Two experiments were conducted with young pigs to determine the efficacy of ornithine (Orn) or citrulline (Cit) as precursors of arginine (Arg). In Exp. 1, pigs were individually fed an Arg-deficient, semipurified diet (.18% Arg) supplemented with .3% Arg or an equimolar quantity of Orn or Cit. Supplemental Arg or Cit increased rate and efficiency of weight gain, but Orn addition was without effect. Free Arg in plasma 3 h post-prandial was increased by addition of either Arg or Cit to the basal diet. Liver Arg was elevated by dietary addition of Arg, Orn or Cit; kidney Arg and Orn were elevated only in pigs receiving supplemental Cit. Arginine or Cit addition to the diet increased Arg concentration in muscle tissue, but muscle Orn was unresponsive to any of the supplements fed. In Exp. 2, pigs were again fed the Arg-deficient, semipurified diet supplemented with .3% Arg or four times an isomolar quantity of ornithine. Arginine addition to the diet increased weight gain and feed efficiency, while Orn supplementation was without effect. Plasma Orn was increased by excess Orn, while plasma Cit was unaffected by supplemental Arg or Orn. Moreover, excess Orn increased free Orn and proline (Pro) in liver, kidney and muscle. Free Cit, however, increased only in liver from feeding excess Orn. In addition, excess Orn decreased both plasma ammonia and free glutamine (Gln) concentration in brain. Arginase activity was roughly 10, 40 and 100 times greater in hepatic tissue than in renal cortex, renal medulla or intestinal mucosa, respectively, while hepatic ornithine transcarbamoylase (OTC) activity was about 15 times greater than the activity present in mucosa tissue. The catabolism of arginine (Arg) by mixed rumen bacteria (B), mixed rumen protozoa (P), and their mixture (BP) was quantitatively investigated in an in vitro system in order to confirm the metabolic pathway of Arg and provide basic information for enzymatic and molecular studies as well as an understanding of the quantitative distribution of metabolites. Rumen microbial suspensions (B, P, and BP) collected from fistulated goats were anaerobically incubated with or without 1 mmol/L Arg at 39°C for 12 h. Arg and other related compounds such as citrulline (Cit), ornithine (Orn), proline (Pro) and 5‐aminovaleric acid (5AV) in both supernatant and hydrolyzates of B, P, and BP suspensions were analyzed by HPLC. The metabolic pathways of Arg in mixed rumen bacteria and mixed rumen protozoa were considered to be as follows: rumen bacteria, Arg → Cit → Orn → Pro → 5AV → VFAs + NH3; rumen protozoa, Arg → Cit → Orn → Pro → 5AV. The disappearance of Arg (1 mmol/L) was approximately 52.9 and 88.2% in B, 33.9 and 55.6% in P, and 52.8 and 85.2% in BP during 6 and 12 h incubations, respectively. When expressed in units of ‘per gram (g) of microbial nitrogen (MN)’, the net degradation rate of Arg in BP (50.3 µmol/g MN/h) was approximately 46% lower than that of B during a 12 h incubation period. The presence of protozoa tended to inhibit the production of Orn from Cit and the production of 5AV from Pro which were thought to be rate‐limiting steps of Arg metabolism in rumen microorganisms. As a result, protozoa appeared to have a saving effect on Arg metabolism, that is, protozoa protected Arg from wasteful exhaustion in the rumen.  相似文献

Biosynthesis of arginine (Arg) from citrulline (Cit), ornithine (Orn), proline (Pro), and 5-aminovaleric acid (5AV) by mixed rumen bacteria (B), protozoa (P), and their mixture (BP) was quantitatively investigated in an in vitro system from the standpoint of protein nutrition in ruminants. Rumen microorganisms, collected from ruminally fistulated goats, were anaerobically incubated with or without 1 mmol/L each of substrates at 39°C for 12 h. Arginine and other related compounds, produced in both supernatants and acid-hydrolyzates of microorganisms in B, P, and BP suspensions, were analyzed by high-performance liquid chromatography. Arginine production from Cit in BP, when expressed with a unit of 'μmol/g microbial nitrogen', was approximately 70% and 94% higher than that in B and P, respectively, in a 12-h incubation period. In the case of Orn, the values were approximately 30% and 75%. Both rumen bacteria and protozoa could produce Cit and Orn from Pro, so it is assumed that they can produce Arg from Pro. Rumen protozoa were unable to degrade 5AV and it was the final product in the metabolism of Cit, Orn and Pro in P suspension. A trace amount of Orn and Pro produced from 5AV in B and BP suspensions indicated that the reversible reaction of 5AV formation was performed only by rumen bacteria. This is the first quantitative report on Arg biosynthesis from its precursors by rumen microorganisms.  相似文献

