Amino sugars are useful indicators for the accumulation of microbial residues. A 14-day incubation experiment with C4 and C3 sucrose additions was carried out to investigate the relationships between amino sugar-specific shifts in δ13C values and those of CO2 production, microbial biomass C, K2SO4 extractable C and soil organic C (SOC). High performance anion exchange chromatography (HPAEC-IRMS) was able to measure amino-sugar specific δ13C values for muramic acid (MurN), galactosamine (GalN), and glucosamine (GlcN) in the range of natural abundance. At day 7, the initial application of C4 sucrose significantly increased the δ13C value of MurN by 1.0‰ in comparison with the non-amended control treatment, whereas that of GalN and GlcN remained unchanged. This significant increase had disappeared by day 14. This means that the HPAEC-IRMS method is not useful for short-term incubation experiments in the natural abundance range, as the pool size, especially of GalN and GlcN, was too large for a significant response in δ13C values. The δ13C values significantly decreased in the order MurN (−23.2‰) > GalN (−25.7‰) > GlcN (−26.5‰) in the control treatment. Similar δ13C values were measured in GlcN, microbial biomass C, and SOC. MurN exhibited δ13C values similar to the K2SO4 extractable fraction. These results may be caused by differences in the access of bacteria and fungi to different SOC fractions or differences in metabolic fractionation in bacteria and fungi. C3 sucrose application without further nutrient supply seven days after C4 sucrose application together with N and P led to strong mineralization of freshly formed microbial residues.  相似文献

Microbial biomass, β-glucosidase and β-glucosaminidase activities, and availability, storage, and age of soil organic C were investigated after 26 years of conversion from sugarcane (Saccharum officinarum) to forest (Eucaliptus robusta or Leucaena leucocephala), pasture (mixture of tropical grasses), and to vegetable cropping (agriculture) in a vertisol in Puerto Rico. Soil organic C (SOC) at 0–100 cm was similar under Leucaena (22.8 kg C/m2), Eucalyptus (18.6 kg C/m2), and pasture (17.2 kg C/m2), which were higher than under agriculture (13.0 kg C/m2). Soil organic N (SON) at 0–100 cm was similar under the land uses evaluated which ranged from 1.70 (under agriculture) to 2.28 kg N/m2 (under Leucaena forest). Microbial biomass C (MBC) and N (MBN) of the 0–15-cm soil layer could be ranked as: pasture > Leucaena = Eucalyptus > agriculture. The percentages of SOC and SON present as MBC and MBN, respectively, were nearly 1% in pasture and less than 0.50% in forest under Leucaena or Eucalyptus and agricultural soil. The activity of β-glucosidase of the 0–15-cm soil layer could be ranked as: Leucaena = Eucalyptus > pasture > agriculture; while β-glucosaminidase activity was ranked as: Eucalyptus > Leucaena = pasture > agriculture. The soil δ 13C changed from 1996 to 2006 in forest under Eucalyptus (18.7‰ to 21.2‰), but not under Leucaena (20.7‰ to 20.8‰). The soil under Leucaena preserved a greater proportion of old C compared to the forest under Eucalyptus; the former had an increased soil mineralizable C from the current vegetation inputs. The soil under agriculture had the lowest enzyme activities associated with C cycling, lowest percentage of SOC as MBC, highest percentage of SOC present as mineralizable C, and highest percentage of MBC present as mineralizable C compared to the other land uses.  相似文献

Spatial patterns of soil δ13C were quantified in a subtropical C3 woodland in the Rio Grande Plains of southern Texas, USA that developed during the past 100 yrs on a lowland site that was once C4 grassland. A 50 × 30 m plot and two transects were established, and soil cores (0–15 cm, n = 207) were collected, spatially referenced, and analyzed for δ13C, soil organic carbon (SOC), and soil particle size distribution. Cross-variogram analysis indicated that SOC remaining from the past C4 grassland community co-varied with soil texture over a distance of 23.7 m. In contrast, newer SOC derived from C3 woody plants was spatially correlated with root biomass within a range of 7.1 m. Although mesquite trees initiate grassland-to-woodland succession and create well-defined islands of soil modification in adjoining upland areas at this site, direct gradient and proximity analyses accounting for the number, size, and distance of mesquite plants in the vicinity of soil sample points failed to reveal any relationship between mesquite tree abundance and soil properties. Variogram analysis further indicated soil δ13C, texture and organic carbon content were spatially autocorrelated over distances (ranges = 15.6, 16.2 and 18.7 m, respectively) far greater than that of individual tree canopy diameters in these lowland communities. Cross-variogram analysis also revealed that δ13C – SOC and δ13C-texture relationships were spatially structured at distances much greater than that of mesquite canopies (range = 17.6 and 16.5 m, respectively). These results suggest fundamental differences in the functional nature and consequences of shrub encroachment between upland and lowland landscapes and challenge us to identify the earth system processes and ecosystem structures that are driving carbon cycling at these contrasting scales. Improvements in our understanding how controls over soil carbon cycling change with spatial scale will enhance our ability to design vegetation and soil sampling schemes; and to more effectively use soil δ13C as a tool to infer vegetation and soil organic carbon dynamics in ecosystems where C3–C4 transitions and changes in structure and function are occurring.  相似文献

