Q fever is a zoonosis caused by the intracellular bacterium Coxiella burnetii. Both humoral and cellular immunity are important in the host defence against intracellular bacteria. Little is known about the immune response to C. burnetii infections in domestic ruminants even though these species are the major source of Q fever in humans. To investigate the goat’s immune response we inoculated groups of pregnant goats via inhalation with a Dutch outbreak isolate of C. burnetii. All animals were successfully infected. Phase 1 and Phase 2 IgM- and IgG-specific antibodies were measured. Cellular immune responses were investigated by interferon-gamma, enzyme-linked immunosorbent spot test (IFN-γ Elispot), lymphocyte proliferation test (LPT) and systemic cytokines. After two weeks post inoculation (wpi), a strong anti-C. burnetii Phase 2 IgM and IgG antibody response was observed while the increase in IgM anti-Phase 1 antibodies was less pronounced. IgG anti-Phase 1 antibodies started to rise at 6 wpi. Cellular immune responses were observed after parturition. Our results demonstrated humoral and cellular immune responses to C. burnetii infection in pregnant goats. Cell-mediated immune responses did not differ enough to distinguish between Coxiella-infected and non-infected pregnant animals, whereas a strong-phase specific antibody response is detected after 2 wpi. This humoral immune response may be useful in the early detection of C. burnetii-infected pregnant goats.  相似文献

Since there is no registered anthelmintic drug available for use in goats, extra-label use of drugs is a common practice in most countries. The aim of the present study was to compare the pharmacokinetic disposition of levamisole (LVM)-oxyclozanide (OXZ) combination in sheep and goats following per os administration. Goats (n = 8) and sheep (n = 8) 12- to 16-months-old were used for this study. The animals received tablet formulation of LVM and OXZ combination orally at a dose of 7.5 mg/kg and 15 mg/kg body weight, respectively. Blood samples were collected by jugular vein at different times between 5 min and 120 h after drug administrations. The plasma concentrations of LVM and OXZ were analyzed by HPLC following liquid-liquid phase extraction procedures. The plasma concentrations and systemic availabilities of both LVM and OXZ in goats were lower and the plasma persistence of LVM was shorter compared with those observed in sheep. Terminal half-lives (t1/2λz) of both molecules are shorter in goats compared with those in sheep. Goats treated with LVM-OXZ combination at the recommended dose for sheep may result in a reduced efficacy, because of under-dosing, which may increase the risk of drug resistance in parasites. Increased or repeated dose could be a strategy to provide higher plasma concentration and thus to improve the efficacy against the target parasites in goats compared with sheep. However, some adverse reactions may occur since LVM has relatively very narrow therapeutic index due to its nicotine-like structure and effect.  相似文献

A trial was conducted during the anestrous period in female goats to determine: (a) whether estrus can be induced in anestrous goats by administration of equine chorionic gonadotropic hormone (eCG) and PGF under pen conditions and (b) whether these sexually active female goats can elicit sexual arousal in sexually inactive bucks. One hundred and fifteen pluriparous, nonlactating mixed-breed female goats were randomly assigned to one of four treatment groups: (1) administration of a single dose of 240 IU of eCG, 50 μg PGF i.m., and 25 mg progesterone (P4) (eCG; n?=?30); (2) administration of P4 and exposure to female goats treated with eCG–PGF (P4; n?=?39); (3) administration of 0.5 ml saline and P4 (Sal; n?=?23); and (4) P4 plus exposure to female goats treated with saline (Con; n?=?23). After hormone administration, all goats were put together with adult sexually inactive bucks for 15 days. The percentage of goats in estrus during these 15 days was similar in eCG-treated animals and untreated animals exposed to the eCG animals (97 and 95 %). Pregnancy rate was also similar (63 vs. 64 %) between these two groups. eCG-treated goats exhibited estrus earlier (P?<?0.05) than the treated goats in contact with the eCG goats. Furthermore, eCG-treated goats had larger litters (1.9?±?0.2 vs. 1.6?±?0.1, P?<?0.05) than the untreated goats in contact with the eCG goats. These results show that fertile estrus can be induced in anestrous female goats by exposing them to female goats induced to estrus with eCG. This female–female interaction triggers the stimulation cycle leading to the sexual arousal of bucks.  相似文献

