A technique for transcervical cannulation of the uterus of the bitch for the collection of samples for cytology and microbiology was developed. This technique involved passing a cannula into the uterus with the aid of an endoscope. The uterus was cannulated successfully in mature bitches (over 10 kg) 110 times in 144 attempts. Success of cannulation was influenced by the experience of the operator and ease of cannulation by the stage of the reproductive cycle, the size of the bitch and the type of cannula. Complications associated with the procedure were endometritis, vaginitis and vaginal tears. Uterine cannulation was generally possible in all bitches studied at all stages of the reproductive cycle. Techniques for the collection of samples from the uterus for cytology and microbiology and for the processing of samples for uterine cytology are outlined. Hysteroscopy was performed on seven occasions in post partum bitches and was found to be possible at least until day 17 after whelping.  相似文献

An oocyte recovery procedure was developed and evaluated to determine whether a transcervical embryo recovery procedure is feasible with our method, which includes estradiol-17beta (E2) and oxytocin (OT) treatments, for dilating the cervix in ewes. On d 6 of an estrous cycle, oocytes were recovered either transcervically or with a laparotomy procedure. In the laparotomy group, ovulation rate was determined during the procedure and was used to calculate the percentage ofoocytes recovered. The laparotomy procedure was a standard uterine flush, and 12 mL of PBS was used to flush each uterine horn. In the transcervical group, the ovaries in each ewe were evaluated ultrasonically to determine ovulation rate. For transcervical recovery, 100 microg of E2 were injected i.v. on d 5 to increase cervical OT receptors, and 100 USP units of OT were injected i.v. 10 to 12 h later to dilate the cervix. Approximately 25 min after OT, ewes were placed in dorsal recumbency in a Commodore cradle, and a modified Foley catheter was passed through the cervix and into the uterus for injection (80 to 210 mL) and aspiration of PBS. The PBS was aspirated with a vacuum pump. The percentage of PBS recovered was greater (P<.01) at laparotomy than with the transcervical procedure (85.8 vs. 36.2%). Despite that difference, oocyte recovery did not differ significantly between the two groups (67% for laparotomy vs. 50% for transcervical; [oocytes recovered/number of corpora lutea] x 100), and there was no evidence that the transcervical procedure damaged the oocytes; the zona pellucida remained intact around all of the oocytes. In conclusion, a procedure that includes E2-OT-induced cervical dilation, passage of a modified Foley catheter into the uterus, and incremental infusion and aspiration of media through the catheter can be used to recover oocytes transcervically from ewes. This procedure may make transcervical embryo recovery feasible for sheep.  相似文献

Samples for microbiological culture were collected from the uterus of bitches using transcervical uterine cannulation (31 samples, 23 bitches) and from the uterus, cervix and vagina post mortem (19 bitches) at all stages of the reproductive cycle. Samples were cultured for aerobic and anaerobic bacteria and for aerobic mycoplasmas. Bacteria were always found in the uterus during prooestrus and oestrus (12 positive in 12 cultures) and rarely at other stages of the reproductive cycle: during anoestrus (one in 14) and other stages (none in 24). When microorganisms were detected at three sites post mortem, those found in the cervix and vagina were always of the same species as those found in the uterus. In six out of 13 instances, microorganisms were found in the cervix or vagina when none were found in the uterus. The mean number of isolates, number of bacteria seen in uterine cytology and bacterial growth were greater (P < 0–005) during oestrus and pro-oestrus than at other stages. Bacteria isolated from the uterus, in order of frequency, were Escherichia coli, Haemophilus species, α-haemo-lytic streptococci, Corynebacterium species, Streptococcus canis, Alcaligenes faecalis, Bac-teroides species, Pasteurella species and Proteus mirabilis. No mycoplasmas were cultured from the samples. This study indicates that the uterus of the normal bitch has a uterine microflora during pro-oestrus and oestrus that is similar to that of the vagina and cervix.  相似文献

