The plasma concentration of glucose was monitored for six hours in lambs fed either ewe colostrum, cow colostrum or milk replacer by stomach tube at one hour of age. The feeding of ewe colostrum resulted in an elevation of the mean (+/- se) plasma glucose concentration from 2 . 6 +/- 0 . 3 mmol/litre before feeding to 7 . 3 +/- 0 . 92 mmol/litre three hours after feeding and this elevation was maintained. A similar rise was observed in the lambs fed cow colostrum but this was of shorter duration. A marked hyperglycaemia was observed after the feeding of milk replacer with a maximum value of 23 . 4 +/- 1 . 33 mmol/litre three hours after feeding. The practical implications of these results are discussed.  相似文献

The transference of immunoglobulins from six New Zealand Romney ewes to their lambs was examined. Immunoglobulin levels were determined in ewe plasma, colostrum and lamb plasma shortly after birth and before the lambs fed, in lamb plasma 2 days after birth, and lamb plasma, ewe plasma and milk 30 days after parturition. Levels of total IgE, and IgE, IgG1, IgG2, IgM, and IgA with specificity for Trichostronglus colubriformis third stage larval secretory/excretory products (TcL3E/S) were determined. Mean levels of total IgE were three times higher in colostrum than in parturient ewe plasma while only trace amounts were detected in milk at 30 days after birth (107.7, 34.3, and 0.2U ml(-1), respectively, differences between means P< or =0.01). Mean total IgE in lamb plasma rose from being undetectable before suckling to levels comparable to those of the ewes by 2 days after birth (21.7U ml(-1)) and then declined to low levels by 30 days (0.4U ml(-1)). Total IgE levels in lamb plasma were significantly correlated with levels in ewe plasma and colostrum (r=0.91, P< or =0.01; r=0.96, P< or =0.003, respectively). The transference of TcL3E/S-specific IgE, IgG1 and IgA was substantial with mean levels of these antibodies in lamb plasma at 2 days comparable to that in parturient ewe plasma (absorbance levels in lamb plasma of 0.283, 0.537, and 0.334, respectively). Proportionally less maternal IgM and IgG2 appeared to be transferred to the lambs (absorbance of 0.112 and 0.081, respectively). Levels of TcL3E/S-specific IgE and IgG1 in lamb plasma at 2 days were significantly correlated with levels in parturient ewe plasma and colostrum (r=0.89 and 0.82, 0.85 and 0.96; all P< or =0.05, respectively). These results indicate that IgE is concentrated in ewe colostrum and that substantial amounts of maternal IgE are transferred to lambs via colostrum. Further, the results suggest that humoral immunity against gastro-intestinal nematode parasites and potentially other parasites in colostrum-fed lambs may approximate that of the ewe. The implications of the transference of humoral immunity through colostrum in ruminants for the passive protection and the development of active immunity against parasites remains to be fully explored.  相似文献

Newborn piglets were fed either a 5 per cent glucose/electrolyte solution or a commercial milk replacer for 24 or 36 hours before they were given colostrum, or they were given colostrum after birth. The effects of these treatments on their ability to absorb macromolecules was assessed by measuring the uptake of 125I-polyvinylpyrrolidone (125I-PVP) and the increases in the concentrations of total proteins and immunoglobulins in serum. The piglets which received colostrum at birth absorbed 50 per cent of a test dose of 125I-PVP within six hours, and there was a rapid increase in the concentration of immunoglobulins in their blood. The piglets which received the 5 per cent glucose/electrolyte solution absorbed 40 per cent of the test dose of 125I-PVP administered 24 or 36 hours after birth and nearly normal quantities of immunoglobulins, but the piglets given the milk replacer absorbed only 10 per cent of the test dose of 125I-PVP, and virtually no immunoglobulins.  相似文献