BackgroundAttenuated Salmonella strain can be used as a vector to transport immunogens to the host antigen-binding sites.ObjectivesThe study aimed to determine the protective efficacy of attenuated Salmonella strain expressing highly conserved Brucella immunogens in goats.MethodsGoats were vaccinated with Salmonella vector expressing individually lipoprotein outer-membrane protein 19 (Omp19), Brucella lumazine synthase (BLS), proline racemase subunit A (PrpA), Cu/Zn superoxide dismutase (SOD) at 5 × 109 CFU/mL and challenge of all groups was done at 6 weeks after vaccination.ResultsAmong these vaccines inoculated at 5 × 109 CFU/mL in 1 mL, Omp19 or SOD showed significantly higher serum immunoglobulin G titers at (2, 4, and 6) weeks post-vaccination, compared to the vector control. Interferon-γ production in response to individual antigens was significantly higher in SOD, Omp19, PrpA, and BLS individual groups, compared to that in the vector control (all p < 0.05). Brucella colonization rate at 8 weeks post-challenge showed that most vaccine-treated groups exhibited significantly increased protection by demonstrating reduced numbers of Brucella in tissues collected from vaccinated groups. Real-time polymerase chain reaction revealed that Brucella antigen expression levels were reduced in the spleen, kidney, and parotid lymph node of vaccinated goats, compared to the non-vaccinated goats. Besides, treatment with vaccine expressing individual antigens ameliorated brucellosis-related histopathological lesions.ConclusionsThese results delineated that BLS, Omp19, PrpA, and SOD proteins achieved a definite level of protection, indicating that Salmonella Typhimurium successfully delivered Brucella antigens, and that individual vaccines could differentially elicit an antigen-specific immune response.  相似文献

OBJECTIVE: To evaluate vaccine safety, antibody response, and nonspecific lymphocyte blastogenesis following inoculation of a commercial monovalent live attenuated bluetongue virus (BTV) serotype 2 vaccine in goats. ANIMALS: 12 nonpregnant and nonlactating Saanen goats. PROCEDURE: 6 goats were inoculated with the monovalent live attenuated BTV serotype 2 vaccine, which has been widely used in Italy during the proceding 2 years. The other 6 goats were unvaccinated and represented negative controls. Nonspecific lymphocyte blastogenesis was evaluated 14 and 7 days before and 7, 21, and 49 days after vaccination by measuring DNA synthesis in peripheral blood mononuclear cells (PBMCs) stimulated with phytohemagglutinin, concanavalin-A, and pokeweed mitogen. On the same days as lymphocyte blastogenesis, blood samples were taken to determine serum concentrations of anti-BTV antibodies. RESULTS: During the 7 weeks following vaccination, PBMCs obtained from vaccinated goats had a significantly decreased response to mitogens in terms of DNA synthesis, compared with PBMCs from the same goats before vaccination. Conversely during the experiment, no significant change was found in the response of the PBMCs obtained from unvaccinated goats. Starting from 21 days after vaccination, serum from vaccinated goats had anti-BTV antibodies. No anti-BTV antibodies were detected in the serum from unvaccinated goats. CONCLUSIONS AND CLINICAL RELEVANCE: Inoculation of goats with the monovalent live attenuated BTV serotype 2 vaccine described herein resulted in a profound depression of nonspecific lymphocyte blastogenesis, which might compromise the resistance of vaccinated goats to pathogens.  相似文献

The serological response, lymphocyte reactivity, and dermal hypersensitivity reactions of goats vaccinated with high and low doses of Brucella melitensis Rev 1 organisms to Brucella antigens were studied. Antibodies to soluble antigen A2 were detectable by immunoelectrophoresis (IE), appeared later than agglutinating antibodies but disappeared faster in most vaccinated animals. Anti-A2 antibodies appeared earlier in goats which received the high dose. Antibodies to polysaccharide B antigen were not detected. Lymphocytes reactive to Brucella antigens in lymphocyte transformation assays appeared at variable times after vaccination. In contrast to the humoral response to antigen A2, the appearance of circulating, reactive lymphocytes was not dependent on vaccine doses. All vaccinated animals demonstrated dermal hypersensitivity reactions to one of the antigenic extracts. Skin reactions peaked at 24 hrs post inoculation. The reactions were elicited when all but one goat had undetectable anti-A2 antibodies and no circulating, reactive lymphocytes as measured in whole blood lymphocyte transformation assay.  相似文献

