The study aimed to compare the acid–base balance and steroid concentrations between follicular fluids (FF) of pre‐ovulatory follicles derived from a spontaneous oestrus (SO), synchronized or induced oestrus (IO) and follicular cysts (CYS) and between FF and blood in dairy cows. Forty‐two dairy cows were included in this study. The animals were allocated to three groups: SO (n = 23); IO (n = 11) using GnRH at day 0 and day 9 and PGF2α at day 7; and animals with CYS (n = 10). The follicular fluids (FF) were aspirated from the cyst/pre‐ovulatory follicles (? ≥ 15 mm) after SO and after second GnRH dose in IO by transvaginal ultrasound‐guided ovum pick‐up technique. Blood samples (BL) were collected in heparinized vacutainer tubes. The oxygen tension (pO2) in FF of IO was higher (p < 0.05) than in SO and CYS groups. There were negative correlations (p < 0.001, r = ?0.89) between FF and blood pO2. The carbon dioxide tension (pCO2) and lactate level in FF of CYS group were higher (p < 0.05) than in SO and IO groups. There were negative correlations (p < 0.01, r = ?0.73) between blood and FF pO2. Oestradiol‐17β concentration in pre‐ovulatory follicles and plasma of the SO group was higher (p < 0.001) than in IO and CYS groups. Progesterone concentration in pre‐ovulatory follicles and plasma of the SO and IO groups was lower (p < 0.01) than in CYS group. Plasma androstenedione concentration in SO and IO groups was higher (p < 0.05) than in CYS group. In conclusion, acid–base parameters, E2 and P4 levels in the follicular fluid of both IO and CYS groups were deviated greatly from the physiological level (disturbances of intrafollicular/intracystic environment), which may affect the quality of both the oocyte and the granulosa cells.  相似文献

The intensity of heat signs in buffaloes is generally low and the incidence of suboestrus varied from 15 to 73% (Buffalopedia). The objective of this study was to investigate the feasibility of monitoring the changes in some milk constituents, oestradiol levels and electrical conductivity of vaginal mucus during peri‐oestrous period in prediction of the timing of oestrus in buffaloes. Twenty‐one Egyptian buffaloes aged 3–9 year, 1st–6th lactations, were examined by oestrous detector and ultrasonographically for monitoring the ovarian and uterine activity for 7 days around the time of standing oestrus. Sodium, potassium, chloride and lactose were assayed in aqueous phase of milk; besides, oestradiol was estimated in serum. Current results declared highly significant acute changes in milk constituents at the time of oestrus characterized by peaking of chloride and sodium levels and lowering of potassium and lactose values. The alternation in milk composition when arranged in decreasing order of magnitude, sodium was the highest (77.78 ± 0.69%), followed by chloride (61.60 ± 1.52%) and potassium (?58.14 ± 10.89%). Concomitantly, milk lactose decreased by 26.07 ± 7.97% compared to baseline levels. Synchronously, vaginal electrical resistance (VER) showed a significant (p < 0.01) decrease, but serum oestradiol 17β levels surged (59.93 ± 7.29 pg/ml) on day of oestrus. Serum oestradiol level was negatively correlated with VER (r = ?0.577), potassium (r = ?0.661), positively correlated with chloride (r = 0.707) and sodium (r = 0.579) and not correlated with lactose levels. These results for the first time suggested that the changes in constituents of milk during peri‐oestrous period may be used as a practical non‐invasive indicator for oestrous detection and prediction of ovulation in Egyptian buffaloes.  相似文献

The effect of a modified eros centre on weaning to oestrus interval, follicle size, ovulation and farrowing rate and total born litter size was investigated. In modified eros centre 94.4% and in group housing 79.1% of the sows (p < 0.01) expressed oestrus within 10 days post‐weaning. Weaning to oestrus interval was shorter (p < 0.001) for sows kept in modified eros centre. The interval from onset of oestrus to the time of ovulation was longer for sows in group housing (p=0.05). The time of ovulation was negatively correlated (r=?0.50) with the interval from weaning to oestrus (p=0.005). The time of ovulation after onset of oestrus was significantly (p < 0.05) shorter for sows expressing oestrus within 2–4 days of weaning, compared with the animals that expressed oestrus between days 5 and 6 post‐weaning and was shortest for sows expressing oestrus after day 6 post‐weaning. Farrowing rate was not affected by a modified eros centre. Litter size tended to be smaller in group‐housed weaned sows (p=0.10). The timing of last artificial insemination relative to time of ovulation did not affect litter size (p > 0.10). The implication of these results is that a modified eros centre may improve some of the post‐weaning oestrous parameters of the sow.  相似文献

