The isolation of Francisella tularensis subsp. holarctica biovar II (strain 06T0001) from a European brown hare (Lepus europaeus) from Thuringia, Germany, is described for the first time. Identification of the microorganism was carried out by phenotypic characterisation, partial sequencing of the 16S rRNA gene and specific PCR using the primers TUL4-435/TUL4-863 and FtC1/FtC4. The epidemiology of tularemia in Germany is discussed and a risk assessment for humans is made.  相似文献

Recently, a change of hepatitis E from being a typical travel‐associated disease to an autochthonous zoonosis in Germany was observed. An increasing number of autochthonous infections with the hepatitis E Virus (HEV) have been recognized in developed countries. Venison from wild boar is already known to be a potential source of infection, if not prepared properly by the consumer. In Germany, certain wild animals are known to be a reservoir for HEV. However, current information is missing about European brown hares (Lepus europaeus) and wild rabbits (Oryctolagus cuniculus). Thus, a total of 833 hunting‐harvested animals (European brown hares n = 669; wild rabbits n = 164) were tested for the occurrence of HEV RNA and HEV antibodies. For this, liver and blood specimens were taken after hunts in six German federal states. HEV antibodies were found by ELISA in 2.2% (624/14) of European brown hares, but no HEV RNA was detectable by nested real‐time RT‐PCR. In contrast, a seroprevalence of 37.3% (126/47) was observed for wild rabbits, and 17.1% (164/28) of the samples were HEV RNA positive. Genomic analysis revealed that these partial sequences clustered within the rabbit clade of HEV‐3 genotype. In addition, one rabbit sequence segregated into subtype 3g of HEV‐3. Highest seroprevalences for hares and rabbits were detected in the federal states of Bavaria and of Schleswig‐Holstein, respectively. Comparing urban, rural and insular areas, the highest seroprevalence was shown for wild rabbits in rural areas and for European brown hares on the northern island Fehmarn. This study provides evidence that European brown hares and wild rabbits from Germany can be infected with HEV. The different prevalences indicate that wild rabbits are a potential reservoir for HEV in Germany, whereas European brown hares seem to be only of minor importance for the epidemiology of HEV.  相似文献

A recently dead adult female European brown hare (Lepus europaeus Pallas) was found in the wild with no evidence of trauma or external injury. Necropsy revealed symptoms consistent with endotoxic shock and a haemorrhagic jejuno-jejunal intussusception with a necrotic intussusceptum. Small, unattached nodules were present extensively in the jejunal lumen and within the intussuscipiens. These nodules proved to be masses of unsporulated Eimeria leporis oocysts. Review of the literature reveals E. leporis to be a common parasite of wild hares, with protracted pathogenic potential under some circumstances. This is the first report of intussusception in this species and the most severe lesion seen associated with E. leporis. This case suggests that E. leporis may be more harmful to the wild European brown hare population than previously supposed.  相似文献

Background:  Many causes of mortality in the European brown hare, such as bacterial and viral infections, anticoagulant poisoning, and trauma, may result in hemorrhage. There are, however, no reference values concerning blood clotting in this species.  Objectives:  The aim of this study was to determine reference values for blood coagulation times and related parameters in healthy European brown hares.  Methods:  Blood samples from 30 clinically healthy adult hares (15 males and 15 females) were obtained. Hares were physically restrained for blood collection from the cephalic vein into tubes containing citrate and EDTA.  Results:  Mean ± SD were obtained for thrombin time (TT) (13.97 ± 1.37 seconds), prothrombin time (PT) (13.32 ± 2.15 seconds), activated partial thromboplastin time (APTT) (16.73 ± 1.86 seconds), fibrinogen concentration (2.98 ± 1.06 g/L), and platelet count (355.28 ± 128.73 × 109/L). Conclusions: Reference values for blood coagulation times and other parameters associated with blood clotting will be useful in the laboratory evaluation of hemorrhage in the European brown hare.  相似文献

European brown hare syndrome (EBHS) is a viral hepatitis that affects European brown hares (Lepus europaeus) and varying hares (Lepus timidus). The disease occurs today in free-living and farmed hares in most European countries (Gustafsson et al. 1989, Gavier-Widén & Mörner 1991). In Sweden and Denmark the EBHS was first observed at the beginning of the eighties, however, in Finland and Norway the disease has not been reported yet.  相似文献