In vitro experiments were conducted to examine the synthesis of arginine (Arg) from argininosuccinic acid (ASA) and citrulline (Cit) by crude enzymes of cattle kidney cortex. Kidney samples, collected from Japanese black cattle, were homogenized in KCl solution (ice‐cold), and centrifuged at 27 000 × g for 20 min at 4°C, and the supernatant fluid was used as a crude enzyme solution. The enzyme solution was incubated at 39°C in Tris HCl buffer with 15 mmol/L ASA or with 10 mmol/L Cit in the presence of 10 mmol/L aspartic acid (Asp), 10 mmol/L ATP and 5 mmol/L MgCl2 to examine the activities of two enzymes, argininosuccinate lyase and argininosuccinate synthetase, which work at the terminal steps of Arg biosynthesis. The production of Arg from ASA, or ASA and Arg from Cit by argininosuccinate lyase and argininosuccinate synthetase activities, respectively, were determined directly by the HPLC method. The optimum pH for argininosuccinate lyase activity was 7.85. Unfortunately, the optimum pH for argininosuccinate synthetase activity could not be determined because no inhibitor of argininosuccinate lyase was used in the Cit incubation, so the ASA produced from Cit spontaneously converted to Arg during incubation with Cit. The maximum production of ASA from Cit was found at pH 6.45 under these conditions. We observed the optimum pH for the synthesis of Arg from Cit at 7.7. The production of Arg from ASA or Cit was quantitatively determined as 14.4 or 8.83 µmol/g kidney tissue/h, respectively, at the optimal pH values. This suggests that the daily production of Arg from ASA or Cit by the kidney might be sufficient to cover the daily requirement of Arg in cattle.  相似文献

We describe the macroscopic anatomy of the intestine of the giraffe (Giraffa camelopardalis). The small intestine was divided into duodenum, jejunum and ileum as usual. The caecum was attached to the ileum by a long ileocaecal fold, and to the proximal ansa of the ascending colon by a caecocolic fold. The ascending colon was the most developed portion of the gross intestine and had the most complex arrangement with three ansae: the proximal ansa, the spiral ansa and the distal ansa. The proximal ansa completely encircled the caecum, describing a 360° gyrus, and represented the widest portion of the intestine. The spiral ansa was formed by three and a half centripetal gyri, a central flexure and three centrifugal gyri. The last centrifugal gyrus left the spiral and described nine flexures of different form and direction over the left side of the mesentery. The two portions that formed each of these flexures ran parallel to each other. The last part of this gyrus ran parallel to the jejunum. When compared with domestic cattle, giraffe had a comparatively short small intestine and a comparatively long large intestine, with a resulting small ratio of small:large intestine. Reasons are presented why this should be considered a peculiarity of cattle‐like ruminants rather than a different representative of a browser–grazer dichotomy in general.  相似文献

