Study was carried out to determine the spoilage association of traditionally fermented ugba, product of African oil bean seed. Samples were collected from three markets within Enugu metropolis in Eastern Nigeria. Microbial population of ugba was isolated, characterized and identified. Effect of time on product quality, total viable cell count and individual growth pattern of isolates was evaluated. Microorganisms isolated were Proteus sp., Klebsiella sp., Staphylococcus epidermidis, E. coli, Bacillus licheniformis and Bacillus subtilis. Initial viable cell count increased from 4.6 x 10(8) -6.2 x 10(8), 6.0 x 10(9) -6.9 x 10(9) and 3.9 x 10(8) -5 x 10(8) cfu g(-1) by the 4th day for samples from Mayor, Kenyatta and Garki markets, respectively. Organoleptic changes in texture and colour were seen to be a factor of time of storage. A decline in growth of Proteus sp., E. coli, Klebsiella sp. and S. epidermidis by the 4th day of storage was observed for all samples. Viable cells of B. licheniformis increased from 6.1 x 10(7) -17.7 x 10(7) cfu g(-1), 13.9 x 10(8) -20.0 x 10(8) and 8.7 x 10(7) -15.5 x 10(7) by the 4th day of storage for samples from Mayor, Kenyatta and Garki markets respectively. Growth of B. subtilis increased from 9.2 x 10(7) -19.9 x 10(7), 14.9 x 10(8) -21.2 x 10(8) and 11.5 x 10(7) -17.2 x 10(7) cfu g(-1) for samples from Mayor, Kenyatta and Garki markets respectively. Our results indicate that ugba spoilage is primarily a result of the continued activity of African oil bean seed fermentative organisms B. subtilis and B. licheniformis.  相似文献

The survival of vegetative and sporulated cells of the Bacillus cultures on the seeds of the crop plants was tried in different combinations. One milliliter inoculum with 1 mL adhesive combination or sterile water showed better results followed by 1.5 mL inoculum with 0.5 mL adhesive or sterile water. The population of 5.5x10(5) cfu seed(-1) on black gram, 10.5x10(5) cfu seed(-1) on soybean and 6.5x10(5) cfu seed(-1) on maize were observed after 12 h of incubation in 1 mL sporulated inoculum mixed with 1 mL of rice gruel. The sporulated inoculum along with rice gruel favoured the adherence of the regenerated cells as rice gruel is rich in nutrient content.  相似文献

Indole Acetic Acid (IAA) producing bacterium was isolated from the Rhizosphere soil and identified as Rhizobium sp. and Bacillus sp., Optimization of Indole acetic acid production was carried out at different cultural conditions, such as pH, temperature and substrate with Rhizobium sp., Bacillus sp. and Rhizobium sp., produced higher amount of Indole acetic acid (6.1 mg mL(-1)) than the Bacillus sp., (4.4 mg mL(-1)) at pH 7 and 37 degrees C in the Bengal gram substrate. Partial purification of Indole acetic acid was done by Thin Layer Chromatography (TLC). In conclusion Rhizobium sp., appear to be a suitable soil microorganism for high level of IAA production.  相似文献

For mass production of V. lecanii, three types of cultivation methods including liquid, solid and diphasic production systems were investigated. In the liquid state of production, six media were tested in stationary culture conditions. Among the six media tested, Molasses Yeast Broth (MYB) supported maximum sporulation (8.33 x 10(8) spores mL(-1)) and biomass production (746 mg/100 mL). In the MYB, 4% molasses concentration was found to produce highest spore count (8.56 x 10(8) spores mL(-1)) and biomass (776 mg/100 mL) followed by 5 and 6% molasses. Among the six solid substrates tested, rice grains supported highest spore production (1.14 g/100 g). In diphasic state of production, combination of MYB and rice grains produced the greatest amount of spores, (1.70 g/100 g). Results of this study indicated that diphasic method using MYB as liquid medium and rice as solid substrate are the best method and media for mass production of V1-7 isolate of V. lecanii.  相似文献

