Little is known about the levels of resistance to Orobanche crenata available in narbon vetch. A germplasm collection of 200 accessions of narbon vetch (Vicia narbonensis L.) originating from different countries was screened for resistance to crenate broomrape under field conditions. Thirteen accessions were selected for more detailed screening under controlled conditions and for additional field testing. Resistance to O. crenata was manifested by lower germination of broomrape seeds, reduced emergence of Orobanche shoots and fewer root attachments per narbon vetch plant. Differences in pod yield among narbon vetch accessions were also found in response to Orobanche infestation. Finally, eight accessions were selected and identified as new sources of narbon vetch resistant to broomrape.  相似文献

Racial evolution of sunflower broomrape (Orobanche cernua) has been very rapid in Spain during recent years, in which resistance has been overcome several times and there has been an important increase in areas infested with this parasitic angiosperm. In order to find resistance to a highly virulent population of sunflower broomrape that could be used directly in breeding programmes, three different sets of cultivated plant material composed of 429 entries were tested by artificial inoculation. All evaluated inbred lines from Moden, Canada, were fully susceptible. Out of the 240 P.I. accessions tested, only 10 segregated for resistance to broomrape, the rest being susceptible. From the 160 USDA breeding lines evaluated, 5% were resistant and 19% segregated for resistance to O. cernua. These lines traced back mainly to crosses of RHA 274 and RHA 801 with Russian, Turkish and Romanian hybrids. The origin of P.I. accessions that segregated for resistance were primarily derived from the former USSR and from Romania.  相似文献

Phylogenetic relationships of 15 genotypes of the genus Lens and 7 of their interspecific hybrids were determined by morphological (quantitative and qualitative) characters. The first multivariate analysis was conducted on quantitative characters and second analysis was conducted on qualitative characters. In the first analysis on the basis of two canonical variates, accessions of Lens nigricans, L. odemensis and L. culinaris ssp. orientalis grouped together, while L. ervoides and L. culinaris ssp. culinaris formed two different groups. The second analysis yielded different results based firstly on two canonical variates where L. nigricans and L. culinaris ssp. culinaris formed two separate groups but the other three (sub)species clustered together when the third canonical variate was included. L. odemensis separated from this larger group and formed a separate group. The phylogenetic relationships of parents and their hybrids have also been discussed. Genomic DNA from an inbred line from the Orobanche crenata resistant cultivar 'Giza 402' was restricted and enriched for AG-containing simple sequence repeats (SSRs). The overall SSR enrichment rate was 54.6%. Of 73 selected SSR markers that successfully amplified in 'Giza 402' 54 were polymorphic among a group of 10 Egyptian and Spanish faba bean genotypes extensively used in Orobanche crenata resistance/susceptibility studies and linkage map development. The polymorphism information content ranged from 0.16 to 0.72 and Unweighted Pair-Group Method using Arithmetic averages cluster analysis based on these loci placed all resistant genotypes descended from the parental line F 402 in the same group. Successful amplification from 28 primer pairs was also observed in five European faba bean genotypes. The SSR loci described here will be extremely useful in studies focusing on Orobanche crenata resistance, genetic diversity studies and will provide additional co-dominant loci to complement existing faba bean linkage maps based primarily on dominant markers.  相似文献

Lentil (Lens culinaris Medik.) is an important cool‐season legume that can be severely damaged by rust (Uromyces viciae‐fabae (Pers.) Schröt.) in many parts of the world. In this work, a collection of L. culinaris germplasm accessions was screened under growth chamber conditions for response to U. viciae‐fabae. Several sources of incomplete resistance have been identified, both of hypersensitive and of non‐hypersensitive nature. Incomplete hypersensitive resistance was due to a late‐acting host cell necrosis that resulted in an intermediate infection type (IT). Incomplete non‐hypersensitive resistance was due to a hampered cell penetration not associated with host cell necrosis, reducing the number of haustoria per colony and therefore hindering the growth of the colony and reducing and retarding disease development in spite of a high IT.  相似文献

