为探讨SD大鼠乙型肝炎病毒(HBV)和戊型肝炎病毒(HEV)的感染情况,本试验采集了53例SD大鼠肝脏,采用免疫组织化学染色、HE染色和Mallory三染色法对上述肝脏样品中的HBsAg、HBcAg和HEV进行了检测,同时对其病理形态学变化进行了观察。结果如下:在53例肝脏样品中有47例呈HBsAg阳性反应,阳性率为88.68%;有38例呈HBcAg阳性反应,阳性率为71.70%;HBV的双阳性率为66.15%,总阳性率为96.23%;而HEV的阳性率为100%。镜下HBsAg阳性反应物在肝细胞胞核、胞浆和胞膜上均有分布,而HBcAg阳性反应物主要分布于肝细胞胞核和胞浆内;HEV阳性反应物分布在肝细胞胞核、胞浆和小叶间胆管上皮中。组织病理学观察结果显示,在上述免疫组化阳性反应肝脏样品的HE染色切片中,均有不同程度的病理变化,主要表现为肝细胞固缩、变性、坏死、中央静脉淤血、淋巴细胞浸润、小叶间胆管增生。Mallory三色染色结果证明有22%的肝脏样品有纤维结缔组织增生。  相似文献

目的:研究IL-4、VCAM-1、VEGF和VEGFR2四种蛋白在MDCK细胞中的表达与分布,探讨四种蛋白与MDCK细胞成瘤性的相关性。方法:采用免疫组化SP(streptavidin-perosidase)法观察四种蛋白在MDCK细胞中的表达分布特点。结果:IL-4、VCAM-1、VEGF与VEGFR2四种蛋白在MDCK细胞中均有表达。结论:IL-4定位于MDCK细胞胞浆中,在胞核及胞质中表达,呈弥漫性分布;VCAM-1、VEGF与VEGFR2三种蛋白定位于MDCK细胞胞膜上,在胞膜及胞质中表达。IL-4、VCAM-1、VEGF和VEGFR2蛋白均为细胞因子并参与细胞成瘤进程,可能与MDCK细胞成瘤性存在相关联系。  相似文献

为了检测山羊迷走神经结状神经节中是否有促黄体生成素受体(LHR)的存在,探讨促黄体生成素(LH)是否能够影响结状神经节内脏感觉神经元的功能活动。本试验采用免疫组织化学SP法观察LHR在结状神经节中的分布特征,利用IPP 6.0图像半定量分析系统分析LHR在结状神经节中阳性神经元和阳性非神经元上的表达量差异。结果显示,LHR主要分布在神经元细胞膜和细胞质中,呈棕黄色为强阳性。在带核的神经元中,神经元细胞核呈圆形空泡状,LHR为阴性反应。神经元群间的神经纤维呈淡黄色,LHR为弱阳性。LHR在神经元胞体上的相对表达量极显著高于节内其他阳性非神经元结构(P0.01)。结果表明,山羊结状神经节内脏感觉神经元是LH作用的主要靶细胞,提示LH有可能通过作用于结状神经节内脏感觉神经元胞体上的LHR参与调节内脏器官感觉信息的传导。  相似文献

运用免疫组织化学超敏 SP法对山羊胎儿脊髓发育中神经生长因子 (nerve growth factor,NGF)及其高亲和力受体 Tr KA的表达及其功能进行了系统的研究和探讨。结果显示 ,山羊胎儿脊髓灰质中存在 NGF及其受体 Tr KA,于 6周龄胚就可检测到 ,随胚龄增加 ,其表达范围及免疫反应着色程度逐渐增强。 NGF主要分布于腹角和背角的神经细胞 ,反应产物主要定位于胞质和突起 ;Tr KA的分布主要以腹角及胶状质为主 ,反应产物主要定位于胞核 ,后期胞质及突起也可见到阳性反应。在山羊胎儿脊髓白质中也可观察到 NGF及 Tr KA免疫阳性反应 ,其发育后期更为显著 ,阳性反应主要分布于神经胶质细胞核、神经纤维的轴索及雪旺氏细胞。结果提示 ,NGF不仅对交感和感觉神经元的发育起作用 ,而且还与腹角运动神经元的发育有关  相似文献

