This study examined the ability of late stage (instar XXVI) Japanese spiny lobster Panulirus japonicus phyllosoma larvae to uptake seven l-type amino acids (AAs; aspartic acid, Asp; threonine, Thr; glutamic acid, Glu; glycine, Gly; alanine, Ala; valine, Val; and methionine, Met) from the rearing medium. The time course uptake during incubation for 5 h in solutions containing 1, 2, 5, 10, or 20 μM of each AA was determined using high performance liquid chromatography. There was considerable individual variation in uptake rates and even occasional release of AAs, but larvae showed net uptakes of all seven AAs. Two polar acidic AAs (Glu and Asp) were readily taken up whereas neutral AAs, including three considered as essential for crustaceans (Val, Met, and Thr), had lower uptake rates. Transient releases of AAs by the larvae were common for the AAs with the lowest uptake rates at 1–2 h of incubation. Uptake rates increased with increasing AA concentration but less so for those taken up in larger amounts. Larvae took up AAs to a total of 16.4 μmol/g/h for incubation in the 20 μM solution. The ability to uptake nutrients directly from the medium may be important for the well-being of the larvae.  相似文献

The time of first feeding is an important factor for establishing successful exogenous feeding in aquaculture hatcheries as it is crucial for larval survival and growth. The effects of delayed first feeding by the sea cucumber Apostichopus japonicus larvae were investigated. Groups of larvae were fed immediately after completion of the digestive tract (fed control) or starved for 24, 48, 72, 108 and 120 h post fertilization. The growth rates of A. japonicus larvae were the same as those of the control group when first feeding was delayed for less than 72 h. When the period of starvation was beyond 72 h, growth rates were reduced. The fed control group had the highest percentage of late auricularia larvae (70.0%) on day 11, followed by the groups starved for 24, 48, and 72 h, respectively, which had 35.00%, 43.48% and 41.67% of late auricularia larvae. When larvae were starved for more than 72 h, the percentage of late auricularia larvae never exceeded 12.0%. Most larvae halted in development and were deformed before reaching metamorphosis when starved for 108 and 120 h. Prolonged exposure to starvation (more than 48 h) resulted in a significant decrease in percentage of larvae reaching settlement. The point‐of‐no‐return (PNR) of A. japonicus larvae was calculated 110.4 h. Ratios of stomach length to total length, stomach width to total width were more sensitive to starvation than other larval morphometrics. To avoid mortality and obtain adequate growth and development, A. japonicus larvae in commercial culture should be fed within 48 h of the completion of the digestive tract.  相似文献

The combined effects of stocking density (0.2, 0.5, 1, 2, 4 and 8 inds/ml) and dietary microalgal ration (20,000 and 40,000 cells/ml) and the sole effect of temperature (10, 12, 14, 16, 18 and 22°C) on the specific growth rate (SGR), per cent survival and per cent metamorphosis of auricularia larvae of the California sea cucumber, Parastichopus californicus, were evaluated in two separate experiments. The SGR was not significantly affected by stocking densities in the range of 0.2 to 4 inds/ml, but was significantly reduced at 8 inds/ml. The SGR of larvae fed 20,000 cells/ml was significantly reduced in comparison to those fed 40,000 cells/ml. Larvae had significantly higher per cent survival and per cent metamorphosis when reared at densities of 0.2 and 0.5 inds/ml compared with those reared at 2–8 inds/ml. Microalgal ration level did not significantly impact survival or metamorphosis. Larvae reared at 16 and 18°C had significantly higher SGRs and per cent metamorphosis than those held at all other temperatures, while per cent survival was highest at 16°C. Based on these results, we recommend rearing auricularia larvae of P. californicus at a stocking density at or below 0.5 inds/ml, a dietary ration of 40,000 cells/ml, and a temperature of 16°C.  相似文献

