Potato storage tuber rots caused by the late blight and pink rot pathogens at harvest can cause severe economic losses, warranting the need for effective post-harvest fungicide applications. The purpose of this study was to evaluate the efficacy of select post-harvest fungicides in reducing tuber infections by the late blight and pink rot pathogens when applied at various post-inoculation time intervals. ‘Russet Burbank’ potatoes were inoculated by submersion in an aqueous suspension ofPhytophthora infestans orPhytophthora erythroseptica zoospores at 0, 1, 2, 4, and 6 h prior to receiving a post-harvest treatment. Products evaluated were zoxamide (various rates and formulation), phosphite (335 g a.i./MT), and a hydrogen peroxide/peroxyacetic acid mixture (HPPA, 9 g a.i./MT), all applied at 2.08 L/MT of tubers as a low pressure spray prior to storage. Zoxamide and phosphite significantly reduced late blight and pink rot incidence and severity when applied immediately after inoculation. HPPA was less effective at controlling disease development. Phosphite was effective at reducing late blight development at all time intervals up to 6 h post-inoculation (7% vs 80% in untreated). Zoxamide appeared to have good post-harvest disease control if applied soon after inoculation. The maximum time intervals between inoculation and treatment where significant reductions in pink rot incidence were observed was 0 h for HPPA (28%), 2 h for zoxamide (55%; 64 g a.i./MT) and 6 h for phosphite (13%) compared to the untreated (73%). Phosphite provided consistent disease control even when applied several h after inoculation and has potential to be a reliable post-harvest fungicide for the potato industry.  相似文献

The Mainestay potato variety is high-yielding and late-maturing producing attractive, round, white-skinned, white-fleshed tubers with medium-shallow eyes. Its major use is expected to be as a maincrop tablestock variety. Mainestay appearance ratings are similar to those of Katahdin, and greening in Mainestay tubers is less than in Kennebec. Most boil and bake qualities were rated equal to Katahdin in at least half of the tests, and Mainestay usually has a low incidence of internal defects. However, purple streak, heat necrosis, vascular discoloration, and heat sprouts have been seen on occasion. Mainestay tubers do not show the net necrosis caused by potato leafroll virus, and have shown very little hollow heart or blackspot bruising. Mainestay is susceptible to common scab and acid scab, but moderately resistant to verticillium wilt (Verticillium, albo-atrum andV. dahliae), early blight (Alternaria solani), rhizoctonia (Rhizoctonia solani), dry rot (Fusarium sambucinum), and pink rot (Phytophthora erythroseptica). It is moderately susceptible to the common race of late blight (Phytophthora infestans). Mainestay tubers are slow to mature after vine kill, and additional time is required prior to harvest in order to allow for proper periderm development and to reduce storage losses.  相似文献

Glycoalkaloids are anti-nutritional compounds commonly found in wildSolanum species used as resistance sources to major potato pathogens. It is therefore important for breeding purposes to know whether selecting for resistance using such species necessarily selects also for high glycoalkaloid contents in the tubers. To test this hypothesis, we used six partial progenies from crosses betweenSolanum tuberosum and accessions ofS. andigena, S. berthaultii, S. phureja, andS. vernei to investigate the possible correlation between resistance toPhytophthora infestans and/or toErwinia carotovora subsp.atroseptica and the concentration of glycoalkaloids in tubers. Concentrations of α-solanine and α-chaconine in the tubers segregated in each progeny, as did resistance to each pathogen. Some, but not all, clones from each progeny showed hypersensitive reactions to the isolate ofP. infestans used. Furthermore, clones within each progeny also differed for components of partial resistance toP. infestans, suggesting that all four wild species could be used as sources of both race-specific and partial resistance to late blight. With the exception of low, but statistically significant, correlations between concentration of α-solanine and two late blight resistance components (incubation period and spore production per unit lesion area) in progenies derived fromS. vernei, and despite a trend towards higher glycoalkaloid concentrations in the tubers of the clones most resistant to soft rot within progenies derived fromS. berthaultii andS. vernei, no consistent relationship between resistance to either disease and concentrations of α-solanine and/or α-chaconine was observed. These results indicate that neither race-specific nor partial resistance to late blight and soft rot in the accessions used as progenitors of resistance depend on high solanine or chaconine concentrations. These resistance sources could thus prove useful in breeding programs for improved behaviour againstP. infestans and/orE. carotovora.  相似文献

