Tests were performed to study the inhibitive capacity of protein-protease inhibitors, 17-beta estradiol, and progesterone in the blood serum of 21 heifers returning to oestrus which had been treated intramuscularly with 400 microgram LH/FSH-RH. Four selected heifers were examined as to the relationships between the concentrations of 17-beta estradiol, progesterone and the inhibitive capacity of the serum. Heifer no. 1 had low concentrations of 17-beta estradiol and higher concentrations of progesterone (average levels 7.8 and 657.4 pg ml-1 of serum). On the other hand, heifer no. 4 was found to have higher concentration of 17-beta estradiol and lower values of progesterone (average levels 28.3 and 325 pg ml-1 of serum). Having shown heat, the heifers were fertilized and remained in calf. Heifer no. 2 with a low concentration of progesterone (292 pg ml-1 of serum) and 17-beta estradiol (12.4 pg ml-1 of serum), as well as heifer no. 3 with a high concentration of both (830 pg ml-1 of serum and 22.7 pg ml-1 of serum, respectively) showed no heat and they were not mated. Heifers with significant differences in the concentrations of progesterone and 17-beta estradiol and with low values of protein-protease inhibitory capacities remained in-calf, whereas heifers with lower progesterone and 17-beta estradiol values, or adversely, with higher inhibition values, showed no heat. Synthetic LH/FSH-RH was found not to increase the amount of protein-protease inhibitors in heifer serum, as distinct from the cervical mucus of breeding cows treated with gestagens in heat synchronization.  相似文献

Oestrus cycles were investigated in seven polar foxes: the investigation consisted of evaluation of clinical symptoms of heat (vulva enlargement), measurement of vaginal mucus electrical resistance and determination of progesterone concentrations in the blood plasma of foxes; the objective was to determine optimum insemination time. The females were inseminated with fresh semen intravaginally by means of a pseudopenis, a day following the record of maximum electrical resistance of vaginal mucus and at the minimum progesterone concentration in blood plasma of 30 ng.ml-1. The females were reinseminated on the following day. The insemination dose had a volume of 1 ml with sperm concentrations of 150.10(6). Clinical symptoms of heat were observed in six out of the seven test foxes. Heat detection by means of measuring vaginal mucus electrical resistance was successful also in six females. Progesterone test enabled to detect heat in five females. Out of six inseminated females, five foxes became pregnant (83.3%) and a total of 33 cubs were born; this is 5.5 cubs per female.  相似文献

