Juvenile Litopenaeus vannamei farmed at 3.0 psu were fed five diets containing glucose, sucrose, wheat starch, corn starch or potato starch as the carbohydrate (CBH) source. Shrimp were fed for 50 days to explore the effect of dietary CBH source on growth, body composition and ammonia tolerance. The specific growth rate of body length of shrimp fed glucose was the highest and significantly higher than those fed potato starch. The survival rate of shrimp fed glucose was 89.44%, and it was the highest and significantly higher than those fed wheat starch. Whole shrimp body crude protein and lipid of the corn starch group were 140.2 g kg?1 and 10.1 g kg?1 respectively. And they were significantly higher than those fed wheat starch. Shrimp fed potato starch had higher hepatopancreas and muscle glycogen. Shrimp fed sucrose had higher glucose‐6‐phosphate dehydrogenase and lower pyruvate kinase activities (P < 0.05). Besides, shrimp fed starch produced more B cells in hepatopancreas tubules than those fed glucose or sucrose. Shrimp fed different sources of CBH differed in the number of R cells. After 96‐h of ammonia nitrogen challenge, the survival rate of the treatments from high to low in turn was glucose, wheat starch, corn starch, sucrose and potato starch, and no significant differences were observed among all treatments. Based on shrimp growth and the economic problems of practical production, we recommend wheat starch as CBH source in practical diets for L. vannamei farmed at low salinities.  相似文献

Raptor, a member of the target of rapamycin complex 1 (TORC1), participates in the formation of complex proteins related to the mechanistic target of rapamycin (mTOR) signalling. In this study, a 5,020 bp cDNA of Raptor with an open reading frame (ORF) of 3,804 bp encoding for 1,267 amino acids was cloned from Litopenaeus vannamei. The protein contains three conserved domains: Raptor N, HEAT and WD40 domains. The expression of Raptor gene was detected by qRT‐PCR in different tissues of L. vannamei, including hepatopancreas, intestinal, stomach, eyestalk, gill and muscle. The mRNA expression profiles of Raptor in muscle were also analysed under suppression or stimulation of mTOR signalling pathway. The level of Raptor mRNA significantly increased either at 0.5–6 hr after an injection of rapamycin (RAPA) or after 3 days starvation. Leucine or arginine alleviated the up‐regulation of Raptor gene expression caused by RAPA or starvation. The Raptor gene was successfully suppressed using RNA interference (RNAi) technology, and the gene expression and the protein phosphorylation level of 4EBP1 and S6K were significantly decreased. The results of the study suggested that the expression of Raptor was sensitive to the immunology status of L. vannamei and participated in nutritional metabolism.  相似文献

Because of the increasing market potential of Pacific white shrimp (Litopenaeus vannamei) and limited availability of coastal areas for production, culture of L. vannamei at low salinity is a growing trend throughout the world. Dietary manipulation could improve the growth performance of L. vannamei at low salinity. This study reared L. vannamei with glucose, sucrose and corn starch as dietary carbohydrate sources, respectively, at a low salinity. The results indicated that the sucrose and glucose feeding groups showed better growth performance. An Illumina‐based sequencing method was used to examine the intestinal bacterial composition and the results indicated that Proteobacteria were the most prevalent members, but abundance of Actinobacteria decreased while Firmicutes increased in the corn starch‐fed group. Furthermore, bacteria related to complex carbohydrate degradation were in lower abundance, whereas the abundance of opportunistic pathogenic bacteria increased in corn starch‐fed group than the other two groups, suggesting that the diet imposes selective pressure on the intestinal microbiota. Complex carbohydrates were not the ideal energy sources for L. vannamei at low salinity because the host has higher energy demand in the stressful conditions while the complex carbohydrate degradation efficiency of the gut microbiota in L. vannamei is limited.  相似文献