Maté, L., Virkel, G., Lifschitz, A., Sallovitz, J., Ballent, M., Lanusse, C. Phase 1 and phase 2 metabolic activities along the small intestine in adult male sheep. J. vet. Pharmacol. Therap. 33 , 537–545. Metabolic activities of several xenobiotic metabolizing enzymes were evaluated in both hepatic and enteric subcellular fractions obtained from Corriedale × Merino crossbreed rams by using a biochemical approach. Microsomes obtained from the different segments of sheep small intestinal mucosa displayed cytochrome P450 (CYP)‐dependent N‐demethylations but not O‐deethylase activities apparently occurred. CYP‐mediated N‐demethylations neither decreased nor increased along the small intestinal mucosa. Percentages of activity for erythromycin N‐demethylase in the small intestine were between 29% (duodenum) and 45% (ileum) from that measured in the liver, whereas those determined for triacetyl‐oleandomycin N‐demethylation ranged between 10% (duodenum) and 15% (jejunum) of the same hepatic activity. Conversely, metabolic rates for aminopyrine and chlorfeniramine N‐demethylations in the gut mucosa ranged between 3% and 7% compared to their respective hepatic enzyme activities. Sheep enteric mucosa also displayed metabolic reactions typically mediated by flavin‐containing monooxygenases (FMOs), carbonyl reductases (CBRs), carboxylesterases (CES), glutathione S‐transferases (GSTs) and uridine diphosphoglucuronyltransferases (UGTs). The FMO‐mediated sulfoxidation of methimazole was 2.6‐fold higher (P < 0.01) in the ileal compared to the duodenal mucosa. Percentages of activity for the microsomal CBR‐dependent biotransformation of menadione were between 12% (ileum) and 19% (duodenum–jejunum) of the total activity measured in the liver; metabolic rates measured in duodenum and jejunum were ~1.7‐fold higher (P < 0.05) than that observed in the ileum. The microsomal CES activity (using p‐nitrophenyl acetate as substrate) was around twofold higher in duodenum (P < 0.05) and jejunum (P < 0.01) in comparison to the ileum. Cytosolic GST‐dependent activities (toward 1‐chloro, 2,4‐dinitrobenzene) were similar in the mucosa of duodenum, jejunum and ileum. Microsomal UGT activities (toward 1‐naphthol) in duodenum and jejunum were three‐ and fourfold higher, respectively, compared to that measured in the ileum. The small intestinal mucosa may play a critical defensive role due to its involvement in the detoxification of toxic compounds prior to absorption. In addition, gut metabolic reactions may contribute to the presystemic metabolism of orally administered drugs. These results are a further contribution to the understanding of the relevance of the extra‐hepatic metabolism of xenobiotics in ruminant species.  相似文献

Samples of the following parts of the gastrointestinal tract were collected from 20 slaughter cattle: - oesophagus, reticulum, omasum, rumen, abomasum, duodenum, jejunum, ileum, colon (beginning and end). There were considerable differences in mineral content, related to the differing functions of each part. Forestomach mucosa contained high concentrations of Ca and Mg, attributable to accumulation and separation of mineral salts. In the three segments of small intestine there was generally more Mg than Ca, with high concentrations of Na and K. The tabulated results are intended to provide a basis for comparison with pathological states.  相似文献

The contractile effects of the tachykinins eledoisin, substance P and neurokinin A and B were investigated in vitro on circular and longitudinal muscle strips from horse duodenum, ileum and colon. Circular smooth muscle of the small intestine was highly responsive, large intestine circular smooth muscle less so, while longitudinal muscle from all gut segments was much less sensitive. pD2 values and intrinsic activities on small intestine circular muscle indicated differences in receptor distribution between the duodenum and ileum: NK3 and a smaller number of NK2 receptors being present in the duodenum, and NK2 receptors predominating in the ileum. Notwithstanding this, eledoisin and neurokinin B were the most active substances on duodenum and ileum, respectively. These findings suggest that tachykinins may play a role in equine gastrointestinal pathophysiology.  相似文献

The presence and distribution of glucose-dependent insulinotropic polypeptide or gastric inhibitory polypeptide (GIP), gastric-releasing peptide (GRP) and glucagon immunoreactivity were studied in the small intestine of the New Hampshire chicken using immunohistochemistry. This is the first report of the presence of GIP-immunoreactive (ir) cells in avian small intestine. GIP, GRP and glucagon immunoreactivity was localized in the epithelium of the villi and crypts of the duodenum, jejunum and ileum. In particular, both in the duodenum and in the jejunum immunoreactive endocrine cells to GIP, GRP and glucagon were observed. In the ileum, we noticed GIP-ir and glucagon-ir cells. GRP-ir was found in nerve fibres of all three segments of the small intestine. The distribution of these bioactive agents in the intestinal tract of the chicken suggests that GIP and glucagon may play a role in the enteropancreatic axis in which intestinal peptides modulate pancreas secretion.  相似文献

