Cross sectional serological surveys were conducted between March and December 2009 to determine the distribution of foot-and-mouth disease and also to validate the current passive surveillance system in Bhutan. A total of 1909 sera collected from cattle, goats, sheep, and pigs, from 485 herds in 106 villages, were tested using a foot-and-mouth disease non-structural protein 3ABC ELISA. The true prevalence at the animal-level for all species was 15% (95% CI: 13.5, 16.7) using the sensitivity (97.2%) and specificity (99.5%) for cattle. The true prevalence for cattle, goats, sheep and pigs were 17.6 (95% CI: 15.6, 19.5), 11.9% (95% CI: 5.6, 18.3), 11.9% (95% CI: 1.3, 25.1), and 1.9% (95% CI: 0.0, 3.8), respectively. The sub-districts that shared border with India had significantly (p=0.03) higher seroprevalence than the interior sub-districts. Villages located in the sub-tropical zone had significantly (p<0.0001) higher seroprevalence than those located at high altitude zones. Herds with known outbreaks of FMD were 3.6 times more likely (p<0.001) to be seropositive than those with no history of outbreaks of FMD. The study showed the usefulness of population-based serological surveys in detecting circulation of active infection in populations which were, until now, considered to be free of disease based on a passive surveillance system. The study also highlighted the benefits of conducting serological and questionnaire surveys, simultaneously, to ascertain the infection status of herds and animals. Some of the findings from this study could be considered for strengthening of the current FMD control program in Bhutan.  相似文献

We collected blood samples from 756 > or =2-year-old cattle in 54 herds in Yucatan, Mexico, and used all of those to determine the antibody seroprevalences (in an indirect enzyme-linked inmunosorbance assay) to bovine respiratory syncytial virus (BRSV) and risk factors for animal-level seropositivity. We used 728 of the same samples (from 52 of the same herds) to do the same for parainfluenza virus-3 (PIV3). Cattle were selected by two-stage cluster sampling. Herd-level and animal-level risk factors were obtained through a personal interview. We analyzed the data by using a random-effects multivariable logistic regression model for clustered observations. All herds had at least 3 (BRSV) or 5 (PIV3) seropositive animals. The animal-level true seroprevalences were: 90.8% (86.5, 95.2%) and 85.6% (80.9, 90.4%) for BRSV and PIV3, respectively. Animals in large herds and old animals had the highest odds of being seropositives to BRSV, and those risk factors plus animals born on the farm for PIV3 infection.  相似文献

The results of a serological survey of livestock in Kazakhstan, carried out in 1997--1998, are reported. Serum samples from 958 animals (cattle, sheep and goats) were tested for antibodies to foot and mouth disease (FMD), bluetongue (BT), epizootic haemorrhagic disease (EHD), rinderpest (RP) and peste des petits ruminants (PPR) viruses, and to Brucella spp. We also investigated the vaccination status of livestock and related this to changes in veterinary provision since independence in 1991. For the 2 diseases under official surveillance (FMD and brucellosis) our results were similar to official data, although we found significantly higher brucellosis levels in 2 districts and widespread ignorance about FMD vaccination status. The seroprevalence for BT virus was 23%, and seropositive animals were widespread suggesting endemicity, despite the disease not having being previously reported. We found a few seropositives for EHDV and PPRV, which may suggest that these diseases are also present in Kazakhstan. An hierarchical model showed that seroprevalence to FMD and BT viruses were clustered at the farm/village level, rather than at a larger spatial scale. This was unexpected for FMD, which is subject to vaccination policies which vary at the raion (county) level.  相似文献

OBJECTIVE: To assess the impacts of the introduction of foot-and-mouth disease (FMD) and various FMD control programs in southern Thailand. ANIMALS: A native population of 562,910 cattle and 33,088 buffalo as well as 89,294 animals legally transported into southern Thailand. PROCEDURES: A quantitative risk assessment was used to ascertain the probability of FMD introduction, and an intrinsic dynamic model was used to assess impacts. Value for the transmission rate (beta) was estimated. Five scenarios created to assess the impacts of nonstructural protein (NSP) testing, mass vaccination, and culling were examined. Impacts were assessed through an examination of the estimated annual cumulative incidence (ACI) of FMD. The ACIs of various scenarios were compared by use of the Tukey Studentized range technique. RESULTS: beta was estimated at 0.115. Approximately 35,000 cases of FMD would be expected from the baseline situation. A 30% reduction of ACI was detected with the introduction of NSP antibody testing. Prophylactic vaccination resulted in an 85% reduction of ACI. Concurrent use of NSP antibody testing and vaccination reduced the ACI by 96%, and the addition of an eradication policy resulted in a slightly greater decrease in the ACI (98%). CONCLUSIONS AND CLINICAL RELEVANCE: The study used epidemiologic models to investigate FMD control interventions. Results suggested that vaccination has more impact than the use of NSP testing. Use of the NSP test reduced ACI during peak seasons, whereas vaccination diminished the underlying incidence. The best mitigation plan was an integrated and strategic use of multiple control techniques.  相似文献

