Grain yield-related traits and grain quality-related traits are important for rice cultivars. The quantitative trait loci (QTLs) involved in controlling the natural variation in these traits among closely related cultivars are still unclear. The present study describes the development of a novel chromosome segment substitution line (CSSL) population derived from a cross between the temperate japonica cultivars Yukihikari and Kirara397, which are grown in Hokkaido, the northernmost limit for rice cultivation. Days to heading, culm length, panicle length, panicle number, brown grain weight per plant, thousand brown grain weight, brown grain length, brown grain width, brown grain thickness, apparent amylose content, and protein content were evaluated. Panicle length, brown grain length and amylose content differed significantly in the parental cultivars. Thirty-five significant changes in the evaluated traits were identified in the CSSLs. A total of 28 QTLs were located on chromosomes 1, 2, 3, 4, 5, 6, 8, 9, 10, 11 and 12. These findings could be useful for breeding rice cultivars in the northernmost limit for rice cultivation.  相似文献

Panicle‐related traits are important agronomic traits which directly associated with grain yield. In this study, we investigated quantitative trait loci (QTLs) associated with panicle‐related traits using a set of 265 introgression lines (ILs) of common wild rice (Oryza rufipogon Griff.) in the background of Indica high‐yielding cultivar Guichao 2 (O. sativa L.). A total of 39 QTLs associated with panicle‐related traits including panicle length (PL), primary branch number (PBN), secondary branch number (SBN), spikelet number per panicle (SPP) and spikelet density (SD), were detected in the ILs with single‐point analysis. The alleles of 20 QTLs derived from wild rice showed positive effects, and some QTLs, such as, QPl1b for PL, QPbn8 for PBN, QSd4 and QSd11b for SD and QSpp4 for SPP showed larger positive effects, providing good candidates and useful information for marker‐aided improvement of yield potential of rice. Most of the QTLs controlling SPP, SBN and SD were located in cluster or closely linked on chromosomes, and the directions of their additive effects were consistent, which explained the genetic basis of significant correlations between their phenotypic characters.  相似文献

Hybrid breakdown (HB) is an important type of post-zygotic reproductive barrier that inhibits hybrid production during the process of cross-breeding. A novel low temperature-dependent HB was identified in a chromosomal segment substitution line (CSSL) library derived from a cross of two rice (Oryza sativa L. japonica) cultivars, Yukihikari and Kirara397. A set of weakness symptoms in a target CSSL was observed at 23°C, but was rescued at 27°C and/or 30°C. Genetic analysis of HB using an F2:3 population of a cross between a target CSSL and Kirara397 found that a recessive temperature sensitive hybrid breakdown1 (thb1) gene from Yukihikari caused HB in the genetic background of Kirara397. Molecular mapping showed that thb1 was located within a 199-kb fragment on chromosome 6. A genetic study of F2 populations of reciprocal crosses between Yukihikari and Kirara397 confirmed that this HB was induced by the interaction of two recessive genes. These results provide important clues to further dissect the mechanism of generation of a novel temperature sensitive HB in rice intrasubspecific crosses and suggest that these linked markers will useful in rice breeding.  相似文献

The barley (Hordeum vulgare L.) mutant erectoides‐k.32 (ert‐k.32) was isolated in 1947 from an X‐ray‐mutant population of cultivar ‘Bonus’. The mutant was released as a cultivar in 1958 with the name ‘Pallas’ – one of the first cereal crop cultivars developed from induced mutants. ‘Pallas’ is a semi‐dwarf barley cultivar known for its culm stability and resistance to lodging. In total, eight allelic ert‐k mutants are known that show different phenotypic strength concerning culm length and spike architecture. They represent alternatives to the widely used, but pleiotropic ‘Green Revolution’ alleles of the Sdw1 (semidwarf1/denso) and Uzu1 (semi‐brachytic1) genes in breeding of robust elite barley cultivars. In the present study, we locate Ert‐k to a 15.7‐cM region in the centromeric region of chromosome 6H. Although the interval is estimated to contain approximately 700 genes, the work provides a solid foundation for the identification of the underlying mutations causing the ert‐k lodging‐resistant phenotype. In addition, the linked markers could be used to follow the ert‐k mutant genotype in marker‐assisted selection of new lodging‐resistant barley cultivars.  相似文献

