Seventeen isolates of the free-living soil fungus Trichoderma spp., collected from Meloidogyne spp. infested vegetable fields and infected roots in Benin, were screened for their rhizosphere competence and antagonistic potential against root-knot nematodes, Meloidogyne incognita, in greenhouse pot experiments on tomato. The five isolates expressing greatest reproductive ability and nematode suppression in pots were further assessed in a typical double-cropping system of tomato and carrot in the field in Benin. All seventeen isolates were re-isolated from both soil and roots at eight weeks after application, with no apparent crop growth penalty. In pots, a number of isolates provided significant nematode control compared with untreated controls. Field assessment demonstrated significant inhibition of nematode reproduction, suppression of root galling and an increase of tomato yield compared with the non-fungal control treatments. Trichoderma asperellum T-16 suppressed second stage juvenile (J2) densities in roots by up to 80%; Trichoderma brevicompactum T-3 suppressed egg production by as much as 86%. Tomato yields were improved by over 30% following the application of these biocontrol agents, especially T. asperellum T-16. Although no significant effects were observed on carrot galling and yield, soil J2 densities were suppressed in treated plots, by as much as 94% (T. asperellum T-12), compared with the non-fungal controls. This study provides the first information on the potential of West-African Trichoderma spp. isolates for use against root-knot nematodes in vegetable production systems. The results are highly encouraging, demonstrating their strong potential as an alternative and complementary crop protection component.  相似文献

The potential of the invertebrate communities in agricultural and natural soils to suppress Meloidogyne incognita on coleus (Coleus blumei) was evaluated in two greenhouse experiments. Soil from adjacent natural and agricultural habitats was collected from four locations that had very different soil types (loamy sand, muck, sand and rock). Soil of each type was placed into pots, planted with coleus seedlings, inoculated with 2000 eggs of M. incognita, and arranged in a 4 × 2 factorial design. Root-knot nematodes were suppressed in natural soil compared to agricultural soil for loamy sand, sand and rock soils, but not for muck soil. Plants grew larger and had less root galling in natural soils than in agricultural soils. Free-living nematodes (bacterivores, fungivores, omnivores and predators) were monitored but did not show population patterns consistent with the suppression of root-knot nematodes observed in natural soils. Reasons for the opposite result (higher root-knot levels in natural than in agricultural soil) on muck soil are unknown, but this soil type was very different from the other soils in its unusually high organic matter content and very low levels of omnivorous and predatory nematodes. The agricultural muck in particular was unusually low in fungivorous nematodes and extremely high in levels of sodium and macronutrients. Despite the exception on the muck soil, the relative suppression of root-knot nematodes in natural compared to agricultural soils of the other three soil types (loamy sand, sand and rock) support the idea that potential for nematode suppression may be greater in natural than in agricultural soils.  相似文献

连作番茄根区病土对番茄生长及土壤线虫与微生物的影响   总被引:4,自引:0,他引:4  
探索连作番茄根区病土对番茄根结线虫病的诱导效果及引起连作障碍的微生态机制,可为深入了解番茄连作障碍发生机理及探究番茄连作障碍防治方法提供科学依据。本研究利用盆栽试验,测定了番茄在健康土壤及接种病土土壤中生物学特性变化及根结线虫侵染状况,并分析鉴定了土壤中微生物及线虫的种类与数量。结果表明,接种连作番茄根结线虫病株根区病土会对番茄生长及根结线虫侵染产生影响:1)番茄苗期根系根结数达9个?株~(-1),健康土壤无根结;土壤线虫数量较健康土壤增加390.4%;收获期番茄根结线虫侵染率达62.7%,病情指数为80.0%。2)番茄生长受到抑制,叶片防御酶活性降低,收获期茎叶及根系鲜质量较健康土壤分别减少50.2%及33.1%,苗期番茄叶片PPO活性较健康土壤降低15.8%,POD活性较健康土壤增加24.0%,差异均达显著水平(P0.05)。3)番茄根系更易感染有害菌,根系内病原菌甘蓝假单胞菌数量较健康土壤增加463倍,根区土壤细菌、真菌及放线菌总数分别增加46.3%、94.5%及134.0%。4)食细菌线虫、食真菌线虫及植物寄生性线虫数量分别为健康根区土壤的3.3倍、1.6倍及7.3倍,其中的植物寄生线虫95.6%为根结线虫。综上所述,接入连作番茄根结线虫病株根区病土不仅导致番茄遭受根结线虫侵染,而且会导致土壤线虫总量及植物寄生线虫所占比例大幅增加,并使番茄根系内有害细菌数量显著增加,对番茄生长造成显著抑制作用,同时影响番茄的生理生化特性,受线虫侵染番茄防御性酶活性降低,使其更易被根结线虫及病原菌侵染,番茄根区土壤线虫、微生物及根系内优势细菌的种类与数量及其之间的作用发生改变。  相似文献