A retrospective study on the outbreaks of foot-and-mouth disease (FMD) in Bhutan, between the years 1996 and 2008, based on the data collected through passive surveillance, was undertaken. A total of 230 outbreaks of FMD at sub-district level were recorded in 299 villages located in 19 out of the 20 districts in the country. There were no significant differences between the years (P = 0.998) or months (P = 0.989) on the incidence of FMD. The sub-districts in the north (altitude >1,000 m above mean sea level) had significantly (P = 0.008) higher incidences of outbreaks in winter than in summer. The sub-districts that shared border with India had significantly more outbreaks than those that didn't (P = 0.001). Cattle were the most predominant species affected being involved in all of the outbreaks reported. Serotype O, which constituted 70.6% of the outbreaks typed was the most predominant serotype prevalent in Bhutan followed by A (16.7%), Asia 1 (8.8%), and C (3.9%). Cattle density was significantly positively correlated (P = 0.023) with the incidence of disease. Three waves of outbreaks of epidemic proportions were reported in 1997/1998, 2002/2003, and 2007/2008 due to the PanAsia strain of the O serotype. The study highlights the incursion of the PanAsia strain of the O serotype into the country, possibly, through the transboundary movement of animals and the need for active surveillance of FMD, especially at the border areas. The study also highlights the significance of the O serotype and cattle as the main indicator species in the epidemiology of FMD in Bhutan. The findings from this study can be used as baseline epidemiological data for further research to understand the epidemiology of FMD in Bhutan.  相似文献

Serological investigation to detect foot-and-mouth disease (FMD) virus circulation in the domestic small ruminant population of India was conducted. A total of 4407 and 4035 serum samples from sheep and goats, respectively were collected at random covering majority of the states across the country during 2010–2012. These samples were analyzed for antibodies against the non-structural proteins (NSP) of FMD virus in an indirect 3AB NSP ELISA and against the structural proteins (SP) in a liquid phase blocking (LPB) ELISA. A total of 20.35% sheep and 13.60% goats were found to be positive for 3AB NSP antibodies providing a serological evidence of extensive viral activity. In LPB ELISA, only 4.54% sheep and 6.27% goats were found to have protective antibody (log10 titre of ≥1.8) against all three serotype strains in the vaccine, which correlates with “no or sparse vaccination” scenario in these species in the country. Hence, to check silent amplification and dissemination of virus in a mixed farming set up, small ruminants may be brought under the ambit of routine vaccination and surveillance programmes.  相似文献

Non-structural protein (NSP) 3ABC antibody is considered to be the most reliable indicator of present or past infection with foot-and-mouth disease virus (FMDV) in vaccinated animals. An indirect ELISA was established, using purified His-tagged 3ABC fusion protein as antigen, for detection of the antibody response to FMDV NSP 3ABC in different animal species. The method was validated by simultaneous detection of the early antibody responses to NSP and structural protein (SP) in FMDV Asia 1 infected animals. The performance of the method was also validated by detection of antibody in reference sera from the FMD World Reference Laboratory (WRL) in Pirbright, UK, and comparison with two commercial NSP ELISA kits. The results showed that the antibody response to SP developed more quickly than that to NSP 3ABC in FMDV infected animals. In contact-infected cattle, the antibody response to NSP 3ABC was significantly delayed compared with that to SP antibody. The early antibody responses to SP and NSP 3ABC in FMDV inoculated cattle and contact-infected or inoculated sheep and pigs were generally consistent. In pigs, 3ABC antibody was linked to the presence of clinical signs; however, in sheep, subclinical infection was detected by the development of 3ABC antibodies. Therefore, the antibody responses to 3ABC varied between host species. Eight out of 10 positive serum samples from FMD WRL were tested to be positive at cutoff value of 0.2. The rate of agreement with the ceditest FMDV-NS and the UBI NSP ELISA were 98.05% (302/308) and 93.2% (287/308), respectively. The prevalence of 3ABC antibodies reached 71.4% in some diseased cattle herds. The further work is required to evaluation the performance of this method in different animal species and different field situations.  相似文献

