Domesticated and wild-type tepary beans (Phaseolus acutifolius A. Gray) were grown with or without inoculation with rhizobia in pots under bacteriologically controlled conditions in a temperature-controlled glasshouse. Seeds were inoculated with a mixture of seven strains isolated from nodules collected from domesticated field-grown tepary bean in Arizona, USA, or with a commercial inoculant strain for Phaseolus vulgaris (CC511). Different degrees of plant reliance upon N2 fixation for growth were generated by supplying the inoculated plants throughout growth with nutrients containing a range of concentrations of 15N-labeled NO3 (0, 1, 2, 5 or 10 mM). An uninoculated treatment that received 10 mM 15N-labeled NO3 was included to provide data for plants solely dependent upon NO3 for growth. Six weeks after sowing, shoots were harvested for dry matter determination and subsequent 15N analysis, root-bleeding xylem sap was collected, and nodulation assessed. With regard to shoot biomass production, domesticated lines were more responsive to inoculation, but less responsive to applied N than wild types. All inoculated plants were nodulated, but the field isolates from tepary bean were more effective in N2 fixation than strain CC511. It was concluded that tepary bean requires a specific inoculant to benefit from fixation of atmospheric N2. Xylem sap samples were analysed for ureides (allantoin and allantoic acid), amino acid content (α-amino-N), and NO3 concentration. The amount of ureide-N present in xylem sap was expressed as a percentage of total solute N, described as the relative abundance of ureide-N (RUN), for each N treatment and was compared to the proportion of plant N derived from N2 fixation (%Ndfa) calculated using a 15N dilution technique. The RUN values ranged from 8% for saps collected from uninoculated plants provided with 10 mM NO3 in the nutrient solution (%Ndfa=0) to 86-91% for nodulated plants grown in the absence of externally supplied NO3 (%Ndfa=100). These data indicated that ureides were the principal product of N2 fixation exported from the nodules to the shoot in xylem sap. Since RUN values were closely related to %Ndfa, it was proposed that N-solute analysis of xylem sap could provide a valuable analytical tool to monitor the symbiotic performance of tepary bean.  相似文献

The genetic relationships among six strains of rhizobia, including three strains of Rhizobium fredii and three strains of Bradyrhizobium japonicum, was determined using random amplified polymorphic DNA (RAPD) technique. In this study, 46 arbitrary 10mer primers were employed for RAPD, generating a total of 251 informative fragments. A dendrogram of phylogenetic relationships among the six strains was constructed. The results indicated that geographical distribution may affect phylogeny, as there were closer relationships among the four Taiwanese strains, SB138, SB562, SB368 and SB651, than between these strains and USDA192, which originated from mainland China. The strain USDA110, obtained from the United States, was used in the parsimony analysis. The greatest similarity (55.6%), existed between two strains of B. japonicum, SB562 and SB138, which both, and the lowest R. fredii (44.4%) between two strains of R. fredii, SB368 and USDA192. We also found a RAPD marker specific to the four Taiwanese SB strains used in the study. There is little information available about soybean aphid (Aphis glycines Matsumura) effects on the physiology and mineral nutrition of soybean (Glycine max [L.] merr.). Controlled-environment studies were conducted to measure soybean aphid infestation effects on dry weight, nitrogen (N) fixation, ureide-N, and nitrate-N concentration and accumulation. Plants grown in perlite using –N nutrient solution culture were infested at the 3rd trifoliolate (V3) stage and measured for N fixation, nodule characteristics, and ureide-N concentration at the full pod (R4) stage. When compared to uninfested control plants, aphid infestation reduced total nodule volume per plant by 34%, nodule leghemoglobin per plant by 31%, plant N fixation rate by 80% and shoot ureide-N concentration by 20%. Soil-grown plants were infested at the first trifoliolate (V1) stage and shoots were measured for dry weight, nitrate-N, and ureide-N at the full bloom (R2) stage. Infestation reduced shoot dry weight by 63%, increased nitrate-N concentration by 75%, but did not significantly affect ureide-N concentration. Because nutrient concentration is a single-point measurement that results from the integration of two dynamic processes, nutrient accumulation and dry matter production, we conclude that aphid-induced reductions in N fixation, coupled with decreased dry weight accumulation, caused shoot ureide-N concentration to remain unchanged in aphid-injured plants when compared to uninfested plants. Because nitrate-N concentration was greater in aphid-damaged shoot tissue, we further conclude that nitrate-N accumulation was less sensitive to aphid injury than dry weight accumulation.  相似文献