Three isonitrogenous diets containing 60 g kg–1, 90 g kg–1 or 120 g kg–1 lipid were formulated and fed to the Litopenaeus vannamei (2.00 ± 0.08 g) under two salinities (25 or 3 psu) in triplicate for 8 weeks. Shrimp fed 90 g kg–1 lipid had higher weight gain and specific growth rate than shrimp fed the other two diets regardless of salinity, and the hepatosomatic index increased with increasing dietary lipid at both salinities. The shrimp at 3 psu had significantly lower survival and ash content, higher condition factor, weight gain and specific growth rate than the shrimp at 25 psu. Increasing dietary lipid level induced the accumulation of serum MDA regardless of salinity, and at 3 psu, it reduced the serum GOT and GPT activities and the mRNA expression of TNF‐α in intestine and gill of L. vannamei. The hepatopancreatic triacylglycerol lipase (TGL) and CPT‐1 mRNA expression showed the highest value in shrimp fed 90 g kg–1 lipid diet at 3 psu. This study indicates that 120 g kg–1 dietary lipid may negatively affect the growth and induce oxidative damage in shrimp, but can improve immune defence at low salinity; 60 g kg–1 dietary lipid cannot afford the growth and either has no positive impact on the immunology for L. vannamei at 3 psu.  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of ascorbic acid (AsA), in the form of l ‐ascorbyl‐2‐polyphosphate (LAPP) on growth performance, body composition, antioxidative capacity and salinity stress tolerance of juvenile Pacific white shrimp, Litopenaeus vannamei. Five practical diets (46% crude protein and 7.6% lipid) supplemented with graded levels of AsA (14.64, 48.55, 84.98, 308.36 and 639.27 mg kg?1 diet) were fed to five replicate groups of L. vannamei (mean initial wet weight 0.57 g). No significant differences were found on growth performance among all treatments. However, whole body lipid content significantly decreased with dietary AsA levels increasing. Activities of total antioxidant capacity, glutathione reductase and glutathione peroxidase were significantly affected by dietary AsA levels. Shrimp fed LAPP‐free diet had higher malondialdehyde content than those fed the diets supplemented with LAPP. Dietary AsA levels higher than 308.36 mg kg?1 diet increased the survival of shrimps after 1, 2 and 3 h of acute salinity change. Broken‐line regression analysis on survival after 3 h of salinity stress and second‐degree polynomial regression analysis on glutathione reductase data indicated that the optimal dietary AsA requirement of L. vannamei was estimated to be 306.39, 319.75 mg kg?1 diet respectively.  相似文献

Juvenile Litopenaeus vannamei were fed for 8 weeks with diets containing four ratios of protein to carbohydrate (CBH) at P26 : C30, P30 : C25, P34 : C19 and P38 : C14, respectively, at 3.0 g L?1 salinity. Shrimp weight gain of P34 : C19 group was the highest and differed from the shrimp fed the P26 : C30 or P30 : C25 diet. Shrimp fed the P26 : C30 diet obtained higher survival than those fed other diets. Shrimp fed the P34 : C19 diet contained the highest body protein and lipid, which were significantly higher than those fed the P38 : C14 diet. Shrimp fed the P30 : C25 diet had the highest haemolymph glucose content, which was significantly higher than those fed the P26 : C30 or P38 : C14 diet. Shrimp muscle glycogen of the P26 : C30 group was the highest. Hepatopancreas B‐cell number of shrimp fed the P26 : C30 diet was lower than those fed other diets, and the R cell number was the highest in the shrimp fed the P30 : C25 diet. This study indicates that the protein‐sparing effect by CBH occurred in the P30 : C25 and P34 : C19 groups because these proteins to CBH ratios can support normal growth. Within the range of basic energy demand, the high dietary CBH to protein ratio can improve L. vannamei survival at low salinity.  相似文献

A 4‐week feeding trial was conducted to determine the effects of oxidized fish oil (OFO, POV: 234.84 meq kg?1) on growth performance and oxidative stress of Litopenaeus vannamei. Five diets containing various OFO levels (0, 25, 50, 75 and 100 g kg?1) with the same dietary lipid level were fed to L. vannamei. The results showed that the body weight gain and the specific growth rate of the shrimp fed with 50, 75 and 100 g kg?1 of OFO diets decreased significantly (< 0.05), whereas the hepatosomatic index increased significantly (P < 0.05). The malondialdehyde concentrations in the serum and muscle of the shrimp fed with 50, 75 and 100 g kg?1 of OFO diets were significantly higher than that of the shrimp fed with fresh fish oil (P < 0.05). The total antioxidant competence decreased significantly compared with the control group. Therefore, dietary OFO affects the growth performance and increases the oxidative stress of shrimp.  相似文献

