从中华倒刺鱼巴(Spinibarbus sinensis)血清分离到一种天然的凝集素,能够凝集多种不同的抗原物质,其组成成分为糖蛋白,具有耐热和耐酸碱性,对β-巯基乙醇敏感。凝集反应受温度、PBS离子浓度以及时间等因素的影响。经糖的专一性抑制实验显示,蔗糖和乳糖对凝集反应有抑制作用,说明这2种糖分子是凝集素在识别和凝集过程中起一定作用的糖分子。  相似文献

取日本沼虾血淋巴液,利用血凝实验和糖抑制实验测定其血清凝集素的凝血活性和糖专一性,对血清凝集素热稳定性、pH值、盐度、重金属离子、EDTA-Na2、有机磷农药、黄酮和多糖类物质对其活性的影响进行了探讨。结果显示:日本沼虾血清凝集素可以对兔、鸡、小鼠、蟾蜍和鲢鱼5种动物血红细胞产生凝集反应,以对小鼠和鸡血红细胞的凝集效价较高。能够专一性结合D-葡萄糖,D-果糖而产生抑制效果。在50~60℃范围内凝集活性最强,80℃及以上凝集素发生变性,无凝集活性。该凝集素pH值作用范围广泛,在pH值2~12都可以使鸡红细胞发生凝集,pH值3~7范围内凝集效果较好。盐度为13~15时,凝集活性最强。凝集素活性能被浓度高于16mmol/L(含16mmol/L)的EDTA-Na2完全抑制,对重金属离子Cu2+和Zn2+敏感性强,其活性被完全抑制。乙酰甲胺磷和敌百虫对日本沼虾血清凝集素活性有一定的影响,黄酮和多糖类物质可显著增强虾血清凝集素的凝集作用。  相似文献

通过血凝试验及糖抑制试验,测定褶纹冠蚌的血清及肌肉提取液的凝血活性及糖抑制专一性,同时进行热稳定性、pH及Ca2+影响试验.结果表明,血清和肌肉凝集素都可以凝集多种脊椎动物的红细胞,其中对鸡红细胞的凝集效价均为最高,分别为25和21;血清和肌肉凝集素都不能凝集大肠杆菌,均能凝集嗜水气单胞菌、枯草杆菌、变形杆菌和保加利亚乳杆菌.血清和肌肉凝集素都有一定的热稳定性,血清凝集素60℃处理10 min丧失凝集活性,而肌肉凝集素50℃处理10 min凝集活性丧失.血清凝集素具有较广泛的pH适应性,在pH 6~10内对鸡红细胞均有凝集活力,而肌肉提取液凝集素在pH<7或pH>9时凝集活性消失.血清凝集素能被乳糖和麦芽糖所抑制,肌肉凝集素不能被受试的7种糖所抑制.血清凝集素凝集活性明显地依赖于Ca2+等2价离子,而肌肉凝集素对Ca2+等2价离子没有依赖性.因此,认为褶纹冠蚌血清和肌肉提取液所含凝集素类型不同.用嗜水气单胞菌对褶纹冠蚌进行免疫刺激后,血清和肌肉凝集素对鸡红细胞的凝集效价并没有发生明显变化.  相似文献

通过用7种红细胞对3种海洋贝类褶牡蛎(Crassostrea plicatula)、缢蛏(Sinonova culaconstricta)和紫贻贝(Mytilus edulis)的凝集素进行凝血活性检测。结果表明:3种贝类凝集素对7种红细胞都有凝集活性,其中紫贻贝的凝集活性最高,家兔红细胞较为敏感。3种贝类凝集素对兔血细胞的凝集活性都可被10种糖不同程度地抑制,亦可被EDTA抑制。3种贝类凝集素在pH=4、pH=5的条件下仍然对兔血细胞显示出较高的凝集活性。缢蛏和紫贻贝的凝集素在80、90℃下处理10分钟,对兔血细胞反而表现出更高的凝集活性,而褶牡蛎的凝集素则在40℃时即丧失活性。  相似文献

