Pale, soft, exudative (PSE) pork occurs, for the most part, from environmental stress on the pig. Amino acid intake may be related to stress susceptibility through hormone and neurotransmitter induction. Two experiments were conducted to determine whether supplementation of 5 g of tryptophan (TRP) or 10 g of tyrosine (TYR) per kilogram of a 14% CP diet would alter the response of pigs to stress as measured by hypothalamic neurotransmitter concentrations and incidence of PSE. Twenty-four (Exp. 1) and 36 (Exp. 2) 92-kg pigs were offered one of three diets: control, TRP-, or TYR-supplemented for 5 d before slaughter. Dietary TRP or TYR supplementation in Exp. 1. doubled (P less than .05) plasma TRP and TYR concentrations, respectively, and increased (P less than .05) 5-hydroxytryptamine, dihydroxyphenyl ethylamine, dihydroxyphenyl acetic acid, and homovanillic acid concentrations in the hypothalamus. Pigs that exhibited stress at slaughter had lower (P less than .05) hypothalamic concentrations of epinephrine, norepinephrine, and 5-hydroxytryptamine. In Exp. 2, pigs were trucked 55 km to a commercial meat packing facility and slaughtered without a rest period. This handling procedure was designed to invoke a high incidence of PSE pork and thus be a strong test of treatments. Supplemental dietary amino acids seemed to alter the frequency distribution of the severity of PSE pork. These data indicate that dietary manipulation of amino acid precursors of neurotransmitters may offer a practical means of reducing stress response in swine.  相似文献

This study was conducted to evaluate the effect of dietary vitamin E (VE) on broiler meat quality, especially focused on PSE (pale color, soft and exudative), under chronic heat stress (HS) conditions. Twenty‐eight‐day‐old female Ross broilers were kept in independent cages with a controlled temperature of 24°C (normal temperature: NT) or 30°C (high temperature: HT). The NT chickens were fed basal feed. The HT chickens were fed basal feed (HT) or VE (200 mg/kg) added feed (HT + E). Broilers were weighed and slaughtered at 38 days old. The breast muscle was removed immediately and then the samples were used for determination of meat color, pH, water holding capacity (WHC) and shear force value (SFV). Body weight gain and feed intake were significantly decreased in the HT and HT + E groups compared to the NT group. VE supplementation did not affect the growth performance. Chronic HS at 30°C for 10 days may cause deterioration of meat quality such as PSE. The effects of chronic HS on meat quality were most significant in the toughness of broiler breast meat. Supplementation of VE in broiler feed would be effective to prevent the extent of PSE on broiler meat by chronic HS.  相似文献

1. Textural and rheological differences among broiler breast meat ranging from pale, soft and exudative (PSE) to dark, firm and dry (DFD) in their fresh and frozen (and thawed) forms were investigated. 2. The PSE meat showed significantly higher lightness values and lower pH and water holding capacity values than normal and DFD meats; DFD meat was also significantly different from normal meat. 3. During cooking, PSE meat lost significantly more liquid and produced a softer gel than normal or DFD meats; texture profile analysis parameters were lower for the PSE meat. 4. The storage modulus values (G', rigidity of elastic response of the gelling material) showed that DFD meat produced a more rigid gel during cooking (especially above 54 degrees C) and later during cooling (back to 30 degrees C) compared with the PSE meat. 5. Freezing resulted in a trend of lower G' values before, during and after cooking. The results indicated that meat proteins were damaged during freezing and PSE meat was more severely affected, or that more protein denaturation occurred in the PSE meat.  相似文献