To achieve higher titer of rabies virus higher density of host cells will need. In this study, capability of FibraCel disks packed in 500 mL spinner basket versus Cytodex-1 in 500 mL spinner flask was investigated for propagation of Vero cells and PV rabies virus proliferation. Minimal Essential Medium (MEM) + 10% Foetal Calf Serum (FCS) and Virus Production- Serum Free Medium (VP-SFM) +4 mM L-glutamine were used in growth phase and MEM+ 0.2% Bovine Serum Albumin (BSA) and VP-SFM were used in virus production phase. Adapted Vero cells grown in VP-SFM were used in all SFM experiments while batch and stepwise perfusion modes were applied and compared in growth stage. The highest Vero cell density were achieved in the trials with 10 g FibraCel disk in stepwise perfusion mode equal to 6.12 x 10(6) and 5.87 x 10(6) cells mL(-1) in MEM and VP-SFM, respectively while with 2.73 g Cytodex-1 lower density equal to 4.2 x 10(6) and 4.0 x 10(6) cells mL(-1) were achieved. The highest titer of rabies virus and overall virus production rate were resulted in VP-SFM and on 10 g disks equal to 2.9 x 10(7) Fluorescent Focus Unit (FFU) mL(-1) and 0.14 FFU/Cell/h, respectively versus 1.7 x 10(7) FFU mL(-1) and 0.08 FFU/cell/h on cytodex-1 in similar conditions. The second harvest of virus was also satisfactory in experiment with 10 g disks (1.7 x 10(7) FFU mL(-1)) in compare to Cytodex-1 (0.51 x 10(7) FFU mL(-1)). An equal surface area at 6600 and 12000 cm(-2) were provided in all comparable trials with seeding density of 12.5 x 10(3) cells cm(-2). Adapted Vero cells grown in VP-SFM were used in all SFM experiments while batch and stepwise perfusion modes were applied and compared in growth stage.  相似文献

The occurrence of pathogens in the internal parts of vegetables is usually associated with irrigation water or contaminated soil and could pose risk to consumers as the internalised pathogens are unaffected by external washing. This study was carried out to assess the rate of internalisation of microbes in common Ghanaian vegetables. Standard microbiological methods were employed in microbial enumeration of vegetables collected at the market and farm levels, as well as irrigation water and soil samples. The overall mean counts of vegetables were 4.0 x 10(3) cfu g(-1); 8.1 x 10(2) cfu g(-1); 2.0 x 10(2) cfu g(-1); 3.5 x 10(2) cfu g(-1) for total bacteria, coliform counts, faecal coliform counts and yeast counts, respectively. The rate of internalisation of coliforms in vegetables irrigated with stream/well water was 2.7 times higher than those irrigated with pipe water. The mean coliform counts (4.7 x 10(7) cfu g(-1)) and faecal coliform counts (1.8 x 10(6) cfu g(-1)) of soil samples were similar to those of stream water suggesting both sources exerted similar contamination rates on the vegetables. Generally, there were no significant variations between the rates of internalisation of microbes at the market and farm levels at p < 05, indicating that internalisation of microbes in the vegetables mainly occurred at the farm level. The study has shown that microbial contamination of vegetables in Ghana is not limited to the external surface, but internal vegetable parts could harbour high microbial loads and pose risk to consumers. Safety practices associated with the commodity should therefore not be limited to external washing only. There is the additional need of heating vegetables to eliminate microbes both externally and internally before consumption.  相似文献

Corn distillers dried grains with solubles (DDGS) were fermented with bacteria or yeast. The microbial growth on three DDGS media-unmodified DDGS, enzyme-pretreated DDGS, and supplemented DDGS - was compared with standard media. Comparative growth profile indicated peaks in microbial growths at different times. The highest specific growth rates (SGR) of 0.6, 0.54 and 0.51 h−1 were observed for Saccharomyces Boulardii on unmodified DDGS, yeast extract, and enzyme-modified DDGS, respectively. Pediococcus Acidilactici and Lactobacillus Plantarum had the SGR of 0.17 and 0.07 h−1, respectively, on control DDGS. However, L. Plantarum on unmodified DDGS demonstrated the highest cell count (2.91 × 1011 CFU/mL), followed by S. boulardii at 1.5 × 109 CFU/mL on YEPD, and P. acidilactici at 1.16 × 1010 CFU/mL on MRS (de Man, Rogosa and Sharpe) media. Protein digestibility of DDGS improved slightly due to fermentation, and resulted in reduced phytic acid content for some treatments. Production of lactic acid was highest on unmodified DDGS.  相似文献

