Juvenile hihi or stitchbirds (Notiomystis cincta) and saddlebacks (Philestumus carunculatus) are particularly prone to systemic protozoal infections when kept in high density populations. Granu-lomatous lesions in the intestine, liver and spleen are associated with proliferating intracellular schizonts. Affected birds become anorexic and lose weight or die suddenly without clinical signs. The protozoan in hihi appears most likely to be Atoxoplasma spp whereas that in saddlebacks is more like Plasmodium spp.  相似文献

AIMS: To undertake disease surveillance for Chlamydia psittaci in native birds as part of a pilot study to examine pathogen diversity on Hauturu-o-Toi/Little Barrier Island. To retrospectively review the Massey University post-mortem database to determine previous cases of avian chlamydiosis in New Zealand.

METHODS:Mistnetting of forest birds was conducted across an elevational gradient on Hauturu-o-Toi/Little Barrier Island. Minitip culture swabs were used to collect cloacal samples from native birds. These swabs were screened for Chlamydia family DNA using two PCR methods. Positive results were sequenced. A retrospective review of the Massey University post-mortem database of all avian cases from 1990 to 2011 was conducted.

RESULTS:Ten native birds including four bellbirds (Anthornis melanura), three rifleman (Acanthisitta chloris), two hihi (Notiomyces cincta), and one whitehead (Mohoua albicilla) were sampled and one otherwise healthy female hihi was positive by both PCR screening methods for Chlamydophila. Sequencing confirmed 99–100% genetic similarity to C. psittaci. A retrospective review of the Massey University post-mortem database revealed no previous diagnoses of avian chlamydiosis in wild native New Zealand birds although it has been detected in captive parrots, and wild and captive exotic pigeons.

CONCLUSIONS:This is the first report of the detection of C. psittaci from a wild native bird in New Zealand. The bird was a Passeriforme from an endangered species that was captured free-living on Little Barrier Island. The incidence of avian chlamydiosis in native birds in New Zealand appears to be very low, based on the retrospective review of the post-mortem database.

CLINICAL RELEVANCE: It is unlikely that avian chlamydiosis is a significant problem for hihi population health. The detection of this organism has greater significance for other more susceptible species on Little Barrier Island and for human health, particularly for conservation workers involved in wildlife translocations. It further suggests that passerine birds may be a reservoir for C. psittaci in New Zealand ecosystems.  相似文献

AIMS: To identify network measures with relevance to disease spread in a network of movements derived from the Department of Conservation (DOC) translocation records from 1970 to mid-2014, and to identify conservation sites that should be prioritised for surveillance activities and improvements to data collection to make the best use of network analysis techniques in the future.

METHODS: Data included the source and destination of translocated specimens, the species and the dates the translocations were expected to occur. The data were used to construct a directed, non-weighted network in which a translocation event represented a tie in the network. Network density, in-degree (movements entering a node of interest) and out-degree (movements leaving a node of interest) and reciprocity were calculated.

RESULTS: The data analysed consisted of 692 unique translocations between 307 sites, with the majority (518; 73%) being for birds. The constructed network for bird, reptile and frog translocations comprised 260 nodes, with 34/260 (13%) having two-way movements and 47/260 (18%) non-reciprocal movements. The median degree score (sum of in- and out-degree) was two (min 0, max 36) with a mean of 3.5 in a right skewed distribution. Most sites acted as receivers or senders of consignments with only a few having both high in- and high out-degree, and thus had characteristics that made them sites of interest for surveillance activities. These included the National Wildlife Centre at Mount Bruce, Tiritiri Matangi Island and Te Kakahu (Chalky Island).

CONCLUSIONS: The presence of linking sites that join larger clusters within the network creates the potential for rapid disease spread if a pathogen were to be introduced. The important sites that supply or receive specimens for translocations are already well recognised by those performing translocations in New Zealand, and this paper provides further information by quantifying their role within the network.  相似文献