The objective of this study was to determine the effects of lairage time (short vs. long), behavior (aggressive vs. non‐aggressive) and gender (gilts, barrows and boars) on stress and meat quality parameters in pigs (n = 100). In the group with short lairage, aggressive behavior of gilts and boars was observed. At exsanguination, blood samples were collected and levels of blood lactate and plasma cortisol were determined. Post mortem pH value, temperature, drip loss, sensory and instrumental color and meat quality class were determined on the muscle Longissimus dorsi. Higher blood lactate concentration (P < 0.05) and skin blemish score (P <0.001), lower temperature (P < 0.05) and drip loss (P < 0.01), as well as darker, less red and yellow meat color (P < 0.001) were found after long lairage compared to short lairage. Gilts had higher plasma cortisol concentrations than barrows (P < 0.05), while barrows had lower skin blemish scores (P < 0.05) than gilts and boars after long lairage. The highest incidence of PSE (pale, soft and exudative) meat was observed in barrows (100.00%) and boars (91.67%) after short lairage, while the lowest incidence of PSE meat was found in barrows (28.89%) after long lairage. Aggressiveness affected meat temperature, as aggressive pigs had higher meat temperature (P < 0.05) than non‐aggressive pigs.  相似文献

The experimental materials comprised 44 hybrid [female (Polish Large White x Polish Landrace) x male Duroc] growing-finishing pigs. The animals were randomly divided into two groups: 24 pigs were slaughtered immediately after transport and 20 pigs were slaughtered after a 24-hour rest period in the lairage. The meat content of pork carcasses, carcass dressing percentage, the proximate chemical composition, physicochemical and sensory properties of meat and shear force values were determined. Serum creatine kinase activity and cortisol levels were determined in blood samples collected before transport and during carcass bleeding. Pigs slaughtered immediately after transport, compared with those slaughtered after a 24-hour rest period, were characterized by a higher meat content of the carcass and a higher carcass dressing percentage. Pre-slaughter handling had no effect on pork quality. The incidence of normal-quality meat, partially PSE (pale, soft, exudative) meat and PSE meat was similar in both groups. Chemical analysis showed that the content of dry matter, total protein, fat and minerals in meat was comparable in both groups. As regards the functional properties of the pork, samples from the carcasses of pigs that had rested before slaughter had a higher contribution of the red color component. Meat from pigs slaughtered immediately after transport had more desirable sensory properties. Pre-slaughter resting had a significant effect on those analyzed physiological parameters which were found to be good indicators of pre-slaughter stress. Serum creatine kinase activity and cortisol levels were higher in blood samples collected after transport (during carcass bleeding) than in samples collected before transport, pointing to a strong stress response of animals to pre-slaughter treatment. The decrease in serum cortisol levels in blood samples collected during bleeding from the carcasses of pigs slaughtered after a 24-hour rest period, compared with samples collected from animals slaughtered immediately after transport, suggests that rest before slaughter alleviated stress induced by pre-slaughter handling operations.  相似文献

Keeping pigs in lairage provides a buffer to supply the slaughter line and allows the pigs to recover from the stress of transport. It has implications for the pigs' welfare by influencing their behaviour and ease of handling. In terms of pork quality, optimal lairage times for most pigs in the UK are probably one to three hours. Shorter lairage times are associated with more pale, soft, exudative (PSE) meat and longer times are associated with more dark, firm, dry (DFD) meat, more skin blemishes caused by fighting, and a lower carcase yield. However, when pigs have not been previously stressed by handling procedures, resting them in lairage may have no beneficial effects on the pigs' welfare or meat quality. At very high environmental temperatures there is evidence that a holding period of less than one hour may help to reduce the number of PSE carcases. At temperatures above 10 degrees C showering the pigs with water is beneficial. Fasting times need to be controlled to minimise the potential loss of carcase yield while reducing the volume of gut contents to reduce the risk of carcase contamination. Lairages can act as reservoirs of infection by pathogenic bacteria and there is evidence that longer holding times increase the risk of cross-contamination.  相似文献

The Chato Murciano pig is a breed catalogued as a Special Protection breed in danger of extinction. At present it is commonly reared in an intensive system. We have studied the mineral composition and other characteristics of its carcass and meat. Samples from the Musculus longissimus lumborum were taken from a total number of 43 animals, divided into two groups, Chato Murciano and Chato Murciano crossed with Iberian, reared in a conventional indoor system. The resulting meat of the Chato Murciano pig and its cross with Iberian was especially rich in iron and copper, rich in phosphorus, and marginally poor in calcium and sodium. The evolution of the pH of the carcass was that of pigs which do not produce PSE (pale, soft and exudative) meat. Some relevant positive correlation coefficients between minerals (P ≤ 0.05) have been found.  相似文献