Using random amplified polymorphic DNA (RAPD) analysis we assessed the genetic relationships between 16 accessions and cultivars of lentil (Lens culinaris ssp. culinaris) in the Australian lentil breeding program. All lines exhibited polymorphism with a maximum dissimilarity value of 0.36. This indicated a limited degree of genetic variation. Polymerase chain reaction (PCR) with primers based on the flanking regions of the 5S rRNA gene from Pisum sativum amplified the non-translated spacer (NTS) region from within the 5S rRNA gene of Lens. Three distinct amplification banding patterns differentiated between restricted genomic DNA of Lens spp. L. culinaris ssp. culinaris and L. culinaris ssp. orientalis shared similar markers of two distinctly different NTS sizes. L. nigricans and L. odemensis shared the same amplification pattern of a single sized NTS region. However, L. ervoides contained two separate sizes of NTS, distinct from other Lens species. In an effort to widen the genetic base of cultivated lentil, these species-specific molecular markers may be used to follow potential introgression between species. The inheritance of resistance to a highly virulent population of sunflower broomrape (Orobanche cernua) has been studied in R-41, a Spanish sunflower restorer inbred line. Using the cytoplasmic male-sterile inbred line HA-89 (cms; very susceptible to this population of broomrape) as a female parent, progenies of the cross with R-41, i.e. Fl, F2 and BC1 to both parents, as well as the parental lines, were analysed for their reaction to the broomrape population EC-94. The goodness of fit of the observed vs expected segregation ratios indicated that the inheritance of resistance to broomrape in line R-41 is conferred by two independent dominant genes.  相似文献

Genetic resistance to broomrape (Orobanche cumana Wallr.) race F in sunflower line J1, derived from the wild perennial species Helianthusgrosseserratus Martens and Helianthus divaricatus L., has been reported to be controlled by dominant alleles at a single locus, Or6. However, deviations from this monogenic inheritance have been observed. The objective of the present study was to gain insight into the inheritance of resistance to broomrape race F in the sunflower line J1. F1, F2, F3 and BC generations from crosses between J1 and three susceptible lines, P21, NR5 and HA821 were evaluated. F1 hybrids showed both resistant (R) and moderately resistant (MR) plants, the latter having a maximum of five broomrape stalks per plant compared with >10 in the susceptible parents. This indicated incomplete dominance of the Or6 alleles. F2 plants were classified as R, MR or susceptible (more than five broomrape stalks per plant). Three different segregation ratios were observed: 3 : 1, 13 : 3 and 15 : 1 (R + MR : S), suggesting the presence of a second gene, Or7, whose expression was influenced by the environment. A digenic model was confirmed, based on the evaluation of F2:3 families.  相似文献

Orobanche and Phelipanche, commonly known as broomrape, are dicotyledonous holoparasitic flowering plants that cause heavy economic losses in a wide variety of plant species. Breeding for Orobanche resistance is still one of the most effective management strategies for this weed. However, previous efforts to find broomrape-resistant tomato (Solanum lycopersicon) genotypes have been unsuccessful. Here, we report on the isolation and characterization of a fast-neutron-mutagenized M-82 tomato mutant, Sl-ORT1. The Sl-ORT1 mutant showed resistance to Phelipanche aegyptiaca as compared to cultivar M-82; segregation analysis suggested a single recessive ort1 allele. Sl-ORT1 broomrape resistance was reflected in a lower number of broomrapes per plant, reduced P. aegyptiaca fresh weight per plant, and the absence of broomrape’s negative effect on plant host growth and yield. Sl-ORT1 was shown to be resistant to high concentrations of P. aegyptiaca seeds, and to another three broomrape species: Phelipanche ramosa, Orobanche cernua, and Orobanche crenata. Grafting experiments demonstrated that roots, rather than shoots, are necessary for Sl-ORT1 broomrape resistance. In addition, Sl-ORT1 was shown to be resistant to broomrape under field conditions. Since yield parameters are slightly affected by the mutation, this resistance gene should be introduced into tomato varieties with different genetic backgrounds; this newly identified Orobanche-resistant mutant may be further utilized in breeding programs for Orobanche resistance.  相似文献