This study was conducted to investigate the effects of slope aspect and position on microbial biomass C (MBC) and some hydrolytic enzyme activities involved in soil N, P, and S cycles in a rangeland ecosystem of west central Iran. Soil samples were collected from three slope positions (summit, backslope, and footslope) of contiguous north- and south-facing slopes. Results indicated higher silt and clay content, soil organic C (SOC), total N (TN), and C/N ratio on the north-facing slope. Furthermore, MBC, alkaline phosphomonoesterase (ALP), acid phosphomonoesterase (ACP), arylsulfatase (ARS), urease (URS), L-asparaginase (LAS), and L-glutaminase (LGL) activities were greater by 46.1, 65.9, 58.6, 59.6, 52.6, 62.8, and 65.7%, respectively, on the north-facing slope compared to the south-facing one. Higher ratios of enzyme activities to MBC were observed on the north-facing slope. In contrast, per cent of inorganic N and microbial quotient were greater on the south-facing slope. The activity of ALP, ACP, ARS along with SOC, TN, and MBC values decreased from summit to footslope. Overall, our findings indicate that north-facing slope and summit position support greater microbial biomass and hydrolytic diversity.  相似文献

Radiation protection planning in urban areas after a radioactive fallout requires knowledge of decontamination caused by storm water transport. This report elucidates the transport of 137Cs from a roof and three storm sewers in Uppsala during the first rainfall after those, on 29–30 April 1986, causing the Chernobyl contamination. Runoff and concentration Of 137Cs in storm water were determined with an accuracy of 10 to 15%. The origin of storm water was determined from its content of 18O. Surface contamination was measured on 3 July 1986. Total fallout was 25 kBq m?2. During the rainfall on 11 May, 4 kBq m?2 were transported from the roof and approximately 1 kBq m?2 from the sewered areas. From 30 April to 4 July there was a decontamination of 13 to 20 kBq m?2. Measurements from another project showed that the fallout rain events washed off 10 to 16 kBq m?2. The wash-off by the small rainfall on 11 May constituted one third of the remaining decontamination occurring up until 4 July. The transport of 137Cs during the 11 May event increased when the runoff increased, but was less efficient as the event proceeded. The relations between 137Cs concentrations and runoff implied that the wash-off of 137Cs in Uppsala was totally dominated by that bound to particles.  相似文献

Abstract

This study describes an economical and simple apparatus and methodology for preparing plant material for δ13C analysis by the combustion of material in sealed Pyrex tubes at 550°C. The largest difference in δ13C for samples combusted at 550°C in Pyrex tubes compared with those combusted at 800°C in the more expensive Vycor tubes was 0.65 δ units. Pyrex combustion tubes are approximately 7% of the cost of Vycor tubes and are easier to handle. Excellent reproducibility was observed with an average standard deviation for samples combusted at 550°C and 800°C of 0.12 and 0.13 units of δ13C, respectively. During the cryogenic distillation of CO2 from the other gasses generated during the combustion, equilibration and evacuation times were varied between 10 s and 120 s. Sample size was varied between 0.4 mg and 2.2 mg. Variations in evacuation time, equilibrium time, and sample size did not significantly affect the value for δ13C. The total duration for the cryogenic distillation of one sample was approximately 5 min.

In inter‐laboratory comparisons, the values for δ13C of various plant samples were found not to be significantly different.  相似文献

Vitamin C, tocopherols, and tocotrienols in berries of wild and cultivated sea buckthorn (Hippopha? rhamnoides L.) of different origins and harvesting dates were determined with HPLC. Wild berries of subsp. sinensis, native to China, contained 5-10 times more vitamin C in the juice fraction than the berries of subsp. rhamnoides from Europe and subsp. mongolica from Russia (4-13 vs 0.02-2 g/L juice). Genetic background and berry-harvesting date were two primary factors determining the vitamin C content in the berries. Crossing different subspecies influenced the vitamin C content to some extent. For bushes cultivated in southwest Finland, the best berry-harvesting date for high vitamin C content was the end of August. The seeds of subsp. sinensis contained less tocopherols and tocotrienols (average 130 mg/kg) compared with seeds of subsp. rhamnoides (average 290 mg/kg) and mongolica (average 250 mg/kg). The fruit flesh of sinensis berries had contents of tocopherols and tocotrienols 2-3 times higher than those found in the other two subspecies (120 mg/kg vs 40 mg/kg in rhamnoides and 50 mg/kg in mongolica). The fresh whole berries of subsp. sinensis were clearly the best source of total tocopherols and tocotrienols. The total content of tocopherols and tocotrienols in the soft parts of the berries reached the maximum level around early- to mid-September, whereas the content in seeds continued to increase until the end of November. The excellent combination of the highest content of vitamin C and tocopherols and tocotrienols makes the berries of subsp. sinensis an optimal raw material for nutritional investigation as a candidate for functional foods with special antioxidative properties.  相似文献