The neuropeptide cubifrin-I and its derivative cubifrin-L have recently been shown to be potent substances that induce in vitro oocyte maturation and spawning in the Japanese common sea cucumber Apostichopus japonicus. We have examined the reproductive behavior of A. japonicus in vivo following the injection of cubifrin-L into the body cavity with the aim of determining the practical value of using this peptide to induce spawning in the hatchery setting. The responsiveness of ovarian fragments with oocytes >155 μm in diameter to cubifrin-L in vitro was well correlated with the spawning success induced in vivo by a cubifrin-L injection. Mature sea cucumbers injected with cubifrin-L displayed sequential reproductive behaviors, which comprised climbing up the side wall of the tank toward the water surface, waving of the head, and shedding of gametes. Gamete shedding started about 60 and 80 min after the injection of cubifrin-L in males and females, respectively, and was completed almost simultaneously in the two sexes about 2 h post-injection. Repeated injections of cubifrin-L at intervals of about 10 days successfully induced multiple spawnings in males and females. These results demonstrate that cubifrin-L can be used as an effective inducer of spawning in Japanese sea cucumber cultivation.  相似文献

The growth rate, survival rate, development and setting rate of larval sea cucumber Apostichopus japonicus were measured under four light intensities (0, 50, 500 and 2,000 lx), and the growth rate and metabolism of the juvenile sea cucumbers were investigated under four light intensity treatments (0, 100, 1,000 and 2,500 lx). The light requirements (i.e. intensity) of the sea cucumber changed as they grew. Better growth performance, development and survival rate were observed in embryos and larvae under 500 lx treatment. However, reduced light intensity (50 lx) increased the settlement rate of the larvae. The highest specific growth rate occurred in juvenile sea cucumbers under 1,000 lx. The sea cucumbers in this group also had the lowest oxygen consumption rate and ammonia excretion rate, but the highest O:N ratio, indicating that the optimal light intensity for the juvenile sea cucumber culture was 1,000 lx. Therefore, appropriate light intensities were suggested to provide larval and juvenile sea cucumbers with better growth and development conditions.  相似文献

The aim of this study was to determine the effect of photoperiod on growth and survival of winter flounder, Pseudopleuronectes americanus, from hatch until metamorphosis. Sixteen 100 L upwelling tanks were stocked with larvae at an initial stocking density of 20 larvae/L. Larvae in eight tanks were provided with continuous light (24 hr light/0 hr dark), while those in the remaining eight tanks were grown under a photoperiod of 14 hr light/10 hr dark. Tanks were sampled for larval standard lengh from 6 to 28 days post‐hatch, and the number of live larvae, live metamorphosed and dead metamorphosed fish were recorded at the termination of the experiment (starting 46 dph). Results indicated that larvae provided with continuous light were larger than those sampled from 14 hr light/10 hr dark treatment. No effect of photoperiod on the number of non‐metamorphosed fish recovered at the end of the experiment was detected. On the contrary, over three times as many fish survived and reached metamorphosis under continuous light. An ANCOVA model showed a significant interaction between light and age. Both regressions were significant and provided the following growth equations: SL24 hr light/0 hr dark = 0.101 × age + 2.55 (r2 = 0.96) and SL14 hr light/10 hr dark = 0.094 × age +2.45 (r2 = 0.97). Using 6.36 ± 0.059 mm SL as the mean size at metamorphosis, larvae experiencing continuous and 14 hr light/10 hr dark photoperiods would reach metamorphosis in 38 and 42 days respectively.  相似文献

The red race of the sea cucumber Apostichopus japonicus was introduced into China from Japan for large-scale seed production because of its economic value. This paper reports the effects of stocking density, temperature, and salinity on survival and growth of early larvae before and after feeding, in order to establish conditions for optimal larval growth and production. To maximize the yield per unit of space, densities of 0.5–1 larvae/ml are recommended for non-feeding larvae, while 0.1–0.2 larvae/ml are best for feeding larvae. Higher survival and growth values were obtained for both non-feeding and feeding larvae at temperature ranges from 21 to 24°C. Larvae reared at a salinity of 30‰ always showed maximum growth and survival. Based on results of this study, a temperature range from 21 to 24°C and a salinity of 30 are considered optimal for early development of the red A. japonicus.  相似文献

