Two experiments (Exps. 1 and 2) were performed to study the influence of Trichuris suis infection and type of dietary carbohydrates on large intestine morphology, epithelial cell proliferation and mucin characteristics. Two experimental diets based on barley flour were used; Diet 1 was supplemented with resistant carbohydrates from oat hull meal, while Diet 2 was supplemented with fermentable carbohydrates from sugar beet fibre and inulin. In Experiment 1, 32 pigs were allocated randomly into four groups. Two groups were fed Diet 1 and two groups Diet 2. Pigs from one of each diet group were inoculated with a single dose of 2000 infective T. suis eggs and the other two groups remained uninfected controls. In Experiment 2, 12 pigs were allocated randomly into two groups and fed Diet 1 or Diet 2, respectively, and inoculated with a single dose of 2000 infective T. suis eggs. All the pigs were slaughtered 8 weeks post inoculation (p.i.). The worm counts were lower in pigs fed Diet 2 in both experiments, but not significantly so. Both diet and infection status significantly influenced the tissue weight of the large intestine. In both experiments, pigs fed Diet 2 had heavier large intestines than pigs fed Diet 1 and in Experiment1 the infected pigs of both diets had heavier large intestines than their respective control groups. Diet and infection also significantly affected the morphological architecture and mucin production in both experiments. Pigs fed Diet 1 had larger crypts both in terms of area and height than pigs fed Diet 2 and T. suis infected pigs on both diets in Experiment 1 had larger crypts than their respective control groups. The area of the mucin granules in the crypts constituted 22-53% of the total crypt area and was greatest in the T. suis infected pigs fed Diet 1. Epithelial cell proliferation was affected neither by diet nor infection in any of the experiments. The study showed that both T. suis infection and dietary carbohydrates significantly influence the morphological architecture and the production and composition of mucins in the large intestine of pigs and suggests that both factors are important in large intestine function and that carbohydrates may play a role in the susceptibility to intestinal helminth infections.  相似文献

This study tried to evaluate the effect of highly fermentable fiber on the incidence and severity of swine dysentery (SD) after experimental oral infection with pure cultures of Brachyspira (B.) hyodysenteriae. Forty eight growing pigs were allocated to two groups and treated until slaughter as follows: Group 1 (n = 24): infected with B. hyodysenteriae and fed with a food containing 9.6% highly fermentable neutral detergent fiber. Group 2 (n = 24): infected with B. hyodysenteriae and fed with a food containing 6.1% low fermentable neutral detergent fiber. Pigs of each group were intragastrically inoculated on each of three consecutive days with pure culture of 1.8 x 10(10) B. hyodysenteriae. All pigs were monitored daily until slaughter. Faecal shedding of B. hyodysenteriae by polymerase chain reaction, antibody response by IFA, clinical signs, growth performance and extents of gross and microscopical lesions specific for swine dysentery were determined. Faecal shedding of B. hyodysenteriae and antibodies specific for B. hyodysenteriae were detected at day 30 post infectionem. Significant (p < 0.05) milder clinical signs typical for swine dysentery were detected in group 1, fed with 9.6% high fermentable fiber compared to group two fed with a food containing 6.1% low fermentable neutral detergent fiber. Daily weight gain differed significantly (p < 0.05) between the groups (group one 780 g vs. group two 760 g). Food conversion efficiency showed in group one a significant (p < 0.05) better (3.28) result than in group two (3.38). Feed consumption presented significantly (p < 0.001) better results in group one compared to group two (2.38 kg vs. 2.25 kg). From our experimental findings we conclude that in production units suffering of swine dysentery high levels of highly fermentable fiber in diet may increase health and performance.  相似文献