本试验旨在研究葛根素对饲喂氧化大豆油饲粮黄羽肉鸡小肠黏膜形态结构、紧密连接蛋白基因表达及抗氧化能力的影响。试验采用2×2因子设计,因子包括油脂质量(新鲜大豆油和氧化大豆油)和葛根素添加水平(0、750 mg·kg-1)。选取健康1日龄雌性黄羽肉鸡360只,随机分成4个处理组,分别为新鲜大豆油饲粮组、新鲜大豆油+葛根素(750 mg·kg-1)饲粮组、氧化大豆油饲粮组和氧化大豆油+葛根素(750 mg·kg-1)饲粮组,每组6个重复,每个重复15羽。在28和56日龄时,每个重复随机选取1只鸡,取十二指肠、空肠、回肠检测小肠形态结构和测定小肠黏膜紧密连接蛋白基因表达及抗氧化指标。结果表明:1)饲喂氧化大豆油显著降低28日龄肉鸡十二指肠的绒毛高度和绒毛高度/隐窝深度(V/C)、回肠V/C和claudin-1 mRNA表达量以及56日龄肉鸡3个肠段的绒毛高度及V/C (P<0.05),显著升高56日龄3个肠段的隐窝深度(P<0.05)。饲粮中添加葛根素显著提高28日龄肉鸡空肠绒毛高度和V/C、回肠闭合小环蛋白1(ZO-1) mRNA表达量和56日龄肉鸡十二指肠V/C、空肠和回肠的绒毛高度及V/C (P<0.05),显著降低3个肠段的隐窝深度(P<0.05)。2)饲喂氧化大豆油显著升高28日龄十二指肠还原型谷胱甘肽(GSH)含量和空肠还原型谷胱甘肽过氧化物酶(GSH-Px)活性(P<0.05),并显著降低28日龄回肠GSH含量、超氧化物歧化酶(SOD)活力和总抗氧化能力(T-AOC)(P<0.05)。饲粮中添加葛根素显著提高28日龄肉鸡十二指肠GSH-Px活性(P<0.05)。饲喂氧化大豆油显著降低56日龄回肠SOD活性和T-AOC (P<0.05),饲粮中添加葛根素显著提高56日龄肉鸡十二指肠SOD和回肠SOD、GSH-Px活性(P<0.05),显著降低56日龄肉鸡空肠GSH含量、SOD活性、T-AOC和回肠T-AOC (P<0.05)。综上所述,饲喂氧化大豆油破坏肠道黏膜形态结构、降低紧密连接蛋白基因的表达量和抗氧化能力,添加葛根素可提高肉鸡小肠黏膜紧密连接蛋白基因的表达量,改善氧化损伤条件下肠道黏膜形态结构,提高抗氧化酶的活性而提高其抗氧化能力。  相似文献

选择3头条件一致的荷斯坦奶牛,安装永久性的瘤胃瘘管、十二指肠瘘管和回肠瘘管,用Co-ETDA做标记物。采用3×3拉丁方设计研究了不同维生素A添加水平对奶牛瘤胃、十二指肠、回肠和血液中维生素A浓度的影响,以及维生素A在小肠中的吸收情况,研究结果表明:瘤胃、十二指肠、回肠和血液中维生素A浓度随着维生素A剂量增加而极显著上升(P〈0.01),其中回肠末端维生素A浓度上升的幅度随着添加剂量的增加而加大,其余三者中维生素A浓度上升的幅度随着添加剂量的增加而逐步降低。小肠中维生素A的吸收量随着维生素A剂量的增加而极显著上升(P〈0.01),但上升的幅度随着添加剂量的增加而逐步降低。  相似文献

