A nation-wide Salmonella enterica surveillance and control programme was initiated in Danish finishing herds over the first quarter of 1995. In Denmark, all swine for slaughter are identifiable by a unique herd code. For each herd code, and depending on the herd's annual kill, random samples ranging from four to more than 60 swine are obtained quarterly at the abattoir. A meat sample from each pig is frozen, and meat juice (harvested after thawing) is examined for specific antibodies against S. enterica using an indirect enzyme-linked immunosorbent assay (ELISA). The ELISA combines several S. enterica O-antigens, and allows detection of antibody response after a variety of different S. enterica serovar infections. Results are transferred to a central database, which each month (based on meat-juice tests obtained in the previous 13 weeks) assigns all herds into three S. enterica infection levels: Level 1, in which the S. enterica prevalence is deemed low and acceptable; Level 2, where there is a moderate prevalence of S. enterica seroreacl.ors (from > 50% in the smallest to > 10% in the largest herds); Level 3, in which S. enterica seroreactor prevalence is clearly unsatisfactory ( > 50% for most herd sizes). Irrespective of Salmonella level, all herds receive a monthly update on the current results of the S. enterica test results. If a herd is categorized in Level 2 or 3, it must receive an advisory visit by a practising veterinarian and a local swine extension specialist, and certain management hygiene precautions must be taken. If a herd is categorized in Level 3, the finishers from the herd must additionally be slaughtered under special hygiene precautions. This is supervised by the veterinary authorities.

During 1995, 604000 samples were tested for S. enterica, corresponding to 3.0% of the total kill. In December 1995, 15522 herds (representing > 90% of the national production) were categorized into one of the three levels: 14551 herds (93.7%) in Level 1; 610 herds (3.9%) in Level 2; 361 herds (2.3%) in Level 3. The proportion of serologically positive meat-juice samples collected during 1995 ranged from a mean of 2.9% in smaller herds (101–200 swine slaughtered per year) to 6.1% in relatively large herds (more than 5000 swine slaughtered per year).  相似文献

An experimental challenge model was developed to demonstrate Lawsonia intracellularis colonization and reproduction of proliferative enteropathy (PE) in naïve weaner pigs. Groups of pigs were orally dosed with between 1010 and 105 L. intracellularis extracted from haemorrhagic PE affected mucosa. Pigs were monitored for clinical signs and intestinal lesions of PE and evidence of bacterial colonization by serology and faecal polymerase chain reaction (PCR). One group of challenged pigs were necropsied after 21 days to confirm the reproduction of PE. L. intracellularis colonization and seroconversion was delayed in pigs dosed with lower numbers of L. intracellularis. When faecal shedding of L. intracellularis ceased to be detected in all of the challenged pigs, they were re-dosed orally with approximately 1010 L. intracellularis and monitored for evidence of re-colonization and clinical disease. This study demonstrated that pigs previously challenged with L. intracellularis were protected from re-colonization and clinical disease on subsequent exposure 10 weeks later, regardless of the initial dose of L. intracellularis.  相似文献

The national bovine herpesvirus 1 (BHV-1) seroprevalence (apparent prevalence) in the Belgian cattle population was determined by a serological survey that was conducted from December 1997 to March 1998. In a random sample of herds (N=556), all cattle (N=28 478) were tested for the presence of antibodies to glycoprotein B of BHV-1. No differentiation could be made between vaccinated and infected animals, because the exclusive use of marker vaccines was imposed by law only in 1997 by the Belgian Veterinary Authorities. Twenty-one percent of the farmers vaccinated continuously against BHV-1.

In the unvaccinated group, the overall herd, individual-animal and median within-herd seroprevalences were estimated to be 67% (95% confidence interval (CI)=62–72), 35.9% (95% CI=35.0–36.8) and 33% (quartiles=14–62), respectively.

