Histological Study on Soybean Somatic Embryogenesis   总被引:4,自引:0,他引:4  
Soybean somatic cell could induce the development of embryoid which was similar to embryo morphologically and structurally. Somatic embryogenesis system of soybean was used to conduct genetic transformation of soybean because of its several advantages such as higher transformational efficiency,beetter synchronism and fewer plant chimeras among transgenic plants. After infected with agrobacterium tumefaciens,the initiation,differentiation and development of young cotyledon embryogenic cell of soybean which was cultured on selective culture medium with kanamycin were investigated through histological study. The result showed that somatic embryo was differentiated in non-bud differentiation way. The embryogenic cells were differentiated from epidermis of explant or cells in 1 layer or 2 layers,with the division of embryogenic cells and degradation and disorganization of surrounding cells,the embryogenic cells would form embryoid with analogous suspensor structure.Later,globular embryoid would extrude from epidermis then developed into heart-shape embryo.The experiment was expected to provide theoretical reference for the construction of high transformational system of using plant somatic embryogenesis induced by young cotyledon of soybean.  相似文献

In an attempt to propagate and conserve the rare, showy bulbous plants of Iris bismarckiana, newly recorded to the flora of Jordan and to contribute to the conservation the wild lris species in Jordan, a simple rapid, time consuming protocol has been developed using plant regeneration via somatic embryogenesis. Somatic embryogenesis was induced in zygotic embryo culture on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (8 mg/L) as the sole plant growth regulator, where both embryogenesis calli and somatic embryos were induced. Separation of embryos from embryo clusters was necessary to enhance the frequency of germination. Germination was stimulated by separation of embryos successfully from embryo clusters and transferred onto fresh MS medium. Data obtained were analyzed as a complete random design with three replications. Calli fragments that were transferred to embryogenesis induction medium (EIM) produced white embryo-like globular structures within two weeks. Within three more weeks, clusters of structures at various stages of development could be found on the same callus. The applied technique is rewarding and encouraging for further research on the endangered wild species of Iris in Jordan.  相似文献

The development of strawberry fruit,two cultivars-Gelila and Xiaoshi,was systematically studied by anatomical and embryological methods.The growth dynamics of fruit showed that the two cultivars performed similar changes with their fresh weight volume and structure,it included the first slow growth stage,the rapid growth stage and the second solw growth stage.The fruit was a typical polyachene with a ovule which belonged to a Poligonum type,the growth process of pollen tube from stigma to embryo sac was clearly observed by flourescence microscopy.Embryo development was similar to that of most dicotyle,and the mature embryo was orthotropous;Endosperm development was a nuclear type,and gradually disappeared with development:Seed coat consisted of 3-4 layer cells,and pericarp was composed of 6-8 layer cells which differentiated from ovary wall cell;The structure of receptacle was a typical stem which consisted of epidermis,cortex and vascular cylinder.With development,the cortex and pith of fruit contained plentiful parenchyma tissue,and they were mainly edible sections.  相似文献

[Objective] The study aimed to reveal the effect of osmosis-regulating substances and organic appendices on somatic embryogenesis in wheat. [Method] The suitable concentration combination of appendices was optimized by adding different concentrations of osmosis-regulating substances including mannitol, sorbitol and organic appendices such as Gln, CH and LH, into the somatic embryogenesis in wheat. [Result] The mannitol or sorbitol lower than 40 g/L was helpful for improving somatic embryogenesis; there was no significant difference in the induction rate of somatic embryogenesis when 300-500 mg/L Gln、CH or LH was respectively added into the induced medium, while somatic embryogenesis could be enhanced dramatically in the presence of 500 mg/L Gln together with 300 mg/L CH. [Conclusion] Somatic embryogenesis could be improved to some extent by different concentrations of osmosis-regulating substances and organic appendices, which laid foundation for establishing a more perfect system of somatic embryogenesis in wheat.  相似文献

Abstract: Somatic embryogenesis from lily bulb scales has not been studied in details, although tissue culture methods have been applied to the propagation for decades. The effects of different kinds and concentration of auxins for oriental lily somatic embryogenesis were investigated (Lilium hybrida var. Sorbonne). 2, 4-dichlorophenoxyacetic acid (2, 4-D), thidiazuron (TDZ) and α-naphthaleneacetic acid (NAA) media with benzyladenine(6-BA) and lactalbumin hydrolysate (LH) were used for embryogenic callus in the darkness. The best response on embryogenic callus formation was obtained on MS media supplemented 2, 4-D 2.0 mg·L^-1, 6-BA 0.5 mg·L^-1 and LH 300 mg·L^-1. Transfer embryogenic callus to the media with TDZ, 6-BA, kinetin (KT) supplemented 2, 4-D. The highest number of somatic embryos has been produced on medium with 0.5 mg·L^-1 2, 4-D and 0.3 mg·L^-1 KT. Germinated embryos with shoot axes were changed to MS media with 6-BA 0.5 mg·L^-1. The results suggest that in vitro culture of somatic embryogenesis from lily bulb scales can be used for plant regeneration.  相似文献

