Common beans are rich in phenolic compounds, which can provide health benefits to the consumer. The objective of this work was to study the relationship among antimutagenicity, antioxidant and enzymatic activities of methanolic extract and trolox by principal components multivariate analysis. Antimutagenicity of phenolic compounds present in methanolic extract from the seed coat of common beans (P. vulgaris, Flor de Mayo Bajío cultivar) and trolox against AFB1 mutagenicity were evaluated in the Salmonella typhimurium microsuspension assay. Antioxidant capacity of methanolic extract and trolox were evaluated using β-carotene and 1,1-diphenyl-2-picryhydrazyl (DPPH) in vitro model assays. Cythrome P450 activity was measured by fluorometric assay. For phenolic extract, trolox and phenolic extract + trolox, the inhibition on AFB1 mutagenicity in tester strain TA100 was 47, 59 and 69%, respectively. While in TA98 was 39, 48 and 68%. The inhibition of phenolic compounds, trolox and phenolic compounds + trolox on cytochrome P450 (CYP450) activity was 48, 59 and 88%, respectively. Correlation analysis showed that phenolic extract and trolox have high antimutagenic and antioxidant activity and also inhibited enzymatic activity. The results suggest that the primary mechanism of action of phenolic compounds in beans against AFB1 mutagenicity may be extra-cellular in the microsuspension assay. 相似文献

In Thailand, beverages containing mulberry leaf (Morus alba L.) are believed to promote good health, especially in people with diabetes. We examined the effects of long-term administration
of an ethanolic extract of mulberry leaf (MA) on blood glucose, oxidative damage, and glycation in streptozotocin-induced
diabetic rats. Daily administration of 1 g/kg MA for six weeks decreased blood glucose by 22%, which was comparable to the
effect of 4 U/kg insulin. Lipid peroxidation, measured as malondialdehyde and lipid hydroperoxide concentrations (3.50 ± 0.33
and 3.76 ± 0.18 μM, respectively) decreased significantly (P < 0.05) compared to nontreated control diabetic rats (8.19 ± 0.45 and 7.50 ± 0.46 μM, respectively). Hemoglobin A1C, a biomarker for chronic exposure to high concentration of glucose, was also significantly decreased in the MA-treated group
(6.78 ± 0.30%) in comparison to untreated group (9.02 ± 0.30%). The IC50 of in vitro antiglycation and free radical scavenging activities of MA were 16.4 ± 5.6 μg/ml and 61.7 ± 2.1 μg/ml, respectively. These
findings support that long-term administration of MA has antihyperglycemic, antioxidant and antiglycation effects in chronic
diabetic rats, which may be beneficial as food supplement for diabetics. 相似文献

This study was designed to examine the in vitro antimicrobial and antioxidant activities and the amount of total phenolics of the methanol extracts of Ballota rotundifolia L. and Teucrium chamaedrys C. Koch. In the case of antimicrobial activity tests, polar sub-fractions of the methanol extracts of plant species exhibited
weakest antimicrobial activity when compared with the non-polar ones. While, non-polar sub-fraction of B. rotundifolia showed moderate activity against A. lwoffii, C. perfringens and the yeasts, T. chamaedrys performed excellent activity pattern against all of the tested microorganisms. The sub-fractions were also screened for their
possible antioxidant activities by two complementary tests, namely DPPH free radical-scavenging and β-carotene/linoleic acid assays. Non-polar extracts of the plant species remained inactive in both test systems. On the other
hand polar extracts showed remarkable antioxidant activities. In DPPH system, free radical scavenging effect of T. chamaedrys was measured as 18.00 ± 1.42 μg.mg−1. It is extremely important to point out that, polar sub-fraction of T. chamaedrys is found as effective as the positive control BHT. Non-polar sub-fraction of T. chamaedrys found to have the highest total phenolic amount (97.12 ± 1.28 μg/mg). Results obtained from this experiment confirm the relationship