Knowledge of pathogenesis of sexual dysfunctions at altered thyroid activity is limited by the knowledge of multiple and ubiquitous action of its hormones throughout the organism. One of the possibilities of modulatory influence of thyroid hormones on sexual functions can be realized through the participation of thyroxine and triiodothyronine in the synthesis and metabolism of primary substrate of steroid synthesis--cholesterol. The presented work is aimed at the study of simultaneous dynamic changes of concentrations of thyroxine (T4), triiodothyronine (T3), 17 beta-estradiol (E2), progesterone (P4) and cholesterol (Chol) during synchronization of the rutting period and gravidity at parallel correlative evaluation of mutual relations of the followed parameters in ten Merino sheep in the seasonal period. Synchronization was achieved by chlorsuperlutin (Agelin--vaginal swabs, Spofa; 20 mg of chlorsuperlutin/swab) and PMSG (500 I. U./animal). Blood was sampled by means of a jugular vein puncture at the time of swab insertion (-13th day) and after three (-10th day) and seven (-7th day) following days, at the removal of swabs and application of PMSG (-3rd day), on the day of insemination (zero day), on the 7th, 14th and 17th day and in the middle of the 2nd, 3rd, 4th and 5th month of gravidity. In the phase of oestrus synchronization a significant increase of E2 concentrations on days -7 and -3 of the experiment (0.47 +/- 0.079 and 0.542 +/- 0.177 nmol.l-1 of serum, P less than 0.001; P less than 0.001) was observed compared to the E2 values on day -13 (0.084 +/- 0.036 nmol.l-1 of serum). Parallel to these observations, marked intermittent changes of T4 (Tab. I, Graph 1) were recorded with the lowest values of this parameter observed on days -10 (41.75 +/- 20.23, P less than 0.05) and -3 (50.22 +/- 18.77, P less than 0.05) and the highest on day -7 (96.77 +/- 17.51 nmol.l-1, P less than 0.01) and day zero (85.40 +/- 19.59 nmol.l-1 of serum, P less than 0.05) in comparison with the -13th day (67.22 +/- 18.29 nmol.l-1 of serum). Concentrations of P4 (Tab. I, Graph 4) declined to the lowest values on day zero observation (0.09 +/- 0.08 nmol.l-1 of serum, P less than 0.05 vs 3.40 +/- 3.61 nmol.l-1 on day -13). No significant changes of concentrations of T3 (Tab. I, Graph 2) and Chol (Tab. I, Graph 5) were observed during oestrus synchronization. During gravidity, concentrations of E2 (Tab. The suppressive effects of three different low dosages of dexamethasone (5, 10 and 15 micrograms kg-1) on serum cortisol concentrations were evaluated in 10 normal cats. On four different days, serum was collected before and at two, four, six and eight hours after the intravenous administration of saline or dexamethasone. Following the administration of saline, no significant difference in mean serum cortisol concentrations was noted between the basal or postinjection values. In contrast, mean serum cortisol concentrations decreased significantly (P less than 0.05) by two hours and remained significantly below mean basal values eight hours after injection of all three dosages of dexamethasone. The degree of cortisol suppression became progressively greater as the dosages of dexamethasone were increased. After administration of the highest dose of dexamethasone (15 micrograms kg-1), serum cortisol decreased to below 5 ng ml-1 by two to four hours and remained suppressed (under 5 ng ml-1) eight hours after injection in all cats. In contrast, two of the 10 cats showed a slight escape from cortisol suppression by eight hours after injection of dexamethasone at the dosage of 10 micrograms kg-1, whereas a dosage of 5 micrograms kg-1 failed to suppress cortisol concentrations below 10 ng ml-1 at any of the sampling times in one cat and was associated with increasing serum cortisol concentrations at eight hours after injection in three cats.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Female llamas ovulate in response to copulation, and progesterone secretion by the corpus luteum indicates recent ovulation (mating) and, or, pregnancy. The plasma progesterone concentration was 0.9 to 1.4 ng/ml in five non-pregnant llamas and 7.4 to 9.2 ng/ml in three llamas in the last month of pregnancy. After ovulation had been induced in nine of 10 llamas by a single intramuscular injection of 500 or 750 iu of human chorionic gonadotrophin, the plasma progesterone concentration increased after two days from 0.5 to 1.2 ng/ml to 4.6 to 10.3 ng/ml after six to nine days and returned to basal values after 10 to 13 days, reflecting the life-span of a corpus luteum in the absence of conception. After a male llama had been introduced into a group of 13 females, 10 matings which resulted in eight conceptions occurred in the first 11 days, and 11 of the llamas became pregnant. The llamas' progesterone concentrations increased after mating and remained high if conception had occurred: 6 to 12 ng/ml in months one to four, and 5 to 9 ng/ml in months five to nine of the 11-month gestation. Two of the 13 llamas had high concentrations of progesterone although they did not become pregnant.  相似文献