The genetics of resistance of common vetch (Vicia sativa L.) to broomrape (Orobanche crenata Forsk.) was studied for two years by using the P1, P2, F1, BC1, BC2, F2 F3, and F4 generations obtained from crosses between resistant and susceptible lines. Resistant lines were selected by screening a world collection m a naturally infested plot. Resistance was tested both under field and greenhouse conditions. The best index to measure resistance was the number of emerged broomrapes per host plant. The results fit the additive-dominance model. The main component of the variation was additivity; dominance and interaction effects seemed to depend on the environment. When dominance is expressed, a low number is dominant over a high number of broomrapes per host plant.  相似文献

A collection consisting of 140 accessions of chickpea (Cicer arietinum L.) and 109 accessions of related wild species (Cicer spp.) was screened for resistance to chickpea rust (Uromyces ciceris-arietini (Grognot) Jacz. & Boyd). Varying levels of partial resistance were identified, based on a reduced disease severity and area under disease progress curve without any macroscopically visible host cell necrosis. Higher levels of resistance were observed in wild Cicer species, but neither associated with hypersensitivity. Components of resistance have been macros- and microscopically studied in selected C. arietinum accessions. Resistance was expressed as longer latent period and lower infection frequency, which were associated with increased percentage of early aborted colonies, reduced number of haustorial mother cells and haustoria per colony, and reduced colony size.  相似文献

Interspecific crosses were made between the cultivated sunflower inbred line HA89 and accessions of five wild perennial Helianthus species (H. giganteus L., H. laevigatus T. & G., H. resinosus Small, H. pauciflorus Nutt. and H. decapetalus L.) resistant to broomrape ( Orobanche cernua Loefl.). Using the genetic male-sterile isogenic version of that line as female, successful reciprocal crosses were also obtained with all these wild species except with H. decapetalus. Five-day-old hybrid embryos were excised and cultured in vitro. In all cases, few mature plants were obtained from embryos in early developmental stages (early heart and globular) but up to 28% mature plants were obtained from embryos in later stages of development. Very immature embryos were difficult to excise without damage. Hybrid embryos and mature plants were obtained from all five wild species. Interspecific hybrid embryos from different wild species showed distinct developmental potentials, the proportion of hybrid embryos in different developmental stages varying among species. Differences in the proportion of hybrid embryos at the several developmental stages were also obtained for the reciprocal crosses (cultivated × wild), which showed higher proportion of fully developed embryos. Hybrids involving H. giganteus and cultivated sunflower were difficult to obtain without the use of embryo culture. Embryo culture proved to be an useful tool to overcome post-zygotic hybrid incompatibility in different Helianthus spp., and facilitated interspecific transfer of resistance to O. cernua. Lathyrus sativus has high potential both as food and fodder crop but can be severely infected by rust. This study assessed the resistance level of a collection of L. sativus accessions against three rust species ( Uromyces pisi , U. ciceris-arietini and U. viciae-fabae ) and characterized, for the first time, macro and microscopically the resistance to U. pisi . All L. sativus accessions were highly resistant to U. ciceris-arietini and to U. viciae-fabae but showed a compatible interaction with U. pisi. However, in spite of this compatible interaction, high levels of rate reducing resistance were identified with significant disease severity reduction, fitting the Partial Resistance definition. Detailed histological observations showed that resistance was mainly because of haustorium formation restriction with high-level of colony early abortion, and reduction in the haustoria number per colony. This was occasionally associated with reduction on the infection hyphae intercellular growth, in a clear example of prehaustorial resistance. Differences in germination, orientated germ tube growth and appressoria differentiation were observed but were, generally, of marginal importance to explain the observed resistance levels.  相似文献