A 60-day glasshouse experiment was conducted to assess the influence of two strains of Pseudomonas fluorescens (GRP3 and PRS9), organic manure, and inorganic fertilizers (urea, diammonium phosphate (DAP), muriate of potash and monocalcium phosphate) alone and in combination on the multiplication of Meloidogyne incognita and growth of tomato. Pseudomonas fluorescens GRP3 was better at improving tomato growth and reducing galling and nematode multiplication than PRS9. Organic manuring resulted in less galling and nematode multiplication than occurred with DAP. However, DAP was found better in reducing nematode multiplication and improving plant growth than urea. Muriate of potash was the inorganic fertilizer least effective in reducing galling and nematode multiplication. Pseudomonas fluorescens GRP3 with organic manure was the best combination for the management of M. incognita on tomato but improved management of M. incognita can also be obtained if DAP is used with the GRP3 strain of P. fluorescens.  相似文献

Information about the interaction between arbuscular mycorrhizal fungi (AMF) and the false root-knot nematode Nacobbus aberrans (Thorne, 1935) Thorne & Allen, 1944 is scarce. The effect of Glomus intraradices Schenk & Smith on tomato (Lycopersicon esculentum L.) cv. Platense inoculated with nematode juveniles from Lisandro Olmos (Argentina) was studied under greenhouse conditions. Six treatments with five replications were performed. After 80 days, nematode reproduction and percentage of AMF colonization in roots were estimated. Some plant growth parameters were also measured. In general, plants with AMF and AMF plus nematodes grew as well as the control without AMF and without nematodes. Furthermore, G. intraradices was beneficial in reducing nematode-induced damage in roots (lower number of galls) as well as in having a suppressive effect on parasite reproduction. This is the first study on the use of G. intraradices as a possible strategy in the control of N. aberrans in tomato.  相似文献

Abstract

A greenhouse experiment was carried out to investigate the influence of Glomus clarum (mycorrhiza) on the growth of tomato seedlings grown in both sterilized and non-sterilized soils. Highest growth parameter values were recorded in tomato plants inoculated with mycorrhiza but grown in sterilized soil, followed by those grown in non-sterilized soil but inoculated with mycorrhiza also. Sterilized but non-inoculated tomato plants also had growth and were closely followed by non-sterilized, non-inoculated tomato plants. There was no significant difference in all the treatments when girth of the tomato plants used was measured in this study. Nutrient uptake (N,P,K) was significantly found highest in the inoculated sterilized tomato plants while it was found lowest in the non-sterilized, non-inoculated tomato plants. Generally, mycorrhizal-inoculated tomato plants (whether sterilized or non-sterilized) showed better growth in all the treatments used.  相似文献