Two hundred eighteen ewes were used in experiments 1) to develop a progesterone supplementation regimen capable of sustaining serum concentrations of progesterone at about 2.0 ng/ml for a period of 50 d (Exp. 1) and 2) to determine the effects of progesterone supplementation (d 6 to 50 after mating) on pregnancy and embryo survival rates in mated ewes (Exp. 2). In ovariectomized ewes in Exp. 1, s.c. administration of four cylindrical (9.5 x 60 mm) silastic implants, containing 20% (1.1 g) progesterone by weight, sustained mean serum concentrations of progesterone of 1.9 +/- .07 ng/ml compared with 1.03 +/- .05 ng/ml in ewes bearing two implants. In Exp. 2 each ewe (n = 159) was mated to two fertile rams at a spontaneous estrus (d 0) during mid-breeding season. Mean ovulation rate, determined on a subgroup of 46 ewes, was 1.45 +/- .05. On d 6, ewes were assigned randomly to control (two implants containing no progesterone) or progesterone-treated (four implants similar to those used in Exp. 1) groups. From d 7 to 50 after mating, progesterone concentrations in serum were greater (P less than .001) in progesterone-treated (four implants similar to those used in Exp. 1) groups. From d 7 to 50 after mating, progesterone concentrations in serum were greater (P less than .001) in progesterone-treated (3.50 +/- .06) than in control (2.65 +/- .05) ewes. In six heifers without sexual organs the effects were compared of the superanalogues LH-RH [(D-Tle6) and (D-Trp6)] on the secretion of luteinizing hormone (LH) and follicles of stimulating hormone (FSH). The superanalogues were given i. m. at dosages of 10, 50 and 100 micrograms pro toto. Dosages of 50 and 100 micrograms of both superanalogues had a greater effect on the secretion of LH than the dosage of 100 micrograms. The highest average concentration of LH during the time of application (D-Tle6) was recorded in the dosage of 10 micrograms (7.84 ng.ml-1). The average concentrations of LH after application (D-Trp6) were within the range of 5.46 to 9.29 ng.ml-1. Because of the great variability of the concentrations of LH no significant differences were ascertained due to the influence of dosage. From a comparison of the concentrations of LH with the same dosage it emerged that (D-Trp6) after an application of 50 micrograms significantly increased the concentration of LH from the 60th to the 240th minute and thus had a more protracted effect. With dosages of 10 and 100 micrograms of the superanalogues no statistically significant differences were recorded after two hours from application. With (D-Trp6) the higher stimulative effect on the secretion of LH was statistically confirmed. The highest concentrations of follicles of stimulation hormone (FSH) (204.6 and 228.6 ng.ml-1) were found from the 40th to the 100th minute after the application of dosages of 50 and 100 micrograms (D-Tle6). The protracted effect was greatest with a dosage of 100 micrograms (270 mins). (D-Trp6) at a dosage of 100 micrograms caused the greatest effect on the secretion of FSH (226.8 ng.ml-1 for a period of 180 mins). The lower dosages of analogues scarcely differed in response. The dosage of superanalogues has an influence on the concentration of FSH in peripheral blood and on the duration of the protracted effect.  相似文献

Six ewes of the Merino breed were subjected to radioimmunological assay (RIA) for the study of free 17-beta estradiol during the instillation of vaginal tampons soaked with 30 mg chlormadinone acetate (CAP), and 17 and 34 days after removal of the tampons. On the day before the administration of the preparation, the concentrations of free 17-beta estradiol were comparatively low, ranging within the limits of 38.6 +/- 5.6 pg ml-1 of plasma. These values showed no significant changes during the instillation of the tampons. When the tampons were removed, three periods of increased 17-beta estradiol levels were recorded: 1. on the 1st to 3rd day after removal of the tampons with a peak of 93.2 +/- 26.7 pg ml-1, 2. on the 13th to 16th day with the highest value of 59.7 +/- 23.6 pg ml-1, 3. on the 30th to 33rd day after removal of the tampons 76.2 +/- 23.5 pg ml-1. The results showed that the endocrine activity of the ovaries, expressed by a change in the concentration of plasma 17-beta estradiol after removal of the tampons (CAP), repeated in similar time intervals as under natural conditions and was probably associated with repeated growth and ripening of the follicles of ovaries and their ovulation.  相似文献

