为评价增精宝对雄性动物生精作用的影响,采用环磷酰胺致小鼠生精障碍为动物模型,考察不同剂量增精宝对环磷酰胺致小鼠生精障碍的保护作用。结果显示,高剂量组对小鼠的附睾、睾丸、储精囊和前列腺均有较显著的促进作用;低剂量组精子总密度和精子活率均差异显著(P<0.05);高剂量组精子活率差异显著(P<0.05);对小鼠睾丸各倍体生精细胞比例的影响,高剂量组小鼠睾丸生精小管较为饱满,管腔内存在大量精子,生精上皮较厚,可见各级生精细胞排列整齐,界膜明显,睾丸间质细胞排列整齐。结论表明,增精宝两种剂量均能够显著促进环磷酰胺导致小鼠生精障碍的保护作用,高剂量组对雄性小鼠的作用尤为显著。  相似文献

利用形态学、组织学方法,对初生至300日龄连城白鸭睾丸生长发育进行了初步研究,以探讨其胚后发育和性成熟特点。结果显示:连城白鸭睾丸大小、重量、睾丸指数及睾丸曲精小管组织结构均呈明显的日龄性变化。随着日龄增长,连城白鸭双侧睾丸体积、双睾重、睾丸指数逐渐增大,曲精小管管径和上皮高度逐渐增大,细胞层数增多,间质组织先增多,而后又减少,白膜厚度也先增大后减小。随着性成熟的开始,精原细胞不断分裂分化,出现了不同发育阶段的生精细胞。连城白鸭睾丸的胚后发育过程分为4期:30日龄之前为精原细胞增殖和曲精小管形成期,30~60日龄为初级精母细胞增殖期,60~75日龄为次级精母细胞增殖期,90~120日龄为精子形成期。  相似文献

[目的]研究亚慢性砷中毒对大鼠睾丸组织AQP-8及survivin蛋白表达的影响。[方法]将40只健康成年清洁级SD雄性大鼠随机分为对照组以及低、中、高剂量砷染毒组,每组10只;采用自由饮水方式染毒,低、中、高剂量染毒组饮水中亚砷酸钠浓度分别为2.4、12.0、60.0 mg/L,连续染毒15周,对照组饮用蒸馏水;染毒试验结束后,对各组大鼠进行精子计数,计算精子存活率;制备睾丸组织切片进行病理学观察;利用免疫组化及Western blotting法测定大鼠睾丸组织中AQP-8及survivin蛋白的表达水平。[结果]与对照组相比,中、高剂量染毒组大鼠精子计数与精子存活率均降低,且差异显著(P<0.05)。低剂量染毒组大鼠生精小管无明显变化;中剂量染毒组大鼠生精上皮层数减少,小管间隙增宽;高剂量染毒组大鼠部分生精小管出现基膜溶解,生精上皮细胞层次紊乱,细胞间隙增大,间质出现水肿、渗出。免疫组化分析结果显示,AQP-8蛋白主要表达于各级生精细胞的细胞膜和精子细胞,与对照组相比,各剂量染毒组大鼠睾丸组织中AQP-8阳性细胞平均光密度值显著(P<0.05)升高;survivin蛋白...  相似文献

为了研究三丁基锡对雄性生殖系统的影响,试验以昆明小鼠为试验动物,经0,0.2,2,20μg/m L(分别为对照、低剂量、中剂量和高剂量组)的三丁基锡暴露45 d后,观察其对雄性小鼠睾丸、附睾、输精管等组织结构的影响。结果表明:三丁基锡暴露后,睾丸的组织结构发生明显改变,而且损伤程度随三丁基锡剂量的升高而加重。高剂量组生精细胞排列杂乱无序,各级生精细胞间出现间隙,睾丸间质没有显著变化。生精小管中精母细胞和精原细胞数目在各剂量组间无显著差异;而精子数目发生显著变化,低剂量组和对照组存在显著差异,中剂量组和对照组、低剂量组之间无显著差异,高剂量组和对照组、中剂量组存在显著差异。附睾管上皮细胞排列疏松,管间结缔组织减少、间隙变大,损伤程度随剂量升高而加剧,高剂量组最为严重,输精管形状发生变化,组织形态发生改变、腔壁脱落;中剂量组表现最明显,输精管的管状结构几乎完全消失。说明45 d的三丁基锡暴露对昆明小鼠雄性生殖系统的组织结构有影响且具剂量效应。  相似文献