Quantitative determinations of IgG1 and IgG2, in one group of colostrum-fed and one group of colostrum-deprived neonatal goats revealed that the occurrence of the IgG1 subclass preceeded that of the IgG2 in both cases. In the colostrum-fed animals the IgG2 appeared, on an average, in the fourth week of life whereas in the colostrum-deprived animals the IgG2 was detected as early as three weeks after birth. At the age of twelve weeks the mean concentrations for IgG, and IgG2 were higher in the animals deprived of colostrum. The immune response to human gamma globulin was studied in colostrum-fed and colostrum-deprived neonatal goats which were immunized at birth and again after four and eight weeks. Following the first two antigen administrations a significantly higher response was obtained in the colostrum-fed neonates. However, the third injection determined a similar response in both groups. A marked suppressive effect on the immune response was observed in colostrum-fed neonatal goats when specific antibodies were present in the colostrum after preimmunization of the mothers with human gamma globulin.  相似文献

Two groups of male, 9-week-old goats (5 goats/group) were vaccinated subcutaneously with formalized exotoxin of Corynebacterium pseudotuberculosis, with Freund's incomplete adjuvant. Each goat was given 2 vaccinations, 2 weeks apart. At each vaccination, each group 1 goat was given 0.5 ml of toxoid, and each group 2 goat was given 1 ml of toxoid. Twenty days after the 2nd vaccination, vaccinated goats and 5 nonvaccinated 12-week-old goats (controls) were inoculated intradermally (challenge exposed) with live C pseudotuberculosis, monitored for 13 weeks, and euthanatized. At necropsy, 5 of the 10 vaccinated goats did not have C pseudotuberculosis lesions, 3 had abscesses limited to the inoculation site and draining lymph node, and 2 had disseminated bacterial lesions. Of the 5 nonvaccinated controls, 4 had disseminated abscesses and 1 had a single abscess in an internal node. Serologically, 9 of the 10 vaccinated goats developed positive (greater than or equal to 1:8) antibody titers against the exotoxin within 1 week after inoculation; the 10th goat seroconverted 2 weeks after inoculation, whereas control goats required 3 weeks to develop a positive antibody response. Therefore, early during an infection with C pseudotuberculosis, antibodies against the exotoxin may protect a goat against spread of the organism. Very few, mostly old, and only preliminary serological studies of brucellosis in goats exist in Ecuador. In order to assess the current epidemiological situation, we performed a cross-sectional serological study in the goat populations of Carchi (n?=?160 animals), Pichincha (n?=?224 animals), and Loja provinces (n?=?2024 animals). Only two positive serological results (RB negative and SAT-EDTA ≥400 IU/ml) were obtained in lactating goats from the same farm in Quito (Pichincha province). Additionally, milk was sampled from 220 animals in Pichincha province. The present study indicates a low apparent prevalence in Pichincha province and absence in Carchi and Loja provinces. A total of 25 positive milk ring tests (MRT) were obtained in Pichincha province yielding a prevalence of MRT of 11.16%. Subsequent culture was performed on the positive MRT samples. All results were negative, apart from a single sample, obtained from a serologically positive goat in Quito, that was positive for Brucella abortus strain 19 (B19). Several hypotheses are forwarded concerning this unexpected result. The most likely hypothesis is the possible accidental use of a needle, previously used for vaccination of cattle with the said vaccine, for the administration of drug treatment to the goat. Tuberculosis in goats (caused by Mycobacterium caprae and M. bovis) has become a significant concern in recent years because of its high prevalence in certain caprine herds in Spain and other European countries, and also due to the potential transmission to other animals and human beings. In the present study, a transthoracic model of tuberculosis infection was performed on goats. Animals were selected based on the serological response used to detect paratuberculosis in goats (negative and positive results). The kinetics of the immune response was evaluated using the interferon-γ (IFN-γ) assay, skin tests and serology of paratuberculosis during nine months post-challenge. At the end of the study the animals were necropsied, tuberculosis-lesions were scored and culture (M. caprae and M. avium subsp. paratuberculosis) was performed to determine the true infection status. Animals were positive to the IFN-γ assay 15 days post-challenge and the values were fluctuating throughout the study. A varied performance of the assay was observed between tuberculosis and tuberculosis-paratuberculosis mixed infection regarding both the number of positive results and the OD values obtained after stimulation with bovine and avian PPDs. Furthermore, the single intradermal comparative cervical tuberculin test did not detect all M. caprae-infected animals. At necropsy, a positive correlation between pathology score and bovine PPD specific IFN-γ response was found.  相似文献