Fourteen apricot genotypes grown under similar cultural practices in Trans-Himalayan Ladakh region were studied to find out
the influence of genotype on antioxidant capacity and total phenolic content (TPC) of apricot kernel. The kernels were found
to be rich in TPC ranging from 92.2 to 162.1 mg gallic acid equivalent/100 g. The free radical-scavenging activity in terms
of inhibitory concentration (IC50) ranged from 43.8 to 123.4 mg/ml and ferric reducing antioxidant potential (FRAP) from 154.1 to 243.6 FeSO4.7H2O μg/ml. A variation of 1–1.7 fold in total phenolic content, 1–2.8 fold in IC50 by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 1–1.6 fold in ferric reducing antioxidant potential among the examined
kernels underlines the important role played by genetic background for determining the phenolic content and antioxidant potential
of apricot kernel. A positive significant correlation between TPC and FRAP (r = 0.671) was found. No significant correlation was found between TPC and IC50; FRAP and IC50; TPC and physical properties of kernel. Principal component analysis demonstrated that genotypic effect is more pronounced
towards TPC and total antioxidant capacity (TAC) content in apricot kernel while the contribution of seed and kernel physical

This study describes the antioxidant activities and antigenotoxic effects of garlic extracts prepared by different processing
methods. Aged-garlic extract (AGE) showed a significantly higher total phenolic content (562.6 ± 1.92 mg/100 g garlic acid
equivalents) than those of raw garlic extract (RGE) or heated garlic extract (HGE). The SC50 for DPPH RSA in HGE was significantly the highest at 2.1 mg/ml. The SC50 for SOD-like activity in garlic extracts was, in decreasing order, RGE (7.3 mg/ml) > AGE (8.5 mg/ml) > HGE (9.2 mg /ml).
The ED50 of AGE was the highest (19.3 μg/ml) regarding H2O2 induced DNA damage and its inhibition rate was 70.8%. The ED50 of RGE for 4-hydroxynonenal (a lipid peroxidation product) induced DNA damage was 38.6 μg/ml, followed by AGE > HGE. Although
the heat treatment of garlic tended to decrease the TPC and SOD-like activity and increased DPPH RSA, garlic, in general,
has significant antioxidant activity and protective effects against oxidative DNA damage regardless of processing method. 相似文献

We report for the first time the antioxidant and antimutagenic activities of fractions from Randia echinocarpa fruit, which is a Rubiaceae plant native to Sinaloa, Mexico. This fruit has been traditionally used in the prevention or treatment of cancer, among other
diseases. The pulp of the fruit was sequentially extracted with solvents of different polarity (i.e. hexane, chloroform, methanol
and water). A high extraction yield was obtained with methanol (72.17% d.w.). The aqueous extract showed the highest content
of phenolics (2.27 mg/g as ferulic acid equivalents) and the highest antioxidant activity based on the β-carotene bleaching
method (486.15). The commercial antioxidant BHT was used as control (835.05). Antimutagenic activity of the aqueous extract
(0–500 μg/tube) was evaluated using the Salmonella microsuspension assay (YG1024 strain) and 1-NP as the mutagen (50 and 100 ng/tube). The aqueous extract was neither toxic
nor mutagenic and the percentage of inhibition on 1-NP mutagenicity was 32 and 53% at doses of 50 and 100 ng/tube, respectively.
The results of the double incubation assay suggest that the extract inhibited the mutagenicity of 1-NP by a combination of
desmutagenic and bioantimutagenic effects. 相似文献

Porophyllum tagetoides is an annual warm-weather herb that has an intense typical smell. Its leaves are commonly used in soup preparation and traditional
medicine for treatment of inflammatory diseases. Its volatile compounds and antioxidant properties were evaluated in crude,
aqueous and ethanol leaf extract and an oil emulsion using different antioxidant assays in vitro, such as: DPPH radical scavenging activity, redox potential, polyphenol content, reducing power and optical density. A high
antioxidative activity was found when comparing leaves with stems. The crude extract from leaves showed a very high reducing
power (2.88 ± 0.20 O.D.) and DPPH radical-scavenging activity (54.63 ± 4.80%), in concordance with a major concentration of
vitamin C (23.97 ± 0.36 mg/100 g). Instead, the highest polyphenol content (264.54 ± 2.17 mg GAE/g of sample) and redox potential
(561.23 ± 0.15 mV) were found by the ethanol and aqueous extract, respectively. Aldehydes and terpenes such as nonanal, decanal,
trans-pineno, β-myrcene and D-limonene were the major volatiles found. This study suggests that Porophyllum tagetoides extracts could be used as antioxidants. 相似文献

