Modern rice varieties that ushered in the green revolution brought about dramatic increase in rice production worldwide but at the cost of genetic diversity at the farmers’ fields. The wild species germplasm can be used for broadening the genetic base and improving productivity. Mining of alleles at productivity QTL from related wild species under simultaneous backcrossing and evaluation, accompanied by molecular marker analysis has emerged as an effective plant breeding strategy for utilization of wild species germplasm. In the present study, a limited backcross strategy was used to introgress QTL associated with yield and yield components from Oryza rufipogon (acc. IRGC 105491) to cultivated rice, O. sativa cv IR64. A set of 12 BC2F6 progenies, selected from among more than 100 BC2F5 progenies were evaluated for yield and yield components. For plant height, days to 50% flowering and tillers/plant, the introgression lines did not show any significant change compared to the recurrent parent IR64. For yield, 9 of the 12 introgression lines showed significantly higher yield (19–38%) than the recurrent parent IR64. Four of these lines originating from a common lineage showed higher yield due to increase in grain weight and another three also from a common lineage showed yield increase due to increase in grain number per panicle. For analyzing the introgression at molecular level all the 12 lines were analyzed for 259 polymorphic SSR markers. Of the total 259 SSR markers analyzed, only 18 (7.0%) showed introgression from O. rufipogon for chromosomes 1, 2, 3, 5, 6 and 11. Graphical genotypes have been prepared for each line and association between the introgression regions and the traits that increased yield is reported. Based on marker trait association it appears that some of the QTL are stable across the environments and genetic backgrounds and can be exploited universally.  相似文献

Near-isogenic Rht lines of ten modern bread wheat (Triticumaestivum L.) and six durum wheat (T. turgidum L.) cultivars weredeveloped and evaluated in replicated trials under three soil moisturetreatments for two years in northwestern Mexico. The three soil moisturetreatments were created by providing one, two or six irrigations during eachcrop season. Grain yield and other traits were measured for each line ineach trial. Mean grain yields of short and tall T. aestivum or T.turgidum isolines were similar in the lowest yielding environment whenmean grain yields (0% grain moisture) of T. aestivum and T.turgidum were 2,232 and 1,870 kg ha-1, respectively. Mean grainyield of dwarf T. aestivum was significantly higher than that of tallgenotypes in another five trials with moderate to high yields. Theperformance of dwarf and tall T. turgidum isolines was unpredictablein moderate yielding trials, and the dwarf isolines yielded significantly morein trials that received six irrigations. Given that the tall isolines producedsignificantly more straw than their shorter counterparts, cultivation of tallwheats may be beneficial in semiarid environments where farmers' yields areclose to 2.5 t ha-1 or lower, and straw has value.  相似文献

To facilitate the use of wheat wild relatives in conventional breedingprograms, a wheat pre-breeding activity started at ICARDA in 1994/1995season. Preliminary results of gene introgression from wild diploidprogenitors, Triticum urartu, T. baeoticum, Aegilops speltoides andAe. tauschii and tetraploid T. dicoccoides are described. Crosseswith wild diploid Triticum spp. yielded high variation in plant andspike morphology. Synthetic hexaploids were produced from crosses of alocal durum wheat landrace `Haurani' with two Ae. tauschiiaccessions. Both Ae. tauschii accessions carry hybrid necrosis allelesthat gave necrotic plant phenotypes in crosses with some bread wheats.Backcross progenies with agronomical desirable traits, i.e. high spikeproductivity, short plant stature, earliness, drought tolerance and highproductive tillering, were identified in crosses of durum wheat with wild Triticum spp. and in a cross of one of the hexaploid synthetics with alocally adapted bread wheat cv. `Cham 6'. Resistance to yellow rust wasfound in durum wheat crosses with the three wild Triticum spp. andAe. speltoides and leaf rust resistance was identified in crosses withT. baeoticum and Ae. speltoides. The results show that wheatimmediate progenitors may be a valuable and readily accessible source ofnew genetic diversity for wheat improvement.  相似文献

