Commercial selenium pellets, manufactured after CSIRO workers drew attention to the significance of grain size on the rate of release of selenium, were tested in 27 sheep grazing a low-selenium New Zealand pasture. The pellets were shown by microscopy to contain mainly 10-20 pm particles of selenium, often agglomerated into larger lumps. There was a considerable variation in the length of time pellets maintained blood selenium levels above the deficiency level of 250 nmol/e (20 microg/l). Whereas four animals given pellets had blood levels below 250 nmol/l after only 343 days, two animals had levels of 375 and 400 nmol/l after 651 days when the level in control sheep was 125 +/- 32 nmol/l. The pellets were recovered from all but one animal and had varying degrees of surface coating which was assumed to be mainly calcium phosphate. Two pellets recovered from sheep at 386 and 484 days, when blood selenium levels were 175 and 1813 nmol/l respectively, were sectioned and examined by light and electron microscopy. Both pellets still contained unreacted selenium but differed in the degree of surface coating. The pellet recovered at 386 days had a solid and continuous coating whereas the coating on the pellet recovered at 484 days was not continuous and consisted of an open lattice of interlocking needles. It appears that it is the extent of this coating which limits the effective life of the pellet in sheep.  相似文献

The efficacy and retention of prototype and commercial (Cosecure) soluble-glass boluses containing selenium, cobalt and copper and having a similar size, density and composition were evaluated in separate 12-month trials with sheep grazing low selenium pastures but with adequate cobalt and copper levels. In both trials, sheep confirmed by X-ray as containing a bolus grew at a significantly greater rate than control sheep. This was attributed to the correction of a selenium deficiency. Although liver copper, and liver and serum vitamin B12 levels were significantly greater in treated sheep than controls on some occasions, the differences were not as great or consistent as with blood selenium levels. The rate of bolus loss was high and in both trials, approximately 40% of the sheep had lost the bolus after 6 months. No prototype boluses were present after 12 months. Intact Cosecure boluses were recovered from 3 sheep out of 30 after 12 months whilst one animal contained a part bolus. The recovered boluses were approximately 45% lighter than when originally administered. Bolus loss did not appear to be due to complete dissolution. In sheep which had lost the bolus, blood selenium levels fell with a half-life of 43 +/- 10 days.  相似文献

Weaned merino lambs, grazing pastures low in selenium, were used to investigate the effect of selenium status on immunity to trichostrongylids. Six weeks following selenium supplementation to 14 of the 27 sheep using intraruminal selenium pellets, 5000 Ostertagia circumcincta and 5000 Trichostrongylus colubriformis larvae were administered orally to all sheep. At four weeks after infection, the mean total worm burden in the selenium supplemented sheep (5537 +/- 343, n = 14) was not significantly different (P greater than 0.05) from that in the unsupplemented sheep (5614 +/- 374, n = 12) and faecal worm egg concentrations were also similar in the two treatment groups. At this time, mean red cell glutathione peroxidase activities in the supplemented and unsupplemented groups were 430 and 11 U g-1 haemoglobin, respectively, and clinical white muscle disease had been observed in the latter group. These results suggest that increasing selenium status of selenium deficient sheep by the use of intraruminal selenium supplementation, has a negligible effect on resistance to an artificial challenge infection of O circumcincta and T colubriformis.  相似文献