Abstract

An improvement to the Walkley‐Black wet digestion method for the rapid determination of organic carbon over the range 0.2–5.5% in air‐dry soil is described. It permits total recovery of the organic‐C in finely ground soil samples digested with the heat of dilution from mixing N K2 Cr2 O7 with concentrated H2SO4. in test tubes followed by external heating from a hot‐plate digestor. The organic‐C concentrations are determined directly, as the Cr product in diluted soil digests, by absorptiometry at 600 nm with calibration against similarly treated sucrose standards in solution. For the soils tested, there were negligible interferences from carbonates, wood charcoal, coke, Fe+2 and readily reducible Mn; Cl does not interfere with the organic‐C assay in non‐saline soils but for saline soils a correction based on 1/12 Cl assay of the soil is necessary. The present method is compared with Tabatabai and Bremner's dry combustion procedure and Allison's manometric adaptation for calcareous soils. The procedure described here does not require carbonate to be determined and is therefore simpler. In addition it is cheaper, faster and more effective in controlling interferences than dry combustion procedures.  相似文献

Bambara groundnut (Vigna subterranea L. Verdc) is the second most important indigenous food legume in Africa. The aim of this study was to evaluate plant growth, N2 fixation, N contribution, C accumulation, and plant water relations of Bambara groundnut grown in 26 farmers’ fields in Mpumalanga Province of South Africa. The data revealed marked (p?≤?0.05) differences in plant dry matter (DM) yield, N concentration and content, δ15N, the proportion of N derived from symbiotic fixation (%Ndfa), and actual amounts of N-fixed between and among the 26 farms surveyed. Bambara groundnut plants obtained 33–98 % (mean?=?72 %) of their N nutrition from symbiotic fixation and contributed 4–200 kg N-fixed ha?1 (mean?=?102 kg N-fixed ha?1). Plant density correlated positively with %N (r?=?0.31***), δ15N (r?=?0.126***), and amount of N-fixed (r?=?0.15*), indicating that the high %Ndfa values obtained for Bambara groundnut in this study and the low symbiotic N yield associated with some farms were due to low plant density rather than poor symbiotic functioning. Bambara groundnut obtained more N from soil (e.g., 173 kg N ha?1) than from symbiosis (e.g., 135 kg N-fixed ha?1) in some fields, implying that the N2-fixing efficacy of the microsymbionts nodulating Bambara groundnut was low at some locations in South Africa. The data from this study showed δ13C values ranging from ?28.01 to ?26.20?‰, which indicates differences in plant water use efficiency on the different fields studied. Furthermore, the positive correlations between δ13C and N-fixed (r?=?0.15*) and between δ13C and N content (r?=?0.14*) suggest a functional relationship between water use efficiency and N2 fixation, just as the positively significant correlations between δ15N and DM yield (r?=?0.24***), N-fixed and DM weight (r?=?0.76**), and N content and DM yield (r?=?0.99*), as well as N-fixed and C content (r?=?0.76**) also indicate a functional relationship between N2 fixation and photosynthesis. In the same way, the positive correlation between δ13C and DM weight (r?=?0.14*), or δ13C and C content (r?=?0.15*), also implies a functional link between water use efficiency and plant growth. Thus, an increase in water use efficiency in Bambara groundnut, whenever it occurs, seems to functionally enhance plant growth, symbiotic N2 fixation, and photosynthetic activity, just as N2 fixation in nodules also seems to stimulate leaf photosynthesis.  相似文献

Soils were collected from 2-year (2-y) and 3-year (3-y) old red-pine seedling plots in two tree nurseries, Hayward in the north and Wilson in the southwestern part of Wisconsin State respectively, and equilibrated with 0.01 M Ca(NO3)2 for soil solution Zn and Mn (solu-Zn and Mn), and with 0.01 M Ca(NO3)2+0.005 M EDTA for soil adsorbed Zn and Mn (ad-Zn and Mn). Buffering capacity of soil Zn and Mn (b-Zn and Mn) was obtained from the ratio of ad-Zn and Mn to the solu-Zn and Mn.