Spikelet sterility in rice (Oryza sativa L.) induced by high temperatures is a major concern given global warming predictions. We studied differences among eight rice cultivars in spikelet fertility at five different temperature levels in temperature gradient chamber (TGC) experiments. Six japonica and two indica cultivars were exposed to high‐temperature gradients in TGCs during the 2005 flowering season. Spikelet sterility increased with temperature in TGCs and differed among cultivars because of both variations in temperature tolerance and timing of heading. The correlation between spikelet fertility of individual panicles and both air temperature and panicle temperature during flowering was analyzed to compare tolerances among cultivars. The temperature (T75) at which spikelet fertility was 75 % of maximum ranged from 34 to 39 °C air temperature and differed significantly among cultivars. Indica varieties had higher T75 values than japonica varieties. The T75 values based on panicle temperature also differed among cultivars, but the difference between indica and japonica varieties were less significant. We concluded that the higher temperature tolerances of indica cultivars in our experiments could be attributed to lower spikelet temperatures, and cultivars with similar spikelet temperatures still had different heat tolerances due to differences in pollination ability.  相似文献

Heterosis is a phenomenon whereby hybrids of inbred lines produce favourable phenotypes that exceed those of their parents. Traits of interest are higher yield and stronger stress tolerance. The two‐line super‐hybrid rice ‘Liangyoupei9’ (LYP9) shows superiority to both its elite inbred line ‘93‐11’ and ‘Pei'ai64s’ (‘PA64s’) parents and conventional hybrids. However, the genetic basis of its hybrid vigour, especially yield determination, remains elusive. In the present study, a set of 156 chromosome segment substitution lines (CSSLs) carrying overlapping segments from ‘PA64s’ in a genetic background of ‘93‐11’ were constructed and planted in six environments. Three major agronomic traits, viz. panicle length (PL), heading date (HD) and plant height (PH), and five yield‐related traits, viz. grain weight per panicle (GWP), number of grains per panicle (GPP), 1000‐grain weight (TGW), seed set (SS) and number of panicles of per plant (PPP), were evaluated over 3 years. Quantitative trait loci (QTL) analysis was conducted using a likelihood ratio test based on stepwise regression. Forty‐six putative QTL distributed on 11 chromosomes were detected in more than one year. Remarkably, GWP of four CSSLs carrying positive yield QTL outperformed the recurrent parent ‘93‐11’ by more than 15%, in at least two environments. These results indicate that CSSLs are effective in identifying yield‐associated traits, and lines harbouring such QTL will be rich in resources for future molecular breeding programmes.  相似文献

The inheritance of agronomic traits from the barley dwarfing gene donors ‘Xiaoshan Lixiahuang’ and ‘Cangzhou Luodamai’ was studied. The results indicated that dwarf plants, six‐row and short spikes, dense spikelets and naked kernels, respectively, were controlled by one pair of recessive genes, but a toothed awn was determined by one pair of dominant genes in both barley cultivars. The genes for the six characters in ‘Xiaoshan Lixiahuang’ were allelic to those in ‘Cangzhou Luodamai’. Genetic linkage was found among the genes for plant height, spike length and spikelet density. They were located on the long arm of chromosome 3 (3HL) in the order: plant height, spikelet density, spike length. The genes for naked kernels, six‐row spikes and tooth awns were independent of each other, and carried on the long arms of chromosomes 1(7H), 2(H) and 7(5H), respectively. The dwarfing genes were the same as the gene uz in Japanese and Korean barley cultivars.  相似文献