The influence of transport stress on rectal temperature, respiratory rate and serum concentrations of cortisol, thyroxine and triiodothyronine (T3) in normal pigs (NN) and in heterozygous (Nn) pigs with Halothane gene (n), a major gene that causes the pale, soft, exudative meat (PSE), were examined in a hot environment (32–34°C). Easily releasable myofilament content of Longissimus dorsi taken by needle biopsy was also measured as an index of myofibrillar disassembly. Rectal temperature and respiratory rate were significantly increased by the transport. However, the influence was higher (P < 0.01) in Nn pigs. Serum concentration of cortisol was increased (P < 0.01) by the transport despite the genotypes. Serum concentration of T3 was not changed by the transport in Nn pigs, whereas in NN pigs it was significantly decreased after the transport (P < 0.01). Easily releasable myofilament content after the transport was higher in Nn pigs than that of NN pigs (P < 0.05). These results suggest that response of thyroid function might relate to the stimulation of muscle proteolysis and PSE in Nn pigs under stressful conditions.  相似文献

The objectives of the current experiment were to determine whether boars heterozygous for the mutation in skeletal ryanodine receptors (sRyR), known to cause porcine stress syndrome, differed from wild-type boars in hypothalamic-pituitary-adrenal axis (HPA) function. We have examined basal plasma ACTH, cortisol, and corticosteroid-binding globulin (CBG) concentrations; plasma ACTH and cortisol responses to a nose-snare stressor and at slaughter; dexamethasone suppression of plasma ACTH and cortisol concentrations; and glucocorticoid receptor (GR) density in the pituitary gland, hippocampus, hypothalamus, and frontal cortex. We have also examined carcass yields, composition, and meat quality to determine whether differences in HPA activity were accompanied by an increased incidence of meat quality characteristics associated with pale, soft, exudative (PSE) meat. Thirty boars either heterozygous or wild-type (n = 15 per genotype) for mutated sRyR were tested for HPA function at 7 mo of age. Heterozygous boars had lower basal plasma ACTH (P < .05) and cortisol (P < .04) concentrations. Integrated basal plasma ACTH and cortisol levels were also lower (P < .05 and P < .005, respectively). Genotype had no significant effect on basal CBG, stressor-induced (nose snare or slaughter) or dexamethasone suppression of plasma ACTH or cortisol concentrations. No differences in immunoreactive GR levels were found in the pituitary gland or any brain region examined. We did find a significant, negative correlation (r = -.62, P < .02) between peak (0800) basal plasma ACTH concentrations and hippocampal GR levels. The alterations in basal HPA function in heterozygous boars were accompanied by lighter body weights (P < .03), decreased carcass fat depth (P < .04), and increased carcass lean yields (P < .02). There was a higher incidence of meat quality characteristics associated with PSE meat in heterozygous boars indicated by higher carcass temperatures (P < .04) and meat brightness (P < .0001) with lower carcass pH at slaughter (P < .03) and after chilling (P < .003). In conclusion, we have found differences in basal and not stressor-induced HPA function between boars heterozygous and wild-type for mutated sRyR. This altered basal HPA activity was accompanied by an increased incidence of meat quality aspects associated with PSE meat in heterozygous boars.  相似文献

A total of 567 young pigs entering one of the national testing stations were tested for sensitivity to halothane between October 1976 and May 1978. They included 443 Landrace, 76 Large White and 48 Irish Welsh. The average weight at testing was 25.6 kg. A total of 24 pigs reacted positively to the test, 21 (4.6%) of the Landrace and 3 (6.3%) of the Irish Welsh. No reactors were found in the Large White breed.Meat quality estimates in the form of the pH in the longissimus dorsi muscle 45 min post-mortem (pH1) were recorded on the 324 slaughtered pigs. Reactor pigs (ten) had significantly lower values than non-reactors, indicating a greater tendency to develop quality problems such as pale, soft, exudative (PSE) meat. There was an indication that reactor pigs might have slightly reduced daily gains and lower backfat thickness.  相似文献

