The development of pyometra in cows depends largely on the state of local immunity of the uterus. The objective of the study was to evaluate the concentration of the following proinflammatory cytokines: tumour necrosis factor (TNF‐α) and interleukin‐6 (IL‐6); anti‐inflammatory cytokine: interleukin‐10 (IL‐10); and acute‐phase proteins (APPs): haptoglobin (Hp) and serum amyloid A (SAA), in serum and uterine washings in cows with pyometra and healthy animals. The study was performed on 20 cows divided into two groups based on the results of cytological and ultrasonographic tests: a pyometra and a healthy group (10 cows per group). Experimental material consisted of blood serum and uterine washings. The levels of the following cytokines, TNF‐α, IL‐6, IL‐10 and APPs – Hp and SAA, in the study material were determined by ELISA. The results showed that the values of TNF‐α, IL‐6, IL‐10 as well as SAA and Hp were significantly higher in serum of cows with pyometra compared to controls (p < 0.001). The uterine washings had significantly higher levels of IL‐6, IL‐10, and Hp in pyometra cows compared to the control (p < 0.001). Our results indicate that it is possible to monitor the course of pyometra in cows based on the evaluation of the concentration of cytokines and Hp in the serum and uterine washings. Simultaneous evaluation of selected indicators of antagonistic interaction can be helpful in determining the current status of local immunity of the uterus. On this basis, it could be possible to properly select an adjunctive therapy in the form of immunomodulating preparations.  相似文献

Nisin, a lantibiotic bacteriocin, has been used for years as a natural food preservative. In addition to its antimicrobial activity, nisin also shows immunomodulatory properties, and the nisin‐producing Lactococcus lactis strain has been successfully tested as a probiotic in weaned piglets. However, the impact of nisin on porcine immune cells has not yet been explored. The objective of the present study was to examine the in vitro immunomodulatory effect of nisin on porcine peripheral blood leucocytes. The whole heparinized blood samples or freshly isolated peripheral blood mononuclear cells (PBMCs) were incubated with different nisin concentrations (0, 1.56, 3.125, 6.25, 12.5, 25 or 50 µg/ml) for 1, 24, 48 or 72 hr. Escherichia coli bacteria were used to stimulate blood phagocytes, while concanavalin A and lipopolysaccharide from E. coli were used as mitogens. Control cells remained unstimulated. MTT colorimetric assay was used to evaluate PBMCs viability and mitogenic response. Phagocyte activity and T‐cell proliferation were measured by flow cytometry. Flow cytometer was also used for immunophenotyping of T cells. Cytokine levels in the culture media were determined using commercial immunoassay (ELISA) kits. The highest concentration of nisin exhibited proliferative activity (p ? 0.05), stimulated interleukin‐1 beta (IL‐1β) and interleukin‐6 (IL‐6) production (both at p ? 0.001), and increased the percentage of CD4+CD8+ T cells (p ? 0.001) among unstimulated leucocytes. After cell stimulation, however, the highest nisin concentration showed antiproliferative activity (p ? 0.05), decreased phagocytic functions (p ? 0.05) and inhibited the synthesis of IL‐6 (time‐ and concentration‐dependent effect). As a typical bacterial product, nisin had a stronger impact on innate immune cells, and its effect on T cells was likely a consequence of the modulation of the activity of antigen‐presenting cells. Nisin may be a good candidate as an immunomodulator in pig breeding.  相似文献

