Around 20,000 samples are analysed each year for the presence of antibacterial residues in Danish finisher pigs, and between zero and five samples are detected positive above the maximum residue level (MRL). The intention was to develop a risk-based surveillance programme involving fewer samples while ensuring equal sensitivity. Therefore, risk indicators were searched for. Data were obtained from the Danish slaughterhouse database covering the period from July 2010 to December 2012. Residues were found or suspected in 17 incidents. In nine of these, the farmer had called in to prevent the pigs from being slaughtered. Hence, eight suspect cases were found through the surveillance programme, and two of these were above MRL. For these eight case herds, the number of pigs slaughtered and the number in which each of the following lesions were found were included in a statistical analysis: chronic pleuritis, tail bite, chronic pericarditis, chronic pneumonia, chronic peritonitis, osteomyelitis, abscess in hindquarters, abscess in leg/toe and abscess in forequarters. Only chronic pleuritis was associated with the presence of residues. Next, data from all herds delivering pigs for slaughter to the same abattoir were included covering a 3-month period prior to the residue finding. The prevalence of chronic pleuritis was on average 1.7 times higher in the eight case herds compared to all other herds. In two herds, the prevalence was significantly higher (P ≤ 0.05), and in one herd substantially higher, but only borderline significant (P = 0.1). In the remaining herds, the prevalence did not differ from the other herds delivering pigs to the abattoir. This indicates that chronic pleuritis might be considered as a risk indicator for use in surveillance. Other risk indicators/factors – reported in the cases where the farmers called in – were inadequate marking of treated animals and incorrect use of medication dispensers. These factors are not suited for use in surveillance and should be dealt with otherwise.  相似文献

The aim of the present paper was to determine an influence of the presence and a level of intestine parasites infection on the quality of pork carcass expressed by the content of meat in carcass (meatiness) in pigs. The experimental part of the study was conducted on pigs farm produced in a closed cycle. The population in the study included 120 fattening pigs maintained in two keeping systems: group I--60 individuals kept on slatted floor, and group II--60 individuals kept on deep litter. All the experimental animals were treated in the same manner. The analysed fatteners were slaughtered in Meat Processing Plant when their body mass reached 110 kg, and the post-slaughter assessment was conducted according to the EUROP classification of pigs carcass using the Ultra-Fom 300 device. The study concerning the internal parasites were conducted basing on coproscopic quantitative McMaster method. As a results, the eggs of three nematode taxa were isolated and identified: Oesophagostomum spp., Ascaris suum and Strongyloides ransomi. Overall prevalence of infection of fatteners kept on litter was lower (25%±11.2) as compared to those kept on slatted floor (38.3%±12.6), however the differences were not statistically significant (χ(2)=2.465; df=1; P=0.116). The mean value of meatiness for pigs free from parasites was 53.68, while in the case of infected pigs the meatiness was statistically lower and was 52.12 (t=2.35; P=0.02). The analysed pigs were classified into three categories and conducted analysis of an influence of parasites on meatiness demonstrate the relationship that is statistically significant. The analysis of correlation between meatiness and an average number of helminth eggs also demonstrated the negative, statistically significant, relationship (F=5.52; P=0.020), i.e. in fatteners with higher EPG value the meatiness was lower.  相似文献

Three different parasites of the phylum Parabasala (Tritrichomonas foetus, Trichomitus rotunda and Tetratrichomonas buttreyi) have been described in pigs. In a previous study (Mostegl et al., 2011) approximately 47% of 91 paraffin wax-embedded intestinal samples of pigs which were Trichomonas-positive by in situ hybridization using a probe with a broad reactivity spectrum contained other species than T. foetus. Out of these, intestinal trichomonads from three pigs (pigs 1-3) were further analyzed by gene sequencing of a part of the 18S ribosomal RNA (rRNA) gene using primer walking. Subsequently, the partial sequences achieved by the different primer pairs were combined to a sequence of about 1000 bp for each trichomonad. In all three pigs unique sequences were acquired which showed only moderate similarities to sequences available in the GenBank. Alignments and the BLAST analysis showed a high degree of homology between sequences of trichomonads from pig 1 and pig 3 with only 1% difference. These sequences were found to be 92% similar to Hypotrichomonas acosta, a trichomonad isolated from squamate reptiles. The trichomonad sequence detected in the intestine of pig 2 showed about 10% nucleotide differences compared to pigs 1 and 3. This sequence was 97% similar to two Trichomitus batrachorum (a frog symbiont) sequences. A phylogenetic analysis using the neighbor-joining and maximum likelihood methods supported the data of the BLAST analysis. These results suggest the presence of at least two as yet undescribed trichomonad species in the intestinal contents of pigs.  相似文献