为探讨来曲唑对香猪睾丸细胞波形蛋白表达及分布的影响,试验将14日龄、体重3 kg的12头雄性香猪随机分为3组,对照组不做任何处理,试验组分别经口投喂0.1、0.2 mg/kg来曲唑,每3 d投喂1次,连续投喂15次,最后一次投喂7 d后取左侧睾丸,应用免疫组织化学方法检测波形蛋白在香猪睾丸细胞中的表达与分布。结果表明:波形蛋白在睾丸生精小管细胞胞浆中表达,试验组睾丸生精小管细胞总数较对照组减少(P0.01);而波形蛋白表达细胞阳性率上升(P0.01),且随着来曲唑剂量的增加而增加(P0.01)。来曲唑可通过降低雌激素水平来调节睾丸生精小管细胞数及波形蛋白表达,说明来曲唑可能促进了香猪睾丸生精小管细胞中波形蛋白的表达,进而影响睾丸生精小管细胞的生长发育,但其作用机制机理还需进一步研究。  相似文献

为了研究杨梅黄酮对环磷酰胺所致雄性小鼠生精障碍的保护作用及作用机制,采用腹腔注射环磷酰胺(50 mg/kg·bw)连续7 d建立雄性小鼠生精障碍模型,杨梅黄酮组分别灌胃杨梅黄酮(100 mg/kg·bw、200 mg/kg·bw、400 mg/kg·bw),连续30 d。观察雄性小鼠的精子密度、精子畸变率,血清中还原型谷胱甘肽(GSH)的含量,睾丸组织中乳酸脱氢酶(LDH)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)及核因子κB(NF-κB)等指标,并观察睾丸组织的病理改变。结果显示:与模型组比较,杨梅黄酮中、高剂量组的精子密度增加、精子畸变率减少,杨梅黄酮中、高剂量组睾丸组织中TNF-α及NF-κB含量降低,杨梅黄酮中、高剂量组血清中GSH水平增加,杨梅黄酮低、中、高剂量组睾丸组织中MDA含量降低、LDH活性增强。病理可见模型组睾丸组织生精上皮明显变薄,生精细胞层次、数量减少,多数生精小管腔未见精子形成,而杨梅黄酮可改善环磷酰胺所致的生精细胞的损伤。表明杨梅黄酮对环磷酰胺诱导雄性小鼠的生精障碍有一定保护作用,作用机制与抗氧化、抗炎有关。  相似文献

试验旨在探究不同月龄山羊睾丸生精小管、固有层的结构及成分变化,及睾丸中α-平滑肌肌动蛋白(α-SMA)蛋白的分布情况。选用1月龄和2月龄(幼龄)、12月龄(成年)山羊睾丸,利用特殊组织染色、免疫荧光、透射电镜并结合形态学计量统计分析,研究性成熟前后山羊睾丸生精小管、固有层细胞的结构成分及变化。结果显示:随着睾丸发育,生精小管管径增大,生精细胞种类、数量增多;支持细胞发育完善,体积增大,但轮廓不明显;睾丸间质细胞逐渐发育成熟,胶原纤维广泛分布于生精小管固有层基膜和睾丸间质中。免疫荧光染色观察可见:在1月龄、2月龄山羊睾丸中,α-SMA在管周肌样细胞、小动脉血管管周平滑肌细胞中表达阳性,在支持细胞、各级生精细胞、睾丸间质细胞中无表达;在12月龄山羊睾丸中,α-SMA在管周肌样细胞、小动脉血管管周平滑肌细胞中表达阳性,在支持细胞、各级生精细胞、睾丸间质细胞中无表达。本试验为深入研究睾丸的生理功能提供了基础形态学依据。  相似文献

为研究三苯基锡(TPT)对雄性哺乳动物生殖的毒性作用,将20只雄性青春期Sparague Dawley(SD)大鼠随机分为3个不同剂量的试验组(0.5、5、50μg/kg)和1个对照组(生理盐水)。每3d灌胃1次,暴露54d,末次灌胃24h后用100g/L水合氯醛麻醉并处死大鼠,取其睾丸和附睾。采用石蜡切片和HE染色方法观察睾丸和附睾组织学;采用分光光度法测定睾丸中碱性磷酸酶(AKP)、总抗氧化能力(TAOC)和γ-谷氨酰转肽酶(γ-GT)水平。结果显示,大鼠睾丸内曲精小管直径随药物剂量的增加而减小,曲精小管生精细胞和管腔内精子数量均有不同程度减少;低、中剂量组AKP高于对照组,高剂量组低于对照组,但差异不显著(P0.05);低剂量组γ-GT活力和T-AOC显著高于对照组,中剂量组与对照组相比变化不明显,高剂量组显著低于对照组(P0.05)。试验证实了TPT在一定程度上影响了雄性大鼠性腺生长发育与精子发生,推测TPT对雄性哺乳动物具有生殖毒性。  相似文献

