The pine sawyer beetle Monochamus galloprovincialis (Olivier) (Coleoptera Cerambycidae) gained importance as a pest in Europe after the introduction in Portugal of the pine wood nematode Bursaphelenchus xylophilus, the agent of pine wilt disease. As the nematode is transmitted by the adult beetles during both maturation feeding and oviposition, effective methods of monitoring of the longhorn beetle populations are needed for the surveillance of the disease spread in Europe. Although in the last years many studies focused on the efficacy of different trapping methods, the general results are still controversial. In this study, we compared the efficiency of two trap types, cross-vane and multi-funnel, and two attractive blends, a kairomonal lure (??-pinene, ipsenol and methyl-butenol) and a commercial kit specific for M. galloprovincialis (pheromone plus kairomonal lure), in order to identify the trap/blend combination more appropriate for the surveillance of the M. galloprovincialis populations. Moreover, the trap efficiency was tested also in a pine stand colonized by M. galloprovincialis after a hailstorm and in a control stand. The results showed that the multi-funnel traps baited with the specific commercial kit was the most effective combination. Interestingly, trap catch was similar in the two pine stands, despite a different density of the target species. As the trapping method was effective also at low population density of M. galloprovincialis, it may be adopted as a general tool for beetle surveillance in the forest and at ports of entry.  相似文献

Ralstonia solanacearum, which consists of five races/biovars, is considered a “species-complex” and is an important phytopathogen that causes wilt disease in more than 200 plant species. R. solanacearum race 1 biovar 4 (R1bv4) has caused yield losses of 30–80 % in the vegetable sweet potato (VSP) in the last decade in Taiwan. To identify the source of the initial inoculum of R1bv4 in VSP fields, soil and cuttings from these fields were examined from 2009 to 2010. The results of the investigation indicated that the population of R1bv4 was generally distributed throughout the natural soil of VSP fields at a density ranging from 1.3?×?102 to 9.5?×?105 cfu/g soil; however, the incidence of bacterial wilt was not significantly associated with the density of the R1bv4 population in soils (R2?=?0.084). In contrast, densities of R1bv4 ranging from 2.3?×?103 to 5.9?×?105 cfu/g tissue were detected in the vine tissue of asymptomatic plants in the fields. Additional experiments demonstrated that R1bv4-free VSP cuttings without visible symptoms planted in infested soils in the greenhouse setting could carry approximately 3.1?×?105 R1bv4 cfu/g tissue, which suggests the existence of a latent period for R1bv4 in VSP plants. The results of a BIO-PCR analysis showed that R1bv4 was detected in 2.0 to 98.0 % of the VSP cuttings used for propagation in fields; in addition, the percentage of VSP cuttings carrying R1bv4 and the incidence of bacterial wilt in fields were positively correlated (R2?=?0.909). The inoculation experiments conducted in greenhouses and in fields showed that the cutting inoculum (CI) contributed more to the incidence of bacterial wilt in VSP plants than the soil inoculum (SI). In the field experiments conducted in 2010, an incidence of disease of 27.1 to 38.5 % was detected in healthy field cuttings 8 months after transplantation; in contrast, the incidence of disease in field cuttings carrying R1bv4 was 49.0 to 68.8 %.  1987-1988年在江苏南京-镇江松材线虫疫区内采(征)集143份自然枯死或濒死马尾松样本,在94份(65.7%)样本中查见松材线虫。有关林分中马尾松的病枯情况不一,在句容桥头一下蜀一线、江宁汤山和六合方山等处有过相当多的马尾松发病死亡。表明松材线虫是当地马尾松自然枯死的主要原因。人工测定了无黑松林区5年生浙江富阳马尾松苗及混栽黑松林区4-5年生江苏马尾松苗对两种松材线虫分离物的反应,结果指出,供试松苗都表现中度感染或高度感染,并有20%或70-80%的病枯株。接种林间马尾松,初步证实马尾松对松材线虫的抗性随树龄变化。1-2年生马尾松苗高感松材线虫,病枯株率高达85.7%;树龄25年以上的成材树病枯株率约30%;而10年左右的幼树则无一发病死亡。  相似文献