The present study which includes two feeding experiments was performed to investigate a possible performance enhancing effect of rare earth elements (REF) in piglets. This performance enhancing effect has been described in the Chinese literature for a long time, however, it was never tested under "western conditions". In the first feeding experiment 72 piglets at a mean BW of 7.3 kg were allotted to a control and to 4 REE groups at different levels of lanthanum chloride or an REE mixture containing mainly chlorides of lanthanum, cerium and praseodymium. The experimental period lasted 5 weeks. Positive effects of REE were found on body weight gain as well as on feed conversion ratio of the piglets. Compared to the control group, the daily weight gain was improved by 2 to 5% and feed conversion was better by up to 7%. These effects were, however, not significant. In the second feeding experiment, piglets (mean BW 17.3 kg) were fed for 8 weeks with a similar REE mixture. Significant positive effects of REE were found on both body weight gain and on feed conversion ratio by 19% and 10%, respectively. This is the first time that a performance enhancing effect of REE in pigs under western feeding conditions has been shown. Since the use of antibiotics as growth promoters in animal feed has been restricted in the European Union recently, rare earth elements might be of interest as new, safe and inexpensive alternative performance enhancers.  相似文献

Background

The major indication for antibiotic use in Danish pigs is treatment of intestinal diseases post weaning. Clinical decisions on antibiotic batch medication are often based on inspection of diarrhoeic pools on the pen floor. In some of these treated diarrhoea outbreaks, intestinal pathogens can only be demonstrated in a small number of pigs within the treated group (low pathogen diarrhoea). Termination of antibiotic batch medication in herds suffering from such diarrhoea could potentially reduce the consumption of antibiotics in the pig industry. The objective of the present pilot study was to suggest criteria for herd diagnosis of low pathogen diarrhoea in growing pigs.Data previously collected from 20 Danish herds were used to create a case series of clinical diarrhoea outbreaks normally subjected to antibiotic treatment. In the present study, these diarrhoea outbreaks were classified as low pathogen (<15% of the pigs having bacterial intestinal disease) (n =5 outbreaks) or high pathogen (≥15% of the pigs having bacterial intestinal disease) (n =15 outbreaks). Based on the case series, different diagnostic procedures were explored, and criteria for herd diagnosis of low pathogen diarrhoea were suggested. The effect of sampling variation was explored by simulation.

Results

The diagnostic procedure with the highest combined herd-level sensitivity and specificity was qPCR testing of a pooled sample containing 20 randomly selected faecal samples. The criteria for a positive test result (high pathogen diarrhoea outbreak) were an average of 1.5 diarrhoeic faecal pools on the floor of each pen in the room under investigation and a pathogenic bacterial load ≥35,000 per gram in the faecal pool tested by qPCR. The bacterial load was the sum of Lawsonia intracellularis, Brachyspira pilosicoli and Escherichia coli F4 and F18 bacteria per gram faeces. The herd-diagnostic performance was (herd-level) diagnostic sensitivity =0.99, diagnostic specificity =0.80, positive predictive value =0.94 and negative predictive value =0.96.

Conclusions

The pilot study suggests criteria for herd diagnosis of low pathogen diarrhoea in growing pigs. The suggested criteria should now be evaluated, and the effect of terminating antibiotic batch medication in herds identified as suffering from low pathogen diarrhoea should be explored.

Background

Overconsumption of antibiotics in the pig industry is of concern in relation to antimicrobial resistance. False positive disease diagnosis may result in the treatment of healthy animals. In Denmark, diarrhoea is the most common cause of antibiotic treatment in pigs. Farm personnel are not professional clinicians, which could result in inappropriate antibiotic treatments of diarrhoea.The primary objectives of this pilot study using digital pictures of faecal pools was to evaluate farmers’ diagnostic performance in the assessment of faecal consistency in nursery pigs and to investigate the effect of different co-variables, including practical experience. A secondary objective was to compare the diagnostic performance of farmers with that of veterinarians.At a pig congress, observers (farm personnel and veterinarians) working professionally with pigs participated in a faecal consistency test consisting of 16 pictures of faecal pools (eight diarrhoeic and eight non-diarrhoeic). The faecal pools had previously been collected and subjected to faecal dry matter determination. The true status of the faecal pools was determined by the faecal dry matter content (diarrhoea: faecal dry matter ≤ 18%). The true status was used to evaluate the farmers’ and veterinarians’ diagnostic performance.