To meet weekly breeding targets, it is occasionally necessary to inject exogenous gonadotrophins to induce oestrus in prepubertal gilts. However, the gilt oestrus response to equine chorionic gonadotrophin (eCG) either alone or in combination with human chorionic gonadotrophin (hCG) can be unpredictable. The objective of the present study was to examine possible reasons for this unpredictability. Prepubertal gilts (90 kg and 153 days of age, n = 109) received an injection of either 600 IU eCG or a combination of 400 IU eCG and 200 IU hCG (PG600), or were non-injected controls, and were then exposed to a mature boar for 15 min daily for 7 days for oestrus detection. At the time of injection, real-time ultrasound revealed that the gilt ovaries had primarily 1–2 mm follicles. Blood samples were obtained at time of hormone injection (day 0) and at days 3, 7 and 10 for assay of serum progesterone concentrations. The oestrus responses by 7 days were15.5%, 73.3% and 0%, for eCG, PG600, and control gilts, respectively (p < 0.001). The oestrus response improved (p < 0.05) with increasing body weight. Based on circulating progesterone levels, all oestrous gilts ovulated except for four of the PG600 gilts. Failure to express oestrus in PG600 gilts was not associated with a premature rise in progesterone.  相似文献

Shortened and weakened oestrous signs in dairy cows may cause a failure of oestrus detection and artificial insemination timing error leading to poor reproductive performance. The aims of this study were to investigate the duration of standing oestrus in high-producing dairy cows under a free stall system, to determine the duration of expression of secondary oestrous signs before and after standing oestrus (Expt 1) and to compare the duration and intensity of oestrus between cows and heifers (Expt 2). Cattle were checked for primary and secondary oestrous signs at an interval of 4 h. Heat detection aids were also used. In Expt 1, of 56 cows which were detected in oestrus, 36 cows (64.3%) showed standing oestrus and other 20 cows (36.6%) showed secondary oestrous signs only. Duration of the standing oestrus was 6.6 +/- 6.3 h on average (+/-SD), ranging between 2 and 32 h. The cows in standing oestrus showed secondary oestrous signs during a period from 9.6 +/- 8.1 h before onset of standing to 18.4 +/- 18.8 h after the end of standing oestrus. In the cows that did not show standing oestrus, expression of secondary oestrous signs were observed for 25.7 +/- 20.5 h, which was 7.5 h shorter than the average duration of oestrus in cows showing standing oestrus. In Expt 2, nine (82%) of the 11 lactating cows in oestrus showed standing, while all the 10 heifers exhibited standing oestrus. Average duration of standing oestrus was 6.4 +/- 4.3 h in cows and 6.2 +/- 3.9 h in heifers, respectively. It may be concluded that the duration of standing oestrus is substantially shortened in lactating dairy cows, and more than one-third of cows did not show standing oestrus. In cows showing standing oestrus, duration of expression of secondary oestrous signs before and after standing is not shortened. Duration of standing oestrus in heifers was as short as that in cows.  相似文献