Transmission experiments were carried out in gnotobiotic pigs to determine whether lesions typical of swine dysentery could be produced by oral inoculation of Treponema hyodysenteriae in combination with Bacteroides vulgatus or Fusobacterium necrophorum, or both. Each of the organisms had been isolated from swine with early lesions of the disease. Lesions were not found in 6 pigs inoculated with T hyodysenteriae alone, in 4 pigs given F necrophorum and T hyodysenteriae, or in 4 pigs given B vulgatus and F necrophorum. Lesions typical of swine dysentery developed in 8 pigs given B vulgatus, F necrophorum, and T hyodysenteriae as well as in 3 of 4 pigs given B vulgatus and T hyodysenteriae. In both of these groups, the inoculated bacteria were recovered from the colon, and T hyodysenteriae was demonstrated in the colonic crypts, epithelium, and lamina propria. The pathogenicity of the T hyodysenteriae was shown by the development of characteristic signs and lesions of swine dysentery in 12 of 14 naturally farrowed pigs inoculated with T hyodysenteriae alone.  相似文献

Twenty-six specific-pathogen-free pigs were fed pure cultures of Treponema hyodysenteriae. Five untreated pigs were controls. Distribution of this large spirochete in pigs with swine dysentery was shown by the indirect fluorescent antibody technique. Findings by this method were compared with those from dark-field examination of colonic mucosal scrapings and from tissue sections. The cultures caused mucohemorrhagic colitis which by 10 days after inoculation was indistinguishable from the colitis of swine dysentery. Control pigs remained normal. Pigs killed when spirochetes were first seen in the feces had normal colonic mucosa with only a few spirochetes. At the first sign of diarrhea, however, the colonic mucosa was thicker than normal and had many spirochetes. T. hyodysenteriae was confined to regions of hypertrophy and exudation of the large intestine mucosa throughout the course of disease.  相似文献

The immune status of 29 pigs recovered from swine dysentery (SD) was evaluated after reexposure to Treponema hyodysenteriae. Pigs which had recovered from SD and remained asymptomatic for 4 to 6, 9 to 13, and 16 to 17 weeks after initial inoculation were reexposed to 1.5 X 10(9) viable cells of T hyodysenteriae per pig. Pigs which had recovered from SD were not shedding T hyodysenteriae, as determined by selective cultural examination of feces, before they were reexposed. Of 29 pigs reexposed to T hyodysenteriae, 27 were resistant to SD. In contrast, 23 of 28 control pigs developed signs of SD when exposed to T hyodysenteriae for the first time. Significant differences in immunity were not observed between pigs from the three convalescent periods. Serum agglutinins to T hyodysenteriae were present in recovered pigs for approximately 8 weeks after inoculation.  相似文献

Twenty-four conventionally reared pigs divided into 4 groups were fed a basic ration deficient in selenium. The following daily supplement of selenium was given per pig; Group 1: none, group 2: 0.2 mg, group 3: 0.4 mg and group 4: 0.8 mg. After 51 days all pigs were inoculated orally with a pure culture of Treponema hyodysenteriae, and subsequently observed for 26 days. Clinical signs of swine dysentery were seen in all 4 groups. Criteria such as morbidity rate, incubation time and weight gain showed that the non-supplemented pigs suffered more severely from swine dysentery than the supplemented ones. Best protection was found among the pigs given a daily supplement of 0.4 mg selenium, whereas a supplement of 0.8 mg had a negative influence on the resistance to swine dysentery. The results indicate that selenium plays a more complex role in mucosal defence mechanisms than hitherto anticipated.  相似文献

Six intravenous injections of formalin-inactivated Treponema hyodysenteriae were given to 8 specific-pathogen-free pigs at 6-day intervals. The 8 vaccinated and 8 control pigs were challenged intragastrically with pure cultures of T hyodysenteriae 7 and 8 days after the last intravenous injection. Clinical signs of swine dysentery were observed in all 8 control pigs, but was observed in only 1 of the immunized pigs. Three control pigs died. These findings suggest that parenteral immunization with T hyodysenteriae provided a marked degree of protection against subsequent intragastric challenge exposure with the homologous isolate of T hyodysenteriae.  相似文献

An attenuated strain of Treponema hyodysenteriae was used to immunise 18 pigs in three experiments. Live attenuated spirochaetes were dosed orally and injected intra-peritoneally, and killed spirochaetes were injected intramuscularly with adjuvant. The vaccinated pigs, which developed high serum agglutination titres against T hyodysenteriae, and 18 unvaccinated litter-mates were repeatedly challenged with virulent T hyodysenteriae. Nine vaccinated pigs and 16 control pigs developed typical swine dysentery.  相似文献

