Summary Four potato lines of cv. Désirée that express the pectate lyase (PL) isoenzyme 3 ofErwinia carotovora subsp.atroseptica were examined in a 4-year field experiment with respect to plant development, tuber yield and resistance of tuber tissue toErwinia soft rot. The PL3 degrades plant cell wall pectin into unsaturated oligogalacturonates eliciting plant defence responses. In one line, enzyme expression was controlled by the CaMV 35S promoter (C) and in three lines it was driven by the potato patatin B33 promoter (D). Plant development of the D-lines in field plots was not distinguishable from that of the non-transgenic counterpart. Also tuber yield was not too different. By contrast, plants of the C-line were smaller than those of the nontransformed counterpart and also showed reduced tuber yield. There were no significant differences in dry mass, starch and protein content of tuber tissue between PL transgenic and non-transgenic potatoes. But compared with the latter, field-grown tubers expressing the PL displayed an enhanced resistance toErwinia soft rot. Thus, average rotting caused byEc-bacteria was diminished in tubers of PL-transgenic lines by 34.1%. The resistance of tubers toEc soft rot was significantly correlated with the PPO activity in tuber tissue.  相似文献

Summary Optimum storage conditions to identify resistance to silver scurf among commercially grown potato cultivars were determined in a series of experiments. Inoculation of field-grown tubers with a conidial suspension ofH. solani (concentration 104 conidia per ml of water) and incubation at 15°C with 95% RH for 1 month followed by 2 months at 85% RH produced the greatest differences in disease severity between potato cultivars. Results with glasshouse-grown tubers were comparable with those from field-grown tubers, indicating that the test could be used to identify resistance in wild species of potato and to understand the inheritance of this resistance.  相似文献

The goal of these studies was to evaluate the influence of tuber age on the expression of resistance to Phytophthora infestans in five selected potato genotypes (three cultivars and two clones) varying in tuber resistance and maturity type, and also to select the best time for testing potato tubers for resistance to blight. Resistance was evaluated by inoculating whole tubers at 13 different times as they progressively aged during growth and storage. Tuber age was expressed as weeks after planting. Tubers were rated from week 13 (late July) until week 43 (late February) for tubers planted on 27–28 April, over three consecutive seasons (2001–2003). Analysis of variance performed on tuber resistance data showed significant effects of genotype, year and tuber age. Significant contributions of genotype × year, genotype × tuber age, year × tuber age and year × genotype × tuber age interactions were also detected. A slight increase in tuber resistance with tuber age was observed for cvs Bzura, Sokół and Irga, while the opposite trend was observed for the clones 97-A-63 and DG 92-227. Changes in tuber resistance with age of tested genotypes were not related to their maturity type. The period of most stable expression of tuber resistance was observed when tubers were tested between the 16th and 28th week after planting.  相似文献

Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.  相似文献

Summary Solanum tuberosum potato cultivars with differing levels of resistance to dry rot caused byFusarium sulphureum were hybridised in a half diallel crossing programme. Glasshouse grown tubers from true seedlings and field-grown tubers of the resulting progenies were assessed for their resistance toF. sulphureum. Statistical analysis revealed differences between the progenies forF. sulphureum resistance on both field and glasshouse grown tubers. On the field grown tubers, differences between progenies were due to differences in the general combining ability (GCA) of the parents. Parental phenotypes and GCA values were highly correlated. Overall. GCA was consistent for resistance or susceptibility, and glasshouse grown progenies showed a good correlation for GCA between years (r=0.85***), and with the results of the field grown progenies (r=0.61* in 1998 and r=0.69** in 1999). It appears that resistance toF. sulphureum is heritable, and that this heritability is better assessed on field grown rather than glasshouse grown tubers, although the two are largely in agreement.  相似文献