Radioimmunological investigation (RIA) of the level of pituitary FSH in the peripheral blood of cows after parturition demonstrated that this level increased successively. The FSH value of 32.93 +/- 3.71 ng per 1 ml, recorded the fourth to sixth day post partum, increased to as much as 57.78 +/- 40.98 ng per 1 ml 40 days after parturition. The LH level changed only slightly over the same period (from 1.12 +/- 0.21 ng per ml to 1.72 +/- 1.15 ng per ml). The concentration of progesterone in the blood of the cows was about 0.40 ng per ml during the first 15 days after parturition, but from the 25th day post partum it trebled, on an average (1.53 +/- 1.19 - 1.59 +/- 1.25 ng per ml). The response of the adenohypophysis of the cows to the administration of 200 micrograms of synthetic gonadoreline (spec. Dirigestran inj. Spofa) increased with increasing length of time from parturition. FSH concentration increased less markedly during the first 28 days p.p. and this increase was not uniform in time (the average increase was 1.5 to three times); in the later period the highest increase 300-500% was recorded, as a rule, 120 minutes after GnRH administration. The situation was similar in the increase in LH concentration in peripheral blood, but after the 26th day post partum the average increase in LH accounted for 500 to 600%. The concentration of progesterone in peripheral blood increased by more than 300%, on an average, from the 25th day after parturition. Experimental studies were conducted into ovariectomized pregnant gilts to establish effects of exogenic hormone administration, with endogenic ovarian steroids excluded, upon uterus and fetus development as well as on hormone levels in blood plasma, endometrium, and allantoic fluid. Hormone concentrations in blood plasma were found to depend clearly on hormone doses applied after ovariectomy to preserve pregnancy. 2 to 3 weeks of smooth gravidity, following ovariectomy, were ensured on the 6th or 14th day after KB1 by daily application of very low doses of progesterone only (80 mg) or in combination with estrogens, the ratio being 480:1.  相似文献

Plasma concentrations of oestradiol-17ß, progesterone, 15-keto–dihydro–PGF and luteinizing hormone (LH) were monitored in llamas and alpacas after mating with an intact male. Concentrations of LH and PGF metabolite were high immediately after copulation. Ovulation occurred in 92% of the animals. The first significant increases in progesterone were recorded on day 4 after mating. In non-pregnant animals the lifespan of the corpus luteum was estimated to be 8–9 days. Luteolysis occurred in association with the release of PGF. In pregnant animals, a transient decrease in progesterone concentrations was observed between days 8 and 18 in both species. No significant changes in PGF secretion were registered during this period. Oes– tradiol–17ß concentrations were high on the day of mating, declined to low values on day 4, and started to increase again on day 8. Peak values after luteolysis in non-pregnant animals were significantly higher than those registered in pregnant ones. Furthermore, concentrations of oestradiol-17ß were elevated for a longer period in non–pregnant than in pregnant animals. The results suggest that progesterone from the corpus luteum exerts a negative influence on follicular activity in pregnant animals by reducing oes– tradiol-17ß secretion.  相似文献

Abstract

The variation in progesterone secretion during the luteal phase and pregnancy in blue fox vixens was analyzed. Progesterone was measured in blood plasma once or twice a week using radioimmunoassay. The material was allocated into three groups; five mated, but barren, blue fox vixens, six mated vixens with implantation zonès in the uterus, but no cubs at parturition, and 26 normally parturient vixens. The progesterone profiles for the three different groups of females showed a steady increase in progesterone immediately after mating. Maximum values were observed on days 8–12 of pregnancy. Then the progesterone levels decreased gradually until delivery around day 52. The levels of progesterone were found to be significantly different (P<0.05) between non-pregnant and pregnant females from day 22 after mating. The plasma progesterone level seems to be affected by the presence of conceptuses.  相似文献

Thirteen female Bedouin goats living in arid land of Algeria Sahara desert were used in this study. These goats were pregnant but they sustained an abortion because of unidentified causes. None of the goats showed any signs of general disease. Plasma concentrations of caprine pregnancy‐associated glycoproteins (cPAGs) and progesterone (P4) were determined during pregnancy using radioimmunoassay. The cPAGs concentration was undetectable (<0.8 ng/ml) throughout the first 2 weeks of gestation. From week 3 after mating, cPAGs concentration was detectable with significant individual variations (p < 0.05) reaching a maximum secretion (436.1 ng/ml). Throughout gestation, cPAGs concentration remained relatively constant but decreased few days before abortion, on an average of 9.2 ± 1.2 days (n = 11), except for two females where the concentrations decreased later (1–2 days before abortion). One or two peaks of cPAGs concentrations (in 4/13 and in 9/13 females, respectively) have been measured few weeks before abortion (77–124 days after mating), when a decline of cPAGs was detected. The P4 concentration increased after mating, and was high from the first week till the end of pregnancy. The P4 concentration (9.1 ± 0.9 ng/ml) decreased rapidly (<0.5 ng/ml) after 4 ± 0.7 days (n = 6) or 9.4 ± 1.6 days (n = 7) before abortion. A positive relationship (p < 0.01) was found between P4 and cPAGs concentrations during gestation. Results indicate that cPAGs and P4 measurements can be used for monitoring gestation and for abortion prediction.  相似文献