A 10‐week growth trial was run to evaluate effects of myo‐inositol (MI) on growth performance, haematological parameters, antioxidative capacity and salinity stress tolerance of Litopenaeus vannamei. Six practical diets supplemented with graded levels of MI (designated as MI0, MI600, MI1200, MI2400, MI 3600 and MI4800 for 448.8, 974.2, 1568.0, 2810.6, 3835.5 and 4893.6 mg/kg diet, respectively) were fed to six replicate groups of L. vannamei (mean initial body weight 0.63 ± 0.00 g). The results showed that significant increment of growth performance was observed in shrimp fed MI600 diet than those fed MI1200 diet. Lipid concentration in whole body of the shrimp fed MI600 diet was significantly increased. Shrimp fed MI0 diet had lower total protein (TP) as compared to shrimp fed the MI‐supplemented diets (except MI4800 diet). In general, lower activities of antioxidant enzymes and higher malondialdehyde (MDA) content in haemolymph and hepatopancreas were recorded in shrimp fed MI0 diet, compared to those fed the MI‐supplemented diets. Reduced survival after 7‐h salinity stress was present in shrimp fed MI0 diet as compared to those fed MI4800 diet. Dietary MI requirement for glutathione peroxidase activity of L. vannamei was 2705 mg/kg diet.  相似文献

In a Biofloc Technology System (BFT), there is constant biofloc formation and suspended solids accumulation, leading to effects on water quality parameters that may affect the growth performance of cultured shrimp. This study aimed to analyse during biofloc formation the effect of different total suspended solids (TSS) levels on water quality and the growth performance of Litopenaeus vannamei shrimp in a BFT system. A 42‐day trial was conducted with treatments of three ranges of TSS: 100–300 mg L?1 as low (TL), 300–600 as medium (TM) and 600–1000 as high (TH). The initial concentrations of 100 (TL), 300 (TM) and 600 mg L?1 (TH) were achieved by fertilization before starting the experiment. Litopenaeus  vannamei juveniles with an average weight of 4.54 ± 1.19 g were stocked at a density of 372 shrimp m?3. Physical and chemical water parameters and shrimp growth performance were analysed. After 6 weeks, TSS mean concentrations were 306.37, 532.43 and 745.2 mg L?1 for, respectively, TL, TM and TH treatments. Significant differences (P < 0.05) were observed in TSS, settleable solids, pH, alkalinity and nitrite, especially between the TL and TH treatments. Similarly, differences (P < 0.05) were observed in the growth performance parameters, specifically final weight, survival, feed conversion and productivity. The water quality parameters at lower range of total suspended solids concentration (TL) treatment resulted in a better performance of L. vannamei in the BFT system. The maintenance at range of 100–300 mg L?1 TSS is thus important to the success of shrimp culture.  相似文献

Five different Vibrio parahaemolyticus strains (SH8, SH108, SH58, AH5 and GD10) isolated from the hepatopancreas of moribund shrimp in farms of mainland China were identified and capable of inducing massive mortality of Penaeus (Litopenaeus) vannamei. The immersion challenge results with five isolates indicated variance of virulence, while only GD10 caused massive sloughing of tubule epithelial cells which was recognized as the most significant symptom of AHPND. Differences in immune responses were detected of P. vannamei during 48 h post‐infection (p.i.) by injection or immersion challenge with V. parahaemolyticus (SH8, SH108 and GD10) isolates. When injected SH8 and SH108 isolates, the expression of lysozyme (LSZ) showing statistically significant upregulation at 16 and 48 h p.i. and that of Toll‐like receptors (TLR) showed statistically significant upregulation at 48 h p.i. When immersion challenge with the GD10 isolate, TLR were upregulated after 8 h p.i. challenge with 104 cfu mL?1; however, LSZ was downregulated when challenged with 103 cfu mL?1. The results suggested that LSZ and TLR serve as crucial molecular markers of innate immunity in shrimp against V. parahaemolyticus infection. LSZ is a vital marker for acute bacterial infection, while TLR serves as a crucial marker for chronic infection.  相似文献