A study was performed on the distribution of catecholamines (noradrenaline, dopamine and adrenaline) and L-DOPA in the hypothalamo-hypophysial region, as well as in the region of n. caudatus, which participate in the control of sexual activities in sheep. After isolation in the activated aluminium oxide, the catecholamine concentration was determined spectrophotometrically. Hypothalamus was divided into three regions: rostral, medial and caudal. In the same regions of the cerebroneural system (CNS), the activity of the degradation enzyme monoaminooxidase (MAO) was determined by radiochemical method. 14C-tryptamine was as a substrate and the results were measured by means of scintillation spectrophotometer Packard. The distribution of catecholamines and L-DOPA in the hypothalamus of sheep is different. The largest proportion is that of noradrenaline in caudal hypothalamus (1.84 +/- +/- 0.36). The dopamine levels in hypothalamus are quite balanced though substantially lower than those of noradrenaline (from 0.22 to 0.31 micrograms/g). The concentrations of adrenaline and L-DOPA in the sheep hypothalamus are low. The sheep hypophysis contains more noradrenaline (1.70 +/- 0.38), adrenaline content amounts to 1.30 +/- 0.28. L-DOPA and dopamine occur in this region at low concentrations. In comparison with the other regions of sheep brain, high dopamine concentrations (2.0 +/- 0.58 micrograms/g) and higher L-DOPA levels were determined in n. caudatus. The activity of the degradation enzyme monoaminooxidase in the cerebroneural system of sheep is different. The purpose of the present study was to investigate the variations in histochemical characteristics of muscle samples segregated according to metabolic rates (MR) and pork quality attributes. A total of 231 crossbred Duroc x (Yorkshire x Landrace) pigs was evaluated. Samples of the LM were taken to evaluate histochemical characteristics, postmortem MR, and meat quality. Samples were classified into fast, normal, and slow MR groups based on muscle pH at 45 min and R-value. Drip loss and lightness (L*) were used to assign samples to 1 of 4 quality classes. Pale, soft, and exudative pork belonging in the fast group had the greatest (P < 0.05) percentage of type IIb fibers, and RSE (reddish-pink, soft, and exudative) pork belonging in the fast group had a similar tendency. Additionally, RFN (reddish-pink, firm, and nonexudative) pork belonging in the normal group showed a lower (P < 0.05) percentage of type IIb fibers than PSE or RSE, regardless of MR, and DFD pork had the lowest (P < 0.05) percentage of type IIb fibers. In general, the fast-glycolyzing PSE pork with the lowest pH at both 45 min and 24 h had greater percentages of type IIb fibers than the fast-glycolyzing RFN pork. There were more fiber-type composition differences between quality classes in pork undergoing a fast rate of metabolism compared with pork undergoing a normal rate of metabolism. It can be concluded that muscle histochemical characteristics are associated with early postmortem MR, the extent of glycolysis, and, thereby, pork quality; however, these effects are limited to the pigs exhibiting a fast glycolytic rate.  相似文献

Throughout the chicken production chain, transport from farm to the commercial abattoir is one of the most critical sources of stress, particularly heat stress. The aim of this work was to describe the performance of a new prototype truck container designed to improve the microenvironment and reduce the incidence of pale, soft and exudative (PSE) meat and dead on arrival (DOA) occurrences. Experiments were carried out for four different conditions: regular and prototype truck, both with and without wetting loaded cages at the farm (for bird thermal stress relief) just before transporting. While there was no difference in the DOA index (P ≥ 0.05), the prototype truck caused a reduction (P < 0.05) in the occurrence of PSE meat by 66.3% and 49.6% with and without wetting, respectively. The results of this experiment clearly revealed a low‐cost solution for transporting chickens that yields better animal welfare conditions and improves meat quality.  相似文献