Two experiments were conducted to determine the apparent ileal digestibility of DM and N by young pigs fed diets supplemented with different protein sources or organic acids. Pigs were surgically fitted with silicone cannulas at 2 wk of age. Following surgery, pigs were allowed to recuperate with their dams while suckling normally. After weaning at 24 d, pigs were assigned to treatment diets at 28 d of age. A 3-d adjustment and 4-d collection sequence was followed for the duration of the 4-wk experiment. Four treatment diets were fed in each experiment in a weekly rotation until each diet had been fed to each pig. Diet samples and digesta collected through the ileal cannulas were analyzed for chromic oxide (used as an indigestible marker), DM, and N. Pigs in Exp. 1 were fed isolysinic (1.0%) corn-based diets supplemented with casein, soybean meal, soy protein concentrate, or isolated soy protein. Casein addition resulted in improved DM (P less than .001) and N (P less than .05) digestibility but reduced gain (P less than .05) compared with the average of the soy protein sources. Nitrogen from diets formulated with soybean meal was digested more completely (P less than .05) than N from diets based on soy protein concentrate and isolated soy protein. Experiment 2 was an evaluation of the effect of dried skim milk (25%) and fumaric acid (2%) addition on apparent ileal digestibility of N and DM in corn-soybean meal diets. Addition of dried skim milk improved DM (P less than .01) and N (P less than .05) digestibility and daily gain (P less than .001). Fumaric acid supplementation did not affect nutrient digestibility or gain (P greater than .10).  相似文献

The purpose of this study was to determine whether methods used to control swine dysentery (SD), caused by the intestinal spirochaete Brachyspira (Serpulina) hyodysenteriae, would also be effective in controlling porcine intestinal spirochaetosis (PIS) caused by the related spirochaete Brachyspira (Serpulina) pilosicoli. Weaner pigs in Groups I (n=8) and II (n=6) received a standard weaner pig diet based on wheat and lupins, whilst Group III (n=6) received an experimental diet based on cooked white rice and animal protein. Pigs in Group II were vaccinated intramuscularly twice at a 3-week-interval with a formalinised bacterin made from B. pilosicoli porcine strain 95/1000 resuspended in Freund's incomplete adjuvant. Eleven days later pigs in all groups were infected orally with 10(10) cells of strain 95/1000 on three successive days. One control pig in Group I developed acute diarrhoea, and at post-mortem had a severe erosive colitis with end-on attachment of spirochaetes to the colonic epithelium. All other pigs developed transient mild diarrhoea and had moderate patchy colitis at post-mortem 3 weeks later. B. pilosicoli was isolated from the faeces of all pigs, except for one fed rice, and was isolated from the mesenteric nodes of three pigs from Group I and from one vaccinated pig in Group II. Consumption of the rice-based diet, but not vaccination, delayed and significantly (p<0.001) reduced the onset of faecal excretion of B. pilosicoli after experimental challenge. Vaccination induced a primary and secondary serological response to B. pilosicoli, as measured using sonicated whole cells of strain 95/1000 as an ELISA plate coating antigen. Antibody titres in the vaccinated pigs then declined, despite intestinal colonisation by B. pilosicoli. Both groups of unvaccinated animals also failed to develop a post-infection increase in circulating antibody titres.  相似文献

Pure cultures of Treponema hyodysenteriae given orally to conventional pigs resulted in the development of swine dysentery, whereas identical cultures given to gnotobiotic pigs did not produce the disease. Oral inoculation of gnotobiotic pigs with Vibrio coli and/or a peptostreptococcus in addition to T. hyodysenteriae did not result in dysentery. Neutralization of gastric secretions with NaHCO3 immediately prior to inoculation with T. hyodysenteriae increased the period during which treponemes were evident in the feces, as did the inoculation of this organism via the intracecal route. None of the gnotobiotic pigs with a persistent fecal Treponema population developed signs of dysentery. Factors other than those investigated in this work must play a part in the etiology of swine dysentery.  相似文献