Avocado seed contains elevated levels of phenolic compounds and exhibits antioxidant properties. We investigated the effect
of Avocado Seed Flour (ASF) on the lipid levels in mice on a hyperlipidemic diet. The concentration of phenols was determined
by high-performance liquid chromatography, antioxidant activity was evaluated using the Trolox equivalent antioxidant capacity
method, and dietary fiber was measured using the Association of Official Analytical Chemists (AOAC) method. The LD50 of ASF was determined using Lorke’s method and hypolipidemic activity was evaluated in a hypercholesterolemic model in mice.
Protocatechuic acid was the main phenolic compound found in ASF, followed by kaempferide and vanillic acid. The total phenolic
content in the methanolic extract of ASF was 292.00 ± 9.81 mg gallic acid equivalents/g seed dry weight and the antioxidant
activity resulted in 173.3 μmol Trolox equivalents/g DW. In addition, a high content of dietary fiber was found (34.8%). The
oral LD50 for ASF was 1767 mg/kg body weight, and treatment with ASF significantly reduced the levels of total cholesterol, LDL-C,
and prediction of the atherogenic index. Therefore, the antioxidant activity of phenolic compounds and dietary fiber in ASF
may be responsible for the hypocholesterolemic activity of ASF in a hyperlipidemic model of mice. 相似文献

Organic solvent (methanol, ethanol, and acetone) extracts and water extracts of cherry (Prunus serrulata var. spontanea) blossoms were prepared, and antioxidant activities of the extracts were evaluated. Methanolic CBE (100 μg/ml) showed the
highest total phenol content (104.30 μM), radical scavenging activity (34.2%), and reducing power (0.391). The effect of CBE
on DNA damage induced by H2O2 in human leukocytes was evaluated by Comet assay. All CBE was a potent dose dependent inhibitor of DNA damage induced by
200 μM of H2O2, methanolic CBE showed the most strong inhibition activity. The methanolic CBE of 500 μg/ml showed 38.8% inhibition against
growth of human colon cancer cell line HT-29. These results indicated that cherry blossoms could provide valuable bioactive

To identify the potential of green leafy vegetables (GLV) as antioxidants, methanolic extracts of Amaranthus sp., Centella asiatica, Murraya koenigii and Trigonella foenum graecum were studied for their antioxidant activity in different systems at multiple concentrations. Total antioxidant activity assessed
by phosphomolybdenum method, free radical scavenging activity by 1,1-diphenly-2-picryl hydrazyl (DPPH), reducing power and
ferrous ion chelating activity were determined. The GLV were analyzed for ascorbic acid, total and β-carotene and total polyphenol
contents. The ascorbic acid, total carotene, β-carotene and total phenolic content (tannic acid equivalents) of the GLV ranged
between 15.18–101.36, 34.78–64.51, 4.23–8.84 and 150.0–387.50 mg/100 g GLV, respectively. The extracts were found to have
significantly different levels of antioxidant activities in the systems tested. The total antioxidant activity was highest
in Murraya koenigii (2,691.78 μmol of ascorbic acid/g sample) and least in Centella asiatica (623.78 μmol of ascorbic acid/g sample). The extract concentration causing 50% inhibition of DPPH (IC50) was determined (M. koenigii < C.asiatica < Amaranthus sp. < T. graecum). The maximum DPPH scavenging activity and reducing power was exhibited by Murraya koenigii. Multiple regression analysis showed that the relationship of total antioxidant activity, free radical scavenging activity,
and reducing power with polyphenol and total and β-carotene was highly significant.

