The μ-opioid receptor (MOR) is expressed in bovine, human, equine and canine oocytes, and in seasonal breeders, it is expressed with higher intensity during the anoestrous phase. Supplementation of in vitro maturation (IVM) medium with opioid agents, agonists or antagonists, was shown to affect oocyte maturation in several species such as rat, bovine and equine. This study reports the effects of supplementing IVM medium with naloxone (Nx), an opioid antagonist, on nuclear and cytoplasmic maturation rate of oocytes recovered from anoestrous bitches. Cytoplasmic maturation was examined in terms of mitochondrial (mt) distribution. In order to confirm the receptor-mediated action of Nx, in oocytes of anoestrous bitches, MOR expression was analyzed by Western blot. Cumulus–oocyte complexes, recovered from the ovaries of bitches in anoestrous, were cultured in vitro and Nx was added at the concentrations of 1 × 10−6, 1 × 10−8 and 1 × 10−10  m . The rate of oocytes resuming meiosis after culture in presence of 1 × 10−6  m Nx (29%) was significantly higher than that of oocytes of control group (12%; p < 0.05). However, treatment with Nx did not affect mt distribution pattern. In denuded oocytes and in corresponding cumulus cells, a doublet of 65 and 50 kDa was observed. We conclude that, in oocytes of anoestrous bitches, MOR is expressed and Nx significantly improves nuclear maturation rate. Further studies should be performed to elucidate the expression of other opioid receptors, such as δ and κ, and possible interactive effects of their antagonists on canine oocyte maturation.  相似文献

Various methods can be used to feed dairy calves that may influence calf performance and these include changing frequency of feeding or age at weaning. Two trials were conducted to determine effects of feeding frequency and weaning age on calf growth, health and rumen development, where 124 Holstein heifer and bull calves were weaned at 3, 4, 5 or 6 weeks of age. During Trial 1, milk replacer (12.5% DM; 22% CP, 15.6% fat) was fed at 10% BW twice per day until 1 week prior to weaning when intake was reduced to 5% BW. During Trial 2 calves were fed at 10% BW in two feedings until 14 days, then at 10% BW once daily until 1 week prior to weaning when milk replacer was reduced to 5% BW. Blood glucose and urea nitrogen, BW 4 h post-feeding, heart girth, hip height, and withers height 4 h post-feeding were obtained weekly. Growth and structural measurements were similar for all treatments up to 8 weeks of age in both trials. Blood constituents and health observations were not different between trials. Similar growth and performance between treatments in both trials through 8 weeks of age indicate that calf performance is not affected by weaning early and feeding once daily.  相似文献

This study evaluated the supplementation effects of three herbal plants (thyme [THY], eucalyptus [EUC] and celery [CEL]), a commercial phytogenic additive ?containing essential oils (PFA‐EO, Digestarom® P.E.P.) and monensin (MON) in calf starter on performance, rumen fermentation and blood metabolites during pre‐ (days 3–55) and post‐weaning (days 56–70). Sixty‐six Holstein dairy calves (3 days of age, 41.2 ± 3 kg of BW) were allocated to one of six starters supplemented with: (i) no additives (CON), (ii) MON (30 mg/kg), (iii) THY (23 g/kg), (iv) CEL (23 g/kg), (v) EUC (23 g/kg) and (vi) PFA‐EO (3 g/kg). All the calves were offered starters ad libitum plus 6 L of whole milk daily. Starter intake tended to be the highest in calves fed PFA‐EO and THY; intermediate in calves fed CON, MON and EUC; and the lowest in those fed CEL. Average daily gain (ADG) and feed efficiency (FE) remained unaffected by dietary treatments during the pre‐weaning. During the post‐weaning period, ADG and FE ?were greatest in calves fed EUC followed by those fed CON, MON, PFA‐EO and THY, and then in those fed CEL. No differences were observed among the treatments in skeletal growth, faecal score, rumen pH or ammonia‐N concentration. Compared to calves fed CON and MON, those fed the herbal plants or PFA‐EO tended to recorded higher molar proportions of acetate and butyrate, and the acetate: propionate ratio. Blood malondialdehyde level did not differ among treatments, but calves on CON had the highest glucose concentration, and those fed PFA‐EO recorded the highest value for β‐hydroxyl butyrate on day 70. In conclusion, the results indicate that the three herbs and PFA‐EO are capable of modulating some of the rumen fermentation parameters and blood metabolites as well as eucalyptus could potentially be a better alternative to monensin for improving post‐weaning performance.  相似文献