As part of a project to investigate the possibility that teratogenic factors are formed in potato tubers infected with the late blight fungus,Phytophthora infestans, a method was devised which allowed quick, efficient inoculation of large numbers of potato tubers. Tubers were placed in specially constructed trays, wounded by puncturing the periderm with flower “pin frogs”, and sprayed with a zoospore suspensionof P. infestans prepared from cultures grown on rye agar at 20C (68F) for 10 days. Approximately 95% of Katahdin and 93% of Russet Sebago tubers inoculated by this method exhibited typical late blight lesions after two weeks incubation at 20C, 80–90% RH. Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.  相似文献

The potato cultivar Defender is high-yielding, white-skinned, and notable for having foliar and tuber resistance to late blight infection caused byPhytophthora infestans (Mont.) de Bary. It was released in 2004 by the USDA-ARS and the agricultural experiment stations of Idaho, Oregon, and Washington. Defender is suitable for processing into french fries and other frozen potato products directly from the field or from storage. Defender also may be used for fresh markets in regions such as California, where cultivars with long tubers and white skin are traditionally grown. Resistances to late blight and other potato diseases make Defender an ideal candidate for organic potato production. Defender consistently produced greater total and U.S. No. 1 yields than ‘Russet Burbank’ in Idaho trials. In early harvest trials conducted in the western U.S., average total yields of Defender were 17% and 23% greater than yields for ‘Ranger Russet’ and ‘Shepody’, respectively. In full-season trials conducted in the western U.S., Defender averaged 10% and 15% higher yields than Ranger Russet and Russet Burbank, respectively. Specific gravity of Defender is consistently high, with values comparable to those of Ranger Russet; tuber ascorbic acid (Vitamin C) levels are also high. In addition to late blight, Defender also is resistant to tuber early blight (Alternaria solani, (Ellis & G. Martin), L.R. Jones & Grout), potato virus X, and net necrosis; it has moderate levels of resistance to Verticillium wilt (Verticillium dahliae, Kleb), pink rot, foliar early blight, corky ringspot, and Erwinia soft rot. Defender is susceptible to scab (common and powdery) and potato leafroll virus; it has moderate susceptibility to dry rot (Fusarium spp.) and potato virus Y. Susceptibilities to internal necrosis, tuber greening, and blackspot bruise also have been noted, but may be minimized through cultural and harvest practices.  相似文献

Annual epidemics of late blight of potato have lead to debate as to the relative importance of overwintering sources of inoculum. Host availability is a key factor for overwinter survival ofPhytophthora infestans and the initiation of epidemics of potato late blight. Temperature within discarded piles of cull potatoes may influence tuber tissue temperature and therefore affect survival of meristematic tissue. Consequently the risk of initiation of an epidemic of late blight from cull piles is closely related to the thermal experience of overwintered potato culls. Temperature monitoring of cull piles over two years indicated that temperature was stable in the interior of the piles regardless of cull pile size (1–15 ton) or year. The possible use of ambient temperature information gathered during winter months in potato-growing regions may not help estimate the risk of an epidemic of late blight initiated from cull piles as, although highly correlated with the surface temperature of cull piles, ambient temperature was not correlated with the temperature within the cull piles. The presence of cull piles in excess of 1 ton may enhance the survival of the host and thus the development ofP. infestans from infected tubers.  相似文献

Due to changes in the pathogen population, late blight of potatoes caused byPhytophthora infestans has become a more difficult disease to manage and there is a need for new potato cultivars with greater and more stable disease resistance. Field studies are required to identify and characterize host resistance to late blight in both foliage and tubers but, as epidemics vary from year to year, combining multi-year results can be difficult. Residual maximum likelihood (REML) was successfully used to analyze data which demonstrated good correlations between foliar and tuber responses to late blight (US-1 genotype). In addition, studies on post-harvest inoculated tubers provided more reliable disease responses than field studies, and the disease resistance with this method correlated well with foliar disease responses. This tuber test would be a useful method of pre-screening selections for disease response prior to testing in the field.  相似文献

