The ultrastructure of the primary and secondary lamellae of gills was investigated in a marine teleost, the white croaker. The following cells were identified and briefly described: pavement cells, mucous cells, mitochondria-rich cells and rodlet cells. These cell types are present throughout the length of the lamellae. They are studied by means of a series of carbohydrate histochemical methods, including lectin procedures. Neutral sugars and substituted sialic acid were detected by means of periodic acid-borohydride reduction-saponification-periodic acid Schiff reaction (PA/Bh/KOH/PAS), saponification-selective periodic acid Schiff reaction (KOH/PA*S) and saponification-selective periodic acid-borohydride reduction-periodic acid Schiff reaction (KOH/PA*/Bh/PAS) histochemical techniques. A battery of seven lectins was used to study binding on tissue sections at the light microscopic level to characterize glycoconjugates in gills. The reaction to Canavalia ensiformis agglutinin (Con-A), Triticum vulgaris agglutinin (WGA), and Ricinus cummunis agglutinin-1 (RCA-1) was weak in pavement cells; unlike Con-A, the reaction to WGA and RCA-1 was more intense in mucous cells. Arachis hypogaea agglutinin (PNA) lectin showed a strong reaction in mucous cells. Ulex europaens agglutinin-1 (UEA-1) lectin was negative in all cell types. The lectin pattern was similar for both primary and secondary lamellae, except for PNA reaction, which was weak in the pavement cells of the secondary lamella and negative in the pavement cells of the primary lamella.  相似文献

An ultrastructural study was performed on the gills of juvenile Argentinian silverside, Odontesthes bonariensis. The gills are composed of two sets of four holobranchs and, in turn, each holobranch consists of a gill arch and two rows of caudolaterally projecting branchial filaments. From the dorsal and ventral surfaces of each filament, branchial lamellae radiate out as foldings of the epithelial layer. Gill rakers are present on each of the gill arches, on the anteromedial side of the arch opposite to the filaments. Gill rakers, gill arches and branchial filaments are covered by a stratified epithelium, whereas branchial lamellae essentially consist of a thin epithelial envelope containing capillaries. In the stratified epithelium, mucous cells, rodlet cells, granular cells, pavement epithelial cells and mitochondria-rich cells are identified. The thin epithelium that lines the lamellae comprises two cell types, outer and inner epithelial cells, and the capillary walls on the inside of the epithelial envelope are defined by pillar cells. The ultrastructure of all these cell types is described and our findings are discussed in light of the existing data on fish gill morphology. In the gills of juvenile Argentinian silverside is of particular interest the characteristics showed by mitochondria-rich cells, such as their arrangement in clusters of 2-3 cells and their small and depressed surface in contact with the aquatic milieu, features which strongly resemble those of euryhaline species.  相似文献

A series of histochemical techniques have been employed to localize alkaline phosphatase, acid phosphatase, non-specific esterase, catalase and peroxidase; and to visualize and characterize glycoprotein (GPs) moieties in the epithelium of gill arch, gill filaments and secondary lamellae of an angler catfish Chaca chaca. The epithelium of gill arch and gill filament shows strong alkaline phosphatase activity in the deeper layer epithelial cells; strong non-specific esterase activities in the outer layer epithelial cells; and weak acid phosphatase activity throughout the epithelium. The activity of these enzymes in the secondary lamellae is weak. The catalase and peroxidase show strong activities in the blood cells of the secondary lamellae. Various classes of GPs have been identified and characterized in the mucous secretions of the gill epithelium of C. chaca. These include—GPs with oxidizable vicinal diols, GPs with sialic acid residues without O-acyl substitution and GPs with O-sulphate esters. The functional significance of different enzymes in gill epithelium and the GPs in the mucus secreted on the surface has been discussed with the physiology of the gills in relation to the characteristic habit and habitat of the fish.  相似文献

