Folates (B vitamins) are essential for the proper function of many bodily processes. Although a rich natural source are vegetables, the literature lacks data on the effect of the pre-treatment and freezing technologies used in vegetable processing and frozen storage time on the folate content in these materials. Moreover, since folates are very unstable nutrients, the amount available in processed and stored foods can be significantly lower than in raw products. In tested vegetables (green beans, yellow beans, peas, cauliflower, broccoli and spinach), one folate form was identified, 5-methyltetrahydrofolate (5-CH3-H4folate). It was observed that pre-treatment and freezing technology significantly (p?<?0.05) decreased 5-CH3-H4folate content only in vegetables with the largest degree of fragmentation (cut and briquetted spinach) and the smallest size (peas). In all analyzed samples, the 5-CH3-H4folate content decreased with the time of frozen storage. In frozen cauliflower, the 5-CH3-H4folate loss exceeded 95 % compared to the fresh product just after the third month of frozen storage. Meanwhile, in green and yellow beans, significant 5-CH3-H4folate losses (at the level of 75 % and 95 %, respectively) were observed no earlier than after the 9th month of frozen storage.  相似文献

High-performance liquid chromatography was used to study the stability of folate vitamers in two types of rye breads after baking and 16 weeks of frozen storage. Bread made using sourdough seeds contained less total folate (74.6 μg/100 g dry basis, expressed as folic acid) than the whole rye flour (79.8 μg/100 g dry basis) and bread leavened only with baker’s yeast (82.8 μg/100 g dry basis). Most importantly, it was generated by a significant decrease in 5-CH3-H4folate form. The baking process caused some changes in folate distribution. Storage of breads at -18°C for 2 weeks did not have a significant effect (p < 0.05) on total folates compared to the content directly after baking. After a 5-weeks storage period, a significant decrease (p < 0.05) in the content of total folates was recorded and it dropped on average by 14% for both type of breads. After a longer period of storage (16 weeks), a 25% loss of folates in the bread made with baker’s yeast and a 38% loss in the bread fermented with sourdough seeds was found. Retention of 5-CH3-H4folate and 10-HCO-H2folate forms were much lower in the bread made with a sourdough addition than with baker’s yeast only.  相似文献

Wholegrain cereals are good sources of folate. However, little is known about folate in barley. In this study, total folate was analysed in five hulled Finnish barley cultivars from three harvesting years (2006–2008). In addition, different fractions were produced from two hulled cultivars by scarification and from mixes of hulled cultivars by industrial milling. Total folate was determined in all samples with a microbiological method and vitamer distribution in scarification and industrial milling fractions, using HPLC or UPLC. Limited variation was observed in the total folate content of the five cultivars. The average contents of the three harvest years ranged from 563 to 773 ng/g dm. Fractions containing germ and outer layers were the richest in folate. The highest total folate content in the scarification fractions was up to 1710 ng/g dm. The total folate content of the hull fraction obtained by dehulling 10% of the grains with an industrial pearler was almost 4-fold compared to the corresponding dehulled grain. This hull fraction is normally not used as food. The main folate vitamers in the fractions were 5-HCO-H4folate, 5-CH3-H4folate, 5,10-CH+-H4folate, and 10-HCO-PGA. Folate content could be enhanced naturally by introducing folate-rich fractions of barley into cereal products.  相似文献