Glycolytic potential of red, soft, exudative pork longissimus muscle   总被引:7,自引:0,他引:7  
Red, soft, exudative (RSE) pork is red as "normal" pork and exudative as PSE pork. The origin of RSE quality is unknown. In the present study, we determined whether the occurrence of RSE pork is related to the presence of the RN (Rendement Napole) gene. A glycolytic potential [= 2 x (glucose + glycogen + glucose-6-phosphate) + lactate] of > 180 micromol lactate/g of meat was used as indicator for presence of the RN gene. At 1 d postmortem, pork loin samples were collected at a commercial slaughter plant, and meat quality characteristics and glycolytic potential were determined. Glycolytic potential of RSE pork (n = 42) was higher (P < .01) than that of RFN (red, firm, nonexudative) pork (n = 25): 137 vs 110 micromol lactate/g of muscle. The glycolytic potential of PSE pork (n = 28) was 161 micromol lactate/g of muscle, which was higher (P < .01) than the glycolytic potential of RFN or RSE pork. Using a glycolytic potential of 180 micromol lactate/g of muscle as suggestive for the presence of the RN gene, four (10%) of the RSE samples were from RN carriers. By the same criterion, the PSE group contained nine (32%) RN gene carriers. These results suggest that the occurrence of RSE quality is not related to the presence of the RN gene.  相似文献

Sixty-nine entire male pigs with different halothane genotype (homozygous halothane positive — nn-, n = 36; and homozygous halothane negative — NN-, n = 33) were fed with a supplementation of magnesium sulphate (Mg) and/or l-tryptophan (Trp) in the diet for 5 days before slaughter. Animals were housed individually and were submitted to stressful ante mortem conditions (mixed in the lorry according to treatments and transported 1 h on rough roads). Individual feed intake was recorded during the 5-d treatment. At the abattoir, pig behaviour was assessed in the raceway to the stunning system and during the stunning period by exposure to CO2. Muscle pH, colour, water holding capacity, texture and cathepsin activities were determined to assess meat quality. The number of pigs with an individual feed intake lower than 2 kg/day was significantly different among diets (P < 0.05; Control: 8.7%; Mg&Trp: 43.5%; Trp: 17.4%) and they were considered to have inadequate supplement intake. During the ante mortem period, 15.2% of pigs included in the experiment died, and this percentage decreased to 8.7% in those pigs with a feed intake > 2 kg/day, all of them from the stress-sensitive pigs (nn). In general, no differences were observed in the behaviour of pigs along the corridor leading to the stunning system and inside the CO2 stunning system. During the stunning procedure, Trp diet showed shorter periods of muscular excitation than control and Mg&Trp diets. The combination of a stressful ante mortem treatment and Mg&Trp supplementation led to carcasses with high incidence of severe skin lesions. Different meat quality results were found when considering all pigs or considering only those with adequate supplement intake. In this later case, Trp increased pH45 (6.15) vs Control diet (5.96) in the Longissimus thoracis (LT) muscle (P < 0.05) and pH at 24 h (Trp: 5.59 vs C: 5.47) led to a higher incidence of dark, firm and exudative (DFD) traits in SM muscle (P < 0.05). Genotype affected negatively all the meat quality traits. Seventy-five percent of LT and 60.0% of the SM muscles from nn pigs were classified as pale, soft and exudative (PSE), while none of the NN pigs showed these traits (P < 0.0001). No significant differences were found between genotypes on the incidence of DFD meat.Due to the negative effects observed in the Mg&Trp group in feed intake and carcass quality, the utilization of a mixture of magnesium sulphate and tryptophan is not recommended.  相似文献

Although a number of problems in connection with the use of shot biopsy have yet to be solved, it can be stated that by this technique biopsy samples of sufficient quantity (up to 1.5 g) can be obtained from the musculus longissimus dorsi of pigs of about 80 kg liveweight. The technique offers data on the metabolic status (biophysical and biochemical values) of muscles, provided that the postmortal processes have been simulated (incubation of bioptate). The use of sensitivity tests on isolated subcellular fractions of the bioptate can lead to more precise results and consequently, to a remarkable reduction in the incidence of PSE syndrome (pale, soft, exudative meat). The first results achieve under conditions of Slovakia (Czechoslovakia) in testing boars show that this method could find a permanent use in the practice. Research institutions in a number of countries are now investigating possibilities of using shot biopsy for prediction of skeletal muscle disorders in pigs and it is excepted that many new results will be published soon.  相似文献