Twenty Holstein-Friesian male calves were obtained within 4 hours after bith (colostrum deprived) and allotted to 1 of 4 groups, each given a different feeding: colostrum, milk replacer, polyvinylpyrrolidone (PVP), and saline solution (0.85% NaCl). Each calf was fed 2 L of the respective diets every 12 hours. Rectal temperatures were recorded and blood samples were collected immediately before each feeding. At approximately 27 hours of age, all calves were inoculated orally with 1.5 X 10(10) viable organisms of a septicemia-producing Escherichia coli serotype O26: K60:NM. Within 8 hours, all calves had diarrhea. Coli-septicemia (E coli cultured from liver, spleen, and cardiac blood) was present in 1 of the 5 calves fed colostrum, in 5 or the 5 calves fed milk replacer, in 5 of the 5 calves fed PVP, and in 4 of the 5 calves fed saline solution. At necropsy of the calves (12 to 48 hours after oral inoculation), the same organism was isolated by cultural technique from small intestines of 19 of the 20 calves. Serum immunoglobulin G concentrations increased (P less than 0.01) in calves fed the colostrum diet in sharp contrast to the agammaglobulinemia occurring in calves fed the milk replacer, PVP, or saline solution. Results indicate that colostrum fed to the calf soon after birth provides protection from colisepticemia, but does not prevent the diarrhea of colibacillosis.  相似文献

To investigate the influence of maternal Se supply and plane of nutrition on lamb morbidity, mortality, and passive transfer of IgG, pregnant ewe lambs were used in 2 experiments with 2 × 3 factorial treatment arrangements. Supplementation of Se began at breeding and was either adequate Se (ASe, 9.5 μg/kg of BW) or high Se (HSe, 81.8 μg/kg of BW) in Exp. 1 or ASe (11.5 μg/kg of BW) or HSe (77.0 μg/kg of BW) in Exp. 2. On d 50 or 40 of gestation for Exp. 1 or 2, respectively, ewes were assigned randomly to 1 of 3 nutritional planes: 60% (RES), 100% (control, CON), or 140% (HI) of NRC requirements. This resulted in the following treatments: ASe-RES, ASe-CON, ASe-HI, HSe-RES, HSe-CON, and HSe-HI. Upon parturition, lambs were separated from their dams and serum samples obtained. Lambs were fed artificial colostrum for the first 20 h and then placed on milk replacer and grain pellets until completion of the study (Exp. 1, 57 d; Exp. 2, 21 d). Twenty-four hours after parturition, lamb serum samples were collected for IgG analysis. All lambs were reared similarly and morbidity and mortality assessed. Main effects were considered significant when P ≤ 0.05. In Exp. 1, there was a Se × plane of nutrition interaction (P ≤ 0.01) for lamb morbidity from birth to weaning and for 24-h IgG concentration. Lambs from ASe-RES and HSe-HI ewes were treated more frequently (P < 0.01) for respiratory and gastrointestinal disease, and lambs from HSe-HI ewes had the smallest (P < 0.01) 24-h serum IgG concentration. In Exp. 1, lambs from HI ewes also had the greatest (P < 0.01) mortality rates from birth to weaning compared with lambs from CON and RES ewes. In Exp. 2, there was an effect (P < 0.01) of maternal plane of nutrition with lambs from RES ewes having increased 24-h IgG compared with lambs from CON and HI ewes. There was no effect of maternal Se supplementation on lamb 24-h IgG in Exp. 2; however, there was a Se × plane of nutrition interaction (P < 0.01) for morbidity. From birth to 21 d of age, lambs from ASe-CON ewes had fewer (P < 0.01) treatment days compared with lambs from any of the other treatment groups. There also tended (P = 0.08) to be an effect of maternal Se supplementation on lamb mortality with increased mortality observed in lambs from HSe ewes. Results from the studies show a restricted maternal plane of nutrition can increase lamb serum IgG concentration. Selenium results were not consistent between the 2 experiments and may be due to differences in maternal Se.  相似文献

To examine effects of nutritional plane and Se supplementation on colostrum quality and mammary development, individually fed, pregnant Rambouillet ewe lambs were allotted randomly to 1 of 6 treatments in a 2 x 3 factorial arrangement. Main effects included dietary Se level, which began at breeding (d = 0) [adequate Se (9.5 mug/kg of BW) vs. high Se (81.8 mug/kg of BW)], and plane of nutrition, which began at d 50 of gestation [60% (RES), 100% (CON), and 140% (HIGH) of requirements]. Upon parturition, lambs were immediately separated from dams and weighed. Three hours after lambing, colostrum yield was determined, and samples were obtained for components and immunoglobulin G (IgG) analysis. Ewes were slaughtered within 24 h of parturition, and mammary tissues were collected for determination of alveolar secretory epithelial cell proliferation index and luminal area. Gestation length was reduced (P < 0.01) in HIGH ewes compared with RES and CON ewes. Although birth weights were reduced (P < 0.01) in RES and HIGH compared with CON ewes, there was little effect of diet on placental size. Mammary gland weight was reduced (P /= 0.15) on mammary gland weight, colostrum quantity, or IgG concentration in pregnant ewe lambs. Improper nutrition from mid to late pregnancy in ewe lambs altered colostrum quality and quantity and reduced offspring birth weight, which may have negative implications for lamb health and survival during the early postnatal period.  相似文献