The aim of this study was to evaluate the efficacy of Cervidil®, a prostaglandin E2 (PgE2)-releasing vaginal insert used for induction of cervical ripening and labour in women, to enhance the ease of transcervical artificial insemination (TCAI) in anestrous ewes (June). It was hypothesized that the use of Cervidil® prior to AI would cause dilation of the cervix, and thus alleviate the difficulty associated with traversing the cervix for semen deposition in sheep. Cervidil® was inserted 12 h before insemination in six Rideau-Arcott ewes; six ewes served as untreated controls.Semen was deposited into the uterus of all six treated ewes but TCAI was possible only in four of the six control ewes. It can be concluded that Cervidil® facilitated transcervical semen deposition in anestrous ewes. The treatment with Cervidil® has the promise of a technique to improve transcervical AI and to enable non-invasive embryo transfer procedures in sheep.  相似文献

In a previous study we observed that it is possible to reach the cervix in all queens with a 1 mm diameter probe only. So, we developed both a new technique and a catheter (1 mm diameter) to allow transcervical insemination [Zambelli and Castagnetti 2001]. The aims of this study were to investigate vaginal and cervical anatomic modifications during the various stages of the oestrus cycle and to test the previously described technique of transcervical catheterization during the various stages of the oestrus cycle. In experiment 1, silicon impression moulds were obtained from the reproductive tracts of 21 queens' cadavers and vaginal and cervical measures were taken. The results showed that there are some significant anatomic modifications during the various stages of the oestrus cycle in vaginal and cervical anatomy, principally related to the dorsal medial fold increase induced by the follicular phase. In experiment 2, transcervical catheterization was attempted in 95 queens at various stages of oestrus cycle both during reproductive and non-reproductive season. After catheterization, methylene blue solution was injected through the cervical catheter. Successful catheterization was assessed during surgery, when colour was observed in the uterine horns. It was possible to perform transcervical catheterization during non-reproductive season in 16 of 20 anoestrus queens and in 12 of 15 induced oestrus queens; during reproductive season in nine of 21 interoestrus queens, in eight of 13 dioestrus/pregnancy queens, in four of 18 oestrus queens and in seven of eight queens in first oestrus during lactation.  相似文献

The purpose of the current study was to investigate the suitability of an isobaric laparoscopic procedure, using a single port, for obtaining serial kidney and liver biopsy samples from standing steers. The samples were used in support of a pharmacokinetic tissue–fluid correlation study. Laparoscopic access was performed 3 times in each of 8 healthy Holstein steers, alternating from the right side to the left side and then to the right side again. The surgery was performed in standing stocks after the animals were given 3 doses of sulfadimethoxine sulfate intravenously and fasted for at least 18 h. Sedation and analgesia were achieved with acepromazine and xylazine. Lidocaine 2% was injected at the center of the paralumbar fossa (left or right), and an incision was made for introduction of a trocar–cannula assembly. Room air was allowed to enter the abdomen through the cannula at the time of insertion. Once the peritoneal cavity was reached, an operating endoscope was inserted. No pressurized insufflation was performed. A biopsy forceps was introduced into the operating channel of the endoscope to obtain a 100-mg kidney or liver sample. No complications were encountered. The 24 laparoscopic procedures provided 24 kidney and 16 liver samples. The results suggest that the isobaric (gasless) single-port laparoscopic technique is feasible for kidney and liver biopsy on standing steers. The procedure can be performed in a reliable and efficient manner in the sedated standing bovine.  相似文献

Eleven healthy horses underwent 5 repeated abdominocenteses, with either a sharp‐tipped spinal needle or a blunt‐tipped teat cannula to investigate possible differences in success rate, sample volume, depth at which a sample was obtained, length of procedure, complications and cytological variables. Variables were analysed with a repeated‐measures ANOVA or Fisher's exact test (α = 0.05). Success rate, sample volume, length of procedure, occurrence of haemorrhage during the procedure and incidence of grossly visible blood contamination were not different between techniques or over time. Depth at which samples were obtained was greater using a cannula than a needle (P<0.02), and samples were obtained with either technique at a greater depth than abdominal wall thickness assessed via ultrasound (P<0.014). Peritoneal fluid total and differential nucleated cell counts, and total protein concentration did not differ between techniques or over time. Red blood cell count in the least blood contaminated fraction of each sample was not affected by time, but it was lower after needle abdominocentesis than after cannula abdominocentesis (P = 0.04). Swelling of abdominocentesis sites increased with both techniques over time (P<0.05) and was more severe in horses undergoing cannula abdominocentesis (P<0.05). Enterocentesis occurred with a spinal needle in one horse, but no subsequent complications were noted. Both techniques appear to be safe and reliable for abdominocentesis in healthy horses. Using a blunt‐tipped cannula, as opposed to a needle, is likely to result in greater subcutaneous swelling. Both the cannula and needle must be long enough to penetrate well beyond the thickness of the abdominal wall to achieve successful peritoneal fluid collection.  相似文献