Assuming a test sensitivity and specificity of 99 and 99.7%, respectively, the true herd, individual-animal and median within-herd prevalence for the unvaccinated group of herds were estimated to be 65, 36 and 34%, respectively. The true herd prevalence for dairy, mixed and beef herds were respectively, 84, 89 and 53%; the true individual-animal prevalence for those types of herds were, respectively, 35, 43 and 31%; whereas, the true median within-herd prevalences were 36, 29 and 38%.  相似文献

Midwest U.S. herds (n = 63) were studied to identify risk factors for harboring Salmonella enterica among slaughter-weight pigs. Samples collected on farms (feces) and at slaughter (distal colonic content, cecal content and ileocolic lymph nodes) were cultured using conventional means. Approximately 15 pigs were studied per herd, for a total of 3754 samples. The proportion of pigs positive in one or more samples was calculated for each herd. Herd characteristics were described by a combination of interview and written survey. Logistic regression was used to detect relationships between the detection of Salmonella and potential herd-level risk factors. The mean individual pig prevalence was 5% for feces, 4% for distal colonic content, 15% for ileocolic lymph nodes, and 17% for cecal contents. One or more Salmonella isolates were detected in at least one sample type in every herd. The five most common serovars were S. Agona, S. Derby, S. Schwarzengrund, S. Typhimurium and S. Senftenberg, with 25 additional serovars detected. Salmonella prevalence estimates were positively correlated among all samples except distal colonic content and ileocolic lymph nodes. Pigs with culture positive fecal samples were at increased odds of being detected positive for each of the slaughter-collected samples examined, namely distal colonic content (OR = 30.5), ileocolic lymph nodes (OR = 12.9) and cecal content (OR = 23.2). Herds with positive fecal sample(s) had increased odds of having positive cecal content (OR > 1.5), distal colonic content (OR = 15.3) and ileocolic lymph nodes (OR = 12.7). Pigs from herds with at least some bowl drinkers had eight-fold higher odds of testing Salmonella positive than did pigs from herds with only nipple drinkers. Pigs from herds with only dry feeders had five-fold higher odds of testing Salmonella positive when compared with pigs from herds with combinations of wet/dry style feeders. Interventions at these two points should be considered when designing growing pig facilities to reduce Salmonella shedding.  相似文献

Two type/reference strains of Brachyspira (B.) hyodysenteriae, 14 Belgian and German indole negative, and 14 Belgian, German and Swedish indole positive field isolates of strongly β-haemolytic intestinal spirochaetes were compared by pulsed-field gel electrophoresis (PFGE) patterns, biochemical reaction patterns, 16S rDNA sequences and MIC determinations of six antibacterial substances. Three tests for indole production, including a spot indole test, were compared with congruent results. All field isolates were classified as B. hyodysenteriae due to a high genetic and phenotypic similarity with the type strains. The Belgian and German indole negative isolates had identical and unique PFGE patterns for the tested restriction enzymes MluI and SalI, as well as identical 16S rDNA sequences, and they could not be differentiated by any of the methods used. Seven unique PFGE patterns were achieved from the 14 indole positive field isolates. The patterns were identical and unique for epidemiologically related isolates. Type/reference strains and isolates without known relation to other tested isolates showed unique banding patterns. The MICs of tylosin, tiamulin, erythromycin, clindamycin, carbadox and virginiamycin were determined in broth for all isolates. In contrast to Belgian and German isolates, the majority of the Swedish field isolates were susceptible to tylosin, erythromycin and clindamycin. Probable pathways of infection for some of the Swedish isolates were determined. The PFGE patterns of epidemic clones of B. hyodysenteriae remained stable for a period of up to 8 years. In vivo development of resistance to macrolide and lincosamide antibiotics due to use of tylosin was clearly indicated for two epidemic clones.  相似文献

Pigs in four groups were fed fine non-pelleted (F-NP), fine pelleted (F-P), coarse non-pelleted (C-NP) or coarse pelleted (C-P) diets. DNA fingerprints, T-RFLP, revealed a strong dietary effect on bacterial community structure in the stomachs. Pigs fed the C-NP diet had the highest microbial diversity in stomach and the T-RFLP fingerprints suggested that bacteria tentatively identified as Lactobacillus delbrueckii, L. mucosae, L. reuteri, L. amylovorus/L. sobrius, Mitsuokella multiacida and Megasphera elsdenii were specifically stimulated. Lactobacilli may be responsible for the previously reported increase in stomach lactate levels of pigs fed the C-NP diet [Mikkelsen, L.L., Naughton, P.J., Hedemann, M.S., Jensen, B.B., 2004. Effects of physical properties of feed on microbial ecology and survival of Salmonella enterica serovar Typhimurium in the pig gastrointestinal tract. Appl. Environ. Microbiol. 70, 3485–3492.]. Moreover, earlier observations of butyrate and propionate accumulation in the stomach of pigs fed C-NP diets could be due to stimulation of M. elsdenii and S. ruminantium according to the T-RFLP fingerprints.  相似文献