Thirty days before expected time of parturition, 20 Holstein cows were divided into ?Cr and +Cr groups. From day 25 before parturition (BP ) up to day 30 after parturition (AP ), +Cr cows received 10 mg of Cr (chromium‐enriched yeast) daily. Muscle and adipose tissue samples were taken at days ?30, ?10, +7 and +10 related to parturition, when body condition score (BCS ) was also determined. Hepatic tissue samples were taken at days ?10 and +7. Tissue samples were used for determination of the insulin signalling pathway protein expressions. Intravenous glucose tolerance test (IVGTT ) was performed at days ?28, ?7, +10 and +30. Milk yield was recorded during first 14 weeks AP . Milk composition was obtained at days 7 and 28 AP . At day 10 BP , protein content of β ‐subunit of insulin receptor (IR β ) was significantly higher (p ? 0.05) in muscle, and phosphorylation of insulin receptor substrate 1 at serine 307 (pIRS ‐1 Ser307) was significantly lower (p ? 0.05) in hepatic tissue of +Cr group. After parturition, pIRS ‐1 Ser307 was significantly lower in muscle tissue at days 7 and 28 (p ? 0.05 and p ? 0.001, respectively), while phosphorylation of Akt at serine 473 (pA kt Ser473) was significantly higher (p ? 0.01) in hepatic tissue at day 7 AP in +Cr group. Chromium had opposite effect on insulin kinetics during IVGTT s obtained BP and AP . Insulin secretion was significantly reduced at day 7 BP and significantly enhanced at day 10 AP , when NEFA concentration was also significantly increased. Milk yield and ECM value were depressed in +Cr group. DMI and BCS were significantly enhanced in +Cr group at day 7 BP . In conclusion, chromium modulates insulin signalling pathway in dairy cows, but targeted signalling molecules are different in antepartal then post‐partal period, probably due to duration of exposure to chromium and different energy status between those periods.  相似文献

An experiment was conducted to examine the effect of progesterone prior to a GnRH‐PGF2α treatment on oestrus and pregnancy in seasonally anoestrous Awassi ewes. Twenty‐four ewes were randomly assigned to three groups to be pre‐treated with 60 mg medroxyprogesterone acetate sponges (group A), 600 mg progesterone sponges (group B) or blank sponges (group C) for 4 days. All ewes were injected with 100 μg of GnRH 24 h after sponge removal followed, 5 days later, by 20 mg PGF2α injection. Ewes were exposed to three fertile rams at the time of PGF2α injection (day 0, 0 h) and were checked for breeding marks at 6‐h intervals for 5 days. Blood samples were collected from all ewes 1 day (day ?10) prior to sponge insertion, at the time of sponge removal (day ?6), 1 day following sponge removal (day ?5, at the time of GnRH injection) and at the time of PGF2α injection (day 0) for analysis of progesterone. Progesterone concentrations on days ?10 and ?5 were basal and averaged 0.2 ± 0.04 and 0.2 ± 0.2 ng/ml, respectively. Progesterone concentrations on day ?6 were elevated only in group B ewes and were higher (p < 0.0001) than those of groups A and C. Progesterone concentrations on day 0 were higher (p = 0.002) in groups A and B than group C. Oestrous responses occurred only in ewes of groups A and B (p > 0.05). Induced oestrus conception rate was greater (p < 0.01) in group A than groups B and C. Ewes returned to oestrus 17–20 days following day 0 were two of eight, six of eight and three of eight of groups A, B and C, respectively, all of which eventually lambed. The overall lambing rate was 82% in progesterone‐primed ewes compared with only 38% non‐progesterone‐primed ewes (p < 0.05). Progesterone priming apparently sensitizes GnRH‐PGF2α‐treated seasonally anoestrous ewes and increases their response in oestrus and pregnancy rates.  相似文献

Blood seleno‐dependent glutathione peroxidase (SeGPX ) activity is widely used as a metabolic indicator of systemic antioxidative status despite inconsistent responses in the literature. This study aimed to compare SeGPX activity profiles in different blood fractions, expressed with different reference units, and assess their impact on interpretation of results. Two studies on selenium (Se) metabolism in gilts, including long‐term and peri‐oestrus SeGPX activity profiles, were submitted to analysis of variance with double repeated measures, after data set standardization. Differences between studies were experimental period (three post‐pubertal oestrus or five post‐pubertal oestrus +30 days of gestation) and sample type (whole blood or blood plasma). No difference was observed between whole‐blood long‐term profiles (three oestrus) for SeGPX activity/mg haemoglobin (SeGPX hb) vs. SeGPX activity/ml whole blood (SeGPX wb; p = 0.29). No long‐term difference was observed in whole blood between profiles according to dietary Se provision (basal and dietary Se‐supplemented groups; p ≥ 0.12). Blood plasma long‐term profiles (five oestrus + 30 days gestation) for SeGPX /mg blood plasma protein (SeGPX pro) were different from SeGPX /ml blood plasma (SeGPX pla) according or not to Se provision (p ≤ 0.007 and p < 0.001 respectively). However, regardless of Se provision (p ≥ 0.80), when excluding gestation from the model, blood plasma profiles were similar. During the peri‐oestrus period (day ?4 to +3), regardless of Se provision, SeGPX activity profiles differed according to reference units in both studies (p < 0.001). However, considering Se provision, similar profiles were observed in whole blood and blood plasma (p ≥ 0.27) for basal Se groups, whereas in Se‐supplemented groups they differed for both sample types (p ≤ 0.02). In conclusion, reference units influence interpretation of SeGPX activity according to physiological state. During oxidative stress periods, this effect depends upon dietary Se provision.  相似文献