Soybean (Glycine max L. Merr.) cultivars Akisirome and Peking were inoculated with Bradyrhizobium japonicum Is-21 and Rhizobium fredii USDA 194, respectively, and were grown in cylindrical pots containing sterilized vermiculite which were aerated with CO2-free air or ambient air to study the effects of CO2 deficiency in the rhizosphere on plant growth, nodulation, and nitrogen fixation. The repressive effects of CO2-free air treatment were more conspicuous in Peking than Akisirome, and nodule number, nodule mass, amount of biologically fixed N and plant growth of Peking were reduced remarkably by the CO2-free air treatment.

Acetylene reduction activity (ARA) of Peking inoculated with USDA 194 and Akisirome inoculated Is-21 was assayed in the absence and presence of CO2, ARA of Akisirome was not affected by the absence of CO2, while that of Peking decreased drastically.

Based on these results, it was concluded that nitrogen fixation by Akisirome inoculated with B. japonicum Is-21 was not appreciably influenced by CO2 deficiency in the rhizosphere, while that of Peking inoculated with R. fredii USDA 194 was severely repressed, and the decrease was estimated to be due to both the reduction of the nodule mass and specific nitrogen-fixing activity.  相似文献

Abstract

Cobb and Coker 488, late‐season (maturity group VIII) cultivars of soybean [Glycine max(L.) Merr], were grovn under irrigated and non‐irrigated conditions on a Norfolk loamy sand in a two‐year field experiment. Each cultivar was inoculated withBradyrhizobium japonicumstrains [USDA 3I1b110; Brazil 587; NifTAL 184 and 102 (NifTAL cultures of Brazil 587 and USDA 110, respectively); and North Carolina 1001, 1004, 1005, 1010, and 1029). Drought conditions were present both years, and irrigation significantly increased the overall yield (2.49 vs 1.92 Mg ha‐1). Coker 488 was significantly higher in seed yield than Cobb (2.55 vs 2.02 Mg ha‐1). Strain ofB.japonicumalso affected seed yields. NC1010‐inoculated soybean was significantly higher in seed yield rank than all other soybean at the P<0.01 level, when compared by single degree of freedom contrast (sdfc). The yield ranking of soybean inoculated with NC1001 was significantly lower than soybean inoculated with all other strains, when compared by sdfc (P<0.10). Other strains differed in responses which ranged from good to poor inoculants under specific water management conditions. For instance, under nonirrigated conditions, soybean inoculated with strains ofB.japonicumfrom North Carolina was significantly higher in seed yield than those inoculated with the cultures of USDA 110, B587, or the control, when compared by sdfc (P>0.03, 0.05, 0.06, respectively). Since soybean inoculated with either strain of USDA 110 was generally high in yield rank under irrigated conditions, their response to irrigation was large relative to soybean inoculated with the NC strain (P<0.04). Neither seed nitrogen nor xylem water potential was highly correlated to seed yield. Since seed yield and N content were not highly correlated, the amount of N accumulated in soybean dry mass and that removed in seed were not highly correlated. Sulfur (S) and Nitrogen (N) metabolisms in plants are interacted and it is known that S deficiency decrease N absorption and metabolism. In leguminous plants S deficiency also decreases N2 fixation by rhizobia in the nodules. Deep placement of a controlled-release N fertilizer is a good method to provide nitrogen to soybean without inhibiting N2 fixation; thus, it was hypothesized that this method is able to provide nitrogen effectively to sulfur-deficient soybean plants. In this study effects of deep placement of coated urea on S-N physicological interaction, growth and productivity in soybean plants were examined using pot experiments. Soybean plants were grown with sulfate concentrations of 30, 100, or 1000 μM, with or without deep placement of coated urea. Shoot weights at the developing stage were not affected by S deficiency. SPAD values of leaves during the flowering stage decreased with S deficiency and increased with the deep placement of coated urea. S deficiency decreased seed weight per plant at the harvesting stage, but this decrease was attenuated by the deep placement of coated urea. N and S content in shoots at the developing stage increased with the deep placement of coated urea, whereas in seeds, only the N content increased. N2 fixation activity based on the relative ureide-N content in xylem sap indicated that the deep placement of coated urea did not inhibit N2 fixation activity at the early flowering stage. Without deep placement of coated urea, the relative ureide-N content decreased under S deficiency at the seed filling stage. These results suggest that the deep placement of coated urea is an efficient method to supply N to support soybean yield under S deficiency.