采用单侧睾丸动脉结扎/再通以研究其对小鼠睾丸生精功能以及血清中抗精子抗体IgG、IgM含量的影响。实验选择健康成年BALB/c雄性小鼠于显微镜下行单侧睾丸动脉结扎。再于2h、4h、6h后开通血管。ELISA法检测血清抗精子抗体IgG、IgM水平,光镜观察睾丸生精小管内变化。结果表明:与非术侧相比,术侧睾丸生精小管内生精细胞不同程度地凋亡坏死,生精上皮脱落。各手术组血清中抗精子抗体IgG、IgM含量均有明显增长(P〈0.05)。实验结论:单侧睾丸动脉结扎/再通可引起睾丸生精细胞的凋亡坏死,同时血清抗精子抗体IgG、IgM含量增加。  相似文献

本实验旨在探究鸟苷酸交换因子15(ARHGEF15)基因在小鼠睾丸中的表达模式及其对精子发生的影响,本研究通过免疫组化和免疫荧光双染色实验检测ARHGEF15蛋白在5日龄和成年(8~9周龄)小鼠睾丸中的表达模式,并利用Cre-LoxP系统特异性敲除小鼠生精细胞内的ARHGEF15基因。结果显示:在5日龄小鼠睾丸中,ARHGEF15蛋白主要表达于精原细胞的细胞质和/或细胞膜,在成年小鼠睾丸中ARHGEF15蛋白主要定位于支持细胞的细胞核;条件性敲除(cKO)组小鼠睾丸的形态大小、组织学结构、精子的运动性能及形态与对照组相比无显著差异,且cKO组小鼠的繁育力也不受影响。因此,ARHGEF15基因主要在发育早期小鼠睾丸的生精细胞中表达,敲除生精细胞内的ARHGEF15基因不影响小鼠的繁殖能力和精子发生。  相似文献

60只雄性小鼠随机分为4组,每天分别腹腔注射0.2mL的0.9%生理盐水或7.5、15、30g/L CA,于14、21d测定曲细精管精子的生成及附睾内精子特性的相关数据。结果显示,随柠檬酸处理剂量的增加精子的生成数减少。14d时,对照组与15、30g/L CA组间、7.5、30g/L CA组间曲细精管内精子的生成差异显著(P〈0.05);21d时,对照组与柠檬酸处理组间及7.5、15g/L CA与30g/L CA处理组间曲细精管内精子的生成有显著差异(P〈0.05)。柠檬酸对附睾精子品质的影响表现为:对照组与柠檬酸处理组间及柠檬酸处理组间精子密度和活动率差异均显著(P〈0.05);对照组和15、30g/L CA处理组及15g/L CA与30g/L CA处理组间精子活力差异显著(P〈0.05);精子的畸形率随柠檬酸浓度的增加而增加,15g/L CA处理组精子畸形率显著高于对照组(P〈0.05),30g/LCA处理组显著高于对照组和7.5g/L CA(P〈0.05)处理组。组织形态学观察结果表明柠檬酸能破坏睾丸正常组织形态结构。柠檬酸高剂量组的生精小管内生精细胞排列疏松、紊乱,生精细胞间出现空隙;生精小管中的生精细胞脱落或退化,精原细胞、精母细胞和精子数量明显减少。研究表明,外源柠檬酸雄性小鼠具有显著的生殖遗传毒性。  相似文献

目的探讨3-甲基-4-硝基酚(PNMC)对雄性小鼠的生殖毒性作用。方法 20只小鼠随机分成阴性对照组和PNMC组(分0.9、9和90mg/kg·bw组),经口灌胃染毒30天,观察睾丸和精子的形态变化。结果染毒组小鼠各剂量睾丸生精细胞发生坏死,曲细精管腔内出现细胞团,精子细胞分化异常,形成均质红染颗粒,睾丸曲细精管生精上皮/直径比值在90mg/kg组明显高于对照组;染毒组小鼠精子畸形率明显高于对照组(P<0.05),并且随着染毒浓度的增加,畸形率有增加的趋势。结论 3-甲基-4-硝基酚不仅可以引起生精细胞坏死和抑制精子形成,还可以诱导精子畸变,从而对生殖系统造成损伤。  相似文献