The efficiency of treatments for unobserved oestrus and their effect on the reproductive performance of a dairy cattle herd with low oestrous detection rate till 60 days post‐partum (dpp), attributed to the declivous and slippery concrete floor were investigated. The herdsman requested advice in order to improve the mean days open of the herd, but no investments were allowed because a new unit was about to be built. Due to the low oestrus detection rate of the herd, the breeding policy was to inseminate at the first detected post‐partum oestrus. Cows were examined at 20–30 dpp to assess uterine involution, ovarian activity and prevalence of reproductive disorders and, at 60 dpp if no previous oestrus was detected. Each examination included palpation per rectum, ultrasound scanning and collection of a blood sample for plasma progesterone (P4) measurement. Cows with unobserved oestrus till 60 dpp were allocated either to a treatment group (n=139) or to a control group (n=139). Three treatments were used: (a) injection of PGF (PG) upon detection of a corpus luteum (CL; n = 30), cows not observed in oestrus being re‐injected 11–12 days later. AI was at oestrus; (b) PRID (n=35) or Crestar (n=74) devices kept in situ for 12 and 9 days, respectively, were associated to an injection of PG and of equine chorionic gonadotrophin (eCG) at device removal. Cows were double‐fixed time‐inseminated at 48 and 72 h after device removal. All treated cows were examined at 48–72 h after treatment to confirm oestrus. The percentage of cows detected in oestrus up to 60 dpp remained unchanged through the trial (35 and 47% for years before intervention: 1994–95; 51 and 48% for years of intervention: 1996–97). In contrast, the oestrous detection rate was high both in treated (93%) and control (100%) cows. This possibly resulted from an improvement in the oestrous detection efficiency of the herd's personnel and from examination of cows at 48–72 h after treatment. Treated and control cows had identical conception rate (CR; 36 and 37%, respectively) and reproductive performance. However, the mean days open of the herd in 1996 was significantly improved in comparison with previous years (mean ± SEM: 134 ± 6, 126 ± 5, 110 ± 4 and 114 ± 5 days, for years 1994, 1995, 1996 and 1997, respectively, p < 0.05, ANOVA ). Conception rate to AI up to 40 dpp was significantly reduced, compared with the period between 60 and 100 dpp but, mean days open were significantly improved in cows inseminated up to 60 dpp, compared with thereafter (p < 0.05).  相似文献