Branchial epithelium of Pseudophoxinus antalyae was lined by both a thick stratified epithelium lining gill arches, gill rakers and primary filaments and a thin epithelium lining the lamellae. Mucous, chloride and rodlet cells, interspersed between pavement cells, were present in the branchial epithelium. With histochemical procedures for the characterization of glycoconjugates, mucous cells showed a strong positive reaction with Periodic acid-Shiff and Alcian Blue at pH 2.5, although with Alcian Blue at pH 0.5 and pH 1.0 the reaction was much weaker. When the combined Alcian Blue (pH 2.5) – Periodic acid-Shiff reaction was performed, most mucous cells were stained purple, whereas by the combined Aldehyde Fuchsin/Alcian Blue (pH 2.5), most cells showed a positive reaction only to Aldehyde Fuchsin. Methylation/ Alcian Blue (pH 2.5) and Methylation/ Saponification/ Alcian Blue (pH 2.5) methods showed the presence of sulphated and carboxylated glycoconjugates in mucous cells. Mucous cells were also detected to stain all metachromatically with Toluidine Blue.  相似文献

The characteristics of the mucous cells located in the gills of the fish Micropogonias furnieri were investigated. Using histochemical procedures that included methods for localization and characterization of glycoproteins (GPs), no differences were detected between the mucous cell contents of the primary and secondary lamellae. The GPs were identified with (a) oxidizable vecinal diols; (b) sialic acids and some of their chain variants, C7 or C9; (c) carboxyl groups and (d) sulphate groups. The electron microscope showed large mucous globules of different electro densities from mucous cells located deep in the epithelium between the other epithelial cells; the release of mucus by exocytosis was observed. GPs secreted on the surface of the mucous cells was suggested to be important for the lubrication, protection and inhibition of microorganisms. It is possibility that GPs could have similar roles in Micropogonias furnieri gills.  相似文献

Abstract

Specific-pathogen-free channel catfish Ictalurus punctatus were exposed to sediment and mud from a pond containing channel catfish with proliferative gill disease. In one experiment, fish were to exposed to mud and sediment for 2 months in water maintained at 19°C. Fish were necropsied weekly, and certain tissues were examined histologically and ultrastructurally. Four trials were conducted with sediment samples from different epizootics of proliferative gill disease. In a second experiment, fish were exposed to sediment for 7 d in water maintained at 16, 19, or 26°C; the fish were then moved to clean water held at 16, 19, or 26°C. Fish were necropsied before transfer to clean water and weekly thereafter for 2 months. Channel catfish held at 19°C developed proliferative gill disease within 2 d of exposure to sediment. Primary cells of a uninucleate myxosporean parasite were present in the gills at the base of lamellae. These developed into plasmodia with numerous secondary cells, and some primary cells disintegrated, releasing their internal secondary cells. Similar development was observed in internal organs 1 week after appearance of the parasite in gills. Complete sporogony did not occur over the 2 months of this study. Plasmodia became necrotic and were not detected after 60 d. In fish exposed to sediment for 7 d at 16, 19, and 21°C, similar organisms were detected, but clinical disease occurred only at 19 and 26°C. Proliferative gill disease may be attributed to extrasporogonic stages of a myxosporean resembling Sphaerospora spp.  相似文献

This study used 22 tench of 25-30 cm in length and weighing roughly 200 g. After sacrifice, the gill arch was extracted and fixed in 5% glutaraldehyde for structural, ultrastructural and morphometrical study. Tench gills consist of primary lamellae which are perpendicular to numerous secondary lamellae in which the respiratory barrier is located. A study was made of the structures which make up that barrier and of the multi-layered epithelium situated in the interlamellar space, which is closely linked to maintaining of internal balance in the fish.  相似文献