AIM: An intensive 2-year field study of 65 hihi (or stitchbirds) on Mokoia Island provided an opportunity to investigate the causes of mortality of this endangered species in a free-living environment. METHODS: The birds were observed daily during the breeding season (October to March) and every 4 weeks during the remainder of the year. Any abnormalities in behaviour, voice and body weight were recorded and all sick or dead birds which could be recovered were taken to the laboratory for necropsy. RESULTS: Thirty-one birds died during the period of this study. Aspergillosis was found to be the cause of death in six of nine adult birds examined post mortem. Some of the affected birds experienced subtle voice changes before becoming ill and all birds had granulomatous lesions in airsacs or lungs at necropsy. A further eight birds for which carcasses could not be found or which were autolysed showed similar clinical signs before death or disappearance. CONCLUSION: Because Aspergillus is an opportunist pathogen it seems likely that affected birds were immunosuppressed. This may have occurred during the breeding season which in this species is highly stressful. The presence of fungal material in bronchial exudate and the occurrence of the disease in mates, raises the possibility that aspergillosis in hihi may be contagious.  相似文献

AIM: To describe the microscopic and immunohistochemical findings in a case of pulmonary hyalohyphomycosis in a Kemp's ridley sea turtle (Lepidochelys kempi).

METHODS: Samples of lung, liver and kidney from a stranded, dead Kemp's ridley sea turtle were routinely processed for histopathological studies. Two monoclonal antibodies that reacted specifically with antigens of Aspergillus spp and the Mucorales (Zygomycetes) group, and a panel of polyclonal antibodies raised against Aspergillus fumigatus, Candida albicans, Geotrichum candidum, Fusarium solani, and Scedosporium apiospermum were used for immunohistochemical or immunofluorescence staining.

RESULTS: Histologically, a severe multifocal granulomatous pneumonia associated with fungal infection was diagnosed. All hyphae were identified as Fusarium spp because a strong and uniform reactivity was obtained only with a heterologously-absorbed polyclonal antibody raised against somatic antigens of Fusarium solani.

CONCLUSIONS: Fusarium spp should be included in the differential diagnosis of mycotic pneumonia in Kemp's ridley sea turtles.

CLINICAL RELEVANCE: This is the first report of a pulmonary mycotic infection in a sea turtle diagnosed using immunohistochemistry. Wildlife rehabilitators and pathologists should be aware of the availability of immunohistochemical techniques for identifying fungi in sea turtles.  相似文献

Six Thoroughbred mares developed vulvo vaginal candidiasis following oral administration of altrenogest, a synthetic progestogen, 12.5 mg/day for fifteen days. Vaginal congestion, whitish discharge and intertrigo were observed. Candida albicans was cultured from the vagina and the vulva of all six mares. Intravaginal administration of clotrimazole, 500 mg/day for five consecutive days, resulted in rapid disappearance of the vaginal symptoms, healing of the cutaneous lesions and negative cultures. Upon clearing of the infection, the mares were returned to normal breeding practices. Five of them became pregnant within three months after completion of the anticandida treatment.  相似文献

Abstract

CASE HISTORY: A sub-adult female olive ridley sea turtle (Lepidochelys olivacea) was found floating off the coast of Gran Canaria, Canary Islands, Spain.

CLINICAL AND PATHOLOGICAL FINDINGS: The turtle was lethargic and weak, and died one day after it was found. Necropsy was performed and selected samples of lung and liver were routinely processed for histopathological and immunohistological studies. Grossly, round, ulcerative lesions were observed on the rear flippers. The liver, spleen, coelomic wall, gastric serosa, lungs and urinary bladder were covered by various amounts of fibrin and small granulomas. Histopathologically, a severe fibrinous and granulomatous polyserositis (liver, spleen, stomach, urinary bladder, coelomic wall), severe fibrinous pleuritis and severe multifocal granulomatous pneumonia were diagnosed. Numerous fungal hyphae were observed associated with the lesions. Specific monoclonal and heterologously absorbed polyclonal antibodies served as the primary reagents for identification of aspergillosis, candidiasis, fusariosis, geotricosis, dermatophytosis, scedosporiosis, and zygomycosis. All hyphae were identified as Trichophyton spp., because a strong and uniform reactivity was obtained only with an heterologously absorbed polyclonal antibody raised against somatic antigens of Trichophyton spp.

DIAGNOSIS: Systemic mycosis caused by Trichophyton spp.