The aim of this study was to investigate the effect of applying a progesterone‐based oestrous synchronization protocol at 51–57 days postpartum in high‐producing dairy cows. The data analysed were derived from 1345 lactating cows. Cows between 51 and 57 days postpartum were assigned to the groups: control, PRID (receiving a progesterone‐releasing intravaginal device for 9 days, and prostaglandin F 24 h before PRID removal) or GnRH–PRID (the same as the PRID group plus GnRH at PRID insertion). Oestrus was detected by using pedometers and confirmed by examination of the genital tract at AI. Oestrous and conception rates before days 71–77 postpartum, pregnancy loss in early pregnant cows or the cumulative conception rate registered on day 120 postpartum were considered as the dependent variables in four consecutive logistic regression analyses. Based on the odds ratios, the oestrous rate increased by a factor of 1.73 in cows showing oestrus before treatment for each unit increase in the number of previous oestruses; decreased by a factor of 0.44 in the control group with respect to the treatment groups; and by a factor of 0.61 in cows without luteal structures at treatment with respect to cows with corpora lutea. The conception rates of cows inseminated before days 71–77 postpartum remained similar across the groups, whereas the likelihood of pregnancy loss for cows becoming pregnant during this period was 0.11 times lower in the PRID group than in the control. Based on the odds ratio, the likelihood of a higher cumulative conception rate on day 120 postpartum: increased in cows showing oestrus before treatment by a factor of 1.41 for each unit increase in the number of previous oestruses, was reduced 0.56‐fold in control cows compared with treated cows, and was also reduced by a factor of 0.98 for each kilogram of milk production increase recorded at treatment. In conclusion, although oestrous synchronization programmes performed in this study did not improve fertility, cows treated with progesterone could be inseminated earlier than untreated cows, such that the treatments increased the cumulative pregnancy rates determined on day 120 postpartum. In addition, fewer pregnancy losses were observed in early pregnant cows in the PRID group than the GnRH–PRID group.  相似文献

Abstract

Two herds of approximately 50 dairy cows were observed for oestrous behaviour for 6 weeks, 12 times a day for 30 minutes. It appeared that 3.08% of the pregnant cows showed oestrous behaviour during pregnancy (EBP) in such an intensity that they would have been considered in oestrus. With a less rigid oestrus detection threshold, 10.8% of the pregnant cows would have been considered to be in oestrus. Animals showed EBP during all months of pregnancy, but most of the behavior was observed in the middle of the gestation period.  相似文献

This report reviews the most recent developments in prostaglandin‐based oestrous synchronization programmes for postpartum dairy cows and addresses the efficiency of controlled breeding protocols based on such developments for cows with abnormal ovarian conditions. A double prostaglandin protocol applied 11–14 days apart seems to be capable of bringing most cows to oestrus. Because of varying oestrus onset times, improved conception rates are obtained following artificial insemination (AI) at detected oestrus rather than fixed‐time AI in prostaglandin‐treated cows. The administration of oestradiol or human chorionic gonadotrophin, or both these hormones, after prostaglandin treatment, improves the synchrony of oestrus yet does not enhance the conception rate. Progesterone‐based treatments for oestrous synchronization are considered the most appropriate for non‐cyclic or anoestrous postpartum dairy cows; prostaglandin alone being ineffective because of the absence of a mature corpus luteum in these cows. Improved oestrus synchrony and fertility rate have been reported using short‐term progesterone treatment regimes (7–9 days) with or without oestradiol benzoate combined with the use of a luteolytic agent given 1 day before, or at the time of, progesterone withdrawal. The ovulation synchronization (Ovsynch) protocol, based on the use of gonadotrophin releasing hormone and prostaglandin, was developed to coordinate follicular recruitment, CL regression and the time of ovulation. This protocol allows fixed time insemination and has proved effective in improving reproductive management in postpartum dairy cows. However, timed AI following Ovsynch seems to have no beneficial effects in heifers, because of an inconsistent follicle wave pattern, and in anoestrous cows, given their lack of prostaglandin responsive CL. To date, there are several prostaglandin based, fixed‐time insemination oestrous synchronization protocols for use in early postpartum dairy cows with ovarian disorders such as ovarian cysts and acyclicity.  相似文献