Two experimental procedures were investigated in order to assess their usefulness in diagnosing occlusion of the equine uterine tube. The starch grain test, which involves the injection of a starch suspension onto the ovary and the subsequent recovery of starch from the cervix, proved to be safe and reliable. Each tube could be investigated separately. It was found that when starch was injected onto the ovulating ovary on the day of ovulation, there was a delay of four to seven days before starch was recovered from the cervix. The phenolsulphonphthalein (PSP) test, which involves the deposition of dye solution into the uterus and its subsequent identification in urine after peritoneal absorption, was not reliable. Contamination of the urine occurred per vaginum, and the test was not unilaterally specific.  相似文献

Congenital abnormalities of the uterus and the cervix are rare in horses, often associated with infertility, despite normal ovarian activity and sexual behaviour. An abnormality of the cervix in a 5‐year‐old jennet with a history of infertility was described. Clinical and ultrasound examination of the genital tract showed a normal development of the uterus with regular cyclic activity in both the ovaries. Vaginoscopy revealed a normal development of the vagina and a cervix deviated on the left side of the vagina by a thick dorsoventral fold. The lateral deviation was permanent in any stage of oestrus cycle. Furthermore, the cervical os was narrow and digital penetration was not realizable. Using a 5‐mm catheter, the full penetration was not possible even during oestrus; moreover, the inoculation of fluid could be revealed in uterus by ultrasound (fluid was not present before inoculation), demonstrating a minimal patency. An abnormal and permanent lateral displacement of a stenotic cervix was suspected as the primary cause of infertility in the jennet.  相似文献

The aim of this study was to investigate the feasibility of simultaneous application of three cannulations and to test the effectiveness of a modified T-type cannula compared to simple T-type cannula. A ruminal, a simple T-type and a modified T-type duodenal and an ilial cannula were used in sheep. No complication was encountered in ruminal cannulation. Although no grafts were used, leakage of digesta was not seen for at least five months in T-type cannula and seven months in modified T-type cannula after surgery. Fibrous tissue grew between the serosa of the intestine and the peritoneum. No erosion of the tissue between intestine and body wall was observed after slaughter. Especially ilial digesta was easily obtained from modified T-type cannula.  相似文献

An effective long‐term treatment is necessary for mares with pyometra, because the condition tends to recur. In many affected animals, several conformational or anatomical anomalies contribute to impaired uterine clearance. Ovariohysterectomy is the surgical procedure of choice. Conservative therapy consists of draining and flushing the uterus, and systemic anti‐inflammatory and antimicrobial treatment. Uterine secretions tend to accumulate again after local treatment, especially in mares with poor vaginal conformation or cervical adhesions. Herein, we describe three cases in which a cervical stent was used in mares after mechanical or manual dilation of the cervix to achieve permanent draining of the uterus. The mares remained symptom‐free for up to 6 years and exhibited good clinical progress and good performance in competitions. Potential complications of the procedure include loss of the stent and obstruction caused by viscous secretion. A cervical stent is a relatively easy and low‐cost option for the long‐term treatment of pyometra in mares, particularly in cases where excessive costs of surgery and risks of a general anaesthesia are to be avoided.  相似文献

Cecal cannulation is necessary for sampling of intestinal contents for a variety of nutritional or digestive physiology studies. This report describes a 2-stage technique for permanent cecal cannulation in standing horses. For the first procedure, a right flank laparotomy is performed and a small pouch of the cecal base exteriorized and sutured to the body wall. The second procedure is performed approximately 1 wk later. During the second procedure, the exposed cecal pouch is removed and the cannula inserted. Ten horses were cannulated using this technique. After the first procedure, 1 horse developed a cecal impaction unresponsive to medical therapy and ruptured its cecum, whereas 2 other horses developed mild transient colic that responded to medical management. Insertion of the cecal cannula after creation of the stoma in the second procedure resulted in transient colic in 4 of 9 horses, but they responded to analgesic therapy in less than 24 h in all instances. The time to complete healing of the cannula site was approximately 30 d. The technique described in this report decreases the risk of peritonitis due to intestinal leakage and is technically easier to perform than previously described techniques.  相似文献

