The in vitro effect of an isolate of the nematophagous fungus Pochonia chlamydosporia (VC1) on the eggs of Anoplocephala perfoliata was evaluated. The eggs were morphologically analyzed for their integrity using light microscopy (10× objectives), plated on 9.0-cm diameter petri dishes containing 2% WA culture medium with and without fungal isolate (control), grown for 10 days, and 10 replicates were prepared per group. In all, 1000 eggs of A perfoliata were plated on petri dishes containing 2% water agar culture medium with (VC1) and without the fungal isolate (control). After 3, 5, 7, and 10 days, approximately 100 eggs were removed from each plate and classified on the basis of the following parameters: without alteration; type 1, lytic effect without morphological damage to eggshell; type 2, lytic effect with morphological alteration of embryo and eggshell; and type 3, lytic effect with morphological alteration of embryo and eggshell, in addition to hyphal penetration and internal egg colonization and destruction. The P chlamydosporia fungus demonstrated ovicidal activity (P < .05) on the eggs of A perfoliata in the studied intervals presenting type 3 effects of 35%, 42.5%, 53.83%, and 71.17% for the intervals 3, 5, 7, and 10 days, respectively. P chlamydosporia is a potential biological control agent for the eggs of A perfoliata.  相似文献

The ovicidal effect of the nematophagous fungus Pochonia chlamydosporia on eggs of Ascaris suum was tested under laboratory conditions. A. suum eggs were plated on 2% water–agar with seven fungal isolates (Isol. 5, Isol. 31, Isol. 1, VC1, Isol. 12, Isol. 22 and VC4) and control without fungus. After 5, 7, 10, 14, 15 and 21 days of incubation, approximately 100 eggs were removed from the plates and classified according to the following parameters: type 1, biochemical and physiological effect without morphological damage to the eggshell, type 2, lytic effect with morphological alteration of the eggshell and embryo and type 3, lytic effect with morphological alteration of eggshell and embryo showing hyphal penetration and internal egg colonization. The isolates effectively destroyed A. suum eggs and all types of effects were observed during the experiment. There was no variation in ovicidal capacity (type 3 effect) among the isolates (p > 0.05) throughout the experiment. After 21 days, isolate 5 showed the highest percentages of type 3 effect (58.33%). The results indicated that P. chlamydosporia (Isol. 5, Isol. 31, Isol. 1, VC1, Isol. 12, Isol. 22 and VC4) can destroy A. suum eggs and is, therefore, a potential biological control agent of nematodes.  相似文献

The present study aimed to demonstrate by scanning electron microscopy (SEM) the in vitro predatory activity of nematophagous fungi Pochonia chlamydosporia (VC1 and VC4 isolates) Duddingtonia flagrans (AC001 isolate) and Monacrosporium thaumasium (NF34a isolate) on eggs of Trichuris trichiura and infective larvae (L3) of Haemonchus contortus. The work was divided into two experimental tests (A and B). In tests A and B, the predatory activity of nematophagous fungi P. chlamydosporia, D. flagrans and M. thaumasium on eggs of T. trichiura and H. contortus L3 was observed. After 6 h, in test A, isolates P. chlamydosporia (VC1 and VC4) had a role in destroying eggs of T. trichiura. For fungi D. flagrans and M. thaumasium the ovicidal activity on T. trichiura eggs was not observed. Test B showed that D. flagrans (AC001) and M. thaumasium (NF34a) were capable of predating H. contortus L3, but no predation by the fungus P. chlamydosporia was seen. These fungi can offer potential for the biological control of nematodes.  相似文献