This work examined preventive effects of a dietary and a medical intervention measure on postpartum (p.p.) ketogenesis in dairy cows overconditioned in late pregnancy. Sixty German Holstein cows were allocated 6 weeks antepartum (a.p.) to three high body condition score (BCS) groups (BCS 3.95 ± 0.08) and one low BCS group (LC, BCS 2.77 ± 0.14). Concentrate proportion in diet a.p. was higher (60% vs. 20%) and increase in proportion p.p. from 30% up to 50% decelerated (3 vs. 2 weeks) in high BCS groups. High BCS cows received a monensin controlled‐release capsule (CRC) (HC/MO), a blend of essential oils (HC/EO) or formed a control group (HC). Performance parameters and energy status were evaluated in three periods [day (d) ?42 until calving, one until 14 days in milk (DIM), 15 until 56 DIM]. Feed efficiency was 65% and 53% higher in HC/MO than in LC (p < 0.001) and HC groups (p = 0.002) in the second period. Milk fat content was higher in HC/EO (5.60 vs. 4.82%; p = 0.012) and milk urea higher in HC/MO (135 mg/kg) than in LC cows (107 mg/kg; p < 0.001). Increased p.p. levels of non‐esterified fatty acids in serum were found in HC (p = 0.003), HC/MO (p = 0.068) and HC/EO (p = 0.002) in comparison with LC cows. Prevalence of subclinical and clinical ketosis was 54% and 46%, respectively, in HC group. Monensin decreased the prevalence to 50% and 7% respectively. Ruminal fermentation pattern showed higher proportions of propionate (23.43 mol % and 17.75 mol %, respectively; p < 0.008) and lower acetate:propionate ratio (2.66 vs. 3.76; p < 0.001) in HC/MO than HC group. Results suggest that a monensin CRC improved energy status and feed efficiency of transition dairy cows while essential oils failed to elicit any effect.  相似文献

The objectives of the present study were to quantify the effects of a biological chronic stressor (lameness) on the duration and frequency of different oestrous behaviours in parallel with milk hormone profiles. Dairy cows 51.8 ± 1.4 days postpartum (n = 59), including 18 non‐lame control cows, were scored for lameness and closely observed for signs of oestrus having had their follicular phases synchronized by administration of gonadotrophin‐releasing‐hormone (GnRH) followed by prostaglandin F (PG) 7 days later. Lameness shortened the period when herd‐mates attempted to mount the lame cows (1.83 ± 0.69 h vs 5.20 ± 1.53 h; p = 0.042) but did not affect the overall duration of total behaviours (lame 12.3 ± 1.3 h vs non‐lame 15.2 ± 1.3 h). Lameness also lowered the intensity of oestrus [1417 ± 206 points (n = 18) vs 2260 ± 307 points (n = 15); p = 0.029]. Throughout the synchronized oestrous period, lame cows mounted the rear of herd‐mates less frequently (p = 0.020) and tended to chin rest less (p = 0.075). Around the period of maximum oestrous intensity, lameness also diminished the proportion of cows mounting the rear of another cow and chin resting (p = 0.048, p = 0.037, respectively). Furthermore, lame cows had lower progesterone values during the 6 days before oestrous (p ≤ 0.05). Fewer lame cows were observed in oestrus following PG (non‐lame 83%, lame 53%; p = 0.030); however, if prior progesterone concentrations were elevated, lame cows were just as likely to be observed in oestrus. In conclusion, following endogenous progesterone exposure, lameness shortens the period when herd‐mates attempt to mount lame cows but does not affect the incidence of oestrous. However, lame cows are mounted less frequently and express oestrus of lower intensity. This is associated with lower progesterone prior to oestrus but not with abnormal oestradiol or cortisol profiles in daily milk samples.  相似文献

To ensure sufficient numbers of pregnant females, particularly at hotter times of the year, hormonal induction of gilt oestrus may be necessary. However, the gilt oestrus and ovulation responses to gonadotrophin treatment have often proven unpredictable. The objective of this study was to examine possible reasons for this unpredictability. Prepubertal gilts (approximately 150 days of age, n = 63) were assigned to one of three treatments: injection of 300 IU hCG (n = 15); pre-treatment with 100 mg FSH in polyvinylpyrrolidinone administered as 2 × 50 mg injections 24 h apart, followed by 600 IU eCG at 24 h after the second FSH injection (n = 23); or FSH pre-treatment as above followed by 300 IU hCG at 24 h after the second FSH injection (n = 25). To facilitate oestrus detection, gilts were exposed to a mature boar for 15 min daily for 7 days. Blood samples were obtained on the day of eCG or hCG injection and again 10 days later and gilt ovulation responses determined based on elevated progesterone concentrations. The oestrus responses by 7 days were 6.7%, 17.5% and 64.0% for gilts treated with hCG, FSH + eCG and FSH + hCG, respectively (p < 0.001). The oestrous gilt receiving hCG alone and one oestrous FSH + hCG gilt did not ovulate, all other oestrous gilts ovulated. A further two anoestrous FSH + eCG-treated gilts ovulated. These data suggest that FSH pre-treatment facilitated the development of ovarian follicles to the point where they became responsive to hCG, but had little effect on the response to eCG.  相似文献