A collection of 111 barley landraces from the Fertile Crescent was screened for resistance to barley leaf rust in the field and under controlled conditions. Large variation was observed for disease severity under field conditions. Accessions with high resistance because of hypersensitivity were identified. Also segregation was observed in some accessions, with individual plants showing hypersensitive reactions (IT ≤ 6). Partial resistance due to a reduction of infection in spite of a compatible infection was commonly found (19%). Resistance of 12 accessions selected for their low disease severity and high IT, was shown to be due to a prolonged latency period and increased percentage of early aborted colonies not associated with host cell necrosis. A high correlation was observed between the microscopic and macroscopic components of partial resistance.  相似文献

Summary Faba beans are adversely affected by numerous fungal diseases leading to a steady reduction in the cultivated area in many countries. Major diseases such as Ascochyta blight (Ascochyta fabae), rust (Uromyces viciae-fabae), chocolate spot (Botrytis fabae), downy mildew (Peornospora viciae) and foot rots (Fusarium spp.) are considered to be the major constraints to the crop. Importantly, broomrape (Orobanche crenata), a very aggressive parasitic angiosperm, is the most damaging and widespread enemy along the Mediterranean basin and Northern Africa. Recent mapping studies have allowed the identification of genes and QTLs controlling resistance to some of these diseases. In case of broomrape, 3 QTLs explained more than 70% of the phenotypic variance of the trait. Concerning Ascochyta, two QTLs located in chromosomes 2 and 3 explained 45% of variation. A second population sharing the susceptible parental line also revealed two QTLs, one of them likely sharing chromosomal location and jointly contributing with a similar percentage of the total phenotypic variance. Finally, several RAPD markers linked to a gene determining hypersensitive resistance to race 1 of the rust fungus U. viciae-fabae have also been reported. The aim of this paper is to review the state of the art of gene technology for genetic improvement of faba bean against several important biotic stresses. Special emphasis is given on the application of marker technology, and Quantitative Trait Loci (QTL) analysis for Marker-Assisted Selection (MAS) in the species. Finally, the potential use of genomic tools to facilitate breeding in the species is discussed. The combined approach should expedite the future development of lines and cultivars with multiple disease resistance, one of the top priorities in faba bean research programs.  相似文献

Broomrape (Orobanche cernua Loelf.) is a sunflower parasite which has become a potential threat to this crop in Spam with an increasing incidence in recent last years. In order to investigate the racial situation of the parasite in Spam and to evaluate the resistance of differentials and inbred lines currently used in breeding programmes, two experiments were carried out. In the first, two sets of European sunflower differentials were inoculated with three broomrape populations from different geographical origin. The second experiment studied the reaction to the parasite of 38 inbred lines in comparison with resistant and susceptible checks. Reactions of differential lines resistant to known European broomrape races ranged from complete immunity of lines P-1380-2A and ‘Jdanovski 8281’ to complete susceptibility of ‘Record’ and the universal suscepts. These results confirmed the existence of several physiological races of broomrape in Spain and a different racial composition from that of Eastern Europe. Results from the second experiment showed good resistance of half the restorer lines tested and 36 % of the maintainers and cms lines. This material together with the resistant differential lines constitutes good starting material to be used in developing cultivars resistant to broomrape populations in Spain.  相似文献