The ovicidal activity of seven fungal strains: Acremonium alabamense, Alternaria chlamydospora, Cladosporium herbarum, Fusarium solani, Paecilomyces variotii, Paecilomyces viridis and Penicillium verruculosum isolated from urban soil samples from Poland was determined in vitro. The fungal mycelium was co-cultured with Ascaris suum eggs on plates with 2% water–agar for 28 days. Eggs exposed and unexposed (control) to fungal mycelium were observed weekly by light microscopy and the percentage of malformed eggs were determined. The eggs were classified according to following parameters: type 1 – biochemical and physiological effect without morphological damage to the eggshell; type 2 – lytic effect with morphological alteration of the eggshell and embryo; type 3 – lytic effect with morphological alteration of eggshell and embryo with hyphal penetration and internal egg colonization. All examined species of fungi extended embryogenesis, but the retardation of embryonic development was varied and depended on the species. A. alabamense, A. chlamydospora and P. verruculosum exhibited very high inhibitory activity on A. suum egg development. The fungus-exposed eggs revealed morphological alternations in all stages of embryogenesis. Isolates of F. solani, P. variotii and P. viridis showed hyphal penetration and internal colonization of A. suum eggs (type 3 effect). No appressoria were produced and simple hyphal penetrations were most commonly observed. A. alabamense and P. verruculosum demonstrated morphological destruction, with eggshell destruction. The remaining fungi showed type 1 effect. The results demonstrated that examined strains of F. solani, P. variotii and P. viridis may be considered to be potential limiting factors of parasitic geohelminth populations.  相似文献

The predatory capacity of the nematophagous fungus Pochonia chlamydosporia (isolate VC4) after passage through the gastrointestinal tract of dogs was assessed in vivo against Toxocara canis eggs. Twelve dogs previously wormed were divided into two groups of six animals and caged. The treatments consisted of a fungus-treated group (VC4) and a control group without fungus. Each dog of the fungus-treated group received a single 4 g dose of mycelial mass of P. chlamydosporia (VC4). Fecal samples from animals of both groups (treated and control) were collected at five different times (6, 12, 24, 36, and 48 h) after fungal administration, and placed in Petri dishes. Each Petri dish of both groups for each studied time interval received approximately 1000 T. canis eggs. Thirty days after the fecal samples were collected, approximately one hundred eggs were removed from each Petri dish of each studied time interval and evaluated by light microscopy (LM) and scanning electron microscopy (SEM). Microscopy examination of plates inoculated with the fungus showed that the isolate VC4 was able to destroy the T. canis eggs with destruction percentages of 28.6% (6 h), 29.1% (12 h), 32.0% (24 h), 31.7% (36 h), and 37.2% (48 h). These results suggest that P. chlamydosporia can be used as a tool for the biological control of T. canis eggs in feces of contaminated dogs.  相似文献

Biological control of gastrointestinal nematodiasis in ruminants is an alternative to reduce the number of infective larvae. The fungal isolates predatory activity preservation is a basic requirement for the success of this control type. The aim of this work is to evaluate the predatory capacity of the fungus Arthrobotrys robusta (isolate I-31), preserved on silica gel on infective larvae of Haemonchus contortus under laboratory conditions on 2 % water agar (2 % WA). In this essay, A. robusta storage on silica gel showed successful predatory activity on H. contortus L3 larvae (p?<?0.01) compared to the control group. Nematophagous fungi were not observed in the control group during the experiment. There was a significant reduction (p?<?0.01) of 73.84 % in the means of H. contortus (L3) recovered from treatment with isolate I-31 compared to the control without fungi. Results indicate that A. robusta (I-31) could survive stored on silica gel for at least 7 years and keep its predatory activity on H. contortus (L3).  相似文献

Ascaris suum is a gastrointestinal nematode parasite of swines. The aim of this study was to observe Pochonia chlamydosporia fungus on biological control of A. suum eggs after fungus passage through swines gastrointestinal tract. Eighteen pigs, previously dewormed, were randomly divided into three groups: group 1, treated with the fungus isolate VC4; group 2, treated with the fungus isolate VC1 and group 3 did not receive fungus (control). In the treated groups, each animal received a 9 g single dose of mycelium mass containing P. chlamydosporia (VC1 or VC4). Thereafter, animal fecal samples were collected at the following intervals: 8, 12, 24, 36, 48, 72 and 96 h after treatment beginning and these were poured in Petri dishes containing 2% water-agar culture medium. Then, 1,000 A. suum eggs were poured into each dish and kept in an incubator at 26°C and in the dark for 30 days. After this period, approximately 100 eggs were removed from each Petri dish and morphologically analyzed under light microscopy following the ovicidal activity parameters. The higher percentage observed for isolated VC4 eggs destruction was 57.5% (36 h) after fungus administration and for isolate VC1 this percentage was 45.8% (24 h and 72 h) (p > 0.01). P. chlamydosporia remained viable after passing through the gastrointestinal tract of swines, maintaining its ability of destroying A. suum eggs.  相似文献