An efficient plant regeneration system was developed from the immature embryos of Triticum aestivum L. Thinopyrum intermedium alien disomic addition lines, which resistant to powdery mildew. The protocol was based on a series of experiments involving the callus induction and differentiation. The experiment studied the effects ofmature embryos. We found that the embryembryo size on callus induction and differentiation of the immature embryos. We found that the embryo size is critical for the establishment of embryogenic callus. Immature embryos (0.8 ~ 1.5 ram) showed high ability to produce embryogenie callus capable of regenerating green plants. The medium Murashige and Skoog‘ s (MS) added with 2mg/L 2, 4-diehlo-rophenoxyacetic acid (2, 4-D) gave the best embryogenic callus induction, maintenance and regeneration. The embryogenic callus maintained high regeneration during six subcultures in the callus induction medium. Suitable time ofpartialdesiccation could effectively improve the regeneration capacity of the callus cultured for 3-4 month. Bud green spot and root green spot were observed during the differentiation of callus and the difference between them was described. Regenerated shoots were rooted on half-strength MS medium containing 0.2 mg/L Naphthalene acetic acid (NAA). Plants were successfully transferred to soil and grew well. This efficient plant regeneration system provides a foundation for the study of somaclonal variation of Triticum aestivum L.- Thinopyrum intermedium alien disomic addition lines.  相似文献

The leaf development of Podocarpus macrophyllus var.maki Endl.started from the leaf primordium formed by the cell division in shoot apical meristem.During the leaf development,three resin canals from the inferior part of vascular bundles firstly began to differentiate,then protoxylem in vascular bundles differentiated from adaxial side to abaxial side.Meanwhile,a layer of parenchyma cells from the inferior part of protoderm developed into the sclerenchyma,which played an important role in supporting young leaves.When xylem became mature basically,transfusion tissue,phloem and accessory transfusion tissue began to differentiate,so mesophyll tissue finally differentiated into developmental palisade cells and sponge cells.The thicker cuticle was formed in the lateral side of epidermal cells at the same time of differentiation and development for the inner structure of leaves.Therefore,the developmental palisade cells and the cuticle in the leaf structure of Podocarpus macrophyllus var.maki Endl.is greatly significant for photosynthesis and transpiration when it grows in arid and cold regions.  相似文献

A method for the production of somatic embryos and subsequent plant regeneration for fritillaria ussuriensis M.is described.Whole leaflet explants,derived from plantlets grown in vitro,formed light yellowith embryogenic calli within one month of culture in the dark.Somatic embryogenesis was obtained after a 28d incubation on MS induction medium supplemented with 2mg/L 2,4-D 0.5mg/L BA,0.5mg/L KT and 500mg/L CH followed by transfer to a second N medium containing 0.5mg/L KT and 100mg/L CH.Somatic embryos were transferred to MS medium with 0.1mg/L NAA placed in the light for regeneration ,After two weeks,mature somatic embryo developed into whole phantlet.  相似文献

The terminal flower buds of 6-yr-old Hanfu apple were used to study the ovule development, ovular characteristics, cell death of abortive ovules, and dynamic change of starch grain quantity in the embryo sac with paraffin slices and terminal deoxynucleotidyl transferase-mediated fluorescein deoxyuridine triphosphate nick-end labeling (TUNEL) system. Four anatropous ovules in each ventricle could be observed before flowering. With the developing of floral organ, the bulk of normal ovules enlarged in each ventricle, the mature embryo sac differentiated into nucellus, and the egg cell developed into zygote by double fertilization. A large number of starch grains were observed during pollen tube growth and double fertilization, which guaranteed basic nutrient supply in the normal development of ovules. Moreover, abortion phenomenon of runtish ovules emerged at the stages of mature embryo sac, double fertilization and zygote development. The abortion characteristics included deformity of ovule development, degradation of nucellus tissue, separation between funiculus and ovule, abnormality of four-nucleate embryo sac, as well as development interruption of mature embryo sac. TUNEL analysis proved that ovule abortion was programmed cell death.  相似文献

