Identification of ergosterol in vesicular-arbuscular mycorrhizae   总被引:1,自引:0,他引:1  
Summary Ergosta-5,7,22-tri-3-enol (ergosterol) was identified by gas chromatography-mass spectrometry in roots of berseem (Trifolium alexandrinum L., cv. Landsorte) and sweet corn (Zea mays L., cv. Honeycomb-F1) infected with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus intraradices. The fungal-derived compound ergosterol was determined quantitatively in root extracts using reverse-phase high performance liquid chromatography. The concentrations of ergosterol in VAM-infected roots reached 72 g-1 dry material in berseem and 52 g-1 in sweet corn after 80 days of growth, whereas concentrations in non-infected roots remained below 8 g-1 dry weight. Additionally, phytosterols such as -sitosterol, campesterol, and stigmasterol were detected in both infected and non-infected roots. Ergosterol, as a characteristic fungal substance, is proposed as an indicator of fungal biomass in the early stages of VAM infection.  相似文献

Summary Increasing the sulfate concentration and concomitant increases in the organic S concentration failed to exert any effect on organic S mobilization in samples collected from all depths within the mineral soil profile, from 15 sites differing in soil type, vegetation, and geographic location. Mobilization capacities at saturating concentrations of sulfate for organic S formation generally tended to increase with increasing depth. The potentials for the accumulation of organic S with various sulfate inputs exhibited saturation kinetics similar to those observed for organic S formation; values for the former parameter ranged from 3×10-3 to 12.6 mol S g–1 dry weight 24 h-1 for the uppermost (A, E) soil horizons, 3 nmol to 10 mol S g-1 dry weight 24 h–1 for intermediate (primarily AB) soil horizons, and from 3 nmol to 13.4 mol S g-1 dry weight 24 h–1 for the lowermost (B, C) soil horizons. Irrespective of depth, the Fullerton, Tarklin, and Loblolly sites in Tennessee and the Florida site showed the least net accumulation of organic S at saturation (<0.2 mol S g-1 dry weight 24 h–1 for all horizons examined), while the Duke Forest (North Carolina), Douglas Fir (Washington), Whiteface (New York) and the Howland (Maine) sites had the highest potential net accumulation of organic S at saturation (>1.0 mol S g-1 dry weight 24 h-1 for most horizons examined).  相似文献

Summary Plants grown from seed with high (1.5–7.3 g Mo seed-1) and low (0.07–1.4 g Mo seed-1) Mo contents were grown in the presence and absence of Mo in growth media (perlite) or in a flowing-solution culture, in a controlled environment. Neither the high (1.5 g Mo seed-1) nor the low (0.1 g Mo seed-1) Mo content in seed from a small-seeded genotype (BAT 1297) was able to prevent Mo deficiency (reduced shoot, root and nodule dry weight, N2 fixation and seed production) in growth media without an external supply of Mo, whereas both the high (7.3 g Mo seed-1) and the low (0.07 g Mo seed-1) contents in seed were able to prevent Mo deficiency in a large-seeded genotype (Canadian Wonder). Responses to Mo treatment by the Two genotypes were inconsistent between the growth media and solution culture experiments. Seed with a large Mo content (3.5 g Mo seed-1) from the Canadian Wonder genotype was unable to prevent Mo deficiency (reduced shoot and nodule dry weight and N2-fixation) in a solution culture without an external source of Mo, whereas both the large (1.7 g Mo seed-1) and the small (0.13 g Mo seed-1) contents in seed prevented a deficiency in BAT 1297. Growing plants from seed with a small Mo content, without additional Mo, reduced the seed Mo content by 83–85% and seed production by up to 38% in both genotypes. Changes in seed size and increases in shoot, root and nodule dry weight occurred, but varied with the genotype and growth conditions. These effects were also observed in some cases where plants were grown with additional Mo, demonstrating that the amount of Mo in the seed sown can influence plant nutrition irrespective of the external Mo supply. Nodule dry weight, total N content of shoots and seed production were improved by using seed with a small Mo content (1.64–3.57 g Mo seed-1) on acid tropical soils in Northern Zambia. Plants of both the large- and small-seeded genotypes grown from seed with a small Mo content (<1.41 g Mo seed-1) had a smaller nodule weight, accumulated less N and produced less seed. The viability of seed with a small Mo content was lower (germination up to 50% less) than that of seed with a large Mo content.  相似文献