本文以乙酸浸提法获得的猪小肠抗菌肽粗提物为对象,通过纸片扩散法研究其在肠道内的分布、抑菌活性及其与添加水平之间的量效关系。结果表明:空肠抗菌肽的含量高于十二指肠与回肠。来源于十二指肠和回肠的抗菌肽对大肠杆菌的杀灭效果均好于沙门氏菌;在空肠则未见明显差异。说明猪小肠抗菌肽的抑菌效果随添加水平减少而不断下降,呈明显的剂量-效应关系。  相似文献

Development of mechanistic models of digestion is a means to account for interactions between different nutrients, and between nutrients and the animal. Although some digestion models for pigs have been proposed, quantitative information concerning the underlying processes is extremely limiting. The aim of this study was to determine the effect of dietary fibre on the transit of the solid and liquid phases of digesta in different segments of the gastro-intestinal tract (GIT) in growing pigs. Six barrows (initial BW 30 kg and fitted with two simple T-cannulas at the proximal duodenum and distal ileum) were used in a double 3 × 3 Latin square design. Pigs were offered diets differing in total dietary fibre content (17, 22 and 27%) at 4 h intervals. A single meal marked with YbO2 and Cr-EDTA was used to determine the passage kinetics of respectively the solid and liquid phases of digesta in different segments of the GIT.

The mean retention time (MRT) averaged 1 h from the mouth to the proximal duodenum (stomach), 4 h from the proximal duodenum to the distal ileum (small intestine), and 35 h from the distal ileum to the faecal excretion (large intestine). Increase in dietary fibre content had no effect on MRT in the stomach, tended to decrease MRT of solid phase in the small intestine (P = 0.07), and decreased MRT of both phases in the large intestine (43 to 29 h, P = 0.06). For all diets, transit of the liquid phase in the large intestine was 4–8 h less than transit of the solid phase.  相似文献

Background

The provision of feed is a major cost in beef production. Therefore, the improvement of feed efficiency is warranted. The direct assessment of feed efficiency has limitations and alternatives are needed. Small intestine micro-architecture is associated with function and may be related to feed efficiency. The objective was to verify the potential histomorphological differences in the small intestine of animals with divergent feed efficiency.

Methods

From a population of 45 feedlot steers, 12 were selected with low-RFI (superior feed efficiency) and 12 with high-RFI (inferior feed efficiency) at the end of the finishing period. The animals were processed at 13.79 ± 1.21 months of age. Within 1.5 h of slaughter the gastrointestinal tract was collected and segments from duodenum and ileum were harvested. Tissue fragments were processed, sectioned and stained with hematoxylin and eosin. Photomicroscopy images were taken under 1000x magnification. For each animal 100 intestinal crypts were imaged, in a cross section view, from each of the two intestinal segments. Images were analyzed using the software ImageJ®. The measurements taken were: crypt area, crypt perimeter, crypt lumen area, nuclei number and the cell size was indirectly calculated. Data were analyzed using general linear model and correlation procedures of SAS®.

Results

Efficient beef steers (low-RFI) have a greater cellularity (indicated by nuclei number) in the small intestinal crypts, both in duodenum and ileum, than less efficient beef steers (high-RFI) (P < 0.05). The mean values for the nuclei number of the low-RFI and high-RFI groups were 33.16 and 30.30 in the duodenum and 37.21 and 33.65 in the ileum, respectively. The average size of the cells did not differ between feed efficiency groups in both segments (P ≥ 0.10). A trend was observed (P ≤ 0.10) for greater crypt area and crypt perimeter in the ileum for cattle with improved feed efficiency.