To elucidate the importance of different causes of mortality which could explain the downward trend of the hare populations in Switzerland and for monitoring selected zoonoses, the health and reproductive status of 167 perished brown hares (Lepus europaeus) was assessed. Concerning causes of mortality, traumas were by far the most frequent diagnosis, 80% of the hares dying because of injuries. Animals killed by road traffic were highly represented. Predators (such as dogs, domestic cats, lynx, martens, buzzards, and golden eagles) killed 16% of the analysed animals. In juveniles, predation was significantly more frequent than in adults. Infectious diseases led to death in 15% of the animals, and cases of pasteurellosis, brucellosis, pseudotuberculosis, tularaemia, listeriosis, and toxoplasmosis were diagnosed. In 5% of the hares, the cause of death pertained to other categories or remained unclear. Reproductive performance was judged to be normal, since mean litter size was 2.5 per female and pregnancy rate in March-June was 74%. We conclude that neither a specific infectious disease, for which adult hares are particularly susceptible, nor an insufficient reproductive performance are responsible for the decline of brown hare populations in Switzerland. This phenomenon is rather a cause of a reduced survival rate in leverets.  相似文献

The isolation and identification of Yersinia enterocolitica from minced meat on CIN agar medium is still one of the major problems in food microbiology because of the low selectivity of cefsulodin-irgasan-novobiocin (CIN) agar. A total of 198 minced meat samples were collected from commercial establishments (butcher shops and supermarkets) in seven German cities in order to investigate the sensitivity and specificity of three identification techniques suitable for the differentiation of Y. enterocolitica within the rich background flora on CIN agar plates. As expected isolation of Y. enterocolitica from minced meat on CIN agar medium after 72 h enrichment in peptone, sorbitol and bile salts (PSB) broth was difficult because all plates were abundantly covered with numerous 'typical'Yersinia-like colonies of bull's eye appearance as well as with atypical colonies. Based on the phenotype of the colonies it was possible to detect colonies showing Yersinia-like growth on CIN agar in 52 samples (26%). For identification of Y. enterocolitica the API 20E system (bioMerieux, Nürtingen, Germany), the Yersinia identification kit (Merlin, Bornheim-Hersel, Germany) and a 16S rRNA based PCR assay were compared. Only in one sample (0.5%) a Y. enterocolitica strain was detected by all methods. Of the three identification systems tested for routine laboratory diagnostics the API 20E system was found to be the most suitable tool to identify Y. enterocolitica colonies within the rich background flora from minced meat samples on CIN agar plates.  相似文献

The HIT method was used to examine blood serums of the game in Moravia (roebuck, red deer, fallow deer, mouflon, wild boar, brown hare) for the presence of antibodies to arboviruses of these groups: alphavirus (Sindbis-SIN), flavivirus (West Nile-WN), tick-borne encephalitis (TBE) and Bunyamwera (Tahyna-TAH, Calovo-CVO). Antibodies to all viruses were detected, and namely in these frequencies: SIN 0.9%, WN 16.9%, TAH 41.5%, CVO 23.1% and TBE 8.5%.  相似文献

Retrospective investigation was done on Yersinia pseudotuberculosis infection in barn rat populations living in an Yersinia endemic area and in zoo rat populations living in a nonendemic area. The OH agglutinins against Y pseudotuberculosis were normally distributed within both populations. Most of the OH agglutinin titers and many O agglutinin titers were reduced by treatment of sera with acetone. Sera with O and OH agglutinin titers were assigned to 2 groups, one with a titer less than 1:8 and the other with a titer of greater than or equal to 1:8. Rats harboring the organism did not possess O or OH agglutinin titers greater than 1:8 in acetone-treated sera. In most nontreated sera, OH agglutinin titers were higher than were the O agglutinin titers. However, in most acetone-treated sera, O agglutinin titers were equivalent to OH agglutinin titers. Differences by region and age were observed in acetone-treated O agglutinin titers of greater than or equal to 1:8 against Y pseudotuberculosis. Therefore, it is recommended that to detect antibody against Y pseudotuberculosis in rats, sera should be treated with acetone before the test and O antigen should be used.  相似文献