Summary Concentrations of phenamiphos ranging from 0 to 40 rg/g soil were established in a typical Oxisol (Tropeptic Eutrustox), inoculated or uninoculated with Glomus aggregatum. The effect of the nematicide on the development of vesicular-arbuscular mycorrhizal (VAM) symbiosis was evaluated in the greenhouse using Leucaena leucocephala as an indicator host plant. Treatment of soil with phenamiphos did not have a significant influence on the development of mycorrhizal activity measured in terms of subleaflet phosphorus concentrations. Similarly, the nematicide did not have an adverse effect on the level of mycorrhizal colonization or on the P content of shoots, as determined at the time of harvest. However, shoot dry weight was adversely influenced by phenamiphos when the chemical was applied to the uninoculated soil at 20 g/g soil or higher, and when it was applied to the inoculated soil at 40 g/g soil. It is concluded that phenamiphos is not likely to influence the growth of Leucaena or its symbiotic association with VAM fungi if the concentrations applied do not exceed levels known to suppress nematodes.Hawaii Institute of Tropical Agriculture and Human Resources Journal series No. 3146  相似文献

Summary From dried leaves of two mutants ofLycopersicon esculentum Mill., namelyexilis (mutant of the variety Condine Red) andprunoidea (Lukullus), t-tomatine has been isolated in an amount of 0,40% and 0,81%, respectively. The identification of the xylose-free alkaloid glycoside was carried out by paper chromatography and investigation of the products formed by acid hydrolysis. Leaves of the non-mutated controls contained 0,64% and 0,82% tomatine, respectively.

Summary The effect of increasing oxygen concentrations (0, 5, 10 and 20 Vol% O2) on total denitrification and N20 release was studied in model experiments using a neutral pH loamy soil relatively rich in easily decomposable organic matter and supplied with nitrate (300 g nitrate N/g dry soil). The sterilized soil was inoculated with three different denitrifying bacteria (Bacillus licheniformis,Aeromonas denitrificans andAzospirillum lipoferum) and incubated (80% WHC, 30°C). The gas volume was analysed for O2, CO2, N2O, NO and N2 by gas chromatography and the soil investigated for changes in ammonium, nitrite, nitrate, pH, total N and C as well as water-extractable C. WithB. licheniformis andAeromonas denitrificans total denitrification increased remarkably with increasing pO2 as the result of intensified mineralization.Azospirillum lipoferum, however, showed the highest activity at 5 vol% O2. WithB. licheniformis N2O was released only in anaerobic conditions and at 5 Vol% O2 (maximum) or 10 Vol% 02, but not at 20 Vol%, whereasAeromonas denitrificans produced N2O only in the presence of He gas (maximum) or at 5 Vol% O2. In contrast to these bacteria, N2O production withAzospirillum lipoferum was restricted to 10 Vol% O2 (maximum) and to 20 Vol% 02, with some traces at 5 vol% O2. With a certain set of conditions, total denitrification and N2O formation seem to be governed by the mineralization rate of the organisms in question. The increased demand for electron acceptors by a high turnover rate rather than the presence of anaerobic conditions seems to have determined the rate of denitrification.  相似文献

Summary The efficiency of N use in flooded rice is usually low, chiefly due to gaseous losses. Emission of CH4, a gas implicated in global warming, can also be substantial in flooded rice. In a greenhouse study, the nitrification inhibitor encapsulated calcium carbide (a slow-release source of acetylene) was added with 75, 150, and 225 mg of 75 atom % 15N urea-N to flooded pots containing 18-day-old rice (Oryza sativa L.) plants. Urea treatments without calcium carbide were included as controls. After the application of encapsulated calcium carbide, 3.6 g N2, 12.4 g N2O-N, and 3.6 mg CH4 were emitted per pot in 30 days. Without calcium carbide, 3.0 mg N2, 22.8 g N2O-N, and 39.0 mg CH4 per pot were emitted during the same period. The rate of N added had a positive effect on N2 and N2O emissions, but the effect on CH4 emissions varied with time. Carbon dioxide emissions were lower with encapsulated calcium carbide than without. The use of encapsulated calcium carbide appears effective in eliminating N2 losses, and in minimizing emissions of the greenhouse gases N2O and CH4 in flooded rice.  相似文献