The aim of this study was to determine whether blood plasma progesterone is a reliable indicator of pregnancy in mink at an early stage of gestation. We also attempted to establish the threshold value of progesterone as a pregnancy indicator. The analysis was carried out at a production farm on 42 standard female mink aged 1 year, which were grouped both according to the observed success of mating ("mated" and "unmated") and the level of blood serum progesterone measured afterwards ("pregnant" and "nonpregnant"). It was next verified whether a particular female had been assigned to the proper group in the first place. An analysis of accuracy of mating success assessment within the group of unmated females revealed that more than one-third of decisions were wrong, since some females that had been considered unmated ultimately whelped. This suggests that mating supervision by farm workers lacks reliability. A progesterone test for verification of such management decisions should limit the risk of err,or. We suggest that progesterone tests could be carried out using the threshold values 1.9 ng/ml and 20 ng/ml in blood sampled on 25 March and 8 April, respectively, although some level of uncertainty should be taken into account.  相似文献

Sixteen pregnant gilts were ovariectomised (Groups 1 and 2: 11th to 15th days of gravidity, Group 3: 19th to 22nd days of gravidity, Group 4: 34th to 45th days of gravidity). Daily intramuscular injections of 120 mg of progesterone and 250 micrograms of oestradiol benzoate were applied to them, beginning on the day of ovariectomy to preserve gravidity. All experimental animals were sacrificed between the 21st and 25th days of gravidity (Group 1) and between the 31st and 37th days (Groups 2 and 3) as well as between the 52nd and 61st days (Group 4). Living and dead embryos were numerically recorded, and histological als well as biochemical studies were conducted in all uteri. Embryo survival rates were normal, that is between 62.8 and 80 percent in Groups 1, 2, and 3, with the numbers of living embryos being 9.5, 7.5, and 8.0. Gravidity-specific uterus alterations were typical, despite constant hormone substitution. The thickness of endometrium declined with significance over the period under review. Plication was intensified, while the surface epithelium was flattened. Vascularisation increased, particularly in the subepithelial region, and the endometrial stroma became more oedematous. Full functionality was retained by the uterine glands in the period under review (increase in glandular epithelium as well as rising activity of alkaline phosphatase). Significant increase was recorded from the activity of acid phosphatase, whereas glycogen concentrations went down in the myometrium.  相似文献

Female guinea pigs were tested to determine whether they could serve as a model of zearalenone (ZEN) toxicosis during early pregnancy, as observed in domestic swine. Only 1 of 4 female guinea pigs that were given 21 mg of ZEN/kg of body weight orally during the first 8 days after mating was pregnant when examined 22 days after mating. Guinea pigs that were given 7 or 14 mg of ZEN/kg had normal fetal development. Serum concentrations of progesterone were less than 12 ng/ml in all guinea pigs 8 and 15 days after mating. Serum concentrations of progesterone were greater than 100 ng/ml in pregnant guinea pigs on day 22, but remained less than 12 ng/ml in nonpregnant guinea pigs. Three of 5 guinea pigs treated with 20 mg of ZEN/kg and only 1 of 4 guinea pigs given 30 mg of ZEN/kg on days 1 to 3 after mating were pregnant 22 days after mating. Female guinea pigs treated with 20 or 30 mg of ZEN/kg on days 4 to 5 or 6 to 8 after mating and female guinea pigs treated with 60 or 90 mg of ZEN/kg on days 4 and 5 after mating had normal pregnancies. 采用MCD Ⅰ型检侧仪对 16 2 0只海狸鼠进行了早期怀孕诊断 ,并与分娩记录相比较 ,其符合率高 ,最早探测宫血音、胎心音、胎血音时间分别为 8,12 ,16d。宫血音诊断与分娩记录符合率在配种后 10~ 15d ,16~ 2 5d分别为 97 7% ,10 0 % ;未怀孕的符合率为 10 0 %。本试验还采用AlokaSSD 2 10DXⅡ型超声诊断装置 (超声断层扫描、B超 )鉴定了已用MCD Ⅰ型检测仪诊断为怀孕的母海狸鼠 2 3只 ,这两种检测结果一致。超声断层扫描最早可在配种后 8~ 13d对怀孕作出诊断 ,故证明MCD Ⅱ型检测仪检测宫血音的效果是可靠的 ,因此 ,母海狸鼠的早期怀孕诊断可用宫血音阳性和超声断层扫描作为其判定怀孕的主要依据  相似文献