This experiment was aimed to study the effect of quinestrol on the expression of PCNA and Caspase-3 in spermatogenic cells of juvenile mice. 4-week old male mice were randomly divided into four groups. Mice in the control group were injected intraperitoneally with olive oil alone and mice in the experiment groups were injected intraperitoneally with octylphenol resolved in olive oil at 1, 10, 100 mg/kg doses respectively for 1 week (once a day). The testes were immediately removed for weighing and serial cross-sections that were used for histological examination when the mice were 8-week-old. The expressions of PCNA and Caspase-3 in spermatogenic cells were determined by immunohistochemical method. The results showed that the thickness of the convoluted seminiferous tubule decreased, cellular deranged, cell layers decreased and intercellular space increased. Meanwhile, there were a large number of abnormal shedding of spermatocytes and round spermatids, but few elongated spermatids and spermatozoa. The expression of PCNA extremely significantly decreased and the expression of Caspase-3 extremely significantly increased as quinestrol dose increased (P<0.01). Quinestrol caused male reproductive function injury through inhibiting proliferation and promoting apoptosis in spermatogenic cells by regulating the expression of associated proteins.  相似文献

The effect of enrofloxacin on sperm quality in male mice   总被引:1,自引:0,他引:1  
Current study was designed to evaluate toxic effects of enrofloxacin on male mice reproductive system. In the treatment group enrofloxacin was administered subcutaneously to male mice at a fixed dose of 150 mg/kg once daily for 15 days, whereas saline solution was given in the same regimen in the control group for the same period. Mice were sacrificed on day 15 and analyzed for sperm quality. In addition to routine examination of sperm material, spermatogenetic activity and organization of each animal were graded according to Johnsen's scoring to assess the spermatogenesis relying on seminiferous tubule cross-section scores. A significant decrease in both epididymal sperm count and sperm motility besides abnormal spermatozoa rate were observed in enrofloxacin group compared to controls (P < 0.01, for all). Johnsen's score in control mice were better than those in treatment group (P < 0.01). These results suggested that a fixed 150 mg/kg dose of enrofloxacin would lead disruption of spermatogenesis in the testes causing deterioration of motility and content of sperms as well as morphological abnormalities.  相似文献

为研究长链非编码RNA NEAT1对小鼠精子发生的影响,构建针对NEAT1的干扰载体并包装慢病毒,通过将干扰慢病毒注射到成年小鼠睾丸曲细精管和定量PCR,H.&E.染色等方法观测NEAT1对小鼠精子发生的影响。研究结果表明,NEAT1在睾丸组织和GC-1生殖细胞系中表达;成功构建CD513B-U6-NEAT1干扰载体,利用第三代慢病毒包装系统包装获得干扰慢病毒且干扰效果明显(NEAT1转录本水平下调69%);慢病毒注射试验结果显示干扰病毒注射组睾丸指数轻微增加,有精子发生的曲细精管比例由97%减少为86%(P〈0.05),表明小鼠精子发生受到影响,NEAT1具有维持雄性小鼠精子发生的作用。  相似文献

选择体质量为(24.69±1.01)g的雄性昆明小鼠90只,随机分为5组。对照组为不含棉酚的全价饲料组,试验组日粮中添加不同质量浓度的提取棉酚,试验I~Ⅳ组添加量分别为每天40、80、120、160mg/kg。结果显示,试验6周结束后,试验小鼠体质量随日龄增加而相应上升,各组之间差异不显著。但试验Ⅱ组到试验后期体质量开始下降。试验组小鼠在饲喂棉酚2周后出现较明显的生理反应:拉稀、活动减少、被毛松散无光泽;试验结束,试验Ⅲ组小鼠解剖肾脏有出血点、质地变脆。试验Ⅳ组小鼠两侧肠膜变黄,腹膜变薄,胃肠蠕动较慢,肾脏有出血点现象。各组在第2周和试验结束后分别取样,发现试验组小鼠肾脏出现不同程度的病变。试验Ⅱ组小鼠肾脏病变较轻微,可试验Ⅲ组小鼠肾脏有不同程度的肾间质出血和肾小管上皮细胞颗粒变性坏死;试验Ⅳ组小鼠肾梗死、肾小球毛细血管扩张,充血,出血。结果表明:棉酚对小鼠的体增重有一定的抑制作用,但对肾脏的生理和病理损伤较大。  相似文献