This paper reports on a retrospective study of the antibody responses to structural and non-structural proteins of FMD virus O Taiwan 97 in six pig herds in Taiwan in the year after the 1997 Taiwanese FMD outbreak. All herds were vaccinated against FMD after the outbreak as part of the countrywide control program. Three of the herds had confirmed FMD infections (herds N, O and P) and three herds remained non-infected (herds K, L and M). The serum neutralizing antibody titers and the non-structural protein ELISA (NSP) antibody responses in sows and 1-month-old pigs in the infected herds were higher than in the non-infected herds, but over time a number of positive NSP reactors were detected. From the serological studies and the herd monitoring and investigations it was considered that the FMD NSP positive reactors may not have constituted a true reservoir of FMD virus infection especially in herds where susceptible pigs were no longer present post-exposure or post-vaccination. Pigs vaccinated with an unpurified FMD type O vaccines being used at that time also showed false positive responses for NSP antibodies.  相似文献

Large scale surveillance of FMD non-structural protein (NSP) antibody in pigs was conducted to monitor for FMD virus circulation in Taiwan using Ceditest and UBI NSP ELISA kits after recurrence of FMD in 2009. A total of 53,759 serum samples were collected from pigs in the auction markets in 2009. There were 43 farms with positive FMD NSP reactors to both NSP ELISA tests in the nationwide surveillance. After tracing back, clinical examination and the NSP ELISA testing using both Ceditest and UBI on 14 follow-up serum samples from all the herds with confirmed NSP reactors in 2009, there were 4 farms classified as positive on follow-up testing criteria. In this surveillance, we have demonstrated that the NSP ELISA tests of outbreak farms followed by clinical and serological investigation could be used to detect FMD circulation in the pig population in Taiwan even while the national compulsory vaccination program is ongoing.  相似文献

In this study, we investigated whether Cedivac-FMD, an emergency vaccine against foot-and-mouth disease (FMD), is suitable for use conjointly with a screening program intended to confirm freedom from disease in vaccinated herds based on evidence of virus replication in vaccinates. Different sets of sera were tested using the Ceditest FMDV-NS ELISA for the detection of antibodies against non-structural proteins (NSPs) of FMD virus. During a vaccine safety study, serum samples were collected from 10 calves, 10 lambs and 10 piglets following administration of a double dose and a repeat dose of high payload trivalent Cedivac-FMD vaccine. All serum samples collected both 2 weeks following the administration of a double dose as well as those collected 2 weeks after the single dose booster (given 2 weeks after the double dose) were negative in the Ceditest FMDV-NS ELISA. In a series of vaccine potency experiments, serum samples were collected from 70 vaccinated cattle prior to and following exposure to infectious, homologous FMD virus. When testing cattle sera collected 4 weeks after vaccination with a regular dose of monovalent >6 PD(50) vaccines, 1 of 70 animals tested positive in the NSP antibody ELISA. After infection with FMD virus, antibodies to NSP were detected in 59 of 70 vaccinated cattle and 27 of 28 non-vaccinated control animals within 7 days. Cedivac-FMD vaccines do not induce NSP antibodies in cattle, pigs or sheep following administration of a double dose or a repeat dose. FMD-exposed animals can be detected in a vaccinated group within 7-14 days. Because Cedivac-FMD does not induce NSP antibodies, the principle of 'marker vaccine' applies.  相似文献

Forty peptides were synthesized corresponding to hydrophilic clusters of amino acids within the sequences of foot-and-mouth disease virus (FMDV) nonstructural proteins (NSP). Six peptides were studied in more detail and the most promising, a 2B peptide, was evaluated in enzyme-linked immunosorbent assay (ELISA) using sera from naive, vaccinated, and vaccinated-and-challenged cattle as well as bovine sera from field outbreaks. The performance of the new NSP peptide ELISA was compared to that of 4 commercial NSP ELISA kits. Antibody to 2B was detectable from the end of the first week to the second week after infection in most of the nonvaccinated animals and by the second to third week in vaccinated-and-challenged animals. The sensitivity of the 2B peptide ELISA was comparable to the 3ABC Ceditest (Ceditest FMDV-NS, Cedi Diagnostics B.V.; Chung et al., 2002). With some modification and further validation, this 2B test could be useful as a screening or conformational NSP test in postvaccination surveillance for FMD.  相似文献