A survey was conducted during January to December 2006 to assess the status of faunal diversity of Savar Upazila, Dhaka, Bangladesh. A total of 30 species of birds, 24 species of winter birds, 7 species of reptiles, 3 species of amphibians, 15 species of mammalians and 32 species of fishes were recorded. Relative abundance of those species were determined. Of the birds, House Sparrow (Passer domesticus) was abundant while Blyth's Kingfisher (Alcedo hercules), Rock Eagle Owl (Bubo bengalensis), Hooded Pitta (Pitta sordida), Black-headed Oriole (Oriolus xanthornus), White-winged Duck (Cairina seululala) and Duck (Anser indicus) were rare. The relative abundance of winter bird could not be assessed because of their migratory habit. Striped keelback (Amphiesma stolata) and Common Smooth Water Snake (Enhydris enhydris) were very common while Black pond turtle (Geoclyms hamiltonii) and Pond tortoise (Melanochelys trijuga) were recorded as endangered. Common Toad (Bufo melanostictus) were abundant but Bull Frog (Rana tigrina) was rare. Asiatic Wild Dog (Cuon alpinus) and House Mouse (Mus musculus) were abundant while Common Otter, Large Indian Civet, Irrawaddy River Dolphin, Indian Hare were rare. Carpu, Silver carp, Tilapia, Nilotica were abundant while, Freshwater Garfish, One stripe spinyeel and Grey Featherback were rare. Landfilling, deforestation, poaching, industrial effluents and current jal were identified as major threats to the faunal diversity of Savar area.  相似文献

Previous studies have shown that inflammatory factors increases in pregnancy and is associated with several complications of pregnancy. The aim of this study was to assess effects of daily consumption of probiotic yoghurt on inflammatory factors in pregnant women. In a randomized clinical trial, seventy primigravid (the first pregnancy) and singleton pregnant women aged 18-30 years were assigned to two groups. Subjects consumed daily 200 g probiotic yoghurt containing Lactobacillus acidophilus La5 and Bifidobacterium animalis BB12 (10(7) CFU g(-1) for each) or 200 g conventional yoghurt for 9 weeks. Fasting blood samples were collected at baseline (28 weeks of gestation) and after intervention (37 weeks of gestation). Inflammatory factors, hs-CRP and TNF-alpha, were measured by Enzyme-linked Immunosorbent Assay (ELISA). Independent t-test was used to compare the two groups after intervention and paired-sample t-test compared variables before and after treatment. The results showed that the probiotic yogurt brought about a decrease in the serum hs-CRP level, from 10.44 +/- 1.56 to 7.44 +/- 1.03 microg mL(-1) (p = 0.041). There was no significant change in the conventional yogurt group in the serum hs-CRP level (12.55 +/- 1.57 to 14.51 +/- 1.62 microg mL(-1), p = 0.202). The probiotic yogurt had no effect on TNF-alpha (from 73.75 +/- 6.59 to 77.91 +/- 5.61 pg mL(-1), p = 0.633). Serum TNF-alpha did not change in the conventional yogurt group (p = 0.134). In conclusion probiotic yogurt significantly decreased hs-CRP in pregnant women but had no effect on TNF-alpha.  相似文献

以五凤香茗(烘青类)和苍南翠龙(扁型茶类)为研究对象,设定适宜的润茶条件,比较润茶过程对这两种不同加工而成的茶叶主要品质成分溶出的影响。结果表明,润茶有利于茶叶中氨基酸、茶多酚和咖啡碱等品质成分的溶出。不同润茶时间,物质溶出量不同,氨基酸溶出速度快,茶多酚和咖啡碱溶出较缓。润茶时表现为烘青类的"五凤香茗"中物质溶出比扁型茶类的"苍南翠龙"快速,润茶70 s~80 s时,物质溶出量处较高水平。润茶用水量20 m L、35 m L、50 m L时,物质溶出量差异大,两种类型茶润茶用水量由20 m L增加至35 m L时,三种主要品质成分溶出量增幅大于50%;润茶用水量从35 m L增加至50 m L时,物质溶出量的增幅差距缩小。以物质最大幅度溶出量考虑,冲泡3 g干茶,茶水比1∶50时,选择35 m L润茶用水量为合适。茶水冲泡后的3 min~5 min内,茶汤中的主要品质成分物质溶出量随时间延长,溶出量增加,但不同时间段增加速率不同,咖啡碱随时间延长,溶出速率增加,氨基酸和茶多酚则是在冲泡4分钟时,溶出速率最快。经润茶后的茶叶再续水冲泡,茶汤上、下层的均匀程度优于直接冲泡;不同冲泡方式均表现为上、下层茶汤中茶多酚、咖啡碱差异大,氨基酸差异小。  相似文献