Summary To determine the levels of heterosis in F1 hybrids, four current pea (Pisum sativum L.) cultivars from southern Australia were used as female parents and crossed with 18 introduced genotypes. The 22 parents, 72 F1 hybrids and, depending on the environment, either 54 or all 72 F2 families were grown in replicated plots in four environments. Grain yield, total dry matter, harvest index, branches per plant, pods per plant, seeds per pod, hundred seed weight, plant height, onset of flowering and flowering periods were evaluated. For both the F1 and F2 generation, heterosis was determined as the superiority over the mid-parent and also over the better parent. In addition, the superiority over the best commercial cultivar was calculated. Most hybrids were higher yielding than their mid-parent but were less stable in yield across environments. Four F1 hybrids were significantly higher yielding than the best parent, by up to 26%. There were significant correlations between F1 hybrid and mid-parent value for plant height, pods per plant and hundred seed weight but not for yield. Overall, grain yield heterosis was mainly due to more pods per plant in the hybrids. The level of heterosis for yield in a poor yielding environment was higher than that in a high yielding one. Both additive and non-additive gene effects were important in the expression of all studied traits. The average level of heterosis for grain yield and total dry matter in the F2 population was half of that in F1 hybrids. The low level of inbreeding depression from the F1 to the F2 generation suggested that epistatic gene action also contributed to the expression of grain yield. Summary Resistance to septoria nodorum blotch in Triticum monococcum, T. tauschii, T. timopheevii, T. dicoccum and T. durum was evaluated on plants at the three-leaf stage in greenhouse tests. A high frequency of resistant genotypes was found in T. monococcum, T. tauschii and T. timopheevii, but not in T. dicoccum and T. durum. The resistance of F1 plants of crosses of resistant T. monococcum (PI 289599) and T. timopheevii (PI 290518) accessions with susceptible common wheat cv. Park and durum wheat cv. Wakooma, respectively, was evaluated on the basis of percentage leaf necrosis, lesion number, lesion size and incubation period. No dominance was found for long incubation period, but various dominance relationships occurred for low percentage leaf necrosis, low lesion number and small lesion size, depending on the cross. Multiple regression analysis showed that lesion number contributed more to percentage leaf necrosis than lesion size or incubation period. Resistance to septoria nodorum blotch was transferred successfully from T. timopheevii to cultivated durum wheat. Resistant BC1F7 lines, recovered from the T. timopheevii (PI 290518) × Wakooma cross, showed normal chromosome behaviour at meiosis (14 bivalents) and were self-fertile. However, an effective level of resistance was not recovered in lines derived from the other interspecific crosses.  相似文献

Wild relatives of the cultivated tomato (Solanum lycopersicum L.) are major sources of new genetic diversity for tomato improvement. Introgression lines (IL) are near-isogenic lines homozygous for one or several mapped wild DNA fragments in a common recurrent parent. A set of ILs developed by Cornell University from Solanum habrochaites accession LA1777 and recurrent parent E6203 has been made publicly available through the Tomato Genetics Resource Center of the University of California-Davis. Our objective was to identify LA1777 introgressions with potential to increase the marketable fruit yield of tomato grown in the tropics. A subset of ILs were each crossed to CLN2498E (resistant to bacterial wilt and some begomoviruses) to create IL hybrids (ILH). ILH, IL recurrent parent E6203, CLN2498E, and CLN2498E × E6203 (Hchk) were evaluated in replicated trials in Thailand, India, and Taiwan during two dry seasons. Highly significant effects for marketable fruit yield were detected in Thailand and Taiwan. ILH heterozygous for S. habrochaites segments at the bottom of chromosome 1 yielded about 20% than the Hchk at Thailand and Taiwan. Our results agree with previous results from Cornell University researchers who found a segment of S. habrochaites DNA located between TG158 and TG27 associated with increased total fruit yield in previous trials conducted in upper state New York. Yield improvement due to this S. habrochaites introgression can occur over a wide range of environments.  相似文献