The study evaluated dietary supplementation with live yeast (LY) Saccharomyces cerevisiae (CNCM I‐4407, 1010 CFU/g, Actisaf; Phileo Lesaffre Animal Care, France) on rumen fermentation and serum metabolic profile in lactating dairy cows. Fifty Holstein cows received a total mixed ration with (Live Yeast Diet, LYD, n = 25) or without (Control Diet, CD, n = 25) 5 × 1010 CFU/cow/day of LY from 3 to 19 weeks of lactation. Rumen fermentation and serum metabolic profile were measured in eight cows per treatment at 3, 7, 11, 15, 19 weeks post‐partum. LYD showed an increased daily milk yield (+4%) over CD (p < 0.05). Mean rumen pH at 4 hr after morning meal was higher in LYD (6.59) than CD (6.32) (p < 0.01). Total volatile fatty acids (VFA) and acetate molar proportion were higher in LYD (114.24 mM; 25.04%) than CD (106.47 mM; 24.73%) (p < 0.05). Propionate and butyrate molar proportions, acetate to propionate ratio, ammonia levels did not differ between LYD and CD. Ruminal lactate was lower in LYD than CD (9.3 vs. 16.4 mM) (p < 0.001), with a 53% decrease in LYD. During peak lactation, LYD had lower serum NEFA (p < 0.05, 0.40 vs. 0.48 mM) and BHBA (p < 0.01, 0.47 vs. 0.58 mM) than CD, lower liver enzyme activities (AST 1.39 vs. 1.54 ukat/L) (p < 0.05). Serum glucose was higher in LYD at peak lactation (3.22 vs. 3.12 mM, and 3.32 vs. 3.16 mM respectively) (p < 0.05). The results confirmed a reducing effect of LY on lactate accumulation in rumen fluid, associated with an increase in rumen pH. Lower serum levels of lipomobilization markers, liver enzyme activities and higher glucose levels may suggest that live yeast slightly mitigated negative energy balance and had a certain liver protective effect.  相似文献

The study aimed to evaluate pregnancy per artificial insemination (P/AI) of cows subjected to synchronization and resynchronization in ovulation protocols using intravaginal progesterone‐releasing insert (P4) before pregnancy diagnosis (PD) and the relationship of PR with the diameter of preovulatory follicles (ØPOF) before TAI. Cows (n = 378) were distributed into two groups: a resynchronization group with new devices (GRN; n = 185) and resynchronization group with used devices (GRU; n = 193). On Day 0, both groups received a new P4 and estradiol benzoate (EB). On D8, P4 removal + D‐cloprostenol + eCG + estradiol cypionate (EC) was done. On d10, TAI was conducted. On d32, cows were resynchronized and divided into two groups, GRN (n = 185) and GRU (n = 193). The GRN group received a new P4 + EB, and the GRU group received a used P4 + EB. On d40, the P4 was removed + PD. The non‐pregnant cows received D‐cloprostenol + eCG + EC. US was done again on d42 to determine ØPOF before the second TAI. The P/AI of the GRN and GRU groups after synchronization were 56.2% and 57.0% (p = 0.87), respectively, and those after resynchronization were 58.0% and 37.3% (p < 0.008), respectively. The P/AI of the GRN and GRU groups observed after TAI (synchronization + resynchronization) were 81.6% and 73.1%, respectively (p = 0.047). No difference (p = 0.067) in ØPOF between the pregnant and non‐pregnant cows in the GRN was found, whereas the GRU group showed a significant difference (p = 0.003). Resynchronization protocols optimized the P/AI in both groups. New intravaginal devices resulted in greater P/AI and P/AI accumulation in resynchronization as compared with the GRU; the ØPOF was related with P/AI.  相似文献

Congenital chondrodystrophy of unknown origin (CCUO) has been reported in beef cattle worldwide. A trace mineral deficiency in pregnant dams has been suggested as causing the deformities seen in CCUO calves. An extended outbreak of CCUO in Australia between 2002 and 2009 provided an opportunity to examine the pattern of trace mineral status in gestating cows and the effect of parenteral trace mineral supplementation on a herd at risk of CCUO calves. A property with a strong history of CCUO was identified from a previous case–control study. A randomly selected sample of multiparous (8 to 10 years old) Angus and Angus cross cows (= 20) was assigned to control (no injection) or supplement group given three injections at the recommended dose rate of chelated mineral supplement (Mn, Zn, Cu and Se) at 6‐week intervals in mid‐ to late gestation. Liver biopsies were performed to measure liver Mn, Zn, Cu and Fe at each injection time and 1 month post‐calving. Pasture samples were also collected for analysis. Supplemented cows had higher liver Cu levels compared to control cows (p < 0.001), but there was no difference between supplement and control cows for liver Zn, Fe or Mn. Liver Cu (p < 0.001), Fe (p < 0.001) and Zn (p < 0.001) fell during gestation and liver Mn increased after calving (p < 0.001). Three cows from the control group and two from the supplement group had liver zinc levels below the normal values at the end of the trial. Two cows from the control group and one from the treatment group gave birth to cows with signs of CCUO including superior brachygnathia. All cows except one from the control group had rough dry coats with excessive dander. The role of trace mineral status in the birth of CCUO calves, especially the role of zinc deficiency, requires further investigation.  相似文献