AIM: To compare the sensitivity, specificity, predictive values and accuracy of detection of oestrus using a novel oestrus detection strip (ODS) and a camera-software device (CSD) with typical farm management practices of visual observation and use of tail paint in dairy cattle at pasture. METHODS: Dairy cows (n = 480) in a seasonal-calving herd managed at pasture under typical commercial conditions in New Zealand were stratified by age, body condition score and days in milk, then randomly allocated to one of two groups prior to the planned start of mating (PSM). Tail paint was applied to all cows and oestrus detected by visual observation of oestrous behaviour and removal of paint, by farm staff. One group (n = 240) was fitted with ODS and also monitored for signs of oestrus using a CSD, while the Control group (n = 240) was monitored using tail paint and visual observations only. Cows detected in oestrus were artificially inseminated (AI), and pregnancy status determined using rectal palpation and ultrasonography, 51-52 days after the end of a 55-day A period. Results of pregnancy diagnosis were used to confirm the occurrence of oestrus, and the sensitivity, specificity, predictive value and accuracy of detection of oestrus compared between oestrus detection methods. RESULTS: The sensitivity and accuracy of oestrus detection in the Control group, using visual observation and tail paint, were low. Compared with the Control group, detection of oestrus using the ODS and CSD resulted in greater sensitivity (85% vs 78%; p = 0.006), specificity (99.6% vs 98.0%; p < 0.001), positive predictive value (PPV; 88% vs 51%; p < 0.001) and overall accuracy (99.0% vs 98.0%; p < 0.001). Negative predictive value (NPV) did not differ significantly between groups (99.4% vs 99.3%; p = 0.28). Pregnancy rate to first service was higher in the CSD group than in the Control group (72% vs 39%; p < 0.05). Use of the CSD significantly increased the cumulative proportion of cows pregnant to AI over the breeding period (p = 0.044). CONCLUSION AND CLINICAL RELEVANCE: The ODS and CSD was satisfactory for detection of oestrus in seasonal calving dairy herds grazing on pasture and could improve the sensitivity and accuracy of detection of oestrus in herds where these are low.  相似文献

The aim of this study was to examine the possible differences of oestrous intensity between natural oestrus and induced oestrus using the walking activity measuring device. Walking activity was used as an evaluation index of oestrous intensity. A total of 27 Japanese Black cows, more than 40 days after calving and clinically normal, were randomly assigned to three groups. Walking activity was recorded using a commercially available computerized pedometer system. The treatment groups consisted of an Ovsynch (n = 8) and a controlled internal drugs releasing device (CIDR) + Ovsynch (n = 9) group. The control group (n = 10) received no treatment. Walking activity was examined in all groups. Timed artificial insemination (timed AI) was performed at 16 hours after the onset of oestrus in the control group and at 24 h after second administration of GnRH in the treatment groups. Duration of oestrus had a tendency to be shorter in both the Ovsynch and the CIDR + Ovsynch groups when compared with the control group. The time required from the onset of oestrus to the time showing the highest number of steps of walking (the time to peak) showed a tendency to be shorter in CIDR + Ovsynch group. The number of steps of walking at peak and overall walking activities were significantly lower in both treatment groups than in the control group. Both activity and super-activity periods of time in the treatment groups were shorter than the control group. No difference was observed in the conception rate between the control (50.0%; 10/20), Ovsynch (50.0%; 4/8) and CIDR + Ovsynch groups (66.7%; 6/9). This study demonstrates that the oestrous intensity of cows in oestrus was different between natural oestrus and induced oestrus and also between the methods of the synchronization, but no difference was observed in the conception rate among the three groups.  相似文献