Results

A total of 119 farmers and 18 veterinarians were included in the statistical analysis. For the farmers, the mean proportion of faecal pools assessed as diarrhoeic was 0.48, the mean proportion of correctly classified faecal pools was 0.84, the mean diagnostic sensitivity was 0.83 and the mean diagnostic specificity was 0.86. Farmers with less than four years of practical experience detected clinical diarrhoea more accurately than farmers with more than four years of practical experience (p < 0.05). No significantly differences between farmers and veterinarians was observed (p > 0.20).

Conclusions

The results, using digital pictures of faecal pools, suggest that farmers and veterinarians have similar diagnostic performance in relation to diarrhoea. False positive classification of non-diarrhoeic pigs appears to be a larger problem than false negative classification of diarrhoeic pigs under Danish conditions. If these results can be confirmed under practical conditions, training in, and validation of, clinical diagnoses may be an important factor in reducing antibiotic consumption in the pig industry.  相似文献

As clinical toxoplasmosis is not considered a problem in pigs, the main reason to implement a control strategy against Toxoplasma gondii (T. gondii) in this species is to reduce the establishment of T. gondii tissue cysts in pork, consequently reducing the risk of the parasite entering the human food chain. Consumption of T. gondii tissue cysts from raw or undercooked meat is one of the main sources of human infection, with infected pork being considered a high risk. This study incorporates a mouse bioassay with molecular detection of T. gondii DNA to study the effectiveness of vaccination (incomplete S48 strain) in its ability to reduce tissue cyst burden in pigs, following oocyst (M4 strain) challenge. Results from the mouse bioassay show that 100% of mice which had received porcine tissues from vaccinated and challenged pigs survived compared with 51.1% of mice which received tissues from non-vaccinated and challenged pigs. The presence (or absence) of T. gondii DNA from individual mouse brains also confirmed these results. This indicates a reduction in viable T. gondii tissue cysts within tissues from pigs which have been previously vaccinated with the S48 strain. In addition, the study demonstrated that the main predilection sites for the parasite were found to be brain and highly vascular muscles (such as tongue, diaphragm, heart and masseter) of pigs, while meat cuts used as human food such as chop, loin, left tricep and left semitendinosus, had a lower burden of T. gondii tissue cysts. These promising results highlight the potential of S48 strain tachyzoites for reducing the number of T. gondii tissues cysts in pork and thus improving food safety.  相似文献

The mechanism for prostaglandin (PG) F release from pig endometrium after oxytocin (OT) treatment is unknown. OT may rapidly stimulate inositol (1,4,5)-trisphosphate (IP3) and diacylglycerol (DAG) formation, consistent with the concept of rapid activation of a second-messenger system. In support of this hypothesis, endometrial IP3 levels were increased (P < 0.05) within 0.5 min after treatment with 0.1 μM OT. In contrast, increased DAG formation was not detected after treatment with OT. However, similar to the stimulation of endometrial PGF secretion observed after OT treatment (P < 0.001), PGF release was increased (P < 0.01) after treatment with phorbol-12myristate-l3-acetate (PMA), which mimics DAG activation of protein kinase C. Further, stimulation of endometrial PGF secretion did not result from cell death induced by PMA or OT because lactate dehydrogenase, a cytosolic marker of cellular integrity, did not leak into the medium after PMA or OT treatment. In contrast, 0.5% saponin (positive control for cell death and concomitant release of lactate dehydrogenase) increased PGF secretion (P < 0.05) and lactate dehydrogenase release (P < 0.001). These results indicate that OT induces endometrial IP3 production in a rapid manner indicative of a second-messenger system. The finding that increased DAG was not also detected after OT treatment may reflect rapid metabolism or compartmentalized production of DAG involved in the second-messenger stimulation of phospholipase C. The high background of DAG used in the biosynthesis of cellular lipids would obscure the rather small spatially localized changes in DAG levels resulting from the activation of phospholipase C. The finding that DAG was present at approximately 10 to 20-fold higher levels than IP3 in resting cells was consistent with this conclusion.  相似文献

Body condition has been shown to affect the pharmacokinetics of subcutaneously administered macrocyclic lactone anthelmintics but the underlying mechanism is unknown. This study examined the effect of different rates of fat deposition on the pharmacokinetics of moxidectin (MXD) and ivermectin (IVM). All animals initially received a diet with a high linoleic acid content for 7 weeks. One group of animals then received a normal grower diet while the other half received a maintenance ration. Within each diet group, animals were treated with either IVM (n = 4) or MXD (n = 4) or remained as untreated controls (n = 2). There was no difference in the proportion of linoleic acid between the drug treated groups and the untreated controls at any time throughout the study. At 4 and 9 weeks after treatment there was a significantly lower proportion of linoleic acid in the pigs fed the normal ration indicating a greater fat deposition in these animals compared with those that received the maintenance diet. There was an increased persistence of MXD in the plasma of pigs fed the normal ration compared with those fed the maintenance ration. No differences were seen in the kinetic disposition of IVM between pigs fed the maintenance or normal ration. Reducing the rate of fat deposition influenced the pharmacokinetic disposition of the highly lipophilic MXD but did not influence the pharmacokinetic disposition of the less lipophilic IVM.  相似文献