The use of the antimicrobial growth promoters (AGPs) carbadox and olaquindox has been banned in the European Union (EU) since September 1999. We studied the effects of the withdrawal on the health of weaned piglets on two types of piglet-producing farms (farrowing herds and farrow-to-finish herds) from the different regions of Finland. Farms with no major problems with post-weaning diarrhoea were selected for the study to better evaluate the effect of AGPs alone. Data on production, medication and incidence of diarrhoea were collected from 73 farms during 1 year after the withdrawal. On 29 of these farms, the data collection began 4 months before the withdrawal.

The health management of the pigs is considered good in Finland, and special attention has been paid to improve the husbandry practices and management of the farms. Eighty-two percent of the farms in the study were free of both Mycoplasma hyopneumoniae and Sarcoptes scabiei infection. Brachyspira hyodysenteriae infection was not detected in any of the farms. The median number of sows in the herds was 56.0 (IQR = 43.0; 72.5) in 2000.

The level of antimicrobial use in each herd was classified as low, moderate and high when the percentage of weaned pigs treated for diarrhoea during a 4-month period was 0–5%, 6–19% and ≥20%, respectively. Only on four herds (14%), there was an increase in the level of antibiotic use after the AGP withdrawal, when seasonally corresponding 4-month periods were compared. Fourty-one percent of these 29 farms were categorized as low users of antimicrobials, 38% as moderate users and 21% as high users.

The level of antimicrobial use for treatment of diarrhoea after weaning (and the incidence of diarrhoea in weaned piglets) did not increase significantly after the withdrawal of AGPs from weaner feeds according to farmers’ evaluations. In this study, the Escherichia coli infection was the most-common cause of diarrhoea in weaned pigs. The age at weaning did not change after the withdrawal of AGPs.  相似文献

The aim of this study was to assess the probability of detecting Salmonella from pen faecal samples in seropositive classified finishing pig herds. The study involved 77 herds from Denmark (20), The Netherlands (20), Greece (17) and Germany (20). The serological herd status was determined by the blood-sampling of 50 finishing pigs. Bacteriological sampling was performed by 20 pen faecal samples per herd. Over-all, 47% of the blood samples had an OD% larger than 10 and 23% larger than 40. Salmonella was isolated from 135 (9.3%) pen faecal samples in 32 herds (42%). Twenty-eight of these herds (87.5%) had a within-herd seroprevalence larger than 50% at sample cut-off OD% > 10. In our study, there was an increasing probability of recovering Salmonella with increasing within-herd seroprevalence. However, this was only a moderate correlation. A correlation coefficient of 0.62 was found between the proportion of culture positive- and seropositive samples in a herd at cut-off OD%> 10 and of 0.58 at cut-off OD% > 40. Serology is a measure of historical exposure, which may or may not correlate closely to the microbiological burden at the time of sampling. Due to the low sensitivity of culture methods, apparent 'false-positive' serological results may well represent real infections not detected by bacteriological testing. For screening purposes, serological testing provides an indication of exposure to Salmonella, which forms the basis for targeted sampling, intervention and logistic slaughter procedures.  相似文献

Dutch dairy herds closed for at least 3 years with no history of paratuberculosis were recruited for a study on herd-certification. One hundred dairy herds were tested for Mycobacterium paratuberculosis at 6-month intervals by pooled faecal culture (five individual animal samples per pool) with solid media. Ninety of the herds completed 9 herd tests and 10 herds dropped out of the study for reasons other than a paratuberculosis diagnosis. Of the 90 herds completing the full study, 61% eventually were found to be M. paratuberculosis-infected. The number of infected herds detected decreased with each round of testing. Assuming that all infected herds had been detected by the ninth herd test, the observed percentage of herds that were truly noninfected (P-free) after each round of testing was calculated. The observed P-free was compared to the predicted P-free based on a previously reported herd-certification model. The P-free predicted by the model was significantly different from the observed P-free. When a single assumption in the model was changed and a diagnostic sensitivity of 40–50% was selected, the predicted P-free closely approximated the observed P-free for the 90 Dutch dairy herds studied. The critical assumption that was changed for Version 2.0 of the model was within-herd infection prevalence for infected but test-negative herds after each round of serial testing. Model Version 1.0 had assumed a 50% decrease in within-herd prevalence but Version 2.0 assumed a stable within-herd prevalence. Culture of pooled faecal samples provides a high-sensitive, high-specific, low-cost test for herd-certification programs.  相似文献