Wheat is one of the major food crops in the world. It is Australia’s largest crop and most important agricultural commodity. In Australia the crop is grown under rainfed conditions with inherently important regional environmental differences; wheat growing areas are characterized by winter dominant rainfall in southern and western Australia and summer rainfall in northern Australia. Maximizing yield potential across these diverse regions is dependent upon managing, either genetically or agronomically, those factors in the environment that limit yield. The potential of synthetic backcross lines (SBLs) to increase yield in the diverse agroecological zones of Australia was investigated. Significant yield advantages were found for many of the SBLs across diverse environments. Depending on the environment, the yield of the SBLs ranged from 8% to 30% higher than the best local check in Australia. Apart from adaptation to semiarid water stressed conditions, some SBLs were also found to be significantly higher yielding under more optimal (irrigated) conditions. The four testing environments were classified into two groups, with the northern and southern environments being in separate groups. An elite group of SBLs was identified that exhibited broad adaptation across all diverse Australian environments included in this study. Other SBLs showed specific adaptation to either northern or southern Australia. This study showed that SBLs are likely to provide breeders with the opportunity to significantly improve wheat yield beyond what was previously possible in a number of diverse production environments.  相似文献

Resistance to stripe rust (caused by Puccinia striiformis Westend.) of 34 Triticum turgidum L. var.durum, 278 T. tauschii, and 267 synthetic hexaploid wheats (T. turgidum x T. tauschii) was evaluated at the seedling stage in the greenhouse and at the adult-plant stage at two field locations. Mexican pathotype 14E14 was used in all studies. Seedling resistance, expressed as low infection type, was present in all three species. One hundred and twenty-eight (46%) accessions of T. tauschii, 8 (23%) of T. turgidum and 31 (12%) of synthetic hexaploid wheats were highly resistant as seedlings. In the field tests, resistance was evaluated by estimating area under disease progress curve (AUDPC). Synthetic hexaploid wheats showed a wide range of variability for disease responses in both greenhouse and field tests, indicating the presence of a number of genes for resistance. In general, genotypes with seedling resistance were also found to be resistant as adult plants. Genotypes, which were susceptible or intermediate as seedlings but resistant as adult plants, were present in both T. turgidum and the synthetic hexaploids. Resistances from either T. turgidum or T. tauschii or both were identified in the synthetic hexaploids in this study. These new sources of resistance could be incorporated into cultivated hexaploid wheats to increase the existing gene pool of resistance to stripe rust.  相似文献

Summary Wheat (Triticum aestivum L.) cultivars grown in the southern Great Plains of the U.S.A. are exposed to a wide range of moisture conditions due to large fluctuations in the amount and frequency of rainfall. Yield stability under those conditions is therefore a desirable trait for wheat breeders. Our primary objective was to quantify various genetic parameters for grain production in drought-stressed and irrigated environments. We also attempted to predict and measure yield responses when selection is practiced in either drought-stressed or irrigated environments, or both. Seventy F2-derived lines from the cross, TAM W-101/Sturdy, were evaluated at Goodwell, OK, under irrigated and naturally drought-stressed conditions in 1987 and 1988. Genetic variance and heritability estimates were higher in the irrigated environment than in the drought-stressed environment. The genetic correlation coefficient for yields in the two environments was 0.20±0.16, indicating that selection of widely adapted genotypes requires testing in both environments. Based on the genetic variance/covariance structure of this particular population, the linear index which maximized the combined expected gain in both environments was 0.66Y1 + 0.34Y2, in which Y1 and Y2 are yields in the irrigated and drought-stressed environments. This index is not expected to apply across all populations; rather, it further supports the hypothesis that testing in either environment alone (drought stressed or irrigated) may not be most effective for increasing either mean productivity or yield under drought stress.  相似文献