CLINICAL RELEVANCE: There are no known previous reports of Trichophyton spp. causing skin disease or systemic mycotic infection in sea turtles. The usefulness of immunohistochemistry for identifying fungi in sea turtles is reinforced.  相似文献

AIMS: To determine factors that may influence the efficacy of an oral pelleted vaccine containing Mycobacterium bovis bacille Calmette-Guérin (BCG) to induce protection of brushtail possums against tuberculosis. To determine the duration of protective immunity following oral administration of BCG.

METHODS: In Study 1, a group of possums (n=7) was immunised by feeding 10 pellets containing dead Pasteur BCG, followed 15 weeks later with a single pellet of live Pasteur BCG. At that time, four other groups of possums (n=7 per group) were given a single pellet of live Pasteur BCG orally, a single pellet of live Danish BCG orally, 10 pellets of live Pasteur BCG orally, or a subcutaneous injection of live Pasteur BCG. For the oral pelleted vaccines, BCG was formulated into a lipid matrix, and each pellet contained approximately 107 colony forming units (cfu) of BCG, while the vaccine injected subcutaneously contained 106 cfu of BCG. A sixth, non-vaccinated, group (n=7) served as a control. All possums were challenged by the aerosol route with a low dose of virulent M. bovis 7 weeks after vaccination, and killed 7–8 weeks after challenge. Protection against challenge with M. bovis was assessed from pathological and bacteriological findings.

In Study 2, lipid-formulated live Danish BCG was administered orally to three groups of possums (10–11 per group), and these possums were challenged with virulent M. bovis 8, 29 or 54 weeks later. The possums were killed 7 weeks after challenge, to assess protection in comparison to a non-vaccinated group.

RESULTS: The results from Study 1 showed that vaccine efficacy was not adversely affected by feeding dead BCG prior to live BCG. Feeding 10 vaccine pellets induced a level of protection similar to feeding a single pellet. Protection was similar when feeding possums a single pellet containing the Pasteur or Danish strains of BCG. All vaccinated groups had significantly reduced pathological changes or bacterial counts when compared to the non-vaccinated group. In Study 2, oral administration of Danish BCG induced protection against challenge with M. bovis, which persisted for at least 54 weeks after vaccination. Some protection was observed in possums challenged 54 weeks after vaccination, but this protection was significantly less than that observed in groups vaccinated 29 or 8 weeks prior to challenge. There was a strong relationship between the proportion of animals producing positive lymphocyte proliferation responses to M. bovis antigens and protection against challenge with M. bovis.

CONCLUSIONS: Factors considered potentially capable of interfering with vaccination, including feeding dead BCG to possums prior to feeding live BCG, feeding multiple doses of BCG at one time, and changing strains of BCG, were shown not to interfere with the acquisition of protective immune responses in possums. Protection against tuberculosis was undiminished up to 29 weeks after vaccination with BCG administered orally. It is concluded that vaccination of possums by feeding pellets containing BCG is a robust and efficient approach to enhance the resistance of these animals to tuberculosis.  相似文献

1. The aim of the present study was to determine if the 9R-strain of the Salmonella Gallinarum live vaccine was responsible for having fowl typhoid outbreaks in chicken flocks from both chicken and turkey breeders as well as to verify the antimicrobial resistance of the isolates from the outbreaks.

2. The triplex polymerase chain reaction, standard antimicrobial test, beta-lactamase genes identification and Ion Torrent PMG whole-genome sequence were used in the field isolates and in the vaccine strain of S. Gallinarum.

3. The 60 tested isolates were not from vaccine origin and manifested high resistance to drugs from macrolide and quinolone groups. Whole-genome sequencing (WGS) and single nucleotide polymorphism analysis on selected isolates for core genes from Salmonella enterica confirmed the wild origin of these isolates and showed two possible sources of S. Gallinarum in the studied outbreaks.