From 1993 to 1996 three groups of potato genotypes were evaluated for resistance toErwinia spp.: (1) 31 interspecific diploid hybrids (28 resistant and three susceptible), (2) five hexaploid or pentaploid somatic hybrids ofSolanum tuberosum (tbr) ×S. brevidens (brd), and (3) eight cultivars. Two evaluation methods were applied: tuber point inoculation with eitherErwinia carotovora ssp.atroseptica (Eca) orE. chrysanthemi (Ech) to test tuber soft rot resistance and stem base inoculation with Eca to test blackleg resistance. Some resistant diploid hybrids and somatic hybrids oftbr × brd were superior to cultivars for both tuber and stem resistance. Tuber and stem resistance toErwinia spp. in the most resistant diploid hybrids were equal to the highly resistant somatic hybrids oftbr xbrd. Tuber resistance to Eca was highly correlated to tuber resistance to Ech (r=0.815***). In two years of evaluation for stem resistance, three diploid hybrids and a derivative of one of the somatic hybrids (USA M 264) failed to develop symptoms of blackleg following inoculation with Eca, Analyses of variance for tuber and stem resistance indicated significant effects of genotype, year and genotype × year interaction. A positive relationship between tuber and stem resistance toErwinia spp. has been found, however the genetic control of resistance in tuber and stem is partially independent. In the case of Eca the correlation coefficient was r = 0.725***. Therefore it should be possible to obtain resistant genotypes to both blackleg and tuber soft rot. Several resistant diploid hybrids were selected from among those tested, which also have several other characters desirable for potato breeders.  相似文献

Summary A crop rotation experiment was carried out on a light sandy soil in 1979–1986 to study the effects of the frequency of potato cropping on yield, quality and on the occurrence of soil-borne pathogens other than potato cyst nematodes. Tuber yield decreased markedly with increasing cropping frequency, but also depended on what crops were grown in rotation with potato. Growth during the early part of the season, as well as the length of the growing period, were reduced in short rotations. The fungiVerticillium dahliae andRhizoctonia solani, and root-knot nematodes (Meloidogyne spp.) were the most important yield reducing pathogens. The effects of rotation depended on the cultivar used. The percentage of mis-shapen tubers increased with increasing cropping frequency and after application of granular nematicides, but the incidence of common scab (Streptomyces scabies) was not affected.  相似文献

Summary Samples of a sandy soil and a marine clay soil sterilized by steam were put in 55-1 containers insulated with polystyrene and placed outdoors on a brick pavement. Sandy soil was infested singly or in all possible combinations with root-knot nematodes (Meloidogyne spp.) and the fungiRhizoctonia solani andVerticillium dahliae, and the marine clay soil was infested with the root-lesion nematodePratylenchus neglectus and the same fungi to evaluate the effects of these organisms on the yield of potato. The experiments were carried out from 1983 to 1986. Tuber yield was reduced by single infestations of theMeloidogyne spp. andV. dahliae but not significantly byR. solani orP. neglectus. A three-factor interaction: nematode ×R. solani × V. dahliae was found in both experiments.R. solani andV. dahliae showed significant synergistic effects when soil was infested with theMeloidogyne spp. orP. neglectus.  相似文献

Summary Available information has been reviewed on tuber resistance toP. infestans, its inheritance and breeding procedures used to obtain potato cultivars superior in this character. P. infestans is worldwide the most destructive potato pathogen. Tuber resistance is an essential component of potato resistance as this pathogen is often responsible for tuber rot in storage, and infected tubers of susceptible cultivars provide an important way for its overwintering. In Europe many cultivars and advanced breeders selections have been obtained with tuber resistance toP. infestans, but in the last decades no progress is noted in the mean level of this resistance in cultivars. The expression of tuber resistance depends to a large extent on testing conditions and therefore it can be difficult to evaluate. Consequently making progress in breeding is not easy, and also cultivar assessment data from various countries sometimes differ considerably. It is concluded from published data that it should be easier to make progress in breeding potato cultivars with resistant tubers if more attention is paid to combining the various types of resistance which are already known, and if the genetic determination of durable resistance toP. infestans is better understood. Summary Tubers of clones from a diploid hybrid population ofSolanum phureja andS. stenotomum were screened for resistance to tuber soft rot caused by strains ofErwinia carotovora subsp.atroseptica (Eca),E. carotovora subsp.carotovora (Ecc) andE. chrysanthemi (Ech). Significant, positive correlations between resistance to the different strains and species were observed, indicating that screening for resistance to tuber soft rot could be accomplished by using just one of the three pathogens. Strains of Ecc and Ech were found to be much more virulent than strains of Eca. Inoculating with Ecc and Ech resulted in more distinct differences among susceptible, moderately resistant and resistant clones than inoculating with Eca.  相似文献