ObjectiveTo investigate if anaesthesia for canine cancer mastectomy further influences host cell-mediated immunity (CMI) promoting cancer progression.Study designA randomized, controlled, blinded clinical study.AnimalsA total of 20 bitches with malignant mammary tumours of clinical stage II or III undergoing the same type of mastectomy (regional mastectomy).MethodsDogs were randomly allocated to one of two anaesthetic groups (10 per group). The anaesthetic protocol of group A used minimally immunosuppressive drugs (tramadol, robenacoxib, propofol), whereas that of group B (control) used more immunosuppressive drugs (morphine, fentanyl, thiopental, isoflurane). For each animal, measurements of white blood cells (WBCs), neutrophils and lymphocytes, and flow cytometric assessment of T cells (CD3+), helper T cells (CD4+), cytotoxic T cells (CD8+) and CD5low+ T cells were performed prior to anaesthesia (day 0) and on days 3 and 10 postsurgery. Data were analysed using a General Linear Model for repeated measures and presented as mean ± standard deviation, p ≤ 0.05.ResultsIn all animals, on day 3, WBCs and neutrophils were significantly increased (p < 0.0005), while flow cytometry revealed significantly decreased relative percentages of T cells (CD3+) (p = 0.003) and their subpopulations CD4+ (p = 0.006), CD8+ (p = 0.029) and CD5low+ (p = 0.031). Specifically, on day 3, the cytotoxic T cells (CD8+) were significantly decreased (p = 0.05) only in group B, whereas the CD4+ (p = 0.006) and CD5low+ (p = 0.008) T cells in group A. The only significant difference between groups was found preoperatively in the CD4+/CD8+ ratio, which was higher in group A (p = 0.006).Conclusions and clinical relevanceIn dogs with mammary cancer undergoing regional mastectomy, a significant decrease in components of CMI was observed on day 3 postsurgery in both anaesthetic groups. Some indication, however, for better preserved cellular immunity by less immunosuppressive anaesthetic/analgesic drugs was detected, rendering their use advisable.  相似文献

The objectives were to evaluate the reproductive indices and survival analysis of pregnancy outcomes in multiparous repeat breeder Holstein cows (n = 557). The cows were synchronized to ovulate by Ovsynch, new controlled internal drug release device (N‐CIDRsynch), and once‐used CIDR device (U‐CIDRsynch). The pregnancy per AI at 28 days post‐insemination (P/AI 28) in the N‐CIDRsynch group (28.75%) was significantly (COR = 1.49; p = 0.011) greater than that reported in the Ovsynch (23.46%) and U‐CIDRsynch (21.73%) groups. Furthermore, the pregnancy per AI at day 75 post‐insemination (P/AI 75) in the N‐CIDRsynch group was significantly greater than the Ovysync group (COR = 1.35; p = 0.050). The repeat breeder cows received a N‐CIDR device had a significantly higher progesterone level on day 2 and day 4 of CIDR insertion (1.38 and 1.67 ng/ml, respectively) than those received a U‐CIDR device or the control group (p = 0.012 and 0.001, respectively). The Cox regression model recorded significant associations for synchronization protocols, THI at the TAI and season of calving with the hazard of P/AI 28 and P/AI 75 (p = 0.044 and 0.046; 0.001 and 0.005; 0.003 and 0.001, respectively). Multiparous repeat breeder cows (>3) had a lower hazard ratio (HR) of P/AI 28 than that reported in the reference (2nd parity) group (HR = 0.74, p = 0.050). The repeat breeder cows inseminated at 76–80 and >80 temperature‐humidity index (THI) had significantly lower HR of P/AI 28 than those inseminated at the baseline (<70) THI value (HR = 0.73 and 0.30, p = 0.036 and 0.001, respectively). The current results indicate that the use of N‐CIDR synch protocol may achieve satisfactory pregnancy outcomes in repeat breeder cows.  相似文献