We studied the effect of the soil physical properties on soybean nodulation and N2 fixation in the heavy soil of an upland field (UF) and an upland field converted from a paddy field (UCPF) in the Hachirougata polder, Japan. Seeds of the soybean cultivar Ryuho were sown in each field with or without inoculation of Bradyrhizobium japonicum A1017. The soybean plants were sampled at 35 (V3) and 65 (Rl) d after sowing (DAS), and then nodulation and the percentage of N derived from N2 fixation in the xylem sap were determined. The soil physical properties were different between UF and UCPF, especially the air permeability and soil water regime. Nodule growth was restricted in UCPF irrespective of rhizobial inoculation, though rhizobial infection was not inhibited by the unfavorable soil physical conditions. Soybean plant growth was closely related to the nodule mass and N2 fixation activity, and the inoculation of a superior rhizobium strain was effective only at 35 DAS. These results indicate that soybean nodulation and N2 fixation was considerably affected by the physical properties of heavy soil, and that it is important to maintain the N2 fixation activity and inoculate the soybean plants with a superior rhizobium strain at a later growth stage in order to increase soybean production in heavy soil fields.  相似文献

Summary Bacteria isolated from the root zones of field-grown soybean plants [Glycine max (L.) Merr.] were examined in a series of glasshouse experiments for an ability to affect nodulation competition among three strains of Bradyrhizobium japonicum (USDA 31, USDA 110, and USDA 123). Inocula applied at planting contained competing strains of B. japonicum with or without one of eleven isolates of rhizosphere bacteria. Tap-root nodules were harvested 28 days after planting, and nodule occupancies were determined for the bradyrhizobia strains originally applied. Under conditions of low iron availability, five isolates (four Pseudomonas spp. plus one Serratia sp.) caused significant changes in nodule occupancy relative to the corresponding control which was not inoculated with rhizosphere bacteria. During subsequent glasshouse experiments designed to verify and further characterize these effects, three fluorescent Pseudomonas spp. consistently altered nodulation competition among certain combinations of bradyrhizobia strains when the rooting medium did not contain added iron. This alteration typically reflected enhanced nodulation by USDA 110. Two of these isolates produced similar, although less pronounced, effects when ferric hydroxide was added to the rooting medium. The results suggest that certain rhizosphere bacteria, particularly fluorescent Pseudomonas spp., can affect nodulation competition among strains of R. japonicum. Summary Differences between isogenic uptake hydrogenase (HUP) mutants of Bradyrhizobium japonicum in terms of nodule efficiency, N2 fixation and N incorporation into various plant parts were studied in a monoxenic greenhouse experiment in order to confirm previous results with soybeans and beans inoculated with various HUP+ and HUP strains. The HUP+ revertant PJ17-1 of a HUP mutant (PJ17) of strain USDA DES 122 showed a completely restored relative efficiency (100% versus 78±2% for the HUP mutant), higher nodule efficiency (N2 fixed per g nodules), higher ureide-N transport rates, higher N contents in pods and higher N harvest indices. All these observations confirm previous experiments with HUP+ and HUP strains.  相似文献