为筛选出新型的高效松材线虫Bursaphelenchus xylophilus防治药剂,采用浸渍法室内测定了氯氟氰虫酰胺、阿维菌素、甲维盐、灭多威、特丁硫磷、甲基异柳磷和吡虫啉7种药剂对松材线虫的毒杀作用和对其繁殖能力的影响,采用高效液相色谱仪测定氯氟氰虫酰胺在马尾松体内的输导分布,并在林间开展树干注射氯氟氰虫酰胺抗松材线虫病试验。结果表明,7种药剂中氯氟氰虫酰胺对松材线虫的毒力最强,LC_(50)为0.019 mg/L,阿维菌素和甲维盐次之,LC_(50)分别为0.0213 mg/L和0.0214 mg/L。氯氟氰虫酰胺树干注射3个月内能迅速从马尾松基部传导至树梢顶部,树干边材东、南面的药剂含量显著高于西、北面,且树干不同高度的氯氟氰虫酰胺含量均高于其LC50;氯氟氰虫酰胺制剂在林间注干后,对人工接种病原松材线虫松树的防治效果为73.33%,自然感染松材线虫病松树的死亡率降至2.00%,而对照试验中人工接种松材线虫松树和自然感染松材线虫病松树的死亡率分别为93.33%和19.67%。表明氯氟氰虫酰胺制剂能有效预防松树个体的松材线虫病发生与为害,可以推广使用。  相似文献

This study investigated the effect of calcium nutrition on tomato bacterial wilt caused by Ralstonia solanacearum and the regulation of resistance mechanisms. Plants cultured in nutrient solution with calcium concentrations of 0.5, 5.0, and 25.0 mM, were inoculated with R. solanacearum by the root dip method. Severity of disease development, Ca concentration in tomato root and shoot tissues, hydrogen peroxide (H2O2) concentration, peroxidase (POD, EC 1.11.1.7) and polyphenol oxidase (PPO, EC 1.10.3.2) in tomato leaves were analyzed. Disease severities of low, medium and high Ca treatments were 100 %, 77.1 % and 56.8 % respectively. Plant growth in high Ca treatment was significantly better than those in low Ca treatment in height, stem diameter and biomass. Tomato plants absorbed significantly more Ca in roots and shoots as the level of Ca in the nutrient solution increased. In addition, H2O2 level in high Ca treatment rose faster and reached a higher peak with 10.86 μM gFW?1(31.32 % greater than medium Ca plants). The activities of POD and PPO also have a greater increase in high Ca treatment with 99.09 U gFW?1 and 107.24 U gFW?1 compared to 40.70 U gFW?1 and 77.45 U gFW?1 in low Ca treatment. A negative correlation was found between Ca concentration, level of H2O2, POD, PPO in tomato, and disease severity, indicating that they played an important role in resistance of tomato to this disease. These results suggested that Ca was involved in the regulation of H2O2 concentration, and activity of POD and PPO in tomato.  相似文献

Since the beginning of the 20th century, pine trees in Japan have been seriously damaged by the pine wilt disease. This disease is caused by the pine wood nematode, Bursaphelenchus xylophilus, which is transmitted by the Japanese pine sawyer, Monochamus alternatus. The control of disease depends to a large extent on chemicals, but the public is now demanding environmentally friendly control methods. The virulence of B. xylophilus varies very widely. Pre-inoculation of young pine trees in a nursery with avirulent B. xylophilus has induced systemic resistance of trees against a subsequent inoculation with virulent B. xylophilus. This induced resistance was considered a hopeful means for developing a biological control for the disease. The induced resistance by the avirulent nematodes was also expressed in mature pine trees in a forest where the disease was naturally epidemic. However, the effects of induced resistance were not satisfactory for practical biological control. Since the inoculation with higher concentrations of the avirulent B. xylophilus induced the resistance more effectively, the pre-inoculation method will need to be improved to develop the biological control. The induced resistance of pine trees by avirulent B. xylophilus should be one of the candidate biological control methods against pine wilt disease. This induced resistance also provides an experimental system to clarify physiological interactions between the nematodes and pine trees.  相似文献