用人的A、B、O、AB型血和兔、鸡、蛙的共7种血细胞检测4种软体动物凝集素粗制品的凝集活性。结果表明,除缢蛏对人AB型血红细胞,毛蚶对人B型血红细胞不表现凝集活性之外,不同来源的凝集素粗提物对供试血红细胞均有良好的凝集性能。兔血红细胞对各类凝集素最敏感。大多凝集素的pH适应性较广泛。不同凝集素热稳定性不同,缢蛏、毛蚶耐热性较好,80℃处理10min仍能凝集兔血红细胞。不同的糖、金属离子对凝集活性的影响不同,0·02mol/LEDTA可抑制全部凝集素的生物活性。  相似文献

从军山湖中华绒螯蟹(Eriocheir sinensis)获得血清,用人A、B、AB、O型红细胞、鸡红细胞及部分细菌菌体检测了血清的凝集活力,同时进行了糖抑制、热稳定性及pH影响试验。结果显示血清能凝集人四种血型红细胞和鸡红细胞,其中对人A型红细胞凝集活力最高;能够和大肠杆菌发生凝集反应,但不能和枯草芽孢杆菌、金黄色葡萄球菌发生凝集反应;凝集条件为pH5.0~5.8、温度4~37℃;1.0%的D-葡萄糖、D-果糖、蔗糖、乳糖、麦芽糖均能不同程度地抑制血清与人A型红细胞的凝集作用。  相似文献

试验表明九孔鲍血清中存在凝集素,可以凝集多种脊椎动物和人各型的血细胞,尤其对兔血细胞有很高的凝集效价(28)。该凝集素具有一定的热稳定性,40~50℃处理20 m in,其凝集活性稳定,70℃以上处理20 m in,凝集效价完全被破坏;pH 6~12凝集活性均保持最高值;Ca2 、Mg2 和ED-TA不影响其凝集活力;盐度对凝集活性影响不大,盐度为6~48,凝集活性一致;乳糖、D-甘露糖、D-果糖、D-半乳糖、D-葡萄糖、蔗糖、木糖、麦芽糖、L-鼠李糖和山梨糖等10种糖对其不产生凝集抑制作用。  相似文献

运用硫酸铵盐析和GlcNAc-Sepharose 6B亲合层析等方法从中华绒螯蟹的血清中分离出一种天然的凝集素,经SDS-PAGE测得其分子量约为82 kD,由单一亚基组成,由等电聚焦凝胶电泳测得其等电点(pI)为5.4.糖凝集抑制试验中检测到葡萄糖、半乳糖、甘露糖、乳糖、果糖和蔗糖对中华绒螯蟹血清凝集素均没有抑制作用,但N-乙酰氨基糖GIcNAc、GalNAc和ManNAc等能抑制其凝集活性.热变性试验结果表明,温度为10~50℃,凝集素仍保持强的凝集活性,但温度升至60℃后,凝集活性迅速下降,至80℃失去凝集活性,在pH 4.0~8.0的各缓冲液中保持较强的凝集活性,而在此pH范围外,凝集活性均有不同程度的下降.中华绒螯蟹血清凝集素可使外源红血细胞发生凝集,还具有凝集细菌或抗菌作用.  相似文献

用鼠红细胞膜纯化中华绒螯蟹血清凝集素的方法   总被引:1,自引:0,他引:1  
本研究建立鼠红细胞膜吸附法纯化中华绒螯蟹(Eriocheir sinensis)血清凝集素的方法,并与硫酸铵盐析、凝胶过滤层析和离子交换层析等其他提取方法进行比较。实验结果表明,中华绒螯蟹血清经鼠红细胞膜吸附后,再由EDTA把结合的凝集素从膜上解离,可以获得纯化的高活性凝集素。该纯化物在PAGE中出现2个清晰的区带,一个是大分子的b带,另一个则是由3种大小相近的小分子组成的c区带;在SDS-PAGE中,则显示出3个分子量接近、约为100 000的条带。经50%饱和硫酸铵盐析或用Sephadex G200凝胶过滤层析法可浓缩但不能纯化中华绒螯蟹血清凝集素。经DEAE-Sephadex A50离子交换层析法可获得2个蛋白峰,其中只有1个峰有凝集活性,在PAGE中表现为a蛋白带。凝集活性较高、分子量较大的蛋白带。上述实验结果表明,应用鼠红细胞膜法可纯化中华绒螯蟹血清凝集素。  相似文献

