ABSTRACT: BACKGROUND: An endogenous circadian clock controls locomotor activity in common spiny mice (Acomys cahirinus). However, little is known about the effects of constant light (LL) on this activity or about the existence of an additional food entrainable clock. A series of experiments were performed to investigate the effects of LL and DD on tau and activity levels. METHODS: Spiny mice were housed individually and their running wheel activity monitored. One group of mice was exposed to LD, DD and several intensities of LL. Another group was exposed to a restricted feeding (RF) paradigm in light: dark (LD) during one hour before the L to D transition. Significance of rhythmicity was assessed using Lomb-Scargle periodograms. RESULTS: In LD all animals exhibited nocturnal activity rhythms that persisted in DD. When animals were exposed to RF (during L), all of these animals (n = 11) demonstrated significant food anticipatory activity as well as an increase in diurnal activity. This increase in diurnal activity persisted in 4/11 animals during subsequent ad libitum conditions. Under LL conditions, the locomotor rhythms of 2/11 animals appeared to entrain to RF. When animals were exposed to sequentially increasing LL intensities, rhythmicity persisted and, while activity decreased significantly, the free-running period was relatively unaffected. In addition, the period in LL was significantly longer than the period in DD. Exposure to LL also induced long-term changes (after-effects) on period and activity when animals were again exposed to DD. CONCLUSIONS: Overall these studies demonstrate clear and robust circadian rhythms of wheel-running in A. cahirinus. In addition, LL clearly inhibited activity in this species and induced after-effects. The results also confirm the presence of a food entrainable oscillator in this species.  相似文献

The bamboo aphid, Pseudoregma bambucicola, is an important insect pest of bamboo that affects normal bamboo growth and induces sooty molds. The control of P. bambucicola involves the application of chemicals, such as imidacloprid, to which many species are resistant. In this study, we isolate a novel botanical pesticide (9-oxo-10,11-dehydro-ageraphorone) from an Eupatorium adenophorum(Asteraceae: Compositae) petroleum ether extract and test the aphicidal activity of this compound against P. bambucicola in laboratory bioassay and field-based experiments. This ageraphorone compound at a concentration of 2 mg/ml caused 73.33% mortality (corrected mortality [Subtracted the mortality of the negative control]: 70%) of P. bambucicola by laboratory bioassay within 6 h. Even at lower concentrations, this compound caused greater 33% mortality (corrected mortality: 30%) of aphids. Field experiments with naturally infested bamboo plants showed that two applications of 2 mg/ml ageraphorone to infested plants completely cleared infestations within 30 d. These effects were similar to those of the positive control (imidacloprid). These results reveal that 9-oxo-10,11-dehydro-ageraphorone exhibits significant aphicidal activity against bamboo aphids. We suggest that future research be directed at developing this ageraphorone compound from E. adenophorum as an aphicidal agent for biocontrol.  相似文献

Every physiological function in the human body exhibits some form of circadian rhythmicity. Under pathological conditions, however, circadian rhythmicity may be dusrupted. Patients infected with HIV or addicted to drugs of abuse often suffer from sleep disorders and altered circadian rhythms. Early studies in Drosophila suggested that drug seeking behavior might be related to the expression of certain circadian clock genes. Our previous research showed that conditioned place preference with morphine treatment was altered in mice lacking the Period-1 (mPer1) circadian clock gene. Thus, we sought to investigate whether morphine treatment could alter the expression of mPer1, especially in brain regions outside the SCN and in peripheral tissues. Our results using Western blot analysis showed that the mPER1 immunoreactivity exhibited a strong circadian rhythm in the brains of the control (Con), morphine-dependent (MD), and morphine-withdrawal (MW) mice. However, the phase of the circadian rhythm of mPER1 expression in the brains of MD mice significantly differed from that of the Con mice (p < 0.05). In contrast to mPER1 expression in the brain, the circadian rhythm of mPER1 immunoreactivity in the kidneys was abolished after morphine administration, whereas the Con mice maintained robust circadian rhythmicity of mPER1 in the kidney. Therefore, the effect of morphine on the circadian clock gene mPer1 may vary among different organs, resulting in desynchronization of circadian function between the SCN and peripheral organs.  相似文献