Cell numbers of known species of nitrate- and nitrite-reducing bacteria, Selenomonas ruminantium, Veillonella parvula and Wollinella succinogenes , in the rumen of goats (25–30 kg) were estimated by competitive polymerase chain reaction (PCR). The number of S. ruminantium was the largest of the three species examined, and tended to be greater in goats fed a high-concentrate diet (5.6 × 107 cells/mL rumen fluid) than in goats fed a high-roughage diet (1.3 × 107 cells/mL). The number of V. parvula tended to be greater when goats were fed a high-roughage diet (6.7 × 103/mL) than when fed a high-concentrate diet (3.2 × 103/mL). The number of W. succinogenes was below the detectable level (< 1.0 × 102/mL) when a high-concentrate diet was fed, but was significantly increased by feeding a high-roughage diet (1.6 × 103/mL). Addition of potassium nitrate (6 g/day) to the high-concentrate diet tended to increase V. parvula , and significantly increased W. succinogenes , indicating that these two bacteria can be increased by feeding a diet containing nitrate.  相似文献

Records for Afshari sheep were retrieved from data collected between 2000 and 2005 at the Zanjan University experimental flock, at Zanjan, Iran. (Co)variance components and corresponding genetic parameters for birth weight (BW), weaning weight (WW), 6-month weight (W6), average daily gain from birth to weaning (ADGa), from birth to 6 months (ADGb), from weaning to 6 months (ADGc), Kleiber ratio at weaning (WWKR) and Kleiber ratio at 6 months of age (W6KR) were estimated using univariate and bivariate analyses by the DFREML procedure. The Kleiber ratio, defined as growth rate/metabolic weight, has been suggested to be a useful indicator of growth efficiency and an indirect selection criterion for feed conversion. Estimates of direct heritability ( h 2) were 0.23, 0.27, 0.11, 0.22, 0.07, 0.01, 0.13 and 0.06 for BW, WW, W6, ADGa, ADGb, ADGc, WWKR and W6KR, respectively. Maternal genetic effects represented a relatively large proportion of the total phenotypic variance for BW ( m 2 = 0.22), whereas maternal permanent environmental effects were significant for W6 ( c 2 = 0.15), ADGb ( c 2 = 0.16), ADGc ( c 2 = 0.14) and W6KR ( c 2 = 0.16). Results of bivariate analyses indicated the variable genetic correlations between traits. The largest positive genetic relationships were between adjacent measurements. The moderate estimates of h 2 for early growth traits indicate that in Afshari sheep faster genetic improvement through selection is possible for these traits. In order to increase the efficiency of feed conversion, use of Kleiber ratio in selection programmes was recommended.  相似文献

This experiment was conducted to determine the effects of interval feeding of whole sunflower seeds on the performance of beef cows and their progeny. During mid to late gestation, 144 multiparous, spring-calving beef cows (588 kg of initial BW; 5.6 initial BCS; 4 to 13 yr old) were individually fed 1 of 3 supplements 4 d/wk for a 76-d period. Supplements (DM basis) included: 1) 0.68 kg of soybean meal/feeding (NCON); 2) 3.01 kg of a soybean hull-based supplement/feeding (PCON); and 3) 1.66 kg of whole sunflower seeds high in linoleic acid/feeding (WSUN). Supplements were formulated to provide similar amounts of CP and ruminally degraded intake protein; PCON and WSUN were also formulated to be isocaloric. During the supplementation period, cows had free-choice access to bermudagrass (Cynodon dactylon) and tall-grass prairie hay. By the end of the 76-d supplementation period, cows fed PCON (P < 0.01) and NCON (P < 0.01) had gained more BW than cows fed WSUN (33, 23, and 10 kg, respectively). However, from the end of this supplementation period to the beginning of the breeding season 84 d later, cows supplemented with PCON had lost more (P < 0.01) BW than cows supplemented with WSUN (-123 kg vs. -111 kg). Cow BW change through weaning (-50 kg, P = 0.43) and final cow BW (536 kg, P = 0.70) at weaning were not different among supplement groups. Furthermore, cow BCS was similar among supplement treatment groups at the end of the supplementation period (5.3, P = 0.09), at the beginning of the breeding season (4.8, P = 0.38), and at weaning (4.7, P = 0.08). No difference among treatments was detected for calf birth weight (36 kg, P = 0.42), calf weaning weight (235 kg, P = 0.67), percentage of cows exhibiting luteal activity at the beginning of the breeding season (57%, P = 0.29), or pregnancy rate (88%, P = 0.44). However, first service conception rate was greater (P = 0.01) for cows fed PCON (79%) and tended (P = 0.07) to be greater for cows fed WSUN (74%) than for cows fed NCON (53%). After weaning, all steer calves were placed in a feedlot and fed a high-concentrate finishing diet for an average of 188 d. Supplements fed to dams during gestation did not influence feedlot performance or carcass characteristics. Prepartum energy supplementation, regardless of energy source or prepartum energy balance, resulted in improved conception rate, but other measures of reproduction, calf and feedlot performance, and carcass characteristics were not affected.  相似文献