土壤线虫和微生物群落结构与作物健康密切相关。采用田间试验,利用传统分类和高通量测序技术,研究了水稻–番茄轮作(FS)、番茄连作(FF)和休耕(CK)3种种植制度土壤线虫及微生物群落结构差异。结果表明,水旱轮作土壤中线虫种类少,自由生活线虫占比高。土壤中植物寄生线虫属的检出率,轮作最少,仅有5个,连作最多,共13个,且大部分为常见属;水旱轮作同时能降低土壤中植物寄生线虫的种类,包括主要危害番茄的根结线虫属,占比仅2.1%,而连作为29.7%。不同种植制度下真菌和细菌类群OUTs和群落结构差异明显;与连作相比,轮作和休耕真菌、细菌种群丰富度更高,共享占比更多,群落构成更相近;水旱轮作Alpha多样性指数显著高于连作;轮作土壤中植物寄生线虫拮抗微生物类群Chaetomium,Talaromyces,Anaerolineaceae和Acidobacteria相对丰度高于连作。水稻–番茄轮作能大幅减少植物寄生线虫种类,增加微生物群落多样性,改善土壤质量,是番茄生产中防控线虫病害的理想措施。  相似文献

Glasshouse experiments were conducted to assess the influence of arbuscular mycorrhizal (AM) fungi (Glomus mosseae and Gigaspora margarita) and organic fertilizers (cow dung, horse dung, goat dung and poultry manure) alone and in combination on the reproduction of the nematode Meloidogyne incognita and on growth and water loss of tomato. Meloidogyne incognita decreased water loss of tomato from the first week onwards after inoculation. AM fungi increased water loss of both nematode-infected and uninfected plants. Glomus mosseae was better at improving growth of tomato and reducing galling and nematode multiplication than G. margarita, but the rate of water loss was similar with both fungi. Addition of organic fertilizers improved growth of tomato, but decreased the rate of water loss. There was less galling and nematode multiplication with poultry manure, which improved tomato growth more than goat dung, while cow dung was the least effective organic fertilizer. Greatest plant growth and least nematode reproduction were observed in plants inoculated with M. incognita along with G. mosseae and poultry manure.  相似文献

The potential effect of rhizobial inoculation on root knot nematodes in chickpea, mungbean and pigeonpea were studied under field condition. The seed treatment with respective rhizobium strains increased the nodulation, leghemoglobin content, bacteriod population, plant growth, yield and nitrogen uptake of three three food legumes compared to the plants without the rhizobium treatment. The nematode (1500?juveniles/kg soil) incited oval galls on the roots of the three legumes, and suppressed plant growth and yield. The galling, egg mass production and soil population of the nematode was greater on the plants without the rhizobium treatment. The pure culture and culture filtrate of the rhizobium strains suppressed the egg hatching and induced mortality to the juveniles of M. incognita over control. The nematode infection reduced the nodulation, bacteroid population and leghemoglobin contents of the nodules and NPK uptake by the plants. Hence, the rhizobia treatment shall be integrated to common agronomic practice of food legume cultivation so as to enhance crop productivity and to protect roots from nematode attack.  相似文献

Disease complex of carrot (Daucus carota L.) involving root knot nematode Meloidogyne incognita and two fungi Alternaria dauci and Rhizoctonia solani were studied in three soil types. More plant growth, chlorophyll, carotenoid and proline contents were found in carrot grown in fly ash mix soil than plants grown in sand mix soil and loamy soil. Inoculation of M. incognita, R. solani, and A. dauci reduced plant growth, chlorophyll and carotenoid but increased proline contents. Inoculation of M. incognita 20 days prior to a fungal pathogen caused a greater reduction in plant growth, chlorophyll and carotenoid than fungal pathogen was inoculated prior. Inoculation of A. dauci prior to R. solani or vice versa had a similar effect on plant growth, chlorophyll, and carotenoid. Nematode multiplication and galling was higher in plants grown in sand mix soil followed by loamy soil and fly ash mix soil. Both fungi had adverse effects on galling and nematode multiplication. Blight disease index caused by A. dauci was 3 and crown rot index by R. solani was also recorded 3. These disease indices were 5 when pathogens were inoculated in combinations.  相似文献