Antioxidant activity of pressurized low polarity water (PLPW) extracts of cow cockle seed and extraction residues were determined
using DPPH, ABTS, and FRAP assays. The effect of extraction conditions (temperature (125, 150 and 175 °C) and time) on the
antioxidant activity and the relationship amongst the antioxidant activity and extract composition (total phenolics and saponin
content) were determined. The antioxidant activity of PLPW extracts increased with extraction temperature. Increasing activity
with time was also observed at 175 °C. PLPW extraction residues had the highest activity suggesting antioxidant compounds
were not completely extracted by PLPW. Antioxidant activity correlated well with total phenolics content of samples (R
2 ≥ 0.94), however no correlation was observed with the saponin content. A strong correlation was observed between the antioxidant
activity values obtained using different methods (R
2 ≥ 0.94). These results point to the potential of PLPW extraction as a method to modify the activity of biological materials

There is evidence that a diet rich in fruit and vegetables may reduce the risk of cancer and other degenerative diseases.
However, potential health impact of bioactive phytochemicals is limited by their low amount and relatively poor bioavailability.
It has been suggested that the health benefits associated with fruit and red wine consumption could be due to the whole antioxidant
pool of the diet microcomponents. In this study, the antioxidant activities of trans-resveratrol, pterostilbene and quercetin, and the effect of their combination were investigated in human erythrocytes in vitro. H2O2-induced lipid peroxidation was assessed by measuring the amount of thiobarbituric acid reactive species. Quercetin and pterostilbene
protected erythrocyte membranes against lipid peroxidation (IC50 values = 64 ± 8.7 μM and 44.5 ± 7.8 μM, respectively). Resveratrol was significantly less effective. However, the three compounds
protected the erythocytes against hemolysis and GSH (reduced glutathione) depletion to the same extent. Combinations consisting
of two compounds (molar ratio 1:1) influenced lipid peroxidation in a concentration–dependent manner. At lower concentrations,
resveratrol with quercetin or pterostilbene inhibited synergistically the oxidative injury of membrane lipids At higher concentrations,
an additive effect was observed. These protective effects may partially explain the health benefit of these bioactive microcomponents

Bromelia pinguin L. is a plant native to Sinaloa, Mexico, where its fruit is used as food or as a phytotherapeutic agent. The fruits of B. pinguin were characterized and they could be considered as a functional food. These fruits show an average weight of 13.7 g and a
yellow color of high luminosity (b* = 43.2, L* = 74.5). The values for acidity (4.6%, as citric acid) and pH (3.7) of B. pinguin fruit are similar to those of citrics. The edible portion is characterized by a high content of vitamin C (126 mg/100 g),
ash (10.6 g/100 g d.w.), crude fiber (3.4 g/100 g d.w.), calcium (1290 mg/100 g d.w.), magnesium (500 mg/100 g d.w.), manganese
(2.95 mg/100 g d.w.) and it is a good source of zinc (2.8 mg/100 g d.w.). Polar fractions extracted from the pulp fruit showed
activity against several genera of human pathogenic-bacteria (Staphylococcus, Streptococcus, Enterococcus, Salmonella, Shigella, Escherichia and Pseudomonas). 相似文献

Screening of natural biodiversity for the better quality traits are of prime importance for quality breeding programs. The
objective of this investigation was to select candidate accession of bean having high concentrations of protein as well as
macro and micro minerals with good cooking quality for use as parents in breeding programme for these compounds. Thirty-five
accessions of bean (Phaseolus vulgaris L) were field grown and their seeds were analyzed for their cooking quality and nutritional composition. Wide variations
were observed in most of the measurements e.g. protein (18.7–26.2%), iron (79.4–137.6 ppm) and hardness after cooking (4.65–9.88 Kg)
suggesting that there are considerable levels of genetic diversity. Across all accessions the concentration of potassium was
negatively correlated with protein (r = −0.43, P < 0.05). Concentrations of protein was significantly greater in accessions VIII, XIII and XIX compared to other accessions
analyzed. Iron concentrations were greatest (137 ppm) in XIX and lowest (79 ppm) in XXVII. Lines with less cooking time were
line III, X, XXVI, XXX and XXXI. Bean line XIX contains high protein (24.9%) with high zinc (33.3 ppm) and highest iron (137.6 ppm),
but it has high hardness after cooking (7.32 kg). Four clusters were computed by cluster analysis that explained quite a good
variation in the traits. The great variability for these attributes suggests that these selected accessions may be useful
as parents in hybridization programs to produce bean with value-added traits. This information was also potentially useful