About 4 different predominant adapted fungal strains (screened from effluent sample) Aspergillus sp., Penicillium sp., Fusarium sp. and Mucor sp. and 4 predominant non-adapted strains (screened from soil, water and fungal fruiting bodies) Aspergillus sp., Penicillium sp., Fusarium sp. and Rhizopus sp., with potential dye decolorization ability on Reactive black 5, Amido black-10B, Red 5B, Reactive red 120 and Anthraquinone violet R were isolated. These organisms were used to develop a consortium which was used in analyzing the bioremediation efficiency on textile effluents containing a mixture of azo dyes. There was about 67% of reduction in color along with 34% of COD reduction by non-adapted fungal consortium while effective bioremediation efficiency was observed in adapted fungal consortium (Color 75% and COD 50%). The regression co-efficient for Langmuir and Freundlich adsorption isotherms were found to be higher for adapted fungal consortium (R2 = 0.97 and R2 = 0.92) than the non-adapted consortium (R2 = 0.97 and R2 = 0.85) proving that both monolayer and multilayer adsorption of dyes were observed on treating the samples with the adapted fungal consortium. On analyzing the results observed through chi-square test, the calculated value (28.712) was higher than the tabulated value (9.49) at a 4 degree freedom hence the hypothesis was rejected. So, there was an association between adapted fungal consortium and non-adapted fungal consortium and hence the adapted fungal consortium could be considered potentially useful for the bioremediation of textile effluent.  相似文献

采用稀释平板法和平板划线法从8个不同品种的香蕉果实上分离纯化酵母菌,并将其与香蕉炭疽病原菌进行平板对峙培养及离体防效试验,以筛选优势拮抗酵母菌.结果表明:从香蕉果实上共分离纯化酵母菌329株;平板对峙培养后获得3株拮抗香蕉炭疽病原菌作用明显的酵母菌;离体防效试验筛选出1株拮抗效果较好且稳定的优势拮抗酵母菌Z-BR-16,以无菌水为对照,接种7d后,其1.0×108 CFU/mL菌液对香蕉炭疽病的抑制率为75.94%.  相似文献

This study investigated the effects of different additives on fermentation quality and aerobic stability of Leymus chinensis silage. Treatments included (i) no additive, (ii) 3 mL kg?1 formic acid (FA), (iii) 6 mL kg?1 FA, (iv) 5 mL kg?1 acetic acid (AA), (v) 10 mL kg?1 AA, (vi) 2 mL kg?1 propionic acid (PA), (vii) 4 mL kg?1 PA, (viii) 5 mL kg?1 butyric acid (BA), (ix) 10 mL kg?1 BA, (x) 1.0 g kg?1 potassium sorbate (PS), (xi) 1.0 g kg?1 sodium benzoate (SB), (xii) 1 × 108 colony‐forming units (cfu) kg?1 Lactobacillus (Lb) plantarum LP (LP), (xiii) 1 × 108 cfu kg?1 Lb. brevis LB (LB) and (xiv) 1 × 108 cfu kg?1 Lb. buchneri NCIMB40788 (Fresh). Each additive treatment was based on fresh matter (FM). Results showed that all additives decreased pH values. All additives except Fresh decreased ammonia‐N content (< .001). Both LP and LB increased lactic acid content (< .001). Butyric acid content increased with FA (3 mL kg?1) and BA, but decreased with PA, PS, SB, FA (6 mL kg?1), AA, LP, LB (< .001). FA (3 mL kg?1), AA (10 mL kg?1), PA, BA, PS, SB and Fresh improved aerobic stability (< .001). After 8 days exposure to air, the pH value and yeast count were lower in FA (3 mL kg?1), BA (10 mL kg?1), SB treatments than in other treatments. Overall, AA outperformed all other additives in improving fermentation quality. Sodium benzoate and AA could be used as an effective additive to improve aerobic stability of L. chinensis silage.  相似文献