The price of hops in Spain is given per kilogram of dry product and according to their content of alpha-acids. If any aphids are noted inside cones there is a penalty of up to 10% of the dry weight. Experiments were performed in a hop garden in 2002, 2003 and 2004 to investigate population development of the damson-hop aphid, Phorodon humuli, and its natural enemies in hop (Humulus lupulus) cones. Furthermore, we investigated if the alpha-acid content was affected by aphid populations. Aphids were first observed within hop cones at the beginning of August and numbers rose sharply from mid- to late August onwards. Aphid infestation of cones reflected the numbers observed on foliage in July, with a greater population halfway up the bines (3 m height) than at the top (6 m height). The alpha-acid content of dried hops was unaffected by the aphid population on leaves, but aphid contamination reduced the economic value of the crop because of arbitrary commercial criteria related to the presence of aphids in cones. The hop aphid’s main natural enemies within cones were anthocorids, all belonging to the genus Orius. The anthocorid population in cones grew in tandem with the aphid population, suggesting that these predators are worthy of consideration in an integrated management approach.  相似文献

Aphid parasitoids are common in Brazilian wheat fields, and parasitize aphids at the wheat tillering stage. However, there is little information available about when this natural enemy occurs in wheat crops. This study investigated the initial occurrence of aphid parasitoids in four commercial wheat crops in northern Paraná during the 2009 crop season. We installed two Malaise traps at each wheat farm, and 400 tillers were assessed weekly in each field for aphid abundance. During this study, we captured 4,355 aphid parasitoids and 197 aphids. Three species of braconid parasitoids were identified, including Aphidius colemani (Viereck 1912), Lysiphlebus testaceipes (Cresson 1880), and Diaeretiella rapae (McIntosh 1855). The aphids species identified were Rhopalosiphum padi (Linnaeus 1758) and Sitobion avenae (Fabricius 1775). This study showed that aphid parasitoids are present in wheat crops even when aphid densities are low, and in one farm, occurred before the aphids colonization. These reports can justified the high efficiency of these natural enemies against aphids in wheat fields.  相似文献

The commercial mycopesticide, Vertalec® based on Lecanicillium longisporum, was evaluated for simultaneous control of aphids and powdery mildew on cucumbers in a greenhouse setting where temperature and RH were allowed to fluctuate within normal operating ranges. Five to six week old cucumber plants were inoculated with either Sphaerotheca fuliginea (Sf) spores, cotton aphids (Aphis gossypii) or both. Vertalec, Vertalec containing irradiation-inactivated blastospores (II Vertalec) or sterilized water (control) were applied to the plants 1, 4, and 7 days later. Vertalec treatment provided complete control of aphids 16 days after aphid inoculation, whereas effects of the II Vetalec were not significantly different from the water-treated control. The number of powdery mildew spots on cucumber leaves and the number of S. fuliginea spores in each spot were significantly lower in Vertalec-treated plants than II Vertalec-treated plants or the controls, whereas numbers in the II Vertalec treatment were lower that the untreated control. These results demonstrate that Vertalec has potential for simultaneous management of both cotton aphid and powdery mildew in greenhouse cucumber production.  相似文献

Soybean aphid (Aphis glycines Matsumura), an exotic species first discovered in the North Central region of the United States in 2000, is a competent vector of severalPotyviridae. Soybean aphid has high fecundity and produces alatae (winged morphs) readily, characteristics typical of proficient virus vectors. When soybean aphids were exposed toPotato virus Y (PVY)-infected potato plants and then clip-caged on healthy potato plants in groups of five or as single aphids, PVY transmission ranged from 14% to 75% across all experiments. Although out-of-lab investigation of the human circadian clock at the clock gene expression level remains difficult, a recent method using hair follicle cells might be useful. While exercise may function as an entrainment cue for circadian rhythms, it remains unclear whether exercise affects human circadian clock gene expression. Efforts to observe apparent effects of exercise on clock gene expression require that several specific conditions be met: intense exercise should be habitually performed at a relatively uncommon time of day over an extended period; and any relative phase shift thereby observed should be validated by comparison of exercise and no-exercise periods. Wake-up and meal times should be kept almost constant over the experimental period. The present study was conducted using a professional fighter who met these strict criteria as subject. Facial hair samples were collected at 4-h intervals around the clock to ascertain rhythms of clock gene expression. During a period in which nighttime training (from 20:00 to 22:00) was habitually performed, circadian clock gene expression was phase-delayed by 2 to 4 h compared with that during a no-exercise period. Maximum level and circadian amplitude of clock gene expression were not affected by the nighttime training. Our trial observations illustrate the possibility that heavy physical exercise might strongly affect the circadian phase of clock gene expression. Exercise might be therefore effective for the clinical care of circadian disorders. The results also suggest that athletes may require careful scheduling of heavy physical exercise to maintain normal circadian phase and ensure optimal athletic performance.  相似文献