Attempts have long been made to study the effect of mineral nutrition on the metabolic substances in excised roots from a numcer of plant species, but very little attention has teen given to an approach to the problem by using the bleeding sap from crop plants. Recently, however, an increasing number of reports about the occurrence of organic substances in bleeding sap from crop plants has completely revised an old view that xylem sap was essentially a rather dilute aqueous solution of inorganic salts. Evidence has already been obtained which suggests a significant role for the root system as a centre of metabolism and an upward transport of metabolites from the root via the xylem to the leaf. Although analyses of the nitrogenous compounds present in bleeding sap from herbaceous plants have shown glutamine and asparagine to be the most important constituents, in some species of plants nitrate nitrogen may be a predominant nitrogenous compound. Besides ami des and nitrate, the presence of amino acids in bleeding sap has ceen shown by Kulayeva, Silina, and Kursanov 1) for pumpkins, Wieringa and Bakhuis 21 for Lupins, and DIE3) for cucumbers and tomatoes.  相似文献

Graminaceous plants can take up iron-phytosiderophore complexes, whereas non-graminaceous plants absorb ferrous ions after the reduction of ferric compounds at the root cell membranes. The iron (Fe) in the roots may be transported to the aerial plant parts through the xylem. We compared the chemical forms in xylem sap collected from the cut stems of three graminaceous plants (rice [Oryza sativa L.], maize [Zea mays L.], barley [Hordenum vulgare L.]) and three non-graminaceous plants (tomato [Lycopersicon esculentum Mill.], soybean [Glycine max Merr.], castor bean [Ricinus communis L.]) grown in composite soils for the concentrations of iron and iron-chelating compounds (nicotianamine, phytosiderophores, citrate). We also fractionated the xylem saps by size-exclusion chromatography to gain insight into the chemical forms of iron. The Fe concentrations in the xylem sap ranged from 9 to 40 μM. Nicotianamine was found in the xylem sap from all the plants examined, with higher concentrations in the non-graminaceous plants. In contrast, phytosiderophores (2’-deoxymugineic acid and mugineic acid) were predominantly detected in the graminaceous plants. The concentrations of free citrate varied greatly (from 4 to 2200 μM) among the six plant species. The xylem sap iron in non-graminaceous plants may form two types of Fe-citrate, whereas in graminaceous plants, the bound Fe forms may be largely two types of Fe-citrate with various Fe-phytosiderophores.  相似文献

This study is aimed to examine if cotyledon node zone may play a role in salt and/or alkali tolerance. Seedlings of halophyte plant Kochia sieversiana and glycophyte plant Lycopersicon esculintum Mill were treated with salt and alkali respectively, xylem sap was collected from above or below cotyledon node zone, and components and contents of inorganic ions in the sap were examined. When compared with that collected from below cotyledon node zone, xylem sap collected from above cotyledon node zone in K. sieversiana contains less Na+ under both salt and alkali stresses, and less chloride (Cl–) under salt treatment. Both Na+ and Cl remain nearly the same in xylem sap collected from below and above cotyledon node zone in L. esculintum Mill. Cotyledon node zone in K. sieversiana selectively restricted ion transport under both salt and alkali stresses, which may represent a novel mechanism of salt and alkali resistance in halophyte plants.  相似文献

In order to select appropriate Bradyrhizobium USDA reference strains for primary grouping of indigenous soybean bradyrhizobia, we systematically constructed phylogenetic trees of 20 USDA strains based on DNA sequence analysis and PCR-restriction fragment length polymorphism (RFLP) targeted to 16S rDNA and the internal transcribed spacer (ITS) region between 16S and 23S rDNAs. The phylogenetic trees of 16S rDNA showed 3 major groups, cluster USDA 110 (USDA 62, 110, 122, 125, and 129), cluster USDA 6 (USDA 4, 6T, 38, 115, 123, 127, 135, and 3622T) and cluster B. elkanii (USDA 31, 46, 61, 76T, 94, and 130), as well as the phylogenetically independent strain USDA 124. The topology of the ITS trees was almost similar to that of 16S rDNA, although the positions of two extra-slow-growing strains, USDA 135 and USDA 3622T were variable among the ITS sequences, PCR-RFLP of the ITS region and 16S rDNA. Only two strains, USDA 110 and USDA 122, harbored hup genes and they fell into the USDA 110 cluster. These results suggest that PCR-RFLP analysis of 16S rDNA and the 16S-23S rDNA ITS region may be useful for the grouping of bradyrhizobia and for the first screening of hup-positive strains. Based on the above results, we propose a minimum set of USDA strains reflecting Bradyrhizobium diversity that includes B. japonicum USDA 6T, B. japonicum USDA 110, B. japonicum USDA 124, and B. elkanii USDA 76T. In addition, an extra-slow-growing strain with the serotype USDA 135 might be necessary for genomic diversity analysis of bradyrhizobia, because their phylogenetic positions were variable.  相似文献