Conclusion

Improved feed efficiency is associated with greater cellularity and no differences on average cell size in the crypts of the small intestine in the bovine. These observations are likely to lead to an increase in the energy demand by the small intestine regardless of the more desirable feed efficiency.  相似文献

The dietary contents of crude protein and free amino acids (AA) may affect the protein digestion and AA absorption in pigs. Trypsin and chymotrypsin activities, AA serum concentrations and expression of AA transporters in the small intestine of pigs fed a low protein, AA‐supplemented (19.2%, LPAA) or a high protein (28.1%, HP), wheat‐soybean meal diet were measured in two 14‐d trials. The LPAA diet contained free L‐Lys, L‐Thr, DL‐Met, L‐Leu, L‐Ile, L‐Val, L‐His, L‐Trp and L‐Phe. All pigs were fed the same amount of feed (890 and 800 g/d for trial 1 and 2 respectively). In trial 1, samples of mucosa (duodenum, jejunum and ileum) and digesta (duodenum and jejunum) were collected from 14 pigs (17.2 ± 0.4 kg); in trial 2, blood samples were collected from 12 pigs (12.7 ± 0.3 kg). The trypsin and chymotrypsin activities in both intestinal segments were higher in pigs fed the HP diet (p < 0.01). Trypsin activity was higher in jejunum than in duodenum regardless the dietary treatment (p < 0.05). Pigs fed the LPAA diet expressed more b0,+AT in duodenum, B0AT1 in ileum (p < 0.05), and tended to express more y+LAT1 in duodenum (p = 0.10). In pigs fed the LPAA diet, the expression of b0,+AT was higher in duodenum than in jejunum and ileum (p < 0.01), but no difference was observed in pigs fed the HP diet. Ileum had the lowest b0,+AT expression regardless the diet. The serum concentrations of Lys, Thr and Met were higher in LPAA pigs while serum Arg was higher in HP pigs (p < 0.05). Serum concentrations of AA appear to reflect the AA absorption. In conclusion, these data indicate that the dietary protein contents affect the extent of protein digestion and that supplemental free AA may influence the intestinal site of AA release and absorption, which may impact their availability for growth of young pigs.  相似文献

The intestinal morphology of 7-week-old pigs was investigated by light (LM) and scanning electron microscopy (SEM). The piglets were fed either a semisynthetic or a cereal-based diet. The shapes of the intestinal villi and crypts of the duodenum, jejunum and ileum were examined. The villi were predominantly tongue-shaped. In the duodenum they were also ridged, branched and folded, and in the jejunum they were also leaf-like and ridged. At places with lymph follicles, the surface of the ileum was rugged with meandering fold-like villi. The crypts of the three segments of the small intestine were mainly coiled and sometimes branched. A novel morphometric evaluation method was introduced using the enlargement factors of each villus and crypt surface. The enlargement factor for the villus surface of the duodenum, jejunum and ileum was 3.13, 3.72 and 2.71, respectively. The factor for the crypt surface of the duodenum, jejunum and ileum was 9.07, 8.94 and 6.53, respectively. Furthermore, the relative proliferation rate and the epithelial renewal index were calculated for the first time. The relative proliferation rate of the duodenum, jejunum and ileum was 32.88, 34.78 and 50.77 proliferations per mm crypt perimeter, respectively. The diets consumed had an influence on the epithelial renewal index being higher for piglets fed the cereal-based diets.  相似文献

The effect of enteritis on the development of the small intestine was examined in newborn, colostrum-deprived piglets infected with a human isolate of Y. enterocolitica (serotype 0:3, biotype 4) soon after birth. The piglets were killed 3 days (n = 6) or 5 days (n = 8) after infection, or antibiotic therapy was commenced on day 5 and the animals killed on day 14 (n = 5). Compared with the non-infected controls, infected animals had reduced mucosal lactase and sucrase, but not maltase activity, while after antibiotic therapy, previously infected piglets had a lower lactase and a higher maltase and sucrase activity. Lactase activity was significantly reduced in the duodenum and jejunum, and mean values were lower in the ileum, but the difference did not reach significance; maltase activity was greater at all ages from the distal jejunum to the mid-ileum; sucrase activity was reduced in all segments up to day 5 but after antibiotic therapy was increased in the jejunum and appeared early in the ileum. Enzyme profiles were more mature along the crypt-villus axis in some segments of the intestine in previously infected piglets. Sodium-potassium-ATPase activity was unchanged. There was a reduced villus height:crypt depth ratio, crypt hyperplasia and increased crypt cell proliferation. Morphological maturation, indicated by loss of vacuoles and location of the nucleus at the base of the enterocyte, proceeded distally from the duodenum to ileum from 3 to 14 days of age when only the ileum remained immature. In infected piglets, there was reduced vacuolation and earlier location of the nucleus at the base of the cell in the distal intestine. Accelerated maturity of specific disaccharidases and enterocyte morphology in infected piglets appears to be due to physical damage to the mucosa resulting in faster proliferation of crypt cells and migration of enterocytes. It is suggested that this may reduce macromolecular internalisation and impair the ability to utilise dietary carbohydrate and may have long-term effects on growth and immunological responses of the gut.  相似文献

