Single-spore cultures of wheat- and barley-adapted isolates of Septoria nodorum were serially passaged through detached leaves of wheat or barley. Rare instances of change in host adaptation were observed after passaging but associated changes in other characters, including heterokaryon compatibility, identified the changed isolates as contaminants. Inoculation of progressively more dilute spore suspensions to detached leaves revealed that some isolates were highly infective to the host to which they were adapted, with a high probability of a single spore initiating infection. Inocula deliberately contaminated with a small proportion of spores of opposite host adaptation revealed changes when passaged on wheat that mimicked the rare changes observed on passaging pure wheat- or barley-adapted isolates. These results suggest that adaptation to wheat or barley is a stable property of pure cultures of S. nodorum and that the occasional changes in host adaptation observed on passaging result from selection of contaminants.  相似文献

Surveys of 59 and 528 randomly selected winter wheat and winter barley crops in 1982 and 1983 respectively, showed that benzimidazole-resistant strains of the eyespot fungus (Pseudocercosporella herpotrichoides) were common and widespread. The frequency of occurrence of resistant isolates, both in 1982 and 1983. was positively correlated with the number of benzimidazole fungicide applications since 1975. Resistance was much more frequent in R-type than in W-type strains of P. herpotrichoides. Fungicides or fungicide mixtures, applied to winter wheat at growth stage (GS) 31, were compared for control of eyespot at six sites in 1983 and one in 1984 where benzimidazole-resistant strains were readily detected prior to spraying. Carbendazim alone gave no control of eyespot and in some trials actually increased disease levels compared with the unsprayed control. Prochloraz, either alone or in mixture with carbendazim, was the most effective fungicide giving a moderate control of eyespot (33–42% reduction) and cost-effective yield increases. DPX-II6573, tested only in the 1984 trial, was as effective as prochloraz. The practical implications of these findings are discussed.  相似文献

Tapesia yallundae was observed in early July on wheat straws, which had been inoculated with isolates of Pseudocercosporella herpotrichoides (W-type) 9 months previously and placed in plots of winter wheat, to act as inoculum in a resistance screen. A perfect state was not observed on straws inoculated with R-type isolates in the same experiment. One hundred and thirty-four single-ascospore isolates were obtained and observations on morphology in culture, pathogenicity to wheat and rye seedlings, resistance to benomyl and the pattern of restriction fragment length polymorphisms demonstrated that all isolates were W-type, although some grew at half the expected rate on agar, and at least two isolates were involved in the production of apothecia. Apothecia were produced in vitro on straws inoculated with single isolates of P. herpotrichoides only when they were incubated in contact with other straws similarly inoculated with certain other isolates. No apothecia were produced on straws with only isolate present, suggesting the need for at least two strains, possibly representing different mating types, for apothecium formation. Apothecia of T. yallundae were produced in vitro on straw pieces which had been inoculated individually with W-type isolates, 14-16 weeks after straws of different isolates were mixed. Evidence for genetic recombination between isolates was obtained when single-ascopore colonies were analysed for resistance to benomyl in culture and for restriction-length polymorphisms (RFLPs).  相似文献

Isolates of Septoria spp. from cereals were tested for their relative pathogenicity to detached seedling leaves of three cereal species. Each of 23 isolates showed unequivocal adaptation to either oats, or wheat, or barley. Five oat-adapted isolates were from Prince Edward Island, Canada, where S. avenue is prevalent on oats; three were from the UK, where there are few oat crops and where adaptation to oats has not been reported.
Oat-adapted isolates tended to have pycnidiospores of above-average length and could be referred to S. avenae. Wheat-adapted and most barley-adapted isolates had shorter spores and could be referred to S. nodorum. UV-fluorescent pigment was produced on oxgall agar by all wheat-adapted isolates, but only by a minority of oat-adapted and barley-adapted isolates.
It is suggested that adaptation to cereal species is a more useful character than spore length for classifying isolates.  相似文献

