Interpretation of eosinophilia in body fluids or tissues is often not straightforward. Eosinophil counts vary among clinically healthy individuals, and considerable overlap can occur between normal and affected animals in conditions such as allergic airway disease. Parasite exposure is a confounding factor when counts are increased, and in cases where very high counts and dramatic clinical signs make another disease process obvious, the underlying pathology may be uncertain and treatment difficult. Eosinophils are a component of the immune response in many diseases of the horse, but their specific role is often unknown and likely multifactorial. In helminth infections, eosinophils are assumed to be part of the normal host response to a pathogen, whereas in multisystemic eosinophilic epitheliotropic disease (MEED), the predominance of eosinophils likely represents a wildly dysregulated response, or an abnormal response altogether. This distinction is still not clear for other diseases. Understanding the pathways involved in recruitment, activation or suppression of eosinophils is required for more accurate diagnostics, effective therapeutics, and successful strategies for prevention of eosinophil associated diseases. Eosinophils of the horse: Part II reviews published observations on the eosinophil in clinical diseases of the horse. The behaviour of eosinophils in three common and relatively well-studied conditions is presented first, including gastrointestinal helminth infections, non-infectious respiratory disease, and insect bite hypersensitivity. The less common eosinophil-associated diseases such as eosinophilic disease confined to the intestine (EDCI) and MEED are considered, followed by a brief summary of the eosinophil in phycomycosis and neoplasia. In conclusion, a panoramic view of the equine eosinophil as presented in Parts I and II is placed in the larger context of current eosinophil research, and areas of study are identified that may improve our understanding of eosinophil biology in equine health and clinical disease.  相似文献

绿色荧光蛋白(GFP)基因和新霉素磷酸转移酶(NPTⅡ)基因被广泛应用于植物的遗传转化中。为了研究融合标记基因NPTⅡ::GFP在柑橘遗传转化中的筛选效率,本实验将CaMV35S组成型启动子驱动的nptⅡ∷GFP融合标记基因表达载体pNGM和由CaMV35S启动子分别驱动NPTⅡ、GFP两个基因的表达载体pNPTII-GF经农杆菌介导法转化锦橙上胚轴。GFP绿色荧光检测结果显示,GFP在转NPTⅡ∷GFP融合基因植株中强烈表达,荧光强度与非融合GFP转基因植株的表达水平相当。NPTⅡ∷GFP融合标记基因对锦橙的转化效率为10.8%,与非融合NPTⅡ基因的转化效率(11.0%)没有明显差异。研究结果表明,NPTⅡ∷GFP融合标记基因是一个有效的柑桔遗传转化筛选标记基因。  相似文献

Metaldehyde and methiocarb are two snail baits (molluscicides) which are commonly ingested by dogs and cause rapid onset of severe neurological symptoms. Rapid and appropriate treatment is essential for patient survival. This paper summarizes the literature on treatment of metaldehyde and methiocarb toxicity, based primarily on actual case reports from the past 35 years.
There are important differences between metaldehyde and methiocarb in their mechanisms and treatment. Metaldehyde's mechanism is unknown; methiocarb is a carbamate and parasympathomimetic. Pentobarbitone anesthesia and atropine have been the mainstays of treatment in metal-dehyde and methiocarb toxicity, respectively; their limitations and disadvantages are discussed. Other drugs and aspects of treatment, such as acetylpromazine, diazepam, gastric lavage, and hypetihermia, are also discussed.
Clinical signs are similar in both types of toxicity, but treatment may be complicated by the development of hyperthermia and atropine poisoning which also have similar clinical signs.  相似文献

Interspecies scaling is a useful tool for the prediction of pharmacokinetic parameters from animals to humans, and it is often used for estimating a first-time in human dose. However, it is important to appreciate the mathematical underpinnings of this scaling procedure when using it to predict pharmacokinetic parameter values across animal species. When cautiously applied, allometry can be a tool for estimating clearance in veterinary species for the purpose of dosage selection. It is particularly valuable during the selection of dosages in large zoo animal species, such as elephants, large cats and camels, for which pharmacokinetic data are scant. In Part I, allometric predictions of clearance in large animal species were found to pose substantially greater risks of inaccuracies when compared with that observed for humans. In this report, we examine the factors influencing the accuracy of our clearance estimates from the perspective of the relationship between prediction error and such variables as the distribution of body weight values used in the regression analysis, the influence of a particular observation on the clearance estimate, and the 'goodness of fit' (R(2)) of the regression line. Ultimately, these considerations are used to generate recommendations regarding the data to be included in the allometric prediction of clearance in large animal species.  相似文献