Our objective was to generate hypotheses about associations between management, climate, and the presence of Escherichia coli O157 in feedlot–cattle water tanks and in feedlot–cattle feed. Water samples from 710 tanks on 73 feedlots, and feed-samples from a subset of 504 pens on 54 feedlots, in four US states were tested for E. coli O157. Management and climate factors were ascertained by survey and observation. Escherichia coli O157 were isolated from 13% of the water tanks and at least one water tank was positive on 60% of the feedlots. The factors significantly associated with E. coli O157 in water were greater percentage of cattle shedding E. coli O157 in faeces within the same pen, higher concentration of total E. coli in the water, lack of the clarity of the water, the use of fly traps, the reported frequency of rodent sightings in the pen or alley area, and the weather at the time of sampling. Escherichia coli O157 were isolated from 14.9% of the feed samples obtained from the feedbunks. Factors positively associated with E. coli O157 in feed were higher heat index at the time of sampling, the presence of cottonseed meal in the ration, and the feedlot location (state). Coliform counts in feed, presence of E. coli O157 in water tanks and faecal prevalence of E. coli O157 were not associated with the presence of E. coli O157 in feed.  相似文献

Antimicrobial resistance (AR) is an increasing phenomenon but its quantitative estimation remains controversial. The classical resistance percentage approach is not well suited to detect either emergence or low levels resistance. One option is to shift the focus from strains to hosts. This approach is applied to test for phenotypic diversity associated with diminished susceptibility to expanded-spectrum cephalosporins (DSESC) in faecal Escherichia coli from healthy food animals in Spain.

We performed E. coli enumeration in faecal samples of broilers (82 pooled samples) and pigs (80 pooled samples) at the slaughterhouse level, using Coli-ID plates alone and supplemented with cefotaxime at two levels (1 and 8 μg/ml). Antimicrobial susceptibility of isolates was tested by the agar diffusion method. Clustering was carried out using these numerical values and Ward and UPGMA methods.

When using plates supplemented with 1 μg/ml of cefotaxime for DSESC E. coli detection, 93% (76/82) of broiler pooled samples and 36% (29/80) pig pooled samples tested positive. When using 8 μg/ml of cefotaxime, 67% (55/82) of broilers and 13% (10/80) of pigs were positive. Nevertheless, the relative abundance of this phenotype was low in both animal species (range 0–4.3%). Irrespective of the clustering method (Ward or UPGMA), a noticeable phenotypic diversity was detected, especially from the plates containing 1 μg/ml of cefotaxime.

We concluded that: (a) E. coli with phenotype DSESC are common in broilers and pigs but are less frequent in pigs, and (b) the host approach is the most appropriate method for antimicrobial resistance assessment when null or very low levels of antimicrobial resistant bacteria are expected.  相似文献

We determined the prevalence of tuberculosis and brucellosis reactors in the dairy herds in the Mbarara district of Uganda in 2002. This is one of the most important dairy-production areas of the country and includes both pastoral and agro-pastoral zones. A total of 340 (of 11,995) randomly selected herds were tested for tuberculosis, using the intradermal tuberculosis-skin test and 315 (of 10,562) herds tested for brucellosis using the serum Rose Bengal test.

The herd prevalence for tuberculosis reactors was 74.1% (95% confidence intervals 69, 78), the individual-animal prevalence was of 6.0% (5.6, 6.5) and within-herd range was 1–50% (up to 100% if suspicious reactors were included).

The herd prevalence for brucellosis was 55.6% (50, 61.2) individual-animal prevalence 15.8% (14.8, 16.7) and within-herd range 1–90%.

The reactor prevalence increased with the age of the animals for both tuberculosis and brucellosis.