OBJECTIVE: To determine whether passively acquired antibodies prevent development of a protective immune response to live virus in calves. ANIMALS: 18 calves. PROCEDURES: Calves were caught immediately after birth and tested free of bovine viral diarrhea virus (BVDV) and serum antibodies against BVDV. Within 48 hours, 12 calves were fed colostrum that contained antibodies against BVDV and 6 calves received BVDV antibody free milk replacer. Three milk replacer fed and 6 colostrum fed calves were exposed to virulent BVDV2-1373 at 2 to 5 weeks of life when passively acquired serum antibody titers were high. After serum antibody titers against BVDV had decayed to undetectable concentrations (at 7 to 9 months of age), the 3 remaining milk replacer fed calves, 6 colostrum fed calves previously exposed to BVDV2-1373, and 6 colostrum fed calves that had not been exposed to the virus were inoculated with BVDV2-1373. RESULTS: Passively acquired antibodies prevented clinical disease in inoculated colostrum fed calves at 2 to 5 weeks of life. Serum antibody titers did not increase in these calves following virus inoculation, and serum antibody titers decayed at the same rate as in noninoculated colostrum fed calves. Inoculated colostrum fed calves were still protected from clinical disease after serum antibody titers had decayed to nondetectable concentrations. Same age colostrum fed calves that had not been previously exposed to the virus were not protected. CONCLUSIONS AND CLINICAL RELEVANCE: A protective immune response was mounted in calves with passive immunity, but was not reflected by serum antibodies titers. This finding has implications for evaluating vaccine efficacy and immune status.  相似文献

Thirty new born male kids were allotted into three groups to evaluate the effects of diet and age on complement system activity in serum. After the colostrum feeding period, the control group (C) received a commercial milk replacer; the CLA group received 20 g/kg milk replacer DM of conjugated linoleic acid; and the GMK group was fed with goat milk. The kids were fed colostrum for 2 days and then either milk replacer or goat milk from days 3 to 60. Blood samples were taken at 1, 10, 20, 30, 40, 50 and 60 days of age. The complement system activity was higher (P < 0.05) in the GMK group throughout the whole experiment through alternative pathway. C kids did not present complement activity at any time. CLA group complement activity was statistically higher than that of C kids from 30 to 60 days, through alternative pathways. In conclusion, the milk replacer formula should be reformulated because it did not induce any complement system activity in the first two months of life.  相似文献

The purpose of this study was to clarify whether cats have a colostral and milk phase of lactation differentiated by concentrations of immunoglobulins, and whether colostrum ingestion by newborn kittens is essential for optimal transfer of passive immunity. Milk from specific pathogen-free queens was analyzed for IgG and IgA concentrations from parturition through 6 weeks of lactation. Serum IgG and IgA concentrations from birth through 8 weeks of age were determined for colostrum-fed kittens, colostrum-deprived kittens that were fed a milk replacer, and colostrum-deprived kittens that were fostered onto queens in the milk phase of lactation. The total IgG and IgA concentrations in milk were significantly higher on the day of parturition than on day 7 of lactation, indicating cats do have a colostral phase of lactation. The predominant immunoglobulin in both colostrum and milk was IgG. The serum IgG concentrations in colostrum-deprived kittens fostered on queens in the milk phase of lactation were similar to colostrum-deprived kittens fed a milk replacer, and the concentrations were significantly lower than in colostrum-fed kittens for the first 4 weeks of life. The serum IgA concentrations in both colostrum-deprived groups were significantly lower than colostrum-fed kittens on day 2 after parturition, but were similar thereafter. Colostrum-deprived kittens fostered onto queens in the milk phase of lactation had failure of passive transfer of maternal antibodies. Protective concentrations of immunoglobulins can be restored in kittens with failure of passive transfer of immunity by parenteral administration of adult cat serum, but not by fostering on queens in mid-lactation.  相似文献