Fifteen powdery mildew resistance genes and the gene MlaN81 derived from ‘Nepal 81’were found in 76 Czech and Slovak spring and winter barley cultivars when tested for reaction to a set of powdery mildew isolates. Nine cultivars (‘Donum’, ‘Expres’, ‘Jubilant’, ‘Orbit’, ‘Primus’, ‘Progres’, ‘Stabil’, ‘Vladan’ and ‘Zlatan’) are composed of lines with different resistance genes. The Mlat gene is present in nine cultivars and was transferred from the Anatolian landrace ‘A‐516′. The resistances derived from ‘KM‐1192’and ‘CI 7672’were identical and designated Ml(Kr). Five winter barley cultivars possess the Ml(Bw) resistance. The winter barley line ‘KM‐2099’carries the mlo gene. The parental cultivar ‘Palestine 10’was also tested in which the genes Mlk1, MlLa were identified. The German cultivar ‘Salome’, a parent of seven cultivars tested, probably carries the gene MlLa in addition to mlo and Mla7. The gene mlo6 may be present in the cultivar ‘Heris’. Most of the results were confirmed by the pedigrees of the cultivars.  相似文献

Four cold-tolerant rice varieties, viz. ‘Khonorullo’, ‘Namyi’, ‘Abor B’ and ‘Meghalaya-1’ were crossed with two cold-susceptible ones, viz. ‘Pusa 33’ and ‘Subhadra’ (DR92), in all possible combinations to study the inheritance of gene(s) governing panicle exsertion and their allelic relationship among cold-tolerant varieties. F1 hybrids of all the crosses showed complete panicle exsertion indicating dominance of this trait. Segregation pattern of panicle exsertion in F2 and backcrosses show that all the four cold-tolerant varieties possessed a single dominant gene designated as Ctr-1. Absence of segregation for panicle exsertion in an F2 generation obtained from intercrosses of cold-tolerant varieties suggests that the dominant genes in all the four cultivars are allelic.  相似文献

Grain numbers is one of the determinations for rice yield and directly associated with spikelet numbers per panicle and its normal development. Lots of genes responsible for spikelet numbers and spikelet early development have been identified, but the molecular information about the spikelet development at later development is still limited. Here, we isolated a rice spikelet abnormal development mutant, which shows degenerated spikelet at the top panicle and named aborted top spikelet mutant 1(Ats1). The spikelets derived from the middle and bottom branches per panicle of Ats1 show normal development with those of wild type. However, a large number of branches and spikelets with arrested development were often observed only on apex panicle. The abnormality did not appear until the stage In8 when rachises elongate rapidly and reproductive organs get mature, based on observations through SEM analysis. The aborted spikelet could develop the complete floral organs with a pair of rudimentary glume, a pair of empty glume, two lodicule, six stamens and one carpel. But all these floral organs did not develop maturity. Genetic analysis on two F2 populations indicated that the Ats1 was controlled by a single dominant gene. By using bulked segregant analysis of F2 population developed from Ats1 crossing with Songjing6, ATS1 was mapped on chromosome 8 between RM3819 and RM5556. Then, the fine mapping was performed with 1078 F2 population developed from Ats1 and IR36. The ATS1 locus was finely mapped in an 85.7 kb region between RM22448 and STS8‐2 with 8 genes according to the rice annotation project database. Sequence analysis of the candidate genes within the delimited region of the Ats1 and Akihikari showed two‐nucleotide changes, including single‐nucleotide substitutions corresponding to an amino acid substitution from asparagine to lysine acid in exons 3 and a 1‐bp deletion resulting in a premature stop codon in exon 22 at the candidate gene, LOC_Os08g06480. A cleaved amplified polymorphic sequence (CAPS) marker, CAPS‐ats1, was developed from the 1‐bp deletion site. The complete cosegregation of the CAPS genotypes with the matching phenotypes were observed in the F2 populations. This suggested that Os08g06480 is most likely the ATS1 gene. These results will provide more information for better understanding of the molecular mechanism governing top spikelet abortion within a short developmental period.  相似文献