Several strains of Bradyrhizobium, Mesorhizobium and Sinorhizobium strains were tested for their compatibility with an Australian acacia: Acacia holosericea. All bacterial strains induced some nodule formation on roots. However, the rhizobia which greatly increased the development of seedlings belonged to the Bradyrhizobium genus. The root-knot nematode Meloidogyne javanica, strongly inhibited the symbiosis with most of the rhizobial strains. The rhizobia had no effect on nematode multiplication except for bacterial strain ORS 1020 where the final nematode population was higher than in the control treatment. The hypothesis concerning the interactions between nematodes and the nitrogen fixative process are discussed. Moreover, this investigation underlines the decreasing effect of nematodes on the potential benefits that may result from growing A. holosericea inoculated with selected rhizobial strains.  相似文献

Summary We selected two isolates of Rhizobium for cowpea (Vigna unguiculata) with sterilized soil tests and two different isolates by non-sterilized soil testing. The four rhizobia were then paired individually with either Glomus pallidum, Glomus aggregatum, or Sclerocystis microcarpa in separate, sterilized, or non-sterilized soil experiments. The purpose of the experiments was to determine the effect of soil sterilization on the selection of effective cowpea rhizobia, and to see whether these rhizobia differed in their effects on cowpea growth when paired with various vesicular-arbuscular mycorrhizal (VAM) fungi. Our experiments showed that the rhizobia selected in sterilized soil tests produced few growth responses in the cowpea compared to the other introduced rhizobia, irrespective of pairing with VAM fungi in sterilized or non-sterilized soil. In contrast, the two rhizobia initially selected by non-sterilized soil testing significantly improved cowpea growth in non-sterilized soil, especially when paired with G. pallidum. Our results suggest that it is important to select for effective rhizobia in non-sterilized soil, and that pairing these rhizobia with specific, coselected VAM fungi can significantly improve the legume growth response.  相似文献

Nutrient-poor, sandy soils form the prevailing substrate at post-mining sites of the Lusatian region(Brandenburg, Germany) and present a challenge for vegetation development. We studied the organic acid quantity and composition of three commonly occurring pioneer plant species, the legumes Lotus corniculatus L. and Trifolium arvense L. and the grass Calamagrostis epigeios(L.) Roth, to determine if plant growth and exudation differed with(non-sterilized soil) and without(sterilized soil) an indigenous soil microbial community. We investigated whether organic acids were found in the rhizosphere and surrounding soil and whether this influenced nutrient mobilization. This study consists of linked field investigations and a greenhouse experiment. Plants were grown in the greenhouse in either sterilized or non-sterilized sandy soil from a reclamation site in the Lusatian mining landscape(Welzow Su¨d, East Germany). After seven months, the plant biomass, root morphology, organic acids, and water-soluble nutrients and root colonization with arbuscular mycorrhizal fungi(AMF) and dark septate endophytes(DSE) were analyzed. Roots of all three plants in the field and greenhouse experiments were highly colonized with AMF. Calamagrostis epigeios and T. arvense had a significantly higher colonization frequency with DSE than L. corniculatus. The quantity and composition of organic acids strongly differed among plant species, with the highest number of organic acids found for L. corniculatus and lowest for C. epigeios. The quantity of organic acids was greatly reduced in all plants under sterilized soil conditions. However, the composition of organic acids and plant growth in sterilized soil were reduced for both legumes, but not for C. epigeios, which had a higher biomass under sterilized conditions. Changes in nutrient concentrations in the field rhizosphere soil relative to those in the control were measurable after seven months. While the spectrum of organic acids and the growth of legumes seemed to be dependent on a highly diverse soil microbial community and a symbiotic partner, the grass C. epigeios appeared capable of mobilizing enough nutrients without an indigenous microbial community, and might be more competitive on sites where soil microbial diversity and activity are low.  相似文献