Folate (vitamin B?) content was evaluated in 10 varieties of coriander with the aim of enhancing its concentration and stability, because of three reasons: 1) coriander is among a few widely used greens in the world and suits many cuisines, 2) folate deficiency is prevalent in developing countries causing anaemia, infant mortality and neural tube closure defects, and 3) natural folate is preferred due to doubts about health risks associated with the synthetic form. In C. sativum, the highest folate content of 1,577 μg/100 g DW was found in var. GS4 Multicut foliage of mature plants (marketable stage) with an insignificantly higher content (1,599.74 μg/100 g DW) at flowering, which is a stage not preferred in markets. In callus cultures treated with plant growth regulators (GRs) (6-benzylaminopurine, kinetin and abscisic acid) substantial increase in folate occurred after 6 h, whereas elicitors (methyl jasmonate and salicylic acid) caused rapid 2-fold increase of folate, particularly in response to salicylic acid. Based on these observations, foliar applications were done for in vivo plants, where salicylic acid (250 μM, 24 h) also enhanced folate level by 2-folds (3,112.33 μg/100 g DW), although the content varied with diurnal rhythms. Stability of folates in treated coriander foliage was 10 % higher than in untreated foliage when stored at 25 °C and 4 °C. This study has established for the first time that coriander foliage is rich in folates, which can be doubled by elicitation and impart 10 % more stability than control during processing and storage.  相似文献

Mature cactus pears from Opuntia stricta have a dark purple color due to high betacyanin concentration, whose biosynthesis is initiated with the amino acid L-tyrosine as a primary precursor. This study followed the maturation and ripening processes of Opuntia stricta fruits to harvest them at high betacyanin and other antioxidant concentrations. Fruits lasted 9 months for final ripening. Physical and compositional changes at different maturation and ripening stages have been determined. Thus, ripe fruits were around 4.72?±?0.10 cm length, 2.94?±?0.05 cm diameter and 22.71?±?0.20 g weight; moisture and pH were maintained at 87.05?±?0.19 % and 3.37?±?0.12, respectively. Purple pigment production started in the ovary of immature fruits four months after anthesis (MAA). Concentration of all analyzed metabolites increased from immature (4 MAA) until ripe (9 MAA) stage. In ripe fruits, reducing sugars were 4.72?±?0.54 g/100 g ff and total phenols 135.17?±?0.68 mg gallic acid/100 g ff. Metabolites identified by HPLC were the betacyanins: betanin (60.17?±?1.08 mg/100 g ff), isobetanin (7.58?±?0.94 mg/100 g ff) and betanidin (13.48?±?0.87 mg/100 g ff). Also, L-ascorbic acid (35.03?±?1.06 mg/100 g ff) and L-tyrosine (4.43?±?0.73 mg/100 g ff) were determined. Antiproliferative effect of Amaranthus mantegazzianus proteins and peptides released after simulated gastrointestinal digestion (DH% 37.8?±?3.8) was investigated on human colon cancer cell line HT-29. Inhibition of proliferation of HT-29 cells was exhibited after a 24 h treatment with different concentrations of amaranth protein isolate (API) and the peptides released after digestion (DGS), presenting IC50 values of 1.35?±?0.12 and 0.30?±?0.07 mg soluble protein/mL, respectively. Lactate dehydrogenase assay indicated that both samples caused the loss of membrane integrity and cell lysis over HT-29 cells, and DAPI fluorescence microscopies evidenced typical apoptotic features. Moreover, Annexin V-FITC flow cytometry showed a significant increase of early apoptotic and late apoptotic/necrotic HT-29 cells compared to untreated ones, and caspase-3 assay confirmed the apoptosis induction with a 43.0?±?10.3 and 65.8?±?12.7% increase of caspase-3 activity produced by a 2 mg/mL treatment of API and DGS, respectively. In conclusion, amaranth peptides successfully released after simulated gastrointestinal digestion would exert a potential antiproliferative activity over HT-29 tumor cells. This effect was linked to the induction of cell necrosis and apoptosis, supporting the idea of using amaranth proteins as a potential food alternative ingredient for functional foods.