Isolation of Treponema hyodysenteriae from sources other than swine   总被引:3,自引:0,他引:3  
Fecal samples were collected from animals and environments on 3 swine farms and cultured for Treponema hyodysenteriae. Each farm was a farrow-to-finish operation and, at the time of sampling, swine dysentery was enzootic among 8- to 22-week-old pigs. Pathogenic T hyodysenteriae was isolated from pigs on all 3 farms. On farm A, nonpathogenic T hyodysenteriae was isolated from a sample of lagoon water. On farm B, pathogenic T hyodysenteriae was isolated from a waste-holding pit. On farm C, a dog was observed to be eating feces of pigs that had swine dysentery. The dog was diarrheic and a fecal sample yielded a pathogenic isolant of T hyodysenteriae. Further isolation attempts were unsuccessful after the dog was removed from the infected premises. Isolation of pathogenic and nonpathogenic organisms from waste-holding systems emphasizes the need for cultural techniques in detecting pathogenic T hyodysenteriae.  相似文献

Thirty-five crossbred barrows averaging 13.5 kg starting BW were used in a 35-d experiment to compare the availability of P and the nutritional value of two near-isogenic progeny of the barley cultivar 'Harrington'. Low-phytic acid barley (LPB, 0.35% total P, 0.14% phytic acid P) was homozygous for the low-phytic acid 1-1 allele, and the normal barley (NB, 0.35% total P, 0.24% phytic acid P) was homozygous for the normal allele of that gene. Pigs were fed individually twice daily in metabolism pens. Barley was the only source of phytate in semipurified diets, 1 to 3. Diet 1 contained 75% NB, 0.14% estimated available P (aP), and 0.50% Ca. Diet 2 contained 75% LPB, 0.22% aP, and 0.50% Ca. No inorganic P (iP) was added to Diets 1 and 2 in order to measure the animal response to the different concentrations of aP in these cultivars. Diet 3 was NB Diet 1 supplemented with iP to equal the concentration of aP in LPB Diet 2. Practical barley-soybean meal (SBM)-type diets, NB Diet 4 and LPB Diet 5, were formulated to meet all minimum nutrient requirements, and contained 0.30% aP and 0.65% Ca. For the semipurified diets, pigs fed LPB Diet 2 had higher (P < or = 0.05) bone ash weight, bone breaking strength, P absorption and retention, and Ca absorption and retention compared with pigs fed NB Diet 1, with a trend (P = 0.10) for pigs fed LPB Diet 2 to have a higher ADG and gain:feed ratio than pigs fed NB Diet 1. However, pigs fed LPB Diet 2 or NB Diet 3 were not different (P > or = 0.3) in growth performance, fresh bone weight, fat-free dry bone weight, bone ash, bone breaking strength, or N utilization. This indicates that LPB and NB were equal in nutritional value after supplementation of NB with iP to equal the estimated aP in LPB. For the practical barley-SBM diets, there were no differences (P > or = 0.4) between pigs fed NB Diet 4 or LPB Diet 5 for growth performance, fresh bone weight, bone breaking strength, the percentages of P and Ca utilization, or N, DE, and ME utilization. The use of LPB in pig diets reduced P excretion in swine waste by 55% and 16% in our semipurified and practical diets, respectively, compared with NB. Using our in vitro procedure designed to mimic the digestive system of the pig, the availability of P for pigs was estimated at 52% for LPB and 32% for NB.  相似文献

