A comparative analysis of ethanol extracts from peel, pulp and seed of Prunus persica var. platycarpa (Tabacchiera peach) was done. The total phenol, flavonoid and carotenoid content as well as the antioxidant properties by using different in vitro assays (DPPH, ABTS, FRAP, Fe-chelating, β-carotene bleaching test) were evaluated. Pulp extract was subjected to liquid chromatography-electrospray-tandem mass spectrometry (HPLC-ESI-MS/MS). Gallic acid, protocatechuic acid, protocatechualdehyde, chlorogenic acid, p-coumaric acid, and ferulic acid were identified as main constituents. Pulp extract was characterized by the highest total phytonutrients content and exhibited the highest antioxidant activity in all in vitro assays (IC50 values of 2.2 μg/mL after 60 min of incubation by using β-carotene bleaching test and 2.9 μg/mL by using Fe-chelating assay). Overall, the obtained results suggest that P. persica var. platycarpa displays a good antioxidant activity and its consumption could be promoted.  相似文献

The bioactive materials in brown seaweeds hold great interest for developing new drugs and healthy foods. The oil content in brown seaweeds (Saccharina japonica and Sargassum horneri) was extracted by using environmentally friendly supercritical CO2 (SC-CO2) with ethanol as a co-solvent in a semi-batch flow extraction process and compared the results with a conventional extraction process using hexane, ethanol, and acetone mixed with methanol (1:1, v/v). The SC-CO2 method was used at a temperature of 45 °C and pressure of 250 bar. The flow rate of CO2 (27 g/min) was constant for the entire extraction period of 2 h. The obtained oil from the brown seaweeds was analyzed to determine their valuable compounds such as fatty acids, phenolic compounds, fucoxanthin and biological properties including antioxidant, antimicrobial, and antihypertension effects. The amounts of fucoxanthin extracted from the SC-CO2 oils of S. japonica and S. horneri were 0.41 ± 0.05 and 0.77 ± 0.07 mg/g, respectively. High antihypertensive activity was detected when using mixed acetone and methanol, whereas the phenolic content and antioxidant property were higher in the oil extracted by SC-CO2. The acetone–methanol mix extracts exhibited better antimicrobial activities than those obtained by other means. Thus, the SC-CO2 extraction process appears to be a good method for obtaining valuable compounds from both brown seaweeds, and showed stronger biological activity than that obtained by the conventional extraction process.  相似文献

Gloiopeltis tenax (G. tenax) is widely distributed along the Chinese coastal areas and is commonly used in the treatment of diarrhea and colitis. This study aimed at investigating the bioactivities of the volatile constituents in G. tenax. We extracted the essential constituents of G. tenax by supercritical carbon dioxide extraction (CO2-SFE), then identified and analyzed the constituents by gas chromatography-mass spectrometry (GC-MS). In total, 30 components were identified in the G. tenax extract. The components showed remarkable antioxidant activity (radical scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH)), lipid peroxidation inhibition capacity (in a β-carotene/linoleic acid-coupled oxidation reaction), and hydroxyl radical-scavenging activity (by deoxyribose degradation by iron-dependent hydroxyl radical), compared to butylated hydroxytoluene. In microdilution assays, G. tenax extracts showed a moderate inhibitory effects on Staphyloccocus aureus (minimum inhibitory concentration (MIC) = 3.9 mg/mL), Enterococcus faecalis (7.8 mg/mL), Pseudomonas aeruginosa (15.6 mg/mL), and Escherichia coli (3.9 mg/mL). Antioxidant and antimicrobial activities of G. tenax were related to the active chemical composition. These results suggest that the CO2-SFE extract from G. tenax has potential to be used as a natural antioxidant and antimicrobial agent in food processing.  相似文献