应用光镜、电镜观察了 7~8 d 小鼠精原干细胞的形态结构特点;应用细胞化学、免疫组织化学方法研究了体外培养小鼠精原干细胞的细胞化学和免疫组织化学特性。结果显示,精原干细胞呈圆形,直径(9±1) μm ,核大,呈圆形或轻微卵圆形,胞质较少;核内常染色质占绝对优势,异染色质极少;胞质内核糖体、线粒体较丰富,其他细胞器不发达;精原干细胞 A K P染色呈阳性或强阳性,胞质内脂滴很少或没有;精原干细胞表达特异的 ckit 受体;体外培养的小鼠精原干细胞常呈不完全的胞质分裂,细胞间由细胞间桥相连而呈链状或团状的细胞群丛,细胞群常由偶数细胞组成。  相似文献

透射电镜观察表明,比格犬(Beagle)脑垂体远侧部实质中含有6种细胞,即生长激素细胞、催乳激素细胞、促甲状腺激素细胞、促性腺激素细胞、促肾上腺皮质激素细胞和滤泡—星形细胞,它们各有其超微结构特征。生长激素细胞呈圆形;胞核为圆形;分泌颗粒很多,圆形,直径约233~465nm,催乳激素细胞多呈不规则形;胞核为不规则形;分泌颗粒很多,圆形,直径约310~620nm。促甲状腺激素细胞呈圆形或卵圆形;胞核为圆形或卵圆形;分泌颗粒较少,直径约78~232nm。促肾上腺皮质激素细胞呈圆形或卵圆形;胞核为圆形或卵圆形;分泌颗粒较多,圆形,直径约155~310nm,分布于细胞周边。促性腺激素细胞呈圆形或椭圆形;胞核为圆形或椭圆形;分泌颗粒较少,呈圆形、卵圆形、杵状或不规则形,长径约155~434nm。滤泡—星形细胞呈星形或不规则形,有突起伸入相邻细胞之间;胞核为圆形或卵圆形;胞质内无分泌颗粒。  相似文献

采用初生山羊羔股骨骨膜,通过植块培养法、完全消化法和半消化植块法分离、培养成骨细胞。结果表明,植块培养法、完全消化法和半消化植块培养法的贴壁时间分别为4、24、4h;培养4d时的细胞数量分别为少量细胞、少量细胞、大量细胞,细胞汇合时间分别为21、19、10d。传代后第7d即可汇合,第14d出现矿化结节,茜素红染色呈橘红色,Von Kossa染色呈黑色;姬姆萨和HE染色后可见体外培养的细胞呈三角形、多角形,胞质丰富,胞核卵圆形,单核,有1~3个核仁;碱性磷酸酶染色后胞质呈黑色颗粒或块状沉淀;扫描电镜观察胞突发达,伸出的突起与临近细胞的突起相连。体外培养传至15代时细胞的生物学性状未发生变化。试验结果显示,半消化植块培养周期短,细胞损害小,有利于细胞贴壁,是一种可行的骨组织工程研究方法。  相似文献

为了阐明妊娠期垂体中催乳素(PRL)、促卵泡素(FSH),促黄体素(LH)的分布和三者之间协调关系.本试验采用免疫组化SP法检测了PRL、FSH,LH三种激素在妊娠各期山羊的腺垂体细胞中免疫反应产物的分布特点.结果显示,在山羊妊娠过程中,腺垂体中PRL阳性细胞多存在于远侧部,数量逐渐增多,胞质呈强阳性;FSH和LH阳性细胞分布在远侧部和结节部,FSH胞质内颗粒着色深浅不一,LH多核膜着色;相对表达量都逐渐升高,而升高程度不同,表现为妊娠后期LH表达量急剧升高,FSH则变化较小,除了FSH的相对表达量在妊娠前期和中期之间无显著差异(P>0.05)外,3种激素的相对表达量在妊娠前期、中期和后期3个时期之间都存在显著差异(P<0.05),妊娠前、中期FSH的相对表达量均高于PRL和LH,妊娠后期LH的表达量最高.FSH次之,PRL最低.提示PRL,FSH和LH三种激素相互协同以促进胚胎着床并维持妊娠,而且在妊娠早期PRL起主要作用,之后协同FSH和LH维持妊娠.  相似文献

