Eleven nematode-free Beagle pups were inoculated with Ancylostoma caninum and Toxocara canis; infection became patent 13 and 35 days later, respectively. Eight pups were treated with fenbendazole oral suspension (10%) at a dosage of 50 mg/kg of body weight/day for 3 days. The remaining three animals were unmedicated controls. The drug was effective in reducing both ascarid and hookworm burdens, and there was marked improvement in the clinical condition of treated pups as compared with unmedicated control pups. Natural expulsion of worms in control animals was 53% for ascarids and 2% for hookworms. Drug-related toxicosis was not observed in any of the medicated animals. It was concluded that fenbendazole oral suspension (10%) at the 50-mg/kg dosage is easily administered and is an effective drug for reducing nematode burdens in experimentally infected pups.  相似文献

Infections with Ancylostoma caninum are transmitted orally or percutaneously. The transmission of infectious stages with the milk of particular importance for the distribution of the species. It occurs during the dissemination of larvae that follows every infection as well as after reactivation of resting somatic larvae in the bitch at the end of the pregnancy. The galactogenic transmission of larvae occurs even when, due to existing immunity, no patent infections develop in the bitch. Immunity does not or only to a low extent influence impatient infections or the migration of reactivated somatic larvae. It also allows a limited reestablishment of a deposit of larvae in the bitch. Following percutaneous infection dermatitis occurs in the area of larval penetration and the lung is affected by migrating larvae. Intestinal stages of Ancylostoma caninum damage the host by ingestion of the mucosa of the small intestine and withdrawal of blood. Main symptoms of ancylostomiasis are a mucous haemorrhagic diarrhoea and anaemia, that become visible 8 to 10 days post infection. The examination for impatient infections with Ancylostoma caninum can be done by immunofluorescence and ELISA. With both methods antibodies against third stage larvae can be detected from the first or second week post infection onward. Patent infections with Ancylostoma caninum can easily be detected by faecal examination for the presence of the characteristic oval, thin-walled eggs containing few blastomeres. Galactogenic infections with Ancylostoma caninum can be prevented or reduced by a regular treatment of the bitch with albendazole, fenbendazole or oxfendazole during the activation of larvae in the last third of the pregnancy or by repeated treatment with ivermectin shortly before and after birth. To prevent patent infections, galactogenic infected puppies have to be treated early and repeatedly.  相似文献

The aim of this investigation was to examine whether reactivated Ancylostoma caninum larvae can be eliminated by the administration of moxidectin to pregnant bitches. Four pregnant bitches infected experimentally with 20,000 third-stage larvae of A. caninum were treated subcutaneously with 1 mg moxidectin/kg body weight on day 55 of the pregnancy (5-8 days before parturition). Another four experimentally infected pregnant bitches served as controls. The single moxidectin treatment completely prevented lactogenic infections in the puppies. Neither intestinal stages nor somatic larvae could be found. The administration of moxidectin caused no local or systemic side-effects in the bitches. All 22 puppies of the treated bitches were born healthy and remained so during the whole trial period. Beginning during the third week after birth, all 20 puppies of the untreated bitches developed a severe microcytic, hypochromic anaemia and they revealed a total of 8649 intestinal stages of A. caninum after autopsy.  相似文献

Fenbendazole and albendazole, given at a dose rate of 150 mg/kg for 3 days, produced a 90 per cent reduction in the numbers of second stage larvae of Toxocara canis present in the tissues of dogs although no reduction in the number of larvae found in the brains of infected dogs occurred with this treatment. The results suggest that a course of 3 day therapy with these anthelmintics should prevent prenatal infections in puppies. However, if infection is acquired by bitches during late pregnancy or early lactation, the transmammary route of infection becomes important. Therefore, anthelmintic treatment of the bitch prior to pregnancy will not prevent transmission of infection to her puppies should the bitch acquire a new infection of T. canis during pregnancy or early lactation. Alternatively, infection with T. canis can be controlled through the treatment of neonatal puppies for migrating larvae of T. canis. Treatment of newborn puppies with fenbendazole, albendazole or oxfendazole at a dose of 100 mg/kg for 2–3 days produced a 91–99 per cent reduction in the number of adult parasites found. In addition, a single dose of fenbendazole, given at a dose rate of 40 mg/kg, eliminated 93–96 per cent of adult T. canis from the intestines of 4–5-week-old puppies. These latter treatments would need to be repeated to eliminate completely the infection from puppies.  相似文献