Growth rate change in earthworms is considered to be a suitable endpoint when determining sublethal effects. In this study we evaluated growth and maturation in the vermicomposting earthworm speciesEudrilus eugeniae as marker of sublethal toxicity of copper and zinc. We also compared routes of uptake. Apart from exposing worms experimentally for 73 days to contaminated food, a series of contact filter paper tests was also performed to determine LD50 for copper and zinc. Both copper and zinc at sublethal concentrations affected growth and maturation in worms exposed to contaminated food. These worms had a copper content of 34.5 g g–1 after 73 days and a zinc content of 184.9 g g–1, showing a differential uptake. Copper was more toxic than zinc. Also in the contact test worms did take up more zinc than copper and the LD50 (48 h) for copper was 0.011 mg cm–2 and for zinc 0.066 mg cm–2, which translated to body burdens of 6 g g–1 for copper and 131 g g–1 for zinc. Indications were that a regulatory mechanism existed for both metals. Both metals were taken up through the body wall at a relatively fast rate. This study indicated that the skin was the major route of metal uptake. This study also showed a poor relation between the two types of tests for purposes of evaluating lethality of zinc and copper.  相似文献

Summary Strains of Bradyrhizobium influenced root colonization by a species of vesicular-arbuscular mycorrhizae (VAM), and species of VAM influenced root nodulation by strains of Bradyrhizobium in pot experiments. In a field experiment, the effects of VAM on competition amongst inoculated bradyrhizobia were less evident, but inoculation with Bradyrhizobium strains increased root colonization by VAM. Certain VAM/Bradyrhizobium inoculum strain combinations produced higher nodule numbers. Plants grown without Bradyrhizobium and VAM, but supplied with ammonium nitrate (300 g ml–1) and potassium phosphate (16 g ml–1), produced higher dry-matter yields than those inoculated with both symbionts in the pot experiment. The cell-associated adsorption of thorium or uranium from the solution containing each metal only at pH 3.50 using various microorganisms was examined. Among the species tested, high thorium adsorption abilities were exhibited by strains of the gram-positive bacteria Arthrobacter nicotianae IAM12342, Bacillus megaterium IAM1166, B. Subtilis IAM1026, Micrococcus luteus IAM1056, Rhodococcus erythropolis IAM1399, and Streptomyces levoris HUT6156. And high uranium adsorption abilities were found in some gram-positive bacterial strains S. albus HUT6047, S. levoris HUT6156, and A. nicotianae IAM12342. Among these highly efficient thorium and uranium adsorbing microorganisms, S. levoris, which could adsorb the largest amount of uranium from the aqueous solution at pH 3.50, could adsorb about 383 mol thorium and 390 mol uranium per gram dry weight of microbial cells from the solution containing thorium or uranium at pH 3.50. The amount and time course of thorium adsorbed were almost unaffected by co-existing uranium; however, the adsorption of thorium was faster when carried out after the adsorption of uranium. Thorium adsorption also became faster when uranium was added after thorium adsorption. The effect of pH on thorium adsorption was also discussed.  相似文献

Aegilops neglecta Req. ex Bertol. is a forage goatgrass that has tetraploid and hexaploid forms. No morphological trait is known to distinguish between the two forms. Accessions of Ae. neglecta representing diverse germplasm were characterized for stomatal frequency and size on the adaxial surface of leaves. Stomatal frequency among tillers within a plant and among leaves on a tiller was measured. Significant variation was found in stomatal frequency between the ploidy levels for basal leaves 2 and 3 and the penultimate and flag leaves. Mean stomatal frequency for basal leaves ranged from 50.24 to 54.80 per mm2 for tetraploid and from 39.24 to 40.50 per mm2 for hexaploid accessions. For upper leaves, it varied from 52.88 to 53.50 per mm2 for tetraploid and from 46.12 to 46.40 per mm2 for hexaploid accessions. The two ploidy forms also exhibited clear differences for stomatal size on the basal and upper leaves of the main tiller. Mean stomatal size for the basal leaves ranged from 41.42 to 46.09 m for tetraploid and from 53.59 to 54.97 m for hexaploid accessions. For upper leaves, stomatal size varied from 47.29 to 53.29 m in tetraploid and from 64.76 to 66.84 m for hexaploid accessions. None of the individual accession ranges overlapped between the ploidy levels. Stomatal frequency and size were highly negatively correlated. Increased ploidy in Ae. neglecta has resulted in fewer but larger stomata per unit leaf area. Stomatal size and/or frequency on the adaxial surface of leaves could be used to distinguish tetraploid from hexaploid cytotypes in Ae. neglecta in fresh material from Turkey collected in the field.  相似文献

