In vitro laboratory decay tests on Ailanthus excelsa Roxb. wood revealed that I. hispidus exhibits a combination of both white-rot and soft-rot patterns of wood decay. Early stages of wood decay showed dissolution of the middle lamella as well as defibration and localized delignification of fiber walls; vessels, axial, and ray parenchyma remained unaltered. Delignification commenced from the middle lamellae at the cell corners without any marked effect on the primary and secondary wall layers. In later stages of growth, the species produces typical soft-rot decay pattern by forming erosion channels through the S2 layers of fiber walls, transverse bore holes in the cell walls, and erosion channels alongside/following the orientation of cellulose microfibrils. The rays showed signs of cell wall alterations only after the extensive damage to the fiber walls. After 120 days of incubation, the vessels also showed localized delignification, the erosion of pits, and separation from associated xylem elements. The extensive weight losses under natural and in vitro decayed wood as well as the very soft nature of severely degraded wood indicate that I. hispidus alters wood strength and stiffness.  相似文献

The structural changes in the cell wall and delignification pattern caused by Trametes versicolor and Trametes hirsuta in the sap wood of Leucaena leucocephala were examined by light and confocal laser scanning microscopy. The in vitro decay test was conducted for 12 weeks. Both species of Trametes used in this study caused anatomical characteristics specific to simultaneous white rot. Formation of boreholes, erosion troughs, erosion channels with U-shaped notches in tangential sections and thinning of cell walls were evident in the wood inoculated with each of the fungal species. Cell separation due to removal of middle lamellae occurred at the early stages of infection (30 d) with T. versicolor. In contrast, middle lamellae remained intact in wood inoculated with T. hirsuta and showed cell separation due to degradation of the outer layer of the secondary wall and degradation of the middle lamellae observed only in severely decayed wood at late stages. Confocal microscopy revealed the delignification pattern particularly from cell corners and vessel walls at an advanced stage of decay, indicating strong ligninolytic activity of both species in the sapwood of L. leucocephala.  相似文献

Chemical, light and electron microscopic studies were carried out on wood of Oriental beech (Fagus orientalis Lipsky) decayed by the white‐rot fungi Pleurotus ostreatus and Trametes versicolor for 30, 60 and 120 days according to the modified European standard EN 113. Mass loss as well as lignin, cellulose and carbohydrate content were determined before and after fungal attack. There were no significant differences of wood mass loss and chemical composition between both fungi at the end of incubation. After each incubation period, small specimens were stained for microscopic studies. The micromorphology of fungal cell wall degradation was rather similar for both fungi. Both decreased the cell wall thickness to the same extent. The accumulation of hyphae as well as the rupture of cell walls was also similar. The occurrence of hyphae, cavities in the pits and vessel walls followed nearly the same patterns. The parenchyma cells were completely destroyed. Altogether, both fungi produced a simultaneous white rot in Oriental beech wood.  相似文献

Fresh sapwood blocks of Norway spruce (Picea abies) were exposed to 12- and 60-day attack by Stereum sanguinolentum in pure culture and were examined with respect to structural, topochemical and colour alterations. Final stages of degradation by 5. sanguinolentum were studied on naturally decayed spruce wood after 1 year of fungal growth. The development of discoloration in the main anatomical directions of wood was assessed. Furthermore, microspectrophotometrical studies on the decomposition of cell wall polyphenols were performed. Initial signs of sap streak appeared already 8 days after fungal infection. Within the three-dimensional decay pattern, mycelial aggregations associ-ated with the ray tissue proved to be the major cause of discoloration. UV extinction measurements taken at 280 nm of different parts of cell wall transverse sections revealed a specific pattern of lignin distribution and its derivatives during fungal degradation.  相似文献