Nixtamalization process is the first step to obtain maize based products, like tortillas; however, in both the traditional and commercial processes, white grain is generally preferred. Creole maize races, mainly pigmented varieties, have increasingly attention since these are rich in anthocyanins and carotenoids. The aim of this investigation was to evaluate the antioxidant and antimutagenic activity of rich anthocyanins and carotenoids extracts from creole maize races before (grain) and after (masa and tortilla) the nixtamalization process. Most anthocyanins and carotenoids were lost during nixtamalization. Before nixtamalization, blue and red genotypes contained either higher antioxidant capacity and anthocyanin contents (963?±?10.0 and 212.36?±?0.36 mg of cyanidin-3-glucoside eq/100 g, respectively) than the white and yellow genotypes. However, the highest carotenoid levels were displayed by red grains (1.01?±?0.07 to 1.14?±?0.08 μg of β-carotene eq/g extract). Anthocyanins losses were observed when the blue grains were processed into masa (83 %) and tortillas (64 %). Anthocyanins content correlated with antiradical activity (r?=?0.57) and with 2-aminoanthracene -induced mutagenicity inhibition on TA98 and TA100 (r?=??0.62 and r?=??0.44, respectively). For white grains, nixtamalization also reduced carotenoids (53 to 56 %), but not antioxidant activity and 2-Aa-induced mutagenicity. Throughout the nixtamalization process steps, all the extracts showed antimutagenic activity against 2-aminoanthracene—induced mutagenicity (23 to 90 %), displaying higher potential to inhibit base changes mutations than frameshift mutations in the genome of the tasted microorganism (TA100 and TA98, respectively). A simple and rapid method for the determination of naturally occurring, potentially hazardous glycoalkaloids (GAs) in potatoes
and their products has been developed. The procedure is based on the on-line solid-phase extraction of the acetic acid extracts
from potato products and combined with liquid chromatography (LC)-mass spectrometry (MS) in a fully automated system (Symbiosis™,
Spark Holland Instruments, Emmen, The Netherlands). As sorbent material HySphere™ 18HD was used for alkaloid enrichment. GAs
were eluted with the LC gradient and directly analysed by MS. Detection of the analytes was achieved in the sensitive multiple
reaction monitoring mode using two characteristic ions (m/z 98 as a qualifier for GAs and m/z 868.3 as a quantifier for α-solanine or m/z 852.4 for α-chaconine). Typical validation data for method precision (v
k α-solanine = 5.3–6.5, v
k α-chaconine = 3.4–15.4), accuracy (average recovery of α-solanine = 84%, average recovery of α-chaconine = 87%) and linearity
over the range from 1 to 1,000 ng ml−1 (R
2 = 0.9915 for α-solanine, R
2 = 0.9939 for α-chaconine) with detection limits of 0.3 ng ml−1 for α-chaconine and 0.5 ng ml−1 for α-solanine were obtained. GA contents of commercial potato products were determined by the new on-line method and afterwards
compared with those obtained with an established high-performance LC routine procedure. Better performance of the on-line
procedure was obvious from the standard deviations of both methods. Other advantages included a strong reduction of overall
analysis time, human intervention and solvent consumption as well as waste production. The time required for the on-line analysis