The knowledge concerning the humoral immunity is scarce in amphioxus Branchiostoma belcheri. This study measured the humoral parameters including phenoloxidase (PO) activity, lysozyme activity, antimicrobial activity, microbial agglutinin, and hemagglutinin in amphioxus humoral fluids before and after lipopolysaccharide (LPS) challenge. Humoral fluids from unchallenged Branchiostoma belcheri (B. belcheri) had PO activity, lysozyme, antimicrobial, microbial agglutinating, and hemagglutinating activities, which may represent part of the baseline level of innate immunity in this organism. After challenge with LPS, many humoral parameters were all increased significantly including the PO activity, lysozyme activity, growth-inhibiting activities against Gram-negative bacteria Escherichia coli (E. coli) and Vibrio alginolyticus (V. alginolyticus), growth-inhibiting activities against Gram-positive bacteria Staphylococcus aureus (S. aureus) and Bacillus subtilis (B. subtilis), microbial agglutinating activities against Micrococcus lysodeikticus (M. lysodeikticus), B. subtilis, and S. aureus, and hemagglutinating activities against rabbit and human A and O erythrocytes. In contrast, the agglutinating activities against V. harveyi and E. coli and the hemagglutinating activity against human B erythrocytes in the humoral fluids were reduced in response to LPS challenge. It appears that the humoral fluids of B. belcheri contain components that are able to differentiate different microbes and different human blood cell types.  相似文献

本文对栉孔扇贝血清的细菌凝集活性、抑菌活性和溶菌活性进行研究。选用24种细菌和真菌进行凝集试验,发现栉孔扇贝血清能够对金黄色葡萄球菌和代远洋弧菌产生凝集作用,血清的细菌凝集作用经数种细菌刺激没有发生改变,说明这种凝集作用是不可诱导的。栉孔扇贝的血清对某些细菌的生长有一定的抑制作用,但在体外彻底杀灭细菌的效果极差,经大肠杆菌和副溶血弧菌刺激后,抑菌效果基本不变。栉孔扇贝血清还具有一定的溶菌作用,而且大肠杆菌的刺激可以明显提高这种溶菌作用。  相似文献

对锯缘青蟹(Scylla serrate)血清凝集素的凝集特性进行了研究。结果显示:锯缘青蟹血清凝集素对人、兔、鼠、鸽子、中华鳖、鲫红细胞均有凝集作用,其中对小鼠红细胞凝集活性最强,血凝活性可达29;凝集素活性最适pH 6.0~7.4;盐度对凝集活力有较大影响,NaC l浓度在0.4~0.5 mol/L时基本失活;未纯化血清凝集素对Ca2+、Mg2+未表现出依赖性;凝集素活性能被N-乙酰葡萄糖胺特异性抑制。对青蟹血清凝集素进行硫酸铵分级沉淀分离后,发现凝集素活性主要分布在25%饱和度硫酸铵沉淀区,该组分凝集比活比血清高27.44倍,SDS-PAGE结果表明主要蛋白带型为79 ku、75 ku、72 ku。  相似文献