Methane oxidation activity (MOA), methanotrophic bacteria (MOB), and CH4 emission in a paddy field fertilized with anaerobically digested slurry were compared between two varieties: a fodder rice variety Leaf star (LS) and a normal rice variety Kinuhikari (KN). Average MOA and MOB per unit dry root were significantly higher in KN (7.1?μmol?g?1 CH4?g?1 dry root h?1 and 3.7?×?107 MPN?g?1 dry root, respectively) than in LS (4.3?μmol?g?1 CH4?g?1 dry root h?1, 2.1?×?107 MPN?g?1 dry root), although MOA in the rice root per whole plant was not significantly different since root biomass of LS (1.5?g dry root plant?1) was significantly higher than that of KN (1.2?g dry root plant?1). MOA in the soil ranged from 0.031 to 0.087?μmol?g?1 CH4?g?1 dry soil h?1, but there were no significant differences in both depths (0–5 and 5–15?cm) between the two rice varieties. MOA in the rhizosphere soil was significantly different between the rice varieties at flowering, but not at tillering. Methane emission in the field was lower and MOA and MOB in the rice root were higher in LS than in KN at tillering, but the reverse results were found at flowering and maturity stages. Total CH4 emission during a growing period was not significantly different between LS (27.4?±?16.9?g CH4?m?2) and KN (24.0?±?19.5?g CH4?m?2). One of the most important strategy in the treatment of obesity includes the development of nutrient digestion and absorption inhibitors. Inhibition of digestive enzymes is one of the most widely studied mechanisms used to determine the potential efficacy of natural products as hypolipidemic and hypoglycaemic agents. In vitro studies here reported were performed to evaluate the inhibitory activity of five species (as hydroalcoholic extracts) of edible plants from Calabria region (Italy) on amylase and lipase by monitoring the hydrolysis of p-NPC and the hydrolysis of glycoside bonds in digestible carbohydrate foods. The formulation obtained from Clematis vitalba L. exhibited the strongest inhibitory effect on pancreatic lipase (IC50?=?0.99 mg/ml) and on α-amylase (IC50?=?31.52 μg/ml). The juçara fruits (Euterpe edulis Martius), native to the Atlantic Forest, are rich in anthocyanins. To preserve the anthocyanins in juçara fruit pulp, this study aimed to evaluate the effectiveness of microencapsulation by spray drying and freeze drying with maltodextrin (dextrose equivalent 16.5 to 19.5) and gum arabic in different proportions. The obtained microparticles were characterized by quantifying the total polyphenol and anthocyanin contents, by performing differential scanning calorimetry, thermogravimetry, and infrared spectroscopy and by using scanning electron microscopy to analyze the morphology of the particles. The total amount of polyphenols in the fruit pulp was 750?±?16.7 mg GAE/100 g of the freeze-dried sample. The total anthocyanins in the fruit pulp was 181.25?±?5.36 (mg/100 g). The microparticles were formed by employing maltodextrin and gum arabic in a 1:1 proportion as the polymeric matrix; the mixtures of pulp and polymeric matrix were prepared in proportions of 2:3 and 2:1, preserving up to 83.69% of the anthocyanin content. Lyophilization of the 2:1 mixture resulted in an anthocyanin content of 116.89?±?4.43 (mg/100 g), whereas lyophilization of the 2:3 mixture resulted in 151.68?±?1.39 (mg/100 g) anthocyanin content, which did not differ from the value obtained by spray drying the 2:3 mixture (150.76?±?5.79 (mg/100 g)). Thermal analyses showed that the microparticles obtained by freeze drying at a ratio of 2:3 presented greater resistance to degradation with increasing temperature. The incorporation of the pulp in the polymeric matrix was demonstrated by IR analyses. Microparticles obtained by freeze drying showed the formation of various-sized flakes, whereas those obtained by spray drying were spherical in shape. Microencapsulation is a possible alternative for improving the stability of the anthocyanins in this fruit.  相似文献