A Beagle bitch was inoculated SC and IM with 1.5 million Neospora caninum tachyzoites on the 35th day of gestation. Eight pups were born alive 28 days after N caninum inoculation of the bitch. Pup 1 did not breathe and died while still enclosed in fetal membranes. Pup 2 died 2 days after birth (DAB). Pups 3, 4, and 5 were euthanatized on 2, 3, and 20 DAB, respectively, because they were hypothermic and not nursing. Pups 6, 7, and 8 remained clinically normal. Indirect fluorescent N caninum serum antibody titers were: less than 50 (pups 1 and 8 at 17 DAB, and the bitch before inoculation), 50 (pups 2 and 3 on 2 DAB), 200 (pups 6 and 7 on 17 DAB), and 800 (bitch on day 17 after parturition). Neospora caninum was recovered in cultured cells inoculated with placenta and tissues from all 5 pups necropsied, and N caninum was seen in histologic sections of the heart of pup 5. Results indicate that N caninum can be transplacentally transmitted in dogs.  相似文献

Coprological examination was used to estimate the prevalence of gastrointestinal parasites in stray and domiciled dogs from Botucatu, S?o Paulo State, Brazil. Risk factors for dog infection were assessed in relation to demographic, husbandry and management data. The dog owners completed a questionnaire survey on some aspects of dog parasitism such as parasite species, mechanisms of infection, awareness of zoonotic diseases and history of anthelmintic usage. Parasites were found in the faeces of 138 dogs, with an overall prevalence of 54.3%. Dogs harbouring one parasite were more common (31.4%) than those harbouring two (18.5%), three (3.2%) or four (1.2%). The following parasites and their respective frequencies were detected: Ancylostoma (37.8%), Giardia (16.9%), Toxocara canis (8.7%), Trichuris vulpis (7.1%), Dipylidium caninum (2.4%), Isospora (3.5%), Cryptosporidium (3.1%) and Sarcocystis (2.7%). Stray dogs were found more likely to be poliparasitized (P<0.01) and presented higher prevalence of Ancylostoma, T. canis and Giardia (P<0.01) than domiciled ones. Toxocara canis was detected more frequently in dogs with <6 months of age (P<0.05) and no effect of sex or breed could be observed (P>0.05). Except for Ancylostoma, that showed a significantly higher prevalence in dogs living in a multi-dog household (P<0.01), parasite prevalences were similar in single- and multi-dog household. The answers of dog owners to the questionnaire showed that the majority does not know the species of dog intestinal parasites, the mechanisms of transmission, the risk factors for zoonotic infections, and specific prophylactic measures. The predominance of zoonotic species in dogs in the studied region, associated with the elevated degree of misinformation of the owners, indicates that the risk of zoonotic infection by canine intestinal parasite may be high, even in one of the most developed regions of Brazil.  相似文献

In five multicentre field trials, the efficacy and safety of a combination of oxantel/pyrantel/praziquantel (Dolpac), Vetoquinol SA) in the treatment of naturally acquired gastrointestinal nematode and/or cestode infestation in dogs was evaluated in northern and southern Europe. Forty-eight investigators from France, Belgium, Germany, Italy and Spain enrolled 329 dogs to be treated with the tested combination; 235 of these dogs complied with the inclusion criteria of the protocol and had a tested helminth identified on Day 0. A pooled analysis was performed on each of the following helminth species: Toxocara canis, Ancylostoma caninum, Toxascaris leonina, Trichuris vulpis, Uncinaria stenocephala, Taenia spp. and Dipylidium caninum, which were isolated on Day 0. The main efficacy criterion was the egg per gram (epg) percent reduction of the nematodes and the absence of proglottids and or eggs for the cestodes. After treatment, dogs were examined on Day 7, Day 14 and Day 21. The efficacy of the combination against Toxocara canis was 99.1%, 98.8% and 98.9% on Day 7, Day 14 and Day 21, respectively. At the same occasions the efficacy was, respectively, 99.2%, 99.2% and 99.3% against Ancylostoma caninum, 97.3%, 97.2% and 98.4% against Trichuris vulpis, 98.4%, 98.8% and 98.8% against Uncinaria stenocephala, 98.9%, 99.5% and 99.9% against Toxascaris leonina, 97.1%, 100% and 100% against Dipylidium caninum and 100% against Taenia spp.  相似文献

Of the gastro-intestinal helminths found in 1 063 dogs from different parts of the country, Dipylidium caninum, the most prevalent cestode, occurred in 19,7% of the animals. Echinococcus granulosus was present in 10 dogs, 8 of which were from urban areas, and Taenia hydatigena in 69 dogs. Taemia multiceps was recovered from dogs only in the Cape Province and the Orange Free State. Toxocara canis was present in 81 of the necropsies carried out on 253 dogs from the Pretoria municipal area, while Ancylostoma spp. were present in 175 dogs. In the Pretoria area Ancylostoma caninum (11,9%) was more prevalent than Ancylostoma braziliense (6,2%).  相似文献