A total of 233 silages were examined and found that Monascus ruber was present in 43 samples with counts between 1 × 103 and 9 × 106 colony-forming units (CFU)/g (mean: 2 × 105 CFU/g). Monacolin KL and the hydroxy acid monacolin KA were detected by liquid chromatography-mass spectrometry in 45 and 50 of 233 samples at levels ranging from 25–15 600 and 28–65 400  μ g/kg, respectively. Citrinin was found with high-performance liquid chromatography-fluorescence detection (FLD) in 14 (6%) samples, the concentrations varied between 2.4 and 64.2  μ g/kg. The concentrations of citrinin were low and toxic effects are not anticipated. Monacolin KA and monacolin KL occur frequently and in considerable amounts in silages. These metabolites are believed to influence the metabolic activity of rumen anaerobic fungi resulting in a poorer digestion of crude fibre.  相似文献

The aim of this study was to determine the effect of nutrition during foetal and lactation periods on calf growth and body composition, and their association with changes in metabolic and endocrine profiles during the calf first year of life on purebred (Hereford and Angus) and cross‐bred (F1) dam offspring. Forty cross‐bred calves and their dams (purebred – PU: Hereford and Angus, and cross‐bred – CR: F1) were used in a randomized block design with a factorial arrangement of herbage allowance of native pastures (high: Hi‐HA and low: Lo‐HA), 4 vs. 2.5 kg dry matter/kg body weight (BW) and dam genotype (PU vs. CR). Calf BW and blood samples were collected monthly from birth to 380 ± 15 days of age, and body composition was estimated by the urea dilution technique at weaning (142 ± 15 days) and 380 days. Calf birthweight did not differ among groups but from birth to 380 days, and BW was reduced (p = 0.046) in Lo‐PU offspring. Although Lo‐CR calves achieved similar BW than Hi‐PU and Hi‐CR offspring, they showed an increased fat in detriment of lean tissue deposition. At birth, plasma total protein was less (p = 0.04), while plasma glucose, insulin or IGF‐I tended or were greater (p < 0.072) in Hi‐HA than Lo‐HA calves. Greater (p < 0.03) plasma total protein and/or glucose concentrations during the first months of lactation were observed in CR offspring associated with the greater dam milk production. Although glucose concentrations did not differ among calf groups after weaning, plasma insulin was greater (p = 0.004) in Hi‐PU than other groups at 380 days. Consistent with the reduced BW, Lo‐PU offspring presented the lowest (p = 0.026) plasma IGF‐I from birth to 380 days. Herbage allowance of native grasslands during calf foetal and lactation periods interacted with maternal heterosis to affect, in the short and/or long term, calf BW or body composition, and metabolic and endocrine profiles.  相似文献

Thirty-seven Holstein and seven crossbred female calves (16.1 ± 4.60 days, and an initial BW of 36.5 ± 3.19) were used to study the effects of conventional (CF) vs enhanced-growth feeding programs (EF) on performance, plasma amino acid (AA) concentrations, and rumen microbial development. After 1 week of adaptation to milk replacer (MR), the CF calves received 4 l/day of MR at 12.5% DM throughout the preweaning period, and the EF calves were offered MR at 18% DM: 6 l/day from 1 to 6 days, 8 l/day from 7 to 26 days, and 4 l/day from 27 days to weaning day (38 days). Calf starter and water were offered ad libitum throughout the study (87 days). Calves fed EF were heavier (P < 0.05) than CF calves at the end of the study (111.7 vs 102.6 ± 1.72 kg, respectively). Until the 27 days, average daily gain (ADG) was greater (P < 0.001) for EF than for CF calves (1.00 vs 0.49 ± 0.061 kg/day, respectively), but it was lower (P < 0.001) from days 27 to 45 of the study (0.32 vs 0.71 ± 0.061 kg/day, respectively), coinciding with the days around weaning. Starter intake was greater (P < 0.001) for CF than for EF calves during the first 45 days of the study (0.60 vs 0.27 ± 0.061 kg/day, respectively) but similar afterwards. As a consequence, EF treatment may have delayed rumen function as suggested by total daily purine derivatives urinary excretion (49.52 vs 33.27 ± 3.095 mmol/day, in CF and EF calves, respectively). Linear regression analyses showed a positive relationship between plasma Trp and Phe concentrations and ADG, and a negative relationship between these two AA and plasma urea concentrations, suggesting that Trp and Phe could be limiting growth in calves fed conventional feeding programs.  相似文献