从海南省乐东、三亚和儋州等市县采集的23份土样中粗筛获得186株具有纤维素降解能力的菌株,11株具有木质素降解能力的菌株,经进一步筛选获得纤维素酶高产菌和木质素酶高产菌各2株,编号分别为12—4、3—5、6—1和3—3。在以香蕉茎叶粉为主要基质的培养基中30℃、120r/min振荡培养5d,12—4和3—5分解羧甲基纤维素钠(CMC)的酶活分别达到979U和226U,分解滤纸(FP)的酶活分别为46.4U和15.1U,6—1和3—3的木质素酶活分别为32.8U和16.4U。经初步鉴定,这些菌株分别属于Trichodermasp.,Penicillumsp.,Streptomycesflavus和ChoanephoraCurreysp.,可用于发酵香蕉茎叶生产饲料的进一步研究。   相似文献

This study was carried out to determine the lethal effect of the entomopathogenic fungi, Beauveria bassaina Vuell. on eggs, young and old nymphs of the whitefly, Bemisia tabaci Genn. Mortality percentage was significantly differed based on stage of B. tabaci and conidial concentrations of B. bassina. Average of the infection level to insect was very low particularly in eggs with only 4.49%, even with higher conidial concentrations (6 x 10(6) conidia mL(-1)). Whereas, it was higher with 1st and 2nd instars (42.045%) and 3rd and 4th instars (35.93%). Three parameters was assessed with B. tabaci eggs, namely; egg infection, egg hatchability and crawlers emergence. Egg mortality percentages averaged 1.2, 4.27 and 8.0% with fungal concentration 2 x 10(6), 4 x 10(6) and 6 x 10(6) conidia mL(-1), respectively. Daily infection percentages were varied depend upon the conidial concentration where the highest infection rate of eggs was occurred with 6 x 10(6), followed by 4 x 10(6) conidia mL(-1). Egg hatch was very high, while the mortality among the emerged crawlers was neglectable compared with the check. Efficiency of B. bassaina on whitefly nymphs also was varied based on the insect instar and fungal concentration. Mortality percentages were obviously higher to young nymphs (1st and 2nd instars) than to older ones (3rd and 4th instars). The results indicated that nymphs were highly susceptible to fungal treatment compared with eggs. Additionally, pathogenicity and virulence of B. bassaina against B. tabaci immatures was not indicated by LC50 only, but also, by the time in days (LT50) required to achieve 50% mortality of an insect.  相似文献

Regarding the importance of the presence of intestinal lactobacilli and their population in infants, four different treatments were evaluated for Lactobacillus isolation efficiency via reduction in the growth of other groups of bacteria capable of growing on de-Man Rogosa Sharpe (MRS) medium from fecal samples of 11 Iranian infants. MRS-Vancomycin (1 mg L(-l)) was used as a base medium and application of lactic acid and aerobic incubation of inoculated plates were performed as selective factors. Each fecal sample was cultivated as duplicate on to the base medium with or without lactic acid to reduce the pH to 5.4 +/- 0.2. Half of the plates were incubated aerobically and the rest of them incubated under 10% CO2 concentration. Total count and Lactobacillus count of all samples were recorded according to the age differences of infants. The counts of false positive colonies were recorded with respect to their cell morphology and gram reaction in all treatments. Anaerobic incubation of lactic acid modified MRS-Vancomycin gave the most Lactobacillus percentage coverage, about 93% among the Lactobacillus positive samples. Using this treatment, the median Lactobacillus count yielded 8.29 log10 cfu g(-1) in the younger and 5.70 log10 cfu g(-1) in the elder group. It could be concluded that lactic acid might be a proper pH reducing agent when enumeration of lactobacilli from fecal samples is of interest.  相似文献