The strategic use of single therapeutic doses of closantel, tetramisole or sustained low-level administration of albendazole in feed pellets in controlling naturally acquired parasitic gastroenteritis in sheep was investigated on a farm in semi-arid Rajasthan, India. A total of 303 5- to 6-month-old sheep were divided into three groups. Two groups were dosed with single therapeutic doses of closantel and tetramisole and the third group was given a low-level medication with albendazole through feed pellets for 30 days. Faecal egg counts revealed significantly lower counts (p<0.001) in the group treated with closantel compared to the other two groups. The faecal egg counts in the group receiving sustained low-level albendazole rose after withdrawal of the medication but remained significantly lower than those in the group treated with tetramisole up to 7 weeks after treatment (p<0.05). On the other hand, in the group treated with tetramisole, the mean faecal egg count rose from 3 weeks after treatment and remained continuously higher than those in any other group up to 12 weeks after treatment. The closantel-treated group gained more body weight but the first six-monthly greasy fleece yield was greater in the group treated with medicated pellets. During the first 3 months of the experiment, three animals in the group treated with tetramisole died of parasitic gastroenteritis. Following sustained low-level administration of albendazole in feed pellets, the plasma disposition curve of both the sulphoxide and sulphone metabolites reached its plateau level by day 5 and remained almost constant thereafter. The comparative cost-effectiveness of the three treatment regimes during the first 3 months of treatment was best for the group treated with closantel followed by the group treated with medicated feed pellets.  相似文献

The investigation comprised 12 herds, totalling 660 sheep, in the counties of Stockholm and Uppsala. The selenium content of specimens of forage, whole blood, muscle, liver, and kidney was determined by a fluorimetric method. Data on the number of born and dead lambs were collected 4—5 months after lambing. Dead and sick lambs were subjected to necropsy and to clinico-chemical analyses, respectively.The selenium content of hay and corn was lower than 60 ng per g, which some authors regard as the minimum level of Se requirement for protection against white muscle disease in sheep. The analyses of forage mixtures containing oil concentrates and beet pulp showed selenium levels that were about 2—5 times as high as this minimum level.In herds in which the animals had not been treated with selenium preparations prior to sampling, there was a significant positive correlation between the selenium content of the forage and that of whole blood. A relationship was also noted between the selenium content of forage and of organs.In 9 out of the 12 herds the selenium concentrations in whole blood were on the level that is considered to be representative of white muscle disease.A high mortality rate was recorded for lambs whose mothers had low selenium levels in their blood.Prophylactic and curative treatment with selenium plus vitamin E orally and/or parenterally probably reduced lamb mortality rate in the herds fed a low-selenium diet.  相似文献

In each of two dairy herds (A and B), rising yearling heifers (Trial 1) and adult cows (Trial 2) were assigned to three treatment groups. Untreated animals were compared to animals treated with either two or four intra-ruminal pellets containing 3 g of elemental selenium. The administration of pellets at the recommended dose (two pellets per animal) was effective in elevating whole blood glutathione peroxidase activity and selenium concentration to over 10 times those of control animals. In Trial 1, a 15% response in liveweight gain (p<0.001) occurred in yearling heifers in the herd with the lowest pre-treatment selenium status. In Trial 2, cows receiving two pellets produced a greater milk volume (p=0.06) and more milk solids (p=0.02) than untreated controls; an increase in volume of 5.4% and 8%, and in milk solids of 6.5% and 6.4%, were noted in herds A and B respectively. There was a trend towards decreasing somatic cell counts in milk from the treated cows when compared to controls, the four-pellet group in Herd A and the two-pellet group in Herd B being significantly different from their respective control group. No between-group differences were noted in calving-first service or calving-conception intervals, nor in the proportion of animals pregnant to first or all services. The administration of selenium at twice the recommended dose rate yielded no additional response above that noted after the administration of the recommended dose. The results of this study support the use of currently recommended Ministry of Agriculture and Fisheries selenium reference ranges in cattle for the prediction of a response to supplementation.  相似文献

Barium selenate was developed as a long-acting subcutaneous injection for selenium treatment. The results from trials using this product show that it is a safe preparation for treating and preventing selenium deficiency in sheep. It produces a satisfactory selenium status which will be maintained for at least six months and, when injected into pregnant ewes, results in the transfer of selenium from the ewe to its progeny.  相似文献