The transfer of immunoglobulin G (IgG) and immunoglobulin M (IgM) from ewe to lamb was quantitated to determine the occurrence of failure in passive transfer. Concentrations of IgG and IgM in ewe serum did not correlate with those in the colostrum. Colostrum from all ewes contained abundant amounts of immunoglobulins when compared with serum values, with IgG being selectively concentrated over IgM. Absorption through the intestinal tract of the lamb appeared to be a nonselective process, lacking predilection for IgG and IgM. All lambs tested 24 hours after birth absorbed colostral immunoglobulins to some extent; however, 13 (14%) of 91 clinically normal lambs demonstrated some failure of passive transfer. In contrast, failure of passive transfer was found in 27 (46%) of 59 lambs dying of natural causes between 24 hours and 5 weeks of age. Evidence presented emphasizes the importance of absorption of adequate amounts of immunoglobulins to enable the newborn lamb to survive the first few weeks of life.  相似文献

The apparent efficiency of absorption and the decrease of specific colostral IgG after its passage into the blood stream were determined in newborn lambs fed with a single dose of colostrum containing anti-peroxidase IgG at 30 minutes, 12 hours and 24 hours after birth. When colostrum was given at 30 minutes after birth, a value of 16.9+/-4.0 per cent of anti-peroxidase IgG ingested appeared in lamb circulation. This percentage was reduced to 9.8+/-0.8 per cent when the feeding was done at 12 hours after birth and no specific IgG was detected in lambs fed at 24 hours after birth. The concentration of anti-peroxidase IgG in lambs' serum declined quickly within 96 hours of age to about 48 per cent of the initial value, and afterwards the level decreased slowly reaching a value of 10 per cent at 32 days of age. This behaviour probably reflects the protein distribution and use of absorbed antiperoxidase IgG.  相似文献

Different methods for the dry preservation of colostrum to be used as a total replacement for bovine colostrum were evaluated. Pooled colostrum from the first and second postpartum milkings from multiparous dairy cows containing immunoglobulin in excess of 40 g/L was freeze-dried, microwave vacuum evaporated, and spray-dried. Spray-drying produced a dried colostrum in which immunoglobulin quantity and function were preserved and was the most cost-effective. Other dehydration methods, although effectively conserving immunoglobulins, were too slow and costly to be used to produce a bovine colostrum replacer. Newborn, colostrum-deprived, dairy calves fed spray-dried colostrum containing 126 grams of immunoglobulin reconstituted in three liters of water as their sole source of immunoglobulin achieved normal mean serum immunoglobulin concentrations. Spray-dried colostrum with high concentrations of immunoglobulin may be produced economically and used as an effective and convenient colostrum replacer in newborn calves.  相似文献

This study examined the time course of rumen metabolic development in the absence of solid feed consumption and the effect of delayed solid feed consumption on sheep rumen development. Twenty-seven lambs consumed milk replacer until slaughter at nine ages from 1 to 84 d (milk group). Three additional lambs consumed milk replacer from 1 to 48 d. From 49 d until slaughter at 84 d, these lambs were weaned onto solid feed (fed group). At slaughter, rumen contents were removed for VFA analysis and rumen epithelium was preserved for morphological examination. Rumen epithelial cells were isolated and incubated in media containing 2.5 mM U-[14C]-glucose or 10 mM 1-[14C]-butyrate. Rumen VFA concentrations did not change with age in lambs given milk replacer. At 84 d of age, intraruminal VFA concentrations were elevated in lambs consuming solid feed compared to 84-d-old lambs given milk replacer (P < .05). The number of ruminal papillae per square centimeter decreased (P < .05) while papillae length and width did not change significantly with age in rumen epithelium from lambs given milk replacer. At 84 d of age, rumen epithelium from lambs in the fed group had fewer and larger papillae/per square centimeter than rumen epithelium from lambs given milk replacer (P < .05). Rates of glucose and butyrate oxidation and acetoacetate and lactate production by rumen cells isolated from lambs given milk replacer did not change with age. Beta-hydroxybutyrate (BHBA) production was undetectable before 42 d of age in lambs given milk replacer and increased to levels found in conventionally raised adults by 84 d. At 84 d there were no differences in rates of glucose and butyrate oxidation or acetoacetate and lactate production by rumen cells between the two treatment groups. Thus, the change in substrate oxidation from glucose to butyrate, indicative of rumen metabolic maturation, does not occur in the absence of solid feed consumption. However, the development of rumen ketogenesis, as evidenced by increased BHBA production, does occur in the absence of solid feed consumption. Delaying the initiation of solid feed consumption results in rumen morphological development but does not stimulate rumen metabolic development. Increased intraruminal VFA concentrations, earlier exposure to VFA, or a longer period of exposure to VFA may be required to induce the genes responsible for rumen metabolic development.  相似文献