The new generation of videoendoscopes uses an electronic, instead of an optical, system for image transmission. Advantages over conventional fibre-optic endoscopes include increased image quality, handling robustness and direct display of the image on a TV monitor for multiple simultaneous viewing. In the present study, hysteroscopy was performed on 14 normal fertile Welsh Pony and Thoroughbred mares at various times during the annual and ovarian breeding cycles. Oestrus was characterised by an oedematous, relaxed cervix lying on the floor of the vagina, diffuse oedema of the endometrium and the occasional accumulation of small amounts of clear secretions in the uterine lumen. In dioestrus, the cervix appeared tight and pale and was located in the centre of the vaginal fornix. The endometrium appeared thinner due to the reduction in interstitial fluid, which allowed the underlying blood vessels to become visible, but it still had a glistening surface as a result of endometrial gland secretions. The appearance of the uterotubal papillae reflected the changes in the cervix, being relaxed, pinkish and oedematous during oestrus but remaining pale, tight and erect in dioestrus (between Days 7 and 12 after ovulation). A flexible polythene cannula was passed into the uterine lumen through the working channel of the endoscope for directed collection of uterine fluid for cytological and bacteriological examinations. Site-directed biopsies of the endometrium were taken, with flexible forceps inserted through the working channel of the endoscope, or with rigid forceps passed through the cervix alongside the endoscope. Both methods enabled accurate visual selection of the sampling area but the rigid forceps were preferred due to the larger size of the piece of endometrium recovered.  相似文献

Cervical anatomy in ewes usually prevents nonsurgical, intrauterine AI and transcervical embryo transfer (ET), which limits their commercial use in sheep. This study was conducted to determine whether oxytocin would dilate the cervix in ewes and permit passage of a stainless steel rod into the uterus. In Exp. 1, at 44 and 52 h after removal of progestogenated pessaries, ewes were injected i.v. with 0 (saline), 200, 400, or 600 USP units of oxytocin. Immediately before and after treatments, stainless steel rods were used to evaluate cervical dilation and determine whether the uterus could be entered. A rod could not be passed through the cervix and into the uterus in any of the saline-treated ewes. All doses of oxytocin given at 44 and 52 h after pessary removal dilated the cervix and permitted easy passage of a rod into the uterus. At both 44 and 52 h, a stainless steel rod was passed into the uterus in 33 of 43 (77%) of the oxytocin-treated ewes. In 93% (40/43) of these ewes, a rod could be passed into the uterus during either the 44-h or during the 52-h attempt. In Exp. 2, on d 9 after pessary removal, ewes were injected i.v. with oxytocin (400 USP units) at 6 or 12 h after i.v. estradiol-17 beta (0, 100, or 200 micrograms). Cervical dilation was evaluated as in Exp. 1. OBJECTIVE: To determine safety of lung biopsy in healthy cattle and compare 3 biopsy instruments for suitability of use. DESIGN: Prospective study. ANIMALS: 60 healthy cows. PROCEDURE: Bilateral lung biopsy through the ninth intercostal space was attempted in all cows. The procedure was performed with cows standing and restrained by an assistant. To study the short-term effects of lung biopsy, 50 cows were slaughtered 1 day after the procedure and examined. To study the long-term effects of lung biopsy, the remaining 10 cows were examined daily for 10 days and then slaughtered. Three biopsy instruments--the Autovac biopsy cannula, the Tru-Cut needle, and the Bard Biopty-System--were evaluated. RESULTS: 2 cows collapsed momentarily during the procedure but recovered quickly. The remaining cows did not develop adverse effects, and general attitude and condition were not affected. Postmortem examination of the 50 cows slaughtered 1 day after the procedure revealed lesions < or = 5.0 mm long and with a surface area < or = 4.0 mm2 at 108 of 154 (71%) biopsy sites involving the pulmonary pleura. Biopsy specimens were obtained during 48 of 50 (96%) procedures in which the Tru-Cut needle was used, during 38 of 39 (97%) procedures in which the Bard Biopty-System was used, and during only 1 of 11 biopsy procedures in which the Autovac cannula was used. At postmortem examination of the 10 cows slaughtered 10 days after the procedure, lesions were seen as small scars; adhesions were not observed. CONCLUSIONS AND CLINICAL RELEVANCE: Percutaneous lung biopsy is a safe procedure in healthy cows.  相似文献