Reasons for performing study: Specific patterns of cytoskeletal filaments reflect a functional state of the cell. In testicular cells intermediate filaments (IFs) are of the vimentin type. Since it is known that Sertoli cells regulate the spermatogenic function in the male gonad, it became important to propose a system that could quantify the state of seminiferous tubular quality. To date, a Johnsen score system has never been used to equine testes. Objectives: To demonstrate the expression pattern of vimentin in testes of mature Arabian stallions and correlate its distribution with grade of seminiferous tubule impairment as indicated by a Johnsen score. Methods: For histological examination by the Johnsen method, routine haematoxylin‐eosin staining was used. Vimentin expression and its presence in testicular sections and testicular homogenates were detected by immunohistochemistry and western blot, respectively. Both analyses were performed qualitatively and quantitatively and further validated by ANOVA tests. Results: Distinct morphology of seminiferous tubules was found in testes harvested from 3 stallions. Vimentin in IFs was immunolocalised to the cytoplasm of Sertoli, Leydig and peritubular‐myoid cells. The intensity and pattern of the IFs staining was different in individual seminiferous tubules suggesting a correlation between vimentin expression and the severity of tubule degeneration. Qualitative results by immunohistochemistry and western blot were confirmed by further quantitative analyses. Conclusions: In equine testes, differential expression of vimentin was found to be correlated with the impairment of seminiferous tubules indicated by a decrease in Johnsen score. Potential relevance: The Johnsen score system may be a useful method to facilitate the identification of tubular alterations in the stallion testes. Combined histological and immunohistochemical approach may provide a detailed phenotypic classification of stallions with decreased fertility.  相似文献

High levels of estrogen produced by boar testes and the presence of estrogen receptors in both interstitial and tubular compartments are consistent with a direct role for estrogen in regulation of testicular cell function. This study investigated the importance of estrogen on hormone production by Leydig cells and seminiferous tubules in the developing boar. Thirty-six 1-week-old littermate pairs of boars were treated weekly with vehicle or 0.1 mg/kg BW Letrozole, an aromatase inhibitor, until castration at 2, 3, 4, 5, 6, 7, or 8 months. Tissue was collected and Leydig cells and seminiferous tubules were isolated. In a separate study, five untreated boars (ages 1.5-4 months) were castrated and Letrozole was added in vitro to Leydig cell and seminiferous tubule cultures. Leydig cells were cultured for 24h with and without porcine LH. Media were assayed for estradiol (E(2)) and testosterone (T) concentrations by RIA. Seminiferous tubules were cultured for 4h with and without porcine FSH; media were assayed for E(2) and immunoreactive inhibin (INH). In vivo aromatase inhibition decreased basal E(2) and increased basal T production by cultured Leydig cells. Basal seminiferous tubule production of E(2) but not INH was reduced. Decreasing estrogen synthesis in vivo did not alter LH-induced Leydig cell E(2) production or FSH-induced seminiferous tubule INH production. INH production decreased with advancing age regardless of treatment. In conclusion, in vivo aromatase inhibition altered baseline steroid production by cultured Leydig cells and seminiferous tubules but had little effect on response to gonadotropins.  相似文献

为探讨枸杞多糖(LBP)对己烯雌酚(DES)致成年雄性仓鼠生殖损伤的保护作用,66只成年雄性仓鼠随机分为空白对照组(A)、DES组(B)、LBP组(C、D、E)及VC+VE组(F)。除对照组注射橄榄油外,其余5组皮下注射DES,连续用药7d,同时C、D、E组分别灌服1,10,50mg/kg的LBP,F组灌服100mg/kg的VC及200IU/kg的VE,A、B组灌服等剂量的生理盐水,于末次给药24h后取材。摘取睾丸、附睾和精囊腺测质量并观察睾丸组织结构,检测血清中SOD、GSH-Px、MDA及T、LH、FSH、E2的水平。结果显示,DES能显著降低仓鼠体质量及生殖器官质量,而10,50mg/kg LBP及维生素对其有显著缓解作用(P〈0.05);DES处理组仓鼠睾丸组织结构严重损伤,应用LBP后其损伤程度显著减轻,其中以10,50mg/kg LBP的作用最为明显,VC+VE的缓解作用与50mg/kg LBP组接近;随着LBP剂量的增加,血清中SOD、GSH-Px的水平增加(P〈0.01),而MDA的水平下降(P〈0.01),维生素组与LBP组的变化趋势一致;LBP对血清激素水平也有不同程度的提高,其中以50mg/kg作用最为显著(P〈0.01)。这表明50mg/kg LBP能显著缓解DES致成年雄性仓鼠睾丸生精损伤。  相似文献