A trial was performed using 20 goats to evaluate the antibody responses to a liposome-adjuvanted Clostridium perfringens epsilon toxoid vaccine (LIPV). The antibody response was compared with those produced by epsilon toxoid vaccines prepared using aluminium hydroxide (ALV) and incomplete Freud's adjuvant (FAV). The animals were allocated to four groups at the beginning of the trial. The animals in group 1 were vaccinated with ALV, while the animals in group 2 received FAV and those in groups 3 and 4 were vaccinated with LIPV. The animals in groups 1 to 3 received three doses of the corresponding vaccine at intervals of three weeks, while those in group 4 received only 1 dose of vaccine at the beginning of the trial. A blood sample was obtained from all the goats at the beginning of the trial and then weekly for 8 weeks. The samples were analysed for epsilon toxoid antibodies by an indirect ELISA technique. No major clinical abnormalities were observed in the animals after vaccination, with the exception of those that received the FAV, which experienced transient lameness. The highest antibody response was observed in the animals vaccinated with FAV, but they presented moderate to severe inflammatory tissue reactions at the injection site. Moderately high antibody responses were obtained with the ALV, with which only minor local reactions were observed. No significant antibody responses were obtained with the LIPV, nor were local reactions observed.  相似文献

An autogenous vaccine was developed, using sonicated bacteria, with a strain of Streptococcus suis capsular type 1/2. The objectives of this study were to evaluate the antibody response following vaccination and to assess the changes in antibody levels in pigs from a herd showing clinical signs of S. suis capsular type 1/2 infection in 6- to 8-week-old pigs. An enzyme-linked immunosorbent assay using the vaccine antigen was standardized. Results from a preliminary study involving 2 control and 4 vaccinated 4-week-old pigs indicated that all vaccinated pigs produced antibodies against 2 proteins of 34 and 43 kDa, respectively, and, in 3 out of 4 vaccinated pigs, against the 117-kDa muramidase-released protein. For the serologic profile, groups of 30 pigs from the infected herd were blood sampled at 2, 4, 6, 8, and 10 weeks of age. The lowest antibody level was observed between weeks 6 and 8, presumably corresponding to a decrease in maternal immunity. A marked increase was seen at 10 weeks of age, shortly after the onset of clinical signs in the herd. For the vaccination field trial, newly weaned, one-week-old piglets were divided into 2 groups of 200 piglets each (control and vaccinated); blood samples were collected from 36 piglets in each group at 2-week intervals for 12 weeks. A significant increase (P 0.05) in antibody response was observed 4 weeks following vaccination and the level of antibodies stayed high until the end of the experiment. In the control group, the increase was only observed at 13 weeks of age, probably in response to a natural infection. The response to the vaccine varied considerably among pigs and was attributed, in part, to the levels of maternal antibodies at the time of vaccination. No outbreak of S. suis was observed in the control or vaccinated groups, so the protection conferred by the vaccine could not be evaluated.  相似文献

Fifty, 2–3 month old dairy goats were each vaccinated orally with 10000 sporocysts of Sarcocystis capracanis and 25 age-matched goats served as uninoculated controls. Groups of vaccinated and control goats were challenged with lethal doses of S. capracanis at 95, 113, 205, and 274 days post-vaccination. Vaccinated goats developed subclinical sarcocystosis. Twenty-three vaccinated goats and 1 control goat died of intestinal coccidiosis and bacterial pneumonia, 15–118 days after vaccination. Myositis and degenerating sarcocysts were seen in muscles of goats necropsied at 90–186 days postvaccination. Very few, or no sarcocysts were seen in goats necropsied at 272 and 332 days post-vaccination. Vaccinated goats survived a lethal challenge inoculation indicating persistent protective immunity.  相似文献