Angus bulls and heifers from lines divergently selected for serum IGF-I concentration were used to evaluate the effects of IGF-I selection line on growth performance and feed efficiency in 2 studies. In study 1, bulls (low line, n = 9; high line, n = 8; initial BW = 367.1 +/- 22.9 kg) and heifers (low line, n = 9; high line, n = 13; initial BW = 286.4 +/- 28.6 kg) were adapted to a roughage-based diet (ME = 1.95 Mcal/kg of DM) for 24 d and fed individually for 77 d by using Calan gate feeders. In study 2, bulls (low line, n = 15; high line, n = 12; initial BW = 297.5 +/- 34.4 kg) and heifers (low line, n = 9; high line, n = 20; initial BW = 256.0 +/- 25.1 kg) were adapted to a grain-based diet (ME = 2.85 Mcal/kg of DM) for 32 d and fed individually for 70 d by using Calan gate feeders. Blood samples were collected at weaning and at the start and end of each study, and serum IGF-I concentration was determined. Residual feed intake (RFI) was calculated, within study, as the residual from the linear regression of DMI on midtest BW(0.75), ADG, sex, sex by midtest BW(0.75) and sex by ADG. In study 1, calves from the low IGF-I selection line had similar initial and final BW and ADG, compared with calves from the high IGF-I selection line. In addition, DMI and feed conversion ratio were similar between IGF-I selection lines; however, calves from the low IGF-I selection line tended (P < 0.10) to have lesser RFI than calves from the high IGF-I selection line (-0.26 vs. 0.24 +/- 0.31 kg/d). In study 2, IGF-I selection line had no influence on performance or feed efficiency traits. However, there was a tendency (P = 0.15) for an IGF-I selection line x sex interaction for RFI. Bulls from the low IGF-I selection line had numerically lesser RFI than those from the high IGF-I selection line, whereas in heifers, the IGF-I selection line had no effect on RFI. In studies 1 and 2, weaning and initial IGF-I concentrations were not correlated with either feed conversion ratio or RFI. However, regression analysis revealed a sex x IGF-I concentration interaction for initial IGF-I concentration in study 1 and weaning IGF-I concentration in study 2 such that the regression coefficient was positive for bulls and negative for heifers. These data suggest that genetic selection for postweaning serum IGF-I concentration had a minimal effect on RFI in beef cattle.  相似文献

We examined the effect of endophyte infection level of tall fescue (Festuca arundinacea Schreb.) used for stockpiled forage on the performance of lactating, fallscalving beef cows and their calves. Treatments were endophyte infection levels of 20% (low; SEM = 3.5), 51%, (medium; SEM = 1.25), and 89% (high; SEM = 2.4; 4 replications/treatment). Five cow-calf pairs grazed in each replicate (n = 60 cow-calf pairs/yr) for 84 d (phase 1) starting on December 2, 2004 (yr 1), and December 1, 2005 (yr 2). After 84 d of grazing each treatment, the cattle were commingled and fed as a single group (phase 2) until weaning in April of each year. Phase 2 allowed measurement of residual effects from grazing stockpiled tall fescue with varying levels of endophyte infection. Pregrazing and postgrazing forage DM yield, forage nutritive value, and total ergot alkaloid concentrations of forage were collected every 21 d during phase 1. Animal performance data included cow BW, ADG, and BCS, as well as calf BW and ADG. Animal performance was monitored during both phases. Endophyte infection did not affect (P = 0.52) apparent intake (pregrazing minus postgrazing forage DM yield) of stockpiled tall fescue, because each cow-calf pair consumed 16 +/- 1.7 kg/d regardless of treatment. Cow ADG during phase 1 was -0.47 +/-0.43 kg for the low treatment, which was greater (P < 0.01) than either the medium (-0.64 +/-0.43 kg) or high (-0.74 +/- 0.43 kg) treatments. However, cows that had grazed the high or medium treatments in phase 1 lost -0.43 and -0.57 (+/-0.24) kg/d, respectively, which was less (P < 0.01) BW loss than the cows in the low (-0.78 +/- 0.24 kg/d) treatment during phase 2. By the end of phase 2, cow BW did not differ (528 +/-27 kg; P = 0.15). Body condition score for cows in the low treatment was greater (P = 0.02) than that of the medium and high treatments at the end of phase 1. Body condition scores did not change appreciably by the end of phase 2, and differences among treatments remained the same as at the end of phase 1 (P = 0.02). In contrast to cow performance, calf ADG was unaffected (P = 0.10) by endophyte level and averaged 0.73 +/- 0.07 kg during phase 1 and 0.44 +/- 0.04 kg during phase 2. Our data suggest that fall-calving herds can utilize highly-infected tall fescue when stockpiled for winter grazing, with little impact on cow performance and no impact on calf gain.  相似文献