Effects of milk replacer composition and intake on the growth of orphan foals were evaluated. Twenty foals were assigned to four treatments: 1) mare-nursed, 2) commercial foal milk replacer at recommended intakes (standard), 3) commercial foal milk replacer at high intakes (high), and 4) acidified replacer at recommended intakes (acidified). Foals fed milk replacer diets were weaned at 12-24 hours postpartum and fed milk replacer for 50 days. Mare-nursed foals were weaned between 52 and 56 days of age. Foals fed replacer diets gained 12% to 28% less weight than mare-nursed foals up to two weeks of age. However, by four months of age, weights of replacer-fed foals were similar to those of mare-nursed foals and 32 other mare-nursed foals at the farm weaned between three and four months postparium. Foals drank 10 to 12 L/100 kg body weight (BW) in fluid replacer daily over the trial period. During the first week, high intake foals consumed 26% more replacer (p<0.05) than foals fed acidified or standard diets. This higher intake resulted in diarrhea earlier (6-11 days vs 11-22 days) and for a longer time (6.3 days vs 2.5-3.6 days) than in foals fed recommended amounts. Mare-nursed foals developed “foal heat scours” in the second week postpartum. After the first week, foals fed high replacer diet voluntarily consumed the same volume of fluid replacer as foals fed the standard intake. Foals ate less than 1 kg grain mix/100 kg BW daily to one month of age, then increased intake to 1.5-2 kg/ 100 kg BW to weaning. Water intake was 20-40% of daily fluid intake and was correlated (r = 0.85) to dry matter intake. Foals in the high intake group ate less (p<0.05) solid feed and drank less water than foals fed the standard and acidified diets. The foal's stomach capacity appears to limit meal size and thus replacer intake. If recommended feeding intervals are used, replacer intakes by foals are less than 15% BW daily. High volume intakes appeared to prolong diarrhea. Normal growth rates occur when replacer and good-quality feeds are fed concurrently.  相似文献

The absorption by the neonatal gut of purified swine IgG isolated from maternal serum was studied in 91 motherless piglets raised without swine colostrum, to investigate some of the factors which influence absorption. Factors studied included the amount of IgG administered, the effect of bovine colostrum fed concurrently, the influence of starvation and the effect of lactose.Administration of iodinated IgG within 3 hours after birth resulted in the appearance of IgG in the piglet circulation in unaltered form. Immediate feeding of bovine colostrum followed by administration of iodinated IgG at 72 hours resulted in the appearance of only IgG digestion fragments in the circulation.Administration of 15 g of lactose during the first 24 hours reduced absorption of IgG by 26% compared to controls while treatment with 54 g of lactose reduced absorption by 94%. On the contrary, when bovine colostrum was administered immediately after purified IgG, the amount of swine IgG absorbed was 50–70% greater than in controls. Mature milk failed to have the same influence. Finally, in experiments in which the amount of purified swine IgG administered varied from 1.0 to 8.0 g, absorption was directly proportional to the amount administered. When these data were compared with data on naturally reared piglets, the same proportionality was seen and indicated a consistent 1:2.5 to 1:3 ratio of ingested IgG (g) to the maximal serum level of immunglobulin (mg/ml) resulting from absorption. IgA and IgM follow a similar pattern.The data are consistent with a regulated absorption mechanism, that appears to be lost very quickly after the initial ingestion of food. Heterologous protein and lactose alone can switch off this absorption mechanism, thus arguing against “gut closure” by a mechanism of receptor saturation by absorbed IgG. Bovine colostrum, when compared to mature milk, is able to augment the absorption of swine IgG by an unknown mechanism.  相似文献