The 15N natural abundance technique is one of those most easily applied ‘on farm’ to evaluate the contribution of biological N2 fixation (BNF) to legume crops. When proportional BNF inputs are high, the accuracy of this technique is highly dependent on an accurate estimate of the 15N abundance of the N derived from N2 fixation (the ‘B’ value). The objective of this study was to determine the influence of soybean variety on ‘B’ value. Plants of five soybean varieties were inoculated separately with two Bradyrhizobium strains (one Bradyrhizobium japonicum and one Bradyrhizobium elkanii) grown in pots of soil virtually free of bradyrhizobia capable of nodulating soybean. The proportion of N derived from BNF (%Ndfa) was estimated in separate pots where a small quantity of enriched 15N ammonium sulphate was added. The %Ndfa was then used with the 15N natural abundance data of the nodulated soybean and non-N2-fixing reference plants, to determine the ‘B’ value for each soybean variety/Bradyrhizobium association. The varieties nodulated by the B. japonicum strain showed significantly greater N content and %Ndfa than those nodulated by the B. elkanii strain, and in all cases the ‘B’ value of the shoot tissue (‘Bs’) was higher. The differences in ‘Bs’ values between varieties nodulated by the same Bradyrhizobium strain were insignificant, indicating that this parameter is influenced much more by the Bradyrhizobium strain than by the variety of the host plant.  相似文献

Functions of sodium (Na+) and potassium (K+) are closely associated. In some crops, Na+ is able to prevent or reduce considerably the occurrence of K+ deficiency. Sugar beet (Beta vulgaris L.) is a natrophilic crop, and positive effects of Na+ applications on yield were observed when K+ was sufficiently supplied. However, it is not known which specific function of K+ can limit the growth of sugar beet when K+ is substituted by an equivalent amount of Na+. Therefore, K+ substitution by Na+ was investigated for sugar beet in hydroponics. Surprisingly, no K+‐deficiency symptoms were observed. However, calcium (Ca2+) concentrations in the leaves were significantly decreased. Moreover, Ca2+ uptake and translocation through xylem sap were reduced in Na+‐treated plants. It is concluded that Ca2+ uptake by roots and its translocation via xylem sap primarily limit the possibility of K+ substitution by Na+ in sugar beet nutrition.  相似文献

Summary Physiological and symbiotic characteristics were identified in Rhizobium fredii isolated from subtropical-tropical soils. The generation times of R. fredii Taiwan isolated-SB 357 and -SB 682 were 1.7 and 2.5 h, respectively. These strains were associated with acid production in yeast-extract mannitol medium. They were able to use hexoses, pentose, sucrose, trehalose and raffinose. Strain SB 357 can resist a high concentration of kanamycin (100 g ml–1 and penicillin (400 g ml–1). It can tolerate up to 2.34% NaCl and 1031.3 mosmol kg–1 (23.4 bars). The growth rate of R. fredii SB 357 under the concentration of approximately 450 mosmol kg–1 (10.2 bars) was not affected by salinity, but responded to osmotic pressure. Both strains (SB 357 and SB 682) isolated from subtropical-tropical soils were able to form an effective N2-fixing symbiosis with the US soybean cv Clark lanceolate leaflet.  相似文献