The available information on Pseudomonas biocontrol inoculants on the non-target fungal and nematode community is scant. The current paper addresses this issue and investigates the effects of biocontrol agents Pseudomonas aeruginosa IE-6 and IE-6S+ (previously shown to suppress several soil-borne plant pathogens) on soil microfungi and plant-parasitic nematodes as well as on the root-knot development and growth of tomato (Lycopersicon esculentum). Furadan, a granular nematicide was included as a treatment for comparative purposes. Treatments were applied to soil at the start of each 52-day-long tomato growth cycle, and their effects on the composition and diversity of rhizosphere and endophytic microfungi and plant-parasitic nematodes were examined at the end of first and fourth growth cycle. Several diversity indices were employed to assess community diversity. A total of 16 genera comprising 23 microfungal species were isolated from the tomato rhizosphere. The most abundant fungal species belonged to the genera Aspergillus, Fusarium, and Penicillium. With a few exceptions, fungi were neither exclusively inhibited nor specifically promoted by the application of treatments at any of the growth cycles studied. However, Paecilomyces lilacinus, an egg and female parasite of root-knot nematode, though exclusively absent in the controls was isolated from the treatments. Both general diversity and equitability of rhizosphere microfungi were greater at first compared to the fourth growth cycle while species richness remained uninfluenced across the growth cycles and treatments. However, Furadan and IE-6S+ treatments considerably abated general diversity and equitability. Of the microfungal species isolated from the rhizosphere seven were also recovered from surface-sterilized root tissue of tomato suggesting that all the endophytes are primarily rhizosphere organisms. Diversity of endophytic fungi was consistently lower compared with that of the rhizosphere. Both general diversity and equitability declined in all three treatments relative to controls in the root tissue but species richness remained unaltered. Diversity and equitability of plant-parasitic nematodes in soil were reduced by all three treatments over the controls at fourth growth cycle whilst species richness did not change at either growth cycle. The biocontrol agents significantly reduced root-knot development and enhanced shoot growth of tomato over the controls. The possible implications of fungal composition and abundance because of biocontrol by Pseudomonas application are discussed.  相似文献

Impact of soil fumigation practices on soil nematodes and microbial biomass   总被引:4,自引:2,他引:4  
This study was designed to understand the impact of methyl bromide (MB) (CHaBr) and its alternatives on both free-living and root-knot nematodes in the soil. A randomized complete block experiment with six treatments and 4 replicates (each replicate in a separate greenhouse) was established in Qingzhou, Shandong Province, China. In addition to MB and untreated control (CK) treatments there were four alternative soil fumigation practices including MB virtually impermeable films (VIF), metam sodium (MS), MS VIF and soil solarization combined with selected biological control agents (SS BCA). Two tomato (Lycopersicum esculentum Mill.) cultivars, cv. Maofen-802 from the Xian Institute of Vegetable Science, China, and cv. AF179 Brillante from the Israeli Hazera Quality Seeds, were selected as test crops. The results indicated that Rhabditidae was the most dominant population with percentage abundance as high as 85% of the total number of identified free-living nematodes, followed by that of Cephalobidae. Methyl bromide and its alternatives except for the non-chemical SS BCA treatment controlled the target pest, root-knot nematodes. Also, the impact of the three chemical alternatives on free-living nematode number and functional group abundance was similar to the impact associated with a typical methyl bromide application. Chemical fumigation practices, especially that with MB, significantly reduced the number of nematodes in the soil and simultaneously significantly reduced the number of nematode genera thereby reducing nematode diversity. All the four soil chemical fumigation activities decreased soil microbial biomass and had an obvious initial impact on microorganism biomass. Furthermore, both plant-parasitic and fungivore nematodes were positively correlated with soil microbial biomass.  相似文献