Summary The severity of late blight tuber rot in five potato cultivars (Green Mountain, Sebago, Dorita, AC Brador and Island Sunshine) resulting from inoculation with US-1/US-8 combinations of Canadian isolates ofPhytophthora infestans was evaluated. In addition, the potential for mixed (A1∶A2) inoculations to result in oospore formation was examined. In studies conducted in 1996 and 1997, cv. Dorita (and Island Sunshine in 1997) developed the least amount of surface or internal necrosis relative to the other cultivars following co-inoculation with US-1 and US-8 isolates ofP. infestans. Differences also were revealed among isolate combinations used for inoculation. The combination of a US-1 isolate from Prince Edward Island with a US-8 isolate from New Brunswick consistently produced the least amount of necrosis in tubers of the various cultivars. Summary The resistance to common scab of nine cultivars and eight advanced breeding lines of potatoes, fiveSolanum andigenum Juz. et Buk. clones, and three potato breeding lines selected for resistance to common scab, was investigated in a field and a pot trial. In the pot trial potaroes were planted in inoculated coarse river sand in plastic pots (150 mm in diameter) placed on irrigated subsoil in a glassfibre house. In a field trial potatoes were planted in an artificially infested field. The harvested tubers were scored individually for scab incidence according to two scales, viz. a 1–5 point scale for percentage surface covered with scab and a 1–3 point scale for lesion type. A scab index value was calculated for each tuber. A significant correlation coefficient of 0.82 was obtained for the results of the two tests.  相似文献

Summary The resistance of ten potato cultivars (Agria, Ajax, Désirée, Liseta, Kennebec, Majestic. Monalisa, Prima, Spunta and Tonda di Berlino) toPhytophthora infestans was analyzed in vitro using 8 fungal strains. An assay based on electrolyte leakage was used for screening leaves and tuber tissues with fungal culture filtrates. With almost all cultivars the resistance of leaves did not correlate with the resistance of tubers. Cv. Ajax appeared the least susceptible in both leaf and tuber tests, while the cv. Summary The kinetics of accumulation of phytoalexins, glycoalkaloids and phenolics was studied in two potato cultivars differing in their degrees of field resistance when infected withPhytophthora infestans. Tuber slices and leaves of cvs Pampeana INTA (high degree of field resistance, free of R genes) and Bintje (susceptible) were infected with race C (complex race 1, 3, 5, 7, 11) ofPhytophthora infestans. Phytoalexins and phenolics accumulated in tuber and leaf tissues which had been inoculated. The levels of these compounds in the susceptible cv. Bintje were relatively low and similar to those found before inoculation. Leaves of cv. Pampeana INTA had a very high glycoalkaloid content, suggesting that glycoalkaloids may play a role in protection of leaves against the fungus. However, we could find no correlation between resistance and glycoalkaloid content of tubers. Our results suggest a major role of phytoalexins, phenolics and glycoalkaloids in the complex mechanisms of field resistance.  相似文献

Summary In artificial inoculations it was found thatFusarium oxysporum f.sp.tuberosi is able to penetrate through potato leaves. The fungus was isolated from the stem of infected plants from all inoculated cultivars. Tubers were completely destroyed if they were inoculated before sprouting. Not any destruction was observed if inoculation was made before young sprouts are longer than 1–3 cm; plants from these sprouts were infected. Lesioned tubers were more sensitive than unlesioned ones, and tuber rot and sprout damage were increased significantly.  相似文献