The effects of calcium (Ca) deficiency on cation uptake and concentration of xylem sap from tomato roots after excision of the aerial parts, were studied. The measurements were made on tomato plants grown on nutrient solutions with +Ca or without‐Ca, over a period of 48 hours. Calcium deficiency entailed a significant increase of the flux of xylem sap between the 6th and 14th hour on the first day after excision. In spite of the lack of Ca in the nutrient solution, the Ca concentration in xylem sap was unaffected in regard to that of excised roots with +Ca. The maintenance of the Ca concentration in xylem sap of plants grown on a Ca deficient solution was related to a reuse of the Ca from the apoplastic root stores. So, this regulation indicates a possible translocation of the Ca available in the root supply and a mobility of this element out of the roots only during the early stages of exposure to a Ca deficiency. The presence of NH4 + in xylem sap with both +Ca and‐Ca treatments confirms the nitrogenous reduction activity of tomato roots. The accumulation of free ammonium 24 h after excision in both xylem saps (+Ca and‐Ca) is likely to be evidence of an alteration process of protein synthesis which is related to the depletion of the root water soluble carbohydrate supply.  相似文献

A relationship among Cu, N, and Rhizobium japonicum was hypothesized because previous research had shown that (a) 35% or more legumes in the Atlantic Coastal Plain have Cu concentrations of 6 mg kg‐1 or less, (b) Cu influences N fixation in some legumes, and (c) irrigated soybean (Glycine max L. Merr.) can accumulate most of its N through fixation. Soybean were grown on a Cu‐deficient Norfolk (fine‐loamy, siliceous, thermic Typic Paleudult) loamy sand with 3 fertilizer sources of Cu, 2 strains of R. japonicum, and with or without 336 kg ha‐1 of N fertilizer. Application of Cu significantly increased the number of pods plant‐1 suggesting pod abortion in determinate soybean may be caused by low Cu, but seed yield was not increased. Fertilization with N increased vegetative growth, but not total biomass or seed yield. Inoculation with R. japonicum strain 110 significantly increased seed yield by 0.3 Mg ha‐1 compared to strain 587. The yield increase was similar with or without fertilizer N application indicating strain response was not totally caused by improved N efficiency. There was no relationship between seed yield and nodule occupancy as measured by the ELISA technique.  相似文献

Soybean plant is characterized by a systemic autoregulatory control system of nodulation (autoregulation) by initial infection with rhizobia, and plants commonly display a systemic acquired resistance (SAR) to pathogenic microbe infection related to salicylic acid (SA) signal transmission. We investigated the effect of exogenous SA supply on soybean nodulation to determine whether SA affects the autoregulation of nodulation. Seedlings of the hypernodulating mutants NOD1-3, NOD2-4 and their parent cv. Williams were treated or not treated (control) with a 100 μmS-SA solution at 5 d before the inoculation of Bradyrhizobium japonicum strain USDA110. The nodule dry weight and the number of nodules of the wild type soybean Williams exhibiting autoregulation drastically decreased by the addition of 100 μm SA. The decrease in the nodule number was not caused by the reduction of the rhizobium number in the medium. Salicylic acid inhibited only early nodule formation and did not affect the growth of formed nodules. The inhibitory effect of SA on the nodulation of NOD1-3 and NOD2-4 was significantly less pronounced than that in Williams. These results indicate that SA is directly involved in signal transmission in the autoregulation, and that SA or the SAR induced by SA stimulates the autoregulation of nodulation in soybean.  相似文献

Summary A field experiment was condutced in a clay loam soil to study the performance of three Bradyrhizobium japonicum strains; USDA 110, USDA 138 and TAL 379, in relation to their N2-fixing potential and competitiveness on two soybean cultivars (Clark and Calland). Inoculation of soybean cultivars with these strains, either singly or in combination, induced significant increases in plant dry weight, N2 fixation and seed yields. However, no significant differences were found between the rhizobial strains and/or their mixtures in N2 fixation and increased seed yield for both cultivars. The two soybean cultivars gave similar responses to inoculation. No significant differences in seed yield were observed between Clark and Calland cultivars. The interaction between inoculant strain and soybean cultivar was not significant. The competition between strains for nodulation was assessed. Strain USDA 110 was the most competitive, followed by USDA 138. Strain TAL 379 was always less competitive on both cultivars. The incidence of double-strain occupancy of nodules varied from 8% to 40%.  相似文献