The kinetics of xylem formation in four-year-old plants of Swietenia mahagoni (L.) Jacq. and Khaya ivorensis A. Chev. were studied by means of high resolution laser measurements (accuracy: ±2 μm) in a spatial resolution of 18.7 to 94.1 μm and a temporal resolution of 1 to 60 s. The radial enlargement of the xylem cells was completed within 0.36 to 8.85 hours in Swietenia and within 0.52 to 12.03 hours in Khaya, while secondary wall formation and lignification lasted days to weeks. Cell enlargement of vessels and ray parenchyma was significantly faster than radial enlargement of fibres and axial parenchyma. The processes of formation of the secondary cell wall and the lignification were slower in fibres than in vessels and parenchyma cells. In Swietenia new secondary cell formation was induced in distinct growth periods almost simultaneously in the whole shoot, while in Khaya new cell formation was restricted to parts of the shoot. Growth stresses in the shoot were studied in terms of released strain. Higher growth stresses were observed in the shoot of Khaya compared to Swietenia. The results indicate that the higher growth stresses in the xylem of Khaya compared to Swietenia originate in the different kinetics of cell development of different cell types in combination with the heterogenous sequence of cell formation within the shoot. ABSTRACT

Structural alterations in the wood cell walls of neem inoculated with by Trametes hirsuta and T. versicolor were studied by microscopic methods. In vitro decayed wood showed extensive weight loss of test blocks (26.7 and 41.38% by T. versicolor and T. hirsuta, respectively) at the end of 3 months. Selective delignification in the initial phase followed by simultaneous removal of lignin was evident in test blocks inoculated with both the species. The separation of middle lamellae and patches of cellulosic polysaccharides stained blue with Astra blue in the delignified region of the fiber wall during early stages indicates selective mode of decay. In contrast, the occurrence of erosion troughs with characteristic U-notch in tangential sections is a characteristic feature of simultaneous rot that was apparent after 3 months of incubation. The decay pattern occurred concomitantly in all the xylem elements irrespective of general resistance pattern shown by vessel and axial parenchyma cells. At an advance stage, both species of Trametes showed formation of erosion channels along the microfibrils angle of cellulose which is considered as characteristics of soft rot decay type. The sharing of white rot and soft rot decay pattern by both the fungi suggest a phylogenetic link between both groups of fungi.  相似文献

Soil affects the anatomy of downy birch wood (Betula pubescens), which raises the question whether the growth of this species differs between mineral and peat soils. The aim of this study was to compare growth, density and structure of xylem of B. pubescens between trees grown in different soils. Both growth and density differed between trees grown in different soils. All measured anatomical characteristics, except double thickness of fibre walls, percentage vessel area and ray number, showed differences between the soil types: the cell dimensions were larger and numbers smaller in trees grown in mineral soil. In peat, high growth rates decreased the wall:lumen ratio of fibres in maturing wood, while no such correlation was observed in trees grown in mineral soil at any studied cambial age. In addition, axial parenchyma may have a different role in trees grown in different soils, as the rapid growth decreased and increased axial parenchyma in mineral and peat soil, respectively. The characteristics affecting wood strength were more similar between trees grown in different soils than those affecting water conductance. The observed differences between trees grown in different soils emphasise rapid growth particularly at young ages and shorter reasonable rotation period in mineral soil.  相似文献

Focussing on four types of parenchyma cell around pith regions of branches of Robinia pseudoacacia L. var. inermis, we examined the timing and role of cell death during heartwood formation. Large parenchyma cells that were located in the inner part of the pith died within a year. By contrast, other parenchyma cells died within 4 years, with the timing of cell death depending on the type of cell. Axial parenchyma cells of the xylem close to the pith died first. Then, small parenchyma cells died in the perimedullary zone in the outer part of the pith. Finally, ray parenchyma cells in the xylem close to the pith died. Variations in the autofluorescence of cell walls, which might have been due to deposition of heartwood substances, were observed first in xylem ray parenchyma cells and small parenchyma cells in the perimedullary zone. Our results indicate that the initiation of heartwood formation occurs within 4 years in pith regions of branches in Robinia pseudoacacia L. var. inermis. Moreover, it appears that not only xylem ray parenchyma cells but also small parenchyma cells in the perimedullary zone might be involved in the synthesis of heartwood substances.  相似文献

