对油菜雄性不育性的分子机理研究作了综合评述。认为油菜的雄性不育性一细胞器DNA结构发生变异,以及线粒体基因重组有关,但与质粒无关。此外,雄性不育与与atp,cox基因的调控,蛋白质、酶变化等有关。已发现油菜的不育性很不稳定,易受温度的影响。  相似文献

研究了大豆分离蛋白的碱性蛋白酶和木瓜蛋白酶水解产物各种加工功能性质的变化趋势,并比较了两种酶在各个不同酶解时间下水解产物的各种功能性质。结果表明:大豆分离蛋白酶解产物比大豆分离蛋白的溶解性、乳化性、发泡能力和吸油性都有所改善;大豆分离蛋白木瓜蛋白酶水解产物保水性、乳化性、吸油性、发泡能力、粘度、凝胶性均优于大豆分离蛋白碱性蛋白酶水解产物。  相似文献

对花生采用水相酶解法制油与蛋白质提取,与水剂法相比能显著提高油反蛋白质得率;在对不同的酶对比使用后发现,使用包含纤维素酶、果胶酶、蛋白酶的复合酶效果比单一酶好;经优化实验得出酶处理的最适参数为:加酶量0.3%,酶反应时间4h、pH6.4、温度49℃,此条件下,花生油反花生蛋白质得率可分别达95.4%和74.6%。  相似文献

稻米中蛋白质对淀粉RVA特征谱的影响   总被引:10,自引:3,他引:7  
通过分析蛋白酶或还原剂二巯基苏糖醇(DTT)分别去除稻米蛋白质或打破蛋白质二硫键后,稻米RVA(Rapid Visco Analyzer)特征谱形成和特征值的变化,研究蛋白质对淀粉RVA特征谱的影响,以及对米饭食味品质的作用。在淀粉糊化过程中,蛋白质使RVA特征谱的峰值黏度、冷胶黏度等值升高,改变RVA线性上升段的斜率。蛋白质可能通过二硫键结合形成的蛋白质网络本身和通过吸水减少淀粉水合的有效水量,协同提高糊化多相体系的浓度,增强分散相与黏稠相的互作。蛋白质影响米饭食味品质,可能通过改变米粒的吸水性而起作用。  相似文献

为探究小麦种子休眠解除期间的成分变化对发芽率的影响,了解种子休眠解除机制,以铭贤169为材料,在收获后不同储藏时间(间隔20 d)取样,通过显微设备观察了种子后熟期间的结构变化,测定了相关生理指标和成分,以探究铭贤169种子休眠性原因。结果表明,铭贤169种子休眠解除过程中,种皮细胞结构、组织结构、淀粉粒结构、蛋白基质等均发生变化,淀粉含量、粒径、糊化特性、热稳定性随贮藏时间延长变化显著。铭贤169的离体胚在花后25 d就具有发芽能力,但种子在花后35 d仍处于休眠。收获后熟期间,种胚活性、种子吸水率、含水量、淀粉含量、淀粉糊化谷值粘度和峰值时间、热稳定性逐步上升,与发芽率正相关。A型淀粉粒表面破损,淀粉粒与蛋白质结合紧密,种皮结构松散;淀粉粒径、淀粉酶活性、可溶性糖含量和沉降值减小;面筋和粗蛋白含量变化不大。推测铭贤169种子的休眠性与种子母体成分有关,休眠解除期间发芽率变化受种皮结构、淀粉的分解程度、淀粉粒与蛋白质的结合方式、可溶性糖含量、淀粉含量和性质以及淀粉酶活性等因素影响。  相似文献

制取低成本、高蛋白含量的大豆浓缩蛋白时,乙醇产生变性作用,从而降低了大豆浓缩蛋白的功能特性,因此本研究采用碱性蛋白酶对醇法大豆浓缩蛋白进行改性.通过对酶浓度、底物浓度、改性时间与改性温度的单因素实验,针对起泡性进行研究,然后进行正交实验,最终得出碱性蛋白酶提高醇法大豆浓缩蛋白起泡性最佳工艺条件:酶用量(E/S)为6%、底物浓度为(W/V)8%、酶解时间为2h、酶解温度为50℃,在此条件下测得的起泡能力为113.3%,起泡稳定性为56.7%,分别提高到酶改性前的2.5、3.6倍.  相似文献