The varied bioavailability and different effects of organic forms of copper on the immune system of poultry have prompted the search for new feed additives based on copper compounds containing modified chelate complexes. The aim of the study was to determine the effect of inorganic and organic forms of copper on selected parameters of the cellular and humoral immune response in broiler chickens by determining the percentages of CD3+CD4+, CD3+CD8+ and CD25+ lymphocytes, cells with MHC Class II expression, and BU‐1+ cells, as well as the concentrations of SOD, IL‐2, IL‐10 and TNF‐α in the peripheral blood. The experiments were conducted using 500 one‐day‐old Ross 308 roosters divided into five groups. Cu was added in inorganic form (CuSO4), in inorganic form with the addition of phytase (CuSO4 + F), in organic form in combination with glycine (Cu‐Gly) and in organic form in combination with glycine and a phytase supplement (Cu‐Gly+F). The results of the study indicate an increase in the percentage of CD3+CD4+ and CD3+CD8+ T lymphocytes, CD25+ T cells, and cells expressing MHC class II molecules, and in the concentration of ceruloplasmin, activity of superoxide dismutase and the concentration of IL‐2 in the groups that received copper, particularly copper‐glycine chelates. Based on the study, we can conclude that supplementation of poultry feed with copper chelates activates mainly the Th1 cellular immune response and the response of peripheral blood T lymphocytes. Furthermore, it promotes secretion of cytokines, which are involved in potentiation and regulation of the immune response in birds.  相似文献

The aim of the present study was to compare the lipid profile in oocytes of indicus and 1/2 indicus × taurus cows with high and low antral follicle count (AFC)/oocyte yields. After an OPU procedure (D0), antral follicles ≥3 mm were counted by ultrasonography (D4, 19, 34, 49, 64), and cows were assigned to groups with either high AFC (≥30 follicles; indicus, NH group; 1/2 indicus × taurus, AH group) or low AFC (≤15 antral follicles; indicus, NL group; 1/2 indicus × taurus, AL group). The lipid profiles of the oocytes were determined by MALDI‐MS. For GI, GII and GIII oocytes, the indicus samples tend to cluster separately from the 1/2 indicus × taurus samples. The lipid species [PC (P‐38:5) + H]+ and/or [PC (P‐36:2) + Na]+, [PC (38:2) + H]+, [PC (38:5) + Na]+ and [TAG (60:8) + NH4]+ were more abundant in indicus (NH and NL groups) than 1/2 indicus × taurus. The higher lipid content in the indicus oocytes likely reflects differences in the rate of lipid metabolism and may contribute to oocyte competence and embryo development.  相似文献

Vitamin E (vit E), an essential antioxidant for maintaining the stability of biological membranes and the function of the immune system, is considered to support adaptive immune responses and performance in cattle. The principal virulence factor of Shiga toxin (Stx)‐producing Escherichia coli (STEC), the eponymous Stx, modulates cellular immune responses in cattle, the primary STEC reservoir. Active and passive immunization of calves with Shiga toxoids (rStxMUT) was recently shown to reduce the STEC shedding. Here, we examined the influence of vit E on calves' serum α‐tocopherol, performance, haematology, blood chemistry and its interaction with rStxMUT immunization. Data from calves having received passive (colostrum from immunized cows) and active (intramuscularly at 5th and 8th weeks of life) vaccination with rStxMUT (n = 24) were compared to unvaccinated controls (n = 24; fed with low anti‐Stx colostrum, placebo injected). For each vaccination group, data were analysed according to the level of vit E supplementation offered by milk replacer (188 IU all‐rac‐α‐tocopheryl acetate daily [VitEM] vs. 354 IU [VitEH]). An increase by 79% in daily vit E supplementation led to slightly higher serum α‐tocopherol level and earlier concentrate intake at the beginning of the experiment without significant differences in live weight gain, haematology, blood chemistry parameters and peripheral CD4+ and CD8+ T‐cell subpopulations. rStxMUT vaccination modulated the CD4+/CD8+ ratio irrespective of vit E supplementation but decreased concentrate intake in VitEH in a time‐dependent manner. Results of our study indicate that an increase in daily vit E supplementation vastly fails to exert effects on laboratory parameters and growth performance. However, observed interactive effects of vit E supply and vaccination on the regulation of feed intake deserves further attention.  相似文献

To determine the effects of puerperal metritis on the immune response, changes in the differential peripheral blood leukocyte counts were analyzed during the peripartum period in cows with or without metritis. Multiparous Holstein cows were examined for uterine health disorders and classified into two groups: healthy (n = 11) or metritis (n = 5) cows. The lymphocyte and monocyte counts and the proportion of CD8+ lymphocytes were higher in cows with metritis compared to healthy cows. Moreover, the effects of puerperal metritis on the lymphocyte counts and CD4+/CD8+ ratio persisted weeks after the uterine inflammation had self‐resolved. Taken together, the findings of the present study indicate the possible long‐term alterations of systemic immune responses in cows with puerperal uterine inflammation. © 2015 Japanese Society of Animal Science  相似文献