Suberization responses to naturally-occurring fungal colonization of living xylem tissues were detected in 30 of 37 woody species examined. These responses comprised the suberization of traumatic parenchyma tissues in compartmentalization wall 4 barrier zones laid down in the vicinity of fungally infected wounds, or the suberization of parenchyma cells, tyloses and vessel linings in reaction zone decay margins in fully differentiated sapwood. Additionally, in a few species, suberization of xylem fibres and tracheids was observed where juvenile xylem tissues, apparently still incompletely differentiated, had been exposed by wounding. These xylem suberization responses are considered to increase the decay resistance of the altered cells, and may protect the hydraulic integrity of adjacent tissue, thus contributing to the antimicrobial defence of the living sapwood.  相似文献

To further our understanding of wood decay in living light red meranti (Shorea smithiana) trees, microscopic characteristics of the cell and cell wall degradations of S. smithiana wood in the presence of the decay fungi, the identity of the causal fungi, and the decay potential and pattern by an isolated fungus were investigated. Cell wall degradations, including cell wall thinning, bore holes formation, rounded pit erosion, and eroded channel opening were clearly observed under light and scanning electron microscopy. In transverse view, many large voids resulting from a coalition of degraded wood tissue appeared in the decayed canker zone. All these observations suggest the well-known simultaneous decay pattern caused by white-rot fungi. By phylogenetic analysis based on the sequences of internal transcribed spacer region of ribosomal DNA, a basidiomycete fungus isolated from the decayed wood was identified as Schizophyllum commune. The degradation caused by this fungus on sound S. smithiana wood in an in situ laboratory decay test was classified as the early stage of simultaneous decay, and showed a similar pattern to that observed in the wood samples naturally decayed.  相似文献

Microscopic examination showed the cell wall decay pattern produced by the brown-rot fungus Coniophora puteana to be different from the degradation pattern known to be typical for brown-rot fungi. Erosion and thinning of cell walls in patterns considered to be characteristic of white-rot decay were observed. In particular, the fungal strain COP 20242 degraded secondary cell wall layers extensively, and also degraded lignin-rich middle lamellae. Some strains of C. puteana produced soft-rot type cavities in the S2 layer. All strains of C. puteana employed in the present work showed a positive reaction to tannic acid in the Bavendamm test, indicating the production of laccase. Microscopic and enzymatic studies provided evidence to suggest that the wood decay by C. puteana is unique both in terms of micromorphological and enzymatic patterns of cell wall degradation. This is because brown-rot fungi are not generally known to form cavities in the cell walls or to produce lignin-degrading enzymes. These observations suggest that lignin degradation capacity of brown-rot fungi may be greater than previously considered.  相似文献

The formation of reaction and barrier zones was studied in the xylem of Eucalyptus globulus and Eucalyptus nitens tree stems after wounding and artificial inoculation with two white rot fungi. The study had two objectives: to describe host responses in Eucalyptus spp. by light microscopy and to determine whether they would differ in a fungal treatment (wounding and inoculation by one of two fungal isolates) when compared to a control treatment (wounding only). Eucalyptus globulus and E. nitens developed similar reaction and barrier zones. The E. globulus barrier zone was characterized by kino vein formation. In both hosts, the reaction zone was primarily influenced by content and distribution of living tracheids and parenchyma cells within the sapwood. By contrast, the anatomy of the barrier zone showed similarities to the basic xylem structure of each host, except for some cell types that were newly formed (sclereids, kino veins) or increased in number (parenchyma cells, tracheids). Other cell types were reduced in number or completely absent. Host response in terms of barrier zone width appeared to be greater in the fungal than control treatment. Both wood decay fungi appeared to induce a wider barrier zone in both species than that associated with non‐specific damage caused exclusively by wounding. However, the small number of replicates available for this study was possibly insufficient to provide statistical evidence for different barrier zone width between fungal and control treatments.  相似文献