The Quaggy Joe potato variety is high-yielding and medium to medium-late maturing with attractive, round-oval, white-skinned, white-fleshed tubers with medium-shallow pink eyes. Quaggy Joe is resistant to the potato leafroll virus, and is also moderately resistant to early blight (Alternaria solani), dry rot (Fusarium sambucinum), and soft rot (Erwinia carotovora subsp.carotovora). The distribution of A1 and A2 mating types, and of metalaxyl-sensitive (MS), metalaxyl-intermediate (MI), and metalaxyl-resistant (MR) strains ofPhytophthora infestans, the causal pathogen of late blight, were determined in Canada in 1997. Potato plants or tubers and tomato plants or fruits, suspected to have late blight infection, were received from seven Canadian provinces in 1997, and were assessed for the presence ofP. infestans. Among 220 samples suspected to have late blight,P. infestans was recovered from 132 samples (60%) received from British Columbia (BC), Manitoba, New Brunswick, Prince Edward Island, and Quebec. From the 132 samples, 364 isolates were obtained and tested for their mating type and response to metalaxyl. Populations ofP. infestans within Canadian provinces were homogeneous for mating types, except within BC, where populations comprised both A1 and A2 mating types both within fields and sometimes within samples. The A2 mating type was found in all provinces where late blight was present. The A1 mating type was found only in Manitoba (1 isolate) and in BC (38 isolates). Unlike the old A1 isolates, those found in BC in 1997 were highly resistant to metalaxyl. These A1 isolates are even more resistant than A2 isolates. Based on assays with 10 ug.ml?1 of metalaxyl, 14% of all tested isolates were MS, 69.8% were MI and 16.2% were MR. In addition, all provinces contained MS, MI, and MR isolates. No specific mating type or metalaxyl sensitivity level were specific to tomato or potato.  相似文献

Summary Host plant resistance is an important component to the management of potato late blight,Phytophthora infestans (Mont.) de Bary. Assessment of potato lines (Solanum tuberosum L.) with various levels of resistance toP. infestans (US8, A2 genotype) were evaluated in field trials, greenhouse controlled environment chambers and inoculated tuber reactions. Five lines (AWN86514-2, B0692-4, B0718-3, Jacqueline Lee, and B0288-17) with strong foliar resistance to late blight were identified in these inoculated field trials. Greenhouse controlled environment chamber studies allowed resistant and susceptible lines to be distinguished, but the 1998 greenhouse results did not correlate well with field data. Four lines (A084275-3, Bzura, MSG007-1, and MSG297-4RD) evaluated by a digital image analysis technique demonstrated tuber resistance based upon average reflective index values in the inoculated tuber studies. Tuber resistance did not correlate with field foliar resistance. Based upon these results, field assessment of foliar reaction toP. infestans provides the best measure for assessing late blight resistance in potato. Tuber resistance to late blight can be identified among lines with varying levels of foliar resistance.  相似文献

The efficacy of Chaetomium globosum as a biocontrol agent against the late blight pathogen Phytophthora infestans was evaluated in potato plants. Among eight Chaetomium isolates evaluated C. globosum isolate Cg-6 showed greater inhibition to mycelial growth of P. infestans in vitro. TLC studies showed that isolate Cg-6 produced an antibiotic called ‘Chaetomin’. Isolate Cg-6 showed greater exo- and endo-glucanase enzyme activity when compared to other isolates. PCR amplification of the ITS region and sequencing of the PCR product confirmed that isolate Cg-6 belongs to the C. globosum group. C. globosum Cg-6 was formulated as a liquid and applied as a tuber, soil and foliar treatment either individually or in combination against Phytophthora infection in potato plants. Among different treatments, combined application of C. globosum as a tuber treatment @ 1 ml/kg of tubers, as a soil application @ 1 ml/kg of Farm Yard Manure (FYM) and foliar spray @ 0.7% resulted in significantly less late blight infection (72%) compared to untreated control (100%) under field conditions. The application of C. globosum resulted in greater tuber yield by reducing late blight infection in two field trials when compared to untreated controls. The study clearly demonstrated the potential use of C. globosum as a biocontrol agent in the management of late blight disease in potato plants.  相似文献