Translocated chromosomes T1BL⋅1RS and T1AL⋅1RS have been widely used in many wheat (Triticum aestivum L.) breeding programs to develop high yielding cultivars. The objective of this study was to evaluate the heterotic effects of T1BL⋅1RS + T1AL⋅1RS, T1BL⋅1RS, and T1AL⋅1RS on yield and yield components of hybrid wheat grown under adequate moisture regimes. Thirteen hybrid wheats and seven parents with different chromosome constitutions relative to T1AL⋅1RS and T1BL⋅1RS were evaluated in a randomized complete block design. Variable performance was observed among the hybrids tested. Two of the three hybrids with T1BL⋅1RS + T1AL⋅1RS, produced 25.26% and 44.64% more grain than the hybrids with only T1BL⋅1RS. This was due to increased biomass, harvest index (HI) and spike density. However, the combination of these two translocations resulted in reduced kernels/spike, spikelets/spike and spike length compared to the T1BL⋅1R Stranslocation alone. When comparing closely related parents, the parent with T1AL⋅1RS produced 23.51% more grain yield than the non-translocated parent. The presence of T1AL⋅1RS resulted in 10.37% heterotic advantage for yield due to increased biomass, KW, and spike density. When the two wheat-rye translocated chromosomes are present in the same hybrid, T1AL⋅1RS seems to have a positive effect on yield through spike density and HI, but masks the effects of T1BL⋅1RS for some agronomic traits. Septoria nodorum blotch is the most important leaf disease of wheat in Western Australia. A potentially useful source of resistance has been identified in an accession of Aegilops tauschii. To study the genetics of resistance of this source a cross was made between the resistant Ae. tauschii accession, RL5271, and a susceptible accession, CPI110889. The resistant parent took significantly longer to develop symptoms, developed significantly fewer lesions and expressed significantly lower levels of disease than the susceptible parent. The F1 mean response for disease severity indicated there was no complete dominance. The F3 families were classified using three approaches. In the first approach the individual F3 plant response was used to classify the F3 families. In the second approach the F3 family means and standard errors were used to classify the F3 families. In the final approach Best Linear Unbiased Predictors of disease score and standard error for each F3 family derived from a REML analysis were used to classify the F3 families. The genotypic ratios generated by each of the approaches suggested that resistance is controlled by a single gene. The effectiveness of the resistance and its simple genetic control in the Ae. tauschii, accession RL5271 may be a useful resistance source for use in a bread wheat breeding program. Cytology and gene expression of an amphiploid between Aegilops tauschiiL., native to China, and Secale silvestre L. were studied to reveal the genomic interaction between the donor species. High frequencies of aneuploids were observed in the progenies of the amphiploid, indicating its cytological instability. Feulgen staining and Giemsa-C banding showed that only the nucleolar organizing region from chromosome 5D of Ae. tauschii existed in the amphiploid (2n = 28). The nucleolus of S. silvestre was not observed. Endosperm storage protein electrophoresis indicated most gliadin and glutenin genes from both parents were expressed in the endosperm of the amphiploid. When inoculated by wheat stripe rust and powdery mildew isolates,the amphiploid did not express the resistance from its Secale parent,suggesting the presence of disease resistance suppressor(s) in the D genome of Ae. tauschii as well as nucleolar organizer suppressors. The main goal of this work was to introduce resistance genes for rust, caused by Uromyces appendiculatus, and anthracnose, caused by Colletotrichum lindemuthianum, in an adapted common bean cultivar through marker-assisted backcrossing. DNA fingerprinting was used to select plants genetically closer to the recurrent parent which were also resistant to rust and to race 89 of C. lindemuthianum. DNA samples extracted from the resistant parent (cv. Ouro Negro), the recurrent parent (cv. Rudá), and from BC1, BC2 and BC3 resistant plants were amplified by the RAPD technique. The relative genetic distances in relation to the recurrent parent varied between 9 and 59% for BC1, 7 and 33% for BC2, and 0 and 7% for BC3 resistant plants. After only three backcrosses, five lines resistant to rust and anthracnose with, approximately, 0% genetic distance in relation to the recurrent parent were obtained. These lines underwent field yield tests in two consecutive growing seasons and three of them presented a good yield performance, surpassing in that sense their parents and most of the reference cultivars tested.  相似文献