Chiltepin, a wild chili mostly used in different traditional foods and traditional medicine in Northwest Mexico, represents a source of polyphenols. However, studies about the bioaccessibility of polyphenols as a parameter to measure the nutritional quality and bioefficacy of them in the fruit after consumption are scarce. Chiltepin showed phenolic acids and flavonoids contents between 387 and 65 μg/g, respectively. Nevertheless, these values decreased after the digestion process. Before digestion, gallic acid, 4-hydroxibenzoinc acid, chlorogenic acid, caffeic acid, p-coumaric acid, quercetin and luteolin were the main polyphenols found in chiltepin by HPLC-DAD and confirmed by FIA-ESI-IT-MS/MS. Gallic and chlorogenic acids were non-detected in the gastric phase, while only p-coumaric acid (5.35?±?3.89 μg/g), quercetin (5.91?±?0.92 μg/g) and luteolin (2.86?±?0.62 μg/g) were found in the intestinal phase. The bioaccessibility of phenolic acids, flavonoids, and total polyphenols after the intestinal phase was around 24, 17 and 23%, respectively. Orange juice is an important source of flavanones in the Western diet. However, little is known of the variation in flavanone content of shop-bought orange juice with pulp (OJP) or without pulp (OJ), nor the impact of pulp on the fate of flavanones in the gut. Total phenols, total flavonoids, antioxidant capacity, hesperidin and narirutin, and dietary fibre were measured in six orange juice brands sold as OJP and OJ. The inclusion of pulp had little impact on fibre content. Apart from total phenols (OJ: 208.4?±?10.7 μg gallic acid equivalents (GAE) ml?1; OJP: 225.9?±?16.7 μg GAE ml?1, P?<?0.05), there were no differences between OJ and OJP. The fate of flavanones in OJ and OJP (Tropicana) were further compared using in vitro gastrointestinal (GI) models. After in vitro upper GI digestion, recovery of hesperidin was higher in OJ compared with OJP (89?±?6 vs. 68?±?3%, P?=?0.033). After 2 h colonic fermentation, hesperidin was 1.2 fold higher in OJP than OJ. However, after 24 h colonic fermentation there was no significant difference between juices in terms of hesperidin, hesperetin, narirutin, naringenin and catabolites. In conclusion, the amount of pulp included in these shop-bought orange juices had little impact on flavanone metabolism in models of the GI tract. The effects of greater amounts of orange pulp remain to be determined.

Cereals are recognised as an important food source of folate, and germinated cereals are reported to contain even more folate. This study examined the effects of germination and oven-drying on folate content in different wheat and rye cultivars. The native folate content in four wheat cultivars ranged from 23 to 33 μg/100 g dry matter (DM) and that in six rye cultivars from 31 to 39 μg/100 g DM. Mean folate content in rye was 25% higher than in wheat. Germination of both cereals resulted in a 4- to 6-fold increase in folate content, depending on cultivar and duration of germination. The highest folate content in both cereals was found after 96 h of germination and was 181 μg/100 g DM for cv. Kaskelott (rye) and 155 μg/100 g DM for cv. Kosack (wheat). Germination increased the amount of 5-CH3–H4folate in both cereals from 45 to 75%. Oven-drying of germinated wheat grains (for 48 and 72 h) at 50 °C did not affect the folate content. In conclusion, germination increases the folate content in wheat and rye cultivars, while subsequent oven-drying does not affect the folate content. Germination can therefore be recommended for producing bakery ingredients with increased folate content.  相似文献

Bacterial spot of tomato (Solanum lycopersicum) caused by Xanthomonas campestris pv. vesicatoria (Xcv) is a devastating disease of tomato world-wide. In the southeastern United States, high summer temperatures and humidity are ideal conditions for this disease, resulting in defoliation, fruit spotting and a significant reduction in fruit yield. Current organic production practices restrict the use of synthetic chemicals for disease control; hence, there is a need for the development of new and effective biopesticides to mitigate plant diseases. Among several biological agents with potential for disease control, Lentinula edodes (shiitake mushroom) has been shown to have antibacterial properties. A controlled-environment study was therefore conducted to validate L. edodes mycelia culture filtrate (Lemcf) foliar application to control bacterial spot of tomato using the cultivar Agriset 761. Lentinula edodes mycelia culture filtrate foliar spray significantly suppressed bacterial spot incidence in tomato foliage in vitro but was not effective in vivo. The phytotoxicity symptoms in Lemcf-treated tomato foliage were attributed to the presence of 422.78 μg of oxalic acid per milliliter of Lemcf (quantified by high-pressure liquid chromatography). Plant height and flowering were normal in Lemcf-treated plants. Additionally, Lemcf seed treatment did not adversely impact tomato germination but significantly enhanced the germination of marginal tomato seeds subjected to biotic stress (Xcv). Our results suggest that after eliminating oxalic acid from Lemcf, the product may be a potential biopesticide for managing bacterial spot of tomato. Future greenhouse or field experiments should be conducted after eliminating oxalic acid from L. edodes culture filtrates.  相似文献