The aphids Macrosiphum euphorbiae (Thomas) and Myzus persicae (Sulzer) (Hemiptera: Aphididae) are responsible for yield reduction in potato (Solanum tuberosum) production by direct phloem feeding and by spreading viruses. Breeding resistant traits from Solanum chomatophilum into the potato germplasm provides alternative means to control aphid infestations. Integrated pest management strategy, using plant resistance, benefits from the characterization of the resistance and of its impact on aphid biology. Our objective was to characterize the resistance of S. chomatophilum by assessing the effects of accessions, plant parts on aphid performance, and by assessing the impact of the resistance factors on different aphid developmental stages and on alate morph production. Detailed aphid performance was obtained by measuring fecundity, survival, percentage of nymphs that reached adult moult, and population growth using whole plant and clip cage experimental designs. Accession and plant physiological age, but not aphid developmental stage, influenced all life-history parameters, except for alate morph production which was not induced on the resistant accessions. Plant part influence was independent of plant species and accession. Both experimental designs resulted in congruent resistance levels at the accession level for each of the two aphid species, supporting the use of any of them in S. chomatophilum resistance screening. PI243340 was resistant to both aphid species, while PI365324 and PI310990 were also resistant to M. euphorbiae and M. persicae, respectively.  相似文献

Insecticidal effect of Ungernia severtzovii bulbs extract has been investigated against the grain aphid Schizaphis graminum. LC50 value of the ethanolic extract of U. severtzovii against the female aphids was 0.235% (2.35 g/l). There was 100% mortality of the subsequent nymphs of the treated females at 0.25-1.0%. The extract was equally active against the nymphs at the highest concentrations. Hexane and ethanolic extracts were the most active against the aphids of all the extracts. Fractionation of the hexane extract by TLC yielded three fractions. Fraction II was the most active (>90% mortality) against the aphids. Xanthoxylin was identified as the major constituent in fraction II of the hexane extract and may be responsible for the insecticidal effect of U. severtzovii.  相似文献

Interactions between an invasive aphid, Metopolophium festucae (Theobald) subsp. cerealium, and Barley yellow dwarf virus (BYDV-PAV) were studied under laboratory conditions. M. festucae cerealium is an economic pest of wheat and barley that has recently been found in high population densities in wheat in the Pacific Northwest of the United States. BYDV-PAV is the most prevalent and injurious species of BYDV worldwide and in the Pacific Northwest. Although M. festucae sensu stricto (Theobald 1917) has been reported previously as a vector of some BYDV isolates, there is no confirmed transmission of BYDV by M. festucae cerealium. Two experiments examined the ability of M. festucae cerealium to transmit BYDV-PAV. The first used single aphids caged to indicator plants of a BYDV-susceptible winter wheat cultivar and the second used multiple aphids on each test plant. M. festucae cerealium did not transmit BYDV-PAV in either experiment, whereas transmission by a known BYDV vector, Rhopalosiphum padi L., was consistently high (≥93%). A third experiment compared the intrinsic growth rate, days until first reproduction and daily reproduction by M. festucae cerealium on sham-inoculated and BYDV-PAV-infected wheat, but detected no differences. The findings are reviewed in light published data on M. festucae species, BYDV transmission, and the potential pest status of this new invading aphid.  相似文献

Populations of the green peach aphid,Myzus persicae (Sulzer), and the potato aphid,Macrosiphum euphorbiae (Thomas), were observed on 27 named varieties of Irish potatoes in the Yakima Valley of Washington intermittently from 1947 through 1968. The Epicure, Great Scot, and B5141-6 varieties produced relatively fewer green peach aphids than other varieties tested; Bliss Triumph and Red Pontiac produced relatively fewer potato aphids. Except for Doebay Red, the red-skinned varieties, Bliss Triumph, Red McLure, Norland, and Earnly Rose, were extremely to moderately productive of the green peach aphid. No correlation was apparent between abundance of aphids and the specific gravity or yield of tubers.  相似文献

Since 1971, we have used roquette,Eruca vesicaria (L.) Cav. subsp.sativa (Mill.) Thell., as a host for prolonged maintenance of colonies of the green peach aphid,Myzus persicae (Sulzer), and the potato aphid,Macrosiphum euphorbiae (Thomas). When roquette plants were kept in a growth chamber with 12 hour light and dark periods and a temperature regime of 20°C light and 15°C dark, or in greenhouse cubicles with temperatures ranging from 22–28°C and light periods ranging from 12 to 14 hours, colonies of these aphids could be maintained on a single plant for 16 weeks. These aphids retained their original characteristics, readily accepting radish, lettuce, and snapdragon as hosts, but accepted chrysanthemum only with great reluctance. Aphids from colonies that were maintained on roquette for 4 years did not lose their ability to transmit viruses.  相似文献