The temporal and spatial distribution of Tomato spotted wilt virus (TSWV) was studied in a 2,085 m2 tomato field with a total of 4,080 plants during 10 weeks from 5 July to 6 September in 2004. First infected plants were detected 4 weeks after the source plants were placed in the field and their number continued to increase by week 7. A total of 147 plants (3.6%) were recorded to be infected according to symptom occurrence and serological test by the end of the experiment. Thrips monitoring was carried out from May 25 to October 25 in the same year. Two thrips species, Frankliniella intonsa (Tryborn) and Thrips tabaci (Lindemann), were determined in the experimental area, with the latter one being the more prevalent (84.9%) species. There was a significant correlation (r?=?0.988, P?<?0.01) between the number of thrips species and the number of infected plants in the field. Majority of infected plants were not located near (0–10 m) an internal virus source within the experimental area, whereas 45.6% of the infected plants were located at a distance of 21–30 m. The number of infected plants at a distance of 21–25 m was significantly higher than at all other distances (P?<?0.05). There was no gradual increase in the distances of TSWV-infected plants within 0–30 m as time progressed. The virus seems to be transmitted by vectors through primary infections and scattered within 30-m distance, but limited secondary infections seem likely to occur after this distance in the case of a small internal virus source.  相似文献

Survival and development of Bursaphelenchus xylophilus (pine wood nematode) was studied for up to 40 weeks in Pinus pinaster sawn wood and branches: 30 boards (1200 × 100 × 25 mm), 30 long‐blocks (1200 × 95 × 95 mm), 10 pine branches with bark and nine branches without bark (1200 mm long). The nematode was found in all materials and through the entire sampling period, with higher abundance in the sawn wood (boards and long‐blocks). In the initial period B. xylophilus reproduced abundantly and a population peak was detected at 8–12 weeks. Subsequently, the populations declined and became dominated by third‐stage resistant larvae (JIII), and in the final sample nematode abundance was very low. Nematode decline in the wood was correlated with a decrease in the moisture content (MC) to below fibre saturation. Survival of the insect vector Monochamus galloprovincialis was also assessed in sawn boards (1200 × 100 × 25 mm, n = 31) and blocks (160 × 95 × 95 mm, n = 40). The majority of the larvae were killed when sawing the wood, although some adults successfully emerged from the boards (10% survival) and blocks (37%). These results represent a contribution to the quantification of the risks of dispersing pine wilt disease through wood packaging materials, confirming that untreated wood can support healthy and abundant B. xylophilus populations for sufficient time for vectors surviving the sawing process to complete their development, to emerge and disperse the nematode.  相似文献

Monochamus galloprovincialis Olivier (Coleoptera: Cerambycidae) is one of the vectors of the pinewood nematode Bursaphelenchus xylophilus (Steiner and Buhrer) Nickle (Nematoda: Parasitaphelenchidae), the causal agent of pine wilt in susceptible pine trees. It is therefore important to study both the biological and ecological characteristics of the beetle. In this study reproductive potentials of female beetles reared on two pine species were investigated under laboratory conditions and compared using fertility tables constructed for beetles reared on each tree species. Beetle population parameters were compared between tree species. The intrinsic rate of increase and associated population parameters of beetles reared on Pinus sylvestris L. were consistently higher than those reared on P. nigra Arnold. This result suggests that P. sylvestris has a significantly greater positive effect on the reproductive potential of M. galloprovincialis populations than does P. nigra.  相似文献

The pine wood nematode, Bursaphelenchus xylophilus, infects pine trees, leading to fatal pine wilt disease. Here, recombinant venom allergen-like protein (VAP) was obtained by expressing Bx-vap-1 in insect cells. Three-year-old Pinus massoniana were inoculated with recombinant VAP, simulating B. xylophilus esophageal gland secretions. Recombinant VAP up-regulated α-pinene synthase gene expression, the trees showed disease symptoms 15 d after inoculation and the xylem pith revealed brown tissue discoloration, indicating that recombinant VAP could damage P. massoniana cells. Recombinant VAP did not, however, lead to cavitation, indicating that the VAP secreted from B. xylophilus acts as a defense response elicitor.  相似文献