This study was made to investigate efficacy of eprinomectin pour-on against to Toxocara vitulorum in calves. In the study, 16 calves naturally infected with T. vitulorum were divided into two groups as treatment (eight calves) and control (eight calves). Eprinomectin (0.5 mg/kg, Eprinex®, Merial) was given to treatment group calves, and eggs per gramme were determined in the faeces on the day of pre-treatment and the second, third, fourth, fifth, sixth, 14th and 28th days of post-treatment. No side effects associated with nervous, respiratory and gastrointestinal systems were observed. In conclusion, eprinomectin was determined to be 100% effective against T. vitulorum. This is the first study to evaluate the efficacy of eprinomectin against a natural T. vitulorum infection in calves.  相似文献

This study was completed to determine the prevalence and distribution of Toxocara vitulorum infection in cattle and buffalo calves and investigate its clinical impact in northern Lao PDR (Peoples Democratic Republic). The results aim to assist decisions on disease control measures that can contribute to increasing cattle and buffalo productivity within smallholder farming systems in tropical areas. A prevalence survey for T. vitulorum in buffalo and cattle calves aged <3 months was conducted between September 2009 and June 2010 in five provinces of northern Lao PDR using a two-stage sampling technique to select 69 villages and 899 calves, with faecal samples collected and examined for T. vitulorum eggs at a local laboratory. At the time of sampling, data on calf morbidity and anthelmintic treatment was also collected. Factors potentially associated with infection and severity of infection were analyzed at univariable and multivariable levels, using T. vitulorum status (positive/negative) and on the positive calves only, faecal egg count levels as outcome variables. The estimated prevalence of T. vitulorum in northern Lao was 22.6 % (95 % confidence interval [CI], 0.17–0.28), and 76.8 % of villages had at least one positive calf. Province was the only significant (p?<?0.05) variable investigated associated with calf infection status. Species (buffalo) was the only variable significantly (p?<?0.05) associated with higher egg per gram of faeces levels among infected calves. Prevalence in calves aged 1–21 days, the reported prepatent period, was 17.5 % (CI 0.11–0.24). Treatment levels were very low (8.2 %) and if treatment occurred it was mostly unsuccessful. The high and wide spread infection of T. vitulorum in cattle and buffalo calves identified in this survey is likely to result in suboptimal cattle and buffalo productivity. Improved management of T. vitulorum infection in cattle and buffalo calves in northern Lao PDR is indicated to reduce potential negative production impacts and enable more efficient development of large ruminant livestock industry as a pathway from rural poverty for smallholder farmers in northern Lao PDR. In addition to quantifying this disease problem in calves, the conduct of this applied participatory research study provided an important opportunity to improve animal health services by increasing the parasite, large ruminant handling and research knowledge and capacity of government animal health staff and farmers.  相似文献

An egg count survey using environmental fecal samples obtained in spring or early summer was conducted to estimate the apparent prevalence of Toxocara vitulorum in unweaned bison calves and of other intestinal parasites in adult bison on 98 farms in Manitoba and Saskatchewan. Calf samples were pooled (maximum 5 samples per pool) by farm and positive pools were examined to determine individual T. vitulorum counts. Toxocara vitulorum eggs were found on 4 farms in Manitoba and none in Saskatchewan. Apparent herd-level prevalence estimates were 12% [95% confidence interval (95% CI): 3.4% to 28.2%] and 0% (95% CI: 0% to 5.7%) respectively. Samples from adult bison contained eggs/oocysts from trichostrongyle species, Eimeria sp., Monieza sp., Capillaria sp., Nematodirus sp. and Trichuris sp. in 100%, 95%, 72%, 13%, 13%, and 5% of herds, respectively. Strongyloides sp. were not found in any herd. Further studies are needed to assess parasite distribution patterns in bison and to evaluate the risk that T. vitulorum may pose to bison, cattle, and wildlife.  相似文献