In South-East Asia, rice distillers’ by-product (RDP) is a widely abundant feedstuff whose adequate incorporation into pig diets is still questionable. Especially, effects of RDP on nutrient digestibility of growing pigs fed corn-soybean meal-based diet are lacking. The objective of this study was to determine nutrient digestibility and energy value of ordinary (ORDP) and glutinous (GRDP) rice distillers’ by-product in growing pigs. Two groups of 12 castrated crossbred barrows (Duroc × Landrace × Yorkshire), about 3 months old, 38?±?1.04-kg initial body weight, were each allocated to a 15-day experiment in which pigs were divided into three isonitrogenous diet-groups. The animals were housed individually in metabolism cages for separated collection of feces and urine. Each diet-group was provided either a control corn-soybean meal diet or a diet in which corn and soybean meal were partly replaced by ORDP or GRDP at 15 or 30% on diet dry matter basis. Glutinous by-product especially showed higher levels in crude protein, neutral/acid detergent fiber, total branched-chain amino acids, and butyric acid. When compared to the control diet, ORDP tented to increase DM intake (P?= 0.054) but decreased energy (P?< 0.001) and crude protein (P?< 0.04) digestibility, while GRDP decreased DM intake (P?< 0.001). Both GRDP and ORDP products negatively affected digestibility of ether extract. The average digestible and metabolizable energy of ORDP and GRDP were 17.0 and 16.6, and 17.7, and 17.1 MJ/kg DM, respectively. In conclusion, these results show that both RDP, and especially GRDP, are highly valuable protein and energy sources for pig production.

-Adrenoceptors were identified and characterized by [3H]dihydroalprenolol ([3H]DHA) binding experiments in muscle membrane preparations from piglets. The [3H]DHA binding was rapid, reversible and stereoselective. Catecholamines competed for specific binding with a rank order of potency (-)-isopropylnoradrenaline > (-)-epinephrine » (-)-norepinephrine, indicating a -subtype of adrenoceptor. Saturation binding experiments with [3H]DHA showed no significant difference for either the number of binding sites or the equilibrium dissociation constants in normal and splayleg pigs.Adenylate cyclase assays indicated that the basal adenylate cyclase activity and the prostaglandin E1 (PGE1)-, (-)-isopropylnoradrenaline (ISO)-, 5-guanylyl-imidodiphosphate (GppNHp)- and sodium fluoride (NaF)-stimulated values were not significantly different in normal and splayleg pigs. In both groups, PGE1 did not affect basal activity, whereas ISO and GppNHp stimulated adenylate cyclase activity significantly (p<0.001) to about 40% above basal level. NaF induced a significant (p<0.001) increase of cAMP in normal and splayleg pigs amounting to 48% and 61% respectively. Significant correlations between absolute adenylate cyclase responses to ISO (r=0.83***), NaF (r=0.72**) and GppNHp (r=0.61*) and basal activity in the splayleg pigs were the most striking findings. In contrast, these correlations could not be detected in the normal pigs. Whether or not this observation reflects an alteration in the signal transduction system needs to be further investigated.To the best of our knowledge this is the first biochemical study which relates an altered -adrenoceptor function and porcine splayleg.  相似文献

Summary

Broiler by‐products (heads, feet, and viscera) mixed with 4% dextrose were pasteurized for 4 min at 90°C core temperature, cooled to 20°C, and fermented with Lactobacillus plantarum as starter culture. These fermented poultry by‐products were fed to 12 individually housed fattening pigs as part (17.6% of the dry matter) of their fattening ration, the remainder composed of compound pig feed. Control pigs received a compound pig feed only. Both groups of pigs were fed restrictively on the basis of body weight. The technical results of the pigs fed the experimental diet showed a significantly improved feed:gain ratio (2.46 vs 2.57), a significantly higher carcass weight (86.1 vs 81.8 kg), a lower meat percentage (50.9 vs 52.5%) and an increased backfat thickness (21.5 vs 18.7%). The bacterial flora in the intestinal tract of the pigs fed the experimental diet differed significantly from the control animals. Decreased colony counts of mesophilic aerobic bacteria, Enterobacteriaceae, enterococci and lactobacilli were found in the rectal content and the prevalence of salmonella was lower.