Black leaf mold (BLM), caused by Pseudocercospora fuligena is a serious threat to tomato production in the humid tropics. Accurate information about the incubation (IP) and latent period (LP) under various host susceptibility and weather favourability circumstances will help to formulate holistic approaches to manage this disease. In this study, effects of temperature, wetness duration, and leaf age on the monocyclic components (IP and LP) of BLM were studied from growth chamber (GC) and greenhouse (GH) experiments as well as detached leaf assays in growth cabins. Linear interpolation and inflection point (of logistic regression model) methods were used to determine IP and LP. These two methods were highly correlated in GC (r 2?=?0.89; P?<?0.0001) and GH experiments (r 2?=?0.90; P?<?0.0001) except when the epidemics were not asymptotic. Thus, IP and LP were estimated according to inflection point method. There was a delay of at least 5 days of IP and LP when plants were left in non-humid open environment than when exposed to wetness durations of 1, 2 or 3 days after inoculation. In general, IP and LP became shorter as the temperature increased from 20–24 and then to 28 °C. In growth chambers, there was more disease and consequently shorter IP and LP on young and unfolded tomato leaves that were 1-, 3-, or 5-week old at the time of inoculation than 7-week old leaves. In the greenhouse, there was about 50 % more disease incidence and sporulation on 1-week than 3-week old leaves. The shortest IP (8–11 days) and LP (12–13 days) were recorded from two out of three GH experiments on 1-week old leaves at an ambient mean temperature of 28.5 °C. This study implicated that fresh market tomatoes planted during warm temperatures in 50-mesh greenhouses and exposed to extended periods of wetness are highly prone to BLM infection at their young stages of growth.  相似文献

为有效防治松材线虫病,研制了注干用10%甲氨基阿维菌素苯甲酸盐 (以下简称甲维盐) 可溶粒剂,并通过在马尾松树干基部钻孔施药,考察了其对松材线虫病的防治效果。以苯甲酸钠、单水乳糖、可溶性淀粉和羧甲基纤维素为水溶性载体,采用混料均匀试验设计及偏最小二乘回归分析法对水溶性载体的配方进行了优化。将水溶性载体与溶于正丁醇和乳化剂OP-10的甲维盐 (油相) 在不同时间下混合并造粒,通过对其性能指标的追踪检测得出粒剂最佳配比和混合时间。结果表明:10%甲维盐可溶粒剂的最佳配比 (质量分数) 为:甲维盐10.0%(折纯),乳化剂OP-10 6.7%,正丁醇12.0%,苯甲酸钠39.0%,单水乳糖20.5%,水溶性淀粉3.0%和羧甲基纤维素4.5%;机械混合时间为10 min,粒剂外观为乳白色圆柱体,各项性能指标合格。注干施药结果表明:在浙江临安东湖村松材线虫病疫区,注干110棵马尾松,1 a后马尾松死亡率为0,对照区为6.1%;在福建泉州仙公山疫区,注干230棵马尾松,1 a后马尾松死亡率为1.7%,对照区10.8%。结果表明,所研制的10%甲维盐可溶粒剂可在松脂分泌旺盛的松树上注干施药,制剂造粒工艺简单,无包装物污染,对松材线虫病防治效果显著。  相似文献

In order to investigate the effects of flagellin secreted by bacteria associated with pine wood nematode (Bursaphelenchus xylophilus) on pine cells, suspension cells of Pinus thunbergii were treated with flagellin of Pseudomonas fluorescens GcM5-1A, and some physiological indices were determined. Superoxide anions radical (·O2 ?) in treated suspension cells increased with prolongation of treatment time, with maximum content of 1,282 μmol g?1 FW at 30 h, but the content of H2O2 gradually decreased as compared with that in the control group. Maximum content of malondialdehyde (MDA) and free proline, which is 1.72 times and 1.49 times of that in the control, respectively, appeared when suspension cells were treated for 12 h, after that both of the contents decreased gradually. The activities of superoxide dismutase (SOD), peroxidase (POD) and catalases (CAT) also increased at the primary stage of treatment and then decreased compared with those of the control.  相似文献