A reduction of arbuscules in roots of grapevines (Vitis vinifera) observed when ring nematodes were added to field microplots led to the hypothesis that nematode feeding suppresses arbuscules by competing for root carbohydrates. Support for this hypothesis was tested by growing ‘Pinot noir’ grapevines in a factorial experiment with three levels of initial nematode densities (0, 0.1, 1.0 nematodes g?1 soil), two levels of light (full sun, 50% sun), and two levels of AMF (nonAMF, +AMF). Effects on plant growth were primarily driven by a light and AMF treatment interaction, such that low light increased stem dry matter accumulation at the expense of roots in +AMF vines only. Nematodes had only a minor influence on plant growth (leaf mass was reduced at the highest nematode density), but nematodes did not affect overall plant dry matter accumulation. Since nonAMF vines were severely limited by P and their growth was so poor, the impact of nematode and light treatments was further analyzed in +AMF plants only. Nematode populations, AMF colonization, and root carbohydrates were differentially affected by initial nematode density or light levels. Root biomass, and reducing sugar and starch concentrations in fine roots were reduced by low light, but the final nematode populations and arbuscule frequencies in roots were unaffected by light. Nematodes reduced arbuscules and starch concentrations in fine roots, but did not affect total colonization by AMF (hyphae, vesicles or arbuscules). Nematodes reduced plant P and K uptake at the highest density, and low light reduced Mg uptake. These findings are consistent with the hypothesis that ring nematodes suppress arbuscules in roots via competition for root carbohydrates. However, the lack of a treatment interaction between light and nematodes in our study suggests that ring nematode–AMF interactions in grape roots are controlled by more than competition for photosynthate.  相似文献

To determine the effect of agricultural management on the dynamics and functional diversity of soil nematode communities in a carrot field at Kibbutz Ramat Hakovesh, Israel, soil samples from 0--10 cm and 10--20 cm depths were collected during the growing season of carrot. Indices were used to compare and assess the response of soil free-living nematode communities to agricultural management. Eighteen nematode families and 20 genera were observed during the growing period, with Cephalobus, Rhabditidae, Aphelenchus, Tylenchus, and Dorylaimus being the dominant genera/families. During the planting, mid-season and post-harvest periods the total number of nematodes at both depths was significantly lower (P < 0.01) in the carrot treatment than in the control plots, while during the harvest period at both depths total nematodes and bacterivores were significantly higher in the treatment plots (P < 0.01). The values of the maturity index (MI) at both depths were found to be significantly lower in the treatment plots than in the control plots during the pre-planting period (P < 0.05). Overall, WI, MI and PPI were found to be more sensitive indicators than other ecological indices for assessing the response of nematode communities to agricultural management in a Mediterranean agroecosystem.  相似文献

To study the influence of fallow on plant parasitic nematodes and soil fertility, the difference in the growth of millet in a greenhouse was compared in sterilized and naturally infested soil. These soils are ferruginous and were taken from a 17-year-old fallow plot and a neighbouring cultivated field, located in the region of Thysse Kaymor in Senegal. The plant-parasitic nematode community of the previously cultivated soil consisted primarily of two species: an ectoparasite, Tylenchorhynchus gladiolatus, and an endoparasite, Scutellonema cavenessi. The soil from the fallow plot was infested with many species, but under the influence of the millet crop, it evolved towards the two species situation observed in the cultivated soil, except that under the experimental conditions (in pots), Pratylenchus pseudopratensis replaced S. cavenessi. Whether or not the soil was infested with nematodes, the previous fallow period had a positive effect on the development of successive millet crops. The impoverishment of the soil through repeated cultivation and the impact of nematodes both reduced crop growth. The two factors evolved in the same way, that is a decrease in the development rate of the millet as the number of crop cycles increased. The symptoms of soil impoverishment can be corrected, either by suppressing the nematodes or by fallowing.  相似文献

Mycorrhizae are fungal symbionts forming mutualistic relationships with plant roots. This study was undertaken to evaluate the overall influence of arbuscular mycorrhizal fungi (AMF) on the overall growth and development of carrot plant. Surface-sterilized seeds of carrot were sown in earthen pots filled with sterile soil. Half of the pots were inoculated with AMF spores; the other half without any AMF inoculation represented control. After germination inoculated plants, along with the controls, were sampled at 20, 40, 60 and 80 days of growth after seedling emergence. There is a progressive net increase in growth with each 20-day interval after seedling emergence. Storage roots of carrot had a higher level of metabolites and nutrients like nitrogen, phosphorus and potassium in mycorrhiza colonized plants than the control. High performance liquid chromatography analysis indicated an increase in the carbohydrate fractions in AMF-inoculated roots. Thus, mycorrhiza has been found increasing the growth, metabolites and nutrition of carrot plant.  相似文献