The inflammatory host response to Schistosoma bovis in young goats was studied at necropsy by light microscopy 34 weeks after primary exposure to 3,000 cercariae (group B, n=6), 34 weeks after primary exposure to 3,000 cercariae followed by challenge with 2,500 cercariae at week 17 (group C, n=5), and 17 weeks after primary exposure to 2,500 cercariae, given on week 17 of the experiment (group D, n=6). Three goats served as uninfected controls. The faecal egg output had been minimal for 17 weeks prior to necropsy in groups B and C and only for the last 2 weeks in group D.Histological studies were carried out on the small intestine, liver, lung and spleen, and tissue egg counts were performed. In sections of the small intestine and liver, a panel of histopathological variables were quantitated to characterize the host response and differences between groups of animals were evaluated with one way analysis of variance. The mean tissue egg count in the small intestine was slightly but not significantly higher in group C than group B and about twice as high in group D (D vs B or C p<0.01). Group means of numbers of inflammatory foci per section of gut wall corresponded well with those of tissue egg counts, suggesting that the rate of inflammatory destruction of eggs did not differ markedly between the groups. Egg material was less commonly seen in granulomas of the small intestine in group B than in group D (p<0.01), suggesting lower passage of eggs through the gut wall during the later than during the earlier phase of patent primary infection. The frequency of eosinophil-rich hepatic inflammatory foci was much higher in group D than in the other groups (D vs B p<0.05, D vs C p< 0.01), and coincided with a high degree of blood eosinophilia in this group at the time of sacrifice. Challenged goats showed a significantly higher frequency of markedly fibrotic inflammatory foci in the liver and of liver granulomas with a marked giant cell component than goats of the other groups. Hepatic portal fibrosis was least prominent in animals with 17- week- old primary infections, implying a possible relation between this change and duration of infection.  相似文献

Angora goats grazing on a paddock contaminated with Ostertagia ostertagi and O. circumcincta were found to have infections consisting of equal numbers of both species. Subsequently, goats were infected with O. ostertagi larvae of bovine origin. The infection became patent by day 28 and egg counts reached a plateau about 7 weeks after infection. Worm numbers did not decline during the 12 weeks of the experiment. It was concluded that Angora goats are suitable hosts for O. ostertagi.  相似文献

The effect of simultaneous infections with trypanosomes (Trypanosoma vivax and Trypanosoma congolense) and gastrointestinal nematodes on the productivity of sheep and goats was studied in 20 animals (6 male goats and 14 male sheep) at a farm near Maputo. The animals were divided into 4 groups which received either treatment against gastrointestinal nematodes, or against trypanosomes, or treatment against both, or no treatment at all.In two-weekly and later in weekly intervals body weight, packed cell volume, body temperature, worm egg burdens and occurence of trypanosomes in the peripheral blood were recorded. Body temperature and packed cell volume did not show the expected close relation to an infection with either trypanosomes or gastrointestinal nematodes, but the differences in the increase of body weight among the 4 groups were very considerable.Animals receiving both treatments gained an average of 13.5 kg in body weight in 40 weeks compared with 5.1 kg (surviving animals with treatment against worm parasites), 6.5 kg (surviving animals with treatment against trypanosomes) and 3.4 kg (surviving animals without any treatment). Of the last 3 groups 1 sheep (21 kg), 2 sheep (50 kg) and 1 sheep (21 kg), respectively, died.One goat, treated against gastrointestinal nematodes, but suffering from an infection with Trypanosoma vivax and Trypanosoma congolense was killed at the end of the experiment. A striking finding during the post-mortem examination was the complete hyperplasia of the red-marrow of the right and left femur.  相似文献

Data on reproduction and mortality were collected over one year from 5100 sheep and 13 300 goats in treated and control flocks. The treated animals received vaccination against peste des petits ruminants (PPR) and anthelmintics twice a year. Productivity parameters (fecundity and mortality rates) obtained with and without prophylaxis were fitted into a benefit–cost economic analysis model and run for project lifespans varying from one to five years. At a 7% discount rate, the overall benefits for a project lifespan of five years were estimated as over 15 million FCFA and 11 million FCFA for sheep and goats, respectively. The benefit–cost ratio ranged from 2.26 to 3.27 in goats and 3.01 to 4.23 in sheep, depending on the project lifespan. It was concluded that PPR and gastrointestinal helminthosis are important causes of economic losses in small ruminants in Cameroon. A national or even a regional vaccination campaign against PPR and strategic anthelmintic treatment of small ruminants are recommended.  相似文献