In this study, colonization and disease development of axenically-grown tomato plants by Clavibacter michiganensis subsp. michiganensis (Cmm), the causative agent of bacterial wilt and canker, was investigated. For this, a spontaneous rifampicin resistant strain of Cmm was tagged with a marker that expressed a green fluorescent protein (GFP) in a stable way and which possessed a similar virulence to the parental strain. In vitro plants were drop-inoculated at the stem base and the population dynamics was determined by dilution pour-plating in a selective medium. At 3 h after inoculation, Cmm was already present in low densities in roots, stems and leaves. At 16 dpi, Cmm was found throughout the entire plant in high densities of ca. 1010 cfu g?1. Symptoms developed in the in vitro plants typical for Cmm, such as canker, wilting and growth reduction. The presence of Cmm in vascular and parenchymatic tissue of in vitro tomato plants was confirmed by epifluorescence stereo- and confocal laser scanning microscopy. This study showed that in vitro tomato plants can be effectively used for detailed studies on interactions between Cmm and its host, in particular if a GFP-tagged strain of the pathogen is used.  相似文献

Seventeen field surveys were done in four commercial orchards during six consecutive fruit-growing seasons from 2006 until 2011 in order to determine the current frequencies of occurrence of M. laxa, M. fructigena, and M. fructicola and their relative contributions to postharvest brown rot in peaches and nectarines in the Ebro Valley. The relative frequencies of occurrence of Monilinia spp. were determined on three sources of primary inoculum and on three sources of secondary inoculum. The major relative frequencies of Monilinia spp. were significantly recorded (P?=?0.05) from mummified fruit on the trees (approx. 42 %) and 7-day-old harvested fruit with brown rot (32 %), followed by that recovered from mummified fruit on the orchard bed (14 %), pruned branches on the orchard bed (8 %) and latent infections of immature fruit (3 %). We found that: (a) the relative frequency of M. fructicola has increased over the years to coexist on the same level as at the time M. laxa, (b) M. fructigena is no longer a cause of brown rot in harvested peaches, (c), a progressive reduction in the time of the first appearance of Monilinia airborne conidia (r?=??0.30, P?=?0.003), and the time of the first latent infection (r?=??0.44, P?=?0.0001) was detected along years after correlation analysis, and (d) these displacements are not associated with an increased incidence of brown rot disease. The M. fructicola increase was due to its significantly increased presence in 7-day-old harvested fruit with brown rot (r?=?0.73, P?=?0.0009), in latent infections of immature fruit (r?=?0.68, P?=?0.002), on pruned branches on the orchard bed (r?=?0.56, P?=?0.018), and on mummified fruit sampled on the trees (r?=?0.53, P?=?0.03). This progressive increase was accompanied by a progressive reduction in the relative frequency of occurrence of M. laxa in 7-day-old harvested fruit with brown rot (r?=??0.55, P?=?0.021) and M. fructigena on mummified fruit sampled on the trees (r?=??0.51, P?=?0.03).  相似文献