Three commercialized ELISA kits for the detection of antibodies to the non-structural proteins (NSPs) of FMD virus were compared, using sera from uninfected, vaccinated, challenged and naturally infected pigs. The kinetics of the antibody response to NSPs was compared on sequential serum samples in swine from challenge studies and outbreaks. The results showed that ELISA A (UBI) and ELISA B (CEDI) had better sensitivity than that of the 3ABC recombinant protein-based ELISA C (Chekit). The peak for detection of antibodies to NSPs in ELISA C was significantly delayed in sera from natural infection and challenged swine as compared to the ELISA A and B. The sensitivity of the three ELISAs gradually declined during the 6-month post-infection as antibodies to NSP decline. ELISA kits A and B detected NSP antibody in 50% of challenged pigs by the 9-10th-day and 7-8th-day post-challenge, respectively. ELISA B and C had better specificity than ELISA A on sequential serum samples obtained from swine immunized with a type O FMD vaccine commercially available in Taiwan. Antibody to NSPs before vaccination was not detected in swine not exposed to FMD virus, however, antibody to NSPs was found in sera of some pigs after vaccination. All assays had significantly lower specificity when testing sera from repeatedly vaccinated sows and finishers in 1997 that were tested after the 1997 FMD outbreak. However, when testing sera from repeatedly vaccinated sows or finishers in 2003-2004, the specificity for ELISAs A, B and C were significantly better than those in 1997. This effect was less marked for ELISA A. The ELISA B was the best test in terms of the highest sensitivity and specificity and the lowest reactivity with residual NSP in vaccinates.  相似文献

The first cases of footrot in Bhutan were reported in sheep in 1990 at the National Sheep Breeding Centre (NSBC), which supplies breeding animals to village sheep flocks throughout Bhutan. Despite the presence of footrot at the Centre the distribution of apparently disease-free sheep continued. Cases of footrot were reported in village flocks soon after the disease was diagnosed at NSBC. A national survey was designed to establish the distribution and prevalence of footrot in Bhutan. This detected footrot in 19/94 village sheep flocks surveyed. The 19 affected flocks were distributed among nine different administrative districts whereas the villages selected were in 13 of a total of 16 sheep growing districts. The highest within-flock prevalences were among the seven flocks sampled in Bumthang district (mean 20.4%). The prevalence of the disease within flocks was generally much lower in other affected districts and in three districts a single affected animal was identified in the sample of 14 sheep examined in each village. Nationally, footrot prevalence was estimated to be 3.1% (95% CI 2.16-4.04%). There was a positive association between the receipt of animals from NSBC and the presence of footrot. The prevalence of the disease was higher in flocks with a migratory system of management than in those using a sedentary system. The relative risk of there being footrot in a migratory flock was nine-times higher than in a non-migratory flock. Only one strain of Dichelobacter nodosus (serogroup B) was identified among the 234 isolates obtained from the 19 affected flocks. Sheep with footrot healed quickly when treated with a vaccine made from this strain.  相似文献

Foot-and-mouth disease (FMD) is a highly contagious and economically devastating disease of cloven-hoofed animals in the world. The disease can be effectively controlled by vaccination of susceptible animals with the conventional inactivated vaccine. However, one major concern of the inactivated FMD virus (FMDV) vaccine is that it does not allow serological discrimination between infected and vaccinated animals, and therefore interferes with serologic surveillance and the epidemiology of disease. A marker vaccine has proven to be of great value in disease eradication and control programs. In this study, we constructed a marker FMDV containing a deletion of residues 93 to 143 in the nonstructural protein 3A using a recently developed FMDV infectious cDNA clone. The marker virus, r-HN/3A93–143, had similar growth kinetics as the wild type virus in culture cell and caused a symptomatic infection in pigs. Pigs immunized with chemically inactivated marker vaccine were fully protected from the wild type virus challenge, and the potency of this marker vaccine was 10 PD50 (50% pig protective dose) per dose, indicating it could be an efficacious vaccine against FMDV. In addition, we developed a blocking ELISA targeted to the deleted epitope that could clearly differentiate animals infected with the marker virus from those infected with the wild type virus. These results indicate that a marker FMDV vaccine can be potentially developed by deleting an immunodominant epitope in NSP 3A.  相似文献

Central Luzon is the number one pig-producing region in the Philippines and was affected by Foot-and-Mouth disease (FMD) in 1995. In this paper, the impact of FMD on the Central Luzon meat market from 1995 to 1999 was examined. Employing the error correction model (ECM) and historical decomposition, the impact of FMD on the Central Luzon pork and chicken meat market was quantified. The following findings were observed: (a) pig farm and pork wholesale prices dropped 11.8% and 15.7%, respectively, after the initial FMD outbreaks in January, 1995; (b) in February, 1995, chicken farm and wholesale prices declined by 21.1% and 14.2%, respectively (while chicken retail prices also went down by 10.5%); (c) the margins of pig and chicken traders were also adversely affected at some point; and (d) FMD caused changes of dynamic interdependence among prices by meat type at different levels of the meat supply chain.  相似文献