The effects on cow and calf performance of replacing grass silage with brewers grains in diets based on barley straw and fed to pregnant beef cows are reported. Using a 2 × 2 factorial arrangement of breed and diet, cows pregnant by artificial insemination (n = 34) of two breeds (cross‐bred Limousin, n = 19 and pure‐bred Luing, n = 15) were fed diets ad libitum which consisted of either (g/kg dry matter) barley straw (664) and grass silage (325; GS) or barley straw (783) and brewers grains (206, BG) and offered as total mixed rations. From gestation day (GD) 168 until 266, individual daily feed intakes were recorded and cow body weight (BW) and body condition score (BCS) measured weekly. Calving date, calf sex, birth and weaning BW, and calf age at weaning were also recorded. Between GD 168 and 266, cross‐bred Limousin cows gained more weight than Luing cows (p < 0.05) and cows offered BG gained more weight than cows offered GS (p < 0.001). Luing cows lost more BCS than cross‐bred Limousin cows (p < 0.05), but diet did not affect BCS. There were no differences in dry matter intake as a result of breed or diet. Calf birth BW, however, was greater for cows fed BG than GS (44 vs. 38 kg, SEM 1.0, p < 0.001) with no difference between breeds. At weaning, calves born to BG‐fed cows were heavier than those born to GS‐fed cows (330 vs. 286 kg, SEM 9.3, p < 0.01). In conclusion, replacement of grass silage with brewers grains improved the performance of beef cows and increased calf birth and weaning BW. Further analysis indicated that the superior performance of cows offered the BG diet was most likely due to increases in protein supply which may have improved both energy and protein supply to the foetus.  相似文献

Ghrelin action, which stimulates growth hormone (GH) secretion, may alter during the weaning period in calves. Our objective was to compare the effects of intravenous ghrelin injection on plasma GH, insulin and glucose concentrations in calves around the weaning period. Four Holstein bull calves were fed whole milk and allowed free access to solid feeds, and weaned at 7 weeks of age. Measurements were performed at weeks 1, 2, 4, 6, 7, 9, 11 and 13, when calves were intravenously injected with ghrelin (1.0 μg/kg body weight (BW)) through a catheter, and jugular blood samples were obtained temporally relative to the injection time. Estimated digestible energy intake per metabolic BW transiently decreased at week 7 because of low solid intake immediately after weaning, and thereafter gradually increased. Plasma insulin and glucose concentrations were not affected by ghrelin injection at all ages. In contrast, plasma GH concentrations increased with ghrelin injection at all ages. The incremental area of GH at week 7 was greatest and significantly higher compared with weeks 2, 4, 6 and 9. This result suggests that nutrient insufficiency immediately after weaning enhances GH responsiveness to ghrelin.  相似文献

The aim of this study was to assess the effects of particle size (PS) of alfalfa hay on growth characteristics and rumen development in dairy calves at two levels of alfalfa supplementation. Fifty newborn dairy calves (42.7 ± 2.2 kg BW) were used in a 2 × 2 factorial arrangement with the factors supplementation level (low, 8%; or high, 16% on DM basis) and PS (medium, 2.92 mm; or long, 5.04 mm as geometrical means) of alfalfa hay. In addition, a control group without alfalfa hay was used. Hence, treatments were: control (C); low level with medium PS (LM); low level with long PS (LL); high level with medium PS (HM) or high level with long PS (HL). Growth performance of alfalfa‐fed calves did not differ from control calves, but alfalfa supplementation decreased corneum thickness of the rumen wall. In alfalfa‐fed calves, post‐weaning starter intake was greater for LL calves than for LM calves. During the entire rearing period, starter intake was 26–32% higher for LL and HM calves than for LM calves. Pre‐weaning average daily gain was higher for LL and HM calves than for HL calves, but this effect was not persistent over the entire rearing period. Final body weight decreased from 86 to 79 kg when the level of long PS alfalfa hay increased from 8 to 16%, but increased from 78 to 87 kg when the level of medium PS alfalfa increased from 8 to 16%. Regardless of PS and level, morphometric characteristics of rumen wall were generally similar among alfalfa feeding groups, but corneum thickness decreased from 8.7 to 6.1 μm with greater PS at the low level. These results indicate that adequate, but not excessive, physical stimulation is required for appropriate rumen development and growth performance of dairy calves.  相似文献