Random amplified polymorphic DNA (RAPD) markers were used to evaluatethe levels and patterns of genetic diversity in ten Elymusalaskanus populations, which were collected from Canada, USA,Greenland and Russia. Ten arbitrarily chosen decamer primers were used in thisstudy. The results revealed high levels of variation. The mean number of allelesper locus (Ap) was 1.5, ranging from 1.4 to 1.6, the meanpercent of polymorphic loci (Pp) was 49.5%, ranging from35.1% to 64.9%, and the mean gene diversity (Hep) was0.162, varying from 0.142 to 0.262. The total variation wasH T = 0.403. When partitioned(G ST), 60% of the total variation was foundamong the populations. Although the genetic diversity values obtained with RAPDsare much higher than for allozymes, they are similar regarding how the geneticvariation is distributed among populations. In addition, a similar geneticpattern of population differentiation, where populations from Greenland andthe USA (violaceus and latiglumis)were clearly separated from the others (hyperarcticus,komarovii and sajanensis), wasrevealed by both the cluster and principal coordinates analyses.  相似文献

Installation of heating cables for warming soil was used to evaluate the effect of disturbance on soil solution chemistry within a northern hardwood forest (Adirondack Mountains, New York). Differences in response among treatments suggested the importance of both the depth and timing of cable installation. There were increases (p>0.05) in many solutes within pilot study plots in which surrogate cable was installed at 15 cm depth. Most notably, mean nitrate concentrations for the 1st year following disturbance were 744 eq l-1 at 15 cm depth compared to 7 eq l-1 for the non-disturbed control. A comparison of pilot plots with 5 cm cable depth and an unheated soil-warming control plot with the same cable disturbance showed that the seasonality of soil disturbance may have a key role in response to disturbance. The soil solution response was diminished if installation occurred during the spring, a period of rapid uptake of nitrogen by vegetation. Mean nitrate concentrations were 176 eq l-1 for 5-cm pilot plots (installed in fall 1991) versus 6 eq l-1 for 5-cm, unheated soil-warming control plots (installed in spring 1992). Disturbance effects were attenuated over time and not generally apparent 1 year after installation.  相似文献

Summary Physiological and symbiotic characteristics were identified in Rhizobium fredii isolated from subtropical-tropical soils. The generation times of R. fredii Taiwan isolated-SB 357 and -SB 682 were 1.7 and 2.5 h, respectively. These strains were associated with acid production in yeast-extract mannitol medium. They were able to use hexoses, pentose, sucrose, trehalose and raffinose. Strain SB 357 can resist a high concentration of kanamycin (100 g ml–1 and penicillin (400 g ml–1). It can tolerate up to 2.34% NaCl and 1031.3 mosmol kg–1 (23.4 bars). The growth rate of R. fredii SB 357 under the concentration of approximately 450 mosmol kg–1 (10.2 bars) was not affected by salinity, but responded to osmotic pressure. Both strains (SB 357 and SB 682) isolated from subtropical-tropical soils were able to form an effective N2-fixing symbiosis with the US soybean cv Clark lanceolate leaflet.  相似文献

Summary Soil mesofauna (sensu Ghilarov, i.e., larger than 1 mm) was sampled from under four shrub species (Lygos raetam, Thymelaea hirsuta, Lycium shawii, and Josonia candicans) growing on the littoral oolitic sand dunes at Gharbaniat, 53 km W of Alexandria, Egypt, for a period of more than 30 months, spanning 3 years. Population density (PD) was calculated in relation to area of shrub canopy. Seasons were considered, according to the prevailing climatic conditions as well as to earlier physiological studies on the animals concerned, as: four winter months, four summer months and two months for each of the transitional seasons spring and autumn. The PD values obtained for each season thus defined were combined for the 41 taxa that were sampled. The table of these data was treated by correspondence analysis (CA) and ascending hierarchic classification (AHC), by the Roux DATA-VISION programme (suitable for Apple). Results from such treatment show that fauna as related to shrub species are differentiated along the first factorial axis, while as related to seasons they are differentiated along the second axis. Taxa common to all shrubs and to all seasons are centrally placed and cause some confusion in the resolution. Summer taxa are more distantly placed than those related to the other three seasons. Fauna under Lygos is richer, more diverse, and more complementary as regards trophic levels (with predators), and could probably be the source for replenishing the fauna of the other shrubs at all seasons. Seasonal changes in microbial biomass and nutrient flush in forest soils   总被引:14,自引:0,他引:14  
Microbial biomass and N, P, K, and Mg flushes were estimated in spring, summer, autumn, and winter samples of different forest soils. The microbial biomass showed significant seasonal fluctuations with an average distribution of 880±270 g C g-1 soil in spring, 787±356 g C g-1 soil in winter, 589±295 g C g-1 soil in summer, and 560±318 g C g-1 soil in autumn. The average annual concentrations of C, N, P, K, and Ca in the microbial biomass were 704, 106, 82, 69 and 10 g g-1 soil, respectively. Microbial C represented between 0.5 and 2% of the organic soil C whereas the percentage of microbial N with respect to the total soil N was two-to threefold higher than that of C; the annual fluctuations in these percentages followed a similar trend to that of the microbial biomass. Microbial biomass was positively correlated with soil pH, moisture, organic C, and total N. The mean nutrient flush was 31, 15, 7, and 4 g g-1 soil for N, K, P, and Mg, respectively, and except for K, the seasonal distribution was autumn spring winter summer. The average increase in available nutrient due to the mineralization of dead microbial cells was 240% for N, and 30, 26, and 14% for P, K, and Mg, respectively. There was a positive relationship between microbial biomass and the N, P, K, and Mg flushes. All the variables studied were significantly affected by the season, the type of soil, and the interaction between type of soil and season, but soil type often explained most of the variance.  相似文献