将阿维拉霉素按照一定的浓度添加到饲料中,投喂经注射接种福尔马林灭活的嗜水气单胞菌(Aerom onashydrophila)菌苗的鲤,于免疫后的第7 d、14 d、21 d、28 d和35 d检测供试鲤血液中的白细胞吞噬活性、血清中溶菌酶活性、抗体效价、补体C3含量及免疫保护率(RPS),探讨了阿维拉霉素对受免鲤免疫应答的影响。结果表明,饲料中添加阿维拉霉素(4 mg/kg和8 mg/kg),投喂较长时间(≥28 d)会降低受免鲤的血液白细胞吞噬活性、血清中的溶菌酶活性和抗体效价,减少受免鲤血清中补体C3的含量,减弱鲤抵抗嗜水气单胞菌人工感染的能力,每公斤饲料中添加8 mg的阿维拉霉素其免疫抑制作用更强。  相似文献

Uptake and clearance of Edwardsiella ictaluri in the peripheral blood of channel catfish Ictalurus punctatus fingerlings were monitored for 216 h after exposure to E. ictaluri for 4 h and 8 h under static conditions. Most fish exposed to E. ictaluri developed bacteriemia 24 h post-exposure, and by 72 h post-exposure E. ictaluri was recovered from all the blood of all exposed fish. The number of E. ictaluri colony forming units (CFU) in the blood of moribund fish ranged between 1.7 × 103 to 1.6 × 105 CFU/50 μL whole blood. Clearance of bacteria from the blood was observed by 216 h post-exposure and all fish surviving bacterial exposure developed agglutinating antibody against E. ictaluri . The pathogenesis of the infection was accompanied by the shedding of viable E. ictaluri into the water which may serve as a mechanism by which fish to fish transmission occurs.  相似文献

Gilthead sea bream, Sparus aurata, were subjected to different stressful situations and the haemolytic activity mediated by the alternative complement and the agglutinating serum activity were tested as indicators of immunocompetence. The results show that both parameters were significantly compromised after chronic or repeated acute stress as well as in groups fed with vitamin E or 3 highly unsaturated fatty acids (HUFA) deficient diets and in infected fish. These results suggest that both haemolytic and agglutinating activity are suitable indicators for immunocompetence as they are non-specific responses induced by non-specific stressors. In addition, a comparison is performed with other immune indicators subjected to the same stressors.  相似文献

Enterococcus seriolicida strains were divided into two groups, agglutinating and nonagglutinating, by a slide agglutination test using antiserum against the YT-3 strain. Intraperitoneal injection of agglutinating and nonagglutinating strains into yellowtail, Seriola quinqueradiata Temminck & Schlegel, revealed that nonagglutinating strains were more virulent than agglutinating strains. Two nonagglutinating and highly pathogenic strains SS91–014 and SS91–092 were subcultured 30 times in brain heart infusion broth, and the agglutination titres of 50 colonies from subcultures 1, 2, 3, 4, 5, 6, 11, 16, 21, 26 and 30 against anti-YT-3 serum were determined. Transformation from a nonagglutinating (1:<4) to an agglutinating (1: ≥4) pattern was first observed at the sixteenth subculture, and the ratio of agglutinating to nonagglutinating substrains rose until the thirtieth subculture. At this time, 70% of the SS91–014 population and 52% of the SS91–092 population were transformed to an agglutinating pattern. When the pathogenicity of four transformed substrains with different agglutination titres was tested in yellowtail, the nonagglutinating substrain showed higher pathogenicity than the agglutinating substrains, but no relationship between LD50 values and the agglutination titres of transformed substrains was observed. The pathogenicity of E. seriolicida appears to be related to the agglutination pattern, although it was not demonstrated that this property is solely responsible for pathogenicity.  相似文献

选取对鲤鱼毒力强的嗜水气单胞菌菌株制成灭活疫苗,研究疫苗的免疫保护效应.采用甲醛灭活制备疫苗,分别对鲤鱼进行注射和浸泡试验,测定白细胞吞噬活性、淋巴细胞转化能力、血清、皮肤黏液的抗体效价及活菌攻毒后的免疫保护率.结果表明,受免鱼各项指标均明显高于对照组,注射组免疫保护率达90%,其中浸泡组各项略低于注射组.嗜水气单胞菌灭活疫苗对鲤鱼有显著免疫保护效应,可作为预防细菌性败血症感染的疫苗.  相似文献