Microalgae are well known for their biotechnological potential, namely with regard to bioactive lipidic components—especially carotenoids and polyunsaturated fatty acids (PUFA), well-known for therapeutic applications based on their antioxidant capacity. The aim of this work was to evaluate the influence of four distinct food-grade solvents upon extractability of specific lipidic components, and on the antioxidant capacity exhibited against both synthetic (2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+•)) and biological reactive species (O2 and NO). A eukaryotic microalga (Scenedesmus obliquus (M2-1)) and a prokaryotic one (Gloeothece sp.) were used as case studies. Concerning total antioxidant capacity, the hexane:isopropanol (3:2) and acetone extracts of Sc. obliquus (M2-1) were the most effective against DPPH and ABTS+•, respectively. Gloeothece sp. ethanol extracts were the most interesting scavengers of O2, probably due the high content of linolenic acid. On the other hand, acetone and hexane:isopropanol (3:2) extracts were the most interesting ones in NO assay. Acetone extract exhibited the best results for the ABTS assay, likely associated to its content of carotenoids, in both microalgae. Otherwise, ethanol stood out in PUFA extraction. Therefore, profiles of lipidic components extracted are critical for evaluating the antioxidant performance—which appears to hinge, in particular, on the balance between carotenoids and PUFAs.  相似文献

Surface-associated marine bacteria are an interesting source of new secondary metabolites. The aim of this study was the isolation and identification of epiphytic bacteria from the marine brown alga, Bifurcaria bifurcata, and the evaluation of the antioxidant and antimicrobial activity of bacteria extracts. The identification of epiphytic bacteria was determined by 16S rRNA gene sequencing. Bacteria extracts were obtained with methanol and dichloromethane (1:1) extraction. The antioxidant activity of extracts was performed by quantification of total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and oxygen radical absorbance capacity (ORAC). Antimicrobial activities were evaluated against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Salmonella enteritidis, Staphylococcus aureus, Saccharomyces cerevisiae and Candida albicans. A total of 39 Bifurcaria bifurcata-associated bacteria were isolated and 33 were identified as Vibrio sp. (48.72%), Alteromonas sp. (12.82%), Shewanella sp. (12.26%), Serratia sp. (2.56%), Citricoccus sp. (2.56%), Cellulophaga sp. (2.56%), Ruegeria sp. (2.56%) and Staphylococcus sp. (2.56%). Six (15.38%) of the 39 bacteria Bifurcaria bifurcata-associated bacteria presented less than a 90% Basic Local Alignment Search Tool (BLAST) match, and some of those could be new. The highest antioxidant activity and antimicrobial activity (against B. subtilis) was exhibited by strain 16 (Shewanella sp.). Several strains also presented high antimicrobial activity against S. aureus, mainly belonging to Alteromonas sp. and Vibrio sp. There were no positive results against fungi and Gram-negative bacteria. Bifurcaria bifurcata epiphytic bacteria were revealed to be excellent sources of natural antioxidant and antimicrobial compounds.  相似文献

Activators of sesquiterpene synthase (STS) gene expression and sesquiterpene production in Piper betle L. were examined using quantitative real time PCR and gas chromatography mass spectrometry methods, and the allelopathic activity of untreated and Fusarium solani-treated betel extracts was tested on seed germination and on the shoot and root growth of Thai rice variety PSL2 (Oryza sativa cv. Phitsanulok 2) and three dominant paddy weeds (Eclipta prostrata, Echinochloa crus-galli and Chloris barbata). The results demonstrated that F. solani dramatically upregulated STS expression and productions of β-cubebene, β-caryophyllene and germacrene D sesquiterpene when compared with the untreated control, and that betel extracts had a greater inhibitory effect on weeds than on rice. The effects were more clearly detected on seed germination and root growth than on shoot growth, and they were found to be dose-dependent. It is also noted that F. solani-treated extract had stronger effects than the untreated extract. The species most sensitive to the allelopathic effects was C. barbata, germination of which was completely inhibited even at a dose of 0.1 mg/mL untreated extract. With regards to rice, although betel extract at 1.0 mg/mL showed no inhibition on germination, it affected the elongation of rice roots, in addition to those of the tested weeds. The obtained data suggested that F. solani has potential as an activator of sesquiterpene allelochemical production via STS expression, the latter leading to the treated betel extract having a stronger phytotoxic effect. These results were beneficial in the promotion of natural herbicide production using biotechnology.  相似文献