[目的]通过测定和分析广西融水种源红心杉木优树材质性状指标,以了解该地区红心杉木材性状变异规律。[方法]以广西融水种源60株20年生的红心杉木优树为研究材料,测定单株材积、红心率、基本密度、组织比量、管胞性状、微纤丝角等10个材质性状指标,分析各材性性状指标分布和变异规律及性状间的相关关系。[结果]表明:红心率、基本密度、木射线比量、管胞比量、管胞长、管胞宽、管胞长宽比等性状数据分布服从正态分布。融水两个地区红心杉优树轴向薄壁细胞比量的变异系数分别为35.08%和44.97%,变异较大。管胞比量变异系数分别为3.28%和3.56%,变异较小。红心率、木射线比量、管胞长、管胞宽、微纤丝角等性状差异极显著(P0.01),轴向薄壁细胞比量差异显著(P0.05)。10个材质性状间存在12对表型显著相关。[结论]测定的10个性状均是连续性数量性状。早、晚材的管胞长度、管胞宽度和管胞长宽比等性状数据呈从心材至边材逐渐增加的规律。作为重要经济性状的红心率与木射线比量呈极显著正相关(P0.01),单株材积与轴向薄壁细胞比量呈显著正相关(P0.05),而单株材积与基本密度的相关性不显著,这使红心杉木生长量与材性相结合的遗传改良成为可能。  相似文献

Samples of pine (Pinus sylvestris) and spruce (Picea abies) were impregnated with a low-viscous epoxy resin using a vacuum process. The epoxy was cured in situ and the specimens sectioned. Deposits of the cured epoxy was then observed in the wood cavities using a scanning electron microscope. The investigation concentrated on tracing the transverse movements of a viscous liquid in the wood, and special attention was therefore given to the cross-field area between ray cells and longitudinal tracheids. A damage hypothesis is proposed based on the results obtained in the present investigation in combination with those from earlier studies on linseed oil-impregnated pine: In addition to the morphology of the bordered pits, viscous liquid flow in wood is dependent on damage that occurs during the impregnation procedure. For pine sapwood, liquid flow is enabled through disrupted window pit membranes, which divide the longitudinal tracheids and the ray parenchyma cells. A mechanism accounting for the reduced permeability of pine heartwood is believed to be deposits of higher-molecular-weight substances (extractives) in the ray parenchyma cells and on the cell walls. In spruce the thicker ray cells in combination with the smaller pits, which are connected to the longitudinal tracheids, reduce permeability considerably.  相似文献

Acacia hybrid (Acacia mangium × Acacia auriculiformis) clones are widely planted in Vietnam with a total of approximately 400,000 ha to meet the demand for pulpwood, sawn timber and wood chip exports. Silvicultural techniques such as pruning and thinning have been applied to improve productivity and sawlog quality of Acacia hybrid plantations. However, those techniques may also create opportunities for wood decay fungi to enter the Acacia hybrid stems through wounds and cause stem defects that reduce sawlog quality and the value of the plantation. The presence of fungal decay agents in Acacia hybrid trees was examined in two Vietnamese plantations. In July 2011, just prior to a second thinning, discoloured wood samples were taken from a three‐year‐old Acacia hybrid plantation at Phan Truong Hai for the isolation of fungi. In July 2012, approximately 18 months after pruning and thinning treatments, discoloured wood samples were taken from a three‐year‐old Acacia hybrid plantation at Nghia Trung for the isolation of fungi. DNA sequencing of the rDNA ITS identified the isolates. In May 2015, approximately 4 years after thinning and fertilizer treatments, discoloured and decayed wood samples were taken from the above (7‐year‐old) Acacia hybrid plantation at Phan Truong Hai for fungal identification. DNA was extracted directly from discoloured and decayed wood samples and fungal rDNA ITS amplicons sequenced on a Roche 454 sequencer. The results showed that silvicultural treatments did not affect the fungal communities associated with discoloured and decayed wood of Acacia hybrid plantation at Phan Truong Hai. A total of 135 fungal species or OTUs (operational taxonomic units) were identified, including 82 members of Ascomycota and 52 Basidiomycota.  相似文献