Swine dysentery (SD) is a common disease among pigs worldwide, which contributes to major production losses. Antimicrobial susceptibility testing of B. hyodysenteriae, the etiological agent of SD, is mainly performed by the agar dilution method. This method has certain limitations due to difficulties in interpretation of results. The aim of this study was the analysis of antimicrobial susceptibility of Brachyspira hyodysenteriae (B. hyodysenteriae) Polish field isolates by broth microdilution procedure. The study was performed on 21 isolates of B. hyodysenteriae, collected between January 2006 to December 2010 from cases of swine dysentery. VetMIC Brachyspira panels with antimicrobial agents (tiamulin, valnemulin, doxycycline, lincomycin, tylosin and ampicillin) were used for susceptibility testing of B. hyodysenteriae. The minimal inhibitory concentration (MIC) was determined by the broth dilution procedure. The lowest antimicrobial activity was demonstrated for tylosin and lincomycin, with inhibition of bacterial growth using concentrations > 128 microg/ml and 32 microg/ml, respectively. In the case of doxycycline, the MIC values were < or = 2.0 microg/ml. No decreased susceptibility to tiamulin was found among the Polish isolates and MIC values for this antibiotic did not exceed 1.0 microg/ml. The results of the present study confirmed that Polish B. hyodysenteriae isolates were susceptible to the main antibiotics (tiamulin and valnemulin) used in treatment of swine dysentery. Further studies are necessary to evaluate a possible slow decrease in susceptibility to tiamulin and valnemulin of B. hyodysenteriae strains in Poland.  相似文献

Groups of five pigs were vaccinated at three to four weeks old with either formolised Treponema hyodysenteriae in oil adjuvant alone, formolised T hyodysenteriae in oil adjuvant plus formolised Campylobacter coli in oil adjuvant, or sterile medium in oil adjuvant (as a control). Each group was challenged four weeks after vaccination by oral dosing on two consecutive occasions with pure cultures of the homologous strain of T hyodysenteriae plus direct contact with two pigs exhibiting severe swine dysentery. The disease was observed in two of five pigs immunised with T hyodysenteriae alone, three of five pigs immunised with T hyodysenteriae plus C coli and all five controls; haemorrhagic diarrhoea was exhibited only by the control group. Each pig immunised with T hyodysenteriae (alone or with C coli) recovered spontaneously, whereas four controls died. Vaccination with T hyodysenteriae also markedly reduced the severity and duration of clinical signs and of weight loss. No differences were observed in response to challenge between pigs immunised with T hyodysenteriae alone and pigs immunised with T hyodysenteriae plus C coli.  相似文献

When pure cultures of Treponema hyodysenteriae were orally inoculated into pigs, severe disease characteristic of swine dysentery developed. Less severe lesions were produced by oral inoculation of infective minced colon. Noninoculated pigs were used as controls. Inoculations of surgically isolated porcine colonic loops with either pure cultures or infective minced colon produced lesions only in the injected loop; the adjacent noninjected colon remained normal. Pigs and other experimental animals, including rabbits, guinea pigs, and mice, inoculated with washed cultures by various parenteral routes remained normal.  相似文献