向日葵列当是向日葵生产中重要的寄生性杂草。新疆阿勒泰地区和内蒙古乌拉特前旗是国内向日葵列当发生的重灾区。为筛选并为抗列当育种工作提供优质抗性材料,从根本上防治向日葵列当,利用田间自然发生向日葵列当的田块,采用随机区组设计设置小区试验;通过寄生率、寄生程度2 个指标判定对列当的抗感性。于2012 年在新疆阿勒泰对56 个向日葵品种(系),2013—2014 年在内蒙古乌拉特前旗分别对22 个品种(系)和20 个品种(系),进行抗列当鉴定。结果表明:在新疆22 个油葵品(系)中‘陇葵杂1 号’、‘F08-1’、‘辽丰F53’、‘赤CY101’、‘S26’、‘MGS’、‘S31’等16 个品种,34 个食葵品种(系)中‘JK106’品种,表现为免疫;在内蒙古乌拉特前旗2013—2014 年24 个油葵品种(系)中,‘赤CY102’、‘F08-1’等2 个品种表现为免疫,‘MSG’、‘新葵杂5 号’、‘赤029×115R’、‘赤128A×116R’等4个品种(系)表现为高抗,而18 个食葵品种(系)中,没有免疫品种,只有‘JK103’和‘JK106’表现为抗列当,其余16 个品种(系)均表现为感病和高度感列当。其中油葵品种‘赤CY102’在新疆和内蒙古抗列当鉴定中均表现免疫。在2 个不同地区进行向日葵品种抗列当鉴定,新疆地区表现免疫和高抗的品种较内蒙古地区多,说明内蒙古乌拉特前旗列当生理小种级别高于新疆阿勒泰地区列当生理小种。另外,油葵品种对列当的抗性较食葵高。  相似文献

Summary Twenty three accessions of nine Portuguese cabbage and kale land races from different geographic origins were tested at the seedling stage for resistance to several important brassica diseases. Resistance to downy mildew (Peronospora parasitica), expressed as necrosis of the cotyledon mesophyll, was found in all the accessions. Type A resistance to cabbage yellows (Fusarium oxysporum f. sp. conglutinans race 1) was present in most of the landraces. Resistance to clubroot (Plasmodiophora brassicae race 6) was found in one accession of the Portuguese tree kale. High resistance to blackleg (Leptosphaeria maculans) and white rust (Albuco candida) was not detected, although several accessions showed 20 to 30% of plants with intermediate expression of resistance. All Portuguese cole accessions were susceptible to blackrot (Xanthomonas campestris pv. campestris).  相似文献

Summary Twenty five tomato cultivars and one accession of wild tomato were evaluated for their resistance to branched broomrape (Orobanche ramosa) infestation. Tomato cultivars were found different in their resistance to Orobanche based on different growth parameters. Total Orobanche number, haustoria development and number of emerged Orobanche shoots were all different between tomato cultivars. Differences in the growth and fruit yield among tomato cultivars were also found in response to Orobanche infestation. Ranking tomato cultivars for Orobanche resistance indicates the existence of different resistance mechanisms in these cultivars. Relatively high to moderate levels of resistance were obtained in Tiny Tim, Acora, Castler, Pomodora, Orient, Red Alert and the accession LA 1478 of L. pimpinellifolium. The cultivar Tiny Tim showed the highest level of resistance for all measured parameters and in all experiments.  相似文献

The wild tetraploid wheat species Tr$$ (Zhuk) Zhuk Var. araratieum is a source of pest resistance genes for T$$ aesti$$ L. Our objectives were to describe the breeding behaviour of T.arartuititm when backcrossed to common wheat and transfer resistance to leaf rust (caused by Pu$$) and powdery mildew (caused by Blumeria $$wheat. Crosses were made between five wheat genotypes and $$ accessions. Fertifity and chromosome numbers of BC$$; plants were determined. Resistance to leaf rust was transferred toBC2 -derived families from 10 different T’ararati$$an accessions. Leaf rust resistance genes in nine T. araratieum accessions can be assigned to at least four loci. Leaf rust resistance transferred from three accessions was inherited in the hexaploid derivatives as a single. $$ gene in each case. Resistance to powdery mildew was also detected in the T. araratie$$ backcross derivatives. Fertile hexaploid derivatives expressing T’araratieum-derived resistance genes can be recovered after two backcrosses to wheat cultivars.  相似文献