Abstract

Unusual short-term associations provide insight into the long-term development or modification of parasitism. A superinfestation of a trap-held crevalle jack Caranx hippos by juvenile cymothoid isopods Cymothoa oestrum suggested loss of the mechanism controlling numbers of isopods on a host, and overcrowding on the host overcame this isopod's strict site specificity. Crabs (Decapoda) have not been reported as parasites of fishes, but unidentified crab zoeae superinfested the gills of a trap-held gray angelfish Pomacanthus arcuatus. Adult burrowing crabs Raninoides lamarcki invaded the gills of trap-held gray snapper Lutjanus griseus; this suggested how a relatively large crustacean could become an opercular chamber parasite and how wound feeding could occur. An apparent prey-to-predator transfer of the cymothoid isopod Anilocra acuta to a free-swimming king mackerel Scomberomorus cavalla also represented a move from an external to a pharyngeal-attachment position in this site-specific isopod. Trap confinement may have protected the infestations of juvenile C. oestrum and crab zoeae from organisms that serve as cleaners. New associations are usually begun by immature stages, but may also be initiated by adult crustaceans, as in the cases of A. acuta and R. lamarcki. The ability of crustaceans to survive, adapt, and be observed makes them excellent subjects for use in discerning complex parasitological processes.  相似文献

Abstract

Proliferative gill disease (PGD) was first reported in channel catfish Ictalurus punctatus at commercial farms in 1981 and is caused by the myxozoan parasite Henneguya ictaluri. The disease affects the gills and is characterized by severe branchial inflammation, epithelial hyperplasia, lamellar fusion, and lysis of chondrocytes. Presumptive diagnosis is based on the presence of lytic areas in the cartilage of the primary lamellae on microscopic examination and is confirmed histologically by the presence of the organism. In these trials, PGD was induced by exposing channel catfish fingerlings to fresh or aged infectious water collected from a pond containing fish diagnosed with severe PGD. The severity of disease was graded by histological scoring and microscopic examination of wet mounts to determine the percentage of gill filaments containing chondrolytic lesions. Exposure of fish to infectious pond water was shown to produce pathological lesions consistent with PGD, and the percentage of gill filaments containing chondrolytic lesions was positively correlated with histological scoring of gill pathology. The number of trophozoite stages in the gills was shown to increase with the severity of the disease. In most cases, however, parasitic cells were not observed in tissue samples with chondrolytic lesions during the early stages of infection. These observations indicate that pathology and lysis of chondrocytes can occur prior to detection of the organism by histopathology. Exposing fish to infectious pond water that was aged for 1 d produced negligible gill pathology and implies that the infectivity of the H. ictaluri actinospore stage is short lived. Removing fish from the source of infection promoted repair of damaged gill tissue; within 14 d of fish transfer to clean water, gill pathology associated with the acute infection was negligible.  相似文献

Gills from Atlantic salmon with experimentally induced amoebic gill disease (Neoparamoeba spp.) were examined with transmission electron microscopy to assess pathology and host-cell responses. Amoebae were found either on the surface epithelium or with pseudopodia extending deeply into invaginations of epithelial cells. The amoebae had various densities along the plasma membrane and contained electron-dense deposits within their cytoplasm. Surface epithelial cells sloughed from the gills and had features consistent with apoptosis, including rounded shape, loss of surface microridges, and hypercondensation of nuclear chromatin. Affected areas of gills had fusion of secondary lamellae with interlamellar spaces occupied by mitotic epithelial cells and eosinophils. Eosinophils contained abundant fusiform-shaped granules that measured approximately 1 microm long and 360 nm wide. The granule consisted of an electron-dense matrix with a central inclusion that was less electron-dense, consisting of particulate and fibrillar material. In many instances, the central inclusion appeared empty and 90% of the eosinophils had morphology suggestive of piecemeal degranulation. Also observed within affected areas were a few neutrophils, mucous cells releasing mucus, and a small number of dendritic-like cells.  相似文献

Abstract

Gill neoplasms have been infrequently described for teleost fishes. Nine of 51 cases of primary cardiac endothelial cell neoplasms of a teleost fish species, mangrove rivulus Rivulus marmoratus, also possessed endothelial cell neoplasms in their gills. These neoplasms occurred in the afferent arterioles of the gill filaments. They were made up of round cells or spindle cells that had nuclei similar to typical endothelial cell nuclei. The large gill neoplasms, with more anaplastic cells, penetrated the walls of the gill arterioles and invaded adjacent gill tissues. Supporting evidence for the metastatic nature of these gill neoplasms includes the following: (1) they were secondary in timing, numbers, and size to the advanced sarcomas of the heart; (2) cell phenotypes of both gill and cardiac neoplasms were similar in cytoplasmic and nuclear features; (3) the cardiac neoplasms appeared to be shedding neoplastic endothelial cells into the lumen of the bulbus arteriosus with a direct route via the circulation of the ventral aorta to the afferent arterioles of the gills; and (4) direct evidence that the gill neoplasms arose from gill endothelium was lacking.  相似文献