The 21 field isolates of Treponema hyodysenteriae which were tested were sensitive to 3-acetyl-4'-isovaleryl tylosin (AIV); the minimal inhibitory concentration was 0.25 to 16 micrograms/ml. 3-Acetyl-4'-isovaleryl tylosin administered prophylactically to pigs at concentrations of 5 to 100 mg/kg of feed and tylosin at 110 mg/kg of feed for 28 or 31 days prevented swine dysentery induced by tylosin-sensitive T hyodysenteriae strain SQ2; 15 nonmedicated, inoculated control pigs had bloody diarrhea, and 9 pigs died. In 2 additional trials, AIV administered prophylactically for 28 days at 55 or 110 mg/kg of feed prevented swine dysentery induced by tylosin-insensitive T hyodysenteriae strain B204. All of the inoculated principal pigs medicated with AIV at 55 or 110 mg/kg of feed or carbadox at 55 mg/kg of feed and the noninoculated sentinel pigs for each group had solid feces throughout the 56-day trial. In the nonmedicated, inoculated control groups, bloody diarrhea began at 4 to 5 days after inoculation was done, and 9 of 10 principal pigs and 6 of 9 sentinel pigs had dysentery; 2 pigs died. In the groups medicated with AIV at 27.5 or 5.5 mg/kg of feed, all 5 principal pigs and 3 or 4 sentinel pigs in each group had dysentery; 3 or 4 pigs in each group died. In the group medicated with tylosin at 110 mg/kg of feed, 7 of 10 principal pigs and all 9 sentinel pigs had dysentery; 1 pig died.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Thirty-five crossbred barrows averaging 14.5 kg initial BW were used in a 5-wk experiment to compare the P availability and nutritional value of a low-phytate hybrid corn (LPC, 0.26% total P, 0.08% phytic acid P) homozygous for the lpa 1-1 allele with a nearly isogenic normal hybrid corn (NC, 0.25% total P, 0.20% phytic acid P). The pigs were fed individually twice daily in metabolism pens. Three semipurified diets were created in which corn was the only source of phytate. Diet 1 contained 72% NC, 0.15% estimated available P (aP) and 0.55% Ca. Diet 2 contained 72% LPC, 0.24% aP, and 0.55% Ca. The only differences between Diets 1 and 2 were the source of corn and the levels of aP. No inorganic P (iP) was added to these diets in order to measure the animal response to the different levels of aP in the corn hybrids. Diet 3 was NC Diet 1 supplemented with iP to equal the level of aP in LPC Diet 2. Diets 4 and 5 were practical corn-soybean meal diets formulated with each corn to meet all minimum nutrient requirements and contained 0.30% aP and 0.65% Ca. For the semipurified diets, pigs fed LPC Diet 2 had higher (P < 0.01) growth performance, bone breaking strength, P absorption and retention, Ca absorption and retention, and N retention than pigs fed NC Diet 1. However, when the NC diet was supplemented with iP to equal the aP in the LPC diet, most criteria were similar (P > or = 0.2), indicating an equal nutritional value for both corn hybrids after adjusting for phytate level. The only treatment difference, other than P excretion, between the practical corn diets supplemented with soybean meal was a higher (P < 0.05) bone breaking strength for pigs fed LPC Diet 5 compared with NC Diet 4. The use of LPC in pig diets reduced P excretion in swine waste by 50 and 18.4% in the semipurified and practical diets, respectively, compared with NC. Using our in vitro procedure designed to simulate the digestive system of the pig, the availability of P for pigs was estimated at 56% for LPC and 11% for NC.  相似文献

SUMMARY A prototype vaccine that is being developed for the control of swine dysentery (SD) was tested in two groups of experimental pigs. Vaccination induced high circulating antibody titres against the aetiological agent, Serpulina (Treponema) hyodysenteriae .
Pigs in the first trial were vaccinated twice before being challenged orally with the bacteria. Five of 6 unvaccinated animals developed dysentery within a fortnight of challenge, but only 1 of 6 vaccinated pigs showed signs of disease at this time. Unexpectedly, 1 mo after challenge, the surviving unvaccinated pig and 2 remaining healthy vaccinated animals succumbed to the disease. The reason for the development of this late-onset form of dysentery was not clear.
In the second trial, 8 pigs were vaccinated 3 times. Only 2 of these animals (25%) developed severe dysentery after being mixed with infected pigs, whereas 7 of 8 (88%) unvaccinated control pigs in the same pen became diseased. The late-onset form of dysentery was not observed.
The prototype vaccine for SD provided a useful level of protection, and could be used in programs to control the disease in Australia.  相似文献

Swine dysentery was induced in pigs and in ligated colonic segments by inoculation of pure cultures of, or colonic contents containing, Treponema hyodysenteriae. The mildest changes, best seen in ligated segments 48 or 72 hours after inoculation, were congestion and leucocytic margination in mucosal capillaries and depletion of mucigen from goblet cells lining the base of the crypts of Lieberkühn. Superficial mucosal necrosis and crypt cell hyperplasia were later changes. Perfusion studies with India ink did not demonstrate occlusive mucosal ischemia in acute swine dysentery. Mucosa with lesions of swine dysentery contained at least 10(5) colony forming units of T. hyodysenteriae per gram. Mucosa without lesions had 10(5) or fewer T. hyodysenteriae per gram. Segments with acute swine dysentery were distended with clear mucoid fluid with electrolyte composition indicative of net colonic secretion. No increase in the concentration of volatile fatty acids was detected in content from intact colons or colonic segments with lesions of acute swine dysentery.  相似文献