OBJECTIVE: To evaluate the respiratory excretion and elimination kinetics of organic and inorganic selenium after oral administration in sheep. ANIMALS: 38 crossbred sheep. PROCEDURES: Selenium was administered PO to sheep as a single dose of 0, 1, 2, 3, or 4 mg/kg as sodium selenite or selenomethionine. Expired air was collected and analyzed from all sheep at 4, 8, and 16 hours after administration. RESULTS: Clinical signs consistent with selenium intoxication were seen in treatment groups given sodium selenite but not in treatment groups given the equivalent amount of selenium as selenomethionine. However, a distinct garlic-like odor was evident in the breath of all sheep receiving 2 to 4 mg of selenium/kg. The intensity of odor in the breath did not correlate with clinical signs in affected animals receiving sodium selenite treatment. CONCLUSIONS AND CLINICAL RELEVANCE: The concentration of selenium in expired air was greater in sheep receiving selenium as selenomethionine than sodium selenite. The concentration of selenium in expired air from sheep receiving high doses of selenium (3 and 4 mg of selenium/kg) was larger and selenium was expired for a longer duration than the concentration of selenium in expired air from sheep receiving low doses of selenium (1 and 2 mg of selenium/kg).  相似文献

Slow-release devices composed of elemental selenium and iron or a soluble-glass containing selenium, cobalt and copper were effective in significantly raising blood and tissue selenium levels of sheep at pasture. Three out of six animals given a soluble-glass bolus lost them over the four months of the trial. In contrast, all six animals retained their iron-selenium pellets. Selenium levels in the kidney cortex four months after administration of the devices, when peak levels were most likely to occur, were approximately 12.7 micromol/kg (1mg/kg). This value is 50% of the maximum permitted level in edible tissue in New Zealand. There was no obvious pattern of distribution of selenium throughout the liver. Variations in concentration from one site to another were, in some cases, almost two-fold. Therefore the analysis of liver biopsy samples gives only an approximate assessment of the mean hepatic concentration.  相似文献

Three flocks of sheep in typical selenium-deficient areas were injected with 2 ml Deposel® (100 mg selenium as barium selenate) in the autumn of 1982 and/or 1983. The selenium status in 10 ewes from each flock, and their offspring, was monitored by selenium analyses of blood samples. The injection induced an adequate increase in blood selenium within 8 weeks. Adequate selenium levels were maintained throughout the lambing season and the following summer. The selenium status in lambs from treated ewes was adequate until they were put out to pasture. Elevated selenium levels were maintained even throughout the next lambing season, and the second injection gave significantly higher blood selenium as compared to the first. The present preparation seems to be suitable and safe for protecting sheep against selenium deficiency. The dosage used seems sufficient to maintain an adequate selenium status for as long as 2 consecutive lambing seasons.  相似文献

SUMMARY Outbreaks of ovine weaner nutritional myopathy occurring in the southwest of Western Australia outside the recognised selenium-responsive areas have been investigated. One hundred and five sheep died in 3 outbreaks following 5 months grazing cereal stubble paddocks. Liver selenium and glutathione peroxidase activities, measured as an index of biological availability of dietary selenium, indicated that the selenium status of the animals was probably adequate. A comparison of liver selenium and α-tocopherol concentrations and glutathione peroxidase activity was made with both selenium supplemented sheep and sheep affected with selenium-responsive nutritional myopathy. In all cases investigated in this study liver α-tocopherol levels in affected animals were low and this may be important in the aetiology of this myopathy.  相似文献