从河南、北京等地采集小麦植株,分离得到202株内生芽孢杆菌,平板拮抗测定获得27株对小麦纹枯病菌拮抗效果明显的菌株。温室盆栽测定了27个菌株对小麦纹枯病的生防效果,其中菌株M-1、W-2和W-3的防治效果分别为60.4%、59.4%和56.6%。对3株生防菌株进行了分类学鉴定,M-1为多粘类芽孢杆菌,W-2为地衣芽孢杆菌,W-3为枯草芽孢杆菌。  相似文献

W-type isolates of Pseudocercosporella herpotrichoides grown on a maize-based agar and exposed to near- ultra-violet radiation at c . 13°C produced a greenish black colour, whilst R-type isolates produced a pink or pale brown colour in the agar medium. More colonies from directly plated lesions or from spore suspensions could be recognized as P. herpotrichoides and could be more easily differentiated as W-type or R-type or as mixtures of both by colour production on maize agar (MA) than by colony morphology on potato dextrose agar (PDA), despite the presence of other fungi. Isolates with intermediate morphology on PDA were positively identified as W-type or R-type on MA; their pathogenicities to wheat and rye seedlings were usually similar to those of W-type or R-type isolates with typical colony morphology, confirming their identification on MA. Drops of mixed suspensions of W-type and R-type spores on PDA formed fast-growing colonies with smooth margins which sometimes had slow-growing sectors with feathery margins. Drops of the same mixtures on MA formed greenish black colonies which sometimes had pink or pale brown sectors. However, when these mixtures were spread onto MA, W-type and R-type colonies could easily be differentiated by colour.  相似文献

Fifty-one single conidial isolates ofExserohilum turcicum were collected from seven locations in Israel and three in the USA and tested for mating type and aggressiveness. Sixteen isolates mated were mating type a; 29 were mating type A; and six failed to mate with either the A or the a mating type. Isolates differed significantly in their ability to cause lesions of different sizes, and in their ability to produce conidia on the susceptible corn hybrid Jubilee. They also differed in their rate of growth on lactose casein agar. No significant difference in lesion size or conidial production was noted between mating types. Conidial production and radial growth were significantly lower for isolates with no mating capacity as compared with conidial production of defined mating types. Pseudothecia were produced after 14–16 and 21 days when mating occurred onSorghum halepense straw or barley straw, respectively. The average number of pseudothecia produced on 10 cm2 of straw was 17 and 8 forS. halepense and barley, respectively. Pseudothecia were not produced on corn straw.  相似文献

ABSTRACT Septoria speckled leaf blotch (SSLB), caused by Septoria passerinii, has emerged as one of the most important foliar diseases of barley in the Upper Midwest region of the United States. To map and tag genes for SSLB resistance, we developed two populations derived from the resistant accessions CIho 4780 and CIho 10644 and the susceptible malting cv. Foster. Segregation analysis of F(2) plants or F(2:3) families from the Foster/CIho 4780 and Foster/CIho 10644 populations revealed that a single dominant gene conferred resistance at the seedling stage. Bulked segregant analysis identified an amplified fragment length polymorphism marker, E-ACT/M-CAA-170, that co-segregated with the SSLB resistance gene Rsp2 in the Foster/CIho 4780 F(2) population. Southern hybridization analysis with DNA from the wheat/barley addition lines localized E-ACT/M-CAA-170 on the short arm of the barley chromosome 5(1H). Restriction fragment length polymorphism analysis with DNA clones previously mapped to the short arm of chromosome 5(1H) placed Rsp2 at a position flanked by the markers Act8 and ksuD14. A sequence-characterized amplified region (SCAR) marker (E-ACT/M-CAA-170a) was developed that co-segregated with not only Rsp2 in the Foster/CIho 4780 population but also resistance gene Rsp3 in the Foster/CIho 10644 population. This result indicates that Rsp3 is closely linked to Rsp2 on the short arm of chromosome 5(1H). The utility of SCAR marker E-ACT/M-CAA-170a for selecting Rsp2 in two different breeding populations was validated.  相似文献