Objectives : To describe a minimally invasive technique for treating urethral obstructions in male dogs and to review the postoperative results. Methods : All dogs (n=9) had urethral obstruction due to calculi. Obstructions were verified by radiographic and ultrasonographic examinations. Dogs with impaired kidney function were not included in the study. A 5‐mm diameter trocar and cannula were placed in the ventral midline, 2 cm cranial to the umbilicus, allowing placement of a 10‐mm diameter cannula under visual guidance, adjacent to the apex of the bladder. The bladder was then partially exteriorised and sutured to the skin. A 5‐mm diameter cystoscopy sheath was introduced into the bladder lumen and advanced into the urethra. Continuous retrograde flushing was used to dislodge the calculi from the site of obstruction and collect them upstream. Results : The nine dogs were followed up for a minimum of 6 months. No major postoperative complications were identified. One dog exhibited transient macroscopic haematuria (for 3 weeks postoperatively). All urethral calculi were removed in the nine dogs. No recurrence was found during the follow‐up period. Clinical Significance : A minimally invasive approach is used to treat urethral obstructions resulting from calculi in the male dogs.  相似文献

The aim of this study was to describe the technique of rhinolaryngotracheobronchoscopy and oesophagoscopy in small ruminants and to report the normal endoscopic appearance of the structures of the upper respiratory tract and oesophagus. Thirty sheep and 30 goats, all clinically normal, underwent rhinolaryngotracheobronchoscopy and oesophagoscopy using a flexible endoscope (length 100 cm; diameter 4 mm). The anatomical features of the structures and the appearance of the mucosa were investigated. The degree of mucosal pigmentation varied between breeds. The appearance of the pharyngeal tonsil and the shape of the trachea in cross-section showed most individual variation. The technique was easily performed and sedation was necessary in only a few cases. No major complications were encountered. It was concluded that the procedure facilitated more accurate diagnosis and prognosis of conditions of the upper respiratory tract and oesophagus.  相似文献

Collection of ileal digesta to evaluate AA digestibilities has become increasingly important in swine nutrition research. Steered ileocecal valve cannulation of pigs permits total collection of ileal digesta, while still allowing normal digesta flow during noncollection periods. This technique was modified and used with 64 crossbred barrows in five trials. Our procedural changes included preoperative i.v. administration of a broad-spectrum antibiotic and nonsteroidal antiinflammatory drug, sharp incision through the muscle layers of the laparotomy wound, use of a heparinized saline lavage solution, replacement of the guide ring with a stylette, and fixing the outer cannula barrel in place with a hose clamp. The current technique involves a right flank laparotomy, parallel and distal to the last rib, with the pig under general anesthesia. A stainless-steel ring (inner ring = 2.0 mm thick, 35.0 mm i.d.) is introduced into the ileal lumen through an enterotomy proximal to the origin of the ileocecal fold. A nylon string attached to this ring is threaded through the ileum and ileocecal valve into the cecum using a silastic stylette, which encases the string. A second stainless-steel ring (outer ring = 2.0 mm thick, 34 mm o.d.) is fixed in place around the ileum, distal to the inner ring and just proximal to the ileocecal valve. A polyurethane cannula barrel (barrel = 100 mm long, 26 mm i.d., 32 mm o.d.; flange = 70 mm o.d.) is introduced into the cecal lumen via an enterotomy through the lateral cecal band and secured in place with two purse-string sutures. The cannula is exteriorized through an incision caudal and proximal to the intial laparotomy site, where it is plugged using a cylindrical stopper (26 mm o.d., 55 mm long) and held in place by a second cannula barrel (barrel = 43 mm length, 33 mm i.d., 41 mm o.d.; flange = 80 mm o.d.). Procedural changes decreased postsurgical complications, as evidenced by decreased seepage around the cannula and fewer and less severe adhesions noted at necropsy. Based on five trials, this technique is a reliable means of collecting ileal digesta for nutrient analyses.  相似文献
