There has been no analysis of a hopping‐type forelimb lameness syndrome seen in ridden horses. The objectives of this retrospective study were to describe the clinical features of this syndrome, response to diagnostic analgesia and imaging findings and to document post mortem findings. Clinical records from 2002 to 2014 were reviewed and data concerning signalment, history, lame limb(s), lameness characteristics, response to diagnostic analgesia and diagnostic imaging were recorded. There were 46 horses from 4 to 13 years of age, 6 of which had a history of known or suspected trauma immediately before the onset of reduced performance or lameness. Lameness seen when ridden was characterised by an intermittent shortened cranial phase of the step of the lame forelimb at the trot and marked elevation of the head as the affected limb was protracted, with the horse appearing to ‘hop’ (on the contralateral limb) as if trying to break to canter. When lameness was at its worst horses were unwilling to trot. Three horses showed sporadic severe stumbling. Local analgesia of the affected limb did not improve the lameness and in 16 horses lameness deteriorated. Three of 5 horses showed some improvement (≥2/8 grades) in the hopping‐type lameness after intra‐articular analgesia of the articular process joints of the sixth and seventh cervical vertebrae, ipsilateral to the lame forelimb. Radiographic, ultrasonographic and nuclear scintigraphic examinations were inconclusive. Two of 4 horses responded to treatment with gabapentin. In 3 horses post mortem examination revealed mild lymphocytic inflammation within or around the dorsal root ganglia of the fifth and sixth cervical nerve roots, sixth cervical nerve root or second thoracic nerve root ipsilateral to the lame limb. Idiopathic hopping‐type lameness syndrome in ridden horses may be a pain‐related condition ± a neurological component and currently has a guarded prognosis.  相似文献

Hyperkalaemic periodic paralysis (HYPP) is a relatively new genetic disease that, so far, appears to be confined to descendants of the American Quarter Horse Impressive, a stallion identified as the link between the pedigrees of all affected horses, and that predominantly affects Quarter Horses but also Paint horses, Appaloosas and Palominos. This study suggests that an episode of muscle weakness, which was reported in a Criollo Argentino genetically unrelated to Impressive, was related to marked hyperkalaemia. The symptoms in this case were reproducible following a KCI challenge test although DNA testing for HYPP was negative. Based on these findings, a diagnosis of a HYPP‐like syndrome was made.  相似文献

Routine necropsies of 27 asymptomatic juvenile chinchillas revealed a high prevalence of gastric ulcers with microscopic lymphoplasmacytic gastroenteritis and typhlocolitis. Polymerase chain reaction (PCR) analysis using Campylobacter genus‐specific partial 16S rRNA primers revealed the presence of Campylobacter spp. DNA in the faeces of 12 of 27 animals (44.4%). Species‐specific partial 16S rRNA PCR and sequencing confirmed that these animals were colonized with Campylobacter lanienae, a gram‐negative, microaerophilic bacterium that was first identified on routine faecal screening of slaughterhouse employees and subsequently isolated from faeces of livestock. Campylobacter lanienae was isolated from the faeces of six PCR‐positive animals and identified with species‐specific PCR and full 16S rRNA sequencing. Phylogenetic analysis showed that these isolates clustered with C. lanienae strain NCTC 13004. PCR analysis of DNA extracted from gastrointestinal tissues revealed the presence of Clanienae DNA in the caecum and colon of these chinchillas. Gastrointestinal lesions were scored and compared between C. lanienae‐positive and C. lanienae‐negative animals. There was no correlation between colonization status and lesion severity in the stomach, liver, duodenum, or colon. Possible routes of C. lanienae infection in chinchillas could include waterborne transmission and faecal–oral transmission from wild mice and rats or livestock. Based on these findings, the authors conclude that C. lanienae colonizes the lower bowel of chinchillas in the absence of clinical disease. This is the first report of C. lanienae in any rodent species. Campylobacter lanienae isolates from different mammalian species demonstrate heterogeneity by 16S rRNA sequence comparison. Analysis using rpoB suggests that isolates and clones currently identified as C. lanienae may represent multiple species or subspecies.  相似文献

The endozepine‐like peptide (ELP) is a testis‐specific isoform of the acyl‐CoA binding protein (ACBP) and shares the latter’s peptide motif for binding mid‐long chain acyl‐CoA groups. ELP is expressed both as mRNA and protein at high levels in the testes of a wide range of mammals, including rodents, carnivores and ruminants. However, the ELP gene is progressively inactivated through primate evolution, with no protein detectable in a range of primates studied, including human. In nonprimate species, ELP is expressed in very late postmeiotic germ cell stages only, such that its function in these species is probably associated with the metabolism of the mature spermatozoon. Current research is looking at both the function of the ELP protein and the haploid regulation of the gene.  相似文献

Influences of a specific dietary nutrient on glucagon‐like peptide (GLP)‐1‐containing cells in the chicken intestine are not yet clear. Significance of dietary protein level on GLP‐1‐containing cells in the chicken ileum was investigated. Chickens fed control or experimental diets of varying protein levels were examined using immunohistochemical and morphometrical techniques. We show that the protein ingestion had an impact on the activities of GLP‐1‐immunoreactive cells in the chicken ileum. Weight gains declined with decreasing dietary crude protein (CP) levels, but no significant differences were detected in the daily feed intake and villous height. GLP‐1‐immunoreactive cells with a round or oval shape were frequently observed in the lower CP level groups (4.5% and 0%). Frequencies of occurrence of GLP‐1‐immunoreactive cells were 41.1 ± 4.1, 38.5 ± 4, 34.8 ± 3.1 and 34.3 ± 3.7 (cells/mm2, mean ± SD) for dietary CP level of 18%, 9%, 4.5% and 0% groups, respectively and significant differences were recognized between the control and lower CP level groups (P < 0.05). Multiple regression analysis indicated a significant correlation between the daily protein intake and frequencies of occurrence of GLP‐1‐immunoreactive cells. The protein ingestion is one of the signals that influence GLP‐1‐containing cells in the chicken small intestine.  相似文献