It is suggested that the improved feed:gain ratio and the reduced bacterial activity of the measured groups of bacteria is a result of 1) the higher energy content of the diet, and(or) 2) an assumed enhanced digestibility of nutritional components in the diet, and(or) 3) the lower incidence of diarrhea in the pigs fed with fermented poultry by‐products. This resulted in a lower contamination level of enteropathogenic bacteria like, salmonella and Escherichia coli, in the gastro‐intestinal tract of the pigs fed fermented poultry by‐products.  相似文献

The establishment of pregnancy is a complex process that requires a well-coordinated interaction between the implanting conceptus and the maternal uterus. In pigs, the conceptus undergoes dramatic morphological and functional changes at the time of implantation and introduces various factors, including estrogens and cytokines,interleukin-1β2(IL1 B2), interferon-γ(IFNG), and IFN-δ(IFND), into the uterine lumen. In response to ovarian steroid hormones and conceptus-derived factors, the uterine endometrium becomes receptive to the implanting conceptus by changing its expression of cell adhesion molecules, secretory activity, and immune response. Conceptus-derived estrogens act as a signal for maternal recognition of pregnancy by changing the direction of prostaglandin(PG) F2αfrom the uterine vasculature to the uterine lumen. Estrogens also induce the expression of many endometrial genes,including genes related to growth factors, the synthesis and transport of PGs, and immunity. IL1 B2, a pro-inflammatory cytokine, is produced by the elongating conceptus. The direct effect of IL1 B2 on endometrial function is not fully understood. IL1 B activates the expression of endometrial genes, including the genes involved in IL1 B signaling and PG synthesis and transport. In addition, estrogen or IL1 B stimulates endometrial expression of IFN signaling molecules,suggesting that estrogen and IL1 B act cooperatively in priming the endometrial function of conceptus-produced IFNG and IFND that, in turn, modulate endometrial immune response during early pregnancy. This review addresses information about maternal-conceptus interactions with respect to endometrial gene expression in response to conceptus-derived factors, focusing on the roles of estrogen and IL1 B during early pregnancy in pigs.  相似文献

Nutrient composition of brewers’ spent grains (BSG) from two industries and sow and weaner meal (SWM) were different (p < 0.05). In completely randomised design (CRD), thirty (30) Landrace x Pietrain crosses 35 to 40 ± 5 kg were randomly allocated to 10 pens each with 3 pigs based on the initial weight. Five diets were assigned to pig, where BSG replaced SWM at 0% (T1), 25% (T2), 50% (T3), 75% (T4) and 100% (T5) on DM basis. Daily feed intake and weekly live weight, feed conversion ratio (FCR) and the net profit were recorded for 42 days. Feed intake increased from T1-T3 and then decreased (p < 0.05). The average daily gains (ADG) were different (p < 0.05), but T1 to T3 were similar (p > 0.05). FCR was similar (p > 0.05) from T1 to T3 but different for T4 and T5 (p < 0.05). Feed cost decreased with the increase in BSG levels. The net return was highest in T3 (50% BSG) than in T1 and T2 but lower in T5. It was concluded that BSG can replace 50% SWM in grower pigs’ diets without adverse effect on daily gain with good economic returns.  相似文献

Mycobacterium bovis recognizes as hosts a wide spectrum of animal species. In particular epidemiological situations, high prevalence of infection is found also in pigs. In the present study, we evaluated the capability of the interferon-gamma (IFN-γ) assay to identify pigs infected with M. bovis. The results of the immune-diagnosis were correlated to the findings of the post mortem inspection and the bacterial culture of lymph nodes. Blood samples of 146 pigs, belonging to a local breed of Sicily reared in free or semi-free roaming conditions, were collected to assess the specificity and the sensibility of the IFN-γ assay. Thirty-one pigs, from M. bovis free herds, did not react to the IFN-γ assay, yielding a specificity of 100%. The IFN-γ assay identified 15 out of 19 animals positive to the bacterial culture and 22 out of 26 animals with tuberculous lesions, with a sensibility of 78.9-84.6%, respectively. Out of 26 reactors to the test, 15 pigs (57.7%) confirmed to be infected after the bacterial culture and 22 (84.6%) had tuberculous lesions. The IFN-γ assay was able to reveal 4 animals with no visible lesions (NVL). Together, these findings support the feasible use of the IFN-γ assay as an intra vitam tool for the surveillance and management of M. bovis infection in swine populations.  相似文献