A 2 x 2 factorial arrangement of treatments with 78 mature ewes was used to evaluate the effects of supplementing the pregnant ewe's diet with high levels of minerals and vitamin E on immunoglobulin G (IgG) absorption by the lamb and whether any altered efficacy of IgG absorption was due to the colostrum or to the lamb. The ewes were estrus-synchronized in October and housed in wk 10 of gestation. In the final 7 wk of gestation, a grass silage-based diet, offered ad libitum, was supplemented with 500 g of a 19% CP concentrate, and from 1 wk later until lambing, half the ewes was offered 48 g of a mineral/vitamin supplement containing 6.5 g of Ca, 4.9 g of P, 5.9 g of Mg, 4.0 g of Na, 790 mg of Zn, 3.5 mg of Se, 40 mg of I, 200 mg of Mn, 20 mg of Co, and 40 IU of vitamin E. At birth, the lambs were allocated to one of four treatments in a 2 x 2 factorial arrangement, with lamb origin and colostrum origin as the two factors. The lambs born to ewes not offered the mineral supplement were fed colostrum obtained from their own dams or from ewes in the mineral-supplemented treatment, whereas lambs born to ewes given supplemental minerals were fed colostrum obtained either from their dams or from ewes in the control treatment. The ewes were milked at 1, 10, and 18 h postpartum and the lambs were fed using a stomach tube. A 5-mL blood sample was taken from each lamb at 24 h postpartum for IgG analysis. The level of fecal adhesion to the upper tail/rump area of the lamb was subjectively scored at 72 h postpartum. There was no difference in gestation length, lamb birth weight, colostrum yield, or IgG production (P = 0.16 to 0.82). When ewes were fed supplemental minerals, the serum IgG content of the progeny was lower than in their control counterparts (6.8 vs. 16.1 g/L; P < 0.001), regardless of whether the lamb received colostrum from ewes with or without access to supplementary minerals. The difference in serum IgG concentrations at 24 h postpartum was a direct reflection of a compromised efficiency in IgG absorption. The progeny of ewes with access to minerals had higher (P < 0.05) levels of fecal adhesion, which was not related to the origin of the colostrum, indicating altered digestive function in these lambs. We conclude, using the sheep as a model, that high mineral intakes in late pregnancy not only lower serum IgG concentrations in the lamb, but also that high mineral intakes result in the neonate being preprogrammed at birth so that it is born with a compromised ability to absorb colostral IgG.  相似文献

OBJECTIVE: To determine sensitivity and specificity of a cow-side immunoassay kit for assessing IgG concentration in colostrum. DESIGN: Prospective study. ANIMALS: 76 dairy and 11 beef cows of various parities. PROCEDURE: Colostrum from first, second, and third milkings and milk samples were collected, and IgG concentration was determined by means of radial immunodiffusion. The immunoassay was performed according to the manufacturer's instructions, and sensitivity and specificity were calculated by comparing results of the immunoassay (positive vs negative) with results of immunodiffusion (< 50 g/L vs > or = 50 g/L). RESULTS: 135 colostrum or milk samples were collected. Mean +/- SD colostral IgG concentrations, determined by means of radial immunodiffusion for dairy and beef cows were 65.4 +/- 51.4 g/L and 114.8 +/- 42.7 g/L, respectively. Mean IgG concentrations for first-, second-, and third-milking colostrum samples and for milk samples were 92 +/- 49.0 g/L, 74.6 +/- 45.1 g/L, 47.5 +/- 32 g/L, and 6.8 +/- 3.8 g/L, respectively. Sensitivity of the immunoassay (ie, percentage of samples with IgG concentration < 50 g/L with a positive immunoassay result) was 93%, and specificity (ie, percentage of samples with IgG concentration > or = 50 g/L with a negative immunoassay result) was 76%. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that the immunoassay kit was an acceptable cow-side test to identify colostrum samples with IgG concentrations < 50 g/L. This study compared serum immunoglobulin G (IgG) concentrations in calves fed colostrum with those of calves fed a colostrum supplement containing spray-dried serum. Twenty-four Holstein calves were randomly assigned to I of 2 treatment groups (fresh colostrum or colostrum supplement). Each calf was fed 4 L of colostrum (n, = 12) or 4 L of colostrum supplement (n2 = 12) via oroesophageal intubation at 3 hours of age. The concentration of the colostrum supplement fed to calves was twice the manufacturer's recommendation. The median and range values for colostral IgG concentration were 6,430 mg/dL and 1,400-17,000 mg/ dL, respectively. Median serum IgG concentrations at 2 days of age differed significantly (P = .001) between calves receiving fresh colostrum (3,350 mg/dL) and the colostrum supplement (643 mg/dL). Eight percent of calves force fed colostrum had serum IgG concentrations < 1,000 mg/dL, whereas 75% of calves force-fed supplement had IgG concentrations below this threshold. The calculated population relative risks for mortality associated with passive transfer for calves force-fed colostrum and calves force-fed colostrum supplement were 1.09 and 1.90, respectively. Force-fed fresh colostrum is superior to the colostrum supplement studied, but the colostrum supplement has similar efficacy to routine colostrum administration practices.  相似文献