Tuberculosis reactor prevalences were higher in animals from the agro-pastoral zone. However, the individual-animal and herd prevalences of brucellosis seroprevalences were higher in the pastoral zone.  相似文献

Our objective was to generate hypotheses for potential on-farm control strategies for Escherichia coli O157 by identifying associations between management practices and climate, and the presence of E. coli O157 in feedlot cattle. Faeces were obtained from 10,622 cattle in 711 pens on 73 feedlots between May and August 2001. Management and climate information was obtained by questionnaire and observation at the time of sampling. The prevalence of E. coli O157 was 10.2% at the sample level, 52.0% at the pen-level, and 95.9% at the feedlot-level. The factors associated with the presence of E. coli O157 in cattle faeces were the frequency of observing cats in the pens or alleys (most common when observed daily), the presence of E. coli O157 in the water tanks (positive association), the historical use of injectable mass medication (positive association), the use of antibiotics in the ration or water (negative association), the wetness of the pen, number of cattle in the pen (negative association), wind velocity (positive association), and height of the feed bunk (positive association).  相似文献

In control programs for Mycobacterium avium subsp. paratuberculosis (Map), the infection status of the cows in a herd is often obtained by testing (a sample of) the herd with an ELISA that may lack some sensitivity and specificity but that is fast and inexpensive. In New York State (NYS), an unabsorbed kinetics ELISA (KELA) has been used extensively for Map control. The objective of this study was to determine the relative sensitivity and specificity of the KELA for detection of fecal shedding of Map for the NYS dairy cow population, taking into account possible confounders such as different antigen batches and Map prevalence in a herd.

The data for the study consisted of all serum samples from NYS dairy cows with concurrent fecal culture results submitted to the NY Animal Health Diagnostic Laboratory (NYAHDL) between 1991 and 1996 (n = 10,562). The data represented cows with different levels of fecal shedding from herds with different within-herd Map prevalence, including herds that were whole herd fecal culture negative on repeated testing.

The cutoff values were based on the predictive value for fecal shedding obtained with a multiple logistic regression model that included variables for the three antigen batches and the Map prevalence in the herd. The KELA could not distinguish between non-shedders and low shedders (≤30 total colony forming units (TCFU)) and thus the predictive value of the KELA to detect moderate to heavy fecal shedders (>30 TCFU) was modeled. The three cutoff values of 65, 135 and 170 were based on low (<0.2), moderate (<0.80) and high (>0.95) probabilities for moderate to heavy fecal shedding. The sensitivity and specificity values relative to culture were 67% and 95.2%, 31% and 99.7%, and 11% and 99.9% for the three cutoff values, respectively. Cutoff values for the KELA decreased for herds with increasing within-herd Map prevalence. For the best positive predictive value of a KELA for moderate to heavy fecal shedding, the cutoff values should be determined based on the apparent within-herd prevalence in a herd.  相似文献

为深入了解畜禽粪污中金黄色葡萄球菌和大肠埃希氏菌的流行及耐药状况,采用建立的畜禽粪便中金黄色葡萄球菌和大肠埃希氏菌的环介导等温扩增技术(LAMP)检测方法对山东省内牛粪污、猪粪、鸡粪、鸭粪中的金黄色葡萄球菌和大肠埃希氏菌进行检测,进而对检测阳性样品进行分离,并对药敏试验中多重耐药性菌株进行耐药基因预测。结果表明,LAMP方法检测畜禽粪污中金黄色葡萄球菌和大肠埃希氏菌与国家标准方法检测结果符合率为100%,重复性好,特异性高。从80份畜禽粪污样品中分离到的3株金黄色葡萄球菌均对青霉素耐药;34株大肠埃希氏菌对氟苯尼考、利福平的耐药比例高于50%,高于25%以上的还有氨苄西林、四环素、多西环素、磺胺异噁唑、头孢噻吩和头孢噻呋;耐药基因预测结果显示,鸡粪、鸭粪、牛粪和猪粪中的4株大肠埃希氏菌分别携带10、8、20和15种耐药基因;鸡粪中的1株金黄色葡萄球菌携带11种耐药基因。说明山东地区畜禽粪污中金黄色葡萄球菌和大肠埃希氏菌耐药状况严峻,菌株普遍多重耐药,且携带多种耐药基因。  相似文献