Stem rust of wheat (caused by Puccinia graminis f.sp. tritici) gained high international attention in the last two decades, but does not occur regularly in Germany. Motivated by a regional epidemic in 2013, we analysed 15 spring and 82 winter wheat cultivars registered in Germany for their resistance to stem rust at the seedling stage and tested 79 of these winter wheat cultivars at the adult‐plant stage. A total of five seedling stem rust resistance genes were postulated: Sr38 occurred most frequently (n = 29), followed by Sr31 (n = 11) and Sr24 (n = 8). Sr7a and Sr8a occurred only in two spring wheat genotypes each. Four cultivars had effective seedling resistance to all races evaluated that could only be explained by postulating additional resistance genes (‘Hyland’, ‘Pilgrim PZO’, ‘Tybalt’) or unidentified gene(s) (‘Memory’). The three winter wheat cultivars (‘Hyland’ ‘Memory’ and ‘Pilgrim PZO’) were also highly resistant at the adult‐plant stage; ‘Tybalt’ was not tested. Resistance genes Sr24 and Sr31 highly protected winter wheat cultivars from stem rust at the adult‐plant stage in the field. Disease responses of cultivars carrying Sr38 varied. Mean field stem rust severity of cultivars without postulated seedling resistance genes ranged from 2.71% to 41.51%, nine of which were significantly less diseased than the most susceptible cultivar. This suggests adult‐plant resistance to stem rust may be present in German wheat cultivars.  相似文献

Tan spot of wheat is caused by the fungus Pyrenophora tritici‐repentis. On susceptible hosts, P. tritici‐repentis induces two phenotypically distinct symptoms, tan necrosis and chlorosis. This fungus produces several toxins that induce tan necrosis and chlorosis symptoms in susceptible cultivars. The objectives of this study were to determine the inheritance of insensitivity to necrosis‐inducing culture filtrate of P. tritici‐repentis, race 2, and to establish the relationship between the host reaction to culture filtrate and spore inoculation with respect to the necrosis component. The F1, F2, and BC1F1 plants and F2:8 lines of five crosses involving resistant wheat genotypes ‘Erik’, ‘Red Chief’, and line 86ISMN 2137 with susceptible cultivars ‘Glenlea’ and ‘Kenyon’ were studied. Plants were spore‐inoculated at the two‐leaf stage. Four days later, the newly emerged uninoculated third leaf was infiltrated with a culture filtrate of isolate Ptr 92–164 (race 2). Reactions to the spore inoculation and the culture filtrate were recorded 8 days after spore inoculation. The segregation observed in the F2 and BC1F1 generations and the F2:8 lines of all crosses indicated that a single recessive gene controlled insensitivity to necrosis caused by culture filtrate. This gene also controlled resistance to necrosis induced by spore inoculation.  相似文献

Yield is a complex trait. To improve it, the accumulation of the favourable alleles of valuable genes is required for each yield‐related trait. In this study, we used two high‐yielding rice cultivars developed in Japan, indica‐type ‘Takanari’ and japonica‐type ‘Momiroman’, for a genetic analysis of the sink capacity‐related traits. An F2 population showed transgressive segregation for the number of spikelets per panicle. Quantitative trait locus (QTL) analysis detected four QTLs for the trait. Two of the QTLs were most likely identical to previously cloned GN1a and APO1, and their Takanari alleles had positive effects. The Momiroman alleles of the other two QTLs had positive effects, and one of these QTLs was most likely identical to SPIKE/GPS. The QTL on the long arm of chromosome 3 appeared to be novel; it clustered with QTLs for grain length and days‐to‐heading. Substitution mapping revealed that the close linkage of QTLs caused the clustering. These results suggest that the combination of the favourable alleles of detected QTLs could lead to greater sink capacity than that of the parental cultivars.  相似文献