The aim of the present study was to clarify the effect of melatonin (MEL) on the salsolinol (SAL)‐induced release of prolactin (PRL) in goats. Female goats were kept at 20°C with 16 h of light, 8 h of darkness, and orally administered saline or MEL for 5 weeks. A single intravenous (i.v.) injection of saline (controls), SAL, thyrotropin‐releasing hormone (TRH) or a dopamine receptor antagonist, sulpiride, was given to the goats 3 weeks after the first oral administrations of saline or MEL, and the responses were compared. The mean basal plasma PRL concentrations in the control group were higher for the saline treatments than MEL treatments (P < 0.05). SAL as well as TRH and sulpiride stimulated the release of PRL promptly after each injection in both the saline‐ and MEL‐treated groups (P < 0.05). The area under the response curve of PRL for the 60‐min period after the i.v. injection of SAL, TRH and sulpiride in the saline‐treated group was greater than each corresponding value in the MEL‐treated group (P < 0.05). These results show that daily exposure to MEL under a long day length reduces the PRL‐releasing response to SAL as well as TRH and sulpiride in goats.  相似文献

Therapeutic efficacy of a new ‘Indigenous vaccine’ prepared from native highly pathogenic ‘Indian Bison Type’ genotype of Mycobacterium avium subspecies paratuberculosis (MAP) of goat origin has been evaluated with respect to control of clinical Johne’s disease in naturally infected Mehsana breed of goat in North Gujarat. Fifty goats from Sheep and Goats Research Station, Sardarkrushinagar Dantiwada Agricultural University, Sardarkrushinagar, were randomly divided into 2 groups viz.,‘Vaccinated’(n?=?35) and ‘Control’(n?=?15). After vaccination, goats were monitored for physical condition, morbidity, mortality, body weights, shedding of MAP in feces, internal condition, gross lesions and humoral immune responses up to 120 days (at each interval of 30 days). At the end of 120 days trial, there was marked overall improvement in physical condition and body weights of vaccinated goats as compared to ‘Control’ goats. Vaccinated goats gained significantly (P?<?0.05) higher body weights, hardly exhibited any lesions characteristic of JD, had significantly higher (P?<?0.01) antibody titers and shedding of MAP was significantly (P?<?0.01) reduced. Few of the vaccinated goats were positive for MAP DNA in faecal PCR and blood PCR before vaccination. However, all were found as negative at 120 days post vaccination (DPV). Overall vaccine exhibited effective in restriction of MAP infection and significant improvement in production parameters and reduction in mortality and morbidity due to JD. This study was designed to investigate the effect of feeding on the plasma disposition of triclabendazole (TCBZ) in goats following oral administration. A total of eight goats, aged 14–16 months and weighing 20–30 kg were used in this study. The animals were allocated into two groups (fasted and fed groups) of four animals each. The goats in fed group were fed ad libitum but the animals in fasted group were not fed 24 h before and 6 h after drug administration. Commercial oral drench formulation of TCBZ (Endex-K, 5%) was administered orally to animals in two groups at dose of 10 mg/kg bodyweight. Heparinized blood samples were collected between 1 and 192 h after treatment and the plasma samples were analysed by high performance liquid chromatography (HPLC) for TCBZ, TCBZ sulphoxide (TCBZ–SO), and TCBZ sulphone (TCBZ–SO2). Relatively very low concentration of TCBZ parent drug was detected between 2 and 48 h, but TCBZ–SO and TCBZ–SO2 metabolites were present between 2 and 192 h in the plasma samples of fed and fasted animals. Fasting significantly enhanced the plasma concentration of TCBZ and its metabolites. The availability of TCBZ, TCBZ–SO and TCBZ–SO2 in the plasma samples of fasted goats were markedly greater compared to those of fed goats. It was concluded that fasting decreases the digesta flow rate and prolongs the retention of the drug into the gastrointestinal tract, resulting in enhanced quantitative gastrointestinal absorption or systemic availability of TCBZ and its metabolites in fasted goats.  相似文献
