triticum dicoccum Schuebl [Schrank], subsp. dicoccum) and spring bread wheat (Triticum aestivum L.) may be rich in lipophilic antioxidants (tocols and carotenoids), and therefore potential food sources with good nutritional properties. The aim of the present study was to determine the contents of major lipophilic antioxidants beneficial for human health in wheat varieties and landraces for breeding and production. In field experiments over two years, fifteen einkorn, emmer and spring wheat varieties were analysed for the contents of tocols and carotenoids in grain. A high carotenoid content (lutein, zeaxanthin, β-carotene) was typical for the selected einkorn genotypes. Among the analysed wheat species, the highest content was of β-tocotrienol, especially in the einkorn accessions. α-Tocotrienol was abundant in einkorn and emmer wheat species. Higher contents of α- and β-tocopherols were characteristic of spring and emmer wheats. δ-Tocotrienol has been detected for the first time in einkorn and some emmer accessions, although in low concentrations. Significant effects of genotype on the contents of carotenoids and tocols were found with einkorn differing from emmer and spring wheats. The year of cultivation had less effect on the contents of carotenoids and tocols. Selected accessions of einkorn with high contents of carotenoids and tocotrienols as well as spring and emmer wheats with higher contents of tocopherols are good sources of antioxidants with potential health promoting benefits for the production of nutritionally enhanced foods.  相似文献

The effects of processing (soaking, dehulling, fermentation and heat treatment) on the cooking time, protien, mineral, tannin, phytate and in vitro protein digestibility (IVPD) of the african yambean (AYB) were examined. The cooking time ranged from 90–155 minutes. Soaking reduced cooking time by about 50 percent. Soaking for 12 hours was the most appropriate to reduce cooking time, tannin and phytate levels. It improved in vitro protein digestibility (IVPD). Prolonged soaking (24 hours) decreased calcium (Ca) and iron (Fe) values by 19 percent and 35 percent, respectively. Dehulling showed that Ca, Fe, magnesium (Mg) and zinc (Zn) were concentrated in the seed coat of the AYB. The seeds soaked and dehulled retained Mg and Zn. Dehulling reduced tannin but had no significant effect on phytate and the IVPD of the AYB except for seeds soaked for 12 hours before dehulling. Soaking for 24 hours before dehulling significantly increased crude protein content by 16 percent (p<0.05). Blanching and roasting increased the IVPD by 8–11 percent. Fermentation had no effect on the crude protein, Ca, Fe, Mg and Zn but significantly reduced phytate content of the AYB. Fermentation had no advantage over heat treatment with respect to improving the in vitro protein digestibility of the AYB.  相似文献

Pearl millet was decorticated to obtain a bran rich and endosperm rich fraction. The two fractions were soaked in solutions with varying pH. Pearl millet grains were germinated and steamed followed by decortication to obtain two fractions. It was observed that bran rich fractions contained high concentrations of iron, zinc, polyphenols, phytic acid, fibre and flavonoids. Soaking for short duration of 3 h did not result in major mineral losses but decreased the inhibitory factors which depended on the pH. Alkaline soaking decreased flavonoid content by 62.7% in the endosperm rich fraction, while acidic soaking decreased phytic acid content to the maximum in the bran rich fraction. Combination of treatments like germination and heat decreased the phytate content to the maximum in the endosperm rich fraction. Acidic conditions improved zinc bioaccessibility in the bran rich fraction (35%) and iron bioaccessibility (2.5%) in the endosperm rich fraction. Bran rich fraction from germinated grain also had enhanced bioaccessibility of both the minerals but comparatively lesser when compared to soaking under acidic conditions. Soaking the grain components under slightly less than neutral conditions also decreased some of the inhibitory factors and improved the zinc bioaccessibility to some extent in the bran rich fraction.  相似文献

Cereal alkylresorcinols (AR), a group of phenolic lipids mainly found in the outer parts of wheat and rye kernels, are currently being studied for the possibility to use them as biomarkers for the intake of whole grain wheat and rye foods. In this work, AR were localised in grains by using light microscopy and gas chromatographic analysis of hand-dissected botanical and pearling fractions. GC-analysis of hand-dissected fractions showed that more than 99% of the total AR content was located in an intermediate layer of the caryopsis, including the hyaline layer, testa and inner pericarp. Microscopic examination showed that the outer cuticle of testa/inner cuticle of pericarp was the exact location, and that no AR were found in the endosperm or in the germ, suggesting that AR could be used as a selective marker of testa.  相似文献

Six green leafy vegetables and herbs – spinach, amaranth, bengal gram, cauliflower, mint, coriander and carrots – were analyzed for moisture, protein,ascorbic acid, -carotene, total iron, ionizable iron (as % of total iron) in vitro iron (% of total iron), copper, manganese and zinc. Moisture content of the leaves and carrots varied from 75.1 percent (bengal gram) to 95.4 percent (carrot) and protein from 9.83 percent (carrots) to 30.9 (mint) percent. Ascorbic acid, -carotene, total iron and ionizable iron contents were at a maximum in case of bengal gram leaves whereas level of ionizable iron and in vitro iron as a percent of total iron was highest in carrots. In order to investigate the intra-specific variation of wheat grain quality response to elevated atmospheric CO2 concentration (e[CO2]), eight wheat (Triticum aestivum L.)cultivars were grown at two CO2 concentrations ([CO2]) (current atmospheric, 389 CO2 μmol mol−1vs. e[CO2], FACE (Free-Air CO2 Enrichment), 550  ±  10% CO2 μmol mol−1), at two water levels (rain-fed vs. irrigated) and at two times of sowing (TOS1, vs. TOS2). The TOS treatment was mainly imposed to understand whether e[CO2] could modify the effects of timing of higher grain filling temperatures on grain quality. When plants were grown at TOS1, TKW (thousand kernel weight), grain test weight, hardness index, P, Ca, Na and phytate were not significantly changed under e[CO2]. On the other hand, e[CO2] increased TKW (16%), hardness index (9%), kernel diameter (6%), test weight (2%) but decreased grain protein (10%) and grain phytate (11%) at TOS2. In regard to grain Zn, Mn and Cu concentrations and some flour rheological properties, cultivar specific responses to e[CO2] were observed at both sowing times. Observed genetic variability in response to e[CO2] in terms of grain minerals and flour rheological properties could be easily incorporated into future wheat breeding programs to enable adaptation to climate change.  相似文献