蛋白质影响水稻米饭食味品质的机理   总被引:5,自引:1,他引:4  
 以杂交中籼丰两优1号组合为材料,设置不同的播期和收获期,分析稻米主要品质指标(直链淀粉、总蛋白含量、RVA特征值等)和米饭食味品质,并用二硫苏糖醇 (DTT)或蛋白酶(Trypsin)分别打破样品蛋白质二硫键或酶解蛋白质后,观察RVA特征谱和膨胀势的变化,旨在明确蛋白质对不同食味品质样品RVA的影响,进而探索蛋白质影响米饭食味品质的机理。结果表明,第3播期(6月13日)稻米品质主要指标(直链淀粉、总蛋白、膨胀势、RVA特征值)与第4播期(6月23日)样品差异不显著,但食味品质明显好于后者;分别经蛋白酶处理后,第3和第4播期样品RVA上升段斜率差异不显著,但前者后段冷却后回升斜率明显小于第4播期。由于影响RVA特征谱后段凝胶刚性的主要因素是淀粉与水之比,刚性随淀粉凝胶中含水量减少而提高,食味品质较好的第3播期米粉经蛋白酶处理后,凝胶刚性小且减弱程度大,这可能是蛋白质酶解后形成蛋白质碎片的持水能力弱,即淀粉与水之比低,由此推测蛋白质影响米饭食味品质可能在米饭蒸煮后阶段,即可能与蛋白质水合后持水能力差异有关。  相似文献

以豆粕粉为原料,加入高效蛋白酶和胰蛋白酶进行水解研究,对反应温度、底物浓度、反应时间、pH值、酶的用量及酶加入间隔的时间等工艺参数进行优化,并分析了大豆多肽的含量及蛋白质的水解度与各影响因素之间的关系.经过优化得到的反应条件为温度45℃,pH 7.5,液固比7: 1,加酶量为高效蛋白酶和胰蛋白酶各0.2 g,反应时间8 h,加酶间隔时间3 h.在此条件下,酶解后的蛋白质含量达85%、水解度为17.72%.  相似文献

利用中性蛋白酶Protex 7L对大豆分离蛋白进行改性研究,以水解度和蛋白质分散指数为评价指标,确定了中性蛋白酶Protex 7L改性大豆分离蛋白的最佳酶解反应条件:加酶量13.5AU/g大豆分离蛋白(SPI)、反应温度55℃、底物浓度为10%、pH值为7.0,酶解时间为1h.在上述条件下大豆分离蛋白的分散性得到显著改善,蛋白质分散指数(PDI值)达到91.8%.  相似文献

为了除去油茶粕蛋白的色素,添加体积分数为4%的双氧水在40℃下作用蛋白提取液1h可取得较好的脱色效果,但双氧水脱色对油茶粕蛋白结构和功能的影响需要进一步研究。本文通过测定变性温度、表面疏水性和巯基的含量,分析红外光谱、X射线衍射峰、微观形貌等研究脱色前后油茶粕蛋白结构和功能的变化。结果显示:双氧水脱色使油茶粕蛋白的表面疏水性上升,巯基含量下降,二硫键含量基本不变;通过红外光谱分析,双氧水脱色减弱了油茶粕蛋白化学键强度,但没有影响油茶粕蛋白基本骨架,微观结构基本相似,油茶粕蛋白的X射线衍射峰的位置和强度基本一致,热变性温度升高,双氧水没有改变油茶粕蛋白的等电点,但油茶粕蛋白的溶解度、乳化性、乳化稳定性、起泡能力和泡沫稳定性均下降,持油性和持水性上升。本研究结果表明双氧水脱色未明显改变油茶粕蛋白的结构,但其功能性质受到一定影响。  相似文献

The rheological properties of wheat flour under processing such as extrusion (with 28% moisture content, wet basis) are influenced by the molecular changes its components undergo during processing. But, there was no simple relationship between the wheat-flour characteristics and their rheological properties. In order to investigate the quantitative and qualitative effects of the individual flour components on rheological properties, model blends of wheat starch and wheat gluten with different starch/gluten ratios were studied. The effects of gluten and starch quality were also investigated by using different gluten types and by modifying the amylose content of starch, respectively. The shear viscosity of the blends, determined by capillary rheometry under controlled conditions (35% moisture content, 140 °C), was observed to be modified by both gluten and amylose content. The changes undergone by wheat gluten under these conditions were analysed by HPLC, to determine the levels of unextractable polymeric proteins, and by Lab-on-a-Chip analysis of protein composition, to follow the polymerisation of protein under processing. This study indicated that in low hydrated products in the molten state, shear viscosity is affected by the structure of the blends as determined by fluorescence microscopy and by the molecular changes occurring during processing.  相似文献

The main aim of this study was to determine the changes in free sulphydryl content during postharvest wheat and flour maturation. The content of free sulphydryl groups was determined from wet gluten over 50 days of wheat postharvest maturation and over 14 days of flour maturation by varying incubation temperatures (30 and 37 °C) and incubation times (0, 45, 90 and 135 min). The amount of free sulphydryl groups increased during postharvest wheat and flour maturation as well as with the increase in temperature and gluten incubation time. The additional aim of this study was to find the interrelation between the content of free sulphydryl groups and selected parameters of technological quality by means of Principal Component Analysis. During wheat/flour maturation, the strengthening of protein structure was observed manifested by the increase in gluten index and Mixolab protein network weakening (C2), and changes in dough resistance as well. The incubation temperature of 37 °C affected the weakening of protein structure manifested by the decrease in gluten index and resistance, and increase in extensibility. Strong differentiation of examined varieties in terms of selected quality indicators was observed upon pre-defined maturation period, probably due to the expression of their intrinsic properties.  相似文献