The effects of dietary tryptophan on growth performance, energy budget and endocrine response of sea cucumber Apostichopus japonicus (Selenka) were studied, to test whether the tryptophan could mitigate the crowding stress response of the sea cucumber. Four density treatments of the sea cucumber (i.e. 4, 8, 16 and 32 individuals per 40 L water, represented as L, ML, MH and H) were fed with diets containing 0 (control), 1%, 3% and 5% l ‐tryptophan, respectively, for 75 days. The results showed that the specific growth rate (SGR) and feed conversion ratio (FCE) of A. japonicus decreased with increasing in the stocking densities but increased after supplementation of 1% and 3% tryptophan. The energy allocation of the A. japonicus was affected significantly by stocking density and dietary tryptophan treatments (< .05). In general, the energy consumed (C) and energy used for growth (G) decreased with the increasing in the stocking densities. The highest proportion of energy deposited to growth occurred in each 3% treatment (L, 6.38%; ML, 6.12%; MH, 4.56% and H, 4.14%), which were significantly higher than the controls (< .05) respectively. Higher cortisol, glucose and lactate levels of A. japonicus were recorded in the control treatments of MH and H, but l ‐tryptophan supplementation could reduce the levels of cortisol, glucose and lactate of the test animals. The present results proved that the growth performance of the sea cucumber was inhibited by high stocking densities, while dietary supplementation of tryptophan could significantly mitigate the crowding stress, and improve the growth of sea cucumber.  相似文献

Two trials were carried out in the laboratory in order to assess the effect of microparticulated feed (F) and live (Thalassiosira pseudonana, M) diets on the growth of recently set (396 ± 13 μm shell height) and 2 mm Crassostrea gigas postlarvae. Different proportions of M and F (100:0, 75:25, 50:50; 25:75, 0:100) were delivered in a single dose of 3 h d−1 in trial 1. Dietary M:F proportions of 100:0, 50:50, and 0:100 were delivered as a single pulse of 8 h d−1 (P1) or two pulses of 4 h−1 (P2) in trial 2. Maximal daily M ration was 296 cells μl−1 d−1 (trial 1), 150 M cells μl−1 d−1 (trial 2), or their equivalent F dry weight. Shell height (SH), dry (DW), and organic weight (AFDW) were evaluated weekly. Oysters from trial 1 significantly increased their size after 28 days, and exhibited no significant dietary differences in terms of DW (1.21 ± 0.15 to 2.01 ± 0.28 mg) or AFDW (0.091 ± 0.022 to 0.166 ± 0.029 mg). Newly set postlarvae (trial 2) also exhibited significant growth after 25 days. No dietary differences were observed in trial 2, yet P2 oysters attained significantly higher shell heights (825–912 μm) than P1 oysters (730–766 μm) after 25 d. Pulse effects were marginally not significant in terms of AFDW and growth rate. Together, these findings showed that balanced microfeeds have a practical potential for the culture of early C. gigas postlarvae, when they are delivered in pulse-feeding schemes  相似文献

The sea cucumber (Stichopus japonicus) is able to undergo autolysis in response to a variety of environmental and mechanical cues. Within the framework of a long-term study of this phenomenon we have purified a protease from the body wall of the sea cucumber by means of ion-exchange chromatography with DE-52 cellulose and gel filtration chromatography with Sephadex G-100. The final enzyme preparation was nearly homogeneous on polyacrylamide gel electrophoresis, and its molecular weight was estimated to be approximately 35.5 kDa. The purified enzyme exhibited a maximum activity for the hydrolysis of casein at pH 7.0 and 50°C and a remarkable stability at pH 4.0–7.0 and 40–60°C. Based on the inhibition and activation profiles obtained with numerous specific protease inhibitors and an activator, the protease purified from the body wall of the sea cucumber was defined to be a cysteine-like protease.  相似文献