4. S. Gallinarum isolated from fowl typhoid outbreaks in the studied period were not caused by the use of the SG9R live vaccine. The source of strains sequenced was diverse.  相似文献

The increasing incidence of candidiasis has drawn the attention of scientists and clinicians attempting to improve methods of studying Candida yeasts. PCR amplification followed by agarose gel electrophoresis (PCR-AGE) and the manual method (morphological characteristics, biochemical profiles and culturing on CHROMagar-Candida) and VITEK 2 automated method were used to test a total of 30 fungal strains from dog sources. The strains were obtained from cases of dermatitis, otitis externa and from the ears, oral mucosa, vaginal mucosa, prepuce and perianal region of clinically normal dogs. After identification as Candida yeasts by the manual method, the strains were analyzed using both VITEK and PCR-AGE methods. Isolates of C. parapsilosis ATCC 22019, C. krusei ATCC 6258 and C. albicans ATCC 10231 were included as controls. The universal primers ITS1, ITS3 and ITS4 were used in two independent PCR reactions. Of 30 yeast isolates, 3 isolates (Saccharomyces cerevisiae, C. rugosa and C. parapsilosis) that were incompletely identified by the manual method were identified with the PCR-AGE and VITEK methods. The results revealed a 96.7% and 86.7% concurrent identification between the PCR-AGE and VITEK methods versus the manual method, respectively. PCR-AGE showed a greater level of concordance with the manual method, besides being faster and more sensitive than the other methods examined, and is therefore indicated for routine diagnostic testing of Candida spp. strains from veterinary sources.  相似文献

1. In two experiments, no consistent differences in weight gain or food intake appeared between broiler chickens given diets based on whole or rolled barley, but food: gain ratio and gizzard weight as a percentage of live weight were reduced (P<0.05) when barley was rolled. Grit addition did not affect performance or intestinal characteristics.

2. In a third experiment, whole barley diets gave significandy higher weight gain than ground barley diets; this was attributable to the significandy higher food consumption. Gizzard weight as a percentage of live weight increased (P< 0.05) when whole barley was fed, while foodrgain ratio was not significandy (P>0.05) affected by the form of barley.

3. The number of particles in the intestinal contents with sizes larger than 2.4 mm was not significantly (P<0.05) different between broiler chickens fed on whole barley and those fed ground barley, but the number of particles with sizes between 2.4 and 0.7 mm was lower (P< 0.05) when diets with whole barley were fed.

4. Enzyme addition increased (P< 0.05) weight gain and the DM of intestinal contents, and decreased (P<0.05) intestinal viscosity, food:gain ratio and pancreas weight as a percentage of live weight regardless of form of the barley.  相似文献

This study investigated vitamin A compounds in the plasma of healthy free‐ranging Central European raptors with different feeding strategies. Plasma samples of nestlings of white‐tailed sea eagle [white‐tailed sea eagle (WTSE), Haliaeetus albicilla) (n = 32), osprey (Pandion haliaetus) (n = 39), northern goshawk (Accipiter gentilis) (n = 25), common buzzard (Buteo buteo) (n = 31), and honey buzzard (Pernis apivorus) (n = 18) and adults of WTSE (n = 10), osprey (n = 31), and northern goshawk (n = 45) were investigated with reversed‐phase‐high‐performance liquid chromatography (RP‐HPLC). In WTSE, northern goshawks and common buzzards retinol were the main plasma component of vitamin A, whilst in ospreys and honey buzzards, 3,4‐didehydroretinol predominated. The median of the retinol plasma concentration in the nestlings group ranged from 0.12 to 3.80 μm and in the adult group from 0.15 to 6.13 μm . Median plasma concentrations of 3,4‐didehydroretinol in nestlings ranged from 0.06 to 3.55 μm . In adults, northern goshawks had the lowest plasma concentration of 3,4‐didehydroretinol followed by WTSE and ospreys. The plasma of all investigated species contained retinyl esters (palmitate, oleate, and stearate). The results show considerable species‐specific differences in the vitamin A plasma concentrations that might be caused by different nutrition strategies.  相似文献

1. This study was conducted to determine the utility of a volatile oil (VO) mixture in quail diets as a natural growth promoter. Different levels of VO mixtures, derived from rosemary and oregano, were added to a basal diet to determine the effects of the mixture on live weight (LW), live weight gain (LWG), feed intake, feed conversion ratio (FCR), carcase yield (CY), lipid oxidation level in thigh meat samples, and blood constituents.

2. A total of 880 one-day-old Pharaoh (Coturnix coturnix Pharaoh) quails, including both males and females, were divided into 4 groups containing 220 quails and treated as follows: (1) a control treatment with 0?mg VO/kg of diet; (2) 100?mg/kg rosemary VO plus 100?mg/kg oregano VO (50:50%); (3) 140?mg/kg oregano VO plus 60?mg/kg rosemary VO (70:30%); and (4) 60?mg/kg oregano VO plus 140?mg/kg rosemary VO (30:70%). The diets were prepared fresh for each treatment. The experiment was carried out for 42?d.