Forty-five ovariectomised gilts in early gravidity received injections of progesterone doses between 120 mg and 40 mg as well as 250 micrograms/die of oestradiol-benzoate and oral application of 12 mg/die of norgestrel, the latter being administered in addition to the 40 mg dose of progesterone. The animals were killed between the 20th and 24th or on the 35th days of gravidity, before endometrial activities were determined of alkaline and acid phosphatases at various points of the uterus (centre and sides of ampullae and in-between ampullae). Under conditions of sufficient progesterone supply (120 mg/die and 100 mg/die), endometrial activities of both alkaline and acid phosphatases in the centre of ampullae were found to be higher than those in-between them. Activities were lower with inadequate progesterone supply (40 mg/die), and differences between points of sampling were less strongly pronounced for alkaline phosphatase. Endometrial glycogen concentrations in the centre of ampullae were lower than those in-between. Values in response to inadequate progesterone administration were lower than those following sufficient supply. With regard to glycogen concentrations, differences between points of sampling in the myometrium were below those in the endometrium.  相似文献

The effect of electroimmobilisation on the plasma concentrations of beta-endorphin/beta-lipotrophin (beta-EP/beta-LPH), cortisol and prolactin (PRL) has been assessed in sheep. Serial blood samples were collected from control and electroimmobilised animals during the first and fourth of a series of four repeated treatments over two days. After electroimmobilisation the mean (+/- SEM) plasma concentrations of beta-EP/beta-LPH increased significantly from 132 +/- 19 pg ml-1 to 545 +/- 111 pg ml-1; the plasma concentrations of cortisol also increased significantly from 22.3 +/- 3.5 ng ml-1 to 108.0 +/- 12.9 ng ml-1. There was no significant change in plasma PRL concentrations after electroimmobilisation and also no significant difference between the plasma concentrations of PRL in the control and electroimmobilised animals. There was no significant difference between the effects of the first and fourth exposure to electroimmobilisation on the plasma concentrations of beta-EP/beta-LPH, cortisol and PRL. These results suggest that the endocrine response to electroimmobilisation may be specific to the pituitary-adrenal axis.  相似文献

The objective of the present study was to compare the concentrations of 17 beta-estradiol, progesterone, cyclic adenosine monophosphate and cyclic quinosine monophosphate in the largest follicles of cows that persist for seven days after insemination following the preceding synchronization of oestrus and superovulation and in follicles of the luteal phase of cycle (5th-10th days). Animals included in the experiment were selected on the basis of rectal examination. Synchronization of oestrus was achieved in 24 crossbreds of Slovak Pied x Lowland Black Pied breeds (SS x Nc) using two doses of cloprostenol of Czechoslovak provenience Oestrophan Spofa, 500 micrograms in each, within 11 days. Serum gonadotrophin at the amount of 2500 I. U. was administered forty-eight hours before administration of the second dose PGF2 alpha. Experimental animals were inseminated after 72 hours. On the 7th day after mating the cows were killed at a slaughterhouse. Evaluated were only the ovaries of the 14 cows in which the persistent large follicles occurred. Ovaries of the 13 control cows in the luteal phase between the 5th-10th days were obtained at the slaughterhouse by the method after Ireland et al. (1980). Correct determination of the phase of sexual cycle was substantiated by determination of progesterone concentrations in blood serum. Follicular fluid was obtained from the largest follicles by aspiration and centrifuged in a cooled centrifuge at 3000 G. The concentrations of 17 beta-estradiol and progesterone in follicular fluid were determined using kits from URVJT at Kosice, designated RIA-test-ESTRA (SI-125-9) or RIA-test-Prog (SI-125-6).2+ persistent follicles (9.15 +/- 5.47 nmol.l-1).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