To study the radiation utilization efficiency, latent heat flux, and simulate growth of rice during post-flood period in eastern coast of India, on-farm trial was conducted with three water regimes in main plots (W 1 = continuous flooding of 5 cm, W 2 = irrigation after 2 days of water disappearance, and W 3 = irrigation after 5 days of water disappearance) and five nitrogen levels in subplots (N 1 = 0 kg N ha?1, N 2 = 60 kg N ha?1, N 3 = 90 kg N ha?1, N 4 = 120 kg N ha?1, and N 5 = 150 kg N ha?1) on a rice cultivar, ‘Lalat’. Average maximum radiation utilization efficiency (RUE) in terms of above ground dry biomass of 2.09 (±0.05), 2.10 (±0.02), and 1.9 (±0.08) g MJ?1 were computed under W 1, W 2, and W 3, respectively. Nitrogen increased the RUE significantly, mean RUE values were computed as 1.60 (±0.07), 1.78 (±0.02), 2.060 (±0.08), 2.30 (±0.07), and 2.34 (±0.08) g MJ?1 when the crop was grown with 0, 60, 90, 120, and 150 kg ha?1 nitrogen, respectively. Midday average latent heat flux (on clear days) varied from 7.4 to 14.9 and 8 to 13.6 MJ m?2 day?1 under W 2 and W 3 treatments, respectively, at different growth stages of the crop in different seasons. The DSSAT 4.5 model was used to simulate phenology, growth, and yield which predicted fairly well under higher dose of nitrogen (90 kg and above), but the model performance was found to be poor under low-nitrogen dose.  相似文献

Potato yellow vein virus (Crinivirus) causes leaf yellowing, is limited to the phloem and is transmitted by Trialeurodes vaporariorum and tubers. PYVV causes up to 50 % yield reduction in S. tuberosum. The prevalence of PYVV was established in Group Phureja in 2008 in the Colombian states of Cundinamarca, Antioquia and Nariño. The presence (S) or absence (NS) of symptoms was established in 24,930 plants. Symptom prevalence was as follows: Cundinamarca 0.74 %?±?0.03, Antioquia 5.4 %?±?12 and Nariño 6.2 %?±?0.11. Percentages of S plants were highly variable among different states and municipalities. PYVV was diagnosed by RT-PCR in a subsample of 1,545 plants from Cundinamarca and Nariño. RT-PCR showed that 2.4 %?±?8.3?×?10?5 and 41 %?±?0.0009 % of the symptomless plants of Cundinamarca and Nariño respectively were positive. This suggests possible late infection of plants by the vector which did not have enough time to express symptoms, infections with two or more viruses or “virus latency”. PYVV symptomatic prevalence in Group Phureja was lower than that reported for Diacol-Capiro (Group Andigena), but increases if molecular results are taken into account. This is the first known study aimed at determining PYVV prevalence using molecular tools and extensively sampling of S.tuberosum Group Phureja plants in grower’s fields in three states and multiple municipalities of Colombia.  相似文献