Ninety-six White laying hens (64 weeks old) were used to investigate the effect of dietary inclusion of Bacillus subtilis on performance and eggshell quality. Hens were randomly distributed into three treatment groups and fed basal diet (control), basal diet plus 0.5 g/kg B. subtilis (Probiotic-0.5), or basal diet plus 1 g/kg B. subtilis (Probiotic-1) for 10 weeks. Probiotic-1 group exhibited the maximum (P?<?0.05) increase in egg production, egg weight, egg mass, eggshell weight, and eggshell thickness, compared with Probiotic-0.5 and the control groups. During the last week of the study, eggshell densities were 94.9, 88.7, and 65.6 mg/cm2 in Probiotic-1, Probiotic-0.5, and the control group, respectively. The average unmarketable eggs were 1.3 %, 2.2 %, and 6.4 % in Probiotic-1, Probiotic-0.5, and the control group, respectively. Tibia weight, density, and ash content increased significantly. The average time required by B. subtilis to establish a significant effect was 3 weeks for Probiotic-1and 6 weeks for Probiotic-5. B. subtilis improved gut microflora balance, which has the potential to improve gut health and absorption efficiency. It should be possible to improve egg performance and eggshell quality of aged laying hens by dietary inclusion of B. subtilis, which will be of importance for farmers.  相似文献

Toxocara vitulorum infection in large ruminants is endemic in many tropical countries and particularly in South-East Asia. A single treatment of calves with pyrantel at 14–21 days of age effectively controls the parasite. Despite this treatment being readily available, T. vitulorum infection remains common and widespread. To understand drivers of effective control of T. vitulorum infection, we examined treatment practices and knowledge of smallholder farmers of this parasite plus determined annual calf morbidity and mortality and identified potential risk factors for these estimates. Interviews were conducted with 273 smallholder farmers who had calves tested for T. vitulorum 4–6 months earlier. Reproductive rates of 0.6 and 0.4 calf per annum in cattle and buffalo respectively, and annual calf morbidity and mortality of 42.6% (CI 0.38–0.47) and 37.3% (CI 0.33–0.42) respectively, were identified. Interviewed farmers had either none (80.6%) or only minimal (19.4%) knowledge about T. vitulorum and only 2.5% of the farmers treated their calves for T. vitulorum using the recommended control regime. Multivariable logistic regression analyses with random effects showed that the number of adult cattle per household, T. vitulorum infection status of the household herd and farmer knowledge of T. vitulorum were significantly associated with calf morbidity and mortality. Financial analysis using partial budgeting showed a net benefit of USD 3.69, 7.46, 11.09 or 14.86 per calf when treating calves with pyrantel and attributing 25%, 50%, 75% or 100% of morbidity and mortality to T. vitulorum infection. The study identified that poor reproduction, high calf morbidity and mortality combined with very limited farmer knowledge and effective control of endemic Toxocariasis, contribute to suboptimal large ruminant production in mixed smallholder farming systems in South-East Asia. The large net benefit per calf achievable by a single pyrantel treatment should drive implementation of this intervention by smallholder farmers, especially as demand for livestock products continues to increase in this region and forces a change to more production oriented farming. To support this, continued capacity building that ensures knowledge transfer of best practice T. vitulorum control to smallholder farmers is required.  相似文献