Indole derivatives including bromoindoles have been isolated from the South Pacific marine sponges Rhopaloeides odorabile and Hyrtios sp. Their structures were established through analysis of mass spectra and 1D and 2D NMR spectroscopic data. Their potential inhibitory phospholipase A2 (PLA2), antioxidant and cytotoxic activities were evaluated. The new derivative 5,6-dibromo-l-hypaphorine (9) isolated from Hyrtios sp. revealed a weak bee venom PLA2 inhibition (IC50 0.2 mM) and a significant antioxidant activity with an Oxygen Radical Absorbance Capacity (ORAC) value of 0.22. The sesquiterpene aureol (4), also isolated from Hyrtios sp., showed the most potent antioxidant activity with an ORAC value of 0.29.  相似文献

The ectomycorrhizal (ECM) symbiosis that develops between the roots of host trees and the soil ECM fungi is an important factor towards the survival, health and growth of these trees, as it stimulates their water and nutrient uptake. Ectomycorrhizal colonization can result in the deposition of phenolic compounds in peripheral cortex cells and a similar answer can be recognized as one way of plant defense against pathogenic infections. The aim of the present work was the evaluation of antioxidant potential of the ectomycorrhizal fungi, Paxillus involutus and Pisolithus arhizus, in presence and absence of the symbiont - Pinus pinaster - in response to the symbiotic association, under different contact periods (45 days and 48 h). Phenolic contents in mycelia, culture media and plant leaves, stems and roots were determined by Folin-Ciocalteu assay, and their antioxidant properties were evaluated by three in vitro assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, reducing power and lipid peroxidation inhibition through β-carotene bleaching inhibition. Comparing the response of the symbionts to the host species, P. pinaster benefited most with the association, because it generally decreases the phenolics content and the antioxidant activity values in the presence of both ectomycorrhizal fungi along time. P. arhizus also decreased, in general, its phenolics levels and antioxidant properties, alongside with P. pinaster, unlike P. involutus that did not have the same response as it increases its content of phenolics and some of its values of antioxidant activity. These results can be considered as an hypothetical signal of a symbiotic differential compatibility of mycorrhizal fungi for a host.  相似文献

Dried green leafy vegetables constitute a major dietary source of provitamin A carotenoids for people living in semi-arid areas of Tanzania. In the present study, eight vegetable varieties commonly consumed in these areas were either traditionally sun-dried or solar-dried, and the amount of provitamin A carotenoids were determined by HPLC. In the fresh blanched leaves, the contents of -carotene, -carotene, 9-cis--carotene and 13-cis--carotene were in the range of 526–917, 12–39, 80–136 and 16–40 g/g dry matter/basis; dmb, respectively. The average amounts of all-trans--carotene in the leaves after blanching, solar drying and open sun-drying were 662, 502 and 353 g/g dmb, respectively, all significantly different from each other (p <0.001). The average amounts of all-trans--carotene were 21, 15 and 8 g/g dmb (p <0.05) and of 9-cis--carotene 101, 82 and 63 g/g dmb (p <0.05) in the blanched, solar-dried and open sun-dried samples, respectively. Estimation of the amount of retinol equivalents provided by the dried vegetables for preparation of an edible portion of 100 g relish showed that solar-dried vegetables contributed more (between 120–275%) than the recommended level of vitamin A, and on average 43% more retinol equivalents than relishes of open sun-dried vegetables.Thus, it is concluded that the amount of provitamin A-carotenes in traditionally treated vegetables were highly reduced by open sun-drying while solar drying resulted in significantly more retention of the carotenes. All-trans--carotene and 9-cis--carotene were similarly affected as all-trans--carotene by the two drying methods. Consumption of a 100 g portion of solar-dried vegetable relish could provide the recommended daily intake of vitamin A.  相似文献