Housing and feeding are integral to calf rearing, and must meet calf needs while remaining functional for the farmer. This study compared health, behavior, growth, and labor requirements of calves housed in groups indoors and fed via an automatic or manual milk feeding system compared to calves manually fed in individual or group hutches outdoors. Seventy-six (49 Holstein Friesian [HF] and 27 HF × Jersey) dairy heifer calves were balanced for birth weight (35.2 ± 4.95 kg), birth date (1 February ± 7.2 d) and breed. The experiment was a randomized block design with four treatments; 1) indoor group housing with automated feeding (IN_AUTO; 12 calves per pen), 2) indoor group housing with manual feeding (IN_MAN; 12 calves per pen), 3) outdoor group hutch with manual feeding (OUT_G_MAN; 8 calves per pen), and 4) outdoor individual hutch with manual feeding (OUT_I_MAN; 6 calves: 1 per pen). Calves in OUT_treatments moved outdoors at 18 d (± 5.9 d). Each treatment was replicated once. Milk allowance increased gradually from 6 to 8 L/day (15% reconstitution rate) with ad libitum fresh water, concentrates, and hay offered from 3 d old. Gradual weaning occurred at 8 wk old. Measurements were divided into period 1; before movement outdoors, and period 2; after movement outdoors. Health was similar among treatments, regardless of period, with the most frequent score being zero (i.e., healthy). Summarized, standing and lying were observed 24.3% and 29.8%, respectively, in OUT_I_MAN calves, compared to 8.0% and 49.1%, for the other systems, which were similar. No difference in bodyweight (BW) existed between treatments, except at weaning where BW was lower for OUT_I_MAN (67.4 ± 2.84 kg) compared to IN_MAN (74.2 ± 2.01 kg), and day 102 where OUT_I_MAN (94.1 ± 2.85 kg) were lighter than IN_AUTO (101.1 ± 2.10 kg) (P = 0.047). Total labor input was greatest for OUT_I_MAN (00:02:02 per calf per day; hh:mm:ss) and least for IN_AUTO (00:00:21 per calf per day) (P < 0.001). The labor for feeding (00:00:29 per calf per day), feeding inspection (00:00:10 per calf per day), and cleaning equipment (00:00:30 per calf per day) was greatest for OUT_I_MAN. All calves showed good health and growth patterns. Differences in behavior expressed by calves in the OUT_I_MAN, compared to other treatments may indicate compromised welfare. Thus, although outdoor group hutches do not negatively impact calves, indoor housing, particularly using automated feeders, can improve labor efficiency.  相似文献