3. The results of the study showed that the highest concentration of rosemary oil (140?mg/kg) caused a significant increase in live weight, live weight gain and carcase yields during the growing and finishing periods. However, feed intake and FCR were not significantly influenced by treatments.

4. The quails fed with rations containing the VO mixture derived from rosemary and oregano had reduced thiobarbituric acid levels (TBA) in raw breast meat samples at different storage times. There is possibly a synergistic effect between oregano and rosemary volatile oils in preventing lipid oxidation in stored meat.

5. In this study, the haemoglobin (PCV) and haematocrit values and the heterophile/lympohocyte (H/L) ratio increased in the blood samples taken from Treatment 2.

6. In conclusion, a volatile oil containing a mixture of rosemary and oregano oils could be a potential natural growth promoter for quails, depending on the plants from which the VOs were extracted, the dosage and the synergetic effects of the mixture.  相似文献

The aim of this study was to compare the ability of a live incomplete strain (Strain 48) and a live complete strain (Strain 89) of Toxoplasma gondii to protect against abortion and congenital infection following an oral challenge of T. gondii oocysts. Sixty-nine two-tooth ewes were immunised pre-tupping with live Strain 48 of T. gondii tachyzoites and seventy ewes were immunised with Strain 89. Eighty-two serologically negative ewes served as controls. At mid-pregnancy half of the ewes were challenged orally with T. gondii oocysts (2×105/ewe).

The ewes vaccinated with Strain 48 were significantly (p<0.05) protected against the effects of experimental challenge and the rate of congenital infection was also significantly (p<0.15) reduced. The ewes vaccinated with Strain 89 were also significantly (p<0.05) protected.

The serological response to challenge as measured by both the Dye test and the Indirect Haemagglutination test varied considerably between the two vaccinated groups.  相似文献

Summary

Egg transmission of egg drop syndrome 1976 virus (BC14 virus) in fowl was demonstrated in the second and third week after experimental infection. Eggs of BC14 virus infected hens were incubated weekly after disinfection with formaline gas. After 18 days of incubation, eggs with live embryos were homogenized. This egg material was fed to adult hens, housed in isolators.

Seroconversion in these birds demonstrated egg transmission.

It is suggested that egg transmission occurs as a result of viremia.  相似文献

1. The feeding value of sweet White Lupin (Lupinus albus variety Buttercup) for Pekin (Cherry Valley) ducklings was evaluated in isocaloric and isonitrogenous diets formulated to contain 0, 100, 200, 300 and 400 g/kg lupins. Male and female ducklings were housed together and fed on starter diets from 0 to 3 weeks and finisher diets from 4 to 7 weeks.

2. Inclusion of sweet lupins at 400 g/kg diet (containing less than 0.1 g/kg alkaloids) had no deleterious effects (P< 0.05) on live weight, efficiency of food utilisation, dressed carcase weight and carcase fat content.  相似文献

  1. A total 120 laying hens (60 TETRA BLANCA white egg layers and 60 TETRA SL brown egg layers) were selected from 250 TETRA BLANCA and 250 TETRA SL pullets based on their predicted body fat content by means of computed tomography (CT) at 16 weeks of age. Three groups of pullets were chosen for the investigation with the highest (n = 20), lowest (n = 20) and average (n = 20) body fat content.

  2. Changes in the live weight, body fat content, egg production and egg composition of the chosen animals were recorded at 32, 52 and 72 weeks of age.

  3. Based on the results, it was established that differences in starting body fat content of the hens remained the same during the experimental period. The differences between the two extreme groups were statistically significant at each age.

  4. The starting body fat content of the hens affected the rate of egg production, i.e. hens with high starting body fat content produced 11–14 eggs fewer than the hens with a low or average body fat content but had no effect on the composition of the eggs.

  5. Genotype affected almost all of the examined traits: TETRA BLANCA hens had lower live weight and higher body fat content during the experimental period and produced fewer eggs with lower albumen and higher yolk, dry matter and crude fat content than the TETRA SL hens.