The aqueous extracts of four marine algae, Caulerpa racemosa var. macrophysa, Gracilaria tenuistipitata var. tenuistipitata, Sargassum sp., and Ulva lactuca, from the coastal areas in Southern Thailand, were prepared by boiling dried seaweed powder in water for 3 h, and by autoclaving
each sample at 120 °C for 3 h. They were then freeze-dried and evaluated for their antioxidant activities using DPPH (1, 1-diphenyl-2-picrylhydrazyl),
hydroxyl radical (OH•) and superoxide anion (O2•−) scavenging assays. Boiling extracts of the seaweeds, except C. racemosa, were found to have higher total phenolic contents (TPC) than those obtained from the autoclave method. The antioxidant results
also showed that O2•− scavenging activity existed only in the boiling extracts of C. racemosa, G. tenuistipitata, and U. lactuca. In DPPH and OH• assays, however, almost all the boiling extracts were less active than the autoclave ones. Among the four alga species, Sargassum sp. was the most active. Both extracts of this seaweed had the highest TPC and also displayed the strongest DPPH• and OH• inhibitory activities. A strong positive-correlation between the antioxidant potency and TPC of the autoclave extracts was
found, while for the boiling extracts such relation was very weak. This result thus reflected that in addition to the phenolic
compounds, there might be some other active components present in these extracts involved in the antioxidant activity. 相似文献

(WEP) in TB. Lactobacillus bulgaricus and Streptococcus thermophilus were used as starter cultures. Lactobacillus acidophilus and Bifidobacterium lactis were used as probiotics. A concentration of 4% TB in yogurt was determined to be the maximum amount that could be added without
causing synersis. By day 7, the number of bacteria greatly increased in yogurt samples containing TB and maintained higher
viable bacteria counts at the end of the cold storage period, in comparison to controls (P ≤ 0.05). Confirming this data was the lower pH levels and higher TTA values of TB yogurt samples exhibited throughout 28 days
(P ≤ 0.05). Polysaccharide extracts of TB exhibited strong antioxidant activity with an ORAC value of 33.86 ± 2.30 μmol trolox
equivalents (TE)/g of bran. Results of this study suggest that TB may serve as a new prebiotic and antioxidant source for

The Cosmos bipinnatus has been used in a traditional herbal remedy for various diseases such as jaundice, intermittent fever, and splenomegaly.
The present study describes the preliminary evaluation of antioxidant activities and antigenotoxic effect of Cosmos bipinnatus flowers according to four different colors (white, pink, orange, and violet). The antioxidants properties were evaluated by determining TPC, DPPH RSA, ABTS RSA, and RP. The highest TPC of methanolic
CFE (at concentration of 1 mg/ml) showed in violet colored CF (1,013 μM), and IC50 of DPPH RSA, ABTS RSA, and RP were also the lowest in violet colored CFE with values of 0.61, 1.48, and 0.82 mg/ml, respectively.
The antigenotoxic effect of the CFE on DNA damage induced by H2O2 in human leukocytes was evaluated by Comet assay. Pretreatments with CFE produced significant reductions in oxidative DNA
damage at the concentration of 500 μg/ml, except for violet colored CFE. The ED50 value of white colored CFE has shown the highest inhibition (0.40 mg/ml) on H2O2 induced DNA damage, followed by orange > pink > violet color. These results suggested that Cosmos bipinnatus has significant antioxidant activity and protective effect against oxidative DNA damage. 相似文献

Carotenoids are pigments responsible for the yellow-reddish color of many foods and are related to important functions and
physiological actions, preventing several chronic-degenerative diseases. The objective of this study was to confirm the carotenoid
composition of jackfruit by high-performance liquid chromatography connected to photodiode array and mass spectrometry detectors
(HPLC-PDA-MS/MS). The main carotenoids were all-trans-lutein (24–44%), all-trans-β-carotene (24–30%), all-trans-neoxanthin (4–19%), 9-cis-neoxanthin (4–9%) and 9-cis-violaxanthin (4–10%). Either qualitative or quantitative differences, mainly related to the lutein proportion, were found
among three batches of jackfruit. Since the fruits from batch A showed significantly lower contents for almost all carotenoids,
it also had the lowest total carotenoid content (34.1 μg/100 g) and provitamin A value, whereas the total carotenoid ranged
from 129.0 to 150.3 μg/100 g in the other batches. The provitamin A values from batches B and C were 3.3 and 4.3 μg RAE/100 g,
respectively. The carotenoid composition of jackfruit was successfully determined, where 14 of the 18 identified carotenoids
were reported for first time. Differences among batches may be due to genetic and/or agricultural factors. 相似文献