This study aimed to evaluate the efficacy of semi fermented and non fermented Camellia sinensis extracts (Black and Green tea) and comparison between them against Streptococcus mutans ATCC 25175, S. mitis ATCC 9811 and S. sanguis ATCC 10556 that are responsible for dental caries and bacteremias following dental manipulations. Minimum inhibitory concentrations of both tea extracts were assessed by Well diffusion and Broth dilution methods and examination of cell adherence (Biofilm inhibitory concentrations) was observed on glass slides under phase contrast microscope and colony counts from glass beads. Concentration of 1 mg mL(-1) of semi fermented tea extract was completely biofilm inhibitor but biofilm formation by these bacteria was seen 7 days after treatment with 1 mg mL(-1) of non fermented Camellia sinensis on glass beads and BIC for oral streptococci treated with this extract was 1.5, 2.5 mg mL(-1) of semi fermented and 3 mg mL(-1) of non fermented extracts had bactericidal effect on these bacteria. Semi fermented and non fermented Camellia sinensis extracts were able to prevent growth of oral streptococci. Therefore dental caries significantly reduce and the efficiency of semi fermented tea was higher due to rich content of volatile components rather than non fermented extracts.  相似文献

The digestion and Volatile Fatty Acid (VFA) production from rice straw and oil palm fronds by cellulolytic bacteria isolated from the termite Coptotermes curvignathus were investigated. The bacteria were Acinetobacter strain Raminalimon, Enterobacter aerogenes strain Razmin C, Enterobacter cloacae strain Razmin B, Bacillus cereus strain Razmin A and Chryseobacterium kwangyangense strain Cb. Acinetobacter strain Raminalimon is an aerobic bacterium, while the other species are facultative anaerobes. There were significant differences (p<0.05) among the bacteria for Dry Matter (DM) lost and acetic acid production from rice straw and Acinetobacter strain Raminalimon showed the highest activity. The facultative bacteria C. kwangyangense strain Cb (cfu mL(-1) 231 x 10(-6), OD: 0.5), E. cloacae (cfu mL(-1) 68 x 10(-7), OD: 0.5) and E. aerogenes (cfu mL(-1) 33 x 10(-7), OD: 0.5) were used for digestion study with the rumen fluid microflora. The in vitro gas production technique was applied for the comparative study and the parameters measured were pH, gas (volume), dry matter lost, acetic acid, propionic acid and butyric acid concentrations. pH was not significantly (p<0.05) different among the five treatments. The bacterium C. kwangyangense strain Cb showed the highest activity (p<0.05) for DM lost, acetic acid, propionic acid and butyric acid production from rice straw when compared to the other bacterial activities. There was no significance (p<0.05) difference between the three bacteria for the dry matter lost of oil palm fronds but the production of Volatile Fatty Acids (VFA) was significantly (p<0.05) high in the treatment which was inoculated with C. kwangyangense strain Cb. The objective of this research was to evaluate the effect of ozone gas and potassium lactate on lipid oxidation and survival of Salmonella typhimurium on fresh pork. A total of 144 samples of fresh pork samples were cut into pieces approximately (8x5x0.6) cm in size, then (0, 2 and 4%) potassium lactate (KL), inoculated with S. typhimurium was applied by spreading a 0.5 mL cell suspension over each sample. The pork samples were then packed in (6"x8") airtight polyethylene bags with a thickness of 87.5 microm and a volume of 3.5 L with and without ozone. Ozone gas was injected into the plastic bags at a rate of 0, 200, 500 and 1,000 mg h(-1) and the samples later stored at 8 degrees C. Thio-barbituric Acid Reactive Substance (TBARS) and microbial loads were determined on days 0, 5, 10 and 15. Data was statistically analyzed using SPSS software and differences among means detected at the 0.5% confident level using the Scheffe's test. Samples treated with 2 and 4% KL had significantly (p<0.01) lower TBARS value than non-treated samples. Combination of ozone and KL showed inhibitory effects on S. typhimurium in samples. S. typhimurium was sensitive to 4% KL with and without ozone. At 1,000 mg h(-1), ozone improved KL inhibitory effect on S. typhimurium. Ozone and KL are potential substances for inhibition of S. typhimurium.  相似文献
