Antimicrobial peptides (AMPs) are a diverse group of small polypeptides that can inhibit the growth of microbes; the investigation of their cytotoxicity is critical before they can be considered for use. The purpose of the present study was to examine the cytotoxicity of apidaecin at different concentrations (0.0, 10.0, 20.0, and 30.0 μg/ml) on tilapia, Oreochromis niloticus, primary intestinal epithelial cells in vitro through cells morphology observation, methylthiazolyldiphenyl-tetrazolium bromide (MTT) conversion assay, and cellular viability assay. The lactate dehydrogenase (LDH) activity, cytosolic free Ca2+ concentration, and phospholipase A2 (PLA2) activity of primary cells treated with/without apidaecin were also determined. No significant differences (P > 0.05) in MTT assay and viability were found between the treated and the control cells. LDH activities were slightly increased when treated with apidaecin for 24 h compared with the control (19.97 ± 1.24 U/l), but not significantly (P > 0.05). When treated with 20.0 μg/ml apidaecin for 24 h, the concentrations of cytosolic free Ca2+ was the highest among all the treatments (133.65 ± 9.65 nmol/l), but not the difference was not significant when compared with the control (P > 0.05). Similar results were found for the activity of cytosolic PLA2. The current results indicate that apidaecin did not show significant cytotoxicity to primary epithelial cells within 30.0 μg/ml and that it can be used as a new candidate for infection control in fish culture.  相似文献

Abstract –  Feeding tactics, body condition and size structure of two populations of pumpkinseed Lepomis gibbosus from Cabañeros National Park (Guadiana River basin, central Spain) were compared to provide insight into the ecological requirements favouring levels of success/failure in relation to human intervention. Habitat, benthic macroinvertebrates and pumpkinseed were quantified in Bullaque (regulated flow, affected by agricultural activities) and Estena (natural conditions) rivers, from May to September of 2005 and 2006. Significant differences were found in the limnological characteristics between the two rivers. Spatial and temporal variations in diet composition were likely related to opportunistic feeding and high foraging plasticity. Diet diversity was higher in Bullaque River. Electivity of benthic prey showed variation between sized individuals and populations. The body condition of pumpkinseed was better in Bullaque River. Current results show that this invasive species is favoured by human intervention in Bullaque River.  相似文献

The duration of efficacy of emamectin benzoate in the control of Caligus curtus infestations in Atlantic cod, Gadus morhua L., was studied following an administration of 50 μg kg?1 for seven consecutive days. No lice were found on medicated fish when challenged 1 week (challenge 1) or 5 weeks (challenge 2) following termination of medication, whereas the mean abundance of lice among the unmedicated fish was 17.9 and 19.3 lice per fish in challenge 1 and 2, respectively. Muscle concentrations of 19.5 ± 8.2 ng g?1 and 3.4 ± 0.9 ng g?1, respectively, and skin concentrations of 23.1 ± 10.8 and 4.2 ± 1.0 ng g?1, respectively, were found 27 and 55 days following the termination of medication. Tissue concentrations and the duration of efficacy indicate a dosing regime for emamectin in cod, similar to the regime used for Atlantic salmon, Salmo salar L.  相似文献

This study has evaluated the effects of different concentrations of the water and ethanolic extracts of Iranian propolis (WEP and EEP) on the microbiological and sensory parameters of minced common carp meat that was stored at 4°C for 9 days. The chemical compounds of the extracts were identified by gas chromatography–mass spectroscopy (GC-MS) analysis. The results revealed that WEP and EEP extracts were efficient against the spoilage microorganisms, including total plate counts (TPCs), psychrotrophic populations (PTCs), lactic acid bacteria (LAB), and even Staphylococcus aureus. There was a significant difference in the bacteria growth between samples treated with WEP or EEP and the control sample during storage. Inhibitory effects were, respectively, 7% EEP > 5% EEP > 3% EEP > 7% WEP > 5% WEP > 3% WEP. In all treatments, PTC counts were higher than 7 log CFU/g after 6 days as well as TPC, LAB, and S. aureus populations after 9 days. Overall, the results show that EEP and WEP have good antimicrobial activity due to high phenolic and flavonoid compounds, including caffeic acid, tectochrysin, aceacetin, chrysin, pinostrobin, pinobanksin-3-acetate, galangin-3-methyl ester, and pinocembrin chalcone. Sensory evaluation shows very slight improvement in sensory properties (color, odor, texture, and overall acceptability) of the treated samples with the different extracts of Iranian propolis.  相似文献

Juvenile stellate sturgeon Acipenser stellatus were intraperitoneally injected with estradiol-17β (E2; 0 and 5 mg/kg fish) to investigate the possibility of sex reversal and also determine the changes in biochemical parameters. Five-month-old fish (40.9 ± 1.1 g) were injected every 3-week interval during a 190-day trial. At the termination of the experiment, final weight and other growth parameters including weight gain and specific growth rate, hepatosomatic and viscerosomatic indices were not affected by repetitive injection of E2. Hematological features of E2-treated fish showed significant reductions in number of red blood cells, hemoglobin concentration, hematocrit value and mean corpuscular hemoglobin (P < 0.05), but no significant changes were observed in number of white blood cells, mean corpuscular volume and mean corpuscular hemoglobin concentration (P > 0.05). Calcium, phosphorus, glucose, triacylglycerol, cholesterol, total protein and estradiol concentrations were significantly increased in fish injected with E2 (P < 0.001). Plasma progesterone and testosterone levels were noticeably lower in fish injected with 5 mg/kg E2 rather than the control fish (P < 0.001). Histological observations of gonads showed that all fish injected with 5 mg/kg E2 apparently feminized, while 66.6 % of the control group was female. These results revealed that the injection of E2 is an effective method for feminization of stellate sturgeon without having significant inhibitory effects on growth and survival.  相似文献