To test the feasibility of dry milling oats (Avena sativa L.) to concentrate antioxidant activity and phenolic antioxidants, groats were pearled for 5 to 180 s. These treatments removed <1 to 15% of the weight. The material obtained from short pearling times was mostly bran. Longer pearling times increased the amount of starchy endosperm in the pearlings. Antioxidant activity of 80% ethanol extracts, measured by β-carotene bleaching and by reduction of the free radical, 2,2-diphenyl-1-picrylhydrazyl, was highest in the short-pearling-time fractions and decreased as more endosperm tissue was included. Likewise, there was a decreasing concentration of total phenolics, determined colorimetrically, and of several simple phenolic acids, determined by high performance liquid chromatography, as more material was pearled from the groats. In contrast, concentrations of avenanthramides were not correlated with pearling time, indicating that they were more uniformly distributed in the groats.  相似文献

The availability of -carotene from Spirulina as compared to standard all trans -carotene was studied by the liver and kidney vitamin A storage method. After 21 days of vitamin A depletion, the rats were repleted with -carotene from Spirulina and a standard source at two dietary levels (60 and 120 µg/day) for a 10 day period. At lower levels, the liver storage levels of vitamin A and the percent of -carotene absorption were comparable to those of the standard. At higher levels both these parameters of the Spirulina fed group were significantly (P<0.01) inferior to the standard source fed group. However, the Spirulina fed group showed better (P<0.05) growth than the standard fed group did at both low and high levels of feeding.  相似文献

Common beans are rich in phenolic compounds, which can provide health benefits to the consumer. The objective of this work was to study the relationship among antimutagenicity, antioxidant and enzymatic activities of methanolic extract and trolox by principal components multivariate analysis. Antimutagenicity of phenolic compounds present in methanolic extract from the seed coat of common beans (P. vulgaris, Flor de Mayo Bajío cultivar) and trolox against AFB1 mutagenicity were evaluated in the Salmonella typhimurium microsuspension assay. Antioxidant capacity of methanolic extract and trolox were evaluated using β-carotene and 1,1-diphenyl-2-picryhydrazyl (DPPH) in vitro model assays. Cythrome P450 activity was measured by fluorometric assay. For phenolic extract, trolox and phenolic extract + trolox, the inhibition on AFB1 mutagenicity in tester strain TA100 was 47, 59 and 69%, respectively. While in TA98 was 39, 48 and 68%. The inhibition of phenolic compounds, trolox and phenolic compounds + trolox on cytochrome P450 (CYP450) activity was 48, 59 and 88%, respectively. Correlation analysis showed that phenolic extract and trolox have high antimutagenic and antioxidant activity and also inhibited enzymatic activity. The results suggest that the primary mechanism of action of phenolic compounds in beans against AFB1 mutagenicity may be extra-cellular in the microsuspension assay.  相似文献

Carotenoids and phenolic profile, antioxidant activity as well as concentrations of selected macronutrients (K, N, Mg, Ca and Na) and micronutrients (Zn, Cu and Mn) in flesh and peel of peach fruit were recorded at two harvest dates. Predominant mineral was potassium, followed by calcium, magnesium and sodium. The concentration of most micronutrients was greater in the peel than in the flesh especially in early season. The concentration of most elements in flesh and peel decreased during fruit maturation. Total carotenoids content varied with respect to the cultivar. β-cryptoxanthin and β-carotene were the major carotenoids in both tissues and flesh contain the lowest amounts. Neochlorogenic acid, chlorogenic acid, catechin, epicatechin, gallic acid, rutin, quercetin-3-O-galactoside, cyanidin-3-O-glucoside, cyanidin-3-O-rutinoside, were detected in both peel and flesh, with chlorogenic acid and catechin being the predominant components. Peel extracts showed markedly higher antioxidant activities, when estimated by ABTS or DPPH assays, than the flesh counterparts, consistent with the observed higher phenolic content. Overall, total phenolics levels increased at full ripening stage in both peel and flesh. The results found herein provide important data on carotenoids, phenolic and macro- and micronutrient changes during fruit growth, and emphases peach fruit as a potential functional food.  相似文献