为探索猪圆环病毒2型(PCV-2)对仔猪的致病机理,采用免疫组化方法和HE染色对人工感染PCV-2仔猪的呼吸、消化及淋巴系统主要器官进行抗原定位和组织病理观察。结果显示,淋巴器官的组织广泛分布阳性抗原,尤其在淋巴小结较为密集,组织出现不同程度的棕黄色着色,不均匀分布在胞质中。消化与呼吸系统的实质性器官功能性小叶不完整,组织结构紊乱,组织细胞呈不同程度的胞质溶解、胞核固缩及出现包涵体等病理变化。淋巴器官的组织细胞出现不同程度的裂解与消失,严重病例中大多数淋巴细胞成团消失,难以发现较为完整的淋巴小结,并在细胞质中出现球形的包涵体,也具有呈单或团状的类上皮样组织结构,细胞质出现溶解或消失,仅剩细胞膜的结构,出现胞核固缩,导致组织的空泡化现象。  相似文献

检测促黄体生成素受体(LHR)在雌性山羊颈动脉体(CB)中存在及其分布特点,为促黄体生成素(LH)是否对CB起调节作用提供形态学基础。取雌性山羊的CB进行LHR免疫组织化学(SP)法染色观察与分析。结果表明:LHR免疫阳性物质在颈动脉体中分布广泛;镜下可见,在CB的小球结构内,Ⅰ型、Ⅱ型细胞的细胞膜LHR免疫阳性物质呈棕黄色,为强阳性,在其细胞质和细胞核中LHR免疫阳性物质呈黄色,为中等阳性;CB的间质组织中,血管内皮细胞LHR免疫阳性物质呈棕黄色,为强阳性;此外间质还可见许多点状或线状的非细胞阳性结构,呈淡黄色,为弱阳性。对实质细胞群和间质组织平均光密度的分析结果表明,CB的实质细胞是LH作用的主要靶细胞之一,提示LH可能通过CB实质细胞上的LHR来介导生物学效应,影响CB的实质细胞对血液内化学刺激的感知,间接的影响动物机体心血管的功能。  相似文献

采用光镜和电镜技术,观察雏鸵鸟睾丸的显微和超微结构;采用免疫组织化学方法对NPY在睾丸内的表达进行研究,探明NPY在睾丸内的表达规律.光镜下可见,雏鸵鸟睾丸表面有一层由坚韧的结缔组织构成的白膜,它向睾丸实质内形成的结缔组织不发达,不形成睾丸纵隔和睾丸小隔;雏鸵鸟的曲精小管直径为85~90 μm,生精上皮由支持细胞和1~2层生精细胞构成,此时精原细胞已开始分裂增殖,可见明显的分裂相,初级精母细胞数量少,体积大.电镜下可见,精原细胞胞质内有少量线粒体、游离核糖体和内质网;初级精母细胞富含线粒体、游离核糖体、内质网和溶酶体;支持细胞与精原细胞之间界限清晰,胞质内含较多游离核糖体,其他细胞器不发达;间质细胞的胞核形状不规则,内含较多脂滴.免疫组化显示,NPY在曲精小管周围、睾丸小血管周围、精母细胞和睾丸间质内均有表达,在睾丸小血管周围和精母细胞内表达量较高,而在曲精小管周围和睾丸间质内表达量较低.  相似文献

为了探明雄性雏鸵鸟生殖器官的形态学特点,采用石蜡切片和HE染色技术,对6羽45日龄雄性非洲雏鸵鸟的生殖器官进行了研究,并与家禽的相关结构进行了比较。在光镜下观察了雄雏鸟生殖器官的形态结构,结果表明:(1)鸵鸟睾丸没有睾丸纵隔和睾丸小叶,曲精小管中精原细胞排列紧密且整齐,睾丸间质较少;(2)附睾由睾丸网、输出小管和附睾管组成;(3)输精管管壁由黏膜、肌层和外膜3层组成;(4)阴茎由2个圆形的纤维性淋巴体、淋巴间隙和微血管共同构成。  相似文献