Culture performance beyond metamorphosis of larval loach (Misgurnus anguillicaudatus) was examined in a feeding experiment of the early development stage (20 days after hatch; DAH). Total length, dry weight, length- and weight-specific growth rate (SGR) and survival were monitored in different diet regimes. During 20 days, diet treatments included: microparticle diets (A); live daphnia (Moina micrura) (B); live daphnia plus live chlorella (Chlorella pyrenoidosa) (C); and live daphnia plus microparticle diets (D). Fish survival rates during 20 days were 21.23 ± 4.2% (A), 73.19 ± 2.8% (B), 90.76 ± 3% (C) and 91.46 ± 3.1% (D), respectively. Length- and weight-specific growth rate after 20 DAH (final mean SGR; % day−1) were 5.36 ± 0.44 and 15.75 ± 1.52 (A), 9.29 ± 1.25 and 23.47 ± 2.23 (B), 9.42 ± 1.55 and 24.88 ± 2.9 (C) and 9.55 ± 1.23 and 24.40 ± 2.75 (D), respectively. Fish in treatments B, C and D displayed higher growth rates and were significantly longer and heavier than fish in treatment A by the end of the experiment (Ρ < 0.05). Fish in treatment A had highly significant greater (Ρ < 0.001) mortalities than in treatments B, C and D. There were no significant differences in any growth parameter between fish in treatments B, C and D, but the survivals in treatments C and D (90.76% and 91.46%) were significantly higher than in treatment B (73.19%, Ρ < 0.05). The results demonstrated that enriched prey and co-feeding may serve as a potential feeding strategy for loach larvae, and the form of co-feeding reduces the costs and dependence on live foods to a certain extent. We concluded that larval loach should be reared over metamorphosis using either of the following methods: feed with live daphnia supplemented with microparticle diets or with live chlorella. However, a prolonged rearing period of loach larvae is needed to detect nutritional problems and observe remote effects of co-feeding on weaning in the future.  相似文献

We evaluated the effects of different proportions of dietary protein (5%, 10%, 15%, 20%, 25% and 30% protein) on the activity of digestive enzymes of normal and albino Apostichopus japonicus. The experimental diets were fed for 60 days, the optimal conditions for digestive enzyme activity in sea cucumbers were studied. The optimal temperature for protease was 29.3°C and the optimal pH was 1.8. The optimal temperature for amylase was 34.3°C and the optimal pH was 6.7. The optimal temperature for cellulase was 56°C and the optimal pH was 5.9. The activity of intestinal protease increased at first and then decreased as the proportion of dietary protein increased, reaching the maximum when the proportion of protein was 19.7%. The activity of protease in the intestine of normal sea cucumber was significantly lower than that of albino sea cucumber, and the activity of amylase was significantly higher than that of albino sea cucumber. This study is expected to provide a basis for further explaining the ecological difference of albino and normal A. japonicus.  相似文献

为研究饲料中添加水苏糖对刺参生长、消化生理与糖代谢的影响,以初始体重为(11.46±0.03) g的刺参幼参为实验对象,在基础饲料中添加包膜水苏糖,配成水苏糖含量分别为0 (D1,对照组)、0.04%(D2)、0.11%(D3)、0.15%(D4)、0.21%(D5)和0.27%(D6)的6组实验饲料,在循环水养殖桶中进行为期67 d的生长实验。结果显示,(1)随着饲料中水苏糖含量的增加,刺参的增重率及特定生长率均先上升后下降,D2~D5组显著高于对照组,体壁基本营养成分不受水苏糖添加量的影响;(2)肠道蛋白酶、脂肪酶和超氧化物歧化酶活性先上升后下降,D3、D4组显著高于对照组,丙二醛含量先下降后上升,在D3组达最低值,淀粉酶、碱性磷酸酶和酸性磷酸酶活性无显著性差异;D3、D4组肠道皱襞高度显著大于对照组,D6组出现炎症细胞浸润;(3)葡萄糖激酶活性先上升后平稳,D5、D6组显著高于其他组,果糖磷酸激酶和丙酮酸激酶活性先上升后下降,分别在D4、D3组达最大值;磷酸烯醇式丙酮酸羧激酶活性呈上升趋势,D6组显著高于其他各组。研究表明,水苏糖提高了刺参糖代谢效率,改善了机体消化生理和抗氧化能...  相似文献