The fertilization and development process of embryo and endosperm of strawberry,Gelila snd Xiaoshi which were broadly cultivated in Harbin were investigated by optical microscopy, fluorescence microscopy and transmission electron microscopy.The papilla cells ranked in grid orid on its stigma and the flourishing transmitting tissue appeared in the center of its thin and long pistillary.There was only one crassinucellate hemianatropous ovule with Polygonum type embryo sac and single integument in each ovary.The germination of pollen and the pathway of pollen tube growth could be clearly observed with fluorescence microscopy:stigma→transmitting tissue in pistillary→intine of ovary placenta→the surface of ovule→micropyle.The development of embryo was consistent with most dicotyledons:zygote→2-cell proembryo→proembryo shaped like a ball→proembryo shaped like a heart→proembryo shaped like a torpedo→proembryo formed two cotyledons.The embryo was orthotropous and its development was asterad type.The endosperm development beloged to nuclear type as following:primary endosperm nuclcus→free endosperm nucleus→endosperm cell →endosperm assimilated gradually by embryo.There was a special struc-ture around it during the development of Gelila embryo.  相似文献

为深入研究枣体细胞胚胎发生的形态建成过程,以小梨枣幼胚为材料,采用苏木精-伊红法染色和石蜡切片技术对体细胞胚发生、发育进行了组织学观察。结果表明:胚型愈伤组织细胞体积小,细胞核大,细胞质浓,细胞排列紧密;而非胚性愈伤组织细胞体积大,细胞核小,细胞质稀薄。体细胞胚产生于幼胚愈伤组织的表层细胞或内部细胞,且发生不同步,其发育过程与合子胚发育过程相似,即经历球形期、心形期和鱼雷期,最终形成子叶形胚。  相似文献

通过对紫花苜蓿( Medicago sativa L.)外植体愈伤组织进行诱导及对体细胞胚胎发生过程的组织学观察,得出以下结果:1)外植体在 M S附加1.0m g/ L6- B A 和10.0m g/ L I A A 的培养基上愈伤组织生长良好,而愈伤组织转入 M S附加500m g/ L水解酪蛋白的培养基中胚状体的分化能力较强。2)紫花苜蓿的体细胞胚大多是由单个具胚状体发生能力的胚性细胞发育而来,发育过程与合子胚的发育过程相似。在早期原胚发育过程中胚体外一直包围有厚壁。原胚时期和子叶胚时期出现两次淀粉积累现象。  相似文献

为了探寻光照对茶树体细胞植株再生的影响效果,以茶树全胚,具有子叶柄的子叶为材料,研究500,1500lx,黑暗3种光照条件对植株再生的影响。结果表明,胚状体主要发生在子叶柄了子叶片的隆起处,子叶片隆起发生率随光照度增加而降低,子叶发生隆起的子叶培养物在500lx光照度下培养,子叶片的胚状体分化率较高,子叶柄的胚状体,不定芽分化随光照度下降而下降;连续在黑暗下培养,子叶片的畸形胚发生率高,且数量多。  相似文献

研究了4种外植体、10种不同浓度激素组合对薇甘菊(Mikania micrantha H.B.K.)离体再生的影响,并对胚性愈伤组织中的体细胞胚发生发育过程进行了组织细胞学观察.结果表明,所用不同浓度激素组合的培养基均能诱导薇甘菊外植体产生愈伤,培养基以MS+6BA 1 mg/L为最宜;不同种类的愈伤组织再分化结果有明显差异,生长良好的胚性愈伤组织转入分化培养基后可诱导体细胞胚的发生.薇甘菊体细胞胚胎发生的形式为单细胞内起源.体细胞胚起源于胚性愈伤组织的胚性细胞,胚性细胞经过一次不均等分裂产生两个细胞,即胚细胞和胚柄细胞.然后依次经过具胚柄的多细胞原胚,再经过球形胚、心形胚、鱼雷胚阶段,发育成具有子叶的成熟胚状体,然后长成为再生植株.  相似文献

选择我国棉花优良品种泗棉3号进行组织培养,获得了具有高频胚胎发生的愈伤组织、胚状体和小植株。在附加ZT的MS培养基上,泗棉3号的下胚轴、子叶、根均诱导获得了愈伤组织,但其形成率和形成量不同,其中下胚较易诱导获得愈伤组织,根和子叶较差。ZT对愈伤组织形成和生长影响较大,其在愈伤组织诱导中较适的使用浓度为3.0或5.0mg/L。  相似文献

以陆地棉体细胞培养获得的胚性愈伤、体细胞胚和再生植株的根尖为材料,用饱和对氯二苯预处理,固定、酸解、染色,进行染色体制片,观察分析棉花体细胞培养中染色体数目的变异。结果表明:继代培养14 d的胚性愈伤组织在饱和对二氯苯处理3.5 h后制成的染色体涂片具有很好的观察效果;染色体数目变异主要集中在偏向于52条染色体,随着体细胞胚的发生、成熟、植株再生,具有正常染色体数细胞的比例逐渐增加,推测具有非正常染色体的细胞在体细胞胚的发育及植株再生过程中丧失了植株的分化能力。  相似文献