Sequences of the ribosomal DNA (rDNA) internal transcribed spacer (ITS) regions were examined to infer a molecular phylogeny of small-spored Phomopsis isolates, designated W-type (mainly white colony, weakly virulent, bearing both alpha and beta conidia at 25°C on PDA) and G-type (mainly gray colony, highly virulent, bearing only alpha conidia at 25°C on PDA), and P. amygdali from fruit trees. Phomopsis G-type and P. amygdali were a monophyletic group distinct from the W-type. The W-type isolates were divided into two monophyletic groups. Diaporthe citri, D. tanakae, P. asparagi, P. viticola, P. vitimegaspora and D. nomurai, which are morphologically distinguishable from W- and G-types, differed from the W- and G-types in molecular phylogenetic analyses. PCR-RFLP analysis of rDNA ITS regions was useful to distinguish each of the Phomopsis species and groups using three restriction enzymes. In mating tests, W-type isolates from fruit trees were heterothallic and inter-fertile even between isolates belonging to the different monophyletic groups. Isolates of the G-type and P. amygdali collected in Japan were cross-fertile. Some isolates from Lunaria annua, Ulmus glabra and Juglans regia belonged to one of the two monophyletic groups of the W-type and were cross-fertile with W-type isolates from Rosaceous fruit trees. Differences in hrpZ sequences determined by polymerase chain reaction (PCR)–restriction fragment length polymorphism (RFLP) were used to investigate the molecular epidemiology of Pseudomonas syringae pv. syringae (PSS) strains that were isolated from diseased barley and wheat plants in Okayama, Japan. PCR–RFLP using HhaI separated PSS strains into two groups (A and B). Although specific PCR–RFLP groups of PSS strains were not always isolated from specific cultivars or seeds produced in a specific area, many strains isolated from barley and wheat belonged to PCR–RFLP groups A and B, respectively.  相似文献

Twenty-one strains of Botrytis cinerea isolated from six plant species on ten sites throughout Israel, as well as a strain from France, were tested for vegetative and mycelial incompatibility, pathogenicity, resistance to the fungicides carbendazim and iprodione, and colony morphology. Selenate-resistant mutants were isolated from the strains as spontaneous, fast-growing sectors arising from restricted colonies on medium amended with sodium selenate with a mean frequency of 0.04 sectors/colony; 81% of the sectors were sulphate non-utilizing (sul) mutants. One hundred and four sul mutants were divided into two complementary groups: resistant (66 mutants) and sensitive to chromate. Based on compatibility reactions between chromate-resistant and chromate-sensitive sul mutants, 12 strains were compatible only with themselves and were each classified as belonging to different vegetative compatibility groups (VCGs). Nine strains were each compatible with one to three other strains, and were assembled into three multi-member VCGs. Mycelial incompatibility between wild-type strains (barrage), in the form of a zone of dark pigmentation or sparse mycelium with or without dark pigmentation of the agar along the line of confrontation, was observed for 70% of the inter-strain pairings. There was no correspondence in compatibility between strains revealed by two approaches: strains in different VCGs did not necessarily produce a barrage. However, self-compatibility was observed both as heterokaryon formation between complementary sul mutants and as an absence of barrages between mycelia of wild-type strains; wild-type strains belonging to the same VCG did not exhibit strong barrages, although weak antagonistic reactions were observed. Strains in two multi-member VCGs showed the same patterns of resistance to carbendazim and iprodione; the third multi-member VCG contained isolates with different patterns of resistance. Four morphological types were revealed among wild-type strains: conidial (five strains), sclerotial (six strains), intermediate (ten strains), and mycelial (one strain). On bean leaves, conidial strains were more aggressive than sclerotial strains.  相似文献