Bitches with naturally occurring Brucella canis infection were treated with combined antibiotic therapy consisting of tetracycline, dihydrostreptomycin, and trimethoprim-sulfadiazine. After treatment, all but 1 bitch became abacteremic, and serologic titers declined for a variable length of time (3 months to 1 years). Abortion did not occur while these bitches were abacteremic. Although sequential antibiotic therapy for 6 weeks did not eradicate Brucella canis from affected bitches, it did not prevent abortion. The number of live pups whelped and weaned by treated bitches was comparable with that in bitches before they became infected.  相似文献

A bitch was inoculated subcutaneously and intramuscularly with Neospora caninum tachyzoites on Day 35 of pregnancy. Eight pups were born 28 days later. Five pups became ill and necropsies were performed before 20 days of age. Three pups and the bitch remained clinically normal for 7 weeks after parturition when they were intramuscularly injected with 40 mg kg-1 methylprednisolone acetate weekly to activate chronic N. caninum infection. Necropsies were performed 48, 17, 18, and 18 days respectively after administration of corticosteroids. Hepatic necrosis, pneumonia, encephalomyelitis, and myonecrosis were the main changes seen in these dogs.  相似文献

It was evaluated by means of a controlled test to what extent reactivated larvae of Ancylostoma caninum were influenced by single or double treatment with ivermectin. The investigations were done with a total of ten experimentally infected bitches. The single treatment of dams two to ten days ante partum with 0.5 mg or 1 mg ivermectin per kg bodyweight reduced the wormburden of their puppies on an average by 96.6% and 98.5%, respectively. The double treatment with 0.5 mg/kg bodyweight ante and post partum each with an interval of ten days completely prevented lactogenic infections with Ancylostoma caninum. Following a single application of 0.5 mg ivermectin per kg bodyweight the excretion of larvae was markedly delayed and remained on a low level. In addition the infections of the puppies of single treated dams became patent on an average 9.5 and 14.5 days later than the controls. In the bitches no side-effects, neither local nor general, were seen after the treatment with ivermectin. All forty puppies of the treated bitches were born without visible damages.  相似文献

Pyrantel pamoate, formulated in a beef-based chewable tablet, was evaluated for efficacy in dogs against induced and natural infections of Toxocara canis, Toxascaris leonina, Ancylostoma caninum and Uncinaria stenocephala. Dose titration trials were conducted in Canada, the UK and Germany in dogs treated with pyrantel (as pamoate salt) at 0, 2.5, 5 or 10 mg kg-1 body weight. These studies showed that a dose rate of 2.5 mg kg-1, the efficacy of pyrantel against adult T. canis, T. leonina, U. stenocephala and A. caninum was 76.1, 85.6, 100 and 87.9%, respectively. Efficacy at 5 mg kg-1 against the same parasites was 94.2, 92.0, 93.5 and 93.8%, respectively, and at 10 mg kg-1 efficacy was 91.2, 97.6, 98.7 and 91.3%, respectively. No adverse effects due to treatment were seen in any of these trials.  相似文献

The anthelmintic activity of ivermectin was tested in 98 dogs against adult ascarids (Toxocara canis, Toxascaris leonina), hookworms (Ancylostoma caninum, A braziliense), and whipworms (Trichuris vulpis), and against experimentally induced infections (4th-stage larvae) of T canis and A caninum. Dosage levels tested were single subcutaneous injections of 50, 100, 200, or 400 micrograms/kg of body weight with appropriate vehicle-treated controls. A minimum of 4 (usually 5) dogs were tested with each parasite and dosage level. The lowest dosage level, 50 micrograms/kg, and all higher dosage levels expelled greater than 99% of the adult forms of both species of hookworms and intestinal larval forms of A caninum, as determined by worm counts at necropsy. A dosage level of 100 micrograms/kg was required to expel greater than 99% of whipworms and 200 micrograms/kg was necessary to expel adult (91%) and larval (97%) stages of T canis. Ivermectin was only marginally effective (34.2%, 46.2%, 69.2%, and 53.8%) against Toxascaris leonina at 50, 100, 200, and 400 micrograms/kg, respectively, and had no effect against occasional infections with the tapeworms, Dipylidium caninum (14 dogs) and Taenia spp (3 dogs).  相似文献

Toxocara canis (T. canis) is originally a parasite of canine bitches and their pups. The pathogenicity of T. canis infection is enhanced during pregnancy and puppyhood. The aim of this study was to investigate if modification of IFNgamma and IL-10 secretion occurs during infection in pregnant dogs and puppies. Analysis of cytokines secreted could let us hypothesize a role for IL-10 and/or IFNgamma in T. canis infection. We tested T. canis-specific production of IFNgamma and IL-10 by lymphocytes of pregnant dogs and their puppies after in vitro re-exposure to purified excretory/secretory antigen (ESAg) from T. canis. Blood mononuclear cells (BMC) isolated from pregnant dogs and their puppies were cultured in the presence of ESAg. Cultures' supernatants were tested for cytokine levels by ELISA. Results obtained showed that IL-10 concentrations increased during pregnancy in infected animals and in the meantime IFNgamma production decreased. In puppyhood, we observed that, IL-10 concentration decreased with the age of puppies mainly in infected animals while IFNgamma increased. In conclusion, our data suggests that BMC of infected dogs have a particular modification of IL-10 and IFNgamma synthesis. These data could be the basis to design immunotherapeutic approaches.  相似文献