Abstract— Defects in thermal regulatory mechanisms of the new-born dog are described with particular reference to the relative absence of true chemical regulation of body temperature during the first 2–3 days of life. This deficiency is partly compensated by the ability to absorb heat from external sources. I wish to add three important references concerning its first recognition in livestock and humans in New Zealand. The first field cases of leptospirosis in calves due to Leptospira pomona (now L. interrogans serovar pomona) were confirmed in November 1951 on farms in Westland, not in Northland in 1953 as stated in the review. The seminal year for leptospirosis research was 1951 not 1953. Simultaneously, the association with human disease in New Zealand was also recognized and confirmed (Bruere 1952; Kirschner et al 1952). In that year there was a mini-epidemic of cases in Westland involving both calves and humans. It was fortuitous at the time that Kirschner had established methods for the diagnosis of leptospirosis at Otago Medical School. Despite obvious clinical signs, laboratory diagnosis of leptospirosis was not confirmed at Wallaceville (New Zealand's only animal diagnostic station available to veterinarians in 1951) because of logistic impossibilities in both transport and sample preservation. A marked increase in leptospirosis in dogs was observed in 2000, part of an increasing trend observed in previous years in Ontario. The highest frequency of seropositive cases occurred from September to December 2000, with the peak in November. Large breed dogs were particularly affected. Clinical and clinicopathological data for 31 dogs admitted between 1998 and 2000 to the Ontario Veterinary College Veterinary Teaching Hospital were analyzed. Major clinical presenting features were acute onset of anorexia, depression, fever, and vomiting. Ninety percent of dogs, on admission, showed biochemical evidence of injury to several organs, notably combinations in the order of kidney, muscle, pancreas, and liver. Almost all dogs showed increased serum urea and creatinine levels, and the majority had increased total creatine kinase, bilirubin, alkaline phosphatase, and leukocytosis with neutrophilia. One-third were thrombocytopenic. Of dogs with liver-related abnormalities, most had evidence of cholestasis, with or without hepatocellular damage. Based on serologic studies, in the year 2000, the major serovar involved was autumnalis, but bratislava, grippotyphosa, and pomona were also implicated. The microscopic agglutination test often gave a confusing pattern of reactivities to the serovars that were tested. The high reactivity to serovar autumnalis may represent an erroneous or "paradoxical" reaction typical of early leptospiral serology. The year 2000 was the warmest in Ontario in each of the 4 fall months (September-December) of the previous decade, as well as being the third wettest in the fall period in the last decade. The increase in canine leptospirosis, therefore, may, in part, reflect climate change. The number of positive cases declined in 2001 by about one-third of those in 2000, but the number of submissions of sera for diagnosis increased markedly over previous years. Further work is required to isolate and to identify definitively serovars involved in resurgent canine leptospirosis and the common sources for dogs.  相似文献

BACKGROUND: Sodium:potassium (Na:K) ratios are often reported in feline biochemical panels, although the importance of this measurement has not been investigated. OBJECTIVES: The aims of this study were to document the range of feline disease states associated with a decreased Na:K ratio, to determine the prevalence of this biochemical abnormality in a referral hospital population, and to identify any particular disease that was more likely to have a decreased Na:K ratio. METHODS: A group of 49 cats with decreased Na:K ratios was compared with a group of 50 cats with normal Na:K ratios that were randomly selected from the same hospital population. RESULTS: Twelve of the 49 cats (24.5%) had gastrointestinal disease, 10 (20.4%) had urinary disease, 8 (16.3%) had endocrine disease, 8 (16.3%) had cardiorespiratory disease, and 5 (10.0%) had diseases affecting other body systems. Six (12.2%) had artifactually decreased Na:K ratios. No cat was identified with hypoadrenocorticism. Statistical analysis revealed that, although none of these disease states was significantly over- or under-represented in the affected group, a significantly higher proportion of cats with decreased Na:K ratio had body cavity effusions (P = .025). Serum potassium concentrations were significantly higher in the affected group (P < .0001), but there was no significant difference in mean sodium concentration between the 2 groups. CONCLUSIONS: Decreased Na:K ratios frequently occur in cats with diseases other than hypoadrenocorticism, including cats with effusions. These findings should be considered when evaluating cats with this biochemical abnormality.  相似文献

A retrospective study was done of 26 cases of metaldehyde and 30 cases of methiocarb toxicity in dogs. Physical signs and treatment protocols are described for four stages of toxicity: asymptomatic exposure; initial toxicity; convulsions; and post-convulsions.
Diazepam-ketamine combination was used as the primaty anesthetic agent in all cases requiring general anesthesia, and proved effective. Metaldehyde cases required a significantly higher number of sedative and anesthetic doses (p < 0.001). Pentobarbitone was not used. There were no incidents of respiratory or cardiac arrest at the time of anesthesia induction.
Emetics, gastric Iavage, and rectal enemas were productive in removing the molluscicide from the gastrointestinal tract. Ninety percent of the methiocarb patients and 69% of the metaldehyde patients were fit for discharge in 12 hours or less, though there was a wide variation. Both metaldehyde and methiocarb groups had a high survival rate, 100% and 93%, respectively.  相似文献

The nature and predictability of protein digestion in steers were examined in a series of experiments. Bypass and intestinal digestion of supplemental proteins were measured with Angus steers (203 to 254 kg) fitted with dual reentrant intestinal cannulas. Daily feed intakes were 1.8 to 1.9% of body weight. Two trials were conducted with soybean meal, cottonseed meal, a hardened casein, meat meal and two different sources of dehydrated alfalfa meal fed with an 80% concentrate diet. In the third trial, digestion of soybean meal and cottonseed meal were measured with a 60% prairie hay diet. Estimates of ruminal escape or bypass were 43, 50, 36, 76, 57, 62, 24 and 43%, respectively. Results suggest that roughage level alters bypass. A standard reference system for predicting bypass based on measurements of N solubility and in situ disappearance rates was compared with measured bypass. The correlation between observed bypass to the small intestine and bypass predicted by this system was high (r = .91, P less than .01). Combining pepsin solubility, an indicator of true intestinal digestibility, with bypass estimated by the standard reference system, allowed us to predict N disappearance from the small intestine. Predicted and measured N disappearance of fed N from the small intestine were very close.  相似文献