Glycoproteins (GPs) were visualized histochemically in the secretory cells – the mucous goblet cells (the type A and the type B), the rodlet cells and the epithelial cells in different regions of the buccal cavity of Cirrhinus mrigala. The type A mucous goblet cells, the type B mucous goblet cells, the rodlet cells and the epithelial cells elaborate GPs with oxidizable vicinal diols and GPs with sialic acid residue without O‐acyl substitution. The type A mucous goblet cells, in addition, contain moderate amounts of GPs with O‐sulphate esters. The type B mucous goblet cells, in contrast, contain high concentrations of GPs with O‐sulphate esters. The rodlet cells secrete small amounts of GPs with oxidizable vicinal diols. The analysis of the results elucidates interesting differences in the composition and concentration of GPs in the mucus elaborated by the secretory cells indicating the potential importance of the glycoproteins in the buccal cavity. These GPs could be considered to represent a mechanism for modulation of the composition of the protective mucus layer correlated to specific functions.  相似文献

Abstract

An aquaculture research facility experienced high mortality rates in white bass Morone chrysops associated with a monogenean infestation of the gills, but not in striped bass Morone saxatilis in the same facility. All mortalities had pale gills. Monogeneans, identified as Gamacallum macroura (MacCallum and MacCallum 1913) Unnithan 1971, were found on the gills. Pale-gilled and healthy white bass were selected with no particular attention to condition for venipuncture and euthanasia for postmortem examination, including parasite counts from gills. The median packed cell volume (PCV) of fish with gill pallor was 12.5% (range 9–37%) while PVC of fish with more normal color was 30% (27–33%). Association between the PCV and gill pallor score was statistically significant, as was the association between PCV and the number of monogeneans found on the gills of each fish. Median estimated white blood cell count of fish with gill pallor, at 12.05 × 103/μL (range 3.8–24.7), was significantly lower than of apparently healthy fish: 24.7 × 103/μL (17.3–31.5). Histopathology of the gill arches of pale-gilled fish revealed multifocal moderate to severe branchitis, focal areas of dilated hyperplastic lamellae occluded by fibrin, and monogeneans attached to the lamellae. Fish that were apparently healthy had grossly similar histologic lesions, but at lower frequency and severity.

The influence of stress and cheese-whey on intestinal parameters in mice   总被引:1,自引:0,他引:1  
In a mouse model the effects of environmental stress on gastrointestinal parameters and the effects of cheese-whey on the changes induced by stress were studied. Mice were subjected to overcrowding, lack of bedding, overcrowding together with lack of bedding, continuous light, and housing at 30 degrees C. The influence of stress on relative caecal weight, faecal enterobacteriaceae, colonisation resistance (CR), filamentous segmented bacteria in the small intestine, fusiform bacteria in the faeces, and concentration of faecal bile acids was studied. Stress had no influence on relative caecal weight, faecal enterobacteriaceae, and faecal bile acids. Stress did decrease colonisation resistance, presence of segmented filamentous bacteria in the small intestine, and fusiform bacteria in the faeces. Cheese-whey had no positive effect on the microbiological disturbances caused by stress. The number of filamentous segmented bacteria in the small intestine and fusiforms in the faeces decreased by giving cheese-whey.  相似文献