The occurrence of qualitative changes in meat was studied in a set of 1008 pigs slaughtered in four sanitation slaughter houses for the period of 12 months. PSE meat was found in 206 pigs (20.43 per cent) and DFD meat only in 9 pigs (0.89 percent) of the total number of pigs examined. The rule was applied that the defect should be demonstrated by two of three commonly used parameters (the pH value, colour and water loss by dripping). The highest occurrence of qualitative changes in meat of PSE type was registered in the group of animals with a defect of the respiratory organs (32.53 per cent) and the lowest in animals with diseases of sexual organs, including parturition complications (5.00 per cent). A similar trend was recorder even in the DFD meat occurrence. As to the seasons, the highest percentage of occurrence of PSE defects was in July and August (35.41 and 29.06 per cent) and the lowest (8.64 and 10.97 per cent) in March and January. The actual disease is proved to be only one of the secondary stress to induce the PSE meat formation.  相似文献

Meat production with minimum animal suffering is a humanitarian concern. Thus, the objective of this work was to observe the performance of Cobb broiler chickens from 7 to 46 days of age when raised under different installations: dark house system (DHS), conventional yellow system (CYC) and conventional blue system (CBC). The feed conversion ratio for the birds raised on the DHS was 3.8% and 2.7% lower than those for the CYC and CBC systems, respectively. Compared with the CYC and CBC systems, average daily gain under the DHS was 11.4% and 9.3% higher, respectively, and body weight at 46 days was 11.4% and 9.3% higher, respectively (P ≤ 0.05). The birds' welfare was assessed based on their stress by determining the amount of pale, soft and exudative (PSE) meat in the breast fillets. The CYC and CBC birds had 24.3% and 25.3% PSE meat, respectively, whereas the DHS birds had 37.0%. We concluded that the DHS has a greater potential to produce broiler chickens, with superior performance to conventional systems, despite the higher stress faced by the birds during the maneuvers just before slaughter.  相似文献

White light from a xenon arc was focused onto the upper surface of 25-mm-thick transverse sections of pork longissimus muscle. A servo motor moved an optical fiber across the lower surface of the muscle to collect transmitted light, which then was passed through a grating monochromator and onto a photomultiplier for spatial measurements of scattering (SMS) and transmittance spectra. The SMS were calculated as the slope of the logarithm of transmittance relative to path length through the sample (which was calculated trigonometrically). Pale, soft, exudative (PSE) pork was measured with an index that included a subjective evaluation of meat color and objective measurements of reflectance, drip loss, and centrifugation fluid loss. The strongest correlation of SMS with PSE was at 610 nm (r = .86, P less than .005) and the strongest correlation of transmittance with PSE was at 650 nm (r = -.95, P less than .005). This supports the use of a red laser at 633 nm for the detection of PSE pork.  相似文献

Twenty-four barrows were used to investigate the effects of exogenous porcine somatotropin (pST) administration (0 and 100 micrograms.kg-1.d-1) between 30 and 60 kg on longissimus muscle morphology and meat tenderness of pigs grown to 90 kg. Administration of pST was by daily i.m. injection. Pigs were fed a fortified diet in restricted amounts between 30 and 60 kg and had ad libitum access to this diet from 60 to 90 kg. Excipient pigs had fewer alpha R fibers and more alpha W fibers than pST-treated pigs (P less than .05). Administration of pST increased (P less than .05) muscle fiber area for all three fiber types at both the 60 kg (34%) and 90 kg (29%) slaughter weights. Furthermore, pST administration increased shear-force (30% at 60 kg; 19% at 90 kg) of the longissimus muscle, indicating that pST administration reduced meat tenderness. All the pST-treated pigs exhibited the "giant fiber syndrome" and a high proportion (62%) of pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomaly resulted from increased muscle protein accretion rate or fiber degeneration is unknown. Results indicated that the stimulatory effects of pST on muscle growth are sustained following cessation of hormone treatment. However, the incidence of pale, soft, exudative muscle in 62% of the pST-treated pigs indicates that pST potentially has undesirable side effects.  相似文献