When barley, wheat or oat leaf epidermal cells were attacked by their appropriate forma specialis (f.sp.) of Blumeria graminis DC. Speer (f.sp. hordei, tritici and avenae, respectively), many attempted penetrations succeeded, functional haustoria were formed and very few plant cells died. When attacked by either of the two possible inappropriate ff.spp., penetration attempts failed in association with papilla deposition by epidermal cells, attacked cells died, or if visible haustoria were formed the plant cell died very soon afterwards. Double inoculation experiments were performed where each cereal species was first attacked by its appropriate f.sp., as inducer, and later by the different ff.spp. as challenger. Infection by the appropriate inducer profoundly affected cellular responses to challenger attack. Suppression of defensive responses was dramatic within epidermal cells containing the inducer haustorium, evident to some extent in adjacent cells, but undetectable at two cells distance. Suppression of penetration resistance allowed most challenger attacks, even by inappropriate ff.spp., to form a haustorium. Furthermore, death of penetrated epidermal cells was also suppressed so that haustoria of the inappropriate ff.spp. functioned to support colony development. In oat, delayed epidermal cell death prevented full colony development by inappropriate ff.spp., but in barley and wheat, no cell death was apparent by four days after inoculation and colonies of the inappropriate ff.spp. produced extensive hyphae, secondary haustoria and conidial chains.  相似文献

A screening test was developed to study the effect of different quantities of inoculum of Rhizoctonia solani on root rot and growth of cultivars of wheat, barley, oats, rye and triticale in a controlled environment and the field in two seasons. The ranking of relative susceptibility to disease of cultivars within a cereal differed between the controlled environment and field experiments, and between seasons in the field. The ranking of relative susceptibility between cereals also differed between experiments. In the controlled-environment experiment, wheat had the highest disease rating, followed by oats, barley, triticale and rye, but in the field barley had the highest rating followed by wheat and then oats. The overall differences between the cereals were small.  相似文献

Surveys of 528 randomly selected commercial fields in England and Wales in 1983 showed that isolates of Pseudocercosporella herpotrichoides resistant to benomyl were common and widespread on winter wheat and winter barley. During June and July resistant strains were recovered from 16 to 66% of fields and the proportion of resistant isolates was 37–52%. The frequency of occurrence of resistant isolates was positively correlated with the number of applications of carbendazimgenerating (MBC) fungicides during the previous 8 years. Benomyl resistance was found in 21–48% of fields where it was claimed that MBC had never been used, and 7–28% of isolates from those fields, were resistant. Resistance was more frequent in R-type than in W-type isolates of the pathogen. The proportion of R-type isolates decreased as the number of previous winter wheat crops rose and increased with the number of winter barley crops grown and with the number of MBC sprays applied to crops in previous years. The implications of these findings for the control of eyespot in England and Wales are discussed.  相似文献

Biochemical differences between the wheat, rye and couch-grass pathotypes of the eyespot pathogen, Pseudocercosporella herpotrichoides, have been identified by comparing isoenzyme profiles and DNA markers from several isolates. The wheat (W) and rye (R) pathotypes were clearly differentiated by both techniques, with isoenzyme profiling also separating the couch-grass isolates from the W- and R-type isolates. The isoenzyme profiles separated the P. herpotrichoides pathotypes from the two related species, P. anguioides and P. aestiva. The isoenzyme profiles of P. anguioides were closer to those of the R-type isolates of P. herpotrichoides than the W-type isolates, whereas the banding pattern of P. aestiva was very different. The isoenzyme profiles of the two German isolates, originally obtained from Nirenberg's laboratory, P. herpotrichoides var. herpotrichoides and var. acuformis, were similar to those of the wheat and rye pathotypes, respectively, isolated from UK-infected material.  相似文献

Visual estimates of wheat disease severity were compared with actual severities determined using image analysis of tracings of diseased leaves. Septoria tritici , leaf senescence and Erysiphe graminis were studied. Observer estimates were widely scattered about the actual severities (i.e. they were imprecise), differed substantially from the actual severities even after averaging (i.e. they were inacctirate), and varied considerably over short time-scales. Relative bias decreased with increasing disease severity. In a comparison of three seed treatments to control powdery mildew on winter barley, visual assessment errors altered the conclusions of the experiment. Two treatments were statistically indistinguishable on the basis of visual severity estimates, while estimates obtained by image analysis showed that one seed treatment was twice as effective as the other.  相似文献