Lagovirus is an emerging genus of Caliciviridae, which includes the Rabbit Hemorrhagic Disease Virus (RHDV) of rabbits and the European brown hare syndrome virus (EBHSV) of hares that cause lethal hepatitis. In 2010, a new RHDV related virus (RHDV2) with a unique genetic and antigenic profile and lower virulence was identified in France in rabbits. Here we report the identification of RHDV2 as the cause in Sardinia of several outbreaks of acute hepatitis in rabbits and Cape hare (Lepus capensis mediterraneus). This is the first account of a lagovirus that causes fatal hepatitis in both rabbits and hares.  相似文献

The prevalence of human pathogenic Yersinia enterocolitica isolates in livestock farming is of paramount interest. Raw goat milk has been proposed as a source of human yersiniosis; however, no data on the prevalence of human strains of Y. enterocolitica in goat herds are available. Therefore, fecal samples (n = 575) were collected from 24 goat herds from Lower Saxony, northern Germany. Pre-enrichment in peptone, sorbitol and bile salts broth was followed by plating on cefsuloidin irgasan novobiocin agar. Yersinia enterocolitica was isolated from 17 (3%) samples of five (21%) goat herds. All isolates were biovar 1A, but represented various serovars. PCR assays targeting Yersinia adhesin (yad) gene and the yopT gene, both associated with pathogenicity, produced no amplification products. Therefore, the isolates can be regarded as opportunistic apathogenic bacteria. Consequently, milk, cheese or meat from goats should not be considered as an important source for human yersiniosis.  相似文献

Attempts were made to define the Brucella abortus and Yersinia enterocolitica IX infection status of animal populations by means of selected agglutination tests. The Brucella abortus O, the Yersinia enterocolitica IX OH and the Y enterocolitica IX H agglutinin titres were measured in a large number of cattle, goat and pig sera In the goats and, to a much lesser extent, the pigs, the relationships between these titres suggested that Yersinia infection was common. In contrast, the results from the cattle sera were complex and tended to indicate the presence of both Yersinia infection and brucellosis.  相似文献

A polyphasic characterization of atypical isolates of Yersinia ruckeri (causative agent of enteric redmouth disease in trout) obtained from hatchery-reared brown trout Salmo trutta in South Carolina was performed. The Y. ruckeri isolates were biochemically and genetically distinct from reference cultures, including the type strain, but were unequivocally ascribed to the species Y. ruckeri, based on API 20E, VITEK, fatty acid methyl ester profiles, and 16S rRNA gene sequencing analysis. These isolates were nonmotile and unable to hydrolyze Tween 20/80 and were therefore classified as Y. ruckeri biotype 2. Genetic fingerprint typing of the isolates via enterobacterial repetitive intergenic consensus (amplified by polymerase chain reaction) and fragment length polymorphism showed biotype 2 as a homogeneous group distinguishable from other Y. ruckeri isolates. This is the first report of Y. ruckeri biotype 2 in the USA.  相似文献

OBJECTIVE: To determine the temporal pattern of Yersinia infections in three goat flocks and examine the influence of management and seasonal factors on the incidence of those infections over a 1-year period. METHODS: A longitudinal study involving monthly culture of faeces for Yersinia spp. from age groups of randomly selected goats on three farms in the Manawatu region of New Zealand. RESULTS: The incidence of excretion of potentially pathogenic Yersinia (Yersinia pseudotuberculosis and Y enterocolitica biotypes 2, 3 and 5) peaked in winter and fell in summer. In contrast, environmental Yersinia (Y enterocolitica biotype 1A, Y frederiksenii, Y intermedia and Y rohdei) showed no clear pattern of seasonal variation. Pathogenic Yersinia were more prevalent in young animals than in adults, while environmental Yersinia were more prevalent in adults. The same type was isolated from the same animal in two or more successive months in about 20 to 25% of cases, and in the remaining cases there was a gap of at least one month between successive isolations, with many animals yielding a particular type on only a single occasion. A notable difference was that with the potentially pathogenic types, no animal had more than one period of time when it was found to be excreting a particular type, suggesting that immunity develops following exposure. In contrast, it was common for environmental types to be isolated from the same animal throughout the study period. Two goats were suspected to have developed clinical yersiniosis but all remaining infected animals showed no clinical signs of infection. CONCLUSIONS: Asymptomatic Yersinia carriage was common in goats in New Zealand, with a clear seasonal and age group pattern of infection with potentially pathogenic types. There was evidence that immunity developed to potentially pathogenic types. This is the first time that Y rohdei has been isolated from goats.  相似文献