In view of the lack of data on the effect of meloxicam (non-steroidal anti-inflammatory drug) on bovine γδ T cells (WC1+ cells) and very poorly recognized effects of dexamethasone (steroidal anti-inflammatory drug) on these cells, the purpose of the present study has been to determine the in vitro influence of these drugs on CD25highWC1+, CD25lowWC1+ and CD25?WC1+ lymphocytes of the peripheral blood of cattle. Peripheral blood mononuclear cells were treated with the drugs in concentrations reflecting their plasma levels achieved in vivo at therapeutic doses (dexamethasone 10?7 M; meloxicam 5 × 10?6 M) and at ten-fold lower concentrations. It was found out that percentages and absolute counts of CD25highWC1+ and CD25lowWC1+ cells increased in the presence of dexamethasone, and this effect was at least partly attributable to lower mortality of these cells, whose apoptosis was depressed by exposure to dexamethasone. It seems certain that this effect was not a result of increased multiplication of CD25highWC1+ and CD25lowWC1+ cells because their proliferation was reduced in the presence of dexamethasone. Exposure to this drug caused a rapidly occurring and lasting depletion of CD25?WC1+, which was at least partly due to their higher apoptosis. The results seem to suggest that impaired proliferation of these cells was responsible for a more profound expression of this disorder. Paradoxically, the percentage of cells producing IFN-γ, a proinflammatory cytokine, increased in the presence of dexamethasone, whereas the count of cells secreting the key anti-inflammatory and immunosuppressive cytokine, i.e. IL-10, declined. This effect was observed in all analyzed subpopulations of cells. Meloxicam did not interfere so drastically as dexamethasone with the functioning of WC1+ lymphocytes because it did not affect their apoptosis, proliferation, percentage or absolute count. With respect to the effect of meloxicam on counts of particular WC1+ lymphocyte subpopulations, it was only demonstrated that exposure to the drug was correlated with a transient and very weakly expressed decrease in the relative and absolute counts of CD25highWC1+ and CD25lowWC1+ cells, which was most probably a result of a temporary down-regulation of the expression of the CD25 molecule. In the presence of meloxicam, percentages of IFN-γ+CD25?WC1+ cells as well as cells producing IL-10 declined, an effect observed in all analyzed cell populations. These results suggest that care should be taken when administering this medication to animals with bacterial or viral infections, and we should avoid giving it to patients suffering from allergic or autoimmune disorders.  相似文献

Cholera toxin (Ctx) is an important mucosal adjuvant with potential experimental applications in pigs. However, little is known about the direct effects of Ctx on porcine immune cells. Therefore, we analysed the influence of Ctx on mitogen-induced lymphocyte proliferation. Ctx inhibited peripheral blood mononuclear cell (PBMC) proliferation with an IC50 of 34±17 ng/mL. This inhibition was not due to increased cell death. Lymphoblast formation in cultures stimulated with concanavalin A and Ctx was decreased at 24 h, but had reached the levels of control cultures again at 72 and 120 h, indicating that suppression was transient. Analysis of T cell subsets revealed that Ctx treatment specifically reduced the percentage of CD4CD8+ and γδ T cells, whereas the proportion of CD4+CD8 increased. Furthermore, Ctx caused secretion of IL-10 by PBMC cultures, but depressed TNFα secretion.  相似文献

In dairy cows, retained fetal membranes (RFM) affect reproductive performance. The aim of this study was to examine the leukocyte counts and the gene expression of tumour necrosis factor α (TNFα), interleukin 1β (IL‐1β), IL‐8, and IL‐10 in polymorphonuclear leukocytes (PMNs) and peripheral blood mononuclear cells (PBMCs) in cows with (n = 5) or without (n = 5) RFM during the peripartum period. The lymphocyte counts in RFM cows were higher than those in control cows throughout the experiment (p < .05). The expression of IL‐8 in PMNs of control cows was higher (p < .05) compared with that of RFM cows postpartum. In cows with RFM, IL‐1β expression was higher (p < .05) in PMNs at 6 weeks postpartum whereas the expression of IL‐1β was lower (p < .05) in PBMCs at 4 weeks postpartum. The expression of IL‐10 in PBMCs of control cows was higher (p < .05) than that of RFM cows at 2 weeks prepartum and 4 weeks postpartum. Taken together, our data indicate that changes of gene expression of pro‐ and anti‐inflammatory cytokines in RFM cows might be associated with the delayed placental separation and development of uterine inflammation in RFM cows.  相似文献