Objective To compare the effectiveness of combined selenium and α-tocopherol acetate treatments in preventing lupinosis-associated myopathy in sheep.
Design Measurement of plasma muscle and liver enzymes, and histopathological examination of muscle and liver in the treatment groups.
Procedure The treatments were: subcutaneous injections of selenomethionine and vitamin E (sc(SeM+E)) , an intraruminal selenium pellet and oral doses of vitamin E and intramuscular injections of selenomethionine combined with either oral doses of vitamin E (imSeM+orE) or intramuscular injections of vitamin E in an oily carrier. Another group received no supplements, while a control group was given selenium pellets on day 0 and fortnightly oral doses of vitamin E from day 0 to 72. To produce lupinosis-associated myopathy, the sheep were fed a diet low in vitamin E and given repeated injections of a crude extract of Diaphorthe toxica. Groups sc(SeM+E) and imSeM+orE were stressed by dosing with protected polyunsaturated fatty acids from day 56 onwards.
Results Lupinosis-associated myopathy was induced in all unsupplemented sheep. In these sheep the storage of Se increased and that of vitamin E decreased. The subcutaneous treatment was highly effective in preventing lupinosis-associated myopathy and also produced the highest vitamin E concentrations in plasma and liver. Supplemental vitamin E was more efficacious than supplemental Se. Concentrations of vitamin E in the livers of sheep given intramuscular vitamin E were higher than expected based on plasma concentrations. Oral doses of vitamin E proved the least effective method of increasing concentrations in liver. Lupinosis did not affect Se concentrations in liver or muscle.
Conclusion The sc(SeM+E) treatment is highly effective in preventing lupinosis-associated myopathy but needs to be further assessed when selenium and vitamin E are both limiting in the diet.  相似文献

In a study to evaluate the efficacy of clorsulon against Fascioloides magna infection in sheep, 12 ewes were inoculated orally with 100 metacercariae of F magna, and 6 were treated with clorsulon (15 mg/kg of body weight) 8 weeks after inoculation. The sheep were euthanatized 16 weeks after inoculation, flukes were recovered, and the liver and other tissues were subjectively scored for the severity of lesions (0 to 4+). The number of flukes recovered from the clorsulon-treated group (3.8 +/- 1.2 flukes) was significantly (P = 0.025) lower than the number of flukes recovered from the group of untreated controls (10.0 +/- 6.6 flukes). The severity of lesions was significantly (P = 0.004) reduced (45.9%) in the treated group (2.0 +/- 1.1), compared with that in the untreated controls (3.7 +/- 0.5). In the untreated group, 3 sheep died and 1 became moribund 14 to 16 weeks after inoculation. The data suggested that a single treatment with clorsulon at a dosage of 15 mg/kg 8 weeks after inoculation was not effective in preventing F magna infection in sheep, because the survival of only a few F magna is potentially fatal in sheep within 6 months after infection.  相似文献

Effect of ivermectin on feeding by Haemonchus contortus in vivo   总被引:1,自引:0,他引:1  
While several in vitro studies have shown that the anthelmintic ivermectin inhibits feeding by parasites, the relevance of this putative site of action in vivo has not been demonstrated. For this study, techniques to measure feeding by Haemonchus contortus in vivo relied on the blood feeding characteristics of the worm, and utilised tritiated inulin administered to sheep intravenously and subsequently measured in worms recovered from abomasa. Nematodes recovered from sheep treated with ivermectin 4 h prior to the [3H]inulin administration showed equivalent feeding levels (over a 1 h period) to those recovered from sheep not treated with ivermectin. In addition, there was no difference in the radioactivity in nematodes of an ivermectin-susceptible and an ivermectin-resistant isolate recovered from individual sheep with concurrent infections after a dose with ivermectin. Ivermectin, therefore, had no effect on feeding by H. contortus in vivo under these experimental conditions. The results are discussed in relation to the dynamics of the expulsion of H. contortus from sheep following ivermectin treatment.  相似文献

The changes in nematode cholinesterase (ChE) activities were examined in relation to the development of resistance in (1) a flock of young grazing sheep, (2) grazing and penned sheep treated with dexamethasone and (3) penned sheep receiving a single mixed infection. Nematodes from grazing sheep with high faecal egg counts (FECs) had higher ChE activities than those from sheep with low FECs. Female nematodes tended to have higher ChE activities than males, and ChE activities in both tended to decline with increasing age of the sheep. The decline in female Trichostrongylus colubriformis ChE activity was associated with a decline in both worm length and in utero egg count. No decline in nematode ChE activity was observed when grazing sheep were treated with dexamethasone. ChE activity of T. colubriformis established in immunosuppressed penned sheep declined 10-20 fold 8 weeks after cessation of treatment. Nematode burdens in the small intestine and abomasum of grazing sheep were significantly correlated, and in individual species they were also correlated with ChE activities. The development of resistance in sheep and the elimination of adult nematode burdens is discussed in relation to gastrointestinal mucosal globule leucocyte numbers, mucus antiparasite activity and the impairment of nematode metabolic function.  相似文献