When 1-mo-old plants of a wilt-resistant clone ofSolanum phureja (1386.15) were stem-inoculated with three strains ofPseudomonas solanacearum (K60, S123, and S206), the bacteria multiplied rapidly at the point of inoculation and then moved in the vascular system to other parts of the stem. Resistant plants showed a remarkable ability to support relatively high populations of the bacterium in the absence of disease symptoms. Although multiplication in this resistant clone was substantially less than in susceptible Russet Burbank potato plants, large numbers of bacteria (up to 624 × 104 cells of K60 per 5-cm stem segment) reached the base of the stem of plants maintained at high temperature (28°C) for 20 days after stem inoculation. From the base of the stem, the bacteria moved rapidly into the roots and tubers. Strains ofP. solanacearum differed in their ability to cause latent tuber infection in different resistant potato clones. When 11S.phureja ×S. tuberosum hybrids were stem-inoculated, maintained at 28°C for 3 wk and then grown to maturity at 20°C., most of the clones yielded tubers infected by one or more strains. The race 1 strain (K60) was the most infectious; 53.8% of all tubers harvested from all plants inoculated with this isolate carried latent infections. Because one clone (BR 53.1) never yielded infected tubers, there appear to be genetic factors which may be useful in breeding programs aimed at eliminating latent tuber infection.  相似文献

Tubers from eight potato cultivars (cvs) grown at two different locations in Wisconsin were tested for bacterial soft rot resistance using different inoculation techniques. The procedures included 1) point inoculations of tubers with different inoculum levels followed by incubation in ambient or low oxygen condtions, 2) inoculation of mechanically bruised tubers followed by incubation in a mist chamber, and 3) a standard slice inoculation method. The point titration test followed by incubation in dew chamber and the mist chamber-bruise test showed similar patterns of resistance for cultivars that were used in these experiments. These two methods are considered to be effective for screening potato tubers for bacterial soft rot resistance. Point titration methods are very useful if only limited numbers of tubers are available. The mist chamber-bruise test is simpler than the other procedures; however, to obtain reproducible results large numbers of tubers are required. Because of the great variability of the results obtained in inoculation of slices, the reliability of this approach can be questioned as a standardized method for evaluation of resistance. Tubers of somatic hybrids ofS. brevidens andS. tuberosum and their sexual progeny were significantly more resistant to bacterial soft rot than tubers of moderately resistant cultivars when evaluated by each of the assay procedures.  相似文献

Summary The resistance toGlobodera pallida of individual potato seedlings from three progenies derived fromSolanum tuberosum ssp.andigena CPC 2802 was assessed using three inoculation treatments. Plants grown from tubers from the seedlings were used to reassess resistance. All the seedling test inoculation treatments over-estimated the proportion of clones classified as resistant compared to the results from tuber-grown plants. However, the mean relative resistance of progenies estimated by both tests showed good agreement, and it is suggested that seedlings are best used for assessing the resistance of progenies of crosses to identify promising combinations rather than individual clones.  相似文献

Summary Survival of the pathogen causing silver scurf of potato (Helminthosporium solani) in Alberta soils was studied by evaluatingH. solani infection in the progeny ofH. solani-free nuclear seed potato tubers planted in fields where potatoes had never been grown or were grown 1, 2, 3, or 4 years previously. Daughter tubers from all the fields developed silver scurf lesions, andH. solani was isolated from infected tubers. This is the first report of survival ofH. solani in Alberta soils. Soil-borne inoculum appears to have a role in the epidemiology of the disease and in the introduction of the pathogen into silver scurf-free potato seed stock. Of 31 plant species tested, only potato was found to be a host ofH. solani. Most of theH. solani isolates from north central Alberta were more sensitive to thiabendazole than those from southern Alberta, where thiabendazole is much more commonly used.  相似文献

Summary The formation of above-ground tubers on stem cuttings of eight potato cultivars was studied over three seasons. To promote tuber formation in the leaf axils, stems grown from single node cuttings were exposed to short day conditions. Tuber formation underground was reduced by covering the soil with a plastic sheet and by using single node cuttings planted with the leaf axils several centimetres above the soil surface. With all cultivars except Alpha, which produced mis-shapen tubers in all experiments, an average of 11 (maximum 40) tubers per cutting were harvested.