In order to investigate the effects of cecropin and synbiotics on the intestinal mucosal morphology and intestinal mucosal immune cells of AA broilers. 480 of 1-day-old healthy AA broilers were chosen and randomly divided into 4 groups with 4 replicates per group and 30 broilers per replicate. The broilers in group Ⅰ(control group) were fed basal diet,while that in groups Ⅱ to Ⅳ were fed the basal diet supplemented with 0.5% cecropin,0.3% synbiotics,0.5% cecropin+0.3% synbiotics,respectively. The trial lasted for 42 days. The result showed that compared with group Ⅰ,the villus height of duodenum and ileum in group Ⅳ were extremely significantly increased (P<0.01).The crypt depth of jejunum and ileum in group Ⅱ,and that of small intestine in group Ⅲ were significantly or extremely significantly decreased (P<0.05;P<0.01).The villus height/crypt depth of small intestine in groups Ⅱ and Ⅲ,and that of duodenum and ileum in group Ⅳ were significantly or extremely significantly increased (P<0.05;P<0.01).The villus width of jejunum in group Ⅱ was significantly decreased (P<0.05),while the villus width of duodenum and jejunum in group Ⅲ,that of jejunum and ileum in group Ⅳ were extremely significantly decreased (P<0.01).The mucous thickness of jejunum in groups Ⅱ and Ⅲ were significantly increased (P<0.05),while that of small intestine in group Ⅳ was extremely significantly increased (P<0.01).The number of the intraepithelial lymphocyte of ileum in group Ⅱ was significantly increased (P<0.05),and that of duodenum in group Ⅲ and small intestine in group Ⅳ were extremely significantly increased (P<0.01).The number of the goblet cell of duodenum in group Ⅲ,duodenum and ileum in group Ⅳ were extremely significantly increased (P<0.01).The mast cell of jejunum in group Ⅳ was significantly increased (P<0.05).In conclusion,both the cecropin and synbiotics could improve the small intestine mucosal structure and promote the small intestine mucosal immunocompetent cells proliferation in AA broiler, and the effect of combined utilization would be better.  相似文献

1. The aim of this study was to investigate the localisation and expression of the epithelial Ca2+ channel TRPV6 (transient receptor potential vanilloid channel type 6) in different intestinal segments and kidney of laying hens during peak lay. 2. Immunohistochemical analysis of the intestine indicated that TRPV6 was localised to the brush-border membranes of the duodenum, jejunum, ileum, caecum, and rectum. Expression was weaker in the rectum, and little or no expression was found in crypt and goblet cells. In addition, TRPV6 mRNA was quantified amongst different intestinal segments, and expression was highest in the duodenum and jejunum. Furthermore, Western blotting indicated that the duodenum expressed the greatest amount of TRPV6 and the rectum the least with the other segments expressing intermediate levels. 3. In the kidney, distinct immunopositive staining for TRPV6 was detected at the apical domain of the distal convoluted tubules (DCT) and medullary connecting tubules (CNT). Interestingly, distribution of TRPV6 extended to the proximal convoluted tubules (PCT). Furthermore, the kidney expressed lower TRPV6 mRNA and protein levels compared with that in the duodenum. 4. In conclusion, the epithelial Ca2+ channel TRPV6 is strongly expressed in the apical cells of the entire intestine and the renal tubules, suggesting that active Ca2+ transcellular transport plays a crucial role in dietary calcium (re)absorption in laying hens.  相似文献