Five ram-lambs were inoculated into the left conjunctival sac with the 15R isolate of Chlamydia psittaci, recovered from a sheep with keratoconjunctivitis. A sixth ram-lamb was kept in contact with them. The five lambs developed varying degrees of acute conjunctivitis and 14 days later C psittaci could be recovered from the inoculated eyes, from which Branhamella ovis was also isolated. The eyes were examined regularly for four months; C psittaci could not be re-isolated but the eyes developed varying degrees of follicular conjunctivitis. After four months the sheep were treated with corticosteroids in an attempt to reactivate a latent chlamydial infection but no chlamydiae could be isolated. Five months after the start of the experiment the six lambs were inoculated with 15R into the left conjunctival sacs. Acute conjunctivitis developed which was not as severe as after the first inoculation, but C psittaci could only be recovered from the left eyes of three sheep three days after inoculation. The eyes remained chronically affected by follicular conjunctivitis. Six months after the start of the experiment the left eyes were again inoculated with 15R; on this occasion acute conjunctivitis did not develop and chlamydiae could not be isolated. Chronic follicular conjunctivitis persisted until the experiment was terminated three months later.  相似文献

ABSTRACT: We reported previously that sheep affected with footrot (FR) have lower whole-blood selenium (WB-Se) concentrations and that parenteral Se-supplementation in conjunction with routine control practices accelerates recovery from FR. The purpose of this follow-up study was to investigate the mechanisms by which Se facilitates recovery from FR. Sheep affected with FR (n = 38) were injected monthly for 15 months with either 5 mg Se (FR-Se) or saline (FR-Sal), whereas 19 healthy sheep received no treatment. Adaptive immune function was evaluated after 3 months of Se supplementation by immunizing all sheep with a novel protein, keyhole limpet hemocyanin (KLH). The antibody titer and delayed-type hypersensitivity (DTH) skin test to KLH were used to assess humoral immunity and cell-mediated immunity, respectively. Innate immunity was evaluated after 3 months of Se supplementation by measuring intradermal responses to histamine 30 min after injection compared to KLH and saline, and after 15 months of Se supplementation by isolating neutrophils and measuring their bacterial killing ability and relative abundance of mRNA for genes associated with neutrophil migration. Compared to healthy sheep, immune responses to a novel protein were suppressed in FR-affected sheep with smaller decreases in FR-affected sheep that received Se or had WB-Se concentrations above 250 ng/mL at the time of the immune assays. Neutrophil function was suppressed in FR-affected sheep, but was not changed by Se supplementation or WB-Se status. Sheep FR is associated with depressed immune responses to a novel protein, which may be partly restored by improving WB-Se status (> 250 ng/mL).  相似文献

Pregnant sows were injected subcutaneously (s.c.) or intramuscularly (i.m.) with a barium selenate suspension (0.5–1.0 mg Se/kg body weight (b.w.)) and together with control animals fed a commercial diet. No response to the injection was seen either in blood selenium levels or in glutathione peroxidase (GSH-Px) activity in the sows. There was, however, a significant difference in these parameters between piglets born from treated dams and control animals. This status was maintained during the nursing period. In another experiment pigs (20 kg b.w.) on a Se-deficient diet were injected s.c. and i.m. with barium selenate (2.5 mg Se/kg b.w.). The treated groups maintained their blood levels of selenium and GSH-Px activity, although the selenium values in the group treated intramuscularly started to decline after 4 weeks. Organ samples from both groups were equal with regard to selenium at the time of slaughter while the control group showed a rapid decline both in blood selenium levels and GHS-Px activity.  相似文献