Sea cucumber Apostichopus japonicus juveniles acclimated to different environmental conditions (23, 25, and 27°C combined with 25, 30, and 35 psu) were assessed for tolerance to increasing and decreasing levels of salinity at a rate of 2 psu h−1. They were also tested for the LS50 (median lethal salinity) when transferred directly into a series of higher salinity (32–46 psu) and lower salinity (9–25 psu). The CSMax (critical salinity maximum), CSMin (critical salinity minimum), USTL (upper salinity tolerance limit), and LSTL (lower salinity tolerance limit) were positively correlated to the acclimated salinity level but negatively correlated to temperature. The CSMax of A. japonicus was 6.2–10.0 psu higher than the USTL, and the CSMin was 5.5–8.5 psu lower than the LSTL, indicating that gradual changes in salinity resulted in the wide range of salinity tolerance that was observed, but that abrupt changes in salinity resulted in the narrow range of tolerance. Two-way analysis of variance revealed that salinity and temperature had a significant effect on 50% CSMax, 50% CSMin, USTL, and LSTL (P < 0.001). The information obtained in this study will be valuable for the further development of the sea cucumber aquaculture industry in China.  相似文献

The diel feeding rhythm and ontogenesis during early life stage of loach Misgurnus anguillicaudatus were investigated under experimental conditions (light: L 06:00–18:00, D 18:00–06:00 h). Morphological and behavioral developments of loach from newly hatched to 40 days after hatching were observed. Larvae were able to prey on daphnia 3–4 days after hatching at 23 ± 0.5°C. As the larvae grew, they showed an increasing feeding capacity and a distinct feeding rhythm. Feeding intensity and incidence for day-4 larvae were highest at 10:00 and 16:00 h. The highest levels of feeding intensity for day-12 larvae occurred at 08:00, 12:00, and 18:00 h as did feeding incidence. By day 20, when the larvae metamorphosed, the highest levels of feeding intensity occurred at 06:00, 18:00, and 24:00 h and were concurrent with the highest feeding incidence. After metamorphosis, feeding capacity had again increased considerably and, in contrast to the earlier stages before day 20, feeding intensity for day-30 juveniles peaked at 05:00 and 20:00 h, about 1–2 h after the maximum feeding incidence. The feeding rhythm of loach juveniles at day 40 was almost the same as the day-30 juveniles. The estimated maximum daily feeding rates were 43.1%, 33.4%, 19.0%, 12.8%, and 5.8% of body weight on days 4, 12, 20, 30, and 40, respectively. Thus, loach was found to have different feeding rhythms in the pre- and post-metamorphosis stages, with the highest feeding activity in daytime during the larval planktonic stage before metamorphosis, and intensely nocturnal feeding behavior during the juvenile benthic stage after metamorphosis.  相似文献

The effects of mono‐species and bi‐species microalgal diets on body length, survival and metamorphosis of auricularia larvae of the California sea cucumber, Parastichopus californicus, were tested in two separate laboratory feeding experiments. The first examined eight single species of microalgae – Chaetoceros calcitrans (Cc), Chaetoceros muelleri (Cm), Dunaliella tertiolecta (Dt), Isochrysis sp. (Tahitian strain: TISO) (Ti), Pavlova lutheri (Pl), Phaeodactylum tricornutum (Pt), Tetraselmis suecica (Ts) and Thallassiosira pseudonana (Tp). The second experiment took the five most effective single species from the first experiment (i.e. Cc, Cm, Dt, Pl and Ti) and presented them in all possible binary combinations. In the first experiment, Cc was determined to be the best diet on the basis of generating a significantly higher metamorphic rate (65.5 ± 4.2%, mean ± SD) than any other species and leading to one of the best survival rates (72.3 ± 7.6%) and greatest body lengths (831.6 ± 5.6 μm at day 14). In the second experiment, diets containing Cc generally produced the highest survival and metamorphic rates and greatest body lengths (except Cc/Cm) of all mixed diets, although no bi‐species treatment was ever significantly superior to a control of Cc alone. Of the various mono‐species and bi‐species diets evaluated for rearing auricularia larvae of P. californicus, Cc (either alone or mixed with other species) is recommended.  相似文献