Laurel wilt is caused by Raffaelea lauricola, a nutritional symbiont of an Asian ambrosia beetle, Xyleborus glabratus. American members of the Lauraceae plant family are most susceptible and 300 million trees have been killed by the disease in the southeastern USA since 2003. Recently, commercial production of an important crop in the laurel family, avocado (Persea americana), has been affected in southern Florida. We summarize studies in which diverse measures were tested for managing the disease. In all studies, trees were treated with potential laurel wilt control measures and subsequently inoculated with R. lauricola. On potted plants in greenhouse experiments, commercial nutritional products (Greenstim and Keyplex 350) and SAR products (Agri-Fos and Nutri-Phite), when applied as soil drenches or foliar sprays, had either no impact on, or increased laurel wilt symptom development compared to non-treated control treatments. Bark applications of Tilt (a propiconazole fungicide for which emergency registration had been obtained in 2010) in a surfactant (Pentrabark) enabled significant laurel wilt protection in greenhouse studies on small potted plants, but Pentrabark and other surfactants moved little propiconazole into the xylem of fruit-bearing trees in field studies. In efficacy studies in the field, Propiconazole Pro (an injectable formulation of propiconazole), Tilt, and two experimental formulations of another triazole fungicide, tebuconazole, decreased the development of laurel wilt compared to nontreated control trees when applied as undiluted injections into branches and scaffold limbs (microinjection), or injection of dilute fungicide into tree flare roots (macroinfusion). However, symptoms developed in all treated trees by 10–11 months after inoculation with R. lauricola, indicating that trees would need to be re-treated at least on an annual basis. Regardless of how fungicides were applied, insignificant levels of different active ingredients entered fruit. Although fungicide treatment of fruit-bearing avocado trees is not a concern for food safety and several triazole and DMI fungicides can protect avocado trees from laurel wilt, cost-effective measures with which the xylem could be loaded with and protected by these products remains a challenge. Management of laurel wilt in commercial avocado production areas is discussed.  相似文献

Nitric oxide (NO) is an important signal molecule involved in numerous plant responses to biotic and abiotic stresses. The effect of nitric oxide (NO) solution on pathogen infection and defence response of peach (Prunus persica (L.) Batsch) fruit against brown rot disease caused by Monilinia fructicola was investigated. The results showed that 15 μmol l?1 NO solution did not significantly inhibit spore germination, germ tube length or pathogenicity of M. fructicola, but significantly reduced disease incidence and lesion areas in the fruit. Although 100 μmol l?1 NO solution effectively inhibited the spore germination, germ tube elongation and pathogenicity of M. fructicola, the high concentration of NO solution caused damage to the fruit. Moreover, 15 μmol l?1 NO enhanced the activities of chitinase (CHI) and β-1,3-glucanase (GNS) in the fruit. RT-PCR analysis showed that the expression of four genes, CHI, GNS, pathogenesis-related protein 1 and 10 genes (PR-1, PR-10) all increased after NO treatment. Conversely, pretreatment with 100 μmol l?1 NO scavenger, 2-4-carboxyphenyl-4,4,5,5- tetramethylimidazoline-1-oxyl-3-oxide (cPTIO), rendered the fruit relatively susceptible to pathogen infection and inhibited the defence response of the fruit. These results suggest that NO solution treatment can protect peach fruit from pathogen infection by inducing the activities of the defence enzymes and the expression of PR genes.  相似文献

Understanding the distribution pattern of the Fusarium species can help prevent crop diseases and large yield losses. While several approaches have been used to control soil-borne pathogens, soil solarisation has shown promising results in managing these pathogens. The main objectives of this study were to: (i) describe the biogeography of Fusarium species in four different climatic zones in Iran and (ii) explain the effect of soil solarisation on main pathogenic Fusarium species in wheat grains, beans and date palms. A total of 12 sub-samples were collected from four different climatic zones including, Rasht (humid), Zanjan (semi-arid), Isfahan (extra-arid) and Ahwaz (arid). For precise identification, molecular-phylogenetic analyses of the species were also performed. From these four sites 17 Fusarium species were recovered. F. solani complex, F. oxysporum and F. equiseti were the only species found in all four regions; whereas F. compactum, F. sambucinum and F. fujikuroi were restricted to Ahwaz, Zanjan and Rasht, respectively. Furthermore, soil solarisation treatments were applied to F. pseudograminearum, F. solani and F. oxysporum, as the main cause of root rot pathogens and wilt disease of wheat, bean and date palm, respectively. After 6 weeks of soil solarisation application, the population densities of these species were decreased from 900 to 100 CFU g?1 in F. solani, from 600 to 50 CFU g?1 in F. oxysporum and from 550 to 0 CFU g?1 in F. pseudograminearum showing a promising result in controlling soil-borne pathogens. Mycogeography of Fusarium species and the effect of soil solarisation can help improve the management control strategies of these soil-borne fungi.  相似文献