Purulent vaginal discharge (PVD) is a prevalent uterine disease of dairy cows during the puerperium that affects the milk production and affects the profitability of farms. The objectives of this study were to evaluate the biochemical profile, the body condition score, the milk production of cows with PVD and the effects PVD on reproductive performance. A total of 338 Holstein dairy cows aged from 3 to 5 years, from three commercial dairy farms, from Brazil, were used. Blood samples were collected within 25 ± 3 days post‐partum from Holstein dairy cows without PVD (control cows, n = 242) and cows with PVD (n = 96), based on scoring of the vaginal discharge. The body condition score and milk production were recorded on the day of sampling. The biochemical profile encompassed albumin, urea, gamma‐glutamyl transferase, calcium, fibrinogen and cholesterol concentrations. The number of services per pregnancy was lower (p < 0.01), and the number of days until first insemination and the median time to pregnancy were higher in cows with PVD (p < 0.01) when compared with control cows. Milk production and body condition score were lower (p < 0.01) in cows with PVD than in control group. Cows with PVD had lower (p < 0.05) serum albumin, urea, calcium and cholesterol concentrations, and higher serum gamma‐glutamyl transferase activity and fibrinogen concentration than cows without PVD. Our results show that cows with PVD have changes in the biochemical profile and negative effects on production and reproduction performance.  相似文献

The aim of the present study was to evaluate the biochemical profile and reproductive performance in postpartum dairy cows with clinical metritis. The research was carried out using performed on 213 pluriparous lactating cows from two dairy herds in southeast Brazil. Dairy cows were assigned into a group of healthy cows (n = 147) and cows with metritis (n = 66). Blood samples to assess serum albumin, cholesterol, fibrinogen, calcium, gamma-glutammyltransferase (GGT), aspartate aminotransferase (AST) and β-hydroxybutyrate (BHBA) concentrations were performed on day 8 ± 2 postpartum. Metritic cows had lower (p < .05) serum albumin, cholesterol and calcium concentration, and higher (p < .05) GGT and AST activity, and BHBA and fibrinogen concentrations when compared to the healthy group. The calving-to-conception interval and services per pregnancy were higher in metritic cows (p < .01) than in healthy cows and the conception rate at first service was lower in metric group (p < .01). Variable importance in projection plots demonstrated that cholesterol was the main discriminator between metritic and healthy cows. Our results indicated alterations in the biochemical profile and impaired reproduction performance in metritic cows.  相似文献

The cellular immune response to an experimental infection by Haemophilus parasuis, the etiological agent of Glässer’s disease in pigs, was characterized studying changes in peripheral blood mononuclear cells (PBMC) in colostrum-deprived pigs. Five groups were studied, four of those were previously immunized with different formulations and the fifth was maintained as non-immunized control. All groups were challenged with 5 × 109 CFU of H. parasuis serotype 5. The non-commercial bacterin conferred a complete protection, while the OMP-vaccine and the exposure to a subletal dose of 105 CFU of H. parasuis protected only partially, and the recombinant Tbp B-vaccine induced no protection. PBMC were analyzed using monoclonal antibodies against porcine CD45+, CD3+, CD4+, CD8α+, CD25+, CD4+ naïve, αIgM+ and SWC3+ cells in single-colour fluorescence, and CD4+/CD8α+ and CD8α+/CD8β+ combinations in two-colour fluorescence. The different groups showed no significant changes in PBMC subsets following vaccination, and only minor changes were encountered after challenge, consisting mainly of significant increases (P < 0.05) in the relative proportions of monocytes and granulocytes (SWC3+) and B cells (αIgM+), as well as a significant reduction in CD3+ cells (P < 0.05). These changes were similar for the five groups compared, except for the significant increase of CD25+ cells, which was only observed for the bacterin-vaccinated group. These results suggest an increase of trafficking of inflammatory cells and the onset of the adaptive antibody response against H. parasuis infection; in addition, the blood cellular response developed by the different groups was not relevant to protection.  相似文献