-carotene in leaf protein extracted from some species is destroyed in a few days when incubated with salt as a preservative. Its shelf-life can be greatly extended by pretreatment with oxalate, ascorbate mercaptans, phenolic substances, extracts made from leaves in which carotene is more stable, and by tight compaction. Treatment with alum reverses the protective effect of these chemical pretreatments, it also increases the lability of carotene in leaf protein extracted from species which usually give a relatively stable product. In these preparations of leaf protein, which were heated to 90 °C during preparation, carotene is only partially extractable into some solvents which are miscible with water. It seems therefore to be present in more than one state, but differences in extractability could not be correlated with differences in lability.  相似文献

A growing market for novel antioxidants obtained from non-expensive sources justifies educated screening of microalgae for their potential antioxidant features. Characterization of the antioxidant profile of 18 species of cyanobacteria (prokaryotic microalgae) and 23 species of (eukaryotic) microalgae is accordingly reported in this paper. The total antioxidant capacity, accounted for by both water- and lipid-soluble antioxidants, was evaluated by the (radical cation) ABTS method. For complementary characterization of cell extracts, a deoxyribose assay was carried out, as well as a bacteriophage P22/Salmonella-mediated approach. The microalga Scenedesmus obliquus strain M2-1 exhibited the highest (p > 0.05) total antioxidant capacity (149 ± 47 AAU) of intracellular extracts. Its scavenger activity correlated well with its protective effects against DNA oxidative damage induced by copper(II)-ascorbic acid; and against decay in bacteriophage infection capacity induced by H2O2. Finally, performance of an Ames test revealed no mutagenic effects of the said extract.  相似文献

Total antioxidant capacities, 2,2-diphenyl-1-picrylhydrazyl (DPPH·), hydroxyl (HO·), scavenging activities, and total phenolic values were determined in extracts of Cucurbita pepo L. female and male flowers. Powdered C. pepo L. samples were extracted in aqueous ethyl acetate (EA: W1, 17:3), ethanol (E), and water (W) by agitating in magnetic stirrer for 80°C, 15 min and also by in aqueous ethyl acetate (EA: W2, 17:3) at 25°C, 15 min. DPPH·, HO· scavenging capacities and total phenolic values of C. pepo L. female and male were higher in EA:W2 than in other extracts. In addition, all determined antioxidant capacities of female were significantly higher than male.  相似文献

Dust has been widely recognised as an important source of nutrients in the marine environment and as a vector for transporting pathogenic microorganisms. Disturbingly, in the wake of a dust storm event along the eastern Australian coast line in 2009, the Continuous Plankton Recorder collected masses of fungal spores and mycelia (~150,000 spores/m3) forming a floating raft that covered a coastal area equivalent to 25 times the surface of England. Cultured A. sydowii strains exhibited varying metabolite profiles, but all produced sydonic acid, a chemotaxonomic marker for A. sydowii. The Australian marine fungal strains share major metabolites and display comparable metabolic diversity to Australian terrestrial strains and to strains pathogenic to Caribbean coral. Secondary colonisation of the rafts by other fungi, including strains of Cladosporium, Penicillium and other Aspergillus species with distinct secondary metabolite profiles, was also encountered. Our bioassays revealed that the dust-derived marine fungal extracts and known A. sydowii metabolites such as sydowic acid, sydowinol and sydowinin A adversely affect photophysiological performance (Fv/Fm) of the coral reef dinoflagellate endosymbiont Symbiodinium. Different Symbiodinium clades exhibited varying sensitivities, mimicking sensitivity to coral bleaching phenomena. The detection of such large amounts of A. sydowii following this dust storm event has potential implications for the health of coral environments such as the Great Barrier Reef.  相似文献