Summary Interspecific hybridisation has been suggested to improve the resistance of crops to biotic and abiotic stresses. To identify sources of resistance to drought which could be used in a breeding programme, an assessment of a large number of wheat species was carried out. The evaluation was based on the estimation of a drought susceptibility index and stability in total dry matter and grain yields of various species grown across a range of soil moisture conditions. On the basis of stability in total dry matter yield, T. aestivum was relatively more drought resistant. However, on the basis of grain yield stability, T. sphaerococcum, T. vavilovii and T. aestivum cv. C 306 exhibited better yield stability and drought resistance than the other species tested. It is suggested that the above species could be utilized as parental material in a hybridisation programme aimed at improving the drought resistance of wheat.  相似文献

Synthetic hexaploid wheats are of interest to wheat breeding programs, especially for introducing new genes that confer resistance to biotic and abiotic stresses. A group of 54 synthetic hexaploid wheats derived from crosses between emmer wheat(Triticum dicoccum, source of the A and B genomes) and goat grass (Aegilops tauschii, D genome donor) were investigated for genetic diversity. Using the AFLP technique, dendrograms revealed clear grouping according to geographical origin for the T. dicoccum parents but no clear groups for the Ae. tauschii parents. The geographical clustering of the T. dicoccum parents was also reflected in the dendrogram of their derived synthetic hexaploids. Diversity of the T. dicoccum parents and their derived synthetic hexaploids was further evaluated by measuring 18morphological and agronomic traits on the plants. Clustering based on morphological and agronomic data also reflected geographical origin. However, comparison of genetic distances obtained from AFLP and agronomic data showed no correlation between the two diversity measurements. Nevertheless, similarities among major clusters with the two systems could be identified. Based on percentage of polymorphic markers, the synthetic hexaploids had a considerably higher level of AFLP diversity (39%) than normally observed in cultivated hexaploid wheat (12–21%). This suggests that synthetic hexaploid wheats can be used to introduce new genetic diversity into the bread wheat gene pool. Summary Aegilops tauschii (Coss.) Schmal. (2n = 2x = 14, DD), a wild relative of wheat has been considered to be a valuable source of variation for improvement of cultivated wheats. However, undesirable genes can be incorporated into the cultivated varieties from wild relatives. The spontaneous spike shattering caused by the brittle rachis character is of adaptive value in wild grass species, but not in cultivated varieties. The rachis of R-61, which was derived from the cross of T. aestivum cv. Bet Hashita with an accession of Ae. tauschii, was brittle. Using telosomic stocks, the brittle rachis gene Br 61 (tentatively designated) of B-61 was located on the short arm of chromosome 3D and the distance of Br 61 to the centromere was 31.9 cM. The distance of Br 61 from the centromeric marker Xgdm72 was 25.3 cM on the short arm of chromosome 3D. The location of Br 61 was similar to Br 1 whose location was determined by telosomic mapping and microsatellite mapping. Discrepancy of disarticulation type was found between R-61 and Aegilops tauschii suggesting that the recombination around the regions of Br 1 locus and Br t locus created the wedge type disarticulation of R-61.  相似文献