Summary Two different soils were fractionated with three granulometric procedures. Enzyme activity measurements were carried out in four fractions of real (undisturbed), stable (water-dispersed) and structural (water-dispersed and physically disrupted) soil units.A predominance of catalase (67.3%), dehydrogenase (57.6%) and urease (65.3%) activities was observed in soil structural microunits with diameters of less than 50 m. Highest protease activity was found in fractions with a diameter of less than 50 m (66.3%) and in fractions with a diameter ranging from 200 to 2000 m (18.5%) which contain organic matter of a higher and lower grade of humification, respectively.All fractionation procedures result in a loss of total enzyme activities, which was more clear in the physically disrupted soil than in the sieved soil (e.g. 38.4% vs. 5.9% loss of urease activity).  相似文献

Summary Six fast-growing soybean rhizobia (Rhizobium fredii) and thirteen slow-growing soybean rhizobia (Bradyrhizobium japonicum) were examined for resistance to 10 antibiotics. Axenic studies were carried out to determine the competitiveness of dual-strain inocula consisting of fast- and slow-growing rhizobia isolated from subtropical-tropical soils for nodule occupancy on a hybrid of Asian and US soybean cultivars. Nodule occupancy was determined by intrinsic resistance to erythromycin and neomycin. The results showed wide variability in resistance to 10 antibiotics for fast- and slow-growing rhizobia. The intrinsic antibiotic resistance of fast- and slow-growing rhizobia was extremely high against nalidixic acid (400 g ml–1) and penicillin (200 g ml–1). The competitive ability of inoculant strains for nodule occupancy varied for different combination sets and with the plant growing media. Our results show that fast-growing rhizobia nodulate a hybrid of Asian and US soybean cultivars. Fast-growing soybean rhizobia did not completely exclude nodulation by the slow-growing strains, which formed 0–79% nodules, depending on the strain used in the inoculum.  相似文献

Recolonization of soils by organisms is dependent not only on biological factors but also on site factors including soil and vegetation. The total nematode populations in soil sterilized 52 months earlier with methyl bromide ranged from 77 to 123% of those in untreated soil; both vegetation and rainfall appear to have been important factors in the recolonization. In a high-rainfall pasture the dominant plant-feeding nematode Helicotylenchus did not recolonize and was replaced by Paratylenchus. Under high rainfall, Iotonchus failed to recolonize either pasture or forest, but Clarkus recolonized well. While indices such as the maturity index, the summed maturity index, PPI, the ratio of fungal: bacterial feeding nematodes, species richness, the Shannon-Weaver index, and indices of evenness, dominance, and diversity showed various effects, none gave consistent trends; rainfall and vegetation probably exerted too much influence. In the light of trends in our data, in data for other ecosystems, and the reality of r-K strategies in animals, it may not be reasonable to expect a global nematode index of soil health or ecosystem condition. While a nematode index may be useful in local or regional studies, research efforts may be better directed towards elucidating key populations of nematodes for investigating the impact of particular land management practices on ecosystem sustainability.  相似文献

Summary The effects of Cd, Co, Ni and Zn on growth and survival of Macrophomina phaseolina were studied in vitro. Cd, Ni and Co at 500 g ml–1 inhibited growth by 78.8%, 73.6% and 11.8%, respectively, after 4 days at 25 ± 1°C. The mycelial dry weight yield was enhanced by 2.1% at 100 g ml–1 Zn. The population of the pathogen declined in soil amended separately with Cd, Co, Ni and Zn. Cd (4000 mg kg–1) proved to be the most toxic, by completely inhibiting the survival of the pathogen in soil after 20 days.  相似文献