Mesembryanthemum edule L. (sourfig, Aizoaceae) has long been used as food and in traditional medicine. This study was intended to characterize the antioxidant properties and the phenolic compounds of M. edule leaf, stem and root. The approach consisted to evaluate these organs for their antioxidant activities through several in vitro tests, to determine tissue contents in total phenolics, flavonoids and proanthocyanidins and to establish the phenolic composition through RP-HPLC analysis. All studied organs showed a high antioxidant activity as compared to positive control BHT, with maximal efficiency for stems followed by leaves and roots. The highest polyphenolic levels were found in stems and leaves (86.5 and 68.7 mg GAE g−1 DW, respectively), suggesting that their strong antioxidant activity could be attributed to these phytochemicals. The HPLC analysis revealed that the main phenolic compounds were quercitrin and avicularin (1.4 and 1.15 mg g−1 DW, respectively) in the leaves, while catechin and procyanidin B2 (1.66 and 1.54 mg g−1 DW, respectively) were the most abundant phenolics in the stems. Overall, the strong antioxidant activity and richness of M. edule aerial tissues suggest that it could be advantageously used as a functional or nutraceutical food, to prevent or moderate oxidative stress-related diseases.  相似文献

This study was designed to examine the in vitro antimicrobial and antioxidant activities and the amount of total phenolics of the methanol extracts of Ballota rotundifolia L. and Teucrium chamaedrys C. Koch. In the case of antimicrobial activity tests, polar sub-fractions of the methanol extracts of plant species exhibited weakest antimicrobial activity when compared with the non-polar ones. While, non-polar sub-fraction of B. rotundifolia showed moderate activity against A. lwoffii, C. perfringens and the yeasts, T. chamaedrys performed excellent activity pattern against all of the tested microorganisms. The sub-fractions were also screened for their possible antioxidant activities by two complementary tests, namely DPPH free radical-scavenging and β-carotene/linoleic acid assays. Non-polar extracts of the plant species remained inactive in both test systems. On the other hand polar extracts showed remarkable antioxidant activities. In DPPH system, free radical scavenging effect of T. chamaedrys was measured as 18.00 ± 1.42 μg.mg−1. It is extremely important to point out that, polar sub-fraction of T. chamaedrys is found as effective as the positive control BHT. Non-polar sub-fraction of T. chamaedrys found to have the highest total phenolic amount (97.12 ± 1.28 μg/mg). Results obtained from this experiment confirm the relationship between the amount of phenolics and biological activities.  相似文献

The aim of this work was to evaluate the effects of solid-state fermentation (SSF) with three strains of Agaricus (Agaricus blazei Murrill SH26, Agaricus bisporus AS2796 and Agaricus bisporus G1) on the total phenolic contents and antioxidant properties of buckwheat (Fagopyrum esculentum Möench). The results showed that the total phenolic contents of the fermented buckwheat varied with fermentation time and the starter organisms. The total phenolic content of buckwheat fermented with strain SH26 reached its maximum value of 18.07 mg/g after the fermentation of 21 days, which was 3.96 times higher than that of control. The ethanolic extracts of buckwheat fermented by strains SH26 and AS2796 exhibited enhanced antioxidant properties in comparison with the control. However, the ethanolic extracts of buckwheat fermented by strain G1 exhibited less antioxidant properties in comparison with the control. The ethanolic extracts of buckwheat fermented with strain SH26 exhibited the highest reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity and ferrous ion chelating ability, respectively. The ethanolic extracts of buckwheat fermented with strain AS2796 exhibited the highest superoxide anion radical scavenging ability.  相似文献