Summary Many conventional hard red spring wheat (Triticum aestivum L. em Thell) lines, including several North Dakota cultivars, carry a gene (or genes) which restore partial male fertility to male sterile plants with Triticum timopheevi Zhuk. cytoplasm. Since this gene has no fertility restoration function in T. aestivum cytoplasm, the postulation can be made that it is being retained in conventional lines because of pleiotropic effects, favorable linkages or chance. The research reported in this paper examined these possibilities. Forty F6 lines, derived from a single F2 plant which was heterozygous for a gene (or genes) for partial fertility restoration, were evaluated for two years in a yield trial planted at Fargo, North Dakota. The 40 lines were testcrossed to a male sterile line having T. timopheevi cytoplasm, and the mean seed set of testcrosses was used as a measure of a line's fertility restoration potential. Twenty-seven lines had the gene for partial fertility, and 13 lines apparently lacked this gene. The 40 lines differed for heading date, anther extrusion, plant height, grain yield, 200-kernel weight, test weight, and grain protein percentage. However, comparisons of lines having the restorer gene with those lacking the gene did not provide any obvious explanation for the retention of the partial fertility restorer gene in the breeding stocks of the North Dakota conventional hard red spring wheat breeding program. The possibility that the restorer gene was linked with genes for resistance to stem rust or leaf rust also was evaluated by testing lines for their reaction to several races of rust. Summary Fifty-five accessions of Triticum species were analyzed for content of hydroxamic acids (Hx), a natural resistance factor against various organisms. Hx were found in all accessions analyzed. Extreme values were found in wild diploid species: highest in T. speltoides (16.0 mmol/kg fr. wt) and lowest in T. tauschii (0.21). Modern polyploid wheats sharing the same genome did not show substantial variations in Hx levels. The data suggest possible sources of high Hx levels for wheat breeding programs.  相似文献

Fusarium head blight (FHB), caused by Fusarium graminearum and Fusarium culmorum, is a devastating disease in cereals. This study was undertaken to estimate progeny means and variances in each of five winter triticale and winter wheat crosses using unselected F2−derived lines in F4 or F5 generation bulked at harvest of the previous generation. Fifty (triticale) and 95 (wheat) progeny per cross were inoculated in two (triticale) or three (wheat) field environments. FHB rating was assessed on a whole-plot basis. Mean disease severities of the parents ranged from 2.3 to 6.4 in triticale and from 3.1 to 6.5 in wheat on a 1-to-9 scale (1 = symptomless, 9 = 100% infected). The midparent values generally resembled the means of their derived progeny. Significant (P < 0.01) genotypic variance was detected within each cross, but genotype × environment interaction and error variances were also high for both crops. Medium to high entry-mean heritabilities (0.6–0.8) underline the feasibility of selecting F2-derived bulks on a plot basis in several environments. Phenotypic correlation of FHB resistance between generation F2:4 and F2:5 was r = 0.87 (P < 0.01) tested across 150 wheat bulks at two locations. Our estimates of selection gain are encouraging for breeders to improve FHB resistance in triticale and wheat by recurrent selection within adapted materials.  相似文献

In order to test if selection can improve a population's adaptation to diverse environments simultaneously, three cycles of recurrent selection based on grain yield in Iowa, Idaho, and Norway were practiced in an oat (Avena sativaL.) population developed from North American, Scandinavian, and wild species (A. sterilis L.) germplasm sources. Specific objectives were to determine if selection: increased mean yields across environments and within all environments; changed the genetic correlation of yields in different environments; and changed genetic variation for yield within the population. We evaluated 100 to 210 randomly-chosen families from each cycle of selection at three Iowa locations, in Idaho, and in Norway for two years. Grain yield within each location and mean yields across locations increased significantly over cycles of selection. Mean yields across locations expressed as a percent of the original population mean increased at a rate of 2.6% per year. Several families from the third cycle population exhibited both high mean yields across locations and consistently high yields within all locations. Average genetic correlations of yield in different environments were higher in the second cycle than in the original population. A trend of reduced genetic variation and heritability was observed in Iowa only. These results suggest that we successfully improved mean population yield both within and across locations, and yield stability across environments, and in developing families with outstanding adaptation to diverse environments.