The relative resistance of 15 winter barley, three winter wheat and three winter oat cultivars on the UK recommended list 2003 and two spring wheat cultivars on the Irish 2003 recommended list were evaluated using Microdochium nivale in detached leaf assays to further understand components of partial disease resistance (PDR) and Fusarium head blight (FHB) resistance across cereal species. Barley cultivars showed incubation periods comparable to, and latent periods longer than the most FHB resistant Irish and UK wheat cultivars evaluated. In addition, lesions on barley differed from those on wheat as they were not visibly chlorotic when placed over a light box until sporulation occurred, in contrast to wheat cultivars where chlorosis of the infected area occurred when lesions first developed. The pattern of delayed chlorosis of the infected leaf tissue and longer latent periods indicate that resistances are expressed in barley after the incubation period is observed, and that these temporarily arrest the development of mycelium and sporulation. Incubation periods were longer for oats compared to barley or wheat cultivars. However, oat cultivars differed from both wheat and barley in that mycelial growth was observed before obvious tissue damage was detected under macroscopic examination, indicating tolerance of infection rather than inhibition of pathogen development, and morphology of sporodochia differed, appearing less well developed and being much less abundant. Longer latent periods have previously been related to greater FHB resistance in wheat. The present results suggest the longer latent periods of barley and oat cultivars, than wheat, are likely to play a role in overall FHB resistance if under the same genetic control as PDR components expressed in the head. However the limited range of incubation and latent periods observed within barley and oat cultivars evaluated was in contrast with wheat where incubation and latent periods were shorter and more variable among genotypes. The significance of the various combinations of PDR components detected in the detached leaf assay as components of FHB resistance in each crop requires further investigation, particularly with regard to the apparent tolerance of infection in oats and necrosis in barley, after the incubation period is observed, associated with retardation of mycelial growth and sporulation.  相似文献

ABSTRACT Septoria speckled leaf blotch (SSLB) caused by Septoria passerinii is a common disease in barley. SSLB resistance genes Rsp1, Rsp2, and Rsp3 have previously been identified in the United States Department of Agriculture National Small Grains collection accessions CIho 14300, CIho 4780, and CIho 10644, respectively. Populations of 100 to 120 F(2) individuals were evaluated for SSLB resistance in the greenhouse. Inheritance was evaluated in F(2:3)-derived families in the field. Partial molecular maps for three Rsp genes were constructed on F(2) and F(2:3) families derived from crosses between Robust and the resistant accessions CIho 14300, CIho 4780, and CIho 10644. The resistant locus Rsp1 was mapped to the short arm of chromosome 3H with two flanking diversity arrays technology (DArT) markers, bPb-6978 (8.9 cM) and bPb-9945 (16.3 cM), and two random amplified polymorphic DNA (RAPD) markers, OPC2(441R) (3.0 cM) and UBC285(158R) (4.3 cM). The genes Rsp2 and Rsp3 were positioned on the short arm of barley chromosome 1H with two restriction fragment length polymorphism (RFLP), six DArT, and three RAPD markers. An RFLP marker, MWG938, and an RAPD marker, OPAH5(545C), were tightly associated with Rsp2 at a distance of 0 cM. Five DArT markers spanning the short arm of 1H surrounded Rsp3 at a distance of 2.3 and 5.8 cM, while two RAPD markers-OPBA12(314C) (2.4 cM) in coupling and OPB17(451R) (3.5 cM) in repulsion-flanked Rsp3. Molecular marker data associated with Rsp2 and Rsp3 indicated that the two genes are closely linked on chromosome 1HS. A total of 17 of 154 simple sequence repeats (SSRs) tested were associated with Rsp genes on chromosome 1H and 3H, and they were also integrated into genetic linkage maps of the three F(2) Robust populations. Knowledge about the map position of Rsp genes on barley chromosomes will be useful for breeding for SSLB resistance in barley and eventual gene cloning.  相似文献