The present study was to determine the time‐dependent alterations in growth performance, histomorphology, digestive enzymes activities and the genes expression related to target of rapamycin (TOR) signalling pathway involved in hepatopancreas and intestine of Litopenaeus vannamei at 3, 6, 12, 18, 24 and 30 days after Aflatoxin B1 (AFB1) challenge. The results manifested that AFB1 could induce a significant decrease in growth performance and visible variations of the hepatopancreas and intestine structures in shrimp. Meanwhile, the digestive enzymes activities and genes expression level were significantly lower in the experimental group than in the control group and showed a time‐dependent decrease (p < 0.05). The expression levels of tor and s6k were significantly higher in the experimental group than in the control group from days 6 to 24 and showed a tendency to increase first and then decrease as a whole (p < 0.05). Meanwhile, we analysed candidate pathways involved in hepatopancreas and intestine of shrimp in metabolic response to AFB1. The mainly disturbed pathways related to metabolism in hepatopancreas were involved in pyrimidine, glycosylphosphatidylinositol, glycosaminoglycan, vitamin and amino acid biosynthesis and metabolism, while the mainly disturbed metabolic pathways in intestine were glycosaminoglycan, vitamin and amino acid biosynthesis and metabolism.  相似文献

A feeding trial was performed for 28 days to evaluate the effects of replacement of fish meal (FM) with fermented cottonseed meal (FCM) on growth, body composition and haemolymph indexes of juvenile Litopenaeus vannamei. Four isonitrogenous and isoenergetic diets were formulated by using FCM (96.4, 206, 317 and 417 g kg?1) to substitute 25%, 50%, 75% and 100% of FM in a control diet respectively. Each diet was randomly allotted to four tanks with 20 shrimp per tank. The feeding trail was conducted in an indoor flow‐through aquaculture system. Shrimp fed diets containing 317 and 417 g kg?1 of FCM obtained lower (P < 0.05) final weight, weight gain, specific growth ratio, protein efficiency ratio as well as a higher (P < 0.05) feed conversion ratio compared with shrimp fed the control diet. The body ash content decreased (P < 0.05) in shrimp fed the diet with complete replacement of FM than those in other treatments. Moreover, increasing the dietary inclusion of FCM linearly raised (P < 0.05) the concentrations of total gossypol, (?) and (+) gossypol enantiomers in the whole shrimp body. No difference (P > 0.05) was observed in haematological parameters among the treatments. The results suggest that up to 50% of FM can be replaced by FCM without adverse effects on growth and feed utilization of L. vannamei.  相似文献

To investigate the effect of dietary sodium chloride (NaCl) on meat quality of white shrimp (Litopenaeus vannamei) reared in low‐salinity (2 g L?1) water, shrimp were distributed into four groups (treatments T‐1, T‐2, T‐3 and control) with three replicates. All shrimps were completely randomised stocked into 12 tanks at an initial density of 40 shrimps per tank. Diets for the control, T‐1, T‐2 and T‐3 groups consisted of the basal diet supplemented with 0 g kg?1, 10 g kg?1, 20 g kg?1 and 40 g kg?1 of NaCl respectively. After 50 days, shrimps in T‐3 showed significantly better (< 0.05) moisture, crude protein and ash than those of the control and T‐1. Higher muscle Na content was observed (< 0.05) in T‐3 than that of the control. Significant increases (< 0.05) in contents of inosinic acid, total free amino acid (TFAA) and essential free amino acid (EFAA) were also found in T‐3. Texture assays showed significant differences (< 0.05) in hardness, adhesiveness and springiness between group T‐3 as compared with those of T‐1 and control. It indicated that dietary supplementation of NaCl appeared to be a promising practice to improve meat quality of white shrimp reared in low‐salinity waters.  相似文献

Feeding aquatic animals with bacterial encapsulated heat‐shock proteins (Hsps) is potentially a new method to combat vibriosis, an important disease affecting aquatic animals used in aquaculture. Food pellets comprised of shrimp and containing Escherichia coli overexpressing either DnaK‐DnaJ‐GrpE, the prokaryotic equivalents of Hsp70‐Hsp40‐Hsp20, or only DnaK were fed to juveniles of the white leg shrimp Penaeus vannamei, and protection against pathogenic Vibrio harveyi was determined. Maintaining pellets at different temperatures for varying lengths of time reduced the number of live adhering E. coli, as did contact with sea water, demonstrating that storage and immersion adversely affected bacterial survival and attachment to pellets. Feeding P. vannamei with E. coli did not compromise their survival, indicating that the bacteria were not pathogenic to shrimp. Feeding P. vannamei with pellets containing bacteria overproducing DnaK (approximately 60 cells g?1 pellets) boosted P. vannamei survival twofold against V. harveyi, suggesting that DnaK plays a role in Vibrio tolerance. Pellets containing DnaK were effective in providing protection to P. vannamei for up to 2 weeks before loss of viability and that DnaK encapsulated by these bacteria enhanced shrimp resistance against Vibrio infection.  相似文献