The volumetric proportion of the various ducts of the epididymis of the emu and ostrich and the immunohistochemistry of actin microfilaments, as well as cytokeratin, desmin and vimentin intermediate filaments, were studied in the various ducts of the epididymis of the emu and ostrich. The volumetric proportions of various ducts, which are remarkably different from those of members of the Galloanserae monophyly, are as follows: the rete testis, 5.2 ± 1.4% for the emu and 2.4 ± 1.8% for the ostrich; efferent ducts, 14.2 ± 2.3% (emu) and 11.8 ± 1.8% (ostrich); epididymal duct unit, 25.8 ± 5.8% (emu) and 26.1 ± 4.1% (ostrich) and connective tissue and its content, 54.7 ± 5.8% (emu) and 60.0 ± 4.9% (ostrich). Unlike in mammals and members of the Galloanserae monophyly, only vimentin was immunohistochemically demonstrated in the rete testis epithelium of the emu, and none of the cytoskeletal protein elements in the ostrich rete testis. The epithelium of the efferent ducts of the emu co-expressed actin, cytokeratin and desmin in the non-ciliated type I cells, and vimentin in the ciliated cell component. The ostrich demonstrated only cytokeratin in this epithelium. The ratite epididymal duct unit is different from that of mammals in lacking actin (only weaky expression in the ostrich), desmin and cytokeratin, and a moderate/strong immunoexpression of vimentin in the basal cells and basal parts of the NC type III cell in the epididymal duct unit. Immunoexpression of the microfilaments and intermediate filaments varied between the two ratite birds, as has been demonstrated previously in birds of the Galloanserae monophyly, and in mammals.  相似文献

The effects of unilateral castration (UC) and induced unilateral cryptorchidism (CR) on plasma hormones and testis anatomy were studied in 36 Holstein bulls altered at either 3, 6 or 9 mo of age (n = 12). Plasma hormone concentrations were determined in six samples collected at hourly intervals on d 0, 1, 3, 7, 14 and 30, and then at monthly intervals through 6 mo after gonadal manipulation. Although plasma testosterone (T) showed a transient decrease (P less than .05) immediately after treatment, mean plasma concentrations of luteinizing hormone (LH) and T were unaffected by UC or CR over the 6-mo period (P greater than .05). Both hormones increased (P less than .05) in concentration with advancing age. Plasma follicle stimulating hormone (FSH) concentration was greater (P less than .05) in UC than in intact (IN) bulls overall, while FSH in CR bulls did not differ (P greater than .05) from either group. At slaughter, 11 mo after gonadal alteration, mean testis weight, ratio of testis weight to body weight and mean testis sperm cell numbers were increased (P less than .05) in UC bulls compared with mean testis values in intact (IN) bulls. Unilateral castration increased (P less than .05) seminiferous tubuler diameter and seminiferous epithelial cell height from basement membrane to the border of the lumen, but did not alter the ratio of tubuler to interstitial space within the testis. Seminiferous tubuler diameter and epithelial cell height were increased (P less than .05) in CR compared with IN bulls. Unilateral gonadal alteration at 3 mo of age caused a greater (P less than .05) hypertrophy of the scrotal testis in both UC and CR bulls than alteration at 6 or 9 mo of age. Results indicate that unilateral gonadal disruption is followed by rapid compensation in testis T production, little change in systemic LH and a rapid increase in secretion of FSH in the bull within those ages investigated. Further, UC elicits a greater compensatory hypertrophy than CR and the pituitary-testis endocrine axis is more responsive to alteration at 3 mo than at 6 or 9 mo of age in the bull.  相似文献

Classical swine fever (CSF) virus (CSFV) nucleic acid and antigen were detected in 15 pigs with naturally occurring chronic CSF by in situ hybridization and immunohistochemistry. The most consistent and prominent microscopic lesions were perivascular mononuclear cell infiltration and gliosis in the central nervous system of pigs with chronic CSF. Positive cells typically exhibited a dark brown (in situ hybridization) or red (immunohistochemistry) reaction product in the cytoplasm without background staining. A positive signal for both in situ hybridization and immunohistochemistry was detected in mononuclear cells and lymphocytes of lymphoid tissues. Viral nucleic acid was detected in some tissue sections in the absence of viral antigen. The in situ hybridization technique developed in this study was useful for the detection of CSFV RNA in tissues taken from chronically infected pigs and may be a valuable technique for studying the pathogenesis of chronic CSFV infection.  相似文献