Effects of colostral antibody on susceptibility of calves to Cryptosporidium parvum infection were examined. Six calves were fed pooled colostrum that contained C parvum antibody, 6 times daily (at 4-hour intervals) for 7 days and then milk replacer for 7 days. Colostrum was obtained from healthy cows or cows inoculated parenterally with C parvum oocysts before parturition. Antibody content was determined in serum and colostrum whey, using an ELISA for anticryptosporidia immunoglobulin. Six calves were fed colostrum from healthy cows 1 time, and then milk replacer 6 times daily for 14 days. On day 1, all calves were challenge exposed with C parvum, PO, and were monitored daily for diarrhea and oocyst shedding. Bovine colostrum containing specific antibody to C parvum, at ELISA titers up to 10,240, was not effective in protecting calves against challenge exposure to C parvum.  相似文献

The importance of blood and colostrum/milk serum gamma-glutamyl transferase (gamma-GT) enzyme activity was evaluated to assess passive transfer status in healthy lambs. Thirty Akkaraman sheep (3-6 years old) were used which had normal pregnancy period and the same conditions, and the age of the lambs ranged between 0 and 15 days. Blood and colostrum/milk samples were collected from sheep and lambs after birth, before suckling (0) and after on 1st, 3rd, 7th and 15th days. Serum immunoglobulin G (IgG) concentration was determined by the use of Single Radial Immunodiffusion method. Serum gamma-GT activity was measured, using a commercially available kit in blood and colostrum/milk samples. Correlations were carried out between immunoglobulin and gamma-GT levels. Regression models (simple and multiple) were calculated with significant data. Linear correlation was determined between colostrum/milk gamma-GT activity and IgG concentrations and between serum gamma-GT activity and IgG concentrations in lambs on the 0 day. (r: 0.607, P: 0.001), 1st (r: 0.768, P: 0.001) and the 3rd (r: 0.603, P: 0.001) days and on the 1st (r: 0.637, P: 0.001) and 3rd (r: 0.478, P: 0.012) days in the experiment, respectively. Multivariate regression models were developed to estimate sample IgG concentration. Serum and colostrum/milk IgG concentration could be predicted using the formula: lamb serum IgG = 825 + 0.688 (lamb gamma-GT) + 52 (days); colostrum/milk IgG = 832 + 0.505 (colostrum/milk gamma-GT) - 167 (days). The regression models were moderately accurate in predicting serum IgG concentration (R2 = 0.51) and colostrum/milk IgG concentration (R2 = 0.55). Test sensitivity and positive predictive values for serum gamma-GT enzyme activity were found to be 96 and 100% and for colostrum/milk gamma-GT enzyme activity were found to be 100 and 68% to prediction IgG concentration. Serum and colostrum/milk gamma-GT activity can be used to assess passive transfer status of lambs. Along with this, regression models used to calculate serum and colostrum/milk gamma-GT activities found to be useful to estimate sample IgG concentration. The use of serum and colostrum/milk gamma-GT enzyme activity was found useful especially after birth on the 0, 1st and 3rd days.  相似文献