The effect of water-stress conditioning on water relations and histological features ofPinus thunbergii Parl. inoculated with avirulent isolate ofBursaphelenchus xylophilus (Steiner and Buhrer) Nickle, pine wood nematode, were investigated. Pines were kept under 8 days cycle of severe water stress. One-half of the water-stressed pines died as a result of infection by avirulent pine wood nematode and water stress tended to induce increased susceptibility and/or decreased resistance of pines to avirulent pine wood nematode. In dead pines, the water conducting function of xylem was lost, and all of the parenchyma cells died. In surviving pines, the xylem hydraulic conductivity and the xylem water content were significantly reduced (12 to 23% and 77 to 83%, respectively) compared to controls. Safranin dye perfusion of excised axis stem segments indicated that the water conductance was limited to the very narrow peripheral area of xylem. Embolism caused by cavitation in the tracheids occurred in the central part of xylem and in that dysfunctional region of the xylem the axial parenchyma cells surrounding the epithelial cells, and ray parenchyma cells partly degenerated but the epithelial cells survived. The disruption of tracheid shape observed in surviving pines indicates that avirulent pine wood nematode temporarily disturbed cell division of the cambium. Considering the differences in responses between dead pines and surviving pines after inoculation with avirulent pine wood nematode, the death of water-stressed pines apparently resulted from death of cells, in particular the vascular cambium and the loss of xylem hydraulic function by cavitation.  相似文献

运用木材解剖图像分析系统和显微照相的方法对人工兴安落叶松次生木质部的解剖结构进行研究,结果表明:落叶松具正常树脂道和受伤树脂道两种类型,前者常见于晚材。落叶松生长轮内的早晚材在干和枝内急变,在根内缓变。早材管胞呈六边形至多边形,胞壁常见单列具缘纹孔,偶见对列具缘纹孔;晚材管胞多呈矩形,胞壁鲜见具缘纹孔,通常为单列具缘纹孔。落叶松木射线同时具有单列木射线和纺锤形木射线两种类型,纺锤形木射线中仅含一枚纵行树脂道。纵行管胞与木射线交叉形成的纹孔场为云杉型。从根到干再到枝,管胞逐渐细化,管胞长度逐渐减小,木射线分布由密到疏。  相似文献

Changes in the morphology and functions of vacuoles provide useful information about the mechanism of cell death. In the present study, we monitored the morphology and contents of vacuoles during the death of ray parenchyma cells in the conifer Cryptomeria japonica. In differentiating xylem, ray parenchyma cells had large central vacuoles. In sapwood, vacuoles in ray parenchyma cells contained proteins, an indication that one of the main functions of these vacuoles might be protein storage. A dramatic decrease in the protein content of some vacuoles was detected in the intermediate wood before the initiation of vacuole rupture. Although vacuole rupture was detected from the intermediate wood to the outermost heartwood, some vacuoles were obviously enlarged in the inner intermediate wood. Condensed nuclei were first observed after the rupture of these large vacuoles in ray parenchyma cells. It seems plausible that the autolysis of the contents of ray parenchyma cells might be caused by the rupture of the enlarged vacuoles in the inner intermediate wood.  相似文献

Radial variations in wood properties, cell morphologies and cell proportions were investigated for nine 4-year-old Neolamarckia cadamba trees planted in a community forest in East Java, Indonesia. The relationships between stem diameter and wood properties, cell morphologies or cell proportions were determined to ascertain the effects of stem diameter on these properties. In the radial variations, almost all of the wood properties, cell morphol- ogies and cell proportion in N. cadamba were changed at around 4 to 6 cm distance from the pith, except for fibre diameter, and the proportion of fibre and axial parenchyma. For trees with larger stem diameter, the vessel diameter was also found to increase. In addition, the ray parenchyma and cell wall proportions in trees with larger stem diameter were lower than those in trees with smaller stem diameter. However, the wood properties obtained from the larger stem diameter trees did not always show lower values. The wood properties in N. cadamba were correlated to fibre wall thickness and vessel diameter.  相似文献