褐壳蛋蛋壳颜色与蛋品质及产蛋性能的关系   总被引:1,自引:0,他引:1  
在30周龄褐壳蛋鸡群中,收集当日产的正常蛋393权,按蛋壳颜色分为奶黄、浅褐、紫粉和深褐色4个组,逐枚测定各项蛋品质,并根据产蛋个体记录分析各组母鸡的产蛋性能。通过最小二来法进行数据处理。结果表明,不同壳色组之间的蛋形指教、哈氏单位、平均蛋重均差异不显著,而紫粉色和深褐色组的蛋壳厚度和强度均显著大于2个浅色组,以紫粉色组最大(0.382mm和4.67kg/cm2),奶黄色组最小(0.369mm和4.25kg/cm2)。在产蛋性能方面,产紫粉色蛋的母鸡蛋重最大(61.16克),但40周龄产蛋数及总蛋重都是低(90.84枚和5549.77g),而产浅色蛋的母鸡产蛋数最多(98.63)、总蛋重(5961.21g)也最大。因此,在褐壳蛋鸡的育种中必须充分考虑蛋壳颜色与蛋品质性状及产蛋性能的密切关系,通过平衡育种避免因只考虑单方面因素的选择而造成其它性状的不利变化。  相似文献

用电镜观察了牛病毒性腹泻病毒Oregon C24株在侵染新生犊牛睾丸细胞中的形态发生。成熟的病毒颗粒呈直径约为50nm的球形颗粒,内含直径约为30nm的核芯。病毒侵染宿主细胞后,在胞质内复制,通过糙面内质网膜出芽成熟,病毒可通过侵染细胞外排或在细胞死亡后含有病毒颗粒的空泡破溃而释放到胞外。  相似文献

The distribution of lectin bindings in the testis of babirusa, Babyrousa babyrussa (Suidae) was studied histochemically using 10 biotinylated lectins, Peanut agglutinin (PNA), Ricinus communis agglutinin (RCA I), Dolichos biflorus agglutinin (DBA), Vicia villosa agglutinin (VVA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Lens culinaris agglutinin (LCA), Pisum sativum agglutinin (PSA), Concanavalin A(Con A) and Ulex europaeus agglutinin (UEA I). Nine of 10 lectins showed a variety of staining patterns in the seminiferous epithelium and interstitial cells. The acrosome of Golgi-, cap- and acrosome-phase spermatids displayed various PNA, RCA I, VVA, SBA and WGA bindings, indicating the presence of glycoconjugates with D-galactose, N-acetyl-D-galactosamine and N-acetyl-D-glucosamine sugar residues respectively. No affinity was detected in the acrosome of late spermatids. LCA, PSA and Con A which have affinity for D-mannose and D-glucose sugar residues were positive in the cytoplasm of spermatids and spermatocytes. DBA was positive only in spermatogonia. In addition to DBA, positive binding in spermatogonia was found for VVA, WGA and Con A, suggesting the distribution of glycoconjugates with N-acetyl-D-galactosamine, N-acetyl-D-glucosamine, D-mannose and D-glucose sugar residues. Sertoli cells were stained intensely with RCA I, WGA and Con A. In Leydig cells, RCA I and Con A were strongly positive, while WGA, LCA and PSA reactions were weak to moderate. The present findings showed that the distribution pattern of lectin binding in the testis of babirusa is somewhat different from that of pig or other mammals reported previously.  相似文献

隐花色素1(Cryptochrome 1,Cry1)基因作为生物钟调控环路的负调控因子,对生物节律的稳定发挥着重要作用,因此研究Cry1基因在哺乳动物生殖轴系的表达对揭示哺乳动物季节性繁殖的调控机理有着重要意义。本实验应用qPCR和免疫组织化学等方法研究了Cry1基因在雄性绵羊生殖轴系(松果体、下丘脑、垂体、睾丸和附睾)的分布和表达情况,并通过生物信息学分析对Cry1蛋白的结构做了预测。结果显示:在绵羊生殖轴系各组织中均有Cry1表达,其中在下丘脑中的表达最高,睾丸次之,各组数据间差异显著。免疫组化结果显示Cry1蛋白在松果体和下丘脑中成弥散性表达,胞膜、胞质及胞核均有阳性表达。在垂体上Cry1蛋白主要位于毛细血管的管壁细胞和血管周围的腺细胞上。Cry1蛋白还在睾丸组织的基膜和间质细胞,以及附睾管腔上皮细胞、管周肌样细胞和腔面精子上呈阳性表达。生物信息学分析发现Cry1蛋白无信号肽信息和跨膜结构,主要位于细胞质和细胞核中,与牛、鹿的亲缘关系最近。Cry1基因参与了绵羊的繁殖,为生物钟调控哺乳动物的季节性繁殖提供了一定的研究基础。  相似文献