Limited genetic knowledge is available regarding crossability between hexaploid triticale (2n= 6x= 42, 21″, AABBRR, amphiploid Triticum turgidum L.‐Secale cereale L.) and rye (2n= 14, 7″, RR). Our objectives were to determine (1) the crossability between triticales and rye and (2) the inheritance of crossability between F2 progeny from intertriticale crosses and rye. First, ‘8F/Corgo’, a hexaploid triticale, was crossed as a female with two landrace ryes, ‘Gimonde’ and, ‘Vila Pouca’ and two derived north European cultivars, ‘Pluto’ and ‘Breno’. These crosses produced 21.7, 20.9, 5.9, and 5.6%, seed‐set or crossability, respectively, showing that the landrace ryes produced higher seed‐set than the cultivars. Second, ‘Gimonde’ rye was crossed as a male with four triticales for 3 years. The control cross, ‘Chinese Spring’ wheat × rye, produced 80‐90% seed‐set. Of the four triticales, ‘Beagle’ produced 35.7‐56.8% seed‐set. The other three triticales produced less than 20% seed‐set, showing that the triticales differ in crossability with ‘Gimonde’ rye. Third, six FiS from intertriticale crosses (‘8F/Corgo’בBeagle’, ‘Beagle’בCachirulo’, ‘Lasko’בBeagle’, ‘8F/Corgo’בCachirulo’, ‘Lasko’בCachirulo’, ‘Lasko’ב8F/Corgo’) were crossed to ‘Gimonde’ rye. Results indicated that lower crossability trait was partially dominant in the two F1S from crosses involving ‘Beagle’(high crossability) with‘8F/Corgo’ and ‘Cachirulo’(low crossability) and completely dominant in the ‘Beagle’בLasko’ cross, as it happens in wheat. Fourth, segregants in four F2 populations (‘Lasko’בBeagle’, ‘8F/Corgo’בBeagle’, ‘Lasko’ב8F/Corgo’, and‘8F/Corgo’בCachirulo’) were crossed with rye. Segregation for crossability was observed, although distinct segregation classes were blurred by environmental and perhaps other factors, such as self‐incompatibility alleles in rye. Segregation patterns showed that ‘Beagle’, with high crossability to rye, carries either Kr1 or Kr2. The three triticales with low crossability with rye were most likely homozygous for Kr1 and Kr2. Therefore, it is likely that the Kr loci from A and B genomes acting in wheat also play a role in triticale × rye crosses.  相似文献

A sequence‐tagged site marker has been developed from restriction fragment length polymorphism marker probe IAG95 for the rye‐derived powdery mildew resistance Pm8/Pm17 locus of common wheat. This polymerase chain reaction marker enables the amplification of DNA fragments with different sizes from T1AL.1RS and T1BL.1RS wheat‐rye translocation cultivars with chromatin from ‘Insave’ and ‘Petkus’ rye, respectively, and therefore will be very useful in distinguishing Pm8‐carrying cultivars from Pm17‐carrying cultivars. Results obtained with that marker were compared with resistance tests performed on detached primary leaves of 29 wheat lines from two populations derived from doubled haploid production. The molecular assay corresponded well with the resistance tests in all the lines, and therefore will be helpful for the identification of Pm17 in lines in which other Pm genes or quantitative trait loci are present.  相似文献

Ideal plant architecture has become a significant objective in high‐yield rice breeding. In this study, a new semidwarf gene in sdk, a spontaneous dwarf mutant of ‘Kasalath’, was characterized and genetically mapped. The mutant has smaller panicles and seeds, reduced awns, and is 6–8 days earlier in heading than ‘Kasalath’. There was no significant difference in number (No.) of seeds and fertile seeds per panicle. Genetic analysis indicated that the dwarf character of sdk is controlled by a recessive gene, semidwarf k (sdk). From a segregating population of about 65 000 individuals, 4987 sdk‐type individuals were sampled. Gene sdk was located in a genomic region of 2622 kb flanked by markers R‐12 and R‐43 in the centromeric region of chromosome 2. The results provide an opportunity for gaining an understanding of the molecular mechanisms underlying sdk and opening possibilities for its use in rice breeding programmes.  相似文献

A total of 50 wheat (Triticum aestivum L.) cultivars were evaluated for resistance to tan spot, using Pyrenophora tritici‐repentis race 1 and race 5 isolates. The cultivars ‘Salamouni’, ‘Red Chief’, ‘Dashen’, ‘Empire’ and ‘Armada’ were resistant to isolate ASC1a (race 1), whereas 76% of the cultivars were susceptible. Chi‐squared analysis of the F2 segregation data of hybrids between 20 monosomic lines of the wheat cultivar ‘Chinese Spring’ and the resistant cultivar ‘Salamouni’ revealed that tan spot resistance in ‘Salamouni’ was controlled by a single recessive gene located on chromosome 3A. This gene is designated tsn4. The resistant cultivars identified in this study are recommended for use in breeding programmes to improve tan spot resistance in common wheat.  相似文献