Our objectives were to examine the effects of added fat in late-gestation cow diets on neonatal response to cold. In Exp. 1, pregnant fall-calving heifers received control (n = 5), safflower seed (n = 5), or whole cottonseed (n = 5) diets. The hay-based, isonitrogenous, and isocaloric diets, fed for 47 d prepartum, contained 1.5, 4.0, and 5.0% fat for control, safflower, and whole cottonseed diets, respectively. At calving, calf BW and vigor score, as well as fat, lactose, and IgG in colostrum were not affected (P > 0.30) by diet. Heifers fed the safflower diet tended to have greater colostral solids (P < 0.10) than heifers fed the control or whole cottonseed diets. At 6.5 h of age, calves were placed in a 5 degrees C cold room for 90 min. Calf vigor, shivering, body temperature, and blood samples were taken every 15 min. During cold stress, calf body temperature decreased 0.7 degrees C (P < 0.03). Across all diets, shivering and serum glucose concentrations increased (P < 0.05), whereas calf vigor and cortisol concentrations decreased (P < 0.02) during cold exposure. In Exp. 2, pregnant spring-calving cows (n = 98) received a control (n = 47) or whole cottonseed (n = 51) supplement. Hay-based diets fed for 68 d prepartum contained 2.0 and 5.0% fat for control and whole cottonseed diets, respectively. Calf BW, vigor, shivering, dystocia score, time to stand, time to nurse, serum glucose concentrations, and serum IgG were not affected (P > 0.50) by diet. Between 30 and 180 min, body temperature of calves from dams fed the whole cottonseed supplement decreased (P < 0.05) more than calves from dams fed the control supplement. Serum glucose concentrations in calves were not affected by diet (P > 0.30). Serum cortisol concentrations tended (P < 0.09) to be greater for calves from dams fed whole cottonseed than control calves. When ambient temperature was < 6 degrees C, calves born to dams fed whole cottonseed had greater (P < 0.05) BW, tended (P < 0.1) to stand earlier, and had greater serum IgG concentrations. We conclude that calves from dams fed high-fat diets containing safflower or whole cottonseed respond similarly to cold stress, but these responses may not be consistent with greater cold resistance. In addition, high-fat dietary supplementation of late-gestation cows may only be beneficial during calving seasons with prolonged cold weather.  相似文献

Sixteen bull calves were born in the Spanish Central Pyrenees in spring. They were either early (EW, 90 days of age) or normal weaned (NW, 180 days of age). At day 90, EW calves were placed on an intensive diet while NW calves were turned out with their dams to high mountain pastures. After summer, at day 180, NW calves were weaned and placed with EW calves on a common finishing diet until slaughter at the fixed age of 1 year. From birth to early weaning date, no performance differences appeared. However, EW calves gained faster (1.549 kg/day) than their unweaned counterparts (0.783 kg/day) from early to normal weaning date (P < 0.001). During the finishing period, NW calves showed compensatory growth, with a 44% higher ADG than EW calves (P < 0.001), with a similar feed intake and a better feed conversion ratio. Early weaned calves had a longer fattening phase than NW calves (264 vs. 158 days, respectively; P < 0.001) and thus total feed intake and feed costs were greater. When slaughtered at 1 year of age, EW and NW calves attained similar weight (489 vs. 510 kg, respectively; P > 0.05), but dressing percentage was higher for EW calves (56.9%) than for NW calves (55.2%) (P < 0.01), which led EW calves to have heavier carcasses, without differences in fat score or conformation. The different growth paths, the result of weaning management, did not affect meat tenderness, chemical composition and fatty acid profile, but affected meat lightness, with higher values for compensating calves (NW) than calves in continuous growth (EW). In conclusion, advancing weaning age modified calf performance without affecting substantially carcass characteristics, except for an improvement in dressing percentage, or meat quality.  相似文献

Glucagon-like peptide-1 (7-36) amide (GLP-1), secreted by the small intestine, has insulinotropic and glucose-lowering action. Basal plasma GLP-1 concentrations were measured in calves around the weaning period, the effect of short-chain fatty acids (SCFA) on plasma GLP-1 concentrations was examined, and the effects of GLP-1 administration on plasma insulin, glucagon, and glucose concentrations were measured. Thirteen Holstein bull calves were fed whole milk and solid feed and weaned at 7 wk of age. Preprandial plasma samples were obtained from 5 calves once a week from week 0 to 13 to measure basal concentrations of plasma GLP-1 and insulin (experiment 1). Four calves were intravenously administered with a mixed solution of SCFA (2.4 mmol/kg body weight [BW]) in week 2 and 11 to measure plasma GLP-1 concentrations (experiment 2). Another 4 calves were intravenously injected with GLP-1 (1.0 μg/kg BW) to elucidate the response of plasma insulin, glucagon, and glucose concentrations in week 1, 2, 4, 6, 7, 9, 11, and 13 (experiment 3). In experiment 1, age and weaning did not affect preprandial basal concentrations of plasma GLP-1 throughout the experimental period. Preprandial insulin concentrations increased after weaning (P < 0.05), and GLP-1 and insulin were more strongly correlated postweaning than preweaning. In experiment 2, intravenous treatment with SCFA increased plasma GLP-1 concentrations in both week 2 and 11 (P < 0.05.) In experiment 3, intravenous GLP-1 treatment decreased plasma glucose concentrations throughout the experiment (P < 0.05), but increased plasma insulin concentrations only after weaning (P < 0.05). Treatment with GLP-1 did not affect plasma glucagon concentrations, regardless of age. These results indicate that preprandial basal concentrations of plasma GLP-1 in calves are not changed by weaning, but SCFA stimulate GLP-1 secretion. The insulinotropic action of GLP-1 is detected only after weaning, but the glucose-lowering action of GLP-1 is not affected by weaning.  相似文献