The commercial value of yellowtail Seriola quinqueradiata meat is often reduced by color deterioration of the dark muscle during storage. The objective of this study was to investigate the effect of simultaneous dietary supplementation of vitamin C (VC) and vitamin E (VE) to cultured yellowtail on color degradation of the dark muscle during storage. Yellowtail were fed for 6 or 11 days on a diet containing 1% VC and 1% VE (vitamin group) or without these vitamins (control group). The amounts of VC and VE in the dark muscle of the vitamin group tended to be higher than those of the control group. During the storage of sliced meat at 23 and 4°C, the decreases in a* value (redness) of the dark muscle of the vitamin group was slower than those of the control group. The formation of metmyoglobin, which can occur along with myoglobin oxidation, in the dark muscle of the vitamin group was slower than that of the control group. These results suggest that simultaneous feeding of high amounts of VC and VE for a short period to yellowtail before harvesting could suppress the color deterioration of the dark muscle during storage.  相似文献

The distribution of Anisakis larvae in fresh caught hake was studied, and the influence of three infection levels of the viscera (low, medium, high) on quality changes of deep frozen muscle was followed over 220 days. Differences in quality were attributed more to the size of the individuals than to the infection degree of the three groups. The removal of belly flaps diminished the risk of consumers’ infection and sensitization as prevalence and intensity was reduced to 75.6 and 5.9%, respectively. Allergens were detected after 220 days frozen storage, which represents a risk for consumers already allergic to Anisakis.  相似文献

Fishponds exhibit high natural retention potential for phosphorus, which enters the ponds from non-point, diffuse and point sources, as well as from aquaculture management. Results of phosphorus mass balance monitoring of nine large fishponds (60–449 ha) over 2010–2014 revealed total phosphorus retention ranging between ?66 % (sink) and +52 % (release) of the P loads from inflows, i.e. specific P retention varied between ?7.83 g m?2 (sink) and 1.00 g m?2 (release) of surface area, per one fish production cycle. Retention of P was eliminated by application of fertilizers (with simultaneous fish feed application) during fish production process and/or by massive P release from fishpond sediments after previous heavy loads. P retention could be increased by preference of surface water outflow instead of bottom discharge and/or also by minimizing of emissions of suspended sediment particles during a fish harvesting. The real role of fishponds in transport processes of P throughout a watershed could be eruditely assessed only by following newly proposed method called “new approach” when P input (via inflows) and P output (via outflows) are compared with natural fishpond ability to retain P, which is determined by a simple model proposed by Hejzlar et al. (2006). For evaluation of effect of actual fishery management, the “culture-balance” method is suitable too. If the P inputs (stocked fish, fertilizing, feeding) and outputs (harvested fish) derived from fishery practice were in balance, the P retention did not decline. However, in condition of inputs > outputs, the P retention appreciably ceased or was totally eliminated. It was concluded that fishponds could serve as an important sink of P transported throughout a watershed even under semi-intensive fish (common carp) production condition.  相似文献

Genome size (C value) is an important index for phylogenetic studies. Haliotidae (abalones) includes many species widely distributed throughout the world??s oceans, which makes this family interesting for phylogenetic studies. To examine Haliotidae phylogeny, we determined the C value and adenine and thymine base pair content (AT?%) of Haliotis discus hannai and H.?diversicolor aquatilis by using flow cytometry. The C values of H.?discus hannai and H.?diversicolor aquatilis were 1.84 and 1.45?pg, with AT?% of 62.3 and 66.3?%, respectively. These data represent the first report of abalones classified as Pacific Northwest (H.?discus hannai) and Indo-Pacific (H.?diversicolor aquatilis) groups, and provides new validation for previous theories related to Haliotidae phylogeny.  相似文献

Fish stocking is an important element of aquaculture management. A constant stocking rate is a convenient strategy in intensive aquaculture, being also the best under steady physical and economic conditions. If, however, these conditions vary with time, the best stocking rate may also vary. This problem has been addressed in a previous paper, where water temperature, fish price and/or market demand were allowed to vary sinusoidally over the annual cycle, while a constant upper limit on the water-treatment capacity constrained the process. Considering only situations where feeding is always to satiation, sinusoidal stocking rates were shown to perform often significantly better than the best constant stocking-rate strategy.In this paper a further step is taken in that over-stocking (more than hitherto) for certain periods of time is allowed. This leads to restricted feeding at a later time (to avoid overload of the water-treatment system) and to some loss of feeding efficiency. Often, however, the loss in feeding efficiency is more than compensated for by better utilization of the rearing space.Optimal constant and sinusoidal over-stocking strategies were applied to eight sample combinations of sinusoidal temperature and market conditions. Restricted over-stocking often resulted in a considerable improvement of economic performance over the previous results. The potential for increase of yield and profit by over-stocking depends on the amount of under-utilization of the rearing space by the previous solution, namely when only feeding to satiation is allowed. On average (for the system under consideration) out of 10% of under-utilized space when feeding to satiation, 4.4% (almost half) can be recovered by properly applied over-stocking. Expressed in terms of feed processing capability, this 4.4% recovery is equivalent to about 16 kg[feed]/(m3[tank]y), and to an additional production of about 6.6 kg[fish]/(m3[tank]y). The gain in profit is of the order of 25 $/(m3[tank]y) per 10% of previously unutilized capacity.  相似文献