Recessively inherited gene Sr2 has provided the basis of durable resistance to stem rust (caused by Puccinia graminis tritici) in wheat (Triticum aestivum L.) worldwide. The associated earhead and stem melanism or ‘pseudo‐black chaff’ is generally used as a marker for this gene. Sr2 has been postulated in many wheat cultivars of India including ‘Lok 1’, based on associated pseudo‐black chaff in adult plants, and leaf chlorosis in seedlings. However, dominant inheritance of the resistance factor operating in ‘Lok 1’, and a 13 : 3 (resistant : susceptible) F2 segregation in the ‘Sr2‐line’ (‘Chinese Spring’6 × ‘Hope’ 3B) × ‘Lok 1’ cross confirmed that Sr2 was absent in ‘Lok 1’. Susceptible plants with a pseudo‐black chaff phenotype were observed in F2 populations of ‘Agra Local’ (susceptible) × ‘Lok 1’, and the ‘Sr2‐line’ × ‘Lok 1’ crosses. Most of the F3 families derived from the susceptible F2 segregants with pseudo‐black chaff phenotypes were true breeding for the expression of pseudo‐black chaff with susceptibility to stem rust. Thus, linkage of pseudo‐black chaff with Sr2 in wheat can be broken, and hence, caution may be exercised in using pseudo‐black chaff as a marker for selecting Sr2 in breeding programmes.  相似文献

Presence of substantial heterosis and economic hybrid seed production are two most desirable components for success of any commercial hybrid breeding programme. Thermosensitive genic male sterile (TGMS) lines of rice, in this regard, have tremendous potential in realizing further quantum jump in yield and economical hybrid seed cost. Analyses for combining ability and heterosis over optimum (120N : 60P2O5 : 40K2O kg/ha) and high (200N : 90P2O5 : 60K2O kg/ha) fertility environments for six traits were made in 2 years (2001 and 2002) using 120 hybrids of inter‐ and intra‐subspecific nature derived from hybridization of 30 elite indica TGMS lines and four cultivars, viz., ‘Pant Dhan 4’ and ‘Ajaya’ (I = indica), ‘Taichung 65’ (J = japonica) and ‘IR 65598‐112‐2’ (TJ = tropical japonica) in line × tester mating design. Predominance of non‐additive genetic variance suggested good prospects of hybrid breeding. Pooled analysis revealed highly significant variances for lines, general combining ability (GCA), specific combining ability (SCA) and line x tester. TGMS line 365‐8S was the best general combiner for all the six traits including grain yield. Trend of relative mid‐parent heterosis for grain yield, panicle length, grain number per panicle and earliness in flowering was I/TJ > I/J > I/I. For panicle number per plant and 1000‐grain weight, trends were I/TJ > I/I > I/J and I/I > I/TJ > I/J, respectively. Grain yield recorded heterosis of 49.3%, 71.9% and 92.7% for I/I, I/J and I/TJ hybrid groups respectively. Effect of environments on the hybrid performance indicated better response of hybrids at high fertilizer dose. Study suggests greater prospects of combining improved japonica and tropical japonica germplasms having wide compatible gene with indica TGMS lines for exploitation of intersubspecific heterosis.  相似文献

The inheritance of freezing tolerance in F2‐derived F4 populations from all possible crosses of winter wheat cultivars ‘Kestrel’, ‘Eltan’, ‘Tiber’, ‘Froid’ and germplasm line Oregon Feed Wheat #5 (ORFW) was investigated. When frozen to a temperature equal to the LT50 of the least freezing tolerant parent (ORFW), survival frequency distributions were skewed to greater survival in six of the 10 crosses, however, very few of the progeny from the four crosses to ORFW survived. The inheritance of this freezing sensitivity was investigated with freezing of F2:4 populations from the crosses of ORFW to ‘Eltan’ or ‘Tiber’ to the LT50 of the hardier parent. Very few of the F2:4 populations survived as well as ‘Eltan’ or ‘Tiber’, indicating a small number of strongly dominant genetic factors in ORFW that conditioned freezing sensitivity. Molecular analysis indicated these factors were not spring‐type vernalization alleles.  相似文献