Wild potatoes are important sources of genes for resistance to disease and insect pests. A collection of wild Mexican and South AmericanSolarium species from the US potato Genebank was evaluated under laboratory and/or field conditions for their reaction to late blight (Phytophthora infestans), Colorado potato beetle (CPB,Leptinotarsa decemlineata Say), and blackleg (Erwinia carotovora subsp.atroseptica (van Hall) Dye) in order to identify individual genotypes with multiple resistance genes. Late blight inoculations using aggressive isolates (US-8/A2 and US-11/A1 mating types) of P.infestans revealed a wide range of variation for resistance between and within the accessions of the wild species tested. For late blight, susceptible as well as moderately to highly resistant genotypes were observed in all the species tested. However, at least one accession from the three Mexican and one South American wild diploid species tested showed a relatively uniform high level of resistance toP. infestans. These includedS. bulbocastanum, S. pinnatisectum, S. cardiophyllum, andS. circaeifolium. Two accessions from South American speciesS. commersonii were highly susceptible to late blight. For the Colorado potato beetle test, only one species,S. pinnatisectum appeared uniformly resistant to CPB under field conditions. Results of screening for blackleg resistance showed that there were major differences between genotypes in the wild species. Accessions ofS. circaeifolium PI 498119 andS. bulbocastanum PI 243504 were identified as having significantly higher blackleg resistance than cultivated potato and the other wild species tested. However, genotypes from these two accessions were more susceptible to late blight and CPB. Characterization of theP. infestans isolate P1801C.16 used for late blight evaluation and multi-locus isolate tests using US-8/A2 and US-11/A1 races revealed that the resistance inS. pinnatisectum genotypes tested corresponded to a race-non-specific genetic system, which was different from any existing R genes.Solanum pin-natisectum genotypes with both high levels of late blight and CPB resistance as well as blackleg resistance genotypes identified in the present study represent a diverse gene pool that may be useful for development of new potato cultivars with multiple disease and insect resistance. The potential utilization of these valuable sources for improvement of cultivated potato is discussed.  相似文献

In northwest Washington State, fungicidal control of late blight caused byPhytophthora infestons was evaluated for three years. During 1977, a high rainfall year with supplemental sprinkler irrigations, all metalaxyl and chlorothalonil treatments reduced tuber rot at harvest, but only metalaxyl extended control of tuber rot after 4 months’ storage. During 1978, a high rainfall year, one application of metalaxyl sprayed over or dusted on potato seed pieces at planting or applied five times to foliage at 2 or 4 wk intervals gave excellent blight control equal to that with chlorothalonil applied to foliage every 2 wk. In 1979, a low rainfall year,P. infestons did not kill plants in untreated plots until October. Five applications of metalaxyl sprayed every 2 wk on foliage did not control foliage blight. However, metalaxyl reduced foliage blight when dusted on cut potato seed pieces. Mancozeb sprayed every 2 wk also gave excellent control. The possibility that metalaxyl applied to foliage is effective in late blight control only when washed into the soil by rainfall or irrigation for uptake by roots is discussed. Yields and percent U.S. No. 1 grade tubers were not significantly affected by any treatment in these 3-year trials.  相似文献

Corynebacterium sepedonicum was detected in symptomless potato stems and tubers with immunofluorescence using monoclonal antibodies specific for the bacterial ring rot pathogen. The concentration of bacterial cells in potato tissue preparations ranged from >500 cells/microscope field to 1 cell per preparation. Symptomless tubers containing ring rot bacteria planted in field plots yielded plants with ring rot symptoms, plants with latent ring rot infections, or plants with no detectable levels of ring rot bacteria. Tubers with the greatest number of bacteria were most likely to develop plants expressing ring rot symptoms, but even some seed tubers with a low number of bacteria developed into plants with symptoms. Some seed tubers with high levels of ring rot bacteria produced plants with only low numbers ofC. sepedonicum.  相似文献