Crop resistance plays a role in preventing aphid damage, benefiting food production industries, but its effects are limited due to aphid adaptation and phenotypic plasticity. Therefore, furthering understanding of aphid–crop interactions will improve our ability to protect crops from aphids. To determine how aphids adapt to resistant varieties of wheat, Triticum aestivum L. over time, we performed a laboratory experiment to assess the multi-generational effects of three wheat varieties, Batis, Ww2730, and Xiaoyan22, with different resistance levels on the fitness of Sitobion avenae (Fab.) (Hemiptera: Aphididae). The results showed that Ww2730 and Xiaoyan22 were more resistant than Batis to S. avenae, regardless of whether the aphids were newly introduced or had been acclimated before being introduced to the three wheat varieties. However, the effect of resistance on aphid life-history traits was time dependent. Aphid weigh gain increased and they development faster of the acclimated generation compared to the newly introduced generation on all three varieties. And the fecundity on the three varieties and net reproduction rates on Batis and Xiaoyan22 significantly decreased. Aphid fitness in terms of individual life-history parameters improved, whereas aphid fitness in terms of reproductive decreased, and a convergence effect, the difference gaps and standard errors of all life-history traits among the three acclimated populations had narrowed and were less than those in the three first-generation populations, was observed during the 3-mo experimental period. We suggested that S. avenae could rapidly respond to wheat resistance through life-history plasticity.  相似文献

The spread of potato leaf roll virus (PLRV) and the resulting phloem necrosis in the tubers has been a limiting factor in the production of the Netted Gem (Russet Burbank) variety in southwestern British Columbia, Canada. Steckling sugar beets are an important overwintering host for nymphs and adults of the green peach aphid,Myzus persicae (Sulzer). By June, aphids are widely disseminated to the florets and new leaves of sugar beet and other hosts, including potato. Colonies with alates are produced on summer hosts which are not treated with insecticides. The peak of the flight of alates from the colonies on the summer hosts occurs during the first half of August. Those alates that were produced on PLRV-infected potato and which travel to other potatoes transmit the virus as soon as they feed. By the end of August, the sugar beet seed crops and the early and mid-season potato crops have been defoliated or harvested. Thus many colonies ofM. persicae are destroyed and the threat of PLRV spread diminishes. Crop and weed plants which serve as overwintering hosts become infested during late summer with alate aphids. Their offspring survive the winter in numbers which are determined by the weather and survival of host plants.  相似文献

The greater sensitivity to aphid vectored diseases of potato plants originating from plantlets or minitubers compared to plants originating from field grown tubers could not be explained by changes in probing behavior, by increased aphid movement between plants or by greater levels of aphid colonization favorable to virus spread. The number of seconds to first probe, the duration and number of seconds before the long probe and the duration of the long probe were statistically similar on all plant types. The average residency time of green peach aphids on each plant type was also statistically the same. WingedMyzus persicae (Sulzer) (green peach aphid) landed in greater numbers on older than on younger plants regardless of plant type. This suggests that the level of aphid colonization is determined by a combination of factors such as height, surface and age of plants but not by plant origin. Where virus vectors are present early in the season, the greater number of aphids landing on the relatively older and larger transplants may contribute to higher infection levels, but later in the season plant physiological factors favorable to virus multiplication or translocation are more likely factors  相似文献

The cotton bollworm Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) is a widespread pest of many cultivated and wild plants in Europe, Africa, Asia, and Australia. In 2013, this species was reported in Brazil, attacking various host crops in the midwestern and northeastern regions of the country and is now found countrywide. Aiming to understand the effects of different host plants on the life cycle of H. armigera, we selected seven species of host plants that mature in different seasons and are commonly grown in these regions: cotton (Gossypium hirsutum, “FM993”), corn (Zea mays, “2B587”), soybean (Glycine max, “99R01”), rattlepods (Crotalaria spectabilis), millet (Pennisetum glaucum, “ADR300”), sorghum (Sorghum bicolor, “AGROMEN70G35”), and cowpea (Vigna unguiculata, “SEMPRE VERDE”). The development time of immatures, body weight, survivorship, and fecundity of H. armigera were evaluated on each host plant under laboratory conditions. The bollworms did not survive on corn, millet, or sorghum and showed very low survival rates on rattlepods. Survival rates were highest on soybean, followed by cotton and cowpea. The values for relative fitness found on soybean, cotton, cowpea, and rattlepods were 1, 0.5, 0.43, and 0.03, respectively. Survivorship, faster development time, and fecundity on soybean, cotton, and cowpea were positively correlated. Larger pupae and greater fecundity were found on soybean and cotton. The results indicated that soybean, cotton, and cowpea are the most suitable plants to support the reproduction of H. armigera in the field.  相似文献