Vaccination with inactivated vaccine is an important measure to prevent and control foot-and-mouth disease (FMD), however, the immune effect and antigenic purity of inactivated vaccines are two major concerns for the establishment and evaluation of FMD free zones with vaccination. In this study, four groups of FMD type O and type A bivalent inactivated vaccines from 3 FMD vaccine manufacturers (designated as A, B and C) were selected to inoculate healthy juvenile cattle of FMD free. All cattle were immunized 3 or 4 times at a 1-month interval. Serum samples were collected before and after 1 month of every vaccination to determine the level of antibody to structural protein and non-structural protein. Results:(1) The qualified rates of antibody to structural protein: in group a1 (vaccine from company A, different batches), the antibody qualified rate could reach 100% for type O and type A, respectively after each vaccination. In group a2 (vaccines from company A, same batch), the antibody qualified rates were 36.7%, 98.3% and 100% for type O, and 15%, 86.7% and 100% for type A after the first to the third vaccination, respectively. In group b (vaccine from company B, same batch), the antibody qualified rates were 18.3%, 97% and 100% for type O, and 1.7%, 45% and 53.3% for type A after the first to the third vaccination, respectively. In group c (vaccines from company C, same batch), the antibody qualified rates were 26.7%, 96.7% and 100% for type O, and 21.7%, 71.7% and 100% for type A after the first to the third vaccination, respectively. (2) Antibody positive rate to non-structural protein 3ABC (confirmed with second ELISA test): In group a1, the positive rates were 0.7%, 1.4%, 9.5% and 4.8% after the first to the fourth vaccination, respectively; In group a2 and c, no 3ABC antibody-positive animal was detected after 3 repeated vaccination; In Group b, only one animal with a positive rate of 0.6% was detected after the third vaccination. The antibody qualified rates to the structural protein of FMDV in 3 of the 4 groups were far less than 70% after the primary vaccination, however, those were increased significantly after boost and repeated vaccination. The antigen purity of vaccines in three groups (a2, b and c) can meet the requirement of OIE standard on the FMD vaccine, however, the seroconversion to 3ABC antibody was obvious in animals from group a1 after repeated vaccination, which would cause some extent of interference to differential diagnosis. Also, a combination of a primary screening test and a confirmatory ELISA test can further improve the accuracy of differential diagnosis. This study provides an important scientific basis to make a rational program for establishment and evaluation of FMD free zone with vaccination.  相似文献

灭活疫苗免疫是防控口蹄疫的重要措施,但是灭活疫苗的免疫效果及其对感染与免疫鉴别诊断的干扰一直是口蹄疫免疫无疫区建设评估需要明确的重要问题。本研究中选择了3个企业(代号A、B与C)的4组口蹄疫O型与A型二价灭活疫苗,分别免疫口蹄疫抗体阴性健康未成年牛,免疫3~4次,测定免疫前后结构蛋白和非结构蛋白抗体的应答水平。结果显示:(1)结构蛋白抗体合格率:a1组(A企业多批次疫苗)4次免疫后O型和A型均为100%;a2组(A企业同批次疫苗)一~三免O型为36.7%、98.3%与100%,A型为15%、86.7%与100%;b组(B企业疫苗)一~三免O型为18.3%、97%与100%,A型为1.7%、45%与53.3%;c组(C企业疫苗)一~三免O型为26.7%、96.7%与100%,A型为21.7%、71.7%与100%。(2)非结构蛋白3ABC抗体阳性率(两种方法复核结果):a1组一~四免分别为0.7%、1.4%、9.5%与4.8%;a2组和c组三次免疫均未检测到阳性;b组仅三免后阳性率为0.6%。3组灭活疫苗首次免疫牛的抗体合格率远不及70%,但加强免疫后抗体合格率均显著提高;非结构蛋白抗体检测结果表明有3组疫苗的抗原纯净度符合OIE的要求,但a1组灭活疫苗免疫后,仍然对感染与免疫鉴别诊断存在干扰;采用两种非结构蛋白抗体检测方法进行复核检验,可以提高感染与免疫鉴别诊断的准确性。本研究为口蹄疫免疫无疫评价方案的制定提供了科学依据。  相似文献