Antioxidant properties of amaranth extracts isolated sequentially by acetone and methanol/water from defatted plant leaves, flowers, stems and seeds were assessed by ABTS+?, DPPH?, ORAC and total phenols content (TPC) assays. In addition, antioxidant properties of solid plant material were evaluated by the direct QUENCHER method using the same assays. Leaves and flowers of amaranth as well as their extracts possessed the highest antioxidant activities. Radical scavenging capacity in ABTS+? assay for leaves, flowers, stems and seeds evaluated by QUENCHER method were 144.24?±?2.41, 112.33?±?7.45, 19.05?±?1.13 and 21.82?±?1.06 μmol trolox equivalents in 1 g of dry weight, respectively. On-line HPLC-DPPH? assay was used to determine the activity of separated compounds and it was observed that rutin was the main radical scavenger in amaranth extracts. Preliminary screening of extract composition was performed by UPLC/ESI-QTOF-MS and rutin, nicotiflorin, isoquercitrin, 4-hydroxybenzoic and p-coumaric acids were identified by measuring their accurate mass and retention time.  相似文献

There is evidence that a diet rich in fruit and vegetables may reduce the risk of cancer and other degenerative diseases. However, potential health impact of bioactive phytochemicals is limited by their low amount and relatively poor bioavailability. It has been suggested that the health benefits associated with fruit and red wine consumption could be due to the whole antioxidant pool of the diet microcomponents. In this study, the antioxidant activities of trans-resveratrol, pterostilbene and quercetin, and the effect of their combination were investigated in human erythrocytes in vitro. H2O2-induced lipid peroxidation was assessed by measuring the amount of thiobarbituric acid reactive species. Quercetin and pterostilbene protected erythrocyte membranes against lipid peroxidation (IC50 values = 64 ± 8.7 μM and 44.5 ± 7.8 μM, respectively). Resveratrol was significantly less effective. However, the three compounds protected the erythocytes against hemolysis and GSH (reduced glutathione) depletion to the same extent. Combinations consisting of two compounds (molar ratio 1:1) influenced lipid peroxidation in a concentration–dependent manner. At lower concentrations, resveratrol with quercetin or pterostilbene inhibited synergistically the oxidative injury of membrane lipids At higher concentrations, an additive effect was observed. These protective effects may partially explain the health benefit of these bioactive microcomponents when together in the diet.  相似文献

A first cut of timothy, treated with water (untreated), formic acid (FA), cellulase + lactic acid bacteria (CB), cellulase + hemicellulase (CH) or cellulase + hemicellulase + a lignin-modifying enzyme (CHL), was ensiled in pilot-scale silos. Silages, except CB, were fed to four male cattle, each equipped with a rumen and duodenal cannula, in a digestibility trial designed as a 4 × 4 Latin square. The animals were fed a diet of 400 g of concentrate and 600 g of silage at a level of 70 g DM kg?1 live weight (LW0·75). All enzyme-treated silages were well-preserved with a more extensive fermentation than in FA silage. The quality of untreated silage was poorer as indicated by higher pH and ammonia-N content. The amount of effluent from enzyme-treated silages ranged from 116 to 127 g kg?1; for FA and untreated silages values were 101 g kg?1 and 80 g kg?1, respectively. Total DM losses from enzyme-treated silages were higher than from FA silage (P < 0·05). No significant differences were noticed between silages in the apparent digestibility of organic matter (OM), neutral-detergent fibre (NDF), acid-detergent fibre (ADF) or nitrogen (N). The apparent digestibility of cellulose was higher with enzyme-treated silages than with FA silage (P < 0·05). The values for microbial N flow at the duodenum were 80·0, 91·9, 80·7 and 70·5g N d?1, and for the efficiency of rumen microbial N synthesis 38·6, 47·6, 36·9 and 32·5 g N kg?1 OM apparently digested in the rumen for untreated, FA, CH and CHL silages, respectively. In the rumen the molar proportion of propionate was higher (P < 0·01) and that of butyrate lower (P < 0·01) with enzyme-treated silages when compared with FA silage. The proportion of butyrate was also lower with untreated than with other silages (P < 0·01). The rumen residence time of NDF and ADF was longer (P < 0·05) with enzyme-treated silages than with FA silage.  相似文献