This study was conducted to investigate the efficacy of ivermectin (IVM), doramectin (DRM), and moxidectin (MXD) against Toxocara vitulorum in calves. In the study, 20 calves naturally infected with T. vitulorum were divided into four groups: three different treatment groups (n?=?5) and one positive control (n?=?5). The animals in each group received either IVM (Baymec®, Bayer), DRM (Dectomax®, Pfizer), or MXD (Cydectin®, Fort Dodge) by subcutaneous injection at a single dose of 0.2 mg/kg. Fecal egg counts were performed on all animals on days 0, 2, 4, 8, 12, and 16 post-treatment. In conclusion, IVM, DRM, and MXD significantly reduced the fecal egg counts on day 8 post-treatment (99.90%, 98.77%, and 99.57%, respectively). After the 12th day, IVM, DRM, and MXD were found to be 100% effective. There was no significant difference in efficacy between the three treatment groups at any of the sampling dates (P?>?0.05). No side effects associated with nervous, respiratory, and gastrointestinal systems were observed. This is the first study to evaluate the comparative efficacy of subcutaneous administration of ivermectin, doramectin, and moxidectin against naturally infected T. vitulorum in calves.  相似文献

This work was performed to determine the predatory capacity in vitro of the nematophagous fungus Duddingtonia flagrans (isolate AC001) on cyathostomin infective larvae of horse (L3). The experimental assay was carried out on plates with 2% water-agar (2% WA). In the treated group, each plate contained 1.000 L3 and 1.000 conidia of the fungus. The control group without fungus only contained 1.000 L3 in the plates. Ten random fields (4 mm diameter) were examined per plate of treated and control groups, every 24 h for seven days under an optical microscope (10× and 40× objective lens) for non-predated L3 counts. After 7 days, the non-predated L3 were recovered from the Petri dishes using the Baermann method. The interaction there was a significant reduction (p < 0.01) of 93.64% in the cyathostomin L3 recovered. The results showed that the D. flagrans is a potential candidate to the biological control of horse cyathostomin L3.  相似文献

The main goal in this research was to determine the beneficial effect of incorporating biological procedures in parasite control programs for equids in zoological parks. Two trials were developed for Equus quagga, E asinus , and E africanus asinus. The first trial (September 2010 to August 2011) consisted of chemotherapy only (ivermectin plus praziquantel), and the second trial (September 2011 to September 2012) consisted of administration of chemotherapy and chlamydospores of the nematophagous fungus Arthrobotrys (Duddingtonia) flagrans. The effect of these measures was evaluated by the estimation of the reduction in the fecal egg counts (FECR). In the first trial, 100% FECR values were achieved 15 days after treatment in all the animals. The egg reappearance period (ERP) was 2-3 months for the equids, and all of them were passing strongyle eggs in the feces at 2-4 months after their deworming. In the second experiment, the FECR values were 100% in the three species. ERPs of 3 months in the European donkeys, 4 months in the Africans, and 6 months in the zebras were recorded. All the equids had positive results for the coprological flotation test 4-8 months after anthelmintic administration. This preliminary study demonstrates the incorporation of chlamydospores of nematophagous fungus, as A flagrans appears highly promising for reduction of the infective stages of the strongyles affecting captive animals, but the experimental design precludes true determination of whether the treatment is fully efficacious.  相似文献

The present study assessed the pathogenic effect of isolates E9, IBCB425 and IBCB159 of the Metarhizium anisopliae fungus, JAB06, JAB07 and AM09 of Beauveria bassiana, IBCB133 and CB75 of Isaria fumosorosea (=Paecilomyces fumosoroseus) and CG189 and CG195 of Isaria farinosa (=Paecilomyces farinosus) against eggs and larvae of the horn fly Haematobia irritans. Eggs were inoculated with suspensions containing 106, 107 and 108 conidia ml−1 of the fungal isolates and observed after 48 h to determine viability. In the larvae study, eggs were allowed to hatch into fresh bovine feces that had been treated with 108, 107 or 106 conidia mg feces−1. In both studies, 5 days after initial procedures, all formed pupae were transferred to an incubator at 27 ± 0.5 °C until the emergence of the adult flies was complete. The M. anisopliae isolates did not cause the death of H. irritans eggs, but they did promote the death of larvae that hatched from treated eggs, and therefore increased the total mortality. Isolate E9 promoted 100% mortality of treated larvae at a concentration of 108 conidia ml−1. For the B. bassiana isolates, no activity was observed against insect eggs or larvae. Both I. fumosorosea isolates promoted significant mortality (p < 0.05) of eggs at every concentration of conidia. Isolate CG195 of I. farinosa increased the mortality of larvae and pupae that hatched from treated eggs and promoted significant total mortality (p < 0.05) of the insect at every concentration of conidia.  相似文献