The growth response, non‐specific immune activities and disease resistance were measured in sea cucumber, Apostichopus japonicus Selenka (initial average weight 6.80 ± 0.10 g), when fed diets supplemented with graded levels of guanosine from the guanosine‐5’‐monophosphate disodium (GMP) at 0 (control), 0.3, 0.6 and 1.2 g/kg for 8 weeks. The results showed that GMP supplemented at 0.6 and 1.2 g/kg significantly enhanced the growth of sea cucumber. Sea cucumber fed a diet with 0.6 g/kg of GMP had significantly higher intracellular superoxide anion production, nitric oxide synthase activity, lysozyme activity and the total superoxide dismutase (T‐SOD) activity than those in control group (< .05). Increased lysozyme activity and T‐SOD activity were also found in sea cucumber fed GMP at 1.2 g/kg. Moreover, there was significantly lower cumulative mortality after the disease challenge in sea cucumber fed the diets with 0.6 and 1.2 g/kg GMP than that in control and 0.3 g/kg GMP groups (< .05), and no significant difference was observed between 0.6 and 1.2 g/kg GMP groups. These results suggested that feeding GMP at a dose of 0.6 g/kg could enhance growth, non‐specific immunity of sea cucumber as well as its resistance against Vibrio splendidus.  相似文献

为了对导致辽宁大连、山东东营的2家养殖场池塘养殖刺参大量化皮死亡的新的敌害生物进行鉴定并确定其对养殖刺参的危害。本实验通过形态学观察、分子鉴定及系统发育分析确定了涡虫的分类地位,通过生态学方法确定了其生态适应条件,通过切割后培养的方法观测了其再生能力,通过与刺参苗种的共培养实验测试了该物种对刺参的危害及其危害方式。形态学观察结果显示,该涡虫体长0.96~3.26 mm,体宽0.49~1.93 mm,外观黄色或黄褐色,头部钝圆,具一对暗红色棒状眼点,尾部具两条并列的尾垂;显微镜镜检发现其表皮下分布密集的虫黄藻,体表周生纤毛,雌雄同体,口后具有两个生殖孔;对该物种COⅠ及18S r DNA基因片段扩增测序结果进行分析,并构建基于18S rDNA基因的系统发育树,结果显示该生物与澳洲异尾涡虫序列同源性达99.64%,根据其形态学特征,并结合18S rDNA分子鉴定结果,将该生物鉴定为澳洲异尾涡虫;进一步对其生活习性进行了研究,结果显示,该生物具有避光性,其适宜温度为18~24°C,适宜pH为5.5~8.0,适宜盐度为20~40;再生实验表明,该物种具有很强的前后轴极性再生能力;该生物与刺参的共培养实验表明,澳洲异尾涡虫对刺参体表表现出很强的趋向性,可以吸附在刺参体表导致刺参苗种溃疡、化皮甚至死亡,但刺参的体腔、肠道、呼吸树内均未发现虫体寄生。研究表明,澳洲异尾涡虫是营自由生活的池塘养殖刺参的一种新的敌害生物,在养殖过程中需要密切关注并防范该敌害生物。  相似文献

亚硒酸钠对刺参免疫反应中体腔液酶活力的影响   总被引:1,自引:0,他引:1  
为了合理评价硒对水产养殖刺参的作用,实验检测和观察了室内模拟亚硒酸钠处理及病菌感染条件下刺参体腔液免疫酶的变化、组织病理变化和相对保护率。结果表明,随着亚硒酸钠处理强度的增加,刺参体腔液中超氧物岐化酶(SOD)、谷胱甘肽过氧化物酶(GP-X)、溶菌酶(LYZ)、酚氧化酶(PO)及碱性磷酸酶(ALP)活性升高;在同时感染病菌的情况下,SOD、GP-X及LYZ活性表现为先升高后下降的趋势,PO和ALP活性则持续上升,但随亚硒酸钠浓度升高,ALP活性增加的幅度减少,而较高亚硒酸钠浓度时累计发病率上升,相对保护率下降。研究结果表明,适宜浓度的亚硒酸钠可增强刺参对病菌感染的免疫力,而病菌感染可以加剧亚硒酸钠胁迫对刺参造成的免疫功能损伤,所以适度利用硒可减轻刺参养殖病害的发生。  相似文献