Pacific white shrimp Litopenaeus vannamei (1050 individuals with initial weight of 1.01 ± 0.001 g) were fed either control diet or one of six dietary astaxanthin (AX) concentration (25, 50, 75, 100, 125 and 150 mg kg−1) diets for 56 days in 35 tanks (30 shrimp per tank). After 56 days of culture, shrimp‐fed AX125 and AX150 diets had higher (< 0.05) weight gain, specific growth rate, total antioxidant status and lower (< 0.05) superoxide dismutase (SOD), catalase (CAT) than shrimp fed control diet. After low dissolved oxygen stress for 1 h, survival rate of shrimp fed AX75, AX100, AX125 and AX150 diets was higher (< 0.05) than that of shrimp fed control diet. Hypoxia inducible factor‐1α (HIF‐1α), cytosolic manganese superoxide dismutase (cMnSOD) and CAT mRNA expression levels of shrimp fed seven diets were significantly down‐regulated under hypoxia than under normoxia, but their expression levels were higher under hypoxia in shrimp fed AX‐supplemented diets than in shrimp fed control diet. About 70‐kDa heat‐shock protein (Hsp70) mRNA expression level of shrimp fed seven diets was significantly up‐regulated under hypoxia than under normoxia, but its expression level was lower under hypoxia in shrimp fed AX‐supplemented diets than in shrimp fed control diet.  相似文献

This experiment was conducted to study the effects of different forms and levels of manganese (Mn) on the growth performance, antioxidant activities, tissue Mn content and cytosolic manganese superoxide dismutase (cMnSOD) gene expression of Litopenaeus vannamei. Treatments consisted of 0, 10, 20, 30, 40 and 60 mg Mn kg?1 from manganese sulphate (Mn‐S) and manganese methionine (Mn‐Met), providing the actual dietary value of 5.17, 15.62, 25.55, 34.22, 44.48 and 67.90 mg Mn kg?1 Mn‐S, and 5.17, 15.71, 25.36, 35.86, 45.16 and 65.06 mg Mn kg?1 Mn‐Met, respectively. Each diet was fed to triplicate groups of L. vannamei (initial body weight: 1.925 ± 0.002 g) in a recirculated fresh water rearing system for 8 weeks. Weight gain rate (WGR) increased in prawns provided with from 25.55 to 44.48 mg Mn kg?1 Mn‐S and 15.71 to 45.16 mg Mn kg?1 Mn‐Met and then declined above these levels. The lowest protein efficiency ratio (PER) and the highest feed conversion rate (FCR) were observed in prawns fed the control diet (< 0.05) and showed no significant differences among other treatments (> 0.05). Survival rate (SR) was not affected by the dietary treatments (> 0.05). Total SOD and Mn‐SOD activities were higher in the hepatopancreas of prawns fed with Mn‐supplemented diets from 15.71 to 44.48 mg Mn kg?1 (< 0.05). On the contrary, malondialdehyde (MDA) content was lower in the hepatopancreas of prawns fed the basal diet (< 0.05). Mn concentrations in the hepatopancreas and muscles increased with increasing levels of dietary Mn supplementation. Moreover, Mn accumulation was lower in the muscle than in the hepatopancreas of the prawns. The mRNA expression of cMnSOD gene in the hepatopancreas of prawns was upregulated with increasing dietary Mn levels of Mn‐S from 25.55 to 44.48 mg Mn kg?1, Mn‐Met from 15.71 to 45.16 mg Mn kg?1 and then plateaued above these levels. Broken‐line regression analysis of WGR indicated that the optimal dietary Mn requirements for juvenile L. vannamei were 32.26 mg Mn kg?1 Mn‐S and 23.90 mg Mn kg?1 Mn‐Met, respectively.  相似文献