试验旨在通过检测商品鸡蛋在不同储存温度和时间蛋壳表面和内容物中真菌的数量和类别,了解真菌污染的动态变化,为改善鸡蛋储存条件并延长保鲜期提供参考。采取冀南某养殖小区不同养鸡场180枚新鲜鸡蛋,分别在25和4 ℃无菌环境下储存42 d,每7 d进行一次蛋壳和蛋液的真菌数量和类别检测。真菌数量检测参照国家标准GB 4789.15-2016;真菌种类鉴定采用形态观察结合rDNA-ITS序列分析法。试验结果显示,鸡蛋内外真菌检出率和数量均随储存时间延长而显著增长;蛋壳比蛋液真菌检出时间早、检出率高,蛋液处理组真菌检出率在21~28 d快速增长;在相同时间点,25 ℃处理组真菌数量(菌落数)显著高于4 ℃处理组,蛋壳处理组显著高于蛋液处理组(P<0.05);在蛋壳和蛋液中共分离鉴定了168个真菌菌株,分属于18个种(属),其中优势属为青霉属(Penicillium sp.),优势种为草酸青霉(Penicillium oxalicum);蛋液与蛋壳处理组的总菌落数和分离株数分别呈显著(P<0.05)和极显著相关(P<0.01)。综上,在采样养殖场环境下,鸡蛋的真菌污染普遍存在;样品真菌检出率和菌落总数均随储存时间延长呈明显上升趋势;冷藏(4 ℃)是防止鸡蛋真菌污染扩展的有效方法;蛋液的真菌污染可能主要来自蛋壳表面的真菌污染;样品鸡蛋污染真菌的多样性较高,其优势种属多为环境常在菌,但污染真菌中也有部分致病或产毒种类。  相似文献

In two small farms in Sweden young calves were found to be naturally infected with Ascaris suum. One of the calves expelled mature worms with the faeces, one had a great number of worms in the ductus choledochus and two others had worms in the intestine. Most of the worms were mature and at the egg-producing stage. The morphology of the eggs and the adult worms indicated that these parasites were A. suum and that Toxocara (syn. Neoascaris) vitulorum could be excluded.  相似文献

Chelonians have been exploited since ancient times for their meat, eggs, fat, and offal, among other things. Among these animals, there is Scorpion mud turtle (Kinosternon scorpioides) which is an omnivorous turtle with semiaquatic habits and is widely consumed in Brazil. The aim of this study was to evaluate the influence of different feeding management systems on the reproductive performance of captive-bred Scorpion mud turtles, including egg quality and chemical composition. Study animals included 15 males and 48 females, divided into three groups. Animals were fed with either a commercial diet or a commercial diet supplemented with one of two different mixes (wet diet). All animals were fed at 1 % BW/day divided on Mondays, Wednesdays, and Fridays. Treatments were as follows: TR?=?100 % fish food (22 % CP); TRM1?=?70 % fish food?+?30 % mix 1 (bovine offal and fish); TRM2?=?70 % fish food?+?30 % mix 2 (bovine offal, fish, and shrimp). Samples were collected during 7 months, and eggs were identified, weighed, measured, and frozen for later analysis. Statistical analyses, including ANOVA, were performed using the program SAEG 9.0. Measurements taken from the nests were compared using the Tukey’s test (P?<?0.05). The different diets were associated with differential egg-laying performance and eggshell thickness. Animals fed with wet diets containing protein of animal origin displayed the best performance. Furthermore, nests with fewer eggs contained eggs of better external quality (e.g., greater length, width, and eggshell thickness). Finally, animals fed with only commercial feed produced eggs with lower saturated fatty acid content.  相似文献