Summary The severity of late blight tuber rot in five potato cultivars (Green Mountain, Sebago, Dorita, AC Brador and Island Sunshine) resulting from inoculation with US-1/US-8 combinations of Canadian isolates ofPhytophthora infestans was evaluated. In addition, the potential for mixed (A1∶A2) inoculations to result in oospore formation was examined. In studies conducted in 1996 and 1997, cv. Dorita (and Island Sunshine in 1997) developed the least amount of surface or internal necrosis relative to the other cultivars following co-inoculation with US-1 and US-8 isolates ofP. infestans. Differences also were revealed among isolate combinations used for inoculation. The combination of a US-1 isolate from Prince Edward Island with a US-8 isolate from New Brunswick consistently produced the least amount of necrosis in tubers of the various cultivars. Outbreaks of late blight occurred in commercial potato fields in northwestern Washington in 1989 and reached epidemic proportions in 1990. Since most blighted fields reportedly had received 2–4 late-season applications of metalaxyl, the possibility existed that pathogenic strains ofPhytophthora infestans had developed resistance to this systemic fungicide. In testing this hypothesis,P. infestans was isolated consistently from diseased leaves, stems, and tubers of potato when small tissue pieces were surface-disinfected in 0.5% NaOCl for 3 min. Virulence of isolates was assessed on detached leaflets of eight differential (R-gene) potato genotypes. Although simple races (single and double) were recovered, there was also a high frequency of complex race combinations. When tested in metalaxyl-amended media, 81% of the 73 isolates recovered from diseased tissues were highly resistant to 10 μg metalaxyl/ml and 19% yielded intermediate resistance. Sensitive isolates were not recovered. Fifty-five percent of 40 isolates sporulated on potato tuber disks in the presence of ≥10 ug metalaxyl/ml. On leaf tissues, 85% of 20 isolates sporulated in the presence of 10 μg metalaxyl/ml, and 50% sporulated in the presence of 100 μg/ml. Late blight, caused byPhytophthora infestans, is the most important disease of potato worldwide and foliar resistance is an important component of managing late blight in the field. The objective of this research was to identify germplasm for use in breeding cultivars with foliar resistance toP. infestans. More than 500 clones were tested from 1997 to 2002 in inoculated (US8 genotype) field experiments conducted at the Michigan State University Muck Soils Research Farm in Bath, Michigan. All of the current commercial cultivars tested were classified as susceptible toP. infestans. The most resistant clones were A90586-11, AWN86514-2, B0718-3, Jacqueline Lee (MSG274-3), MSI152-A, MSJ307-2, MSJ317-1, MSJ453-4Y, MSJ456-2, MSJ456-4, MSJ461-1, MSK101-2, MSK128-1, NY121, LBR8, LBR9, Tollocan, and Torridon. Some of these resistant selections were from crosses with B0718-3, Jacqueline Lee, and Tollocan suggesting that the resistance toP. infestans was transmissible. These resistant clones will provide the opportunity to breed late-blight-resistant cultivars from a diverse pool of cultivated germplasm. Consistent foliar reaction toP. infestans over years suggested that the Michigan State University Muck Soils Research Farm is a valuable location for North American breeders to assess the reaction of potato germplasm to the US8 genotype of late blight.  相似文献

Response to foliar late blight for Superior and Sebago was not affected by the two potato seed selection and multiplication methods. Kennebec and Russet Burbank potato plants derived throughin vitro tissue culture techniques had significantly more late blight damage on only 4 of 7 and 2 of 7 observation dates, respectively, than plants derived through a clonal selection system. In practical terms, the disease response differences between the two seed propagation methods were minimal. Similarly, disease response differences among the 2 and 3 years of field multiplication for Russet Burbank and Kennebec, respectively, did not demonstrate any significant disease response trends. In general,in vitro culture plots had slightly higher yields than clonal plots but only Kennebec had a significant yield response. The incidence of late blight storage rot was generally not significantly different but clonally selected Kennebec potatoes had significantly more disease than those produced throughin vitro culture.  相似文献