Effects of calving season and finishing system on forage and concentrate consumption and carcass characteristics of calves were compared. In each of 3 yr, two replicates of three growing and finishing systems were compared including 1) spring calves finished on a high-grain diet in a feedlot immediately post-weaning (WF); 2) spring calves backgrounded on a hay-corn gluten diet over winter for 179 +/- 18 d after weaning, grazed for 98 +/- 9 d in cool-season grass-legume pastures, and finished on a high-grain diet in a feedlot (SGF); and 3) fall calves backgrounded on a hay-corn gluten feed diet over winter for 69 +/- 31 d after weaning, grazed for 98 +/- 9 d in cool-season grass-legume pastures, and finished on a high-grain diet in a feedlot (FGF). During the grazing phase, calves on the SGF and FGF treatments were equally stocked with spring-calving cow-calf pairs before grazing by pregnant fall-calving cows in a first-last rotational stocking system at a rate of 1.9 standard livestock units/ha. As designed, retained calves in the FGF system spent 110 fewer days in the drylot during backgrounding than retained calves in the SGF system (P = 0.01), resulting in less feed provided during winter. A greater (P < 0.01) quantity of hay was fed to SGF calves after weaning over winter (1,305 kg of DM per calf) than the quantity fed to FGF calves (305 kg of DM per calf). Quantity of grain (including commercial starter) fed to SGF calves after weaning did not differ (P = 0.28) from that fed to FGF calves (126 vs. 55 kg of DM per calf); however, calves in the FGF system required 80 and 71 kg of DM per calf more concentrate to finish to an equivalent external fat thickness compared with SGF and WF calves, respectively (P = 0.02). Average daily gains in the feedlot were greater (P < 0.01) for SGF and FGF calves than for WF calves during all 3 yr. There were no differences (P = 0.69) in carcass quality grades among calves in all groups, but SGF calves had greater (P < 0.01) hot carcass weight and LM area measurements at slaughter than FGF or WF calves. Although calves in the FGF system were 25 kg lighter than calves in the WF system at slaughter (P = 0.03), and had a lower dressing percent (P = 0.03), other carcass characteristics did not differ between these two groups. Lower stored-feed requirements and similar carcass quality characteristics made retention of a fall calf crop advantageous over retention of a spring calf crop for use as stocker animals before finishing.  相似文献

We investigated the effect of increasing nutrient intake on the responsiveness of the GH/IGF-I system in calves fed a high-protein milk replacer. Fifty-four Holstein bull calves were fed one of three levels (low, medium, and high; n = 18 per treatment) of a 30% crude protein, 20% fat milk replacer to achieve target rates of gain of 0.50, 0.95, or 1.40 kg/d, respectively, for low, medium, and high. Six calves per treatment were slaughtered at approximately 65, 85, and 105 kg BW. Additionally, six calves were slaughtered at 1 d of age to provide baseline data. Plasma aliquots from blood samples collected weekly were analyzed for IGF-I, insulin, glucose, NEFA, and plasma urea nitrogen (PUN). Plasma IGF-I and insulin, measured weekly, increased (P < 0.001) with greater nutrient intake from wk 2 of life to slaughter. Plasma glucose and NEFA also increased (P < 0.05) with nutrient intake. In addition, each calf underwent a GH challenge beginning 4 d before the scheduled slaughter. Plasma from blood collected before the first GH injection and 14 and 24 h after the third injection was analyzed for IGF-I and PUN. Response to challenge, calculated as the absolute difference between the prechallenge and 14-h postchallenge plasma IGF-I concentrations, was significant in calves on all three treatments. Plasma urea nitrogen was not different among treatments as measured weekly but decreased (P < 0.001) following GH challenge in all calves. Results of ribonuclease protection assays showed increased expression of hepatic mRNA for GH receptor 1A and IGF-I with increased intake. The amounts of GH receptor and IGF-I mRNA in muscle and adipose, however, were not affected by intake. In summary, plasma IGF-I was elevated in calves with increased nutrient intake, and the elevations in plasma IGF-I following short-term administration of GH were significant in all calves by 65 kg BW. Data demonstrate that in well-managed milk-fed calves the somatotropic (GH/IGF-I) axis is functionally coordinated and sensitive to nutrient intake and GH.  相似文献