Yersiniosis is considered to be an emerging infection in humans. It is believed that contaminated pork is the main source of infection for man but also beef and cow milk might cause infection in the consumer. Therefore, a survey was carried out to obtain actual data on the prevalence of anti-Yersinia antibodies within cattle. 600 serum samples were taken form 120 farms located in 43 districts of Bavaria, Germany. Antibodies were detected in 396 animals (65.7%) revealing a distribution of 52.3% of anti-YopD, 40.3% of anti-YopM, 24.0% of anti-YopH, 21.2% of anti-YopE and 3.0% of anti-V-antigen antibodies. 21 reactionpatterns out of 32 possible patterns were identified. Anti-YopD and YopM antibodies were sufficient to detect all Yersinia antibody positive sera. No regional differences in the distribution of antibodies were noted.  相似文献

Fecal immunoglobulin-A (IgA) antibodies to Yersinia enterocolitica serovar O3 strain were detected in the mice orally immunized with formalin-killed organisms. Y. enterocolitica O3 organisms were inhibited to colonize in the intestines of the mice producing fecal IgA. The fecal IgA antibodies were detected in the mice orally infected with the bacteria. When IgA was produced in the mice infected, they ceased shedding the organisms in their feces.  相似文献

Brucella and Yersinia enterocolitca O:9 (Y.e.O:9) have antigenic determinants in common causing cross-reactions which interfere strongly in Brucella serology. Hoffmann (1988) observed cross-reactivity between Y.e.O:9, Y.e.O:5, and Y.e.O:6 and used an ELISA for the differentiation between Brucella- and Yersinia antibodies since Y.e.O:5 and Y.e.O:6 produced only low-level cross-reactions with Brucella antigen. The observations of Hoffmann were confirmed and led to the construction of an ELISA with 4 antigens. Sera from natural outbreaks of brucellosis and from artificial infections with Brucella and Y.e.O:9 were properly identifiable by this system. Furthermore, the test material has been used to measure the antibody avidity by a specially designed ELISA. This test produced a further divergence between specific and non-specific reactions.  相似文献

A free-living brown hare (Lepus europaeus occidentalis) trapped during a survey of tuberculous possums in the Ahaura Valley, Westland, was found to have both microbiological and histopathological evidence of infection with Mycobacterium bovis. The mesenteric lymph nodes were enlarged and showed extensive caseation, tuberculous nodules were present in the liver and kidney, and there was a proliferative pleurisy. Histologically, characteristic tuberculous granulomatous foci were seen in the mesenteric lymph nodes, liver, kidney, pleura and lungs, often in association with small numbers of acid-fast organisms. This is the first report of naturally occurring tuberculosis due to M. bovis in the brown hare.  相似文献

Abstract

Juvenile rainbow trout Oncorhynchus mykiss and brown trout Salmo trutta acclimated to freshwater or salinities of 9.0‰ or less were exposed to Yersinia ruckeri, the bacterial pathogen that causes enteric redmouth disease (ERM). Both species of fish were kept in the same recirculating systems after bacterial exposure. Rainbow trout mortality was significantly (P < 0.05) different in each salinity: 96.5% in freshwater, 89.5% in water of 1.1‰ salinity, 81.3% in 3.0‰ salinity, and 75.0% in 9.0‰ salinity (model SE = 1.0). All deaths occurred between 3 and 12 d after exposure to Y. ruckeri. Only 2.3% of brown trout in all salinities died, and differences among treatments were not significant. For both fish species, Y. ruckeri was isolated from liver, spleen, and trunk kidney of fish dying during this experiment, and lesions of rainbow trout were consistent with ERM. Yersinia ruckeri was not isolated from brown trout surviving for 21 d after bacterial exposure but was isolated from 3 of 24 surviving rainbow trout; a polymerase chain reaction assay detected the DNA of Y. ruckeri in 3 additional rainbow trout survivors. Neither the lesions of fish with ERM nor the percentage of surviving fish subclinically infected with Y. ruckeri was affected by salinity. Bacterial growth in vitro was not affected by low (≤9.0‰) salinity; however, bacterial adhesion to polystyrene was significantly reduced as salinity increased. Although mortality caused by Y. ruckeri was significantly lower for rainbow trout in water with slightly increased salinity, none of the salinities tested was effective in preventing serious losses caused by this pathogen in recirculating systems.  相似文献