Gliadin and glutelin are major rice storage proteins. In this study, we evaluated changes in these proteins to determine the influence of pasting properties on rice storage. Notably, the physical and chemical properties of these proteins changed steadily. We analyzed protein oxidation indexes and determined correlations between the protein oxidation index and gelatinization property index. The results showed that significant oxidation of gliadin and glutelin occurred during the process of rice storage (P < 0.05). Protein oxidation had a significant impact on pasting properties. For glutelin, changes in the structure and function of the protein during rice storage had a significant impact on the five rice pasting property indexes, including peak viscosity (PV), holding strength (HS), breakdown, final viscosity (FV), and setback (SB; P < 0.05). In particular, the levels of carbonyl compounds and active sulfur and the surface hydrophobicity of glutelin were significantly correlated with rice pasting property indexes (P < 0.05). However, gliadin only significantly affected three indexes, i.e., PV, HS, and FV (P < 0.05). Thus, these findings suggested that the carbonyl compounds, active sulfur, and surface hydrophobicity of glutelin could be used as sensitive indexes for changes in rice quality evaluation.  相似文献

This paper presents a novel concept for making an elastic dough using a structured protein suspension. The idea behind it is based on the hypothesis that a number of gluten properties originate from a particle structure present in the gluten network. Three different mesoscopically structured whey protein suspensions were produced: whey protein aggregates, a whey protein cold set gel and whey protein particles. Dough mixtures or batters were prepared by mixing the structured protein particle suspension with starch. Farinograph curves, small and large deformation experiments showed that the presence of a mesoscopic protein structure had a large impact on the properties of gluten-free starch mixtures. The whey protein that was structured into a mesoscopic particle suspension changed the starch mixture from a liquid into a cohesive material, having strain hardening properties.  相似文献

为探索白菜型冬油菜抗旱机理,本研究采用双向电泳结合液相色谱-质谱技术分析抗旱冬油菜品系DR-5差异蛋白质组变化,克隆其差异蛋白热激同源蛋白70(heat shock cognate protein 70)基因HSC70-1,研究干旱胁迫对白菜型冬油菜该基因表达的影响。质谱鉴定出23个差异蛋白质,分别参与刺激响应(5)、糖/能量代谢(6)、脂代谢(2)、信号转导(1)、氨基酸/蛋白质代谢(2)、核酸代谢(1)、细胞骨架(1)、光合作用(2)、伴侣蛋白(3)。同源克隆得到1 911bp的HSC70-1基因完整开放阅读框,编码637个氨基酸残基。所编码的蛋白HSC70是稳定的疏水性蛋白质,含有HSP蛋白家族特有的核酸结合功能区及底物结合功能区,有4个活性口袋,氨基酸序列与白菜型油菜(Brassica rapa)HSC70氨基酸序列相似度达98%。实时荧光定量和半定量分析HSC70-1基因干旱胁迫下表达,发现HSC70-1在叶片中上调表达。本研究结果为冬油菜抗旱育种提供了理论基础。  相似文献

Microstructure and rheological properties of mixtures of acid-deamidated rice protein (ADRP) and dextran were studied. The microstructures of the ADRP/dextran mixtures were described using confocal laser scanning microscopy (CLSM), which revealed the effective association among ADRP and formation of a protein network-like structure in the mixture with higher protein concentration. Mechanical properties of the mixtures were observed by rheometer. The steady shear measurements showed a correlation with the CLSM results via a marked increase in the viscosity of the mixtures with protein association. Frequency sweeps further evidenced the build-up of the gelled network-like structure. The differences in fracture forces observed by textural measurements between mixture and single ADRP gel also suggested the difference in microstructures. The formation of network-like structure appeared to have occurred through a phase separation of ADRP and dextran.  相似文献

Inhibition of plant growth by Trinexapac-ethyl, TE, a gibberellin-biosynthesis inhibitor, can produce a shorter stemmed plant, requiring less nutrients and water to grow, while maintaining grain yield. Although TE and other plant growth regulators are commonly used in grain crops, their effects on starch biosynthesis in the grains have not been systematically examined. The changes in the structural and functional properties of starch in grains harvested from TE-treated sorghum (Sorghum bicolor (L.) Moench) were examined, and the results compared with those from the untreated controls. TE treatment had little or no effects on the molecular structures of starch, starch granule morphology, and starch and amylose contents, but increased the protein content of the grains significantly. Consistent with the lack of change in the molecular structure, there were no significant effects on the thermal properties of the starch. The pasting properties of TE-treated sorghum flours, however, showed lower peak viscosity, trough, and final viscosity, which were attributed to their higher protein contents. The TE treatment thus does not have an appreciable effect on the biosynthesis of starch during grain development in sorghum.  相似文献