A total of 796 sows and gilts from 30 Danish sow herds were examined three times at intervals of 6 weeks for serum antibodies to Leptospira bratislava by the microscopic agglutination technique (MAT) test. The prevalence of seroreactors with positive titer values, 1:100, at the three successive tests were 2.7%, 2.5% and 2.9%; 4.5% of the animals were positive in at least one of the three tests, and 2.2% showed a greater than two-fold rise in titer between two consecutive samplings. Of the 30 herds, 21 (70%) had ever-positive within-herd prevalences in sows and gilts of 4–13%. The risk of a herd having one or more positive sow was positively associated with a herd size of > 141 sows, and distinct regional differences in the prevalence of positive herds were observed. The reproductive performance of the 21 herds with seroreactions was poorer than the performance of the nine herds without positive reactions concerning the variables: ‘days from weaning to last service’ (2.7 days more, P = 0.07), ‘percentage of sows returning to heat’ (4.0 percentage units more, P = 0.03), ‘services per farrowing’ (0.04 more, P = 0.04), ‘farrowing percentage’ (4.3 percentage units lower, P = 0.06), and ‘stillborn pigs per farrowing’ (0.16 more, P = 0.02). No association between the MAT serological status of the herd and the incidence of medical treatments of sows and gilts could be found. A high prevalence and low cumulative incidence of seroreactors was demonstrated in first-parity gilts, followed by a low prevalence and cumulative incidence from parity 2 to 3, and a high prevalence and cumulative incidence at the fifth parity.  相似文献

Two studies were carried out: a preliminary study based upon 220 flocks visited twice to collect data on sheep-flock management and losses, and a second intensive 2 year (1984–1986) study carried out on 16 of the 220 flocks in one country.

Mean annual lamb mortalities were 17.6% (1984–1985) and 31.3% (1985–1986), and varied significantly according to nutrition. Pre-parturient deaths represented 27.3% (1984–1985) and 18.9% (1985–1986) of total deaths. The percentage of parturient deaths was very low (0.4%) for both years. Of the lambs necropsied, starvation/exposure complex contributed most to lamb mortality. Enteritis and septicemia represented the second most important cause. Of the enteritis cases, 18.5–63.2% of the samples analyzed were positive for Escherichia coli: 38.5% of the septicemia cases were positive for E. coli in 1985–1986 but none were positive in 1984–1985. E. coli K99 isolates were rare. Only one Salmonella isolate was made in 1985–1986. There were 16 cases (12.2%) of pneumonia in 1984–1985 and two cases only (1.5%) in 1985–1986. Of the cases examined, 18.8% were positive for Pasturella hemolytica and 25% were positive for Pasturella multocida in 1984–1985.

Its appears that lamb mortality can be reduced by improvement of nutrition and health care in many of these flocks. Further work should be done to provide information on the causes of lamb pneumonia as well as the potential for vaccination against lamb diarrhea (i.e. E. coli in these flocks.  相似文献

Prior to establishing a control and prevention program for Johne's disease in cattle in Galicia (northwest Spain), a survey was conducted to estimate the prevalence of the disease. For this survey, 61,069 animals of at least 1-year of age from 2735 randomly selected herds were bled and samples analyzed with a commercial ELISA. The estimated true individual-level prevalences – assuming the manufacturer's reported test sensitivity of 48.5% and specificity of 98.9% – were 3.02% in dairy cattle, 1.03% in beef cattle and 2.83% in animals from farms with both dairy and beef cattle. True herd prevalences (with herds declared positive if one or more animals tested positive) were 10.69% for dairy herds, 0% for beef herds and 2.71% for mixed herds. When herds were declared positive if at least two animals tested positive, true herd prevalences were 14.75% for dairy herds, 1.47% for beef herds and 12.01% for mixed herds. Assuming a higher specificity of 99.4%, true individual-level prevalences increased to 4.03% in dairy herds, 2.07% in beef herds and 3.84% in mixed herds. Herd prevalences were 27.77%/18.79%, 2.78%/2.40% and 5.70%/12.24% (using the one/two-animal cut-offs) in dairy, beef and mixed herds, respectively. In conclusion, these results seem to indicate that a small percentage of cows and a rather high percentage of dairy herds in this region are MAP-seropositive.  相似文献