Eight trials were conducted in dogs to document the efficacy of ivermectin (6 micrograms/kg of body weight) and pyrantel pamoate (5 mg of active pyrantel/kg) in a beef-based chewable formulation against Dirofilaria immitis, Ancylostoma caninum, Uncinaria stenocephala, Toxocara canis, and Toxascaris leonina. Three studies involved induced infection with D immitis, and 5 studies involved induced or natural infection with hookworms and ascarids. In 3 intestinal parasite trials, the efficacy of the combination chewable tablet was compared with each of its components. Results indicated that 1 component did not interfere with the activity of the other. In 1 heartworm and 2 intestinal parasite trials, the efficacy of pyrantel, ivermectin/pyrantel combination, or ivermectin with pyrantel dosage of 10 mg/kg was evaluated. The ivermectin/pyrantel combination was 100% effective in preventing development of D immitis larvae. Efficacy of the combined product against T canis, Toxascaris leonina, A caninum, and U stenocephala was 90.1, 99.2, 98.5, and 98.7%, respectively. In the intestinal parasite trials, each individual component was found not to interfere with the anthelmintic action of the other. Increasing the dosage of pyrantel to 10 mg/kg (2 x that in the combination) did not interfere with the efficacy of ivermectin against heartworm or increase the activity of pyrantel against intestinal parasites.  相似文献

The efficacies of nitroscanate and mebendazole against nematodes and Dipylidium caninum were compared by the examination of the faeces of 155 dogs. In unweaned puppies nitroscanate acted significantly faster and was significantly more effective than mebendazole against Toxocara canis. In adult dogs, both drugs had a low efficacy against Trichuris vulpis but a similarly good efficacy against other nematodes, with a tendency for nitroscanate to be more effective in reducing the prevalence of Ancylostoma caninum. Nitroscanate reduced the prevalence of D caninum infections considerably more than mebendazole.  相似文献

The effects of canine herpesvirus (CHV) on fetuses were studied after IV inoculation of pregnant bitches in the 2nd trimester of gestation. Cesarean sections were performed on 2 bitches that were inoculated with CHV on the estimated 30th day of gestation. Bitch M-1 had 2 mummified fetuses and bitch M-2 had 4 mummified and 2 dead fetuses and 3 live-born pups. Infection by CHV was confirmed histopathologically by the presence of focal areas of necrosis associated with intranuclear inclusion bodies in heart muscle sections of 1 dead fetus; CHV was not recovered from other organs. Abortion occurred between the 2nd and 3rd week after inoculation of another pregnant bitch inoculated with CHV on the estimated 30th day of gestation. Two bitches inoculated with CHV on the estimated 40th day of gestation gave birth prematurely to 10 pups. The detection of characteristic herpesviral lesions in various organs and the reisolation of CHV from the liver, spleen, kidneys, and lungs of premature pups indicated CHV infection. Transplacental infection of fetal pups by CHV resulted in their death and subsequent mummification. It appears that abortion and premature birth also may occur in pregnant bitches infected during the 2nd trimester of gestation.  相似文献

Although there have been numerous microbial examinations of soil for the presence of human pathogenic developmental parasite stages of Ancylostoma caninum and Toxocara canis, molecular techniques (e.g. DNA extraction, purification and subsequent PCR) have scarcely been applied. Here, DNA preparations of soil samples artificially contaminated with genomic DNA or parasite eggs were examined by PCR. A. caninum and T. canis-specific primers based on the ITS-2 sequence were used for amplification. After the sheer DNA preparation a high content of PCR-interfering substances was still detectable. Subsequently, two different inhibitors of PCR-interfering agents (GeneReleaser, Bioventures Inc. and Maximator, Connex GmbH) were compared in PCR. Both substances increased PCR sensitivity greatly. However, comparison of the increase in sensitivity achieved with the two compounds demonstrated the superiority of Maximator, which enhanced sensitivity to the point of permitting positive detection of a single A. caninum egg and three T. canis eggs in a soil sample. This degree of sensitivity could not be achieved with GeneReleaser for either parasite Furthermore, Maximator not only increased sensitivity; it also cost less, required less time and had a lower risk of contamination. Future applications of molecular methods in epidemiological examinations of soil samples are discussed/elaborated.  相似文献