Insulin‐independent actions of glucagon‐like peptide‐1 (GLP‐1) are not yet clear in ruminants. Four Suffolk mature wethers (60.0 ± 6.7 kg body weight (BW)) were intravenously infused with insulin (0.5 mU/kg BW/min; from 0 to 90 min) and GLP‐1 (0.5 μg/kg BW/min; from 60 to 150 min) with both hormones co‐administered from 60 to 90 min, in a repeated‐measure design under euglycemic clamp for 150 min, to investigate whether GLP‐1 has insulin‐independent actions. Jugular blood samples were taken at 15‐min intervals for plasma hormones and metabolites analysis. Compared to baseline concentrations (at 0 min), insulin infusion decreased (P < 0.05) plasma concentrations of glucagon, non‐esterified fatty acids (NEFA), lactate, nonessential amino acids (NEAA), branched‐chain amino acids (BCAA), total amino acids (TAA) and urea nitrogen (UN). Insulin plus GLP‐1 infusion induced a greater increase (P < 0.05) in plasma concentrations of insulin and triglyceride (TG), but decreased (P < 0.05) glucagon, total cholesterol (T‐Cho), NEAA and UN plasma concentrations. GLP‐1 infusion increased (P < 0.05) NEFA, β‐hydroxybutyrate and TG, but decreased (P < 0.05) glucagon, T‐Cho, NEAA, BCAA and UN plasma concentrations. In conclusion, GLP‐1 exerts extrapancreatic roles in ruminants not only insulin‐independent but probably, in contrast to non‐ruminants, antagonistic to insulin effects.  相似文献

Two male, neutered, domestic, shorthaired cats were evaluated for progressive paresis and ataxia. Neurological examinations suggested a spinal cord lesion in each case. Complete blood examination and cerebrospinal fluid analysis were unremarkable in both cats. MRI revealed malformation of the occipital bone with herniation of the cerebellar vermis through the foramen magnum but without syringomyelia. Chiari‐like malformation was suspected in both patients. MRI repeated one year later in both cats because of progression of clinical signs yielded the same findings as the initial scans. Foramen magnum decompression in one cat was associated with resolution of clinical signs.  相似文献

This study examined the effects of supplementation of ES‐like cell culture medium with bone morphogenetic protein (BMP)‐4 (0, 10, 20 or 100 ng/ml) or Noggin (250, 500 or 750 ng/ml) or TGF‐β1 (0, 0.1, 1 or 10 ng/ml) or SB431542 (0, 10, 25 or 50 μm ), an inhibitor of TGF‐β1 signalling, on survival, colony area and expression level of pluripotency genes in buffalo ES‐like cells at passage 40–80, under different culture conditions. BMP‐4 supplementation significantly reduced (p < 0.05) colony survival rate, percentage increase in colony area and relative mRNA abundance of OCT4, whereas that of NANOG and SOX‐2 was increased significantly (p < 0.05). Noggin supplementation did not affect the colony survival rate and percentage increase in colony area in the presence of FGF‐2 and LIF. In the presence of FGF‐2 alone, it significantly reduced (p < 0.05) the relative mRNA abundance of OCT4 and SOX‐2 and increased (p < 0.05) that of NANOG. Supplementation with TGF‐β1 at 1.0 ng/ml but not at other concentrations increased colony survival rate but had no effect on percentage increase in colony area at any concentration. Supplementation with SB‐431542 decreased (p < 0.05) colony survival rate at 50 μm but not at other concentrations. The percentage increase in colony area was lower (p < 0.05) with 10 μm SB‐431542 than that in the controls, whereas at higher concentrations of 25 or 50 μm , SB‐431542 decreased (p < 0.05) the colony size instead of increasing it. In conclusion, these results suggest that BMP‐4 induces differentiation in buffalo ES‐like cells, whereas TGF‐β/activin/nodal pathway may not be playing a crucial role in maintaining pluripotency in these cells.  相似文献

This study was performed to determine the localization strategies of Toll‐like Receptor 4 (TLR4) in digestive tract (oesophagus, bulbodium, foregut, midgut and hindgut) of Blunt snout bream (Megalobrama amblycephala) using immunohistochemical staining method. TLR4 positive cells were observed throughout the digestive tract. In the oesophagus, some positive reactions in lamina propria were found around small blood vessels and there were also some positive cells within the stratified squamous epithelium. Lots of positive cells were observed in the muscular layer of the oesophagus. In bulbodium, foregut and hindgut, the expression of TLR4 was mainly restricted to the apical surface of epithelial cells located at the bottom of the mucosal folds and the mesenchymal cells in lamina propria. It was very interesting that epithelial cells in the midgut, but none in other parts, had many TLR4 positive cytoplasmic granular structures which were also periodic acid Schiff positive. These findings suggested that TLR4 was expressed in a compartmentalized manner in the Blunt snout bream (M. amblycephala) digestive tract and provided novel information about the in vivo localization of pattern recognition receptors.  相似文献