Canine pyometra is a dioestrus period disease in which systemic inflammatory response syndrome (SIRS) is a common outcome due to the response of the body to the bacterial infection. The purpose of this study was i) to differentiate canine pyometra and cystic endometrial hyperplasia (CEH)/mucometra by measuring serum C‐reactive protein (CRP) and prostaglandin F2α metabolite (PGFM) concentrations in blood and ii) to compare serum concentrations of CRP and PGFM in bitches with a pathological uterus (pyometra or CEH/mucometra) to concentrations in bitches with a healthy uterus. Mean CRP concentrations were found significantly higher (p < 0.001) in dogs with pyometra compared to those with CEH/mucometra or healthy uterus. However, no statistical difference could be detected between the groups for mean PGFM concentrations. Mean white blood cell count (WBC), alkaline phosphatase (ALP) and total protein concentrations were found significantly higher (p < 0.001) in dogs with pyometra. Escherichia coli was the most frequently isolated microorganism from dogs with pyometra (64.3%). Edwardsiella spp. was detected in a single case of pyometra for the first time. In conclusion, our results demonstrate that serum CRP concentrations were increased in dogs with pyometra and thus we conclude that serum CRP concentration but not PGFM might be useful as a marker to differentiate a case of CEH/mucometra from pyometra in female dogs. To the authors' knowledge, this is the first report in which Edwardsiella spp. has been isolated in the canine uterus.  相似文献

This study analysed the effect of growth differentiation factor‐9 (GDF‐9) on the in vitro culture of isolated ovine secondary follicles. The follicles were cultured in α‐MEM supplemented with BSA, insulin, glutamine, hypoxanthine, transferrin, selenium, ascorbic acid and FSH (α‐MEM+—control medium) or α‐MEM+ supplemented with 1, 10, 50 or 100 ng/ml GDF‐9. Next, the oocytes were destined to in vitro maturation (IVM). After 12 days of culture, there were no differences regarding the percentage of normal follicles, antrum formation and follicle diameter between the treatments (p > 0.05). The rates of fully grown oocytes (≥110 µm) were higher (p < 0.05) in 100 ng/ml GDF‐9 than other treatments, except for 10 ng/ml of GDF‐9 (p > 0.05). Treatment containing 100 ng/ml GDF‐9 showed higher (p < 0.05) mitochondrial activity than the control group. Moreover, 100 ng/ml GDF‐9 showed more oocytes in MI than α‐MEM+, 1 or 50 ng/ml GDF‐9 (p < 0.05). In conclusion, 100 ng/ml GDF‐9 increased the growth, mitochondrial function and meiotic resumption of oocytes from in vitro grown sheep secondary follicles.  相似文献

Fourty-eight Churra Tensina single male lambs were used to evaluate the effect of different feeding systems on carcass composition and predict the carcass tissue composition from joint composition and carcass measurements. Four treatments were studied: GR, unweaned lambs continuously grazing with ewes; GR + S, the same as the previous group, but lambs had free access to concentrate; DRL-GRE, lambs remained indoors with free access to concentrate and ewes grazed for 8 h/day, thereafter remaining with lambs; and DRL, lambs and ewes were kept in confinement, they had free access to concentrate and dry unifeed respectively. In the DRL-GRE and DRL groups lambs were weaned when 45 days old. Lambs were slaughtered at 22–24 kg live weight. Commercial joints from the half left carcass were obtained, recorded and dissected to determine carcass tissue composition.The feeding system had an effect on the proportion of the carcass tissue composition. GR treatment showed higher muscle (M) (p < 0.05) and bone (B) (p < 0.001) percentages, and a lower total fat (F) percentage (p < 0.001) than the rest of treatments. Hence it revealed higher M/F (p < 0.01) and lower M/B (p < 0.05) and subcutaneous fat/intermuscular fat ratios (SF/IF; p < 0.001). Carcass composition predictions were more accurate for grams than for percentages. In predictions from joint composition, pelvic limb was the most accurate joint for grams (R2 = 0.84, R2 = 0.68, and R2 = 0.77 for muscle, bone, and fat, respectively) as well as for percentage (R2 = 0.65, R2 = 0.56, and R2 = 0.73 for muscle, bone, and fat, respectively). Predictions from carcass measurements were greater in muscle and fat grams (R2 = 0.87 and R2 = 0.86 for muscle and fat, respectively) than in percentage (R2 = 0.58 and R2 = 0.73 for muscle and fat, respectively). Equations to predict carcass tissue that include carcass measurements are a simple method to assess accurately saleable muscle yield from different lamb feeding systems without involving carcass damage.  相似文献