The study was conducted on six Murrah buffalo synchronized and induced to oestrus. An indwelling catheter was placed in the jugular vein of each buffalo 4 days before the expected onset of the oestrus following the induced oestrus and blood samples were collected at 8 h intervals from each animal throughout the oestrous cycle. Plasma immunoreactive inhibin, follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol-17β and progesterone were estimated by radioimmunoassay to study the variations in the peripheral levels of these hormones and their inter-relationships in order to elucidate the feedback systems controlling them during the oestrous cycle of buffalo. Plasma inhibin levels ranged between 391.25 and 631.97 pg/ml during various phases of the oestrous cycle and were found to be higher than reported in cows. Peak LH and FSH levels during oestrus were 38.40 ± 9.21 and 24.04 ± 4.75 ng/ml, respectively and estradiol-17β and progesterone were 19.50 ± 5.51 pg/ml and 0.61 ± 0.25 ng/ml, respectively. The mean plasma inhibin concentration on the day of oestrus was 562.5 ± 18.9 pg/ml. Levels of FSH in the plasma showed three mid-cycle elevations which corresponded to comparatively lower inhibin and elevated estradiol-17β levels during the same period. From this observation it was deduced that both inhibin and estradiol-17β have a feed-back regulatory effect on FSH secretion in buffalo.  相似文献

Herbage feeding with only little input of concentrates plays an important role in milk production in grassland dominated countries like Switzerland. The objective was to investigate the effects of a solely herbage‐based diet and level of milk production on performance, and variables related to the metabolic, endocrine and inflammatory status to estimate the stress imposed on dairy cows. Twenty‐five multiparous Holstein cows were divided into a control (C+, = 13) and a treatment group (C?, = 12), according to their previous lactation yield (4679–10 808 kg) from week 3 ante partum until week 8 post‐partum (p.p.). While C+ received fresh herbage plus additional concentrate, no concentrate was fed to C? throughout the experiment. Within C+ and C?, the median of the preceding lactation yields (7752 kg) was used to split cows into a high (HYC+, HYC?)‐ and low‐yielding (LYC+, LYC?) groups. Throughout the study, HYC+ had a higher milk yield (35.9 kg/d) compared to the other subgroups (27.2–31.7 kg/d, p < 0.05). Plasma glucose (3.51 vs. 3.72 mmol/l) and IGF‐1 (66.0 vs. 78.9ng/mL) concentrations were lower in HYC?/LYC? compared to HYC+/LYC+ cows (p < 0.05). Plasma FFA and BHBA concentrations were dramatically elevated in HYC? (1.1 and 1.6 mmol/l) compared to all other subgroups (mean values: 0.5 and 0.6 mmol/l, p < 0.05). Saliva cortisol, plasma concentrations of serum amyloid A (SAA), haptoglobin (Hp), beta‐endorphin (BE) and activity of alkaline phosphatase (AP) were not different between C+ and C?. In conclusion, herbage‐fed high‐yielding cows without supplementary concentrate experienced a high metabolic load resulting in a reduced performance compared to cows of similar potential fed accordingly. Low‐yielding cows performed well without concentrate supplementation. Interestingly, the selected markers for inflammation and stress such as cortisol, Hp, SAA, BE and AP gave no indication for the metabolic load being translated into compromised well‐being.  相似文献

This study aimed to evaluate the effect of oestrous synchrony between donors and recipients and the embryo quality on the pregnancy rate in beef cow recipients. The experiment was performed over two years at an embryo transfer (ET ) centre in Southern Brazil. Ninety Aberdeen Angus cows were subjected to superovulation (SOV ) protocols, resulting in the recovery of 1,048 transferable embryos. Eleven groups were formed with intervals of 6 hr, from ?30 to +30 hr, with respect to recipient versus donor oestrous detection. Evaluation of embryo quality was according to the IETS guidelines. The overall pregnancy rate was 52%. Effects related to donor and recipient oestrous synchronization on pregnancy rate were observed (=  .01), ranging from 36% to 50%. The embryo quality rate affected the pregnancy rate, where Grade I resulted in 57% and Grade III in 43% of pregnancy (<  .001). The embryonic state (frozen or fresh) showed no (>  .05) effect on pregnancy rate: 53% for fresh embryos and 44% for frozen embryos. The odds ratio for explanatory variables causing pregnancy indicated that Grade III embryos had 31% less chance of conception compared to Grade I. Thus, oestrous synchrony between donor and recipient, considering ±30 hr apart, can affect the pregnancy rate along with embryo quality.  相似文献