Histological findings show that various pathogenic processes are involved in the cases of clinically detected branchionecrosis. The changes in gills occurring at toxic branchionecrosis are a necrobiotic process in their nature; in gill lamellae this process manifests itself as a swelling of the respiratory epithelium and as dilation of lamellar sinuses, and in stratified epithelium of gill arch and filaments it takes on the form of the hypertrophy and hyperplasia of epithelial cells. In severe cases the swollen epithelium forms deposits filling up also the interlamellar space. Toxic branchionecrosis is a term referring to a disease when the pathological processes are caused by ammonia intoxication and autointoxication, the main role being played by pH value. At experimental ammonia intoxication the changes in gills are about the same as in toxic branchionecrosis but the symptoms are not so pronounced. Changes characteristic of spherosporosis involve severe necrotic processes in the respiratory epithelium as well as in the stratified epithelium of gills, and the epithelium harbours different developmental stages of spherospores. The spherospores are easily detected when stained with alcian blue at pH 2.5. The changes in the gills of carp naturally invaded by spherospores with additional ammonia intoxication correspond to findings characteristic of toxic branchionecrosis: degeneration of stratified and respiratory epithelium of gills combined with necrotic changes caused by the developmental stages of spherospores. When water is highly oxygenated with chlorinated lime, large cells with eosinophilic granules severely multiply mainly in the stratified epithelium of gills; this process is considered as an inflammatory process. Parasitic invasions by Trichodina and Chilodonella induce local inflammatory reactions with the swelling of the respiratory epithelium.  相似文献

Seed handling by primary frugivores can influence secondary dispersal and/or predation of post‐dispersal seeds by attracting different guilds of ground‐dwelling animals. Many studies have focused on seeds embedded in feces of mammals or birds; however, less is known about how ground‐dwelling animals treat seeds regurgitated by birds (without pulp and not embedded in feces). To compare the effect of differential seed handling by primary dispersers on secondary seed removal of Chinese yew (Taxus chinensis var. mairei), we conducted a series of exclosure experiments to determine the relative impact of animals on the removal of defecated seeds (handled by masked palm civet), regurgitated seeds (handled by birds) and intact fruits. All types of yew seeds were consistently removed at a higher rate by rodents than by ants. Regurgitated seeds had the highest removal percentage and were only removed by rodents. These seeds were probably eaten in situ without being secondarily dispersed. Defecated seeds were removed by both rodents and ants; only ants might act as secondary dispersers of defecated seeds, whereas rodents ate most of them. We inferred that seeds regurgitated by birds were subjected to the highest rates of predation, whereas those dispersed in the feces of masked palm civets probably had a higher likelihood of secondary dispersal. Seeds from feces attracted ants, which were likely to transport seeds and potentially provided a means by which the seeds could escape predation by rodents. Our study highlighted that primary dispersal by birds might not always facilitate secondary dispersal and establishment of plant populations.  相似文献

Lesions induced in the gills of rainbow trout (Salmo gairdneri) by exposure to acutely lethal aqueous concentrations of inorganic mercury and copper were examined by light and electron microscopy. Lesions were most severe during the first 48 hours of exposure to the metals and were characterized primarily by apoptosis of lamellar epithelial cells and lamellar fusion. The latter process occurred either by simple apposition of adjacent lamellae to each other or through epithelial hypertrophy and hyperplasia. Except for hypertrophy of and increased number of primary lysosomes in lamellar epithelial cells of animals exposed to mercury, branchial lesions were not prominent in fish collected between 48 and 96 hours. The branchial lesions observed in this study are compared with pathological processes occurring in mammals.  相似文献

This study aimed to determine the cellular alterations in the gill of Labeo rohita exposed to lethal temperature maxima (LTMax) and lethal temperature minima (LTMin) by means of transmission electron microscopy (TEM). Acclimation of advanced fingerlings of L. rohita was carried out at 26°C for 30 days. Acclimated fish were subjected to a constant rate of increase or decrease in temperature (0.3°C/min) until the LTMax and LTMin values were reached. Dissected gills were processed for TEM, both at the end of acclimation period at ambient temperature (26°C) and at lethal temperatures. Results indicated that at ambient temperature, the gill tissues appeared normal. However, significant changes were observed at lethal temperatures. The gill tissues at lethal temperature maxima showed severely damaged lamellae, with more vacuolated space. At lethal temperature minima, gill tissues showed increased density of mitochondria. Our prima‐facie report indicated that L. rohita exposed to lethal temperatures exhibited marked ultrastructural changes in the gills.  相似文献
